A one-step synthesis of the betulin dipropionate directly from the birch bark without a separate stage of the betulin preparation is described in this paper. Extracts with different content of the betulin dipropionate were shown to form depending on the conditions of acylation of the upper birch bark with propionic acid. The product with the maximum content of the betulin dipropionate was prepared from the starting fraction of 2?C5 mm of the upper birch bark and the fraction of 10?C20 mm that was preliminarily activated with superheated water vapor. The upper bark extract was analyzed by gas chromatography. The structure of betulin dipropionate was confirmed by element analysis, 1H NMR, 13C NMR, and FTIR spectroscopy.  相似文献

通过比较白桦树皮中白桦脂醇的不同的提取方法,获得一种简单、高效的白桦脂醇提取、纯化方法。通过对不同浸提时间比较、超声震荡方法及旋转蒸发仪回收提取3种白桦脂醇提取方案的比较,进一步利用甲醇-氯仿重结晶法、无水乙醇重结晶法及大孔吸附树脂层析法对获得的白桦脂醇初提取物进行纯化。获得了白桦脂醇提取及纯化的最佳方案。用95%乙醇溶液,液固比50∶1(mL∶g)浸泡白桦树皮样品120 h后,用滤纸过滤,收集滤液,将滤液50℃加热回流5 h,减压蒸馏得到米白色固体粉末。接着以70 mL乙酸乙酯为溶剂,将得到的固体加热回流90 min,趁热过滤。滤液使用坩埚50℃浓缩至干。再以无水乙醇为溶剂进行重结晶,无水乙醇∶固体=30 mL∶1 g,-20℃重结晶,重结晶产率可达38.00%,最高纯度达到99.81%。优化后的白桦脂醇提取纯化方法操作简便,产量较高、试剂安全,是一种可用于规模化提取的有效方法。  相似文献

Betulin-containing products were obtained in a yield of about 40% from absolutely dry birch bark. The content of betulin in the products was 74–75% or 85–89% depending on the presence of sodium or potassium hydroxide, respectively. The one-step method of extraction of high-purity betulin (97.7%) is presented. Betulin was identified by physic-chemical methods, i.e., elemental analysis, IR and NMR spectroscopy, and electron scanning microscopy. The thermal characteristics of betulin were also studied. It was shown that betulin exhibits gastroprotective, hepatoprotective, and capillary-strengthening properties.  相似文献

Background

Birch bark has a long lasting history as a traditional medicinal remedy to accelerate wound healing. Recently, the efficacy of birch bark preparations has also been proven clinically. As active principle pentacyclic triterpenes are generally accepted. Here, we report a comprehensive study on the underlying molecular mechanisms of the wound healing properties of a well-defined birch bark preparation named as TE (triterpene extract) as well as the isolated single triterpenes in human primary keratinocytes and porcine ex-vivo wound healing models.

Methodology/Principal Findings

We show positive wound healing effects of TE and betulin in scratch assay experiments with primary human keratinocytes and in a porcine ex-vivo wound healing model (WHM). Mechanistical studies elucidate that TE and betulin transiently upregulate pro-inflammatory cytokines, chemokines and cyclooxygenase-2 on gene and protein level. For COX-2 and IL-6 this increase of mRNA is due to an mRNA stabilizing effect of TE and betulin, a process in which p38 MAPK and HuR are involved. TE promotes keratinocyte migration, putatively by increasing the formation of actin filopodia, lamellipodia and stress fibers. Detailed analyses show that the TE components betulin, lupeol and erythrodiol exert this effect even in nanomolar concentrations. Targeting the actin cytoskeleton is dependent on the activation of Rho GTPases.

Conclusion/Significance

Our results provide insights to understand the molecular mechanism of the clinically proven wound healing effect of birch bark. TE and betulin address the inflammatory phase of wound healing by transient up-regulation of several pro-inflammatory mediators. Further, they enhance migration of keratinocytes, which is essential in the second phase of wound healing. Our results, together with the clinically proven efficacy, identify birch bark as the first medical plant with a high potential to improve wound healing, a field which urgently needs effective remedies.  相似文献

为了阐明黄龙山白桦不同生境、不同径级、不同器官桦木醇与桦木酸含量,系统采集了陕北黄土高原黄龙山林场阳坡、阴坡、林缘、孤立木4种生境下的白桦不同径级植株的树皮、树枝和树叶样品,采用超声波辅助提取法提取样品,用高效液相色谱法测定了不同生境、不同径级白桦各器官桦木醇、桦木酸含量。结果表明:(1)不同生境下白桦各器官桦木醇、桦木酸含量均存在显著差异(P<0.05),白桦树皮中桦木醇与桦木酸含量、白桦树枝中桦木酸含量在4种生境下均表现为阴坡>孤立木>林缘>阳坡,白桦树叶中桦木醇与桦木酸含量及树枝中桦木醇含量均表现为阴坡>林缘>孤立木>阳坡,总体来看阴坡生境下白桦各器官桦木醇与桦木酸含量都是最高的。(2)在白桦各器官中,桦木醇、桦木酸含量均表现为树皮>树枝>树叶。(3)不同生境下白桦各器官桦木醇、桦木酸含量随着胸径的增大均先增大后减小,均在第Ⅱ径级即胸径10.1~20 cm达到最大。研究认为,阴坡生境有利于白桦各器官积累桦木醇、桦木酸;白桦各器官中桦木醇含量较高,桦木酸含量比较低,而以桦木酸为原料的生物制剂已进入临床应用阶段,因此,以桦木醇为原料制取桦木酸是解决桦木酸在天然植物中含量低、提取困难的有效途径;对白桦各器官的采收应注意把握时机,于白桦生长到胸径10.1~20 cm时为最佳采收期。  相似文献

Betulin and oleanolic acids (pentacyclic triterpenoid secondary metabolites) have broad pharmacological activities and can be potentially used for the development of anti-cancer and anti-AIDS drugs. In this study, we detected the accumulation and the distribution characteristics of betulin and oleanolic acid in various organs of white birch at different ages. We also determined the expression of 4 OSC genes (LUS, β-AS, CAS1 and CAS2) involved in the triterpenoid synthesis pathways by real time RT-PCR. The result showed that the 1-year old birch can synthesize betulin and oleanolic acid. In addition, betulin and oleanolic acids were mainly distributed in the bark, while the content in the root skin and leaf was very low. The content of betulin and oleanolic acid in birch varied in different seasons. The content of betulin and oleanolic acid and their corresponding LUS and β-AS gene expression were very low in 1-year old birch. With increasing age of birch, betulin content was increased, while oleanolic acid was decreased. Similar changes were also observed for their corresponding synthesis genes LUS and β-AS. In the leaf of 1-year old plant, the highest expression of CAS1 and CAS2 occurred at end of September, while expression of LUS and the β-AS was low from June to October. In the stem skin,high expression of β-AS and the LUS genes occurred from the end of July to September. In the root, high expression of the β-AS gene was observed at the end of October. These results indicated that triterpenoid gene expression was similar to the triterpene accumulation. Expression of LUS gene and β-AS gene in birch with different ages were corresponding to the betulinic and oleanolic acid accumulation. Expression of CAS1 and CAS2 genes were elevated with increasing age of birch. This study provides molecular mechanisms of triterpenes synthesis in birch plants.  相似文献

Introduction – Plant extracts are usually complex mixtures of various polarity compounds and their study often includes a purification step, such as solid‐phase extraction (SPE), to isolate interest compounds prior analytical investigations. Molecularly imprinted polymers (MIPs) are a new promising type of SPE material which offer tailor‐made selectivity for the extraction of trace active components in complex matrices. Numerous specific cavities that are sterically and chemically complementary of the target molecules, are formed in imprinted polymers. A molecularly imprinted polymer (MIP) was synthesised in order to trap a specific class of triterpene, including betulin and betulinic acid from a methanolic extract of plane bark. Methodology – Imprinted polymers were synthesised by thermal polymerisation of betulin as template, methacrylic acid (MAA) or acrylamide (AA) as functional monomer, ethylene glycol dimethacrylate as crosslinking agent and chloroform as porogen. Afterwards, MAA‐ and AA‐MIPs were compared with their non‐imprinted polymers (NIPs) in order to assess the selectivity vs betulin and its derivatives. Recovered triterpenes were analysed by HPLC during MIP‐SPE protocol. Results – After SPE optimisation, the MAA‐imprinted polymer exhibited highest selectivity and recovery (better than 70%) for betulin and best affinity for its structural analogues. Thus, a selective washing step (chloroform, acetonitrile) removed unwanted matrix compounds (fatty acids) from the SPE cartridge. The elution solvent was methanol. Finally, the MAA‐MIP was applied to fractionate a plane bark methanolic extract containing betulin and betulinic acid. Conclusion – This study demonstrated the possibility of direct extraction of betulin and its structural analogues from plant extracts by MIP technology. Antiviral agents frequently applied for treatment of herpesvirus infections include acyclovir and its derivatives. The antiviral effect of a triterpene extract of birch bark and its major pentacyclic triterpenes, i.e. betulin, lupeol and betulinic acid against acyclovir-sensitive and acyclovir-resistant HSV type 1 strains was examined. The cytotoxic effect of a phytochemically defined birch bark triterpene extract (TE) as well as different pentacyclic triterpenes was analyzed in cell culture, and revealed a moderate cytotoxicity on RC-37 cells. TE, betulin, lupeol and betulinic acid exhibited high levels of antiviral activity against HSV-1 in viral suspension tests with IC50 values ranging between 0.2 and 0.5 μg/ml. Infectivity of acyclovir-sensitive and clinical isolates of acyclovir-resistant HSV-1 strains was significantly reduced by all tested compounds and a direct concentration- and time-dependent antiherpetic activity could be demonstrated. In order to determine the mode of antiviral action, TE and the compounds were added at different times during the viral infection cycle. Addition of these drugs to uninfected cells prior to infection or to herpesvirus-infected cells during intracellular replication had low effect on virus multiplication. Minor virucidal activity of triterpenes was observed, however both TE and tested compounds exhibited high anti-herpetic activity when viruses were pretreated with these drugs prior to infection. Pentacyclic triterpenes inhibit acyclovir-sensitive and acyclovir-resistant clinical isolates of HSV-1 in the early phase of infection.  相似文献

Betulin is a principal component of birch bark and is known to possess a broad range of biological activities, including antiinflammatory, antiviral and anticancer actions. The present study was carried out in vitro to clarify the influence of betulin on melanocortin (MC) receptor-ergic signalling by using COS-7 cells transfected with corresponding human MC receptor DNA. The results showed that betulin binds to the human melanocortin MC1, three to five receptors with selectivity to the MC1 subtype (K(i) value 1.022 +/- 0.115 microM). Betulin binds to the MC receptors with the following potency order-MC > MC3 > MC5 > MC4. Betulin itself does not stimulate cAMP generation, however, it slightly antagonizes alpha-melanocyte-stimulating hormone (alpha-MSH)-induced cAMP accumulation in the mouse melanoma cell line B16-F1. As a water-insoluble substance, betulin was dissolved in DMSO therefore DMSO competition with the labelled ligand NDP-MSH for the binding to the MC receptors was tested in the identical experimental set-up. We found that DMSO competes for binding to all the MC receptor subtypes, at 20% concentration and above. Selectivity for one or another receptor subtype was not observed. We have demonstrated for the first time, the ability of the plant compound betulin to bind to the MC receptors. One may suggest MC receptor MC1 subtype as the essential target for the antimelanoma action of betulin and its structurally close molecules such as betulinic acid. Moreover, we have found a new non-peptide small molecule MC mimetic, that is betulin. Thus, we report a new chemical motif for the binding to the MC receptors that could be used as a template for the search of more selective MC mimetics.  相似文献

The mechanical, physical properties and decay resistances of particleboard made from particles impregnated with Pinus brutia bark extractives were examined. Properties included were modulus of rupture, modulus of elasticity, internal bond, thickness swelling, and weight loss according to European standards. The results showed that particleboards made from particles impregnated with bark extractives had significantly lower mechanical values than those made from unimpregnated particles. Impregnating wood particles with bark extractives improved the decay resistance and thickness swelling of particleboard. Increasing concentration of the extractives decreased the mechanical properties and improved the thickness swelling and decay resistance of the panels. Particleboards made from 1% P. brutia bark extractives met the specifications for modulus of rupture and internal bond strength for general purposes.  相似文献

Abstract

The aim of this study was to investigate the relationship between nitric oxide (NO) and polyamines (PAs) and to determine their roles in betulin accumulation induced by the endophytic fungus Phomopsis in Betula platyphylla. Treatment of birch plantlets with the endophytic fungus Phomopsis promoted an NO burst and accumulation of PAs and betulin. Birch plantlets were treated with the NO-specific scavenger 2-(4-carboxyphenyl)-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide potassium salt (cPTIO) and the PA synthesis inhibitor d-arginine (d-arg). cPTIO and d-arg inhibited the fungus-induced NO burst and accumulation of PAs and betulin. The exogenous NO donor sodium nitroprusside promoted PA production and betulin accumulation, whereas an exogenous PA, putrescine, promoted an NO burst and betulin accumulation. In addition, d-arg inhibited NO production and cPTIO decreased PA production during fungus-induced betulin accumulation. Our results indicate that NO, PAs, PA-induced NO, and NO-induced PAs mediate fungus-induced betulin accumulation in birch plantlets.  相似文献

Fourier transform infrared spectroscopy (FTIR) and gas chromatography mass spectrometry (GC/MS) were conducted to identify a birch bark quiver from Ergonghe Reservoir Cemetery (the Tang Dynasty, A.D. 618–907) in Xinjiang, Northwest China. White substance on the bark surface was identified as gypsum (CaSO4·2H2O) by FTIR and X-ray diffraction analysis (XRD). The gypsum was probably residual white pigment of colored painting on the birch bark quiver. This is the first time that birch bark has been scientifically identified in China and it also provides the earliest evidence of colored painting on a birch bark artifact in ancient China. The minimally destructive methods are informative and promising for the analysis of ancient samples. Birch species are widely distributed in North China and ancient nomadic people put the bark to use in many aspects of their lives, reflecting the considerable importance of birch exploitation in their local life.  相似文献

We studied the variation in bark chemistry between and within 19 European white birch ( Betula pendula ) clones and its implications to resistance to the mountain hare ( Lepus timidus ). We used one-year-old clonal plantlets originating from randomly selected naturally regenerated parental trees. The same clones were used in both chemical analyses and in feeding experiments. The condensed tannins were analysed by an acid butanol assay, other phenolic components by HPLC-DAD, and triterpenoid components by HPLC-MS. The resistance to hare was tested in open-field feeding experiment. The main phenolic compounds in birch bark were catechin derivatives, rhododendrin, platyphylloside, and condensed tannins, and the main triterpenoids were papyriferic acid and pendulic acid. Most of the variation in the concentrations of the studied compounds was found between clones for the studied phenolics and large variation for triterpenoid components were found both between clones and among plantlets within the same clone. Hares clearly selected among the studied clones. Our results suggest that birch bark chemistry play an important role in resistance to herbivory by hare. The total triterpenoids and total flavonoid-aglycones showed significant negative correlation with hare feeding. It seems that a genetic basis for bark chemistry and birch resistance is strong. Such a high variation in secondary chemistry both between clones and within individual clones indicates that European white birch populations have a good resistance towards variable environmental conditions and varying pressures from herbivory.  相似文献

In this article, the group chemical composition of individual parts of the bark of the Betula pendula (Betulaceae) birch was determined at different tree heights. The part of the bark in the examined birch was found to be 14%, including 5.4% of the outer bark and 8.6% of the inner bark. The content of main groups of substances (cellulose, lignin, extractive and mineral substances, free acids, and carbohydrates) was evaluated in the outer and inner bark depending on the stem height.  相似文献

Alzheimer’s disease (AD) is a neurodegenerative disease induced by cholinergic neuron damage or amyloid-beta aggregation in the basal forebrain region and resulting in cognitive disorder. We previously reported on the neuroprotective effects of Betula platyphylla bark (BPB) in an amyloid-beta-induced amnesic mouse model. In this study, we obtained a cognitive-enhancing compound by assessing results using a scopolamine-induced amnesic mouse model. Our results show that oral treatment of mice with BPB and betulin significantly ameliorated scopolamine-induced memory deficits in both passive avoidance and Y-maze tests. In the Morris water maze test, administration of BPB and betulin significantly improved memory and cognitive function indicating the formation of working and reference memories in treated mice. Moreover, betulin significantly increased glutathione content in mouse hippocampus, and the increase was greater than that from betulinic acid treatment. We conclude that BPB and its active component betulin have potential as therapeutic, cognitive enhancer in AD.  相似文献

Samples of the lichen H. physodes were collected from bark of living trees (pine, spruce, birch, alder, rowan, and willow); from the wood of these trees and of juniper; from bark of dead spruce, alder, and rowan trees; and from the moss Hypnum pallescens. Thalli of this lichen were placed onto medium with carboxylmethyl cellulose (CMC) (water being used as a control). Output of sugars was determined using the Nelson-Somogyi technique. Cellulosolytic activity of samples from the bark of pine and birch was higher than that of samples from the bark of spruce. In thalli of the lichen from wood, from moss, and from bark of living alder and rowan trees, the output of sugars on the medium with CMC was similar to that in the control. The cellulosolytic activity was revealed in samples from the lichen from bark of dead rowan and alder trees. In the lichen from spruce bark, the output of sugars on the medium with CMC was higher in samples from dead trees in comparison with living trees. The results are discussed.  相似文献

为了快速、准确的对白桦多个样品进行基因表达分析,本研究应用荧光定量PCR技术,对白桦持家基因微管蛋白基因(Tu)和泛素基因(UbQ)在不同处理的细胞及不同树龄或不同生长季节白桦植株各部位表达的稳定性进行检测。结果表明,对于茉莉酸甲酯(MeJA)、水杨酸(SA)处理的白桦细胞和不同生长季节白桦植株的基因定量表达分析,可同时使用Tu和UbQ平衡化qRT-PCR数据;而针对不同树龄白桦茎皮中基因表达研究时,单独使用UbQ基因作为内参对照即可获得精确的表达数据。  相似文献

对从全国收集的75份山茱萸种质资源,从叶长、叶宽、果实横径、果实纵径、鲜果百粒重、种子千粒重、种子含水量、枣皮灰分、枣皮水分、水溶性浸出物和马钱素含量等表型性状,分析其变异系数和各性状间的相关性.分析结果显示,75份山茱萸种质资源在叶长、叶宽、鲜果百粒重、种子千粒重、枣皮灰分和马钱素含量等性状上的变异系数均高于10%,说明这些性状变化范围较大,个体间性状不稳定,易受环境因素的影响;而果实横径、果实纵径、种子含水量、枣皮水分和水溶性浸出物的变异系数均低于10%,说明这些性状变异较小,表型性状比较稳定.相关性分析结果显示,各性状间都存在一定的相关性,其中果实纵径和果实横径与鲜果百粒重呈显著正相关(0.396和0.312,P <0.01),说明果实纵横径是影响果实重量的关键因素;水溶性浸出物大小与果实横径(0.481,P<0.01)、果实纵径(0.280,P<0.05)、马钱素含量(0.372,P<0.01)呈现显著性正相关,表明水溶性浸出物含量的高低与山茱芰有效成分马钱素含量和果实大小及形状密切正相关,这将为种质的筛选、评价和新品种选育提供依据.  相似文献
