生物浸矿技术相比于传统的矿物加工技术具有成本低、易操作和污染小的特点,可以用来处理金精矿、低品位金矿、难处理金矿或者是高硫煤炭。为了更好地利用多种浸矿细菌的协同作用,本文综合阐述了生物浸矿的协同作用优势和存在的一些问题,对今后协同浸矿的发展做了预测。本文首先分析了3种主要浸矿细菌,包括氧化亚铁硫杆菌、氧化亚铁钩端螺旋菌和氧化硫硫杆菌等各自的生物学特性,接着重点分析了国内外近些年来浸矿细菌的协同作用研究进展情况和作用机制,最后展望了未来二十年内浸矿技术研究的发展方向。  相似文献

The bacterial genus Collimonas has the remarkable characteristic that it grows at the expense of living fungal hyphae under laboratory conditions. Here, we report the first field inventory of the occurrence and abundance of Collimonas in soils (n = 45) with naturally different fungal densities, which was performed in order to test the null hypothesis that there is a relationship between the presence of Collimonas and fungal biomass. Estimates of fungal densities were based on ergosterol measurements. Each soil was also characterized in terms of its physical and chemical properties and vegetation and management types. Culturable Collimonas was identified in plate-spread soil samples by its ability to clear colloidal chitin, in combination with a Collimonas-specific restriction fragment length polymorphism analysis of 16S rRNA PCR amplified from individual colonies. Using this approach, we found culturable collimonads only in (semi)natural grasslands. A real-time PCR assay for the specific quantification of Collimonas 16S rRNA in total soil DNA was developed. Collimonas was detectable in 80% of the soil samples, with densities up to 105 cells g−1 (dry weight) soil. The numbers of Collimonas cells per gram of soil were consistently lowest in fungus-poor arable soils and, surprisingly, also in fungus-rich organic layers of forest soils. When all soils were included, no significant correlation was observed between the number of Collimonas cells and ergosterol-based soil fungal biomass. Based on this result, we rejected our null hypothesis, and possible explanations for this were addressed.  相似文献

Biochemical consequences of mutational damage to common components of the Erwinia phosphoenolpyruvate-dependent phosphotransferase system (the HPr protein and enzyme I) were studied. The transport of glucose, mannose, fructose, and mannitol inErwinia was shown to require a preliminary induction of proteins of the phosphotransferase system. A drastic decrease in the rate of the transport of these carbohydrates was observed in ptsI and ptsH mutants. A disturbance in the common components suppresses the synthesis of inducible enzymes (-galactosidase, complexes of pectolate lyases and cellulases) and renders it resistant to catabolite repression by glucose, but mutants were shown to retain intracellular cAMP content. Erwinia mutants devoid of common components of the system lack phytopathogenic features. The appearance of an intact ptsI allele in the cell completely repaired pleiotropic disturbances in these mutants.  相似文献

To identify the bacteria that play a major role in the aerobic degradation of petroleum polynuclear aromatic hydrocarbons (PAHs) in a marine environment, bacteria were enriched from seawater by using 2-methylnaphthalene, phenanthrene, or anthracene as a carbon and energy source. We found that members of the genus Cycloclasticus became predominant in the enrichment cultures. The Cycloclasticus strains isolated in this study could grow on crude oil and degraded PAH components of crude oil, including unsubstituted and substituted naphthalenes, dibenzothiophenes, phenanthrenes, and fluorenes. To deduce the role of Cycloclasticus strains in a coastal zone oil spill, propagation of this bacterial group on oil-coated grains of gravel immersed in seawater was investigated in beach-simulating tanks that were 1 m wide by 1.5 m long by 1 m high. The tanks were two-thirds filled with gravel, and seawater was continuously introduced into the tanks; the water level was varied between 30 cm above and 30 cm below the surface of the gravel layer to simulate a 12-h tidal cycle. The number of Cycloclasticus cells associated with the grains was on the order of 103 cells/g of grains before crude oil was added to the tanks and increased to 3 × 106 cells/g of grains after crude oil was added. The number increased further after 14 days to 108 cells/g of grains when nitrogen and phosphorus fertilizers were added, while the number remained 3 × 106 cells/g of grains when no fertilizers were added. PAH degradation proceeded parallel with the growth of Cycloclasticus cells on the surfaces of the oil-polluted grains of gravel. These observations suggest that bacteria belonging to the genus Cycloclasticus play an important role in the degradation of petroleum PAHs in a marine environment.  相似文献

糜酶是一类丝氨酸蛋白酶,主要存在于肥大细胞的分泌颗粒和细胞间质。成熟的酶为一糖蛋白,由226个氨基酸组成,分子量为30kD,其酶活力能被丝氨酸蛋白酶抑制剂所抑制,而不受血管紧张素Ⅰ转换酶抑制剂的影响。糜酶的cDNA和基因组DNA均已被克隆和测序。该酶和神经炎症反应、血管活性肽代谢和细胞外基质代谢等有密切联系,在心脏血管紧张素Ⅱ形成中起重要作用。  相似文献

艾滋病痴呆综合征(acquired immunodeficiency syndrome dementia complex, ADC)是由人类免疫缺陷病毒(human immunodeficiency virus,HIV)侵犯中枢神经系统,造成患者意识、行为和运动能力改变为主要临床表现的综合征。约50%的HIV感染者表现出不同形式的认知功能损伤。ADC的发病机制尚未完全阐明,诊断也无明确的病理和生化指标。目前,蛋白质组学技术以其高灵敏度和高通量的特点广泛应用于传染病研究。本文综述了近年来ADC蛋白质组学研究领域的进展,包括ADC蛋白质组学研究所用样品、蛋白质组学技术及其在ADC研究中的应用,为ADC的诊断和发病机制研究提供新思路。  相似文献

The aims of this work were to develop a high-rate fluidized-bed bioprocess for ferric sulfate production, to characterize biomass retention, and to determine the phylogeny of the enrichment culture. After 7 months of continuous enrichment and air aeration at 37 degrees C, the iron oxidation rate of 8.2 g Fe(2+) L(-1)h(-1) (4.5.10(-12) g Fe(2+) cell(-1) h(-1)) was obtained at a hydraulic retention time (HRT) of 0.6 h. However, oxygen supply became the rate-limiting factor. With gas mixture (99.5% O(2)/0.5% CO(2) (vol/vol)) aeration and HRT of 0.2 h, the iron oxidation rate was 26.4 g Fe(2+) L(-1)h(-1) (1.0.10(-11) g Fe(2+) cell(-1) h(-1)). Leptospirillum sp. was predominant in the mesophilic fluidized-bed reactor (FBR) enrichment culture as determined by fluorescent in situ hybridization, while Acidithiobacillus ferrooxidans was not detected. Denaturing gradient gel electrophoresis (DGGE) of the amplified partial 16S rDNA showed only three bands, indicating a simple microbial community. DGGE fragment excision and sequencing showed that the populations were related to L. ferriphilum (100% similarity in sequence) and possibly to the genus Ferroplasma (96% similarity to F. acidiphilum). Jarosite precipitates accumulated on the top of the activated carbon biomass carrier material, increasing the rate of iron oxidation. The activated carbon carrier material, jarosite precipitates, and reactor liquid contained 59% (or 3.71.10(9) cells g(-1)), 31% (or 3.12.10(10) cells g(-1)) and 10% (or 1.24.10(8) cells mL(-1)) of the total FBR microbes, respectively, demonstrating that the jarosite precipitates played an important role in the FBR biomass retention.  相似文献
