Ultrastructural differences were observed among mitochondria from the corpora allata of healthy, Nosema-infected, and anti-Nosema drug treated honeybees. Mitochondria in the corpora allata of the healthy honeybees were large in size, possessed well defined membrane, numerous mitochondrial granules and less electron-dense matrix. In the corpora allata of Nosema-infected honeybees, mitochondria were small in size and the mitochondrial matrix had a higher electron density. Mitochondria from the corpora allata of Nosema-infected honeybees after treatment with thimerosal, had a lower electron density in the mitochondrial matrix and a larger diameter than those from untreated honeybees. Mitochondria from fumagillin-treated honeybees had the smallest diameter and the highest electron density in the matrix among mitochondria observed in the present study. Image analysis also confirmed the differences in the electron density of the mitochondrial matrix among mitochondria in the corpora allata of four experimental groups of honey bees.  相似文献

Electron-dense secretion granules were numerous in the hypopharyngeal glands of healthy honeybees. In the hyopharyngeal glands of honeybees infected by Nosema apis, these granules appeared to have increased in size and lost their electron density, possessing a core area that consisted of numerous smaller granules, and a slightly electron dense fringe area, which in some cases possessed a crystalline structure. In the hypopharyngeal glands of infected honeybees after treatment with fumagillin, the secretion granules were also large and slightly electron dense, but the granular content was homogenous. Some of these granules were also partially crystallized. These ultrastructural changes in the secretion granules of diseased and fumagillin treated bees, is probably associated with a change in secretory activity of the glands.  相似文献

In addition to glial cells, intrinsic glandular cells and ordinary axons, the corpora cardiaca of Blattella germanica (L.) (Dictyoptera : Blattellidae) contain 4 types of neurosecretory fibers originating from the brain (types 1, 2, 3, and 4), which can be recognized on the basis of the size, form, and electron density of their neurosecretory granules. A comparative ultrastructural study of the corpora cardiaca from normal females and from dopamine-treated (1 μg) females has been carried out at different stages within the first ovarian cycle (freshly emerged: day 0; 2-day-old: pre-vitellogenesis; 4-day-old: beginning of vitellogenesis; 6-day-old: full vitellogenesis; 8-day-old: post-vitellogenesis, period of ootheca transport). Quantitative data on the exocytotic configurations observed in each type of fiber (1–4) have led to the following conclusions: (a) the exocytotic configurations in control specimens are too infrequent to prompt any inference about the dynamics of neurosecretion release in the different types of fibers; (b) dopamine treatment induces a stimulation of the exocytotis phenomenon, whose extent depends on the specimen age and on the type of fiber. Therefore, on days 2 and 6, dopamine: preferentially stimulates neurosecretion release in the fibers of types 3 and 4 respectively.  相似文献

Summary The Falck-Hillarp method reveals yellow and green fluorescent areas in the neurohemal part of the corpora cardiaca. Some neurons and the neuropile of the hypocerebral ganglion appear green. The localization of tritiated serotonin taken up and stored in the corpora cardiaca was studied radioautographically in vivo and in vitro with the use of low concentrations of tritiated serotonin. In the neural part of the corpora cardiaca, fibers with 100 nm dense granules were shown to be indolaminergic. Fibers containing 200 nm dense granules and 250 nm clear vesicles are thought to be catecholaminergic.  相似文献

The ultrastructure of the retrocerebral endocrine-aortal complex of the earwig, Euborellia annulipes has been studied. The space between the inner and outer stromal layers of the aorta is occupied by numerous axon terminals and pre-terminals containing large electron dense granules (NS-I) of approximately 100 to 220 nm and a few axon terminals having small granules (NS-II) of approximately 40 to 90 nm; the former appear to belong to medial neurosecretory A-cells, and the latter to the B-cells of the brain. The corpora cardiaca consist of intrinsic cells with mitochondria and multivesicular bodies. Granules of type NS-II and NS-III are observed in the axon terminals and pre-terminals in the corpora cardiaca. The NS-II are identical to those found in the aorta and are probably the secretions of the lateral B-cells. Granules of type NS-III are 40 to 120 nm and electron dense, and are intrinsic in origin. Similar granules occur in the intrinsic cells of the corpora cardiaca. E M studies have confirmed the rôle of the aorta as a neurohaemal organ for the medial neurosecretory cells, and the corpora cardiaca for the lateral neurosecretory cells of the brain. The corpora cardiaca also act as a reservoir for the intrinsic secretion. The corpus allatum is a solid body consisting of parenchymal cells with prominent nuclei, mitochondria, and endoplasmic reticulum. In between its cells are occasional glial cells and also neurosecretory as well as non-neurosecretory axons. The gland is devoid of A-cell NSM.  相似文献

We have isolated two major 6-kDa peptides from extracts of corpora cardiaca of adult females of Locusta migratoria. These peptides have been characterized by peptide sequencing and liquid secondary-ion mass spectrometry. They are structurally related dimers, one (6278.5 Da) being a homodimer (A-A chains), the other (6280.5 Da) being a heterodimer (A-B chains). A 60% similarity exists between the A and B chains. Both peptides have been chemically synthesized and the synthetic compounds appeared to be identical to the native ones. Polyclonal antibodies raised against each of these peptides demonstrated that they were contained within the secretory granules of the intrinsic cells of the glandular lobes of the corpora cardiaca. The physiological significance of these two peptides is unknown but, using the synthetic peptides, we are currently probing their biological role.  相似文献

The median neurosecretory cells of the protocerebrum of fifth instar larvae and 1- or 2-day-old adults of Oedipoda coerulescens produced more fuchsinophilic material when reared on dark backgrounds than on pale ones. In the latter case, the neurosecretory cell bodies were almost completely devoid of neurosecretion. The same differences were found in the anterior lobes of the corpora cardiaca. Individuals implanted with pars intercerebralis from donors reared on a dark background showed dark pigmentation no matter what their own background colour (and conversely). Black or red pigmentation appeared on absorbent grounds whereas a pale one was produced on reflecting grounds with high levels of incident light predominantly between 576 and 588 mμ. The significance of these results is discussed. A possible scheme for the interaction of environmental factors and pigmentation is suggested.  相似文献

Doppler radar actographs have been used to measure the fall in locomotor activity following feeding in mid-fifth instar nymphs of Locusta migratoria. Filling the crops of the insects with agar via a cannula, reduced activity in a similar manner. Haemolymph from recently-fed donors injected into insects which had been deprived of food, likewise reduced locomotor activity compared with haemolymph from deprived donors. Injections of nutrients were ineffective, but homogenates of the storage lobes of the copora cardiaca from food-deprived donors proved effective compared with similar homogenates from newly-fed donors. It is suggested that neurosecretion released from the corpora cardiaca, known to occur following crop-filling, leads to a reduction in locomotor activity.  相似文献

The Egyptian locust, Anacridium aegyptium, has four protocerebral neurosecretory centres: the A to B neurosecretory cells of the pars intercerebralis (the A cells are rich in fuchsinophil material and the B cells are devoid of fuchsinophil neurosecretion), the voluminous C neurosecretory cells poor in neurosecretion, and the median sub-ocellar neurosecretory cells.From September to the beginning of January, imaginal diapause is characterized by an accumulation of the median neurosecretion in the pars intercerebralis-corpora cardiaca system, by small corpora allata, and, in the female, by a stop in oöcyte development although the male's sexual activity is still not altered. Allatectomy suppresses neither the male's sexual behaviour nor its fecundity. From January, the increase of the photoperiod causes a release of the median neurosecretion in both sexes, an increase of the volume of the corpora allata, and breaks ovarian diapause.In autumn, the implantation of the male's or female's corpora allata of Anacridium does not stimulate ovarian growth of diapausing females. On the contrary, the implantation of corpora allata or of pars intercerebralis or of corpora cardiaca of Locusta migratoria migratorioides (locust without diapause) causes ovarian development of the diapausing females of Anacridium. Thus, in the two sexes of the Egyptian locust, the corpora allata are inactive during the female ovarian diapause. The imaginal diapause of Anacridium affects both sexes (stocking of median neurosecretion, arrest of the corpora allata). If diapause does not seem to affect the male's development, it is because its sexual activity is free from the pars intercerebralis and corpora allata.The corpora allata of Anacridium show a sexual dimorphism in the active adult: they are smaller in the male and have more mitosis in the female. Cephalic nervous connexions were found necessary for oviposition and normal egg maturation in the two species studied. Neurosecretory cells of A type from the brain, suboesophageal, thoracic, and four abdominal ganglia seem to elaborate a stimulating substance for egg laying. This neurosecretion type is not present in corpora cardiaca, corpora allata, the four last abdominal ganglia, or perisympathetic organs which have no clear-cut effect on oviposition. The circadian rhythm of egg laying appears to be entirely controlled by the liberation of this neurosecretion and by an inhibitory factor localized in the head.In Carausius, egg deposition by the valvulae of the ovipositor is regulated by these two factors whereas egg transit through the common oviduct is a distinct preliminary step under nervous control. This nervous effect from the posterior region does not exist in Clitumnus.  相似文献

(Department of Entomology, North Dakota State University of South Carolina, School of Medicine, Columbia, U.S.A.) — Acta zool. (Stockh.) 61(2): 65–77. The corpora cardiaca (CC) and corpora allata (CA) of the pink bollworm, Pectinophora gossypiella, were studied at the ultrastructural level during induction maintenance, and termination of larval diapause. The CC is composed of 5–6 intrinsic secretory (IS) cells, glial cells, and axons that carry electron dense (ED) and electron lucent (EL) granules from the brain. The IS cells produce ED granules with an average diameter of 110 ± 26 nm. During diapause, the axons with ED granules showed large accumulations of neurosecretory granules, but the axons with the EL granules contained lesser amounts. The CA is made up of approximately 25 large cells and axons that pass through the CC from the brain. Most of the axons take up a peripheral position. A characteristic feature of CA cells during diapause was the presence of stacks of convoluted tubules of the smooth endoplasmic reticulum (SER) that may be associated with juvenile hormone synthesis. The IS cells of the CC appeared to be inactive during diapause.  相似文献

Ultrastructural analysis of the corpora cardiaca of the stable fly, Stomoxys calcitrans, and the tsetse fly, Glossina morsitans, revealed the presence of elementary neurosecretory granules (ENG) unique to the intrinsic neurosecretory cells (INC) of these species. In addition to electron‐dense spheres, the INC of the corpus cardiacum of the stable fly contain electron‐dense angular granules, either square or rectangular in shape, while the INC of the tsetse fly contain electron‐dense spindle‐shaped ENG. The distinctive granules of these INC can be traced within nerves to their sites of storage and release, eliminating the need for labeling with artificial probes. Although the INC of the corpus cardiacum of most species have been found to be fuchsinophilic, neither the INC of the stable fly nor the tsetse fly are aldehyde‐fuchsinophilic. These peptigenic cells offer neuroendocrinologists a unique opportunity to study the physiology and biochemistry of neurosecretory cells. J. Morphol. 240:155–168, 1999. Terminal o?cytes containing yolk in both healthy and nosema infected queen honey bees were studied. In the healthy queens the terminal o?cytes exhibited a layer of follicular cells which were covered by a smooth-surfaced ovariole sheath. In the o?plasm were numerous electron-dense yolk granules and lipid yolk droplets. The elecron-dense yolk granules exhibited a crystalline structure. Stacks of endoplasmic reticulum were observed in the yolk granules throughout the o?plasm. Numerous mitochondria possessing well defined cristae were also observed. O?cytes in the ovary of queen honey bees appeared degenerated after 7 days of infection by Nosema apis. The ovariole sheath was wrinkled. In the o?plasm, yolk granules were broken down into small spheres and granular substances. Numerous ribosomes without stacks of endoplasmic reticulate were observed. Lysosomes were abundant and numerous electron-dense materials surrounded by a membrane were detected. The o?cytes appeared to be extensively autolysed. The activity of the substance(s) which are contained in the cephalic endocrine organs of the locust which induce egg diapause in Bombyx mori was examined by implantation and injection of saline extracts of these organs. Extracts from the median and lateral neurosecretory parts of the locust brain were not effective in inducing egg diapause. Extracts of the corpora cardiaca, corpora allata, and suboesophageal ganglion of the locust induced diapause eggs in Bombyx pharate adults from which the suboesophageal ganglion had been removed. The first two extracts could induce egg diapause even in isolated abdomens of pharate adults of Bombyx. In the locust corpora cardiaca, the activity was present only in the glandular lobe and not in the nervous region. This activity decreased when the nervi corporis cardiaci I and II and of nervi corporis allati I were cut. Allatectomy also brought about a decrease in the activity in the glandular lobe which could not be restored by the injection of juvenile hormone. The activity in the corpora allata was enhanced slightly by the disconnection though not significantly.From these results, it is assumed that the corpora cardiaca, corpora allata and suboesophageal ganglion of the locust contain and active principle(s) capable of inducing egg diapause in Bombyx mori. The nervous connections between the brain, corpora cardiaca, and corpora allata are essential for the accumulation of the active substance(s) in the glandular lobes of the corpora cardiaca.  相似文献

Abstract A cluster of five to seven AKH-like immunoreactive cells lie in each lobe of the paired corpora cardiaca of the true armyworm, Pseudaletia unipuncta. These cells form a mesh work of immunoreactive processes within the corpora cardiaca, and immunoreactive tracts projecting posteriorly over the aorta.
Reversed-phase high performance liquid chromatography of extracts of the corpora cardiaca of P.unipuncta revealed a single large U.V. absorbent peak with a retention time identical to synthetic Manduca- AKH. Amino acid analysis of the contents of this peak yielded a composition identical to that of synthetic Manduca-AKH which was analysed in a parallel manner. Furthermore the material within the peak possessed adipokinetic activity when bioassayed in day 2 adult male P. unipuncta. The corpora cardiaca of similar individuals were found to contain approximately 17.6ng (17.6pmol) of Manduca-AKH equivalents per pair.
Injection of Manduca-AKH into 2-day-old adult male P.unipuncta resulted in a dose-dependent elevation in haemolymph lipid levels with a maximum level of 80–90μmg/μl obtained with 5–10 ng of Manduca-AKH. Continuous flight also elevated haemolymph lipid levels in day 4 adult males with a significant elevation evident in the first samples taken after 15 min of flight and lipid levels plateauing at approximately 100 μg/μl by about 60 min of flight.  相似文献

Malpighian tubules of Namib Desert tenebrionid beetles of the genus Onymacris are strongly stimulated by homogenates of the corpora cardiaca. The corpora cardiaca of other arid-adapted tenebrionids also contain diuretic material. Biogenic amines, which could be released during the preparation of corpora cardiaca extracts, do not stimulate fluid secretion in tubules of Onymacris rugatipennis. The diuretic factor in corpora cardiaca extracts is stable to boiling and to incubation with pronase. HPLC separation of the corpora cardiaca of O. rugatipennis gives a single region with diuretic activity in both secretory and electrical bioassays. Diuretic activity can not be detected in the haemolymph of Onymacris, and injection of corpora cardiaca extracts into the beetles does not cause diuresis. Simultaneous injection of corpora cardiaca and the dye amaranth shows that the most of the dye transported by the Malpighian tubules moves anteriorly into the midgut, indicating fluid recycling by this route. The most likely function for this “diuretic hormone” is clearance of metabolic wastes from the haemolymph.  相似文献

The absence of juvenile hormone at the time of head cap slippage during the last-larval moult of the tabacco hornworm, Manduca sexta, causes deposition of premelanin granules into the outer regions of the newly forming endocuticle beginning 13 h later. These granules were found to contain an inactive phenoloxidase which becomes activated about 9 h later, 4 h before body melanization begins. The onset of melanization was not accelerated by melanization and reddish colouration hormone from Bombyx heads, extracts of pharate-adult corpora cardiaca or pharate-larval ventral nerve cords (sources of eclosion hormone), or extracts of pharate-larval suboesophageal ganglia or corpora cardiaca-corpora allata complexes. Instead the fall of the ecdysteroid titre to below 250 ng/ml 20-hydroxyecdysone equivalents appeared to be the cue that allowed melanization about 4.5 h later. Up to, but not after, this time both melanization and ecdysis could be delayed by exogenous 20-hydroxyecdysone in a dose-dependent fashion above 0.1 μg per larva. In vitro studies published elsewhere indicate that 20-hydroxyecdysone prevents the activation of the premelanin granules. Thus the granules can be deposited at the proper time in the newly forming endocuticle but their melanization is regulated by the declining ecdysteroid titre and it thus synchronized with other events occurring just before ecdysis.  相似文献

An attempt has been made to determine the influence of the corpora cardiaca on sustained flight tendency shown by Schistocerca gregaria male imagos reared in dense groups or in isolation, taking into consideration the presence or absence of the corpora allata. Corpora cardiaca either whole or in part (glandular or neurohaemal lobe) from either good or poor flyers were inserted into the abdomen of either good or poor flyers. A small amount of extra corpora cardiaca or neurohaemal lobe from good flyers favours sustained flight. A large amount from good or poor flyers inhibits flight. Allatectomized recipients react like intact animals, but locusts isolated for several generations scarcely react. Glandular lobes are without effect and so are corpora cardiaca taken from good fliers about 15 days after their connexions with the brain have been cut. Organs taken from good fliers just after a long flight are also of little effect. It is concluded that factors are elaborated by the pars intercerebralis, then stored and activated in the neurohaemal part of the corpora cardiaca: these organs alone do not control flight tendency.  相似文献

Abstract.The intracisternal (= ergastoplasmic) granules in the adipokinetic hormone-secreting cells of the glandular lobes of the corpora cardiaca in Locusta migratoria migratorioides represent accumulations of adipokinetic prohormones within cisternae of the rough endoplasmic reticulum. Solitary locusts have more and larger intracisternal granules than gregarious locusts. This coincides with the general locomotor activity and thus the energy metabolism in solitary locusts being quite different from that of gregarious locusts, which apparently has consequences for the amounts of adipokinetic hormones synthesized and/or released and, consequently, for the storage of these hormones in the intracisternal granules. These granules apparently function as supplementary stores of secretory material.  相似文献

Summary In the corpora cardiaca of the insect Leucophaea the administration of serotonin elicits ultrastructural features indicative of the extrusion of neurosecretory material by exocytosis. The response to the stimulus and the process of extrusion seem to occur at considerable speed. Nearly all of the 30 test animals, fixed at various intervals starting as early as 3 min after the injection of the drug, show granules captured at the moment of leaving the axon as well as fully exteriorized secretory material. The fact that many of these granules are much smaller than the typical neurosecretory type speaks for intracellular fragmentation of the latter prior to the discharge of this cellular product. After 25 min or more the extruded electron dense structures show signs of breakdown. The apparent speed of these phenomena accounts for the dearth of omega-type configurations observed in unstimulated specimens of this species. The possible relationship between the membrane phenomena involved in exocytosis and the transient protrusions of bounding membranes of neurosecretory granules described in earlier papers remains to be clarified.Supported by N.S.F. research grant BMS 74-12456  相似文献
