Abstract Two endocrine cell types, PbH+ and PAS+ cells, were recognized in the pars intermedia (PI) of the roach, Leuciscus rutilus, by light and electron microscopy. They can be distinguished by the appearance of their secretory granules. The PAS+ cells stain with toluidine blue in semithin sections as opposed to the PbH+ cells. After 5 days' treatment with metopirone the PbH+ cells are highly activated whereas the PAS+ cells show no response. The PbH+ cells are assumed to produce MSH. In roaches adapted to diluted sea water (10%) the transfer to fresh water for 1—1 1/2 hours or reserpine injections for the same period had no notable effect on neither cell type. Three kinds of axons innervate the PI. Two of these, which have granules averaging 111 nm and 151 nm in diameter, originate in the nucleus preopticus (NPO). The third type of axons has granules of an average size of 74 nm and is probably aminergic. The glia cells in this part of the neurohypophysis (NH) are granular pituicytes, which are innervated by all three axon types. The border between the NH and the PI is composed of a double basement membrane from which a network extends into the NH. This network, the basement membrane and the capillaries are the main sites for the terminations of the axons although nerve fibers also invade the endocrine tissue. Only axon type 3 was observed to synapse frequently with the PbH+ cells.  相似文献

Three different endocrine cell types were identified by electron microscopy in the rostral pars distalis (RPD) of the roach, Leuciscus rutilus. The cells which are characterized by the morphology and sizes of their secretory granules are assumed to secrete prolactin, ACTH and TSH. Prolactin cells occur in two forms which could be separated on account of the sizes of their granules. Intra-peritoneal injections of reserpine had a strong and persistent effect 24 hours after the treatment. The prolactin cells with small granules reacted with a marked release of secretion. The border of the RPD is composed of a so-called double basement membrane in contact with the perivascular spaces and forms extensive ramifications into the rostral neurohypophysis (RNH). Nerve fibers of different categories terminate at the border and its network in the RNH. Substances released from these terminals may influence the endocrine cells of the RPD. The ACTH cells and the prolactin cells are innervated by axons with granules less than 100 nm. The nomenclature of type ‘A’ fibers and ‘B’ fibers is discussed because it was difficult to make a reliable discrimination of the fibers and their nature by the sizes of their granules. However, four types of nerve fibers were distinguished in connection with the RPD although the presence of more types was indicated. In all four types the size variations of the granules are great and overlapping. The average sizes of the granules in these fibers are 157, 114, 119 and 81 nm. The origin of the nerve fibers innervating the RPD is discussed. All morphological data indicate a complex hypothalamic control of the RPD of the roach.  相似文献

Summary The hypothalamus of the teleost fish Leuciscus rutilus was investigated with the Falck-Hillarp technique. The nucleus preopticus (NPO) and the nucleus lateralis tuberis (NLT) contain no fluorescent, i.e. catecholaminergic cells. Green fluorescent fibers probably originating from the paraventricular organ and/or the preoptic recess organ, are intermingled with the cells.The electron microscopical study was based on the three fixatives glutaraldehyde-osmium tetroxide, osmium tetroxide and potassium permanganate. In the NPO two cell types are recognized, characterized mainly by dense core vesicles (dcv) with measured diameter of 130 nm and 170 nm across respectively. The endoplasmic reticulum in the former cell type generally has large dark inclusions measuring from 175 to 375 nm across, which are also found in the neurite. In the NLT, four different cell types are identified, some of which are subject to considerable variations. The rostral and the medial parts of the nucleus include a large cell type (I) with dcv of diameter 170 nm. The medial part also has a small cell type (II) with dcv of 80 nm. The lateral part is characterized by two cell types (III, IV). Cell type III occurs in three forms with dcv of about 140 nm. The fourth cell type (IV) is rare and contains irregularly formed granules, the most circular ones measuring about 130 nm and the most elongated ones 110 nm×210 nm. The ventrolateral part contains the same cell types (except for type II) as those found in the lateral and medial parts.The morphological differentiation of the NLT as well as its different cell types strongly indicates its functional diversity.After permanganate fixation the secretory granules of the different cell types in the NPO and the NLT appear as empty vesicles. This method also reveals that the cell types of the two nuclei have dcv of about 90 nm. The ultrastructure of endocrine cells in the midgut of the adult mosquito, Aedes aegypti, resembled that of endocrine cells in the vertebrate gastro-intestinal tract. Midgut endocrine cells, positioned basally in the epithelium as single cells, were cone-shaped and smaller than the columnar digestive cells. The most distinctive characteristic of endocrine cells was numerous round secretory granules along the lateral and basal plasma membranes where contents of the granules were released by exocytosis. Secretory granules in each individual cell were exclusively of one type, either solid or 'haloed', and for all cells observed, the range in granule diameter was 60-120 nm. The cytoplasm varied in density from clear to dark. Lamellar bodies were prominent in the apical and lateral cellular regions and did not exhibit acid phosphatase activity. The basal plasma membrane was smooth adjacent to the basal lamina, whereas in digestive cells the membrane formed a labyrinth. Some endocrine cells reached the midgut lumen and were capped by microvilli; a system of vesicles and tubules extended from beneath the microvilli to the cell body. An estimated 500 endocrine cells were distributed in both the thoracic and abdominal regions of the adult midgut. In one midgut, we classified a sample of endocrine cells according to cytoplasmic density and granule type and size; endocrine cells with certain types of granules had specific distributions within the midgut.  相似文献

Summary The structure of the neurohypophysis of Poecilia latipinna (green molly, sailfin molly) was studied with the electron microscope. Profile diameters of neurosecretory granules in the non-myelinated neurohypophysial nerve fibres were measured and mathematically corrected for error due to section thickness. Six different types of nerve fibres could be distinguished by statistical classification of their granules and by other ultrastructural features. One fibre-type (type B) contained granules with a mean diameter of 85 nm, and the other five types (types Ala, Alb, A2, A3 and A4) all contained granules with mean diameters greater than 100 nm. Synaptic contacts were observed between type B fibres and all the adenohypophysial cell-types, although in the case of the ACTH cells the synapses were separated from the cell membrane by a continuous double basement membrane. Type A fibres were observed to contact the cells of the proximal pars distalis and pars intermedia, but did not form synapses. However, synapses occurred between type A fibres and pituicytes, and between type A fibres and the pericapillary basement membrane in the interior of the neurohypophysis. The possible roles of the different types of nerve fibres in controlling the adenohypophysial cells are discussed in the context of evidence from other teleosts.We thank Mr. W.A. Thomson and Mr. D.I. The cells of the adenohypophysis of the Japanese quail were studied by both light and electron microscopy after exposure to long photoperiods or injection of lutenizing hormone-releasing hormone (LRH). Six cell types were identified in the adenohypophysis by examining alternate thick and thin sections by light and electron microscopy. In the cephalic lobe, there are four types of glandular cells. They are the prolactin cells, ACTH cells, TSH cells, and gonadotropic cells (FSH?). In the caudal lobe, there are two types of cells, STH cells and gonadotropic cells (LH?). After exposure to long daily photoperiods, gonadotropic cells in both lobes were strongly activated. They became larger and accumulated many granules. ACTH cells became vacuolated; granules were sparse. Synthetic LRH injection (10 mug/0.2 ml/day) for 10 days to the non-photostimulated quail stimulated certain numbers of the gonadotropic cells in the both lobes, although the response of the cells was less than that induced by photostimulation. Summary The pancreatic endocrine tissue of Fugu rubripes rubripes consists of numerous round principal islets (Brockmann bodies) of various sizes scattered around the gall-bladder. The endocrine cells are divided into A-, B-, D-, and Ff-cells. Each cell type was identified by comparing thick and thin sections in both light and electron microscopy. Aldehyde-fuchsin positive B-cells contain numerous round secretory granules (average diameter 300 nm) each of which has a round compact core of moderate density; a narrow space exists between this core and the limiting membrane. Grimelius' silver positive A cells contain round secretory granules (average diameter 360 nm) with a hexagonal or tetragonal crystalline core (average diameter 170 nm) of high density; the silver grains preferentially appear in the space between the limiting membrane and the core. The crystalline core of each -granule often contains an appendix-like structure of variable shape. D cells blackened by the silver impregnation method of Hellman and Hellerström (1960) have round secretory granules (average diameter 320 nm) filled with a flocculent material of low density. The fourth cell type (Ff-cell) has a clear cytoplasm after differential staining for light microscopy. By electron microscopy, this cell has elongated fusiform secretory granules (520 nm average length × 230 nm average width) filled with numerous filaments arranged in parallel with the longitudinal axis. Figures suggesting granule formation in the sacs of the Golgi apparatus were obtained in all of islet cell types. Equivalents of emiocytotic release of secretory granules were encountered in the A and Ff cells.  相似文献

The thyrotropin (TSH) producing cells are distributed in the rostral and proximal pars distalis. This cell type is the smallest and most infrequent cell of the adenohypophysis. Its cytology is similar to the smallest gonadotropic (GTH) cells although the two cell types can be separated by the size of the small secretory granules (diameter less than 200 nm) in the TSH cells. In presmolts and smolts the cells are more numerous than in parr and adult salmon and have cytological features indicating an increased activity. This was also the case after intraperitoneal injections of synthetic TRH. Antisera to carp GTH and salmon GTH cross-reacted with both the GTH and the TSH cells. Anti-human TSH cross-reacted only with the TSH cells which confirms the assumption of antigenic similarity between human and fish TSH.  相似文献

Summary In the gastric mucosa of two teleost species, the perch (Perca fluviatilis) and the catfish (Ameiurus nebulosus) three endocrine cell types were found, located predominantly between the mucoid cells of the gastric mucosa. A fourth cell type is present in the gastric glands of catfish. Each cell type was defined by its characteristic secretory granules. Type-I cells were predominant in both fish. These cells contained round or oval granules with a pleomorphic core. The average diameter of granules was 400 nm for the perch and 270 nm for the catfish. Type-II cells of both species displayed small, highly osmiophilic granules about 100 nm in diameter. The secretory granules of type-III cells (260 nm in the perch and 190 nm in the catfish) were round or slightly oval in shape and were filled with a finely particulate electron-dense material. Type-IV cells of the catfish were found in the gastric glands only. Their cytoplasm was filled with homogeneous, moderately electron-dense granules averaging 340 nm in diameter. The physiological significance of these different morphological types of gastric endocrine cells requires further investigation.  相似文献

Summary In the pancreas of Scyliorhinus stellaris large islets are usually found around small ducts, the inner surface of which is covered by elongated epithelial cells; thus the endocrine cells are never exposed directly to the lumen of the duct. Sometimes, single islet cells or small groups of endocrine elements are also incorporated into acini. Using correlative light and electron microscopy, eight islet cell types were identified:Only B-cells (type I) display a positive reaction with pseudoisocyanin and aldehyde-fuchsin staining. This cell type contains numerous small secretory granules (Ø280 nm). Type II- and III-cells possess large granules stainable with orange G and azocarmine and show strong luminescence with dark-field microscopy. Type II-cells have spherical (Ø700 nm), type III-cells spherical to elongated granules (Ø450 × 750 nm). Type II-cells are possibly analogous to A-cells, while type III-cells resemble mammalian enterochromaffin cells. Type IV- cells contain granules (Ø540 nm) of high electron density showing a positive reaction to the Hellman-Hellerström silver impregnation and a negative reaction to Grimelius' silver impregnation; they are most probably analogous to D-cells of other species. Type VI-cells exhibit smaller granules (Ø250 × 500 nm), oval to elongated in shape. Type VI-cells contain small spherical granules (Ø310 nm). Type VII-cells possess two kinds of large granules interspersed in the cytoplasm; one type is spherical and electron dense (Ø650 nm), the other spherical and less electron dense (Ø900 nm). Type VIII-cells have small granules curved in shape and show moderate electron density (Ø100 nm). Grimelius-positive secretory granules were not only found in cell types II and III, but also in types V, VI, and VII. Abstract The double immunofluorescent method indicates that two types of gonadotropes occur in the proximal pars distalis of the teleost fish Rutilus rutilus. With rabbit anti-carp GTH as the first antibody the two cell types cross-react whereas after rabbit anti-salmon GTH as the first antibody only the so-called globular cell, known to produce gonadotropin in teleosts, cross-reacts. The cytological differences between the two assumed gonadotropic cells are reviewed. The pseudoisocyanine technique gave a selective fluorescent picture of the assumed gonadotropes. Fine structural analysis shows that the gonadotropic cells close to the neurohypophysis are innervated by one type of fiber in contrast to most of the gonadotropic cells, which are not innervated. In the latter case the neuroendocrine information reaches the gonadotropic cells by a median eminence system in which the intervascular channels in the proximal pars distalis are the last parts.  相似文献

Summary Using the electron-microscopic immunogold method, vasotocin, isotocin, somatostatin (SRIF), gonadotrophin-releasing hormone (LHRH) and corticotrophin-releasing factor (CRF)-like immunoreactivities were localized in separate neurosecretory fibres in the pituitary of a teleost fish Poecilia latipinna. Antigenicities were preserved in sections of conventionally fixed tissue, except in the case of LHRH and CRF-like substances which were sensitive to osmium postfixation. Under the same fixation conditions, ultrastructural differences were observed between the 5 fibre types, and morphometric analysis of their granule sizes revealed significant differences in mean diameter except between vasotocin and isotocin fibres.Terminal-like regions of each type were identified on blood vessels, glial cells or other fibres in the neurohypophysis, on the basement lamina of the adenohypophysis, or directly on adenohypophysial endocrine cells. The fibres containing the two neurohypophysial hormones, originating from separate preoptic perikarya, were intermingled with, and may form endings near all the adenohypophysial cell types except those secreting prolactin. Although both types had similar mean granule diameters, the granules in the vasotocin fibres (mean 135 nm) were markedly less electron dense than those in the isotocin fibres (mean 140 nm). SRIF-immunoreactive fibres (mean 101 nm) appeared to form synapse-like endings on the somatotrophs, and a few thyrotrophs in the proximal pars distalis, and near the pars intermedia cells. An LHRH-positive type (mean 103 nm) contacted only the gonadotrophs of the proximal pars distalis. The rarer CRF-like fibres (mean 116 nm) appeared to project mainly towards the pars intermedia, but a few appeared to terminate rostrally near the adrenocorticotrophic cells.The significance of these observations is discussed in relation to the direct neurosecretory control of adenohypophysial function in teleosts.  相似文献

Summary Cholecystokinin (CCK) has been localized by the immunogold technique in a type of endocrine cell of the dog duodenum characterized by small (166±38 nm) secretory granules with fairly dense, homogeneous core separated from its enveloping membrane by a thin clear space. The CCK cell is immunocytochemically distinct and cytologically different from other types of endocrine cells, as the secretin, GIP and motilin cells, already identified in the dog duodenum.  相似文献

The endocrine pancreas of the freshwater teleost Pimelodus maculatus was studied by electron microscopy. Based on the granule morphology 2 cell types were described: Secretory granules of type I cells are rounded, nearly completely filling the limiting membranous sac which measures from 120 to 150 nm in diameter; the type II granules are also rounded and measure from 220 to 270 nm in diameter; they consist of an eccentrical electron dense core separated from the limiting membrane by a wide electron lucent halo. Four endocrine cell types were identified ultrastructurally in the pancreas of the grass lizard, Mabuya quinquetaeniata. These cells were similar in shape, location and frequency to the previously described B-, A-, D- and PP-cells. The secretory granules of the B-cells were round or oval in profile, with an internal core of variable shape. The mean diameter of the B-cell granules was 780 nm (range 350–1000 nm). The A-cell granules were round, oval or irregular in shape and highly electron dense, with a narrow electron lucent space between the core and the limiting membrane. The mean diameter of these granules was 450 nm (range 200–750 nm). The D-cell granules were round, oval or irregular and of moderate electron density, with an average diameter of 340 nm (range 200–500 nm). The limiting membrane was closely apposed to the core or separated from it by a narrow lucent space. PP-cell granules were round with high electron density and with a narrow space between the core and the limiting membrane, and their average diameter was 150 nm (range 50–350 nm); these secretory granules accumulated at the cytoplasmic process. Tracing of the cytoplasmic processes of PP-cells in serially cut ultrathin sections revealed that most of these processes ended in the vicinity of blood capillaries, indicating that the PP-cells were endocrine rather than paracrine.  相似文献

Summary Human duodenal endocrine cells reactive with antibodies to cholecystokinin (CCK) 33 (10–20) and/or gastrin 34 (1–15) were studied by a combination of immunohistochemical and electron-microscopic methods. By immunohistochemistry, three types of endocrine cells were distinguished in human duodenal mucosa, i.e., those only positive for only CCK, those positive for both CCK and gastrin and those only positive for only gastrin. Ultrastructurally, the first cell type is characterized by many secretory granules with an eccentric dense core (mean diameter; 271+-74 nm). The second cell type, which was less frequent than the other two, has ultrastructural features that resemble type-I cells. The last cell type was composed of two types of cells containing small secretory granules identical to those of IG cells (mean diameter; 171+-31 nm) or large secretory granules indistinguishable from those of I cells (mean diameter; 286+-50 nm).  相似文献

Summary The cellular binding sites of anti-oPRL IgG and anti-bSTH IgG were demonstrated in the pituitary glands of Lepidosiren paradoxa, Rana temporaria and Ambystoma mexicanum by means of the unlabeled antibody enzyme method by light and electron microscopy (the latter only in Lepidosiren). With the light microscope PRL or PRL-like substances and STH or STH-like substances were revealed in two different cell types in the distal lobe corresponding to the acidophils. However, as a result of the insufficient differentiation of the acidophils in Lepidosiren after staining with Brookes' procedure it was not possible to distinguish the two types of acidophils in this animal. Treatment with low dilutions of both anti-oPRL and anti-bSTH IgG revealed simultaneous immunocytochemical staining in both types of acidophils in Lepidosiren and in Rana. These results, indicating that there is antigenic cross-reaction between anti-oPRL and anti-bSTH IgG and both PRL and STH in these animals, are discussed.The electron microscopic investigations of Lepidosiren revealed that the specific anti-oPRL IgG reactive cells contain granules ranging in size from 200 to 300 nm, while the specific anti-bSTH IgG reactive cells contain smaller immunoreactive granules ranging from 80 to 160nm.  相似文献

Summary In order to contrast anionic sites, in mouse lung alveoli, two staining procedures were applied: (a) staining with Ruthenium Red and Alcian Blue and (b) staining with Cuprolinic Blue in a critical electrolyte concentration method. The Ruthenium Red-Alcian Blue staining procedure revealed electron-dense granules in the alveolar basement membrane. The granules were closely associated with the epithelial cell membrane and continued to stain even when the procedure was carried out at a low pH, indicating the presence of sulphate groups in the granules.After staining with Cuprolinic Blue, electron-dense filaments, also closely associated with the cell membrane, became visible in the basement membrane of type I epithelial cells. Their length depended on the MgCl2 concentration used during staining. At 0.4m MgCl2, the length was mostly within the range 100–180 nm. Using a modified Cuprolinic Blue method, the appearance of the filaments closely resembled that of spread proteoglycan monomers with their side-chains condensed. The basement membrane of type II epithelial cells also contained filaments positive towards Cuprolinic Blue; their length, however, was smaller in comparison with those of type I epithelial cells. The filaments lay in one plane and provided the whole alveolus with an almost continuous sheet of anionic sites. Cuprolinic Blue staining also revealed filaments in the basement membrane of the capillary endothelial cells. Furthermore, Cuprolinic Blue-positive filaments (average length about 40 nm) became apparent in close contact with collagen fibrils and separated from each other according to the main banding period of the collagen fibrils (about 60 nm), indicating a specific ultrastructural interaction between these two components. Filaments connecting collagen fibrils with each other were also detected.  相似文献
