McMurdo Station, Antarctica, has discharged untreated sewage into McMurdo Sound for decades. Previous studies delineated the impacted area, which included the drinking water intake, by using total coliform and Clostridium perfringens concentrations. The estimation of risk to humans in contact with the impacted and potable waters may be greater than presumed, as these microbial indicators may not be the most appropriate for this environment. To address these concerns, concentrations of these and additional indicators (fecal coliforms, Escherichia coli, enterococci, coliphage, and enteroviruses) in the untreated wastewater, water column, and sediments of the impacted area and drinking water treatment facility and distribution system at McMurdo Station were determined. Fecal samples from Weddell seals in this area were also collected and analyzed for indicators. All drinking water samples were negative for indicators except for a single total coliform-positive sample. Total coliforms were present in water column samples at higher concentrations than other indicators. Fecal coliform and enterococcus concentrations were similar to each other and greater than those of other indicators in sediment samples closer to the discharge site. C. perfringens concentrations were higher in sediments at greater distances from the discharge site. Seal fecal samples contained concentrations of fecal coliforms, E. coli, enterococci, and C. perfringens similar to those found in untreated sewage. All samples were negative for enteroviruses. A wastewater treatment facility at McMurdo Station has started operation, and these data provide a baseline data set for monitoring the recovery of the impacted area. The contribution of seal feces to indicator concentrations in this area should be considered.  相似文献

Weddell seals (Leptonychotes weddellii) are large-bodied, long-lived mammals that live only in the circumpolar Antarctic. As such, they represent an ideal sentinel species for the impacts of environmental change on polar species. However, longitudinal physiological studies of marine mammals are notoriously difficult due to their largely aquatic lifestyle, such that many baseline indicators of health and condition are not well defined. Fifty known-age, adult seals (9-27 years, 24 males, 26 females) in McMurdo Sound were sampled for mass, total body fat, blubber depth and a suite of blood parameters (21 variables) to assess hydration state, nutritional plane, reproductive hormones (females only), organ function and immune status. Total body mass increased with age, whereas variation in blubber depth was best described by mass and/or sex, with an overall greater blubber thickness in female seals (p < 0.001). Ten blood parameters showed a significant effect of one or more model variables, most prominently between the sexes. Serum chemistry suggested that females were better nourished and displayed lower indicators of acute stress compared to males. We noted limited age-related declines that might indicate impaired organ function. Overall, seals demonstrated clinically normal values for large mammals, even at the upper end of the age continuum. Future physiological studies in this species are strongly encouraged to incorporate potential sex effects in experimental design and analysis.  相似文献

The spatial distribution, movement, and impact of the untreated wastewater outfall from McMurdo Station, Antarctica, were investigated under early austral summer conditions. The benthic environment was examined to determine the distribution of Clostridium perfringens in sediment cores and the intestinal contents of native invertebrates and fish along a transect of stations. These stations extended ca. 411 m south of the outfall. The findings revealed that the concentration of C. perfringens decreased with depth in the sediment and distance from the outfall. High percentages of tunicates and sea urchins were colonized with this bacterium along the transect. Coprostanol concentrations were also measured in sediment samples taken from each of the transect stations, and a similar trend was observed. These results are in agreement with the findings of previous studies performed with the water column and collectively provide evidence that the disposal of domestic wastes deserves special consideration in polar marine environments.  相似文献

To find the most suitable indicator of viral and parasitic contamination of drinking water, large-volume samples were collected and analyzed for the presence of pathogens (cultivable human enteric viruses, Giardia lamblia cysts, and Cryptosporidium oocysts) and potential indicators (somatic and male-specific coliphages, Clostridium perfringens). The samples were obtained from three water treatment plants by using conventional or better treatments (ozonation, biological filtration). All samples of river water contained the microorganisms sought, and only C. perfringens counts were correlated with human enteric viruses, cysts, or oocysts. For settled and filtered water samples, all indicators were statistically correlated with human enteric viruses but not with cysts or oocysts. By using multiple regression, the somatic coliphage counts were the only explanatory variable for the human enteric virus counts in settled water, while in filtered water samples it was C. perfringens counts. Finished water samples of 1,000 liters each were free of all microorganisms, except for a single sample that contained low levels of cysts and oocysts of undetermined viability. Three of nine finished water samples of 20,000 liters each revealed residual levels of somatic coliphages at 0.03, 0.10, and 0.26 per 100 liters. Measured virus removal was more than 4 to 5 log10, and cyst removal was more than 4 log10. Coliphage and C. perfringens counts suggested that the total removal and inactivation was more than 7 log10 viable microorganisms. C. perfringens counts appear to be the most suitable indicator for the inactivation and removal of viruses in drinking water treatment.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

A diurnal pattern in haul-out behaviour has been described for Weddell seals at McMurdo Sound, Antarctica, but regional and seasonal variations were previously unknown. Knowledge of activity patterns is important for standardising census methods and census data. This study quantified the diurnal pattern in haul-out behaviour of Weddell seals at the Vestfold Hills from October 1994 to March 1995. Sequential counts of seals on the ice showed that, between 0900 and 1930 hours, seal abundance differed up to 95%. Fewer seals were hauled out in the morning than in the afternoon. The maximum numbers of seals were hauled out at the warmest time of day. The diurnal cycle was less pronounced in the breeding season than in the moulting season. The findings indicated the importance of censusing Weddell seals after 1430 hours and before 1700 hours local time, especially in the moulting season. The diet of adult and juvenile Weddell seals (Leptonychotes weddellii) in McMurdo Sound, Antarctica, was determined from both scat and stable isotope analyses, to ascertain if foraging behavior varied with age, season, or diving pattern. Scats were collected over 6 years and recovered hard parts identified. Stable carbon and nitrogen isotope values were determined for seal blood samples and potential prey items and used to identify primary prey species and assess trophic interactions. Pleuragramma antarcticum remains were recovered from between 70 and 100% of the scats, and there was little evidence for inter-annual or age-specific variation in foraging behavior. However, stable isotope and dive data analyses indicated that while most seals foraged predominantly on pelagic fish and squid, some juveniles concentrated on shallow benthic Trematomus spp. Combining these three methods permitted firm conclusions about diet and foraging behavior to be drawn. Abstract Signature lipids from the phospholipid esterlinked fatty acids (PELFA) of cell membranes were used to describe benthic microbial communities of 4 Antarctic sediments. Metabolic activities of the communities were determined by incorporation of [3H]thymidine into bacterial DNA and sodium [14C]acetate into membrane lipids. Biomass measurements from extractable phospholipid fatty acids per g dry wt. ranged between 6 to 76 nmol, or when converted to number of bacteria, 3.7 × 108 to 4.5 × 109 cells per g dry wt. The West Sound site at New Harbor contained the lowest biomass, while Cape Evans on the East Sound contained the greatest. A marked difference was also noted between sites in their sediment microbial community structure. The East Sound sites at Cape Armitage and Cape Evans contained a greater abundance of diatom marker lipids, whilst both sides of the Sound contained approximately the same relative amounts of bacterial groups distinguished using PELFA. Activity of sediment microorganisms measured by radiolabel incorporation under ambient conditions followed the trends of the biomass measurements. The East Sound sites were more active by an average of 45–73% for [3H]thymidine and possibly also for sodium [14C]acetate.  相似文献

To obtain insight into the occurrence and prevalence of Clostridium perfringens and its major toxins in polar bears (Ursus maritimus), we took fecal samples for bacteriologic analysis from live-captured bears in the Svalbard Archipelago, Norway, in 2001. Clostridium perfringens was isolated from 40 of 92 samples (44%). Thirty strains were further characterized by determining toxin type and were classified to be type A, while one was also positive for the gene encoding beta2-toxin. Despite the fact that C. perfringens type A has been associated with fatal diseases in several animal species as well as in humans, our data indicate that C. perfringens type A is an normal inhabitant of the gastrointestinal tract of polar bears.  相似文献

Seals are known to be periodic breathers with eupneic and apneic phases at rest. We video-recorded a resting Weddell seal (Leptonychotes weddellii) that appeared head up from an ice hole in McMurdo Sound, Antarctica. From the video recorded, the duration of each eupneic and apneic phase was extracted, and heart rate in apneic phase was obtained by counting ripples of the sea surface around the neck of a seal. The results indicated that the distribution of instantaneous heart rate was bimodal. In addition, the higher the apneic heart rate, the shorter the apneic duration as well as the combination of apneic duration and successive eupneic duration (A–E duration). From these analyses, we characterized the periodic breathing of a resting Weddell seal.  相似文献

The McMurdo Dry Valleys of Antarctica form the coldest and driest ecosystem on Earth. Within this region there are a number of perennially ice-covered (3–6 m thick) lakes that support active microbial assemblages and have a paucity of metazoans. These lakes receive limited allochthonous input of carbon and nutrients, and primary productivity is limited to only 6 months per year owing to an absence of sunlight during the austral winters. In an effort to establish the role that bacteria and their associated viruses play in carbon and nutrient cycling in these lakes, indigenous bacteria, free bacteriophage, and lysogen abundances were determined. Total bacterial abundances (TDC) ranged from 3.80 × 104 to 2.58 × 107 cells mL–1 and virus-like particle (VLP) abundances ranged from 2.26 × 105 to 5.56 × 107 VLP mL–1. VLP abundances were significantly correlated (P < 0.05) with TDC, bacterial productivity (TdR), chlorophyll a (Chl a), and soluble reactive phosphorus (SRP). Lysogenic bacteria, determined by induction with mitomycin C, made up between 2.0% and 62.5% of the total population of bacteria when using significant decreases and increases in TDC and VLP abundances, respectively, and 89.5% when using increases in VLP abundances as the sole criterion for a successful induction event. The contribution of viruses released from induced lysogens contributed <0.015% to the total viral production rate. Carbohydrate and protein based organic aggregates were abundant within the water column of the lakes and were heavily colonized by bacteria and VLPs. Alkaline phosphatase activity was detected within the matrix of the aggregates, implying phosphorus deficiency and consortial nutrient exchanges among microorganisms.  相似文献

Background For veterinary management of non‐human primates in captivity, and conservation of wild‐living primates, management of their health risks is necessary. Incidences of pathogenic bacteria in the fecal specimens are considered as one of the useful indicators for non‐invasive health monitoring. Methods We carried out the detection of Clostridium perfringens in feces from captive and wild chimpanzees by the rapid polymerase chain reaction method. Results The bacterium was detected in most fecal specimens (80%) in captive chimpanzees. Contrarily, the detection rate in the wild chimpanzees was low, with 23% (n = 12) of 53 fecal samples from the Bossou group, Guinea, and 1.2% (1/81) in the Mahale group, Tanzania. Conclusions These results show that the intestinal microflora differs between Pan populations under various living conditions, being influenced by their diet and environment.  相似文献

Summary Zooplankton was sampled through a hole in the sea ice off Scott Base from 28 October to 17 November 1987. Vertical hauls through 100 m and 300 m were taken to compare with data from the same place two years earlier. Wide variations of abundance occurred for each taxon identified over each occasion, but there is general consistency between abundance and dominance for each species between years. For Clione limacina antarctica 1987 was demonstrably a poorer year, but better for Limacina helicina and Conchoecia belgicae. Samples taken by nets set to stream with water currents below the ice, showed increasing density of most species (and total zooplankton) with depth, with only Clione antarctica and pilidium larvae more abundant near the surface. Numbers were very low in the top 40 m. Rarer macrozooplankton were caught by using larger nets for longer times at 50 m and deeper.  相似文献

The total viable count, population density of Escherichia coli and coliform bacteria, and nitrogen in the microbiomass (microbiomass-N) in sediments were monitored monthly at 12 points in the Ongagawa River basin from June 2002 to May 2006. The measurement of the sediment microbiomass-N was used for evaluation of the sediment’s microbial population in the river ecosystem. An extraordinarily high population of E. coli was observed during the season when there was stagnant water in the basin, with a high population and an insufficient drain diffusion system, and, thus hydrological water control is indispensable to prevent rapid E. coli growth. Microbiomass-N in sediments showed a negative correlation or independent fluctuation in relation to the bacterial population in the water column of the river. Seasonal changes in extracted nitrogen (N) in river sediments did not show correspondence with microbiomass-N in sediments. The microbiomass-N in sediments changed independently of the bacterial population in the river water, indicating that the high population of bacteria in the water does not lead to a high microbial population in river sediments. Ordination of the microbial parameters by canonical correspondence analysis (CCA) showed that microbiomass-N in sediments was quite different from other parameters. Relatively higher H+ (lower pH), PO4 3− concentration and dissolved oxygen (DO) were the determinant parameters of higher microbiomass-N in sediments. A relative microbial abundance between the water column and sediments as well as each of the microbial populations in the water column and sediments could be a quantitative parameter for evaluating the biochemical processes of stream water.  相似文献

Phytoplankton populations in perennially ice-covered Lake Bonney, Antarctica grow in a unique non-turbulent environment. The absence of turbulence generated by winds or major streams, combined with strong vertical gradients in temperature and nutrients, create vertically stratified environmental conditions that support three discrete phytoplankton populations in the east lobe of this lake. Phytoplankton biomass and photosynthesis were measured in the east lobe of Lake Bonney during the winter-spring transicion (September) to mid-summer (January). During this period, irradiance beneath the ice increased from 0.03 to 1.9 mol quanta m−2 d−1. Chlorophylla concentrations ranged from 0.03 to 3.8 μl−1 within the trophogenic zone (just beneath the permanent ice cover to 20 m) and photosynthesis ranged from below detection to 3.2 μg Cl−1 d−1. Our results indicate: (1) phytoplankton photosynthesis began in late winter (before 9 September, our earliest sampling date); (2) maxima for phytoplankton biomass and production developed sequentially in time from the top to the bottom of the trophogenic zone, following the seasoral increase in irradiance; and (3) the highest photosynthetic efficiencies occurred in early spring, then decreased over the remainder of the phytoplankton growth season. The spring decrease in photosynthetic rates for shallower phytoplankton appeared to be related to nutrient availability, while photosynthesis in the deeper populations was solely lightdependent.  相似文献

Aims

To evaluate and compare the reductions of human viruses and F‐specific coliphages in a full‐scale wastewater treatment plant based on the quantitative PCR (qPCR) and plate count assays.

Methods and Results

A total of 24 water samples were collected from four locations at the plant, and the relative abundance of human viruses and F‐RNA phage genogroups were determined by qPCR. Of the 10 types of viruses tested, enteric adenoviruses were the most prevalent in both influent and effluent wastewater samples. Of the different treatment steps, the activated sludge process was most effective in reducing the microbial loads. Viruses and F‐RNA phages showed variable reduction; among them, GI and GIII F‐RNA phages showed the lowest and the highest reduction, respectively.

Conclusions

Ten types of viruses were present in wastewater that is discharged into public water bodies after treatment. The variability in reduction for the different virus types demonstrates that selection of adequate viral indicators is important for evaluating the efficacy of wastewater treatment and ensuring the water safety.

Significance and Impact of the Study

Our comprehensive analyses of the occurrence and reduction of viruses and indicators can contribute to the future establishment of appropriate viral indicators to evaluate the efficacy of wastewater treatment.  相似文献

Summary During an investigation of the meiofauna communities of the eastern Weddell Sea (Antarctica) the first naupliar stage of a new species of Pseudotachidius T. Scott, 1897 (Thalestridae) was held in culture on board R V Polarstern. These nauplii are extraordinary for a harpacticoid copepod in being globular in shape and densely filled with yolk. They lack mouth, gut tube, anus and the masticatory parts of the appendages, all of which indicate that they do not feed. It is not unlikely that later stages are also able to subsist safely on the large yolk reserves.  相似文献
