The xanthophyll cycle and the water-water cycle had different functional significance in chilling-sensitive sweet pepper upon exposure to chilling temperature (4 °C) under low irradiance (100 µmol m−2 s−1) for 6 h. During chilling stress, effects of non-photochemical quenching (NPQ) on photosystem 2 (PS2) in dithiothreitol (DTT) fed leaves remained distinguishable from that of the water-water cycle in diethyldithiocarbamate (DDTC) fed leaves. In DTT-fed leaves, NPQ decreased greatly accompanied by visible inhibition of the de-epoxidized ratio of the xanthophyll cycle, and maximum photochemical efficiency of PS2 (Fv/Fm) decreased markedly. Thus the xanthophyll cycle-dependent NPQ could protect PS2 through energy dissipation under chilling stress. However, NPQ had a slighter effect on photosystem 1 (PS1) in DTT-fed leaves than in DDTC-fed leaves, whereas effects of the water-water cycle on PS1 remained distinguishable from that of NPQ. , the oxidation level of P700 (P700+) decreased markedly relative to the control and DTT-fed leaves. Both Fv/Fm and NPQ changed little in DDTC-fed leaves accompanied by little change of (A+Z)/(V+A+Z). This is the active oxygen species inducing PS1 photoinhibition in sweet pepper. The water-water cycle can be interrupted easily at chilling temperature. The function of chloroplast ferredoxin quinone reductase (FQR)-dependent flow was examined by comparing a wild type tobacco and a tobacco transformant (ΔndhB) in which the ndhB gene had been disrupted with their antimycin A (AA)-fed leaves upon exposure to chilling temperature (4 °C) under low irradiance (100 μmol m−2 s−1 photon flux density). During the chilling stress, the maximum photochemical efficiency of photosystem (PS) 2 (Fv/Fm) decreased markedly in both the controls and AA-fed leaves, and P700+ was also lower in AA-fed leaves than in the controls, implying that FQR-dependent cyclic electron flow around PS1 functioned to protect the photosynthetic apparatus from chilling stress under low irradiance. Under such stress, non-photochemical quenching (NPQ), particularly the fast relaxing NPQ component (qf) and the de-epoxidized ratio of the xanthophyll cycle pigments, (A+Z)/(V+A+Z), formed the difference between AA-fed leaves and controls. The lower NPQ in AA-fed leaves might be related to an inefficient proton gradient across thylakoid membranes (ΔpH) because of inhibiting an FQR-dependent cyclic electron flow around PS1 at chilling temperature under low irradiance. 相似文献

Chlorophyll (Chl) a and Chl b contents, rate of CO2 gas exchange, quenching coefficients of chlorophyll fluorescence, and endogenous phytohormones have been studied in primary leaves of barley seedlings cultivated under blue (BL) or red (RL) light. Photon flux densities (PFD) were between 0.3 and 12 mol m-2 s-1. Plants grown at PFD of 0.3 mol m-2 s-1 demonstrated in BL tenfold and in RL threefold decreased Chl content compared to plants grown at 12 mol m-2 s-1. Chl a/b ratio increased from 2.3–2.5 to 4.4–4.5 in BL, not in RL, following the decrease in PFD at plant cultivation from 12 to 0.3 mol m-2 s-1. Plants cultivated at weak BL demonstrated severalfold decreased rate of photosynthetic CO2 uptake, whereas decrease in PFD of RL from 12 to 0.3 mol m-2 s-1 caused only 20% de cline in the rate of photosynthesis. Decrease in PFD during a plant cultivation reduced the maximum quantum yield of photosynthesis in BL, not in RL leaves. Light response curves of non-photochemical and photochemical quenching of chlorophyll fluorescence calculated on the basis of absorbed quanta were not affected by PFD of RL during plant cultivation. On the contrary, both non-photochemical quenching and accumulation of QA
-, reduced primary acceptor of Photosystem II, occurred at lower amounts of absorbed quanta in leaves of BL plants grown at 0.3 than at 12 mol m-2 s-1. Two photoregulatory reactions were suggested to exert the light control of the development of photosynthetic apparatus in the range of low PFDs. The photoregulatory reaction saturating by very low PFDs of RL was supposed to be mediated by phytochrome. Phytochrome was proposed to enhance (as related to other pigment-protein complexes of thylakoids) the accu mulation of chlorophyll- b-binding light-harvesting complex of Photosystem II (LHC II). It acts independently of the pigment mediating the second photoregulatory reaction, as evidenced by the results of experiments with plant growth under mixed blue plus red light. The contents of cytokinins and indole-3-acetic acid in a leaf were not significantly affected by either light quality and PFD thus indicating those phytohormones not to be involved into photoregulatory processes. 相似文献

Based on the analysis of fluorescence quenching, the nonphotochemical dissipative processes were investigated in Norway spruce needles during acclimation to winter and spring conditions. The maximum nonphotochemical fluorescence quenching (qNmax) was reached at lower irradiances in winter (up to 310 μmol m-2 s-1) than in spring (about 1130 μmol m-2 s-1), but its values were nearly the same (qNmax = 0.91±0.01) during both winter and spring measurements. In early winter the pronounced initial fluorescence quenching (q0) suggested that nonradiative energy dissipation in the antennae complexes dominated. Significantly lower q0 (by 40-60 % compared to winter needles) during acclimation of needles to spring conditions supported a significant contribution of quenching in the reaction centres. These findings support the hypothesis that the antennae systems and reaction centres cooperate in the protective dissipation of excess excitation energy. Changes in the chloroplast ultra-structure and photochemical function were studied in detached barley (Hordeum vulgare L. cv. Akcent) leaf segments senescing in darkness or in continuous white light of moderate intensity (90 mumol m-2 s-1) for 5 days. A rate of senescence-induced chlorophyll degradation was similar in the dark- and light-senescing segments. The Chl a/b ratio was almost unchanged in the dark-senescing segments, whereas in the light-senescing segments an increase in this ratio was observed indicating a preferential degradation of light-harvesting complexes of photosystem II. A higher level of thylakoid disorganisation (especially of granal membranes) and a very high lipid peroxidation were observed in the light-senescing segments. In spite of these findings, both the maximal and actual photochemical quantum yields of the photosystem II were highly maintained in comparison with the dark-senescing segments. 相似文献

The introduction of a more efficient means of measuring leaf photosynthetic rates under field conditions may help to clarify the relationship between single leaf photosynthesis and crop growth rates of commercial maize hybrids. A large body of evidence suggests that gross photosynthesis (AG) of maize leaves can be accurately estimated from measurements of thylakoid electron transport rates (ETR) using chlorophyll fluorescence techniques. However, before this technique can be adopted, it will first be necessary to determine how the relationship between chlorophyll fluorescence and CO2 assimilation is affected by the non-steady state PPFD conditions which predominate in the field. Also, it must be determined if the relationship is stable across different maize genotypes, and across phenological stages. In the present work, the relationship between ETR and AG was examined in leaves of three maize hybrids by making simultaneous measurements of leaf gas exchange and chlorophyll fluorescence, both under controlled environment conditions and in the field. Under steady-state conditions, a linear relationship between ETR and AG was observed, although a slight deviation from linearity was apparent at low AG. This deviation may arise from an error in the assumption that respiration in illuminated leaves is equivalent to respiration in darkened leaves. The relationship between chlorophyll fluorescence and photosynthetic CO2 assimilation was not stable during fluctuations in incident PPFD. Since even minor (e.g. 20%) fluctuations in incident PPFD can produce sustained ( > 20 s) departures from the mean relationship between ETR and AG, chlorophyll fluorometry can only provide an accurate estimate of actual CO2 assimilation rates under relatively stable PPFD conditions. In the field, the mean value of ETR / AG during the early part of the season (4.70 ± 0.07) was very similar to that observed in indoor-grown plants in the vegetative stage (4.60 ± 0.09); however, ETR / AG increased significantly over the growing season, reaching 5.00 ± 0.09 by the late grain-filling stage. Differences in ETR / AG among the three genotypes examined were small (less than 1% of the mean) and not statistically significant, suggesting that chlorophyll fluorometry can be used as the basis of a fair comparison of leaf photosynthetic rates among different maize cultivars. 相似文献

Leaf discs from spinach were exposed to a photon flux density of 1250 μmol m−2 s−1 at 5°C for 2 or 3 h in ambient air. Photoinhibition of photosystem II (PS II) was measured by means of chlorophyll fluorescence. Recovery of photosystem II was followed at 6°C and 20°C in low light or darkness for periods up to 12 h.
The experimental setup allowed kinetic resolution of different phases of recovery. The experiments revealed a temperature dependent dark recovery phase and two distinct light- and temperature dependent phases: (1) A relatively fast, light dependent recovery phase occurred in parallel with partial recovery of basic fluorescence at 6°C and 20°C. A population of PS II centers with very slow fluorescence induction kinetics, which had accumulated during photoinhibition treatment, disappeared during this phase. This fast recovery phase is proposed to represent reactivation of photoinhibited PS II, without dissassembly or incorporation of new D1 -protein. (2) A relatively slow light-dependent recovery phase took place at 20°C, but not at 6°C. In the presence of the chloroplast translation inhibitor streptomycin, part of the 2nd phase was inhibited. This phase is proposed to involve assembly of new Photosystem II centers, which is partly dependent on de novo synthesis of D1 -reaction center protein, but presumably is also using a preexisting pool of D1 -protein. Cold acclimation of the leaves resulted in a decreased sensitivity for photoinhibition of photosystem II. Recovery of photoinhibited photosystem II at 6°C of the cold-acclimated leaves was faster than in non-acclimated leaves, but this effect can be ascribed to diminished photoinhibitory damage. 相似文献

The effects of low temperature acclimation and photoinhibitory treatment on Photosystem 2 (PS 2) have been studied by thermoluminescence and chlorophyll fluorescence decay kinetics after a single turnover saturating flash. A comparison of unhardened and hardened leaves showed that, in the hardened case, a decrease in overall and B-band thermoluminescence emissions occurred, indicating the presence of fewer active PS 2 reaction centers. A modification in the form of the B-band emission was also observed and is attributed to a decrease in the apparent activation energy of recombination in the hardened leaves. The acclimated leaves also produced slower QA
– reoxidation kinetics as judged from the chlorophyll fluorescence decay kinetics. This change was mainly seen in an increased lifetime of the slow reoxidation component with only a small increase in its amplitude. Similar changes in both thermoluminescence and fluorescence decay kinetics were observed when unhardened leaves were given a high light photoinhibitory treatment at 4°C, whereas the hardened leaves were affected to a much lesser extent by a similar treatment. These results suggest that the acclimated plants undergo photoinhibition at 4°C even at low light intensities and that a subsequent high light treatment produces only a small additive photoinhibitory effect. Furthermore, it can be seen that photoinhibition eventually gives rise to PS 2 reaction centers which are no longer functional and which do not produce thermoluminescence or variable chlorophyll fluorescence.Abbreviations D1

The effects of two light treatments (photosynthetically active photon flux density of either 650 or 1950 µmol m–2 s–1) on the photochemical efficiency of Photosystem II (PS II) (measured as variable to maximum fluorescence ratio) and on the xanthophyll cycle components was studied in wilted Zea mays leaves. For comparison, these parameters were followed under the same light conditions in well-hydrated leaves maintained either in normal or CO2-free air. The net CO2 assimilation of dehydrated leaves declined rapidly as their relative water content (RWC) decreased from 100 to 60% while the PS II efficiency measured after a prolonged dark period of 16 h declined only when RWC leaves was lower than 60%. Furthermore, drought caused an increase in the pool size of the xanthophyll cycle pigments and the presence of a sustained elevated level of zeaxanthin and antheraxanthin at the end of the long dark period. The leaf water deficit enhanced the sensitivity of PS II efficiency to light exposure. During illumination, strong inhibition of PS II efficiency and large violaxanthin deepoxidation was observed in wilted leaves even under moderate photon flux density compared to control leaves in the same conditions. After 2 h of darkness following the light treatment, the PS II efficiency that is dependent on the previous PPFD, decreased with leaf water deficit. Moreover, zeaxanthin epoxidation led to an accumulation of antheraxanthin in dehydrated leaves. All these drought effects on PS II efficiency and xanthophyll cycle components were also obtained in well-hydrated leaves by short-term CO2 deprivation during illumination. We conclude that the increased susceptibility of PS II efficiency to light in wilted maize leaves is mainly explained by the decrease of CO2 availability and the resulting low net CO2 assimilation. 相似文献

Photoinhibition of photosynthesis and its recovery were studied in intact barley ( Hordeum vuigare L. cv. Gunilla) leaves grown in a controlled environment by exposing them to two temperatures, 5 and 20°C, and a range of photon flux densities in excess of that during growth. Additionally, photoinhibtion was examined in the presence of chloramphenicol (CAP, an inhibitor of chloroplast protein synthesis) and of 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU). Susceptibility to photoinhibition was much higher at 5 than at 20°C. Furthermore, at 20°C. CAP exacerbated photoinhibition strongly, whereas CAP had little additional effect (10%) at 5°C. These results support the model that net photoinhibition is the difference between the inactivation and repair of photosystem II (PSII); i.e. the degradation and synthesis of the reaction centre protein, Dl. Furthermore, the steady-state extent of photoinhibition was strongly dependent on temperature and the results indicated this was manifested through the effects of temperature on the repair process of PSII. We propose that the continuous repair of PS II at 20°C conferred at least some protection from photoinhibition. At 5°C the repair process was largely inhibited, with increased photoinhibition as a consequence. However, we suggest where repair is inhibited by low temperature, some protection is alternatively conferred by the photoinhibited reaction centres. Providing they are not degraded, such centres could still dissipate excitation energy non-radiatively, thereby conferring protection of remaining photochemically active centres under steady-state conditions.

In order to evaluate the photosynthetic activity of a C3 leaf from the electron transport rate (ETR) of photosystem 2 (PS2), a new method was devised and examined using leaves of sweet potato. In this method, both surfaces of a leaf were sealed with transparent films to stop the gas exchange between the leaf and the atmosphere; hence the functions of both photosynthetic assimilation (CO2 uptake) and photorespiration (CO2 release) are restricted to the inside of the leaf. After both functional rates became equally balanced, ETR of the sealed leaf (ETRseal) was determined from the chlorophyll fluorescence. The measurements were conducted at different irradiances and leaf temperatures and by using leaves of different age. Under each measurement condition, ETRseal showed a close positive relationship with the photosynthetic potential, or the gross photosynthetic rate measured in the air of 2 % O2 (P

Abstract Comparative studies on the low temperature fluorescence emission of whole cells, purified chlorophyll-protein (CP) complexes and on patterns noted in sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) for chlorophyll-protein complexes and chloroplast membrane polypeptides of Scenedesmus obliquus with several distinct mutant classes has allowed further correlation between the fluorescence emission bands seen at 77K and the known chlorophyll-protein complexes. In mutants deficient in photosystem II (PS-II; total loss of the reducing side) the fluorescence emission spectra showed only two peaks, i.e., 686 and 718 nm, but in the wild type, in mutants lacking the oxidizing side of PS-II and in phenotypes missing the CP-a1 complex (and P-700 activity) all three emission bands at 686, 696 and 718 nm were present. In a mutant lacking the light-harvesting CP-a/b complex the emission peak at 686 nm was strongly reduced and the longer wavelength emissions predominated. Gel electrophoresis studies showed that the PS-II (reducing side) mutants lacked the polypeptides of apparent molecular weight 54 and 51 kilodaltons and the chlorophyll-protein complex, CP-aII, of apparent molecular weight 32 kilodaltons. Contrarily, the loss of the oxidizing side of PS-II did not result in any alteration of these components. Genetic deletion of CP-a1 did not alter significantly the long wavelength emission even though the isolated CP-a1 shows the low temperature-dependent long wavelength emission comparable to that seen in the whole cell. It was deduced that remaining PS-I antennae chlorophylls must account for the emission seen at 718 nm. The absence of the CP-a/b complex and the strong simultaneous decrease of the 686 nm emission strengthens the concept that this complex is the primary emitter of fluorescence at room temperature. Its absence facilitated the detection of the CP-aII complex in SDS-PAGE and enhanced the in vivo fluorescence by the two photosystems. Parallel experiments with two mutants which green and develop, one to the wild-type and the other to the CP-a/b deficient phenotype, provided additional evidence for the source of the low temperature emission bands. 相似文献

Adult trees of Quercus petraea were submitted to controlled water shortage in a natural stand near Nancy, France. Diurnal course of net CO2 assimilation rate (A) was measured in situ together with chlorophyll a fluorescence determined on dark adapted leaves. In 1990, trees experienced a strong water stress, with predawn and midday leaf water potentials below –2·0 and –3·0 MPa, respectively. Diurnal course of A of well-watered trees exhibited sometimes important midday decreases in A related to high temperature and vapour pressure deficit. Decreases in initial (Fo) and maximal (Fm) fluorescence and sometimes in photochemical efficiency of photosystem II (Fv/Fm) were observed and probably revealed the onset of mechanisms for thermal de-excitation. These mechanisms were shown to be sensitive to dithiothreitol. All these effects were reversible and vanished almost completely overnight. Therefore, they may be considered as protective mechanisms adjusting activity of photosystem II to the electron requirement for photosynthesis. Water stress amplified these reactions: A was strongly decreased, showing important midday depression; diurnal reductions in Fm and Fv/Fm were enhanced. The same trends were observed during summer 1991, despite a less marked drought. These protective mechanisms seemed very effective, as no photoinhibitory damage to PS II could be detected in either water stressed or control trees. 相似文献

Blue light effects on the acclimation of energy partitioningcharacteristics in PSII and CO2 assimilation capacity in spinachto high growth irradiance were investigated. Plants were grownhydroponically in different light treatments that were a combinationof two light qualities and two irradiances, i.e. white lightand blue-deficient light at photosynthetic photon flux densities(PPFDs) of 100 and 500 µmol m–2 s–1. The CO2assimilation rate, the quantum efficiency of PSII (PSII) andthermal dissipation activity / in young, fully expanded leaves were measured under 1,600 µmol m–2 s–1white light. The CO2 assimilation rate and PSII were higher,while / was lower in plants grown under high irradiancethan in plants grown under low irradiance. These responses wereobserved irrespective of the presence or absence of blue lightduring growth. The extent of the increase in the CO2 assimilationrate and PSII and the decrease in / by high growth irradiance was smaller under blue light-deficient conditions. These resultsindicate that blue light helps to boost the acclimation responsesof energy partitioning in PSII and CO2 assimilation to highirradiance. Similarly, leaf N, Cyt f and Chl contents per unitleaf area increased by high growth irradiance, and the extentof the increment in leaf N, Cyt f and Chl was smaller underblue light-deficient conditions. Diurnal time courses of net CO2 assimilation rates, stomatal conductance and light-driven electron fluxes were measured in situ on attached leaves of 30-year-old Turkey oak trees (Quercus cerris L.) under natural summer conditions in central Italy. Combined measurements of gas exchange and chlorophyll a fluorescence under low O2 concentrations allowed the demonstration of a linear relationship between the photochemical efficiency of PSII (fluorescence measurements) and the apparent quantum yield of gross photosynthesis (gas exchange). This relationship was used under normal O2 to compute total light-driven electron fluxes, and to partition them into fractions used for RuBP carboxylation or RuBP oxygenation. This procedure also yielded an indirect estimate of the rate of photorespiration in vivo. The time courses of light-driven electron flow, net CO2 assimilation and photorespiration paralleled that of photosynthetic photon flux density, with important afternoon deviations as soon as a severe drought stress occurred, whereas photochemical efficiency and maximal fluorescence underwent large but reversible diurnal decreases. The latter observation indicated the occurrence of a large non-photochemical energy dissipation at PSII. We estimated that less than 60% of the total photosynthetic electron flow was used for carbon assimilation at midday, while about 40% was devoted to photorespiration. The rate of carbon loss by photorespiration (R1) reached mean levels of 56% of net assimilation rates. The potential application of this technique to analysis of the relative contributions of thermal de-excitation at PSII and photorespiratory carbon recycling in the protection of photosynthesis against stress effects is discussed. 相似文献

An optode device for net-photosynthesis measurements, based on oxygen-depending quenching of fluorescence from O2-specific sensors, and PAM fluorometry have been used to study diurnal courses of net-photosynthesis and the Fv/Fm ratio of the submerged plant Lagarosiphon major. Plants were pre-cultivated and studied in large mesocosm flow-through outdoor tanks under 50% and 80% shade cloth, respectively. Growth under the different shade cloths resulted in similar light compensation points (∼20 μmol photons m−2 s−1), but strongly different light saturation levels, with about 150 μmol m−2 s−1 for plants grown under 80% shade cloth and about 350 μmol m−2 s−1 for plants grown under 50% shade cloth. Plants under both growth conditions showed a transient reduction of the maximum Fv/Fm value in the afternoon (down to 70% of the morning control values under 80% shade cloth and down to 85% under 50% shade cloth), which was not accompanied by a reduction of the net photosynthetic rate. This indicated that the fluorescence parameter Fv/Fm must not be a reliable indicator of the rate of photosynthesis under all conditions. The new photo-optical device became evidenced as a valuable tool not only for laboratory experiments, but also for field studies of gas exchange of submerged plants. 相似文献
