Microstomum spiculifer possesses a pair of intracerebral photoreceptors each consisting of a single rhabdomeric sensory cell and two cup or mantle cells. The mantle cells are devoid of pigment. In addition, four so-called ciliary aggregations, presumed to have a light-sensing function, are present. Each ciliary aggregation represents a specialized cell with an internal cavity filled with axonemes of modified cilia. Rhabdomeric photoreceptors consisting of one to three sensory cells and a single pigmented or unpigmented mantle cell are widespread within taxa of the Plathelminthes Rhabditophora. On the contrary, the existence of two mantle cells forming the eye cup is only known for M. spiculifer and a few other species of the Macrostomida. Therefore, at least two hypotheses are possible: (1) two cup cells are a basic characteristic of the Rhabditophora and a reduction from two to one cup cell has occurred secondarily or (2) the stem species of the Rhabditophora possessed rhabdomeric eyes with one cup cell, and two mantle cells have evolved within the Macrostomorpha. The existence of ciliary aggregates has been documented for several taxa of the Plathelminthes Rhabditophora. From their distribution it can not be concluded whether these differentiations are either a basic feature of the Rhabditophora or have evolved several times convergently. The four eyes of the prolecithophoran Allostoma sp. are disposed in two pairs in a dorsolateral position at the periphery of the brain and beneath its capsule. They are rhabdomeric pigment-cup ocelli. Each eye in the anterior pair consists of one pigment cell and one receptor cell; each in the posterior pair is made up of a larger, single pigment cell and two photoreceptor cells. A lens in front of the pigment cell's aperture is formed by electron-dense, refractive, finger-like protrusions which arise from unpigmented cytoplasmic extensions of the pigment-cup margin. Degenerative signs are sometimes visible in the lens.  相似文献

We have tested the hypothesis that developmentally significant cellular subsets are present in the early stages of neural crest ontogenesis. Cultured quail trunk neural crest cells probed with the monoclonal antibodies HNK-1 and R24 exhibited heterogeneous staining patterns. Fluorescence-activated cell sorting was used to isolate the HNK-1+ and HNK-1- cell populations at 2 days in vitro. When these cell populations were cultured, the HNK-1+ sorted cells differentiated into melanocytes, unpigmented cells, and numerous catecholamine-positive (CA+) cells. In contrast, the HNK-1- sorted cells gave rise to melanocytes and unpigmented cells, but few, if any, CA+ cells. When neural crest cells at 2 days in vitro were labeled with R24 and sorted, both the R24+ the R24- sorted cell populations produced numerous CA+ cell, melanocytes, and unpigmented cells. These results provide evidence for the existence of developmental preferences in some subsets of neural crest cells early in embryogenesis.  相似文献

Comparison of actin isoforms in unpigmented goldfish cells (a normal dermal fibroblast-like cell line, and an unpigmented erythrophoroma cell line capable of being induced to undergo melanization) and in normal and neoplastic melanized goldfish cells shows that the melanized phenotype is accompanied by the presence of multiple actin isoforms. In contrast, the unpigmented cells have only beta-actin. The possible significance of this to pigment organelle translocation is discussed.  相似文献

To determine if neural crest cells are pluripotent and establish whether differentiation occurs in the absence of noncrest cells, a cell culture method was devised in which differentiation could be examined in clones derived from single, isolated neural crest cells. Single neural crest cells, which were isolated before the onset of in vivo migration, gave rise to three types of clones: pigmented, unpigmented, and mixed. Pigmented clones consisted of melanocytes only, whereas some unpigmented cells in mixed and unpigmented clones contained catecholamines, identifying them as adrenergic cells. Extracellular matrix derived from quail somite or chick skin fibroblast cultures stimulated adrenergic differentiation and axon formation. These results demonstrate for the first time the existence of pluripotent quail neural crest cells that give rise to at least two progeny, melanocytes and neuronal cells. They also suggest that continuous direct interactions with noncrest cells are not required for the differentiation of these two cell types. However, components of the extracellular matrix derived from noncrest cells may play an important role in expression of the adrenergic phenotype.  相似文献

The phylogenetic position of Orbiniidae within Annelida is unresolved. Conflicting hypotheses place them either in a basal taxon Scolecida, close to Spionida, or in a basal position in Aciculata. Because Aciculata have a specific type of eye, the photoreceptive organs in the orbiniid Scoloplos armiger were investigated to test these phylogenetic hypotheses. Two different types of prostomial photoreceptor‐like sense organs were found in juveniles and one additional in subadults. In juveniles there are four ciliary photoreceptor‐like phaosomes with unbranched cilia and two pigmented eyes. The paired pigmented eyes lie beside the brain above the circumoesophageal connectives. Each consists of one pigmented cell, one unpigmented supportive cell and three everse rhabdomeric sensory cells with vestigial cilia. During development the number of phaosomes increases considerably and numerous unpigmented sense organs appear consisting of one rhabdomeric photoreceptor cell and one supportive cell. The development and morphology of the pigmented eyes of S. armiger suggest that they represent miniaturized eyes of the phyllodocidan type of adult eye rather than persisting larval eyes resulting in small inverse eyes typical of Scolecida. Moreover, the structure of the brain indicates a loss of the palps. Hence, a closer relationship of Orbiniidae to Phyllodocida is indicated. Due to a still extensive lack of ultrastructural data among polychaetes this conclusion cannot be corroborated by considering the structure of the unpigmented ciliary and rhabdomeric photoreceptor‐like sense organs. Larvae of Poecilochaetus serpens, Trochochaeta multisetosum and Polydora ciliata possess almost identical unpigmented, ciliary, presumptive light sensitive organs within the prostomium. The data corroborate hypotheses on the close relationship of Poecilochaetidae, Trochochaetidae and Spionidae and are even congruent with inclusion of Poecilochaetidae and Trochochaetidae within Spionidae. The organs in P. serpens, T. multisetosum and P. ciliata are composed of one monociliary receptor cell, one supportive cell and several associated flask shaped bipolar sensory cells. The receptor cell cilium enters the supportive cell cavity through a thin pore, dilates and then branches into a high number of disordered projections. The associated sensory cells bear one or occasionally two cilia, which run horizontally beneath or within the cuticle. The supportive cell cavity is not sealed by any cell contact from the subcuticular extracellular space. The organs in Magelona mirabilis are composed of a single supportive cell, but several receptor cells. No further sensory cells are associated. Each receptor cell sends one cilium into an own invagination of the supportive cell, and the ciliary branches are highly ordered. The examined organs in P. serpens, T. multisetosum and P. ciliata exhibit a unique organization amongst polychaetes. The organs of M. mirabilis are most probably homologous. A homology to ciliary organs of Protodrilida is conceivable. In the lineage leading to Protodrilida, primary larval organs may have been integrated into the adult body organization by heterochrony.  相似文献

In vitro clonal analysis of quail cardiac neural crest development   总被引:1,自引:0,他引:1  
The developmental potentials of cardiac neural crest cells were investigated by in vitro clonal analysis. Five morphologically distinct types of clones were observed: (1) "pigmented" clones contained melanocytes only; (2) "mixed" clones consisted of pigmented and unpigmented cells; (3) "unpigmented dense" clones consisted of flattened, closely aligned unpigmented cells; (4) "unpigmented loose" clones consisted of a few loosely arranged, flattened cells; and (5) "unpigmented large" clones included a large number of small, stellate cells that were highly proliferative. The binding patterns of antibodies against lineage-specific markers showed that cells in the different clones expressed characteristic phenotypes. The following phenotypes were expressed in addition to pigment cells: smooth muscle cells, connective tissue cells, chondrocytes, and cells in the sensory neuron lineage. Mixed clones expressed all five phenotypes. Unpigmented dense clones contained smooth muscle cells, connective tissue cells, chondrocytes, and sensory neurons. Unpigmented loose clones exclusively consisted of smooth muscle cells, whereas unpigmented large clones contained chondrocytes and sensory neuron precursors. Based on these results, the following conclusions can be drawn: (1) Pigmented and unpigmented loose clones are most likely formed by precursors that are committed to the melanogenic and myogenic cell lineages, respectively. (2) Mixed and unpigmented dense clones are derived from pluripotent cells with the capacity to give rise to four or five phenotypes. (3) Unpigmented large clones originate from progenitor cells that appear to have a partially restricted developmental potential, that is, these cells are capable of generating two phenotypes in clonal cultures. Thus, the data indicate that the early migratory cardiac neural crest is a heterogeneous population of cells, consisting of pluripotent cells, cells with a partially restricted developmental potential, and cells committed to a particular cell lineage.  相似文献

To examine possible changes in cell surface carbohydrates, fluorescent lectins were applied at various times during differentiation of neural crest cells in vitro. The pattern and intensity of binding of several lectins changed as the crest cells developed into melanocytes and adrenergic cells. Considerable amounts of concanavalin A (Con A) and wheat germ agglutinin (WGA) bound to all unpigmented cells throughout the culture period. Melanocytes, however, bound much less of these lectins. Soy bean agglutinin (SBA), unlike Con A and WGA, only bound later in development to unpigmented cells at about the time when catecholamines were detected histochemically. Binding of SBA could be induced in younger cultures by pretreating the cells with neuraminidase. Melanocytes, however, did not bind detectable amounts of SBA even if treated with neuraminidase. The SBA-binding sites were often concentrated on cytoplasmic extensions and on contact points between neighboring cells, even when receptor mobility was restricted by prefixation of the cells or adsorption of lectin at 0 degrees C. All three lectins bound to cell processes resembling nerve fibers in particularly high amounts.  相似文献

Summary The retina of nudibranch eyes contains two types of large cells; pigment cells which comprise about two-thirds of the total, with unpigmented sensory cells making up the remainder. Both pigment and receptor cells carry microvilli on their distal borders, but no traces of cilia were observed among them. The cornea of the eyes of aeolid and dendronotid nudibranchs is composed of a single layer of small cells, unlike the dorids where the cornea is made up of one of more large cells. The latter contain nuclei comparable in size with those of the pigment cells in the retina, but are themselves unpigmented.The elliptical eyes ofAplysia contain three types of retinal cell; the pigment cells and two kinds of receptor cells. The ciliary receptor cells bear equal numbers of cilia (9+2) and microvilli, while the microvillous receptor cells carry long tufts of microvilli with only an occasional cilium among them. The proximal cytoplasm of the receptor cells inAplysia and the nudibranchs contains large quantities of the small spherical vesicles (averaging 660 Å in diameter) which appear to be characteristic of gastropod eyes.  相似文献

The photoreceptors of four polychaete species were investigated by transmission electron microscopy: Eteone longa and Anaitides mucosa (Phyllodocidae), Scolelepis squamata (Spionidae), and Heteromastus filiformis (Capitellidae). Four different types of light-sensitive organs could be distinguished: 1) a simple, unpigmented rhabdomeric type; 2) a simple ocellus composed of a sensory and a pigmented cell; 3) complex eyes with a lens consisting of secretory granules; 4) a simple, unpigmented type with modified cilia. In spite of its simpler organization the fourth type is listed last, because its function as a photoreceptor seems dubious. The first type (unpigmented rhabdomeric receptor) occurs in all four species investigated. It is the only type of photoreceptor in Heteromastus. Additionally, the two phyllodocids Eteone and Anaitides possess another kind of receptor (type 4) in close proximity to the type 1 receptor. Simple ocelli (type 2) are found in Scolelepis. A pair of complex eyes (type 3) is present in both Eteone and Anaitides, but they show important differences in the two species. First, the eyes in Eteone exhibit ciliary rudiments within the sensory processes, but such rudiments are absent in the eyes of Anaitides. Secondly, the sensory cells in Anaitides possess pigment granules, whereas in Eteone they do not. Thirdly, the lens in Eteone is composed of secretion granules of equal electron density, whereas in Anaitides the lens granules show increased electron density centrally. Lens material appears to be secreted from a single corneal cell in Eteone, and from several corneal cells in Anaitides. In both species these corneal cells are located distally outside the lens.  相似文献

Monocelis lineata consists of a complex of sibling species, widespread in the Mediterranean and Atlantic Ocean. Previous genetic analysis placed in evidence at least four sibling species. Nevertheless, this research was not conclusive enough to fully resolve the complex or to infer the phylogeny/phylogeography of the group. We designed specific primers aiming at obtaining partial sequences of the mtDNA gene Cytochrome c Oxidase subunit I (COI) of M. lineata, and have identified 25 different haplotypes in 32 analyzed individuals. The dendrogram generated by Neighbor-Joining analysis confirmed the differentiation between Atlantic and Mediterranean siblings, as well as the occurrence of at least two Mediterranean sibling species. Thus validated, the method here presented appears as a valuable tool in population genetics and biodiversity surveys on the Monocelis lineata complex.  相似文献

Summary The lateral line organ of the spotted shark is characterized by its semi-cylindrical shape. Each organ (neuromast) is so closely apposed to the next that the individual neuromasts are almost continuous. The neuromast is composed of receptor cells, supporting cells and mantle cells. The receptor cells bear one kinocilium and up to 40 stereocilia. Bi-directional arrangement of the receptor cells as occurs in teleosts was demonstrated. Afferent and efferent nerve endings were found at the base of the receptor cells. The supporting cells extend from the basal lamina to the free surface. Long microvilli and a cilium-like ciliary rod project from the top of each supporting cell. The cell contains relatively few elements of the Golgi apparatus and little rough endoplasmic reticulum, but mitochondria and filaments are abundant. The mantle cell limits the lateral margin of the neuromast. It is distinguished from the supporting cell because of its long crescent-shaped nucleus and scarce, short microvilli. Myelinated nerve fibres are found in the subepithelial connective tissue but not in the epithelium.The fine structure of the shark lateral line organ suggests that this organ is in an intermediated step of evolution between that of lamprey and teleost.  相似文献

Photodynamic therapy (PDT) as a regime for melanoma is of limited success due to factors such as the efficacy of the photosensitizer used, penetration depth and the presence of pigment. We characterised a pigmented and an unpigmented melanoma cell line with respect to their phenotypes. Cell viability was assessed after exposure to hypericin, a UVA-activated photosensitizer. Exposure to 3 μM activated hypericin induced a cytoprotective (autophagic) response from both cell lines. However, the pigmented cells accumulated a large amount of glycogen in their cytoplasm. We hypothesise that the treatment induces an initial cytoprotective response through autophagy, but with increased stress results in a different mode of cell death in pigmented melanoma cells from unpigmented cells. These results indicate that hypericin-PDT could be an adjuvant therapy for melanoma.  相似文献

Sunflower broomrape (Orobanche cumana Wallr.) is considered a self-fertilizing species, but there is no indication as to whether it is strictly self-fertilized or that it presents some extent of cross-fertilization. The objective of this research was to measure the rate of cross-fertilization in O. cumana using an unpigmented recessive mutant as a visual marker. A pot and a field experiment in which single unpigmented plants were surrounded by a large number of pigmented plants were conducted. Occurrence of F1 hybrids, readily distinguishable from unpigmented plants in the progenies of unpigmented plants provided a direct measurement of the cross-fertilization rate. Progenies of unpigmented plants contained 21.5 % of F1 hybrids in the pot experiment and 28.8 % in the field experiment. The results revealed that O. cumana is a partially allogamous species, which has great relevance for understanding the genetic structure and dynamics of populations and, ultimately, race evolution in this parasitic plant.  相似文献

 The nephrostome of Enchytraeus albidus exhibits a longitudinal slit-like opening on the dorsal side of a bulbous organ which is mainly composed of three cells, one flame cell situated centrally and endowed with a ciliary flame, and two cells lying superficially, called the mantle cell and the accessory mantle cell. The mantle cell occupies the ventral side of the organ extending on both sides up to the opening to constitute its immediate border on the frontal and lateral sides. Here it forms a kind of crest which is heavily subdivided into many protrusions and infoldings endowed with long cilia which exclusively spread into the coelomic cavity. The accessory mantle cell borders the narrow posterior slit of the opening, forming the roof of the initial canal which is devoid of cilia. From its anterior region a projection arises extending above the opening. The flame cell forms a groove from which the ciliary flame arises which extends into the canal. At its posterior region it replaces the accessory mantle cell displacing it onto the dorsal surface of the organ. It is argued that the mantle cell and the accessory mantle cell have presumbly originated from coelothelial cells. Thus the metanephridium may be a composite organ developing from a nephridioblastic and a coeloblastic source. A discussion of results in other annelid species indicates that metanephridia in the Annelida may have evolved more than once. Accepted: 13 October 1997  相似文献

Mice homozygous for the recessive patchwork (pwk) mutation are characterized by a variegated pigment pattern with a mixture of unpigmented and normally pigmented hairs. The pigmented hair bulbs contain functional melanocytes. By contrast, the unpigmented hair bulbs contain no melanocytes. This lack results from the death of melanoblasts in the hair follicle at the end of embryogenesis. Here, we report that melanoblasts and melanocytes are found in the epidermis of pwk/pwk mice. Furthermore, these epidermal pigment cells are able to colonize new hair follicles after skin wounding. Despite the presence of epidermal pigment cells with a colonization potential, a follicle that had produced an unpigmented hair produces a new unpigmented hair during the successive hair growth cycles. This hair color continuity is also true for the pigmented hair follicles. Thus, in normal conditions, the hair acts as an independent functional unit as regards its pigment cells population.  相似文献

The study was supposed to determine whether leucine-enkephalin (leu-Enk) or delta opioid receptor was presented in mantles and feet of the Octopus Octopus ocellatus. The results showed that they were found in the mantles, wrists, interbrachial membrane, and funnels of the Octopus Octopus ocellatus. The immunoreactivities of leu-Enk and delta opioid receptor were observed in the epithelial cells of dorsal mantle, wrist and interbrachial membrane. Strong one was presented in both the connective tissue which was close to the epithelium of dorsal mantle, wrist and interbrachial membrane and a small amount of connective tissue cells and nerve fibers, while less positive in other parts. A little of immunoreactive material was also observed in the ventral mantle and funnel, for example, weak one was in the epithelial tissue, the adjacent cells and nerve fibers. The different densities of leu-Enk and delta opioid receptor in mantles and feet of the Octopus O. ocellatus may be related to the different functions.  相似文献
