A number of factors which may correlate with the levels of α-glucosidase and β-glucosidase in cultured amniotic fluid cells have been investigated. Fluctuations in enzyme activity occurred as passage numbers increased. Whereas α-glucosidase showed a consistently lower activity in the earlier passages compared to the later ones, the results for β-glucosidase were equivocal. Both enzymes showed an increase in activity correlated with the time taken by the cells to reach confluency in the third passage. When replicate cultures were assayed daily after subculture, neither enzyme showed any change correlated with time. When cultures were grown in parallel in Ham's F10 and Eagle's M.E.M. tissue culture media, the activity of both enzymes was unaffected. Cell strains cultured from serial samples of amniotic fluid from the same woman had differing enzyme levels unrelated to gestational age. 相似文献

In the present study, an established controlled burn wound model was used to test the hypothesis that controlled surface contamination with is capable of generating a noninvasive method for the creation of a reproducible deep tissue burn wound infection. Using a liquid tight-wound chamber in Yorkshire pigs, partial-thickness burns were inoculated with saline-immersed for 24 hours. Noninoculated burns and unwounded skin immersed in normal saline served as controls. Bacterial cultures of wound fluid were performed daily, and tissue biopsies for bacteriological and histological evaluations were performed on days 1, 3, and 5. was only recovered from -inoculated wounds (tissue and fluid), whereas all controls contained endogenous only. The number of colony-forming units per gram of wound tissue did not correlate with the bacterial counts found in the overlying wound fluid for any wounds. Fluid counts were consistently higher than tissue counts by two logs. -inoculated wounds showed three times deeper tissue destruction than control wounds. Obtaining consistently deep tissue colonization without cross-contamination among wounds, this study introduces a noninvasive model for controlled burn wound infection suitable for future investigations regarding the efficacy of topical antibiotic wound treatment in experimental burns. 相似文献

Gamma-Aminobutyric acid (GABA) levels in cerebrospinal fluid were measured in seven patients with tension headache and 12 patients with migraine. GABA was detected only during the migraine attack. The results suggest disordered GABA metabolism in migraine. 相似文献

Serum samples from psoriatic patients were subjected to quantitation of serum IgA, IgG and IgM levels and screening of tissue autoantibodies including ANA, GPA, MTA and SMA. Serum IgA was found to be significantly elevated and incidences of ANA and GPA were also increased. Their significance and possible mechanism were discussed. 相似文献

The concentration of prostaglandin F (PGF) has been measured by radioimmunoassay in follicular fluid collected from follicles at various time intervals after treatment of prepuberal gilts with pregnant mare serum gonadotropin and human chorionic gonadotropin to induce ovulation. A high proportion of animals will ovulate 116 +/- 8 hr after treatment. Pre-ovulatory follicles can be identified on the basis of gross morphological apperance 10-12 hr before the predicted time of ovulation. The concentration of PGF in fluid from follicles judged not to be pre-ovulatory was relatively constant at about 0.45 ng per g and appeared to be independent of the time of sampling. An increase in the concentration of PGF was observed in fluid collected from follicles classified as destined to ovulate. This increase became more pronounced as the time of ovulation approached and reached a maximum at or about the time of follicle rupture. These data provide evidence in support of a role for prostaglandins in the ovulatory process in the pib. 相似文献

Proliferative vitreoretinopathy is the most common reason for failure in retinal reattachment surgery. Since both substance P (SP) and SP receptors were found to be present in the human eye, and as pharmacological studies suggest an importance of SP for ocular functions, we investigated intraocular fluids for the presence of SP in eyes elected for cataract surgery, retinal detachment surgery and retina surgery for severe proliferative vitreoretinopathy (PVR) as well as in eyes with proliferative diabetic retinopathy (PDR). High performance liquid chromatography and radioimmunoassay (RIA) for SP immunoreactivities were performed. The SP mean concentration in intraocular fluid (IOF) of patients for cataract surgery (CS) was 2.2 fmol/ml, for retinal detachment (RD) was 2.7 fmol/ml and for PDR was 1.9 fmol/ml; significantly higher levels (mean concentration of 26.9 fmol/ml) were measured in eyes with PVR. HPLC analysis revealed two immunoreactive peaks coeluting with synthetic SP and SP-sulfoxide, indicating that RIA values represent authentic SP. We conclude that SP may play an important role in PVR. Since SP antagonists are known to inhibit a variety of SP effects in the eye, there might be a useful tool to reveal the importance of SP in this disease and, in this instance, a new possible treatment. 相似文献

Daily fluctuations in cerebrospinal fluid cyclic AMP levels were found in neurologically intact rhesus monkeys. When animals were maintained in continuous dim light as opposed to a 12L/12D regimen, the fluctuations in cerebrospinal fluid cyclic AMP levels appeared to be reduced. Knowledge of these daily fluctuations may be helpful for protocols of future experiments concerning measurements of CSF cAMP. 相似文献

The concentration of ions in plant cells and tissues is an essential factor in determining physiological function. In the present study, we established that concentration gradients of mobile ions exist in both xylem exudates and tissues within a barley (Hordeum vulgare) primary leaf. For K+ and NO3?, ion concentrations generally decreased from the leaf base to the tip in both xylem exudates and tissues. Ion gradients were also found for Pi and Cl? in the xylem. The hydathode strongly absorbed Pi and re‐translocated it to the rest of the plant, whereas Cl? was extruded. The ion concentration gradients developed early during leaf growth, increased as the tissue aged and remained under both high and low transpiration conditions. Measurement of the expression profiles of Pi, K+ and NO3? transporters along the longitudinal axis of the leaf revealed that some transporters are more expressed at the hydathode, but for most transporters, there was no significant variation along the leaf. The mechanisms by which longitudinal ion gradients develop in leaves and their physiological functions are discussed. 相似文献

A new method has been developed for prelabeling tissue ATP pools with 32P inorganic phosphate (Pi) and for the subsequent isolation of [32P]cAMP and [32P]ATP. The new method of prelabeling eliminates the need to separate trace amounts of radioactive cAMP from radioactive breakdown products of adenine formed in tissues prelabeled with [3H]- or [14C]adenine. The effect of epinephrine to increase [32P]cAMP levels in rat ventral prostate tissue fragments has been studied in terms of increase in the ratio of [32P]cAMP/[32P]ATP in the absence and presence of various phosphodiesterase inhibitors. Tissue prelabeling with 32Pi labels GTP as well as ATP (and other nucleoside triphosphates); thus the method lends itself to the isolation of [32P]cGMP as well as [32P]cAMP from the same tissue sample. 相似文献

Nicotinamide administration can elevate plasma and brain choline levels and produce a marginal increase in striatal acetylcholine levels in the rat. We now report that subcutaneous nicotinamide produces a substantial and long-lasting rise in asternal cerebrospinal fluid (CSF) levels of choline in free-moving rats, possibly through the enzymatic formation of N1-methylnicotinamide (NMN) in brain. CSF choline levels peaked 2 hours after nicotinamide administration and were accompanied by increases in striatal, cortical, hippocampal and plasma choline levels. The enzymatic formation of [3H]NMN in rat brain was evaluated by incubating aliquots of rat brain cytosol with unlabelled nicotinamide and the methyl donor [3H]S-adenosylmethionine. High performance liquid chromatography and radiochemical detection demonstrated that [3H]NMN was specifically formed by a brain cytosolic enzyme. The production of [3H]NMN was dependent on exogenous nicotinamide and could be prevented by denaturing the cytosol. The metabolism of nicotinamide to NMN in rat brain may explain the rise in CSF choline levels since NMN, a quaternary amine, can inhibit choline transport at the choroid villus and reduce choline clearance. 相似文献
