好氧反硝化菌的筛选及其脱氮除磷性质的研究   总被引:2,自引:0,他引:2  
利用富集培养基, 从用生活污水驯化后的活性污泥中筛选得到一株具有好氧反硝化兼具除磷功能的细菌。通过形态学及生理生化指标鉴定其为假单胞菌属。利用此好氧反硝化菌处理模拟废水及生活废水, 通过监测总氮、无机磷及CODcr变化确定在C/N摩尔比为3:1、接种量为10%、pH 6.8、30°C条件下处理2 d, 该菌株脱氮、除磷及去除有机物的效果最佳, 活性污泥经此好氧反硝化菌强化后, 对生活废水的处理能力得到明显提升。  相似文献

Two laboratory-scale sequencing batch reactors (SBRs) were operated for enhanced biological phosphorus removal (EBPR) in alternating anaerobic-aerobic or alternating anaerobic-anoxic modes, respectively. Polyphosphate-accumulating organisms (PAOs) were enriched in the anaerobic-aerobic SBR and denitrifying PAOs (DPAOs) were enriched in the anaerobic-aerobic SBR. Fluorescence in situ hybridization (FISH) demonstrated that the well-known PAO, "Candidatus Accumulibacter phosphatis" was abundant in both SBRs, and post-FISH chemical staining with 4,6-diamidino-2-phenylindol (DAPI) confirmed that they accumulated polyphosphate. When the anaerobic-anoxic SBR enriched for DPAOs was converted to anaerobic-aerobic operation, aerobic uptake of phosphorus by the resident microbial community occurred immediately. However, when the anaerobic-aerobic SBR enriched for PAOs was exposed to one cycle with anoxic rather than aerobic conditions, a 5-h lag period elapsed before phosphorus uptake proceeded. This anoxic phosphorus-uptake lag phase was not observed in the subsequent anaerobic-aerobic cycle. These results demonstrate that the PAOs that dominated the anaerobic-aerobic SBR biomass were the same organisms as the DPAOs enriched under anaerobic-anoxic conditions.  相似文献

It has been speculated that the microbial P pool is highly variable in the uppermost layer of various aquatic sediments, especially when an excessive P accumulation in form of polyphosphate (Poly‐P) occurs. Poly‐P storage is a universal feature of many different organisms and has been technically optimised in wastewater treatment plants (WWTP) with enhanced biological phosphorus removal (EBPR). In the recent past, new insights into mechanisms of P elimination in WWTP almost exclusively depended on the development and application of novel methods like 31P‐NMR spectroscopy and molecular methods for identifying Poly‐P accumulating microorganisms (PAO). The aim of the present review is to compile current methods potentially available for detection and quantification of Poly‐P in sediments and to complement it with yet unpublished results to validate their application in natural sediments. The most powerful tool for reliable Poly‐P quantification in sediments is the liquid 31P‐NMR technique which has been successfully used for Poly‐P measurements in a variety of aquatic sediments. But the microorganisms as well as mechanisms involved in Poly‐P storage and cycling are largely unknown. Therefore, we also intend to stimulate future studies focusing on these encouraging topics in sediment research via the implementation of novel methods. (© 2008 WILEY‐VCH Verlag GmbH & Co. A sequencing batch membrane biofilm reactor (SBMBfR) was developed for simultaneous carbon, nitrogen, and phosphorus removal from wastewater. This reactor was composed of two functional parts: (1) a gas-permeable membrane on which a nitrifying biofilm formed and (2) a bulk solution in which bacteria, mainly denitrifying polyphosphate-accumulating organisms (DNPAOs), were suspended. The reactor was operated sequentially under anaerobic condition and then under membrane aeration condition in one cycle. During the anaerobic period, organic carbon was consumed by DNPAOs; this was accompanied by phosphate release. During the subsequent membrane aeration period, nitrifying bacteria utilized oxygen supplied directly to them from the inside of the membrane. Consequently, the nitrite and nitrate products diffused into the bulk solution, where they were used by DNPAOs as electron acceptors for phosphate uptake. In a long-term sequencing batch operation, the mean removal efficiencies of total organic carbon (TOC), total nitrogen (T-N), and total phosphorus (T-P) under steady-state condition were 99%, 96%, and 90%, respectively. In addition, fluorescence in situ hybridization (FISH) clearly demonstrated the difference in bacterial community structure between the membrane biofilm and the suspended sludge: ammonia-oxidizing bacteria belonging to the Nitrosomonas group were dominant in the region adjacent to the membrane throughout the operation, and the occupation ratio of the well-known polyphosphate-accumulating organism (PAO) Candidatus "Accumulibacter phosphates" in the suspended sludge gradually increased to a maximum of 37%.  相似文献

Abstract

A cadmium tolerant strain Cupriavidus sp. H29 could be applied on simultaneous removal of nitrate, phosphorus and cadmium. Response surface methodology (RSM) experiments showed that optimal removal ratios of nitrate, phosphate and Cd(II), which reached 98.89%, 75.23% and 85.01%, occurred at Cd(II) initial concentration of 30.00?mg/L, nitrate initial concentration of 55.20?mg/L, phosphate initial concentration of 50.00?mg/L, initial pH of 7.0 and C/N ratio of 6.0. Studies on gaseous product, precipitations and extracellular polymeric substances (EPS) indicated that the removal of Cd(II) occurred in the extracellular place. Through the coordinated complexation of EPS, strain H29 can achieve the bio-induced phosphate-cadmium removal. Moreover, studies on heated cells, resting cells, permeable cells, cells membrane and cytoplasm demonstrated that the removal of Cd(II) mainly taken place on the cells membrane. This study provided the theoretical basis for the subsequent research of synchronous removal of heavy metals and other pollutants.  相似文献

Propionate, a carbon substrate abundant in many prefermenters, has been shown in several previous studies to be a more favorable substrate than acetate for enhanced biological phosphorus removal (EBPR). The anaerobic metabolism of propionate by polyphosphate accumulating organisms (PAOs) is studied in this paper. A metabolic model is proposed to characterize the anaerobic biochemical transformations of propionate uptake by PAOs. The model is demonstrated to predict very well the experimental data from a PAO culture enriched in a laboratory-scale reactor with propionate as the sole carbon source. Quantitative fluorescence in-situ hybridization (FISH) analysis shows that Candidatus Accumulibacter phosphatis, the only identified PAO to date, constitute 63% of the bacterial population in this culture. Unlike the anaerobic metabolism of acetate by PAOs, which induces mainly poly-beta-hydroxybutyrate (PHB) production, the major fractions of poly-beta-hydroxyalkanoate (PHA) produced with propionate as the carbon source are poly-beta-hydroxyvalerate (PHV) and poly-beta-hydroxy-2-methylvalerate (PH2MV). PHA formation correlates very well with a selective (or nonrandom) condensation of acetyl-CoA and propionyl-CoA molecules. The maximum specific propionate uptake rate by PAOs found in this study is 0.18 C-mol/C-mol-biomass . h, which is very similar to the maximum specific acetate uptake rate reported in literature. The energy required for transporting 1 carbon-mole of propionate across the PAO cell membrane is also determined to be similar to the transportation of 1 carbon-mole of acetate. Furthermore, the experimental results suggest that PAOs possess a similar preference toward acetate and propionate uptake on a carbon-mole basis.  相似文献

Simultaneous nitrification and denitrification (SND) via the nitrite pathway and anaerobic-anoxic-enhanced biological phosphorus removal (EBPR) are two processes that can significantly reduce the energy and COD demand for nitrogen and phosphorus removal. The combination of these two processes has the potential of achieving simultaneous nitrogen and phosphorus removal with a minimal requirement for COD. A lab-scale sequencing batch reactor (SBR) was operated in alternating anaerobic-aerobic mode with a low dissolved oxygen (DO) concentration (0.5 mg/L) during the aerobic period, and was demonstrated to accomplish nitrification, denitrification, and phosphorus removal. Under anaerobic conditions, COD was taken up and converted to polyhydroxyalkanoates (PHAs), accompanied by phosphorus release. In the subsequent aerobic stage, PHA was oxidized and phosphorus was taken up to <0.5 mg/L by the end of the cycle. Ammonia was also oxidized during the aerobic period, but without accumulation of nitrite or nitrate in the system, indicating the occurrence of simultaneous nitrification and denitrification. However, off-gas analysis showed that the final denitrification product was mainly nitrous oxide (N(2)O), not N(2). Further experimental results demonstrated that nitrogen removal was via nitrite, not nitrate. These experiments also showed that denitrifying glycogen-accumulating organisms (DGAOs), rather than denitrifying polyphosphate-accumulating organisms (DPAOs), were responsible for the denitrification activity.  相似文献

摘要:【目的】调查我国部分昆虫螺原体的存在情况,收集我国的昆虫螺原体资源,并研究它们的基本生物学特性,初步确定其分类地位。【方法】螺原体分离、培养方法,应用暗视野显微镜和透射电子显微镜观察螺原体形态,根据16S rDNA构建系统发育树研究螺原体分离菌株可能的分类地位。【结果】从黄道蚜蝇昆虫体内分离到螺原体YY0801,并对其进行了形态学、基本生物学及分子生物学特性研究。分离菌株在R2液体培养基中生长良好,能通过孔径为0.22 μm、0.45 μm的微孔滤膜;在R2固体培养基上呈颗粒状菌落;在对数期呈典型的螺旋状;能利用葡萄糖、D-果糖作为碳源;能强烈代谢精氨酸;不能利用尿素,在含氨苄青霉素钠(2000 U/mL)的R2液体培养基中生长良好。根据16S rDNA构建的系统发育树显示,分离菌株YY0801与血清组Ⅰ的Spiroplasma melliferum 聚类较近。【结论】首次在国内从食蚜蝇科中的黄道蚜蝇(Phytomia zonata)分离到螺原体,分离菌株YY0801可能是Spiroplasma melliferum,但其确切的分类地位需要进行血清学进一步分析。  相似文献

从污泥中筛选出1株对聚乳酸(poly—L—lacticacid,PLA)具有降解活力的细菌DSL09,该菌株对PLA的乳化液、粉末及薄膜都具有降解作用。通过形态学、16SrDNA比对及生理生化特性的分析,鉴定该菌株属于芽胞杆菌属(Bacillus sp.)。为提高该菌株对PLA的降解活力,对其进行了紫外诱变,获得了稳定遗传的突变株DSL09-60b,该突变株的PLA降解活性提高至原始菌株的1.5倍。对该突变株产PLA降解酶的发酵条件进行了优化,经测定DSL09-60b在初始培养基pH为8.0、0.5%酪蛋白为诱导物、接种量6%(体积比)的条件下37℃培养54h时发酵液酶活性最高。  相似文献

从南极普利兹湾深海沉积物中筛选到一株嗜冷杆菌7195。其16SrDNA序列分析表明谊菌株属于嗜冷杆菌属(Psychrobacter),从该菌的全基因组DNA中克隆到编码丙酮酸脱氢酶系E1(PDHcEI)的完整ORF,全长为2817bp,使用DNAMAN5.1对其全长ORF的PDHcEI基因进行分析,PDHcE1基因编码一个由939AA残基组成、分子量预计为100663Da的PDHcEI蛋白质,与Psychrobacter sp.273—4的PDHcE1有78.53%的相似性。  相似文献

In this study, we report a bacterium, Achromobacter sp. TY3-4, capable of concurrently removing Mn (II) and Cr (VI) under oxic condition. TY3-4 reduced as much as 2.31?mM of Cr (VI) to Cr (III) in 70?h, and oxidized as much as 20?mM of Mn(II) to Mn oxides in 80?h. When 0.58?mM Cr (VI) and 10?mM Mn(II) were present together, both Cr(VI) and Mn(II) were completely removed by TY3-4 and the generated precipitates are MnIIIOOH, MnIII,IV3O4, MnIVO2 and CrIII(OH)3. Experiments also show that both biosroption and bioreduction of Mn(II) are the driving forces for Mn(II) removal, whereas bioreduction of Cr(VI) is the driving force for Cr(VI) removal. On the basis of these results, a possible reaction was proposed that TY3-4 concurrently reduces Cr(VI) and oxidizes Mn(II). This study is fundamental for Mn and Cr cycles. 从土壤中筛选到一株产顺式环氧琥珀酸水解酶(ESH)的菌株,经生理生化、Biolog碳源利用试验和16S rDNA序列分析系统发育研究,菌株可能为赤红球菌(Rhodococcus ruber) M1。摇瓶试验确定了最佳碳源、氮源、顺式环氧琥珀酸二钠添加时间和添加量。正交优化试验的最佳培养基和培养时间为:葡萄糖12%,硫酸铵06%,酵母膏05%;顺式环氧琥珀酸二钠投加时间为30h,投加量为358%;菌体培养时间70h。摇瓶试验ESH酶活达750U/g湿细胞。目前该菌株已经应用于固定化细胞连续生产L(+)酒石酸。  相似文献

A laboratory-scale anaerobic–anoxic/nitrification sequencing batch reactor (A2N-SBR) fed with domestic wastewater was operated to examine the effect of varying ratios of influent COD/P, COD/TN and TN/P on the nutrient removal. With the increased COD/P, the phosphorus removals exhibited an upward trend. The influent TN/P ratios had a positive linear correlation with the phosphorus removal efficiencies, mainly because nitrates act as electron acceptors for the phosphorus uptake in the A2N-SBR. Moreover, it was found that lower COD/TN ratio, e.g. 3.5, did not significantly weaken the phosphorus removal, though the nitrogen removal first decreased greatly. The optimal phosphorus and nitrogen removals of 94% and 91%, respectively were achieved with influent COD/P and COD/TN ratios of 19.9 and 9.9, respectively. Additionally, a real-time control strategy for A2N-SBR can be undertaken based on some characteristic points of pH, redox potential (ORP) and dissolved oxygen (DO) profiles in order to obtain the optimum hydraulic retention time (HRT) and improve the operating reliability.  相似文献

A gene library of poly (vinyl alcohol) (PVA)-degrading Pseudomonas sp. strain VM15C was constructed in Escherichia coli with the vector pUC18. Screening of this library with a chromogenic PVA dehydrogenase assay resulted in the isolation of a clone that carries the gene (pdh) for the PVA dehydrogenase, and the entire nucleotide sequence of its structural gene was determined. The gene encodes a protein of 639 amino acid residues (68,045 Da) and in the deduced amino acid sequence, some putative functional sites, a signal sequence, a heme c-binding site, and a PQQ-binding site, were detected. The amino acid sequence showed low similarity to other types of quinoprotein dehydrogenases. PVA dehydrogenase expressed in E. coli clones required PQQ. Ca2+, and Mg2+ stimulated the activity. PVA-dependent heme c reduction occurred with exogenous PQQ in cell extracts of the E. coli clone. The PVA dehydrogenase in the E. coli clone was localized in the cytoplasm.  相似文献

The chemical composition and antioxidant properties of the essential oil and EtOH extract of immortelle (Helichrysum italicum (Roth) G.Don subsp. italicum, Asteraceae) collected in Montenegro were evaluated. The essential oil was characterized by GC/MS analysis, and the content of total phenolics and flavonoids in the EtOH extract was determined using the Folin? Ciocalteu reagent. The free‐radical‐scavenging capacity (RSC) of both the essential oil and the EtOH extract was assessed with the 2,2‐diphenyl‐1‐pycrylhydrazyl (DPPH) method. Moreover, the inhibition of hydroxyl radical (.OH) generation by the EtOH extract of immortelle was evaluated for the first time here. Neryl acetate (28.2%) and γ‐curcumene (18.8%) were the main compounds in the essential oil, followed by neryl propionate (9.1%) and ar‐curcumene (8.3%). The chemical composition of the oils of the examined and additional 16 selected Helichrysum italicum taxa described in literature were compared using principal component (PCA) and cluster (CA) analyses. The results of the statistical analyses implied the occurrence of at least four different main and three subchemotypes of essential oils. Considering the antioxidant properties, the EtOH extract of immortelle exhibited similar potential as propyl gallate and quercetin, while the essential oil exhibited relatively weak DPPH.‐scavenging capacity.  相似文献

以茶树(Camellia sinensis)萌动芽为材料,根据茶树萌动芽芽抑制消减杂交文库中分离得到的肌动蛋白(actin)基因的5′-片段设计引物,利用3′-RACE技术克隆了其cDNA全长序列,该基因cDNA全长1 470 bp,命名为CsActin1(GenBank登录号HQ235647)。序列分析表明,CsActin1开放阅读框长1 134 bp,编码377个氨基酸,5′非编码区100 bp,3′非编码区236 bp。推测的蛋白质分子量为41.70 kD,等电点约为5.31,具有肌动蛋白家族的特征信号序列(YVGDEAQs.KRG和WIAKaEYDE)和肌动蛋白相关蛋白的特征信号序列(LLTEApLNPkaNR)。CsActin1与GenBank中注册的其它植物肌动蛋白核苷酸序列的相似性在80%以上,氨基酸序列相似性在95%以上。与其它植物肌动蛋白的进化树分析结果表明,茶树肌动蛋白与杨树的两个肌动蛋白间的亲缘关系最为密切。并对推导的蛋白结构进行了分析。  相似文献

从两份西藏青稞材料中分离克隆出4个B组醇溶蛋白基因(BH1—BH4),DNA测序结果表明:它们均包含完整的开放阅读框。推断的氨基酸序列与先前报道的大麦B组醇溶蛋白具有相同的蛋白质基本结构。系统分析表明:它们推断的氨基酸序列与栽培大麦中的B组醇溶蛋白具有较高的相关性,与野生大麦和山羊草属的醇溶谷蛋白相似性较低。并且,在4个基因BH1—BH4中,BH1与先前报道的B组醇溶蛋白基因有较低的序列相似性,因此我们对BH1基因进行了原核表达,含该基因的表达载体在大肠杆菌中表达出相对分子质量为28.15kDa并以包涵体形式存在的蛋白,进一步对其在青稞谷粒品质改良中的潜在价值进行了探讨。  相似文献
