We describe the isolation and characterization of 14 microsatellite loci in the striped field mouse from genomic DNA-enriched
libraries in this paper. The 14 new loci were tested in 24 individuals from four populations in Southwest China. These loci
were highly polymorphic with numbers of alleles per locus ranging from 6 to 14 and expected heterozygosities from 0.789 to
0.925. All loci followed Hardy–Weinberg expectations except SFM4, SFM11 and SFM13 loci. No significant linkage association
was found among all loci. The 14 novel polymorphic microsatellite loci will be useful in studying phylogeography and population
genetics of the striped field mouse. 相似文献

Eleven polymorphic microsatellite loci were characterized for the endangered conifer Taxus yunnanensis. Eight loci were isolated through SSR-anchored PCR, one locus was developed by cross-species amplification tests, while the
last two loci were obtained from cross-species microsatellite sequences available in GenBank. Variability of these markers
was tested in 48 individuals collected from its main distribution range in Southwest China. The number of alleles per locus
ranged from 2 to 9, and the observed and expected heterozygosity varied from 0.000 to 0.625 and from 0.062 to 0.853, respectively.
Ten of these eleven loci were significantly deviated from Hardy–Weinberg expectation, except TS07 which showed a distinct
heterozygote excess. The availability of these new polymorphic microsatellite markers will provide an ideal marker system
for detailed population genetics studies in T. yunnanensis and potentially also for closely related species. 相似文献

Piper polysyphonum is an endemic species in southeast Asia, in the narrow habitat located in the Chinese provinces of Guizhou and Yunnan, and
the country of Laos. Recently, loss of forests due to agricultural activity has dramatically reduced the habitat and population
size of P. polysyphonum. In this study, eleven primer sets of polymorphic microsatellite DNA loci were developed for P. polysyphonum. Allele numbers ranged from two to ten, with observed heterozygosities ranging from 0.222 to 0.889. Four loci exhibited a
departure from Hardy–Weinberg equilibrium, possibly due to population admixture. No loci pairs revealed significant linkage
disequilibrium. Among the eleven loci, two with extremely high numbers of TCG repeats were obtained. The polymorphic microsatellite
DNA markers reported here should provide a helpful means to address questions concerning population structure and demographic

The Yangtze sturgeon (Acipenser dabryanus) is an endemic species in China. Using 454 sequencing, eight polymorphic tri‐, tetra‐, penta‐, and hexanucleotide microsatellite loci were isolated in this study. The raw sequence data from a one‐eighth run of 454 sequencings were 38.0 Mbp containing 94 222 reads/sequences. Of 80 microsatellite loci, only eight loci were polymorphic in a population of 30 individuals. The number of alleles per locus ranged from 4 to 14 (mean 7.62), and the observed heterozygosities varied between 0.46 and 0.88 (mean 0.74). Cross amplification was tested in congeneric species Acipenser sturio and Acipenser sinensis. These new microsatellite markers will be useful for further studies on genetic variation, parentage analysis, and conservation management for this critically endangered species. 相似文献

Fifteen polymorphic dinucleotide microsatellite loci were characterized for Erynnis propertius using an enrichment protocol. The number of alleles varied from nine to 28 for a sample of 24 individuals. Observed heterozygosities ranged from 0.25 to 0.96. Homozygote excess was detected for 10 loci. Eleven polymorphic microsatellite loci were obtained from a GA enriched genomic library, constructed from DNA of buck’s‐horn plantain (Plantago coronopus). The microsatellite loci were tested on 24 genotypes. These plants were collected from meadows along the coast, located on 11 sites ranging from the southwest to the northeast of the Netherlands. In this set of plants the isolated microsatellites were highly polymorphic with 3–24 alleles per locus and a maximum observed heterozygosity of 0.91. Some of the microsatellite loci also showed amplification in two other plantain species (P. lanceolata and P. maritima). 相似文献

The animal‐mediated pollination and seed‐dispersal mutualisms of Ficus species give them key roles in tropical ecosystems, but may make them vulnerable to habitat fragmentation. The development of highly polymorphic markers is needed to analyse their genetic diversity and investigate the effects of fragmentation on gene flow. Of thirteen microsatellite loci isolated from Ficus insipida, a monoecious species in French Guiana, eleven were polymorphic (two to six alleles each). High levels of variation were found among loci; expected heterozygosities ranged from 0.151 to 0.715. All markers revealed a broad cross‐species affinity when tested in 23 other Ficus species. 相似文献

We describe the isolation and characterization of 14 microsatellite loci in the Chevrier's field mouse from genomic DNA-enriched libraries. The 14 loci were highly polymorphic with numbers of alleles per locus in 24 individuals ranging from three to 13, observed heterozygosity from 0.381 to 0.867 and expected heterozygosity from 0.448 to 0.931. All loci followed Hardy-Weinberg expectations except for CFM3, CFM10 and CFM12 loci. No significant linkage association was found among all these loci. The 14 polymorphic microsatellite loci will be useful in studying phylogeography and population genetics of the Chevrier's field mouse. 相似文献

A total of 10 polymorphic microsatellite loci from Hemibarbus labeo were isolated and characterized using an optimized protocol to construct a microsatellite‐enriched genomic library. The analysis of variability was performed in 24 specimens of mainland China. The mean number of alleles across loci was 3.10 ± 1.10 and the level of expected heterozygosity varied from 0.0417 to 0.7482. Frequencies of null alleles of the 10 loci are not significantly greater than zero. No linkage disequilibrium was detected between loci in either population. Five primer pairs cross‐amplify the microsatellites in other species, indicating transportability of the markers within the family Cyprinidae. 相似文献

The endangered vermilion darter (Etheostoma chermocki) is endemic to the Black Warrior River system in the Mobile Basin in Alabama. Restoration and conservation of this species require an understanding of its population genetic structure, which can be characterized using microsatellite DNA. Nine microsatellite loci were developed; eight loci were polymorphic. Although observed heterozygosity was lower than expected heterozygosity in most polymorphic loci, only one locus showed significant deviation from Hardy–Weinberg equilibrium. These nine markers were tested in an additional 24 species of Etheostoma and appear to have sufficient allelic variation to be useful in studies of population genetic structure. 相似文献

We developed nine polymorphic microsatellite loci for the field vole, Microtus agrestis. The number of alleles ranged from five to 15 and observed heterozygosities ranged from 0.40 to 1.00. We also tested the microsatellite loci for amplification and polymorphism in the congeneric species Microtus arvalis. Five of the nine loci were successfully analysed in this species. The microsatellite markers will be employed in studies of reproductive success and fine‐scale spatial genetic structure. 相似文献

Odonates were the first group of organisms where sperm competition and last male sperm precedence have been identified. With the development of 10 microsatellites for the emperor dragonfly Anax imperator, the function and priority patterns of the multiple sperm storage organs of females can be studied and compared between species in natural populations. In addition, two microsatellite loci developed for the sister species Anax parthenope, are also highly polymorphic in A. imperator. We isolated and characterized microsatellite loci in Viola mirabilis (Violaceae), an endangered species from South Korea. Twenty‐three polymorphic microsatellite loci were developed and tested in Korean, Chinese and Japanese populations. The number of alleles per locus varied from two to eight. The observed and expected heterozygosities within the three populations were 0.000–0.625 and 0.469–0.695, respectively. A total of six loci in the Korean population, one locus in the Chinese population and seven loci in the Japanese population deviated from Hardy–Weinberg equilibrium. We expect that these newly developed microsatellite markers will contribute to understanding the phylogeography and population genetics of V. mirabilis, which will aid in developing conservation strategies for this species. 相似文献

Twelve microsatellite loci were found from genomic DNA‐enriched libraries of the South China tiger (Panthera tigris amoyensis). The number of observed alleles for each locus in 53 individuals ranged from five to 14; the expected and observed heterozygosity was 0.480 to 0.883 and 0.333 to 0.956, respectively; and the mean polymorphic information content (PIC) was 0.729. Lycopodium fordii Bak. (Lycopodiaceae, Pteridophyta) is a gardening plant with a native, fragmentary distribution in Taiwan. In this study,
