Summary Approximately 600 strains of various bacteria and actinomycetes were tested for their ability to hydroxylate o-acetyltoluidide (AT) at the 4'-position. Nocardia asteroides IFO 3384 and N. globerula IFO 13510 were selected as the best strains to catalyse the hydroxylation of AT. The product was isolated from culture] broth and identified as 4'-hydroxy-o-acetyltoluidide (4'-HAT). When AT was added during the course of cultivation of N. asteroides IFO 3384 and N. globerula IFO 13510, final concentrations of 1.0 mg/ml (a 23% molar conversion yield) and 1.5 mg/ml 4'-HAT (a 34% molar conversion yield) were obtained, respectively, from AT after 7 days cultivation. On the other hand, when resting cells of N. globerula IFO 13510 were used, the concentration of 4'-HAT attained was 4.4 mg/ml, a 69% molar conversion yield, after 4 days reaction.

Ribonucleotide anhydrides have been prepared from corresponding ribonucleoside 5′-S-methyl phosphorothiolates by demethylthiolation with iodine in dry pyridine at room temperature in the presence of appropriate phosphates such as inorganic orthophosphate, inorganic pyrophosphate or glucose 1-phosphate. Thus synthesis of ribonucleotide anhydrides have been achieved and three ribonucleoside 5′-triphosphates (ATP, CTP and UTP), three ribonucleoside 5′-diphosphates (ADP, CDP and UDP) and a pyrophosphate coenzyme (UDPG) have been synthesized and isolated as lithium salts by charcoal treatment followed by ion exchange chromatography. 相似文献

A method for the determination of allethrin and other pesticides in mosquito coils was developed by the combination of shaking extraction and gas chromatography (GC). Allethrin and other pesticides were adequately extracted by shaking for only half an hour with a mixture of toluene and 99% formic acid (5:1). This shaking extraction method was more effective for shortening the extraction time compared with the Soxhlet extraction method, and accurate determination was achieved without the interference from inert materials in the mosquito coils. The recovery of allethrin in various contents from the coils was 96.6 to 97.1%, with a 1.2 to 1.5% coefficient of variation. Furthermore, the recoveries of other pesticides and synergists from the coils were 94 to 102% by this shaking extraction method. 相似文献

The modified nucleosides 4-thio-2-deoxyuridine (s4dU) and 4-thio-2-deoxythymidine (s4dT) are incorporated into dinucleosides, and s4dT is incorporated into a DNA hairpin loop to provide divalent metal ion binding sites. Binding of two different metal ions to these sites is studied, including Cd(II) as an NMR spectroscopy probe and Cu(II) as a reactive metal ion for DNA cleavage. Binding of Cd(II) to 4-thiouridine (s4U) and s4dT nucleosides, s4dU- and s4dT-containing dinucleosides, and a hairpin loop oligonucleotide containing s4dT is monitored by following the change in UV-vis absorbance of the thionucleosides at 340 nm and 21 °C in solutions containing 20.0–40 mM buffer, 1.00 M NaCl, and 15.0 mM BaCl2. Cd(II) binds to the N(3) deprotonated form of s4dT with a binding constant (K=1.1×104 M–1) that is similar to that for Cd(II) binding to d(Tps4T) (K=9.2×103 M–1). Apparent binding constants (Kapp) at pH 7.7 of Cd(II) to dinucleosides d(Gps4T), d(s4TpG), and d(Gps4U) are similar to those of their respective nucleosides s4U and s4dT, suggesting that neither the phosphate diester nor the second nucleoside has a major effect on Cd(II) binding. Binding of Cd(II) to s4U and d(Gps4U) is studied by use of 113Cd NMR and 1H NMR spectroscopy, respectively. Binding strength and stoichiometry of the Cd(II) complex with d(Gps4U) as studied by 1H NMR spectroscopy are similar to that obtained by UV-vis spectroscopy. Cd(II) binds strongly to s4dT in the loop portion of a DNA hairpin loop (Kapp=2.7×103 M–1 at pH 7.7). However, the hairpin loop is moderately destabilized by Cd(II) binding, with a decrease in Tm of 14 °C in the presence of 10.0 mM Cd(II) as determined by optical melting experiments. The patterns of incorporation of d-[G-14C]shikimate and variously labelled 14C-4-(2′-carboxy-phenyl)-4-oxobutyrate into the naphthoquinone nucleus of phylloquinone by maize shoots have been investigated. The results show that (a) the alicyclic ring and C-7 of shikimate give rise to Ring A and either C-1 or C-4, and (b) the phenyl ring, 2′-carboxy and C-4, and C-2 and -3 of 4-(2′-carboxyphenyl)-4-oxobutyrate give rise to Ring A, C-1 and -4 and C-2 and -3. Radioactivity from α-[1-14C]naphthol, 1,4-[1,4-14C]naphthoquinone and [Me-14C]menadione is not incorporated into phylloquinone to any significant extent. 相似文献

Abstract Thymidine was converted to its 5′-epoxy derivative, which was reacted with nucleophiles to give 5′-C-aminomethyl-, 5′-C-bromomethyl-, 5′-C-cyanomethyl, and 5′-C-methoxymethylthymidine derivatives with defined stereochemistry. 5′-C-ally-, 5′-C-hydroxymethyl-, 5′-C-hydroxypropyl-, and 5′-C-(imidazole-4-acetamido)methyl-thymidine derivatives were also prepared. A series of amides, amidines and amidoximes have been made from the corresponding nitrile compounds, to provide potent antagonists and inverse agonists for the CB1 receptor with considerably lower lipophiliciy, higher polar surface area and improved plasma/brain ratios compared to the centrally acting rimonabant. Extensive investigations of ADME and in vivo pharmacological properties led to selection of the amide series and specifically the 4-(4-fluorophenyl)piperidin-4-ol derivative D4. A clear improvement in the peripheral profile over rimonabant was seen, although some contribution of central effect on the pronounced weight reduction in obese mice cannot be ruled out. 相似文献

The production of inosine by microbial conversion of 5′-inosinic acid (IMP) was investigated. Among the various strains of Streptomyces and Bacillus tested, Streptomyces aureus NCIB 9803 was selected for the microbial conversion process due to its high IMP-degrading activity. A maximum conversion yield of 0.43 (86% of the theoretical value) was obtained when IMP was added to the culture medium at 24 h. Kinetic studies with [8-14C] IMP showed that the difference from the theoretical values mainly attributable to the uptake of inosine by S. aureus. 相似文献

Abstract A short multigram synthesis of 2′-O-methylpseudouridine and its phosphoramidite derivative is described which avoids the use of protecting groups on the nitrogens. Abstract Studies on t h e biosynthesis o f the N-nucleoside antibiotics have established that the purine and pyrimidine nucleosides/nucleotides serve as the carbon and nitrogen skeleton, whereas with the C-nucleoside antibioticus, the C-N precursor forthe aglycon is either acetate or glutamate. With the pyrrolopyrimidine nucleoside antibiotics (toyocamycin, tubercidin, and sangi vamycin), either two or three carbons of the N-riboside/ribotide of GTP contribute to carbons 5 and 6 of the pyrrolering and the cyano or carboxamide group.
