来自恶臭假单胞菌的腈水解酶具有高效催化3-氰基吡啶产烟酸的能力,对表达该酶的基因psn进行发酵和产酶条件优化,通过对C源、N源、磷酸盐、金属离子、温度、诱导剂浓度和诱导时间进行单因素考察,获得最适培养基条件(g/L):葡萄糖5、蛋白胨15、酵母粉5、(NH4)2SO45、K2HPO424.5、KH2PO45.76、MgSO40.48;最佳诱导条件:培养2.5 h后添加IPTG诱导,浓度0.2 mmol/L,诱导温度30℃。在该条件下培养,重组大肠杆菌的腈水解酶比酶活可达到45.67 U/mL,比优化前提高了2.26倍。在此基础上,于5 L发酵罐上进行C、N源的补料研究,获得最适分批补料策略,发现其腈水解酶活力可达到75.40 U/mL,是优化前的3.74倍。  相似文献

为从基因水平上改造腈水合酶,进行了诺卡氏菌腈水合酶基因的外源表达研究。在重组大肠杆菌表达系统内,腈水合酶的α亚基几乎不能正常表达,在重组E. coli BL21(DE3) (pET32aNHBAX)中,腈水合酶活性仅为0.04U/mg。构建重组毕赤酵母表达质粒pPIC3.5kNHBAX,采用电穿孔转化法将其转入宿主菌P. pastoris GS115中,经过菌株培养和腈水合酶的诱导表达,筛选获得了优选菌株P. pastoris NH4。对P. pastoris NH4的细胞培养和腈水合酶的诱导表达条件进行优化,结果表明,重组腈水合酶在毕赤酵母中的表达水平可以达到0.52U/mg,但不能稳定积累。  相似文献

通过三步亚克隆 ,将单点突变葡萄糖异构酶 ( GIG1 38P)基因及其调控序列插入链霉菌质粒p IJ40 83,构建重组表达质粒 p IJ40 83- GI1 .用重组质粒转化变铅青链霉菌 TK54原生质体 ,经硫链丝菌素抗性 ( Th R)筛选 ,获得重组菌株 TK54/p IJ40 83- GI1 .酶活力测定和 SDS- PAGE分析表明 ,GIG1 38P基因在变铅青链霉菌中得到高效表达 ,GI1粗酶液比活力为 1 5U/mg,GI1表达量约占菌体可溶性蛋白的 2 5% .同时也研究了重组质粒的遗传稳定性 .重组菌株在无选择压力条件下经液体连续传代培养 ,GI1比活力和 GI1表达量在 2 0 0 h传代时间中呈平缓下降趋势  相似文献

以Lactobacillus casei染色体基因组为模板,PCR扩增获得磷脂酶A2基因pla2,以pET-28a(+)为载体构建重组表达质粒pET-28a(+)-pla2。通过IPTG诱导实现磷脂酶A2在E.coli DE3中的重组表达。对诱导条件初步优化后,重组菌酶活最大可达2.8 U/mL。通过Ni-螯合柱对目的蛋白进行纯化,SDS-PAGE分析重组磷脂酶A2相对分子质量为1.7×104。通过酶学性质分析,最适温度为37℃,最适pH 8,比酶活为110 U/mg。  相似文献

肠膜明串珠菌蔗糖磷酸化酶基因在大肠杆菌中的表达   总被引:1,自引:0,他引:1  
以肠膜明串珠菌基因组DNA为模板,通过PCR扩增得到1 581 bp的蔗糖磷酸化酶(SPase)DNA片段.将该基因克隆到表达载体pET-22b(+)上,构建获得重组质粒pET-SPase.测序结果与GenBank上已公布的基因序列比较,有1个碱基发生变化,但该碱基的改变未引起氨基酸序列的改变.将pET-SPase转化到Escherichia coli Rosetta(DE3)感受态中,IPTG诱导表达后进行SDS-PAGE分析,目的蛋白条带约为55 kD,与预期大小一致,结果表明SPase基因在大肠杆菌中进行了表达.酶活分析,产物的比活为1.8 U/mg,证明了表达产物具有预期的酶活性.进一步考察了IPTG浓度、诱导温度和时间等因素对重组菌表达的蔗糖磷酸化酶的影响.在优化条件下,该蔗糖磷酸化酶的比活可以达到16.6 U/mg,比优化表达条件前的酶比活提高了9.2倍,比已报道的肠膜明串殊菌粗酶液比活(7.1 U/mg)提高了2.34倍.  相似文献

PCR扩增葡萄糖脱氢酶基因(glucose dehydrogenase, gdh),连接到测序载体pUC19,转化大肠杆菌JM109,测序(GenBank登录号:EF626962)后,亚克隆到表达载体,转化大肠杆菌M15,筛选得到GDH高产工程菌M15/pQE31-gdh8.工程菌经IPTG诱导表达,超声波破碎,粗酶液比活力高达15 U/mg.镍凝胶柱亲和层析纯化表达蛋白,超滤、冻干后,比活力达360 U/mg.重组质粒pQE31-gdh8在E .coli M15中稳定程度高达99.8%.工程菌M15/pQE31-gdh8诱导表达的重组酶活力高,易纯化,重组质粒稳定程度高,具有良好的工业应用前景.  相似文献

PCR扩增假单胞菌WBC-3的甲基对硫磷水解酶基因,插入表面展示质粒pYD1的多克隆位点,构建pYD1-MPH重组质粒。重组质粒转化酿酒酵母EBY100,2%半乳糖诱导甲基对硫磷水解酶表达,并利用免疫荧光检测甲基对硫磷水解酶在酿酒酵母细胞表面的表达展示。研究了表面展示甲基对硫磷水解酶的酶学性质和酵母工程菌对水体中甲基对硫磷的降解效果。结果表明成功构建具有全细胞甲基对硫磷水解酶催化活性的酵母工程菌,经2%半乳糖诱导48 h,表面展示甲基对硫磷水解酶比酶活力为18.2 U/mg细胞干重。表面展示甲基对硫磷水解酶的最适作用pH为9.5,最适作用温度为30℃,在p H4.0-10.5之间和45℃以下稳定性较好,Mn2+、Co2+、Zn2+、Ca2+、Hg2+、K+、Ni2+对表面展示甲基对硫磷水解酶活性有激活作用,Na+、Fe3+、Ag+对展示酶活力有抑制作用。工程菌在1 h内对淡水中20 mg/L的甲基对硫磷的降解率在80%以上。  相似文献

以短小芽孢杆菌HZbp总DNA为模板以PCR的方式获得512 bp的脂肪酶基因,并在该基因的两端引入了EcoR1和Sal1的酶切位点,将该基因与大肠杆菌表达质粒pSE380连接,获得重组质粒pSE380-BPL。重组质粒转入大肠杆菌表达细胞株BL21,获得工程菌株BL21-BPL。序列分析显示所克隆的基因具有脂肪酶的保守G-X-S-X-G序列,SDS-PAGE电泳显示该脂脂肪酶的分子质量约为20 kDa。在LB培养基中,IPTG诱导浓度为1.0 mmol/L,33℃诱导培养10 h后,发酵液酶活达到8 U/mL。  相似文献

以丁酸梭菌(Clostridium butyricum)基因组DNA为模板,利用PCR技术扩增得到1,3-丙二醇氧化还原酶基因dhaT,将它连接到pMD18一T载体上,得到重组质粒pMD—dhaT,对此重组质粒进行序列测定,对其DNA序列分析表明,dhaT基因全长为1 158bp。将dhaT基因插入表达载体pSE-380中,构建成重组子pSE—dhaT,并在大肠杆菌JMl09中进行诱导表达。研究表明,以1,3-丙二醇为底物时,基因工程菌在37℃下,以1.0mmol/L IPTG诱导14h,酶活力达到16.28U/mL,比原始菌株提高5、6倍。  相似文献

在甘油厌氧发酵生产1,3-丙二醇的过程中,需要消耗还原当量NADH,NADH的有效供给决定了1,3-丙二醇的产量和得率。采用PCR方法从Candidaboidinii基因组中克隆编码甲酸脱氢酶基因fdh,将fdh基因片段插入载体pMALTM-p2X中,构建表达载体pMALTM-p2X-fdh,并转入1,3-丙二醇生产菌Klebsiella pneumoniae YMU2,获得重组菌Klebsiella pneumoniae F-1。研究了重组质粒的稳定性和IPTG诱导fdh基因过量表达的条件。结果表明,重组质粒具有良好的稳定性;fdh基因表达的蛋白分子量为40.2kDa;IPTG诱导表达研究表明,在IPTG浓度为0.5mmol/L时,诱导4h后甲酸脱氢酶表达明显;发酵过程中甲酸脱氢酶比酶活达到5.47U/mg;与出发菌株K.pneumoniae YMU2相比,重组菌F-1合成1,3-丙二醇的浓度提高了12.5%。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
