CO2 at different concentrations were added to cultures of the eukaryotic microalgae, Chlorella kessleri, C. vulgaris and Scenedesmus obliquus, and the prokaryotic cyanobacterium, Spirulina sp., growing in flasks and in a photobioreactor. In each case, the best kinetics and carbon fixation rate were with a vertical tubular photobioreactor. Overall, Spirulina sp. had the highest rates. Spirulina sp., Sc. obliquus and C. vulgaris could grow with up to 18% CO2.  相似文献

The CO2-concentrating mechanism (CCM) was induced in the green unicellular alga Chlorella when cells were transferred from high (5% CO2) to low (0.03%) CO2 concentrations. The induction of the CCM correlated with the formation of a starch sheath specifically around the pyrenoid in the chloroplast. With the aim of clarifying whether the starch sheath was involved in the operation of the CCM, we isolated and physiologically characterized a starchless mutant of Chlorella pyrenoidosa, designated as IAA-36. The mutant strain grew as vigorously as the wild type under high and low CO2 concentrations, continuous light and a 12 h light/12 h dark photoperiod. The CO2 requirement for half-maximal rates of photosynthesis [K0.5(CO2)] decreased from 40 μM to 2–3 μM of CO2 when both wild type and mutant were switched from high to low CO2. The high affinity for inorganic carbon indicates that the IAA-36 mutant is able to induce a fully active CCM. Since the mutant does not have the pyrenoid starch sheath, we conclude that the sheath is not involved in the operation of the CCM in Chlorella cells.  相似文献

Non-phototrophic CO 2 fixation by soil microorganisms   总被引:1,自引:0,他引:1  
Although soils are generally known to be a net source of CO2 due to microbial respiration, CO2 fixation may also be an important process. The non-phototrophic fixation of CO2 was investigated in a tracer experiment with 14CO2 in order to obtain information about the extent and the mechanisms of this process. Soils were incubated for up to 91 days in the dark. In three independent incubation experiments, a significant transfer of radioactivity from 14CO2 to soil organic matter was observed. The process was related to microbial activity and could be enhanced by the addition of readily available substrates such as acetate. CO2 fixation exhibited biphasic kinetics and was linearly related to respiration during the first phase of incubation (about 20–40 days). The fixation amounted to 3–5% of the net respiration. After this phase, the CO2 fixation decreased to 1–2% of the respiration. The amount of carbon fixed by an agricultural soil corresponded to 0.05% of the organic carbon present in the soil at the beginning of the experiment, and virtually all of the fixed CO2 was converted to organic compounds. Many autotrophic and heterotrophic biochemical processes result in the fixation of CO2. However, the enhancement of the fixation by addition of readily available substrates and the linear correlation with respiration suggested that the process is mainly driven by aerobic heterotrophic microorganisms. We conclude that heterotrophic CO2 fixation represents a significant factor of microbial activity in soils.  相似文献

Seven strains of extremely halophilic bacteria (Halobacterium spp., Halococcus spp., and Haloarcula sp.) fixed CO2 under light and dark conditions. Light enhanced CO2 fixation in Halobacterium halobium but inhibited it in Halobacterium volcanii and Haloarcula strain GN-1. Propionate stimulated 14CO2 incorporation in some strains, but inhibited it in others. Semi-starvation in basal salts plus glycerol induced enhanced CO2 fixation rates. 14CO2 fixation in semi-starved cells was stimulated by NH 4 + or pyruvate and inhibited by succinate and acetate in most strains. No possible reductant was found. In cell-free extracts of H. halobium, NH 4 + but not propionate stimulated 14CO2 fixation. No RuBP carboxylase activity was detected. The main 14C-labeled -keto acid detected after a 2-min incubation with 14CO2 and pyruvate was pyruvate. Little or no -ketobutyrate was detected among the early products of propionate-stimulated CO2 fixation. Glycine was the major amino acid synthesized during a 2-min incubation with NH 4 + , propionate, and 14CO2. Propionate-stimulated CO2 fixation was sensitive to trimethoprim and insensitive to avidin. Earlier labeling experiments have shown that autotrophically grown Acetobacterium woodii assimilates cell carbon via direct acetyl CoA formation from 2 CO2, rather than via the Calvin cycle. Cell extracts contained the enzymes required for biosynthesis starting from acetyl CoA and CO2. Notably, pyruvate synthase, pyruvate phosphate dikinase, and phosphoenolpyruvate carboxytransphosphorylase were present in sufficiently high activities. Ribulose-1,5-bisphosphate carboxylase activity could not be detected. The observed enzyme pattern was consistent with the postulated biosynthetic pathway as deduced from 14C-labeling experiments.  相似文献

Chlorofluexus aurantiacus OK-70 fl was grown photoautotrophically with hydrogen as the electron source. The lowest doubling time observed was 26 h.The mechanism of CO2 fixation in autotrophically grown cells was studied. The presence of ribulose-1,5-bis-phosphate carboxylase and phosphoribulokinase could not be demonstrated. Carbon isotope fractionation (13C) was small, and alanine and aspartate but not 3-phosphoglycerate were the major labelled compounds in short term 14CO2 labelling. Thus CO2 is not fixed by the Calvin cycle.Fluoroacetate (FAc) completely inhibited protein synthesis in cultures and caused a slight citrate accumulation. However, CO2 fixation continued and increased polyglucose formation occurred. Under these conditions added acetate was metabolized to polyglucose, as were glycine, serine, glyoxylate and succinate, but to a lesser extent; little or no formate or CO was utilised.Glyoxylate inhibited CO2 fixation in vivo, indicating that pyruvate is formed from acetyl-CoA and CO2 by pyruvate synthase. Two key enzymes of the reductive TCA cycle, citrate lyase and -ketoglutarate synthase were not detected in cell free extracts, but pyruvate synthase and phosphoenolpyruvate carboxylase were demonstrated. Summary Microautoradiography was used to show that chlorophyllous cells of young Picea abies stem slices are able to fix 14CO2, in the dark as well as in the light. The amount of 14CO2 fixed in the dark is much lower than that in the light. In the dark the concentration of radioactive label is equally high in all chlorophyllous cells of the stem. In the light, however, a gradient of radioactive assimilates extends from the stem surface to its centre, with the highest concentration being located in the phelloderm and the outer one-third of the cortex. This is in spite of even illumination and CO2 supply across the whole stem slice. In the dark, stem slices with and without bark show the same amount of radioactive label in the chlorophyllous cells of xylem, perimedullary region and pith. In the light, however, the concentration of radioactive assimilates in these cells is much higher in stem slices with bark than in stem slices without bark. It is assumed therefore that light fixation products of phelloderm and cortex are transported radially into the tissue inside the cambium.  相似文献

The lichen Peltigera aphthosa consists of a fungus and green alga (Coccomyxa) in the main thallus and of a Nostoc located in superficial packets, intermixed with fungus, called cephalodia. Dark nitrogenase activity (acetylene reduction) of lichen discs (of alga, fungus and Nostoc) and of excised cephalodia was sustained at higher rates and for longer than was the dark nitrogenase activity of the isolated Nostoc growing exponentially. Dark nitrogenase activity of the symbiotic Nostoc was supported by the catabolism of polyglucose accumulated in the ligh and which in darkness served to supply ATP and reductant. The decrease in glucose content of the cephalodia paralleled the decline in dark nitrogenase activity in the presence of CO2; in the absence of CO2 dark nitrogenase activity declined faster although the rate of glucose loss was similar in the presence and absence of CO2. Dark CO2 fixation, which after 30 min in darkness represented 17 and 20% of the light rates of discs and cephalodia, respectively, also facilitated dark nitrogenase activity. The isolated Nostoc, the Coccomyxa and the excised fungus all fixed CO2 in the dark; in the lichen most dark CO2 fixation was probably due to the fungus. Kinetic studies using discs or cephalodia showed highest initial incorporation of 14CO2 in the dark in to oxaloacetate, aspartate, malate and fumarate; incorporation in to alanine and citrulline was low; incorporation in to sugar phosphates, phosphoglyceric acid and sugar alcohols was not significant. Substantial activities of the enzymes phosphoenolpyruvate (PEP) carboxylase (EC 4.1.1.31) and carbamoyl-phosphate synthase (EC 2.7.2.5 and 2.7.2.9) were detected but the activities of PEP carboxykinase (EC 4.1.1.49) and PEP carboxyphosphotransferase (EC 4.1.1.38) were negligible. In the dark nitrogenase activity by the cephalodia, but not by the free-living Nostoc, declined more rapidly in the absence than in the presence of CO2 in the gas phase. Exogenous NH 4 + inhibited nitrogenase activity by cephalodia in the dark especially in the absence of CO2 but had no effect in the light. The overall data suggest that in the lichen dark CO2 fixation by the fungus may provide carbon skeletons which accept NH 4 + released by the cyanobacterium and that in the absence of CO2, NH 4 + directly, or indirectly via a mechanism which involves glutamine synthetase, inhibits nitrogenase activity.Abbreviations CP carbamoyl phosphate - EDTA ethylenedi-amine tetraacetic acid - PEP phosphoenolpyruvate - RuBP ribulose 1,5 bisphosphate  相似文献

The growth yield of Chlorella vulgaris, Y kJ defined as g cells harvested per kJ of light energy absorbed by the cells, was assessed in a turbidostat culture by varying CO2 and O2 partial pressures ( and ). The value of Y kJ ranged from 3.1×10-3 to 5.0×10-3 g cells/kJ under light-limited conditions [ = 1.02.4%, = 065%; total pressure of gas (composed of CO2, O2 and N2)=1 atm]. In the light-limited environment, the algal specific growth rate deteriorated appreciably with the increase of . The deterioration accounts for the above range of Y kJ observed. The growth inhibition due to oxygen that was defined by subtracting from 1.0 the ratio of at given values of to that at = 0% extended from 0.07–0.30 (7–30%). However, glycolate could not be detected in the turbidostat culture. Isotopic experiments on the specific rate of 14CO2 uptake also revealed that the inhibition due to oxygen was from 22–38% when was varied from 0 to nearly 100%. The pathway of autotrophic CO2 fixation in Methanobacterium thermoautotrophicum has been investigated by long term labelling of the organism with isotopic acetate and pyruvate while exponentially growing on H2 plus CO2. Maximally 2% of the cell carbon were derived from exogeneous tracer, 98% were synthesized from CO2. Since growth was obviously autotrophic the labelled compounds functioned as tracers of the cellular acetyl CoA and pyruvate pool during cell carbon synthesis from CO2. M. thermoautotrophicum growing in presence of U-14C acetate incorporated 14C into cell compounds derived from acetyl CoA (N-acetyl groups) as well as into compounds derived from pyruvate (alanine), oxaloacetate (aspartate), -ketoglutarate (glutamate), hexosephosphates (galactosamine), and pentosephosphates (ribose). The specific radioactities of N-acetylgroups and of the three amino acids were identical. The hexosamine exhibited a two times higher specific radioactivity, and the pentose a 1.6 times higher specific radioactivity than e.g. alanine. M. thermoautotrophicum growing in presence of 3-14C pyruvate, however, did not incorporate 14C into cell compounds directly derived from acetyl CoA. Those compounds derived from pyruvate, dicarboxylic acids and hexosephosphates became labelled. The specific radioactivities of alanine, aspartate and glutamate were identical; the hexosamine had a specific radioactivity twice as high as e.g. alanine.The finding that pyruvate was not incorporated into compounds derived from acetyl CoA, whereas acetate was incorporated into derivatives of acetyl CoA and pyruvate in a 1:1 ratio demonstrates that pyruvate is synthesized by reductive carboxylation of acetyl CoA. The data further provide evidence that in this autotrophic CO2 fixation pathway hexosephosphates and pentosephosphates are synthesized from CO2 via acetyl CoA and pyruvate.  相似文献

The activity of two carboxylating enzymes was studied in the green filamentous bacteriumChloroflexus aurantiacus. The carboxylation reaction involving pyruvate synthase was optimized using14CO2 and cell extracts. Pyruvate synthase was shown to be absent from cells ofCfl. aurantiacus OK-70 and present (in a quantity sufficient to account for autotrophic growth) in cells ofCfl. aurantiacus B-3. Differences in the levels of acetyl CoA carboxylase activity were revealed between cells of the strains studied grown under different conditions. The data obtained confirm the operation of different mechanisms of autotrophic CO2 assimilation inCfl. aurantiacus B-3 andCfl. aurantiacus OK-70: in the former organism, it is the reductive cycle of dicarboxylic acids, and in the latter one, it is the 3-hydroxypropionate cycle.  相似文献

In many legume nodules, the H2 produced as a byproduct of N2 fixation diffuses out of the nodule and is consumed by the soil. To study the fate of this H2 in soil, a H2 treatment system was developed that provided a 300 cm3 sample of a soil:silica sand (2:1) mixture with a H2 exposure rate (147 nmol H2 cm–3hr–1) similar to that calculated exist in soils located within 1–4 cm of nodules (30–254 nmol H2 cm–3hr–1). After 3 weeks of H2 pretreatment, the treated soils had a Km and Vmax for H2 uptake (1028 ppm and 836 nmol cm–3 hr–1, respectively) much greater than that of control, air-treated soil (40.2 ppm and 4.35 nmol cm–3 hr–1, respectively). In the H2 treated soils, O2, CO2 and H2 exchange rates were measured simultaneously in the presence of various pH2. With increasing pH2, a 5-fold increase was observed in O2 uptake, and CO2 evolution declined such that net CO2 fixation was observed in treatments of 680 ppm H2 or more. At the H2 exposure rate used to pretreat the soil, 60% of the electrons from H2 were passed to O2, and 40% were used to support CO2 fixation. The effect of H2 on the energy and C metabolism of soil may account for the well-known effect of legumes in promoting soil C deposition.  相似文献

The incorporation of 14CO2 by an exponentially growing culture of the autotrophic bacterium Methanobacterium thermoautotrophicum has been studied. The distribution of radioactivity during 2s–120s incubation periods has been analyzed by chromatography and radioautography. After a 2 s incubation most of the radioactivity of the ethanolsoluble fraction was present in the amino acids alanine, glutamate, glutamine and aspartate, whereas phosphorylated compounds were only weakly labelled. The percentage of the total radioactivity fixed, which was contained in the principal early labelled amino acid alanine, increased in the first 20 s and only then decreased, indicating that alanine is derived from primary products of CO2 fixation.The labelling patterns of alanine produced during various incubation times have been determined by degradation. After a 2 s 14CO2 pulse, 61% of the radioactivity was located in C-1, 23% in C-2, and 16% in C-3. The results are consistent with the operation of a previously proposed autotrophic CO2 assimilation pathway which involves the formation of acetyl CoA from 2 CO2 via one-carbon unit intermediates, followed by the reductive carboxylation of acetyl CoA to pyruvate.  相似文献

Stomatal opening on Vicia faba can be induced by high CO2 partial pressures (10.2%) in dark as well as in light. Stomatal aperture was measured in both cases with a hydrogen porometer. The distribution of 14C among early products of photosynthesis was studied. Comparisons are made with carboxylations occurring when stomata were open in the dark with CO2-free air and in light with 0.034% CO2. Results showed that in high CO2 partial pressure in light, less radioactivity was incorporated in Calvin cycle intermediates and more in sucrose. carboxylations and photorespiration seemed to be inhibited. In the dark in both CO2 conditions, 14C incorporation was found in malate and aspartate but also in serine and glycerate in high CO2 conditions. In the presence of ethylene, which enhances carnation flower senescence, carbohydrates contribute to ovary growth not only from the stem and calyx but also from the petals. With silver thiosulphate and ethanol treatments which delay flower senescence, the petals remain the active sink and ovary development is suppressed. Ethylene stimulated chloroplast development in the ovary wall. However, the calyx plus stem of all treatments showed the greater photosynthetic ability and transported a major portion of the synthesised products to the ovary.  相似文献

In continuous cultures of Chlorella fusca under steady state conditions, the CO2-fixation rate, the ATP-level, the apparent rate of photophosphorylation as calculated from the changes in the ATP-level during light to dark or dark to light transients and the energy charge were measured at various environmental conditions. During growth the energy charge was around 0.64. CO2-assimilation and the apparent ATP-synthesis were strongly dependant on light intensity, however the ATP-level was independant on it. Since the rates of apparent ATP-synthesis and of the CO2-fixation do not seem to be strictly correlated in a logic way when environmental factors are changed and furthermore the stoichiometry of 3 ATP necessary per CO2 fixed was never achieved, the described method frequently used for procaryotes to determine the in vivo rate of phosphorylation does not give valid results in highly compartimented eukaryotic cells.  相似文献

We did this work to discover the pathway of CO2 fixation into sugars in the dark during gluconeogenesis by the cotyledons of 5-day-old seedlings of Cucurbita pepo L. We paid particular attention to the possibility of a contribution from ribulosebisphosphate carboxylase. The detailed distribution of 14C after exposure of excised cotyledons to 14CO2 in the dark was determined in a series of pulse and chase experiments. After 4s in 14CO2, 89% of the 14C fixed was in malate and aspartate. In longer exposures, and in chases in 12CO2, label appeared in alanine, phosphoenolpyruvate, 3-phosphoglycerate and sugar phosphates, and accumulated in sugars. The transfer of label from C-4 acids to sugars was restricted by inhibition of phosphoenolpyruvate carboxykinase in vivo by 3-mercaptopicolinic acid. We conclude as follows. Initial fixation of CO2 in the dark is almost entirely into phosphoenolpyruvate, probably via phosphoenolpyruvate carboxylase (EC 4.1.1.31) which we showed to be present in appreciable amounts. Incorporation into sugars occurs chiefly, if not completely, as a result of randomization of the carboxyl groups of the C-4 acids and subsequent conversion of the oxaloacetate to sugars via the accepted sequence for gluconeogenesis. Ribulosebisphosphate carboxylase appears to make very little contribution to sugar synthesis from fat.  相似文献

In addition to membrane translocation, measured in the dark, it was found that pre-illumination of the chloroplasts resulted in an enhancement of sulfate uptake by 25% and of sulfite uptake by 55% as soon as the concentration of the ion in the incubation medium exceeded 2 mmol l-1. This amount which is additionally taken up after pre-illumination is less readily exchanged for other ions. Kinetics of the uptake in relation to pre-illumination time and to light intensity closely parallel those of titration of SH-groups by 5,5-dithiobis (2-nitrobenzoic acid). As a consequence, 10-6 mol l-1 DCMU completely inhibits the light triggered increase of uptake of both ions. Uncoupling with 10-6 mol l-1 CCCP increases the light induced 35SO 3 2- binding, but decreases that of 35SO 4 2- , demonstrating the need of ATP formation to initiate sulfate reduction. Rates of uptake, measured at different intensities of pre-illumination under nitrogen or in the presence of bicarbonate, suggest that the presence of a carbon skeleton increases the binding rate for both ions. With respect to 35SO 4 2- , the data further indicate a rate limiting step (ATP sulfurylase or adenosine 5-phosphosulfate sulfotransferase) which is activated by light, thus representing a control step to harmonize the rate of CO2 fixation and of sulfate incorporation. On the contrary, 35SO 3 2- is directly bound in relation to the amount of SH-groups, which in turn are created by the photosynthetic electron transport, resulting in Car-S-SO 3 - . Since the formation of SH-groups is maximal already at low light intensities, no effective control step for SO 3 2- incorporation is indicated.  相似文献

Microautoradiography indicated that 1-year-oldFraxinus excelsior L. stem chlorenchyma assimilated external14CO2 in mid-April, when buds were swollen, but before bud-break. The lenticel regions showed the highest amount of radioactively labeled assimilates. Labeled assimilates declined in the tangential direction with increasing distance from lenticels, suggesting that14CO2 entered the stem through the open intercellular spaces of lenticels. In the radial direction, the amount of radioactively labeled assimilates did not constantly decline with growing distance from the lenticel entrance. It was high in all lenticel phelloderm cells, which had high chlorophyll autofluorescence and very small starch grains, highest in the adjacent 4–6 rows of chlorenchyma, which had larger starch grains that increased in size towards the interior rows, and much lower in the inner cortex chlorenchyma, which had large starch grains. We suggest that the main function of the lenticel chlorenchyma (lenticel phelloderm plus 4–6 rows of adjacent cortex chlorenchyma) is the refixation of respiratory CO2 which could easily leave the stem intercellular spaces, rather than the fixation of external CO2. The lenticel chlorenchyma could reduce the loss of respiratory CO2 by its photosynthetic activity.  相似文献
