Contribution of root cap mucilage and presence of an intact root cap in maize (Zea mays) to the reduction of soil mechanical impedance 总被引:4,自引:0,他引:4
BACKGROUND AND AIMS: The impedance to root growth imposed by soil can be decreased by both mucilage secretion and the sloughing of border cells from the root cap. The aim of this study is to quantify the contribution of these two factors for maize root growth in compact soil. METHODS: These effects were evaluated by assessing growth after removing both mucilage (treatment I -- intact) and the root cap (treatment D -- decapped) from the root tip, and then by adding back 2 micro L of mucilage to both intact (treatment IM -- intact plus mucilage) and decapped (treatment DM -- decapped plus mucilage) roots. Roots were grown in either loose (0.9 Mg m(-3)) or compact (1.5 Mg m(-3)) loamy sand soils. Also examined were the effects of decapping on root penetration resistance at three soil bulk densities (1.3, 1.4 and 1.5 Mg m(-3)). KEY RESULTS: In treatment I, mucilage was visible 12 h after transplanting to the compact soil. The decapping and mucilage treatments affected neither the root elongation nor the root widening rates when the plants were grown in loose soil for 12 h. Root growth pressures of seminal axes in D, DM, I and IM treatments were 0.328, 0.288, 0.272 and 0.222 MPa, respectively, when the roots were grown in compact soil (1.5 Mg m(-3) density; 1.59 MPa penetrometer resistance). CONCLUSIONS: The contributions of mucilage and presence of the intact root cap without mucilage to the lubricating effect of root cap (percentage decrease in root penetration resistance caused by decapping) were 43 % and 58 %, respectively. The lubricating effect of the root cap was about 30 % and unaffected by the degree of soil compaction (for penetrometer resistances of 0.52, 1.20 and 1.59 MPa). 相似文献

Intensity of hydrostimulation for the induction of root hydrotropism and its sensing by the root cap 总被引:7,自引:1,他引:6
Roots of Pisum sativum L. and Zea mays L. were exposed to different moisture gradients established by placing both wet cheesecloth (hydrostimulant) and saturated aqueous solutions of various salts in a closed chamber. Atmospheric conditions with different relative humidity (RH) in a range between 98 and 86% RH were obtained at root level, 2 to 3mm from the water-saturated hydrostimulant. Roots of Silver Queen corn placed vertically with the tips down curved sideways toward the hydrostimulant in response to approximately 94% RH but did not respond positively to RH higher than approximately 95%. The positive hydrotropic response increased linearly as RH was lowered from 95 to 90%. A maximum response was observed at RH between 90 and 86%. However, RH required for the induction of hydrotropism as well as the responsiveness differed among plant species used; gravitropically sensitive roots appeared to require a somewhat greater moisture gradient for the induction of hydrotropism. Decapped roots of corn failed to curve hydrotropically, suggesting the root cap as a major site of hydrosensing. 相似文献

Primary roots of Zea mays cv. Ageotropic are nonresponsive to gravity and elongate approximately 0.80 mm h?1. Applying mucilage-like material (K-Y Jelly) to the terminal 1.5 cm of these roots induces graviresponsiveness and slow elongation 28% (i.e. from 0.80 to 0.58mm h?1). Applying mucilage-like material to one side of the terminal 1.5 cm of the root induces curvature toward the mucilage, irrespective of the root's orientation to gravity. Applying a 2-mm-wideband of mucilage-like material to a root's circumference 8 to 10 mm behind the root cap neither induces gravicurvature nor affects elongation significantly. Similarly, applying mucilage-like material to only the root cap does not significantly affect elongation or graviresponsiveness. Gravicurvature of mutant roots occurs only when mucilage-like material is applied to the root/root-cap junction. Reversing the caps of wild-type and mutant roots produces gravitropic responses characteristic of the root cap rather than the host root. These results are consistent with the suggestion that gravitropic effectors are growth inhibitors that move apoplastically through mucilage between the root cap and root. 相似文献

A sequential indole-3-acetic acid (IAA)-zeatin treatment was applied to Pisum sativum hypocotyl explants, resulting in shoot formation from 50% of the explants. Shoots were easily rooted and transplantable plants could be obtained in 3 months. The method has been applicable to the 5 cultivars tested. Histological examination of explants suggests the shoots to be of de novo origin, which would make the system suitable for transformation experiments. 相似文献

Chloroplast ultrastructure changes in Pisum sativum associated with supplementary ultraviolet (UV-B) radiation 总被引:1,自引:2,他引:1
Pea plants (Pisum sativum L. cv. Greenfeast) were grown and exposed to supplementary UV-B radiation from day 17 after planting under growth cabinet conditions. The effects of this exposure on the ultrastructure of chloroplasts and the total soluble sugar and starch concentrations were estimated. Supplementary UV-B radiation was shown to damage the structure of chloroplasts, as manifested by dilation of thylakoid membranes, a progressive disruption of the thylakoid structure and disintegration of the double membrane envelope surrounding the chloroplast, accompanied by the accumulation of large starch grains. Diurnal changes observed in starch concentration suggest that the higher concentration of starch in supplementary UV-B-treated leaves is due to its immobilization, rather than to any increase in starch synthesis: soluble sugars accumulated and remained at a higher level and then later declined. 相似文献

根际袋土培试验研究了玉米幼苗根系分泌物中的可溶性糖、低分子量有机酸和氨基酸对不同芘污染水平(50、200、800mgkg-1,记为T1、T2、T3)的响应差异,探讨芘胁迫下植物根系的生理生态效应。结果表明,较低浓度芘可适当地刺激玉米的生长,高浓度芘处理抑制了玉米的生长,并且抑制作用随芘处理浓度的提高而增强;芘对玉米根系的影响要大于对茎叶的影响。芘胁迫下促进了根系分泌可溶性糖、低分子量有机酸和氨基酸增多。T1、T2和T3处理根系分泌物中可溶性糖、低分子量有机酸、氨基酸含量分别是T0处理的1.14、1.81、1.35倍,1.24、4.31、2.94倍,1.58、5.56、5.40倍。不同芘污染水平下,乙酸分泌量表现为T2T3T1,酒石酸和柠檬酸分泌量表现为T3T2T1,草酸分泌量表现为T3≈T2T1。芘处理对根系分泌氨基酸种类的影响不大,而对各氨基酸分泌量的变化幅度影响较大;芘胁迫处理对于18种常见氨基酸组分的分泌量的影响各不相同。不同芘污染水平下,天门冬氨酸、丝氨酸和丙氨酸分泌量表现为T3T2T1,苏氨酸、谷氨酸、脯氨酸、甘氨酸、胱氨酸、缬氨酸、甲硫氨酸、异亮氨酸、亮氨酸、酪氨酸、苯丙氨酸、赖氨酸、组氨酸、γ-氨基丁酸、鸟氨酸分泌量表现为T2T3T1。 相似文献

We quantified the structural changes accompanying cellular differentiation in root caps of Zea mays cv. Ageotropic to determine the developmental basis for the nongraviresponsiveness of their primary roots. Cells of the calyptrogen and columella of primary roots of the ageotropic mutant have structures indistinguishable from those of caps of primary roots of Z. mays cv. Kys the graviresponsive, wild-type parent of Z. mays cv. Ageotropic. However, the relative volumes of dictyosomes, dictyosome-derived vesicles and starch in the outermost peripheral cells of wild-type roots were significantly lower than were those in peripheral cells of mutant roots. This corresponds to a dramatic accumulation of starch and mucilage-filled vesicles in peripheral cells of mutant roots. Cellular differentiation in root caps of graviresponsive seminal roots of the Ageotropic mutant resembled that of primary and seminal roots of the wild-type cultivar, and differed significantly from that of primary roots of the mutant. We conclude that the mutation that blocks secretion of mucilage from peripheral cells of Ageotropic roots: (1) expresses itself late in cellular differentiation in root caps; (2) is expressed only in primary (but not seminal) roots of the Ageotropic mutant; and (3) is consistent with malfunctioning dictyosomes and dictyosome-derived vesicles being the cellular basis for agravitropism of primary roots of this mutant. 相似文献

Our knowledge of the genetics of resistance to the pink stem borer ( Sesamia nonagrioides ) in maize ( Zea mays ) is restricted to a few crosses among maize inbreds. The objectives of this study were to enlarge our understanding of the genetics of traits related to damage by pink stem borer and yield under infestation and to use generation means analyses to compare per se and testcross performance for detecting epistatic effects. All generations, either per se or crossed to testers, were evaluated in a 10 × 10 triple lattice design under artificial infestation with S. nonagrioides in 2005 and 2006. Most traits fit an additive–dominance model; but evidence for epistasis for resistance and yield under infestation was shown. Epistasis, in general, did not appear to play an important role in the inheritance of yield under pink stem borer infestation. However, the epistasis contribution to maize yield performance could be important in some outstanding crosses such as EP42 × A637. Testcross generation means revealed epistatic effects undetected by the generation means analysis, but neither method was able to eliminate dominance effects that could prevail over epistatic effects. 相似文献

In previous papers we found that the frequency of B chromosomes in native races of maize varies considerably in different populations. Moreover, we found genotypes that control high and low transmission rates (TR) of B chromosomes in the Pisingallo race. In the present work crosses were made to determine whether the genes controlling B-TR are located on the normal chromosome set (As) or on the B chromosomes (Bs). We made female f.0B × male m.2B crosses between and within high (H) and low (L) B-TR groups. The Bs were transmitted on the male side in all cases. The mean B-TR from the progeny of f.0B (H) × m.2B (H) and f.0B (H) × m.2B (L) crosses was significantly higher than that from f.0B (L) × m.2B (L) and f.0B (L) × m.2B (H) crosses. The results show that the B-TR of the crosses corresponds to the H or L B-TR of the 0B female parents irrespective of the Bs of the male parent. This indicates that B-TR is genetically controlled by the 0B female parent and that these genes are located on the A chromosomes. 相似文献

BACKGROUND AND AIMS: Root growth and development are closely dependent upon nitrate supply in the growth medium. To unravel the mechanism underlying dependence of root growth on nitrate, an examination was made of whether endogenous nitric oxide (NO) is involved in nitrate-dependent growth of primary roots in maize. METHODS: Maize seedlings grown in varying concentrations of nitrate for 7 d were used to evaluate the effects on root elongation of a nitric oxide (NO) donor (sodium nitroprusside, SNP), a NO scavenger (methylene blue, MB), a nitric oxide synthase inhibitor (N(omega)-nitro-L-arginine, L-NNA), H(2)O(2), indole-3-acetic acid (IAA) and a nitric reducatse inhibitor (tungstate). The effects of these treatments on endogenous NO levels in maize root apical cells were investigated using a NO-specific fluorescent probe, 4, 5-diaminofluorescein diacetate (DAF-2DA) in association with a confocal microscopy. KEY RESULTS: Elongation of primary roots was negatively dependent on external concentrations of nitrate, and inhibition by high external nitrate was diminished when roots were treated with SNP and IAA. MB and L-NNA inhibited root elongation of plants grown in low-nitrate solution, but they had no effect on elongation of roots grown in high-nitrate solution. Tungstate inhibited root elongation grown in both low- and high-nitrate solutions. Endogenous NO levels in root apices grown in high-nitrate solution were lower than those grown in low-nitrate solution. IAA and SNP markedly enhanced endogenous NO levels in root apices grown in high nitrate, but they had no effect on endogenous NO levels in root apical cells grown in low-nitrate solution. Tungstate induced a greater increase in the endogenous NO levels in root apical cells grown in low-nitrate solution than those grown in high-nitrate solution. CONCLUSIONS: Inhibition of root elongation in maize by high external nitrate is likely to result from a reduction of nitric oxide synthase-dependent endogenous NO levels in maize root apical cells. 相似文献

The close relationship between nitrate depletion of the subsoil and root-length densities found in field experiments could not be explained by mathematical models simulating nitrate uptake (Wiesler and Horst, 1994). The objective of the present study was the validation of some of the assumptions made in these models namely uniform nitrate-uptake rates (NURs) independent on root age and daytime.Different techniques were developed and compared for the measurement of NUR of different root zones: (i) isolated root segments, (ii) compartmented uptake cuvettes, (iii) depletion of nitrate (water) from agarose blocks placed on specific zones of roots growing in nutrient solution and (iv) in rhizotrones filled with soil over the whole growing cycle of maize plants. All methods yielded a similar magnitude of NUR (10 - 30 pmol cm-2 s-1). However, only intact plants growing in nutrient solution as well as in soil, but not isolated root segments, showed higher NUR at apical root zones compared to more mature branching root zones by a factor of 2 - 8. The NUR of the root apex was particularly sensitive to the nitrogen demand of the plant and the assimilate supply from the shoots as affected by light intensity. At suboptimal, but not at optimal light conditions during preculture, NUR was lower in the dark than in the light. As plants matured, NUR of soil grown plants became increasingly dependent on water uptake. But even if nitrate uptake by mass flow was subtracted from total nitrate uptake, mature roots showed a surprisingly high nitrate-uptake capacity.The results indicate that the formation of root-age classes with different NUR and the assumption of lower NUR at night could improve the modelling of nitrate uptake. 相似文献

Sodium butyrate at 5 mM in aerated White's medium reduced the mitotic index in root meristems of seedlings of Pisum sativum to < 1% after 12 h. This effect was lessened as the butyrate concentrations were lowered. The fraction of the root meristem nuclei in G2 increased to ~ 70% after 12 h in butyrate. After 12 h exposure to butyrate, seedlings transferred lo medium without butyrate gradually re-established their normal root meristem mitotic pattern, with a burst of mitosis at 10 h after the transfer. Even a brief exposure to butyrate inhibited DNA synthesis, and nuclei released from butyrate exposure were still unable to resume normal DNA synthesis even after 12 h. This information suggests that butyrate halts progression through the cell cycle by arresting meristem nuclei in G2 and inhibiting DNA synthesis. 相似文献
