Nodulated soya bean (Glycine max L.) plants at the early floweringstage were allowed to assimilate 13CO2 under steady-state conditions,with a constant 13C abundance, for 8 h in the light. The plantswere either harvested immediately or 2 d after the end of the13CO2 feeding, divided into young leaves (including flower buds),mature leaves, stems+petioles, roots and nodules; the 13C abundancein soluble carbohydrates, organic acids, amino acids, starchand poly-ß-hydroxybutyric acid was determined witha gas chromatography-mass spectrometry. The rapid turnover of 13C in the sucrose pools observed in allorgans of the plants showed that sucrose was the principal materialin the translocation stream of primary products of photosynthesis.At the end of the 13CO2 exposure, sucrose in the mature leavesas the major source organs and in the stems+petioles was labelledwith currently assimilated carbon to about 75 per cent, whereasa much higher labelling of sucrose was found in the roots andin the nodules. This suggests the existence of two or more compartmentedpools of sucrose in mature leaves and also in stems+petioles. The relative labelling patterns of individual organic acidsand amino acids were similar in various plant organs. However,the rapid turnover of succinate and glycine was characteristicof nodules. Treatment with a high concentration of nitrate inthe nutrient media increased the turnover rate of amino acidcarbon in shoot organs and roots, while it markedly decreasedthe labelling of amino acids in nodules. The cyclitols, exceptfor D-pinitol, were significantly labelled with assimilated13C in mature leaves, but in nodules, the labelling was verymuch less. In the nodules, which were actively fixing atmospheric nitrogen,a large proportion (80–90 per cent) of currently assimilatedcarbon was found as sucrose and starch at the end of the 13CO2feeding. This was also true of the roots. On the other hand,in young growing leaves, the distribution of currently assimilatedcarbon into sucrose, starch and other soluble compounds wasmuch less. This suggests that a large amount of carbon assimilatedby and translocated to young leaves was used to make up structuralmaterials, mainly protein and cell wall polymers synthesis,during the light period. Tracer amounts of atmospheric [13N]-Iabelled ammonia gas, wereabsorbed by leaves of Lupinus albus and Helianthus annuus inboth the light and the dark. Exogenous [13N]-ammonia was onlyabsorbed in the dark when the feeding occurred shortly aftera period of illumination and the tissue was not depleted ofits carbohydrate reserves (e.g. starch). Incorporation of the[13N]-ammonia appeared to occur via the leaf glutamine synthetase/glutamatesynthase (GS/GOGAT) cycle since 2.0 mol m–3 MSX, an inhibitorof the GS reduced uptake in both the light and dark. Photosyntheticincorporation of 11CO2 was not affected by this treatment Therate of movement of [13N]-assimilates in the petiole of attachedleaves of Helianthus and Lupinus was similar to that of the11Cl-photo assimilates. Export of both [13N] and [11C]-Iabelledassimilates from the leaf and movement in the petiole in boththe light and the dark was inhibited by source leaf anoxia (i.e.nitrogen gas). Translocation was re-established at the samerate when the feed leaf was exposed to gas containing more than2% O2 which permitted dark respiration to proceed. After aninitial feeding of either 11CO2 or [13N]-ammonia at ambient(21%) O2 exposure of the source leaf to 2% O2, or 50% O2 didnot alter the rates of translocation, indicating that changesin photosynthetic activity in the source leaf due to photorespiratoryactivity need not markedly alter, at least during the shortperiod, the loading and translocation of either [11C ] or [13N]-labelledleaf products. Sunflower (Helianthus annuusL.) and oilseed rape (Brassica napusL.) were grown at constant temperatures of 30 ?C (warm) and13 ?C (cold). Maximal rates of photosynthesis between 5 ?C and35 ?C were at higher temperatures in sunflower than rape. Photosyntheticrate over 4 h at the growth temperature declined in warm-andcold-grown rape and cold-grown sunflower, but remained constantin warm-grown sunflower. The stimulation of photosynthesis by2.0 kPa O2 compared to 21 kPa O2 declined with decreasing temperature.At 10 ?C in warm-grown rape photosynthesis was insensitive to2.0 kPa O2. However, sensitivity to low O2 continued at 10 ?Cin warm-grown sunflower. Carbohydrates accumulated in the cold,particularly fructose, glucose and sucrose in warm-grown sunflowertransferred to 13 ?C. By monitoring changes of 14C in leaves after the assimilationof 14CO2, the rates of carbon export from leaves, pool sizesand carbon fluxes between them were estimated. The transferof warm- and cold-grown rape to 13 ?C and 30 ?C, respectively,had little effect on these parameters over 22 h. However, exportof carbon from sunflower leaves at 13 ?C was markedly less thanat 30 ?C, irrespective of the growth temperature, due to slowerexport from the transport pool. The rapid suppression of carbonexport at 13 ?C in warm-grown sunflower may be due to inhibitedtranslocation rather than reduced sink demand in the cold. It is concluded that assimilate utilisation is more depressedin the cold than is photosynthesis; this imposes a greater restrictionon biomass production in sunflower than in rape. A study was made of the incorporation of 14C by intact leavesof Coffea arabica (cultivars Mundo Novo, Catuai, 1130–13,and H 6586–2) and Coffea canephora (cultivar Guarini)supplied with gas mixtures containing 14CO2 under controlledconditions. Samples of the leaves were combusted and the 14Cin the CO2 produced measured using a liquid scintillation counter.The results were used to estimate photosynthetic rates. Theeffects of changing the partial pressures of O2 and CO2 on thephotosynthetic rate were studied and estimates made of the CO2compensation point and photorespiration. The data obtained show differences between the mean net photosyntheticrates of the C. arabica cultivars (6·14 mg CO2 dm–2h–1) and the mean rate for the C. canephora cultivar (3·96mg CO2 dm–2 h–1). The cultivar of the latter speciesphotorespired more rapidly than the cultivar Catuai of C. arabica.Rates of photosynthesis in coffee measured using the 14CO2 methodwere similar to rates obtained by others using an infrared gasanalyser. The 14CO2 method proved to be reliable for photosyntheticmeasurements and the apparatus is suitable for use in fieldconditions.  相似文献

Ten-day old kidney bean plants (Phaseolus vulgaris L. cv. Shin-edogawa)were exposed to 2.0 and 4–0 parts 10–6 NO2, and0.1, 0.2, and 0.4 parts 10–6 O3 alone or in combinationfor 2, 4, and 7 d. The effects of these air pollutants wereexamined with respect to the growth, partitioning of assimilates,nitrogen uptake, soluble sugar content, and root respiration. Decreased dry matter production was significant with all treatmentsexcept 2.0 parts 10–6 NO2 and 0.1 parts 10–6 O3.Exposure to mixtures of the gases produced more severe suppressionof growth than exposure to the single gases. Root/shoot ratiowas significantly lowered at 7 d by the gas treatments otherthan 2.0 parts 10–6 NO2 and 0.1 parts 10–6 O3. Thetotal nitrogen content of plants was increased by all treatments;the higher percent of nitrogen found with O3 exposure will resultfrom the growth retardation which increases the concentrationof nitrogen in the plants because the absorption of nitrogenby roots was unaffected. The combination of O3 with NO2 significantlydecreased the assimilation of NO2 by the plants. The concentration of soluble sugars in roots was decreased bythe gas treatments. There was a strong positive correlationbetween soluble sugar content and dry weight of the roots harvestedat 7 d. Root respiration was relatively unchanged until 5 dand then decreased significantly at 7 d by 2.0 parts 10–6NO2 and 0–2 parts 10–6 O3. Retarded growth of theroots and the decreased root respiration may be due to diminishedtranslocation of sugars from leaves to roots caused by exposureto air pollutants. Changes in growth and yield parameters, and 14CO2 and (U-14C)sucrose incorporation into the primary metabolic pool, and essentialoil have been investigated under Mn-deficiency and subsequentrecovery in Mentha piperita, grown in solution culture. UnderMn-deficiency, CO2 exchange rate, total chlorophyll, total assimilatoryarea, plant dry weight, and essential oil yield were significantlyreduced, whereas chlorophyll a/b ratio, leaf area ratio andleaf stem ratio significantly increased. In leaves of Mn-deficientplants, 14CO2 incorporation into the primary metabolic pool(ethanol-soluble and -insoluble) and essential oil were significantlylower, whereas (U-14C) sucrose incorporation into these componentswas significantly higher as compared to the control. Among theprimary metabolites, the label was maximum in sugars, followedby organic acids and amino acids. A higher label in these metaboliteswas, in general, observed in stems of Mn-deficient plants ascompared to the control. Mn-deficient plants supplied with completenutrient medium for 3 weeks exhibited partial recovery in growthand yield parameters, and essential oil biogenesis. Thus, underMn-deficiency and subsequent recovery, the levels of primaryphotosynthetic metabolites and their partitioning between leafand stem significantly influence essential oil biogenesis. Stands of spring wheat grown in open-top chambers (OTCs) wereused to assess the individual and interactive effects of season-longexposure to elevated atmospheric carbon dioxide (CO2 and ozone(O3) on the photosynthetic and gas exchange properties of leavesof differing age and position within the canopy. The observedeffects were related to estimated ozone fluxes to individualleaves. Foliar chlorophyll content was unaffected by elevatedCO2 but photosynthesis under saturating irradiances was increasedby up to 100% at 680 µmol mol–1 CO2 relative tothe ambient CO2 control; instantaneous water use efficiencywas improved by a combination of increased photosynthesis andreduced transpiration. Exposure to a seasonal mean O3 concentration(7 h d–1) of 84 nmol mol–1 under ambient CO2 acceleratedleaf senescence following full expansion, at which time chlorophyllcontent was unaffected. Stomatal regulation of pollutant uptakewas limited since estimated O3 fluxes to individual leaves werenot reduced by elevated atmospheric CO2, A common feature ofO3-treated leaves under ambient CO2 was an initial stimulationof photosynthesis and stomatal conductance for up to 4 d and10 d, respectively, after full leaf expansion, but thereafterboth variables declined rapidly. The O3-induced decline in chlorophyllcontent was less rapid under elevated CO2 and photosynthesiswas increased relative to the ambient CO2 treatment. A/Ci analysessuggested that an increase in the amount of in vivo active RuBisCOmay be involved in mitigating O3-induced damage to leaves. Theresults obtained suggest that elevated atmospheric CO2 has animportant role in restricting the damaging effects of O3 onphotosynthetic activity during the vegetative growth of springwheat, and that additional direct effects on reproductive developmentwere responsible for the substantial reductions in grain yieldobtained at final harvest, against which elevated CO2 providedlittle or no protection. Well-nodulated soya bean (Glycine max L.) plants were allowedto assimilate 13CO2 for 10 h in the light, under steady-stateconditions in which CO2 concentration and 13C abundance wereboth strictly controlled at constant levels. The respiratoryevolution of 13CO2 from roots and nodules and 13C incorporationinto various metabolic fractions were measured during the 13CO2feeding and subsequent 48 h chase period. CO2 respired from nodules was much more rapidly labelled with13C than that from roots. The level of labelling (percentageof carbon currently assimilated during the 13COM2 feeding period)of CO2 respired from nodules reached a maximum of about 87 percent after 4 h of steady-state l3CO2 assimilation and thereafterremained fairly constant. The absolute amount of labelled carbonevolved by the respiration of the nodules during the 10 h 13CO2feeding period was 1·5-fold that of root respiration.These results demonstrated that the currently assimilated (labelled)carbon was preferentially used to support nodule respiration,while root respiration relied considerably on earlier (non-labelled)carbon reserved in the roots. Sucrose pools were mostly composed of currently assimilatedcarbon in all tissues of the plants, since the levels of labellingaccounted for 86–91 per cent at the end of the 13CO2 feeding.In the nodules, the kinetics and levels of sucrose labellingwere in fairly good agreement with those of respired CO2, whilein the roots, the level of labelling of respired CO2 was significantlylower than that of sucrose. Succinate and malate were highly labelled in both roots andnodules but they were labelled much more slowly than sucroseand respired CO2. The kinetics and levels of labelling of theseKrebs cycle intermediates resembled those of major amino acidswhich are derived directly from Krebs cycle intermediates. Itis suggested that large fractions of organic acids in noduleswere physically separate from the respiration site. Methionine sulfoximine (MSO) greatly reduced the carbon dioxideexchange rate (CER) of detached wheat (Triticum aestivvm L.cv Roland) leaves in 21% O2, but only slightly reduced it in2% O2. A supply of 50 mM NH4Cl had little effect on the CERirrespective of the O2 concentration. A simultaneous additionof glutamine and MSO protected against the inhibition of photosynthesisto a considerable extent and caused the accumulation of moreNH3 than did the addition of MSO alone. Fixation of 14CO2 in wheat leaves was inhibited by MSO treatmentin 22% O2, and there was decreased incorporation of 14G intoamino acids and sugars and increased label into acid fractions.The addition of MSO and glutamine together eliminated the effectof MSO on the photosynthetic 14CO2 fixation pattern. NH4Cl stimulatedthe synthesis of amino acids from 14CO2, especially the synthesisof serine in 22% O2. For the first time, spruce shoots (Picea abies [L.] Karst.)were fumigated in vivo with 13N-labelled NO2 with the aim ofelucidating the mechanism of NO2 trapping in the apoplastof the substomatal cavity. Uptake by the needles could be monitoredon-line, and a quantitative analysis of the activity recordsdelivered a deposition velocity in agreement with the commondry deposition estimates and ruled out rapid export processes.A fast extraction procedure was applied which revealed thatNO2 did not produce any detectable traces of nitrite. In needlesin which the enzymes of nitrate reduction were not induced byprior fumigation with NO2, incorporation of NO2 was partiallyinhibited as compared to the fully induced shoots which tookup and assimilated NO2 as expected from a constant influx. Theonly labelled inorganic species found in the extracts was nitrate(60%), whereas the rest of the label (40%) was assimilated organicnitrogen.A quantitative analysis of the data shows that thereaction of NO2 in the apoplast yields at least three timesmore nitrate than nitrite, so that the existing models aboutthe apoplastic trapping reaction, disproportionation or antioxidantscavenging, which both postulate substantial production of nitrite,have to be reconsidered. The rates of photosynthetic 14CO2 fixation by Chlorella vulgarisllh, grown under high CO2, were determined between 4 to 37°Cwith air containing from 300 to 13,000 ppm 14CO2. When the CO2level was increased, both the rate of photosynthesis and theoptimum temperature for maximum photosynthesis increased. Themaximum photosynthetic rate was reached at 12°C with 300ppm l4CO2. Among the photosynthetic products fromed at 300 ppm 14CO2, glycolatedecreased greatly when the temperature was raised from 20 to30°C. At 3,000 ppm 14CO2 an insignificant amount of glycolatewas formed at all temperatures, whereas 14C-incorporation intothe insoluble fraction, sucrose, and the lipid fraction wassignificantly higher than at 300 ppm 14CO2. The 14C in sucrosewas greatly increased and the radioactivity in the insolublefraction decreased when the temperature was raised from 28 to36°C. (Received April 8, 1980; )  相似文献

The reductive carboxylic acid cycle appears to be the majorcarbon assimilation pathway in green sulfur bacteria, Chlorobiumthiosulfatophilum. While cyanide was relatively ineffectivein inhibiting the bacterial photosynthetic CO2 fixation, photosynthesiswas strongly impaired in an O2-containing atmospheric environment.No glycolate formation was detected in Chlorobium under an O2atmosphere. In the purple sulfur bacteria, Chromatium vinosum,however, photosynthesis was highly sensitive to cyanide, andin a short-term incubation (up to 10 min) photosynthetic CO2fixation was found to be relatively indifferent to an O2-containingatmosphere of up to 100% O2. Significant formation of glycolatewas demonstrated upon a very brief exposure to O2, whereas thetotal photosynthetic CO2 fixation was slightly affected. However,ribulose-1,5-bisphosphate carboxylase activity in Chromatiumextract was competitively inhibited by O2 in a similar mannerto the higher plant enzyme, K1(O2) value being 0.7 mM at pH8.2. The percentage of incorporation of 14CO2 into glycolateand glycine under an O2-containing atmosphere declined withincreasing levels of bicarbonate concentrations in the medium.The Warburg effect and biosynthetic mechanisms involving glycolatein photosynthetic bacteria are discussed. 1 This is paper XXXIX in the series "Structure and Functionof Chloroplast Proteins". Single, seed-grown plants of ryegrass (Lolium perenne L. cv.Melle) were grown for 49 d from the early seedling stage ingrowth cabinets at a day/night temperature of 20/15 C, witha 12 h photoperiod, and a CO2 concentration of either 340 or680µI 1–1 CO2. Following complete acclimation tothe environmental regimes, leaf and whole plant CO2 effluxesand influxes were measured using infra-red gas analysis techniques.Elevated CO2 increased rates of photosynthesis of young, fullyexpanded leaves by 35–46% and of whole plants by morethan 50%. For both leaves and whole plants acclimation to 680µI–1 CO2 reduced rates of photosynthesis in bothCO2 regimes, compared with plants acclimated to 340µll–1. There was no significant effect of CO2 regime onrespiration rates of either leaves or whole plants, althoughleaves developed in elevated CO2 exhibited generally lower ratesthan those developed in 340µI I–1 CO2. Initially the seedling plants in elevated CO2 grew faster thantheir counterparts in 340µI I–1 CO2, but this effectquickly petered out and final plant weights differed by onlyc. 10%. Since the total area of expanded and unexpanded laminaewas unaffected by CO2 regime, specific leaf area was persistently13–40% lower in elevated CO2 while, similarly, root/shootratio was also reduced throughout the experiment. Elevated CO2reduced tissue nitrogen contents of expanded leaves, but hadno effect on the nitrogen contents of unexpanded leaves, sheathsor roots. The lack of a pronounced effect of elevated CO2 on plant growthwas primarily due to the fact that CO2 concentration did notinfluence tiller (branch) numbers. In the absence of an effecton tiller numbers, any possible weight increment was restrictedto the c. 2.5 leaves of each tiller. The reason for the lackof an effect on tillering is not known. A long-term, steady-state 13CO2 assimilation system at a constantCO2 concentration with a constant 13C abundance was designedand applied to quantitative investigations on the allocationof photoassimilated carbon in nodulated soya bean (Glycine maxL.) plants. The CO2 concentration in the assimilation chamberand its 13C abundance were maintained constant with relativevariances of less than ±0.5 per cent during an 8-h assimilationperiod. At the termination of 8-h 13CO2 assimilation by plantsat early flowering stage, the currently assimilated carbon relativeto total tissue carbon (measured by the degree of isotopic saturation)were for young leaves (including flower buds), 13.9 per cent;mature leaves, 15.7 per cent; stems+petioles, 5.9 per cent;roots, 5.4 per cent and nodules, 6.9 per cent, 48 h after theend of the 13CO2 assimilation period, they were 12.3, 7.5, 7.4,6.8 and 6.1 per cent, respectively. The treatment with a highconcentration of nitrate in the nutrient media significantlydecreased the allocation of 13C into nodules. Experiments on13CO2 assimilation by plants at the pod-filling stage were alsoconducted. Labelling by 13C was weaker than at the early floweringstage, but an intense accumulation of 13C into reproductiveorgans was observed. Attached leaves of sunflower (Helianthus annuus L. var. Mennonite)with water potentials of –5 to –18 ? 105 Pa, wereexposed for different times to 300 vpm CO2 containing 14CO2and 21 or 1.5% O2. 14C accumulated linearly with time in bothO2 concentrations and at all stresses. 3-Phosphoglyceric acidwas saturated with 14C after 10 min in unstressed plants atboth O2 concentrations but with increasing stress the rate ofaccumulation and the specific activity decreased. With decreasingleaf water potential there was accumulation of radioactivityin the glycolate pathway intermediates glycine and serine. Otheramino acids contained a slightly larger proportion of assimilatedcarbon as water potential decreased. The specific activitiesof all compounds were smaller with stress. In contrast to theamino acids less radioactivity accumulated in sugars, organicacids, and sugar phosphates and their specific activities decreasedwith stress. The radioactive labelling patterns and specificactivity measurements are interpreted as showing increased carbonflux in the glycolate pathway and inhibition of the metabolismof serine to sucrose. These changes are related to previousresults showing that with stress photo respiration increasesas a proportion of photosynthesis. Lowering the O2 concentrationto 1.5% decreased the accumulation of radioactivity in glycineand stopped photorespiration. It increased the amount of radioactivityin serine and sucrose but did not greatly change specific activities.Oxygen effects were independent of water stress. Patterns of initial photosynthetic CO2 incorporation were determinedfor some seagrasses and were related to activities of primarycarbon fixing enzymes, carbonic anhydrase activities, and 13Cvalues. According to the incorporation patterns, Cymodocea nodosa wasa C4 species while Thalassia hemprichli and Thalassodendronciliatum were C3 plants. Halophila stipulacea showed an unusualincorporation pattern which could be viewed as intermediatebetween typical C3 and C4 pathways. The activity ratios of ribulose-l,5-bisphosphate carboxylase (RUBPcase) to phosphoenolpyruvatecarboxylase (PEPcase) were about 3 for Thalassodendron ciliatumand 1 for Cymodocea nodosa and Halophila stipulacea. The lattervalue, which is intermediate to ratios found in terrestrialC3 and C4 plants, may correlate with the incorporation patternsfound for Halophila stipulacea. Since the C4 seagrass lackedthe Kranz anatomy, it may, in addition, point to a flexibleincorporation potential for these plants. The high 13C values found in these and other seagrasses didnot correlate with their photosynthetic pathways as in terrestrialplants. This discrepancy is probably due to a ‘closedsystem’ type of photosynthesis in which CO2 is efficientlyutilized. The C3 species which utilize CO2 enzymatically must convertexogenous HCO-3 to CO2 internally. Even though carbonic anhydraseactivities were very low, conversion rates seemed to be sufficientfor high rates of photosynthesis. Since enzymatic fixation ratesapproached photosynthetic rates even at CO2 saturation, thelimitation for these seagrasses to express their high photosyntheticpotential is most probably the HCO3 uptake system.  相似文献

Inair-grown cells, O2 evolution dependent on low, but not high,HCO3 concentrations was strongly inhibited by the carbonicanhydrase inhibitor acetazolamide. Cells grown with 3% CO2 exhibitedlow rates of O2 evolution at low external inorganic C; however,after 30 min in air O2 evolution rates at low inorganic C approachedthose of air-grown cells. These results are compatible withthe view that Scenedesmus develops a ‘CO2 concentratingmechanism’ in air, with carbonic anhydrase as an importantconstituent When 3% CO2-grown cells were subjected to air-level of CO2,just a transient decline in NO3 utilization was observed,but in the presence of acetazolamide the rate of the processdecreased drastically in response to the decrease in the CO2level. In CO2-free air NO3 was taken up at high ratesbut in a deregulated manner, leading to release of NH4+. A portionof the NO3 taken up in the absence of CO2 was apparentlyassimilated Cellular nitrate reductase (NR) activity initially decreasedbut subsequently recovered after a transition from 3% CO2 toair. In the presence of acetazolamide, a persistent decreasein NR activity was observed. Cellular glutamine synthetase (GS)activity increased after transition from 3% CO2 to air, theactivity increase being unaffected by acetazolamide. NH4+ releaseto the medium in the presence of L-methionine-D, L-sulphoximine(MSO) transiently increased in 3% CO2-grown cells in responseto a transfer to air. MSO-induced NH4+ release was in fact higherin air-grown cells than in 3% CO2-grown cells. Glycollate wasinitially released after transition from 3% CO2 to air, butthere was no difference in glycollate release between MSO-treatedand untreated cells. Wheat plants were grown in a controlled environment with daytemperatures of 18 ?C and with 500 µ Einsteins m–28–1 of photosynthetically active radiation for 16 h. Beforeanthesis and 2 to 3 weeks after, rates of net photosynthesiswere measured for leaves in 2 or 21% O2 containing 350 vpm CO2at 13, 18, 23, and 28 ?C and with 500 µEinsteins m–2s–1 of photosynthetically active radiation. Also, underthe same conditions of light intensity and temperature, therates of efflux of CO2 into CO2-free air were measured and,for mature flag leaves 3 to 4 weeks after anthesis, gross andnet photosynthesis from air containing 320 vpm 14CO2 of specificactivity 39?7 nCi µmol–1. When the O2 concentration was decreased from 21 to 2% (v/v)the rate of net photosynthesis increased by 32 per cent at thelowest temperature and 54 per cent at the highest temperature.Efflux of CO2 into CO2-free air ranged from 38 per cent of netphotosynthesis at 13 ?C to 86 per cent at 28 ?C. Gross photosynthesis,measured by the 14C assimilated during 40 s, was greater thannet photosynthesis by some 10 per cent at 13 ?C and 17 per centat 28 ?C. Bot. 36: 1835-1841. White clover was grown at 20 and100 Pa p(CO2). The CO2 response of net photosynthesis and differentialuptake of 14CO2 and 12CO2 by leaves were measured at varioustemperatures and at various O2 and CO2 partial pressures andcompared with predictions from ribulose bisphosphate carboxylase/oxygenasekinetics. Discrepancies between the observed gas exchange characteristicsfor the leaves and those predicted from the enzyme kineticswere interpreted as being due to a CO2 concentrating mechanism.Plants grown at 20 Pa p(CO2) showed a higher affinity for CO2than plants grown at 100 Pa p(CO2) when measured at 10 ?C. Nodifference in affinity was found at 30 ?C. The postulated CO2concentrating effect was greater in plants grown at low CO2than in plants grown at high CO2 concentration and occurredonly at low temperature and low CO2 partial pressure. It issuggested that plants grown at the lower CO2 partial pressurehave a higher affinity for CO2 due to a more efficient CO2 concentratingsystem than plants grown at the higher CO2 partial pressure.
