Bud rot disease affecting oil palm in South American countries is reported to be caused by Phytophthora palmivora. P. palmivora is a local pathogen affecting various crops in Malaysia, and this finding caused an alarm, which prompted an investigation of pathogenicity using Malaysian P. palmivora to assess the potentials of this Oomycete to infect oil palm in Malaysia. A total of 11 P. palmivora isolates were obtained from cocoa and durian for the study. Leaf bioassays via artificial inoculation using 50,000 zoospores/ml and mycelial agar disc showed severe necrotic lesions on the infection spot of oil palm (DxP) spear leaves. Mild infection was observed in oil palm spear leaves of OxG hybrids indicating lower susceptibility against P. palmivora infection. Phylogenetic analysis using internal transcribed spacer (ITS) data revealed that Malaysian isolates were genetically similar to Colombian isolates supported by significant bootstrap values. The leaf bioassay results revealed that Malaysian oil palm materials are susceptible towards local P. palmivora infection. The Colombian P. palmivora isolates causing bud rot incidence may have evolved over a long period of time, undergone sequential genetic shift to become more virulent towards Colombian oil palm planting materials.  相似文献

Differences in the reproductive mode (sexual and vegetative reproduction) between populations of a species may produce diverse patterns of genetic structure within and among populations. We analyzed the clonal diversity of populations of a heterostylous plant (Nymphoides indica (L.) Kuntze) having different floral morph ratios in the Higashi-harima area of Japan. The number of MLGs (multilocus genotypes) in populations with equal floral morph ratios was significantly higher than that among populations with biased floral morph ratios and among monomorphic populations. Populations with equal floral morph ratios followed the Hardy–Weinberg equilibrium. A dispersal distance of 0–4.2 km was significantly correlated with Nei's genetic distance, supporting a stepping-stone model of dispersal. Four rare MLGs were included in populations with equal floral morph ratios, distributed among neighboring populations. We concluded that clonal diversity of N. indica in the Higashi-harima area is maintained by the success of seed production and establishment of seedlings, and by the proximate occurrence of habitats around the populations where successful sexual reproduction occurs.  相似文献

Reliance on clonal reproduction is associated with an increased risk of extinction. Grevillea infecunda is a rare, putatively sterile shrub restricted to 11 populations in a localized coastal region of south-eastern Australia. We assessed the genetic diversity, clonal diversity and spatial distribution of clones in all populations of G. infecunda to guide conservation management. Eight chloroplast haplotypes were identified from the trnL – trnF and trnQ – rps16 intergenic spacer regions. All individuals belonged to a single maternal lineage dominated by one haplotype (95/111 samples). Minor haplotypes differed from the common haplotype only by single mutational steps. However, microsatellite markers revealed 89 multilocus genotypes (MLGs) in 38 multilocus lineages (MLLs) of variable size. MLLs were not shared among populations and ramets from different MLLs rarely intermingled physically. New shoots arising after fire were confirmed to belong to previously-existing MLLs, indicating that this species exhibits adaptation to fire. Genetically similar MLGs were more likely to be found in close proximity than less similar MLGs, resulting in significant spatial autocorrelation to ca 350 m. Genetic diversity was moderate but genotypic diversity was low once likely clonality was taken into account. Clonality appears to have arisen several times within the holly-leafed grevilleas and examining G. infecunda is a step towards understanding why and how often clonality occurs, and the long-term evolutionary outcomes of this life history.  相似文献

Juniperus brevifolia is an important woody species endemic of the Azores archipelago (Portugal), found from coastal to mountain environments. Due to colonization and grazing pressure this species has suffered fragmentation, leading to extinction in one island and being threatened in others. The genetic diversity and population structure of J. brevifolia populations was studied to provide guidelines for restoration and conservation programmes. Nuclear Single Sequence Repeats (nSSR) from Juniperus communis and Juniperus przewalskii and Inter Simple Sequence Repeats (ISSR) were tested and results compared to test the transferability of the microsatellites to J. brevifolia. Samples from ten populations over three islands, divided as coastal, mid-altitude and mountain were analysed. Both marker systems revealed results statistically and strongly correlated with each other, and not dependent on population sample size. We observed positive fixation indexes, moderate to high levels of genetic diversity (h = 0.415 for nSSR and h = 0.245 for ISSR), low to moderate φpt genetic differentiation (0.070 for nSSR and 0.129 for ISSR) and high gene flow (Nm > 2.432). Regarding Nei's genetic distance the coastal communities clustered together reflecting the phenotypic plasticity, but no specific clustering was observed regarding φpt values. Therefore no populations with substantial genetic differentiation were identified, once the diversity is mostly observed within populations. However it is advised the continuous monitoring of J. brevifolia.  相似文献

Picconia azorica (Tutin) Knobl. (Oleaceae) is an endangered species, endemic to the Azores. Samples from 31 populations in 8 islands were genotyped using 8 newly developed nuclear microsatellite markers. From the amplified loci, 81% were polymorphic across all populations and the species showed a relatively high total genetic diversity (HT = 0.7). Several populations were close to Hardy–Weinberg equilibrium while others presented positive FIS values (0.02–0.2). The largest proportion of genetic variation (98%) occurred within populations and the level of differentiation between populations, was generally low, although 27% of the population pairwise comparisons showed relatively high differentiation values (0.25  RST  0.65). Relatively high levels of gene flow were also found among most populations. Using the Bayesian clustering method implemented in STRUCTURE we found a particular genetic pattern in Corvo samples, and also similarities between Santa Maria, São Miguel and Flores populations. Considerable levels of genetic admixture within P. azorica populations might have resulted from: (i) fruit dispersal by native birds; and/or (ii) human mediated dispersal between islands. Our results revealed the existence of some genetically depauperate populations needing specific conservation measures, and indicate that arbitrary translocation of individuals between islands should be avoided.  相似文献

Background and Aims

The association of clonality, polyploidy and reduced fecundity has been identified as an extinction risk for clonal plants. Compromised sexual reproduction limits both their ability to adapt to new conditions and their capacity to disperse to more favourable environments. Grevillea renwickiana is a prostrate, putatively sterile shrub reliant on asexual reproduction. Dispersal is most likely limited by the rate of clonal expansion via rhizomes. The nine localized populations constituting this species provide an opportunity to examine the extent of clonality and spatial genotypic diversity to evaluate its evolutionary prospects.

Methods

Ten microsatellite loci were used to compare genetic and genotypic diversity across all sites with more intensive sampling at four locations (n = 185). The spatial distribution of genotypes and chloroplast DNA haplotypes based on the trnQ–rps16 intergenic spacer region were compared. Chromosome counts provided a basis for examining genetic profiles inconsistent with diploidy.

Key Results

Microsatellite analysis identified 46 multilocus genotypes (MLGs) in eight multilocus clonal lineages (MLLs). MLLs are not shared among sites, with two exceptions. Spatial autocorrelation was significant to 1·6 km. Genotypic richness ranged from 0 to 0·33. Somatic mutation is likely to contribute to minor variation between MLGs within clonal lineages. The eight chloroplast haplotypes identified were correlated with eight MLLs defined by ordination and generally restricted to single populations. Triploidy is the most likely reason for tri-allelic patterns.

Conclusions

Grevillea renwickiana comprises few genetic individuals. Sterility has most likely been induced by triploidy. Extensive lateral suckering in long-lived sterile clones facilitates the accumulation of somatic mutations, which contribute to the measured genetic diversity. Genetic conservation value may not be a function of population size. Despite facing evolutionary stagnation, sterile clonal species can play a vital role in mitigating ecological instability as floras respond to rapid environmental change.  相似文献

Eucalyptus urophylla (Timor mountain gum) is an economically important plantation species that occurs naturally in a series of disjunct populations on the volcanic slopes of seven islands in eastern Indonesia. Twelve microsatellite markers were used to investigate the distribution of nuclear genetic diversity among 19 geographically defined E. urophylla populations. High levels of gene diversity were observed throughout the geographic range (H E = 0.703 to 0.776). The level of genetic differentiation among populations was low (F ST = 0.031), but the amount of differentiation increased with geographic distance. A phenogram produced by a neighbor-joining analysis illustrated that populations clustered according to islands. However, a Bayesian clustering approach revealed a more cryptic population structure comprising two genetically homogeneous groups. Gene flow among the populations is likely responsible for the apparent weak influence of geographic insularity on the genetic diversity and structure of the island species. These findings provide direction for conservation and breeding strategies in E. urophylla. The cocoa tree, Theobroma cacao L., suffers large yield losses in Aceh Indonesia due to the disease black pod rot, caused by Phytophthora spp. Despite having the largest area under cacao production in Sumatra, farmers in the Aceh region have low overall production because of losses to insect pests and black pod rot. Trichoderma spp. were isolated from the roots and leaves of cacao trees and screened as potential biological control agents. Isolates used in the study were Trichoderma asperellum isolates T2 and T4, Trichoderma longibrachiatum isolates T15 and T16, and Trichoderma virens isolates T1 and Tv. T1, T2, T4, and Tv completely colonized and destroyed Phytophthora tropicalis and Phytophthora palmivora mycelium in precolonized plate assays. All six isolates reduced P. tropicalis, but none reduced the growth of P. palmivora in dual plate assays. Phytophthora growth was suppressed on MIN media amended with sterile heat inactivated Trichoderma culture filtrates, with Tv best suppressing growth of both Phytophthora spp. T. virens isolate Tv was the only isolate observed coiling around P. tropicalis mycelium and disrupted the formation of P. palmivora sporangia. Of all six isolates, only Tv reduced P. palmivora lesion expansion in a detached pod assay, reducing severity by 71%. Tv also reduced P. palmivora infection on seedlings when applied aerially at 1 × 106 and 1 × 108 conidia/ml, by 19% and 59%, respectively. T. virens isolate Tv is a mycoparasite, antagonizes Phytophthora in a dual plate assay, and shows antibiosis against Phytophthora spp., suggesting that multiple modes of action contribute to its ability to limit Phytophthora lesion expansion on cacao pods and seedlings.  相似文献

The existence of multiple independently derived populations in landlocked marine lakes provides an opportunity for fundamental research into the role of isolation in population divergence and speciation in marine taxa. Marine lakes are landlocked water bodies that maintain a marine character through narrow submarine connections to the sea and could be regarded as the marine equivalents of terrestrial islands. The sponge Suberites diversicolor (Porifera: Demospongiae: Suberitidae) is typical of marine lake habitats in the Indo-Australian Archipelago. Four molecular markers (two mitochondrial and two nuclear) were employed to study genetic structure of populations within and between marine lakes in Indonesia and three coastal locations in Indonesia, Singapore and Australia. Within populations of S. diversicolor two strongly divergent lineages (A & B) (COI: p = 0.4% and ITS: p = 7.3%) were found, that may constitute cryptic species. Lineage A only occurred in Kakaban lake (East Kalimantan), while lineage B was present in all sampled populations. Within lineage B, we found low levels of genetic diversity in lakes, though there was spatial genetic population structuring. The Australian population is genetically differentiated from the Indonesian populations. Within Indonesia we did not record an East-West barrier, which has frequently been reported for other marine invertebrates. Kakaban lake is the largest and most isolated marine lake in Indonesia and contains the highest genetic diversity with genetic variants not observed elsewhere. Kakaban lake may be an area where multiple putative refugia populations have come into secondary contact, resulting in high levels of genetic diversity and a high number of endemic species.  相似文献

Prince Rupprecht's Larch (Larix principis-rupprechtii Mayr.) is one of dominant components of middle and high elevation forests in North China. Shanxi Province is well known as “the Hometown of Prince Rupprecht's Larch” in China. In this study, six natural populations of this species across Shanxi were selected to investigate the genetic variation of the species using amplified fragment length polymorphism (AFLP) markers. Results showed that in comparison with some other species of Larix, higher genetic diversity was revealed at the species level for L. principis-rupprechtii (percentage of polymorphic loci PPL = 71.9%, Nei's gene diversity HE = 0.225, Shannon information index I = 0.341). Most of genetic variation existed within populations (80.5%), while the genetic differentiation among populations was significant (p < 0.001) and higher (Gst = 0.194) than most other species of Larix. The differentiation can be attributed to the limited gene flow (Nm = 1.035) among populations, which could be due to the spatial isolation and habitat fragmentation. The six populations can be divided into three groups based on the Nei's genetic distances between populations (from 0.033 to 0.076). There was no significant correlation (r = 0.268, p > 0.05) between genetic distance and geographic distance among populations. The measures for in-situ or ex-situ conservation should be taken to preserve the genetic diversity of this species.  相似文献

The levels of genetic diversity and gene flow may influence the long-term persistence of populations. Using microsatellite markers, we investigated genetic diversity and genetic differentiation in island (Krakatau archipelago, Indonesia) and mainland (Java and Sumatra, Indonesia) populations of Liporrhopalum tentacularis and Ceratosolen bisulcatus, the fig wasp pollinators of two dioecious Ficus (fig tree) species. Genetic diversity in Krakatau archipelago populations was similar to that found on the mainland. Population differentiation between mainland coastal sites and the Krakatau islands was weak in both wasp species, indicating that the intervening 40 km across open sea may not be a barrier for wasp gene flow (dispersal) and colonization of the islands. Surprisingly, mainland populations of the fig waSPS may be more genetically isolated than the islands, as gene flow between populations on the Javan mainland differed between the two wasp species. Contrasting growth forms and relative 'immunity' to the effects of deforestation in their host fig trees may account for these differences.  相似文献

Invasive populations typically demonstrate genetic isolation which results in a loss of genetic diversity and a reduction in invasion success. This study focused on the genetic population of a successful invasive species of tarantula. Individuals were sampled in two mainland localities of the Yucatan Peninsula (Zoh-Laguna and Raudales), in addition to two island localities (El Cedral and Rancho Guadalupe on Cozumel Island). All populations present high genetic diversity (mean: He = 0.23, P = 99%), with significant differences between the Raudales and Rancho Guadalupe localities. The AMOVA analysis revealed a significant population structure (14.5% variation among populations), consistent with the gene differentiation coefficient (GST = 0.21), and spatial analysis of population structure. Our results suggested that the original introduced population did not suffer a loss of genetic diversity during establishment on the island, possibly a result of different biological conditions. Population structure analysis leads us to suggest that one island population is similar to the original genetic profile, whereas the genotypic profile of the other island population reflects recent introductions from the mainland. We identified a potential risk of extinction for one local mainland population, suggesting that this species may be a successful invader in a new environment but endangered in some parts of its natural area.  相似文献

Phytophthora capsici causes significant loss to pepper (Capsicum annum) in China and our goal was to develop single nucleotide polymorphism (SNP) markers for P. capsici and characterize genetic diversity nationwide. Eighteen isolates of P. capsici from locations worldwide were re-sequenced and candidate nuclear and mitochondrial SNPs identified. From 2006 to 2012, 276 isolates of P. capsici were recovered from 136 locations in 27 provinces and genotyped using 45 nuclear and 2 mitochondrial SNPs. There were two main mitochondrial haplotypes and 95 multi-locus genotypes (MLGs) identified. Genetic diversity was geographically structured with a high level of genotypic diversity in the north and on Hainan Island in the south, suggesting outcrossing contributes to diversity in these areas. The remaining areas of China are dominated by four clonal lineages that share mitochondrial haplotypes, are almost exclusively the A1 or A2 mating type and appear to exhibit extensive diversity based on loss of heterozygosity (LOH). Analysis of SNPs directly from infected peppers confirmed LOH in field populations. One clonal lineage is dominant throughout much of the country. The overall implications for long-lived genetically diverse clonal lineages amidst a widely dispersed sexual population are discussed.  相似文献

Abstract . This paper assesses variation in coral species diversity within the Indonesian archipelago, and the influence of regional species pools, geomorphology and anthropogenic pollution on coral species diversity and occurrence. We obtained transects from 33 sites on 14 reefs in three regions of Indonesia: Ambon (Moluccas), South Sulawesi and the Java Sea. We determined the within‐site species richness by using species‐sampling curves. Cluster analysis and multi‐dimensional scaling showed that land‐based pollution was the primary determinant of coral species diversity and species occurrence on reefs. Relatively unaffected reference sites in eastern Indonesia were approximately 20% more diverse than Java Sea reference sites. Rare species formed a higher proportion of the coral fauna on eastern Indonesian sites, and eastern Indonesian apparent endemic species contributed approximately 25% of the total species pool sampled. Between‐site variation in species occurrence was lower on Java Sea reefs than on eastern Indonesian reefs. A larger species pool in eastern Indonesia than in the Java Sea probably accounted for most of the difference in within‐site species diversity between eastern Indonesian and Java Sea reference sites. High fishing intensity in the Java Sea, including destructive fishing practices, may have also contributed to reduced within‐site species diversity on Java Sea reference reefs. Despite the fact that the Java Sea was exposed during Pleistocene lowstands, and was recolonized by marine organisms only within the last 10 000 years, coral species diversity and assemblage composition on the Java Sea reefs was largely similar to open ocean reefs in eastern Indonesia.  相似文献

Eighteen populations of the endangered aromatic and medicinal plant Mentha cervina (Lamiaceae) were sampled across its natural range, in the western half of the Iberian Peninsula, and inter-simple sequence repeats (ISSRs) markers were used to assess genetic diversity and population structure. M. cervina populations exhibited a relatively low genetic diversity (percentage of polymorphic loci PPB = 14.2–58.3%, Nei's genetic diversity He = 0.135–0.205, Shannon's information index I = 0.08 − 0.33). However, the genetic diversity at species level was relatively high (PPB = 98.3%; He = 0.325; I = 0.23). The results of the analysis of molecular variance indicated very structured populations, with 50% of the variance within populations, 44% among populations and 6% between regions defined by hydrographic basins, in line with the gene differentiation coefficient (GST = 0.532). A Mantel test did not find significant correlation between genetic and geographic distance matrices (r = 0.064), indicating that isolation by distance is not shaping the present genetic structure. The levels and patterns of genetic diversity in M. cervina populations were assumed to result largely from a combination of evolutionary history and its unique biological traits, such as breeding system, low capacity of dispersion, small effective size and habitat fragmentation. The high genetic differentiation among populations indicates the necessity of conserving the maximum possible number of populations. The results also provide information to select sites for ex situ conservation. Optimal harvesting strategies, cultivation and tissue culture should also be developed as soon as possible to guarantee sustainable use of the species under study.  相似文献

Canker disease of Theobroma cacao L. caused by Phytophthora palmivora (Butler) Butler gives rise to serious damage in Fiji and is common in the S.W. Pacific area. Fusarium decemcellulareBiick (imperfect stage of Calonectria rigidiuscula (Berk. & Br.) Sacc.) was constantly associated with P. palmivora in disorders of cocoa but was not found to be a primary pathogen. Most Amelonado cocoa trees were resistant to canker but there were differences in the reactions of other Forastero and of Trinitario types. The F1 generation of a local Trinitario selection, itself resistant, showed segregation resulting in a proportion of very susceptible plants. Isolates of P. palmivora from black pod, canker and green twigs were equally pathogenic, both to pods and seedling stems. Sporangial dimensions, particularly the length/breadth ratio, differed from those reported elsewhere. It is uncertain whether the reason that cocoa canker is more serious in some countries than others is because of differences in the pathogenicity of P. palmivora strains to bark or the genetic differences of the cocoa grown.  相似文献

Understanding the factors that contribute to population genetic divergence across a species' range is a long‐standing goal in evolutionary biology and ecological genetics. We examined the relative importance of historical and ecological features in shaping the present‐day spatial patterns of genetic structure in two related plant species, Beta vulgaris subsp. maritima and Beta macrocarpa. Using nuclear and mitochondrial markers, we surveyed 93 populations from Brittany (France) to Morocco – the southern limit of their species' range distribution. Whereas B. macrocarpa showed a genotypic structure and a high level of genetic differentiation indicative of selfing, the population genetic structure of B. vulgaris subsp. maritima was consistent with an outcrossing mating system. We further showed (1) a strong geographic clustering in coastal B. vulgaris subsp. maritima populations that highlighted the influence of marine currents in shaping different lineages and (2) a peculiar genetic structure of inland B. vulgaris subsp. maritima populations that could indicate the admixture of distinct evolutionary lineages and recent expansions associated with anthropogenic disturbances. Spatial patterns of nuclear diversity and differentiation also supported a stepwise recolonization of Europe from Atlantic‐Mediterranean refugia after the last glacial period, with leading‐edge expansions. However, cytoplasmic diversity was not impacted by postglacial recolonization: stochastic long‐distance seed dispersal mediated by major oceanic currents may mitigate the common patterns of reduced cytoplasmic diversity observed for edge populations. Overall, the patterns we documented here challenge the general view of reduced genetic diversity at the edge of a species' range distribution and provide clues for understanding how life‐history and major geographic features interact to shape the distribution of genetic diversity.  相似文献

Tapiscia sinensis is a Tertiary relict and endangered tree species with unique scientific research value and great economic value. In this study, we assessed the genetic diversity of five wild T. sinensis populations from different geographical regions using 10 polymorphic simple sequence repeat (SSR) markers. Our results reveal that the natural populations of T. sinensis have rich genetic diversity (PPL = 100%, He = 0.6904, I = 1.4368), with Shannon's index indicating that the T. sinensis populations are at a relatively stable stage. Of the genetic relationships among populations, the distance between the Hunan Yanling (YL) and Guizhou Xifeng (XF) populations is the smallest (0.4829); the genetic distance between the Shaanxi Ningshan (NS) and the Guizhou Xifeng (XF) populations is the largest (0.9821). A Mantel test shows that there is no correlation among the populations between geographic distance and genetic distance. AMOVA suggest that 33.3% of the genetic variation arose among the populations, while 66.7% of the variation arose within them. The moderate gene flow among populations (Nm = 0.7274) is not sufficient to counteract genetic drift within the populations and result in significant differentiation (Fst = 0.2987). Our results will benefit the conservation and exploitation of T. sinensis and provide a theoretical basis for further study of the evolution and phylogeography of the species.  相似文献

Celebochoerus is a unique suid having extremely large upper tusks, and which was to date only known from the Pliocene-Pleistocene of Sulawesi Island in Indonesia. Here, we report on the discovery of a canine fragment referable to Celebochoerus from the Cagayan Valley of Luzon, Northern Philippines. We name a new species, Celebochoerus cagayanensis nov. sp., which differs from the Sulawesi species Celebochoerus heekereni in having mesial and distal enamel bands on the upper canines. We see these characteristics as symplesiomorphic in suids and propose a migration route from the Philippines to Sulawesi, possibly out of Taiwan, which would have occurred independently from the better known Pleistocene migration route from India into Java.  相似文献

To obtain accurate estimates of population structure for purposes of conservation planning for wild lotus (Nelumbo nucifera Gaertn.) in central China, genetic diversity among and within six populations, and clonal diversity within another two populations of the species were analyzed. The genetic diversity was high (percentage of polymorphic bands, PPB = 90.0%; Shannon's information index, I = 0.383 ± 0.234) at the species level, but low within individual study populations (PPB = 35.8%; Shannon's information index I = 0.165 ± 0.241). The mean coefficient of gene differentiation (Gst) was 0.570, indicating that 43.0% of the genetic diversity resided within the population. Analysis of molecular variance (AMOVA) indicated that 50.47% of the genetic diversity among the study populations was attributed to geographical location while 12.3% was attributed to differences in their habitats. An overall value of mean estimated number of gene flow (Nm = 0.377) indicated that there was limited gene flow among the sampled populations. The level of clonal diversity found within the populations was considerably high (Simpson's diversity index, D = 0.985) indicating that clonal diversity contributes to a major extent to the overall genetic variation in the genetic structure of N. nucifera. On the basis of the high Gst and D values detected in this study we recommend that any future conservation plans for this species should be specifically designed to include those representative populations with the highest genetic variation for both in situ conservation and germplasm collection expeditions.  相似文献
