Objectives

To develop a more effective dissolved air flotation process for harvesting microalgae biomass, a co-flocculation/air flotation (CAF) system was developed that uses an ejector followed by a helix tube flocculation reactor (HTFR) as a co-flocculation device to harvest Chlorella sp. 64.01.

Results

The optimal size distribution of micro-bubbles and an air release efficiency of 96 % were obtained when the flow ratio of inlet fluid (raw water) to motive fluid (saturated water) of the ejector was 0.14. With a reaction time of 24 s in the HTFR, microalgae cells and micro-bubbles were well flocculated, and these aerated flocs caused a fast rising velocity (96 m/h) and high harvesting efficiency (94 %).

Conclusions

In a CAF process, micro-bubbles can be encapsulated into microalgae flocs, which makes aerated flocs more stable. CAF is an effective approach to harvesting microalgae.

The objective of this work was to assess and compare the removal efficiency of paracetamol and salicylic acid from aqueous medium by a microalgae-based treatment, using either Chlorella vulgaris or Tetradesmus obliquus. Moreover, considering microalgae application in wastewater treatment, the influence of these pharmaceuticals in the algal nutrient removal capacity was evaluated. The removal of paracetamol by T. obliquus (>40 %) was larger than by C. vulgaris (>21 %) in batch culture, and this was also observed for salicylic acid (>93 % by T. obliquus and >25 % by C. vulgaris). Both strains removed nutrients (phosphate and nitrate) almost completely by the end of the batch culture, but T. obliquus showed the highest efficiency at the steady state conditions of the semicontinuous culture. In spite of this, under the flocculants here tested, the efficiency in the recovery of biomass was much higher for C. vulgaris. These results highlight the importance of strain selection in the application of microalgae for wastewater treatment and, particularly, for the removal of pharmaceuticals.  相似文献

The waste management industry is an important employer, and exposure of waste-handling workers to microorganisms is considered an occupational health problem. Besides fungal contamination, it is important to consider the co-occurrence of mycotoxins in this setting. Forklifts with closed cabinet and air conditioner are commonly used in waste industry to transport waste and other products within the facilities, possibly increasing the risk of exposure under certain conditions. The aim of this study was to assess the fungal contamination and mycotoxin levels in filters from the air conditioning system of forklift cabinets, as an indicator to assess occupational exposure of the drivers working in a waste sorting facility. Cytotoxicity was also assessed to understand and characterize the toxicity of the complex mixtures as present in the forklift filters. Aqueous extracts of filters from 11 vehicles were streaked onto 2% malt extract agar (MEA) with chloramphenicol (0.05 g/L) media, and in dichloran glycerol (DG18) agar-based media for morphological identification of the mycobiota. Real-time quantitative PCR amplification of genes from Aspergillus sections Fumigati, Flavi, Circumdati, and Versicolores was also performed. Mycotoxins were analyzed using LC-MS/MS system. Cytotoxicity of filter extracts was analyzed by using a MTT cell culture test. Aspergillus species were found most frequently, namely Aspergillus sections Circumdati (MEA 48%; DG18 41%) and Nigri (MEA 32%; DG18 17.3%). By qPCR, only Aspergillus section Fumigati species were found, but positive results were obtained for all assessed filters. No mycotoxins were detected in aqueous filter extracts, but most extracts were highly cytotoxic (n = 6) or medium cytotoxic (n = 4). Although filter service life and cytotoxicity were not clearly correlated, the results suggest that observing air conditioner filter replacement frequency may be a critical aspect to avoid worker’s exposure. Further research is required to check if the environmental conditions as present in the filters could allow the production of mycotoxins and their dissemination in the cabinet during the normal use of the vehicles.  相似文献

The new paradigm is to view wastes as resources for sustainable development. In this regard, the feasibility of poultry waste and CO2 utilization for cultivation of a filamentous nitrogen-fixing cyanobacterium, Nostoc muscorum Agardh, was investigated for production polyhydroxyalkanoates, the biodegradable polymers. This cyanobacterium showed profound rise in biomass yield with up to 10 % CO2 supply in airstream with an aeration rate of 0.1 vvm. Maximum biomass yield of 1.12 g L?1 was recorded for 8 days incubation period, thus demonstrating a CO2 biofixation rate of 0.263 g L?1 day?1 at 10 % (v/v) CO2-enriched air. Poultry litter (PL) supplementation also had a positive impact on the biomass yield. The nutrient removal efficiency of N. muscorum was reflected in the significant reduction in nutrient load of PL over the experimental period. A maximum poly(3-hydroxybutyric acid-co-3-hydroxyvaleric acid) [P(3HB-co-3HV)] copolymer yield of 774 mg L?1 (65 % of dry cell wt.), the value almost 11-fold higher than the control, was recorded in 10 g L?1 PL-supplemented cultures with 10 % CO2 supply under the optimized condition, thus demonstrating that N. muscorum has good potential for CO2 biomitigation and poultry waste remediation while simultaneously producing eco-friendly polymers.  相似文献

The early life stages of Boreogadus saida and Arctogadus glacialis are morphologically similar, making it difficult to assess differences in their ecological niche. The present study documented for the first time the early life stage ecology of A. glacialis, compared it to that of B. saida, and identified the factors separating the niches of the two sympatric species. The 10,565 larval gadids collected in the Beaufort Sea from April to August of 2004 and 2008 were identified to species either directly by genetics and/or otolith nucleus size, or indirectly with a redistribution procedure. Between 8.0 and 8.7 % of all gadids were assigned to A. glacialis. Larvae of A. glacialis were longer at hatch and experienced lower mortality rates than those of B. saida. The two species shared similar spatiotemporal and vertical distributions, hatching season, and growth rate. Under the ice, feeding incidence of B. saida was low (14 %) relative to A. glacialis (88 %). At lengths <15 mm, both species specialized on different prey. The diet of fish >15 mm overlapped (Schoener’s index = 0.7), with Calanus glacialis and C. hyperboreus providing >50 % of the carbon intake of both species. The higher mortality in B. saida may be explained by the smaller size at age from hatching to metamorphosis and a lower under-ice feeding incidence. The early larval stage appears to be the key period of niche divergence between the two species.  相似文献

Dysoxylum malabaricum (white cedar) is an economically important tree species, endemic to the Western Ghats, India, which is the world’s most densely populated biodiversity hotspot. In this study, we used variation at ten nuclear simple sequence repeat loci to investigate genetic diversity and fine scale spatial genetic structure (FSGS) in seedlings and adults of D. malabaricum from four forest patches in the northern part of the Western Ghats. When genetic variation was compared between seedlings and adults across locations, significant differences were detected in allelic richness, observed heterozygosity, fixation index (F IS), and relatedness (P < 0.05). Reduced genetic diversity and increased relatedness at the seedling stage might be due to fragmentation and disturbance. There was no FSGS at the adult stage and FSGS was limited to shorter distance classes at the seedling stage. However, there was clear spatial genetic structure at the landscape level (<50 km), regardless of age class, due to limited gene flow between forest patches. A comparison of the distributions of size classes in the four locations with published data from a more southern area, showed that large trees (diameter at breast height, DBH, >130 cm) are present in the southern sacred forests but not in the northern forest reserves. This pattern is likely due to stronger harvesting pressure in the north compared to the south, because in the north there are no cultural taboos regulating the extraction of natural resources. The implications for forest conservation in this biodiversity hotspot are discussed.  相似文献

Objective

To explore the combined effects of temperature and Daphnia-associated infochemicals on colony formation of Scenedesmus obliquus to faciliate harvesting the algal biomass.

Results

A three-parameter modified Gaussian model fitted the changes of the number of cells per particle in S. obliquus induced by Daphnia culture filtrate well under any temperature. Decreases in temperature enhanced the induced–colony formation of Scenedesmus. The maximum colony size at 15–25 °C was significantly larger than those at 30–35 °C. An additional 1 or 2 days at low temperature was needed to reach the maximum colony size, which indicates the best harvest time for algal biomass.

Conclusion

Induced-colony formation of Scenedesmus by Daphnia culture filtrate at 15–25 °C is recommended to settle algal cells. This condition facilitates harvesting the biomass.

In order to establish an attractive method for the production of valuable medicinal alkaloids (galanthamine and lycorine), the plants of Leucojum aestivum and L. aestivum ‘Gravety Giant’ grown in bioreactor RITA® were subjected to various concentrations of methyl jasmonate (MeJA), salicylic acid (SA), 1-aminocyclopropane-1-carboxylic acid (ACC) and 2-chloroethylphosphonic acid (ethephon) at different times of culture. The application of MeJA showed a negative effect on L. aestivum and L. aestivum ‘Gravety Giant’ plant growth. We observed that the incubation of plants during 168 h with 100 µM of MeJA resulted above two times lower F.W. (fresh weight) increments compared with control. While SA showed an inhibitory effect only on the growth of L. aestivum cultures. ACC and ethephon had a positive effect on both types of culture. Treatment with 50 µM of MeJA during 168 h stimulated galanthamine and lycorine biosynthesis in L. aestivum and L. aestivum ‘Gravety Giant’ cultures. In addition, the accumulation of galanthamine was increased when 10 µM of ACC were added to both types of culture. 10 µM of ACC stimulated also lycorine biosynthesis by L. aestivum ‘Gravety Giant’. The addition of 10 µM of ethephon had a positive effect only on lycorine production in plants of L. aestivum. SA promoted galanthamine and lycorine biosynthesis in tested plants. Indeed the highest galanthamine (0.8 mg/g dry weight: D.W.) and lycorine (1.53 mg/g D.W.) concentrations were observed in L. aestivum ‘Gravety Giant’ plants treated with 5 µM of SA during 10 h.  相似文献

Objective

The growth characteristics and intracellular hemin concentrations of Lactococcus lactis grown under different culture pH and aeration conditions were examined to investigate the effect of culture pH on the respiration efficiency of L. lactis NZ9000 (pZN8148).

Results

Cell biomass and biomass yield of L. lactis grown with 4 μg hemin/ml and O2 were higher than those without aeration when the culture pH was controlled at 5–6.5. The culture pH affected the respiratory efficiency in the following order of pH: 5 > 5.5 > 6 > 6.5; the lag phase increased as the culture pH decreased. Hemin accumulation was sensitive to culture pH. Among the four pH conditions, pH 5.5 was optimal for hemin accumulation in the cells. The highest intracellular hemin level in L. lactis resting cells incubated at different pH saline levels (5–6.5) was at pH 5.5.

Conclusion

The respiration efficiency of L. lactis under respiration-permissive conditions increases markedly as the culture pH decreases. These results may help develop high cell-density L. lactis cultures. Thus, this microorganism may be used for industrial applications.

Numerous physiological functions of menaquinone-7 (MK-7) act to reduce vascular calcification, suggesting that MK-7 may be a potential therapy for Alzheimer’s and Parkinson’s disease, and in this study, we attempted to increase the concentration of MK-7 synthesized by Bacillus subtilis natto, a standard nattokinase (NK) producing strain. Different Bacillus subtilis isolates demonstrated positive correlations between MK-7 and NK concentrations. Response surface methodology (RSM) was employed to optimize a culture medium for the simultaneous production of these molecules; the optimized medium contained the following components (%, w/v): soybean curd residue, 12.2; soya peptone, 5.7; lactose, 2.6; and K2HPO4, 0.6. The fermentation process was subsequently optimized based on online feedback control of fermentation process parameters. The dissolved oxygen (DO) concentration played an important role in the production of MK-7 and NK. With increased DO concentrations, the cell growth rate and NK activity increased. In contrast, at low DO concentrations, the concentration of MK-7 rapidly increased during the late fermentation stage. Thus, in this study, the production of MK-7 and NK by Bacillus subtilis was accomplished using soybean curd residue through medium optimization and DO control. This novel coproduction strategy was developed by controlling the aeration rate during the fermentation process. The concentrations of MK-7 and NK achieved in this study reached 91.25 mg/L and 2675.73 U/mL, respectively.  相似文献

The present research investigates the effect of Piriformospora indica, an endophytic fungus, on production of protoberberine alkaloids in in vitro cell suspension cultures of Tinospora cordifolia. Although T. cordifolia produces a number of protoberberine alkaloids, the simultaneous production of jatrorrhizine and palmatine in cell suspension cultures of T. cordifolia was observed for the first time with the use of P. indica as biotic elicitor. The cells in suspension cultures were elicitated with P. indica on 14th day of culture initiation and the production of the alkaloids on 16th day was monitored. The autoclaved as well as filter sterilized cultures of P. indica were used in addition to the use of fungal cell extract. The elicitor effect of P. indica was analyzed and compared with other abiotic elicitor (methyl jasmonate) and biotic elicitors (chitin and chitosan). The culture filtrate of P. indica in the filter sterilized (5.0% v/v) form gave better response with enhanced 4.2-fold production of jatrorrhizine (10.72 mg/g DW) and 4.0-fold production of palmatine (4.39 mg/g DW). The production of these compounds was at par with that achieved in methyl jasmonate (at 250 µM) treated cell suspension cultures.  相似文献

Rhododendron tomentosum Harmaja (Ledum palustre), a peat bog plant from Ericaceae family, has been used in traditional medicine as the anti-arthritis agent. Although modern researches confirm its anti-inflammatory properties, it remains threatened by habitat degradation and possibilities to collect this endangered species from its natural environment for further biological activity studies are limited. Therefore, R. tomentosum liquid in vitro cultures were established as the alternative source of that valuable plant material. Schenk–Hildebrandt medium with 24.60 μM 2-isopentenyladenine and 592.02 μM adenine provides intensive growth and proper morphology of the obtained microshoots. The R. tomentosum biomass was scaled up using the various bioreactors (immersion, temporary immersion and spraying systems) for better growth and improved volatile oil production. The largest biomass accumulation (fresh weight?=?250 g l?1, growth index?=?280, dry weight?=?20 g l?1) and essential oil content (0.5% v/m) were achieved with application of commercially available RITA® bioreactor. GC/MS analysis revealed the high content of p-cymene (6.9%), alloaromadendrene (5.5%), shyobunone (8.2%) and ledene oxide (II) (13.0%) in the volatile fraction obtained from RITA® system. The biomass growth parameters and production profile in terms of essential oil and selected terpenoid compounds were determined during the 2 month period. The influence of culture conditions and bioreactor construction on the growth and volatile oil production in R. tomentosum biomasses was discussed.  相似文献

As a new α-linolenic acid (ALA) resource, there has been little known about the relationship between expression levels of fatty acid desaturase (FAD) genes during seed development and fatty acid (FA) composition in herbaceous peony (Paeonia lactiflora Pall.). In this study, oil content and FA composition of nine cultivars were measured at four different stages during seed development. Moreover, five genes including PlFAD2-1, PlFAD2-2, PlFAD3-2, PlFAD6 and PlFAD7 related to the ALA biosynthesis were isolated. Furthermore, the relative expression levels of these genes in seeds were investigated in two cultivars, that is, ‘PL1’ and ‘PL2’ with higher and lower ALA content, respectively. The results showed that oil content was from 17.03 ± 0.15 to 24.51 ± 0.15%; 15 kinds of FA were detected. However, the relative content of ALA was decreased during seed development. Although, the genes PlFAD2-1, PlFAD2-2, PlFAD3-2, PlFAD6 and PlFAD7 were all expressed during the seed development, the expression levels varied significantly. Most of the genes were expressed strongly in the early stage but weakly in the late stage except PlFAD6. Moreover, expression level of gene PlFAD2-2 was the highest while that of PlFAD7 was lowest at S1. In addition, the relative expression level of genes in ‘PL1’ (high ALA content) was higher than those in ‘PL2’ (low ALA content) during the early stage of the seed development apart from gene PlFAD6. This study provides a reliable theoretical basis for improving the contents of ALA by means of genetic engineering in the herbaceous peony seed oil.  相似文献

This study aims to develop a low-cost microalgae culture system which uses a simple closed vessel as photobioreactor to save manufacturing cost, waves for mixing to save energy cost, and high concentration of bicarbonate for carbon supply to avoid the high cost of CO2-bubbling pipeline construction on the ocean as well as to control pH by buffering the effect of bicarbonate/carbonate. To test this idea, the alkalihalophilic cyanobacterium Euhalothece sp. was cultured with 1.0 M NaHCO3 in small-scale floating photobioreactors (PBRs) on 10-cm-high artificial waves at first. The final biomass concentration was up to 0.91 and 1.47 g L?1 for indoor and outdoor cultures, respectively. However, the recorded dissolved oxygen (DO) was occasionally over-saturated (> 500% of air saturation), indicating mass transfer problem. k L a in these PBRs with different culture depth was measured then, and the results showed great variation, from 0.13 to 4.87 h?1. At the scale of 1.0 m2, this floating PBR was made with low-cost membrane and inflatable design. It was placed on the ocean surface and mixed with natural waves. Biomass concentration of 1.63 g L?1 and productivity of 8.27 g m?2 day?1 were obtained in this culture. With these results, the feasibility of a low-cost microalgae culture system was proven, which could systematically reduce the cost of photobioreactor manufacturing, operating, and maintenance.  相似文献

Selective releasing intracellular product in Triton X-100 micelle aqueous solution to prepare whole cell biocatalyst is a novel strategy for biosynthesis of Monascus pigments, in which cell suspension culture exhibits some advantages comparing with the corresponding growing cell submerged culture. In the present work, the nonionic surfactant Triton X-100 was successfully replaced by edible plant oils for releasing intracellular Monascus pigments. High concentration of Monascus pigments (with absorbance nearly 710 AU at 470 nm in the oil phase, normalized to the aqueous phase volume approximately 142 AU) was achieved by cell suspension culture in peanut oil–water two-phase system. Furthermore, the utilization of edible oil as extractant also fulfills the demand for application of Monascus pigments as natural food colorant.  相似文献

Petiveria alliacea L. is a medicinal plant originating from the Amazon region. This study describes an efficient cryopreservation protocol for somatic embryos (SEs) produced from roots of P. alliacea based on the comparison of vitrification, encapsulation-dehydration, and D cryo-plate techniques. With the vitrification technique, SEs treated with PVS2 solution (0.4 M sucrose, 3.3 M glycerol, 2.4 M ethylene glycol, and 1.9 M DMSO) for 30 min displayed high viability (85%) and intermediate proliferation recovery (about 12 adventitious SEs produced from original SEs [SEs/SE] after 90 d of culture). With the encapsulation-dehydration technique, lower viability (70%) and very low proliferation recovery (about two SEs/SE) were achieved with cryopreserved SEs dehydrated for 10 min in a laminar air flow cabinet. The D cryo-plate technique led to high viability (85%) and proliferation recovery (19 SEs/SE) of cryopreserved SEs after 90 min dehydration. In the experimental conditions tested, the D cryo-plate method was the most efficient technique for cryopreservation of P. alliacea SEs.  相似文献

Microalgae are the most promising sources of protein, which have high potential due to their high-value protein content. Conventional methods of protein harnessing required multiple steps, and they are generally complex, time consuming, and expensive. Currently, the study of integration methods for microalgae cell disruption and protein recovery process as a single-step process is attracting considerable interest. This study aims to investigate the novel approach of integration method of electrolysis and liquid biphasic flotation for protein extraction from wet biomass of Chlorella sorokiniana CY-1 and obtaining the optimal operating conditions for the protein extraction. The optimized conditions were found at 60% (v/v) of 1-propanol as top phase, 250 g/L of dipotassium hydrogen phosphate as bottom phase, crude microalgae loading of 0.1 g, air flowrate of 150 cc/min, flotation time of 10 min, voltage of 20 V and electrode’s tip touching the top phase of LBEF. The protein recovery and separation efficiency after optimization were 23.4106?±?1.2514% and 173.0870?±?4.4752%, respectively. Comparison for LBEF with and without the aid of electric supply was also conducted, and it was found that with the aid of electrolysis, the protein recovery and separation efficiency increased compared to the LBEF without electrolysis. This novel approach minimizes the steps for overall protein recovery from microalgae, time consumption, and cost of operation, which is beneficial in bioprocessing industry.  相似文献

The NLN-medium has been successfully used, since 1982, for microspore culture in Brassica napus and other Brassica species. Changes to the media composition were restricted to carbohydrate and nitrogen sources and growth regulators while micro-nutrients have not been optimized. The NLN-medium contains boron at a concentration of 162 µM. Boron is required for diverse physiological and metabolic processes in the cell. This study investigated the effect of seven- and 13-fold increased boron concentration on the induction of embryos in microspore cultures of four genotypes of B. napus. A significant improvement of microspore embryogenesis was achieved by both enhanced boron concentrations in the NLN medium. No effect on the regeneration of embryo to plant conversion was observed.  相似文献

Lessonia trabeculata is one of the most valuable seaweeds in Chile, especially in the northern zone where its harvest has been going on for decades. We carried out population dynamics studies in the Atacama Desert coast (Bahia Chasco), in order to assess its productivity under natural and harvesting scenarios. We found very slow but consistent growth (1.98 cm month?1) and density (3–4 individuals m?2 with no monthly variation) during 18 months of observations in an undisturbed subpopulation. However, after total harvesting, L. trabeculata exhibited different responses. Its recruitment was season-specific, with exceedingly high values in autumn (ca. 80 individuals m?2 in 5 months) and a dramatic reduction of recruits in summer (1–5 individuals m?2 in 7 months, with many areas with no recruitment). Gradually, density values tended to stabilize to growth rates under un-altered conditions. In parallel, pruning systems at three different thallus levels (frond meristem base cuts, removal of half and total canopy) were all inefficient and harmful: (i) Biomass takes longer to be harvested; (ii) pruned individuals die off; and (iii) do not detach easily from the substrata, delaying the recovery by potentially emerging L. trabeculata juveniles. Some of these results agreed with our culture experiments, where 26 months were needed to obtain up to 100 cm long thalli with shrub-like morphology. We conclude that management of L. trabeculata beds must be improved in order to guarantee survival of the industry, and we propose some practices that at some stage should involve the complete removal of older/senescent individuals.  相似文献
