On routine electrophoretic analyses on filter paper and starch gel in an alkaline or neutral medium, no abnormal hemoglobin fractions were found in the blood of 600 newborn infants or their mothers. Trace amounts of hemoglobin Barts were noted in many of the blood samples from newborns when the starch gels (phosphate buffer pH 7.0) were stained with a benzidine/H2O2 reagent. In one infant, precocious cessation of synthesis of hemoglobin F was postulated to account for the small amounts of this hemoglobin found in a cord-blood specimen. Analysis of 15,000 blood samples from adults revealed two instances in which the hemoglobin F level was 20 and 35%, respectively. The former was attributed to a hereditary persistence of hemoglobin F, while the latter was associated with acute leukemia.In an addendum, the finding of an infant with an abnormal hemoglobin variant, resembling in many of its properties hemoglobin F Texas, is reported.  相似文献

将强启动子P43与透明颤菌血红蛋白基因(vgb)通过重叠延伸PCR进行融合,克隆到芽孢杆菌整合表达载体pDG1730中,重组表达载体pDG-P43vgb转化促生防病解淀粉芽孢杆菌FZB42,Wsetern-Blot和CO差光谱分析表明重组菌株FZB42-VHb表达了有活性的VHb蛋白,VHb的表达对重组菌株菌体的生长及抗菌脂肽的产生都有促进作用.在相同培养条件下,重组菌最大菌体密度比原始菌株提高了14.49 %,抗菌脂肽fengycin的产量提高了1.74倍,抗菌脂肽surfactin的产量提高了3.14倍.  相似文献

 为探索一条研制猪血红蛋白 (pHb)为基础的血液代用品新途径 ,开发了干膜超声法将猪血红蛋白和别构效应剂、超氧化物歧化酶等联合包埋于脂质体的技术 .考察了氢化大豆卵磷脂、二肉豆蔻酰磷脂酰胆碱、胆固醇、二硬脂酰磷脂酰乙醇胺 甲氧基聚乙二醇和维生素E等在制备脂质体包埋血红蛋白 (LEH)中的作用 .通过控制磷脂与其他成分的配比 ,制得包埋率为 10 3%,pHb浓度达 16 %的稳定的LEH ;进一步将pHb微囊通过小鼠尾静脉多次注入其体内 ,检测受试小鼠血液中红细胞和白细胞数、抗体滴度、血小板聚集率及肾脏组织学等方面的变化 .小鼠体内试验表明了所制备的LEH具有低免疫原性、对肾脏无明显损害等特点 .脂质体包埋pHb是一种极具开发前景的稳定的人工载氧系统  相似文献

经肌醇四磷酸修饰,血红蛋白能改善氧亲和力。小麦麸皮浸出液经硫酸铵分级沉淀、Sephadex G-50凝胶过滤、Mono Q离子交换柱梯度层析纯化得到植酸酶。用纯化的植酸酶催化植酸(肌醇六磷酸,IP6)水解,可得到一系列肌醇磷酸化合物。控制反应条件使水解终止在肌醇四磷酸(IP4)为主要产物。用国产714树脂填充柱一步纯化得到肌醇四磷酸钠盐,经HPLC反向柱,FAP质谱分析其中不含其他肌醇磷酸化合物。  相似文献

本文介绍一种酸性琼脂电泳方法。它可以比较容易地分开血红蛋白A和血红蛋白F、可将异常血红蛋白分成两大类,即酸性电泳阳性和酸性电泳阴性两类异常血红蛋白。此法在血红蛋白病中比较常用的是鉴别血红蛋白S与其它电泳速度相同的变异物,帮助诊断镰状细胞贫血。在常见病方面,这种方法还能分开血红蛋白A和糖基化血红蛋白,用来帮助诊断糖尿病。  相似文献

Release of hemoglobin into plasma is a physiological phenomenon associated with intravascular hemolysis. In plasma, stable haptoglobin-hemoglobin complexes are formed and these are subsequently delivered to the reticulo-endothelial system by CD163 receptor-mediated endocytosis. Heme arising from the degradation of hemoglobin, myoglobin, and of enzymes with heme prosthetic groups could be delivered in plasma. Albumin, haptoglobin, hemopexin, and high and low density lipoproteins cooperate to trap the plasma heme, thereby ensuring its complete clearance. Then hemopexin releases the heme into hepatic parenchymal cells only after internalization of the hemopexin-heme complex by CD91 receptor-mediated endocytosis. Moreover, alpha1-microglobulin contributes to heme degradation by a still unknown mechanism, with the concomitant formation of heterogeneous yellow-brown kynurenine-derived chromophores which are very tightly bound to amino acid residues close to the rim of the lipocalin pocket. During hemoglobin synthesis, the erythroid alpha-chain hemoglobin-stabilizing protein specifically binds free alpha-hemoglobin subunits limiting the free protein toxicity. Although highly toxic because capable of catalyzing free radical formation, heme is also a major and readily available source of iron for pathogenic organisms. Gram-negative bacteria pick up the heme-bound iron through the secretion of a hemophore that takes up either free heme or heme bound to heme-proteins and transports it to a specific receptor, which, in turn, releases the heme and hence iron into the bacterium. Here, hemoglobin and heme trapping mechanisms are summarized.  相似文献

Various immunological features were measured in 20 patients with epilepsy who had received prolonged treatment with hydantoins. Immunoglobulin A (IgA) was shown to be absent or low in five patients, β1C/A was low in 10 patients. Five patients showed negative reactions to skin tests, and two could not be sensitized to dinitrochlorobenzene. The corresponding features were normal in 14 control patients with epilepsy but without hydantoins. It is suggested that the hydantoins influence humoral immunity, whereas other immunosuppressant agents have been found to affect cellular immunity.  相似文献
