Summary Six strains of fungi: two Aspergilli of the A. glaucus group, Penicillium sp. Trichoderma viride, and two unidentified dark fungi, were found able to use thiourea as a source of nitrogen. One of the Aspergilli and the Penicillium showed the best utilization, but the growth was always slow, and no more than 50% (usually much less) of the thiourea-nitrogen was recovered in the mycelium.Assimilation of thiourea-nitrogen by Aspergillus and Penicillium was accompanied by oxidation of the sulphur to sulphate in roughly approximate stoichiometric proportion. The same fungi were able to produce sulphate from cystine as combined source of nitrogen and sulphur.  相似文献

This article reviews some of the aspects of single cell oil (SCO) production using solid-state fermentation (SSF) by fungi of the genus Mortierella. This article provides an overview of the advantages of SSF for SCO formation by the aforementioned fungus and demonstrates that the content of the polyunsaturated fatty acids (PUFA) depend on the type of fermentation media and culture conditions. Process variables that influence lipid accumulation by Mortierella spp. and the profile of the fatty acids are discussed, including incubation temperature, time, aeration, growth phase of the mycelium, particle size of the substrate, carbon to nitrogen ratio, initial moisture content and pH as well as supplementation of the substrate with nitrogen and oil. Finally, the article highlights future research trends for the scaled-up production of PUFAs in SSF.  相似文献

Aims: To determine in liquid (LF) and solid‐state fermentation (SSF) the effect of medium concentration on growth and Taxol produced by Nigrospora sp., a fungus isolated from the Mexican yew. Methods and Results: Nigrospora sp. was grown at different concentrations of the base culture medium M1D, i.e. two (2×), four (4×), six (6×) and eight times (8×) the base concentration. The titres of Taxol determined by competitive inhibition enzyme immunoassay increased with increasing medium concentration in LF and SSF but were higher in SSF in every medium concentration. The Taxol produced in SSF and LF with 8× medium was 221 and 142 ng l?1. The SSF gave also higher biomass, growth and sugar utilization than LF in every medium. The growth and sugar consumption were modelled by the logistic and the Pirt models, respectively. However, the Luedeking–Piret model was unsuitable for Taxol. Conclusions: The SSF surpassed LF in terms of Taxol, growth and sugar utilization; thus, it has significant advantages over LF. Significance and Impact of the Study: This is the first report on Taxol production by SSF and the first contribution to evaluate the influence of the medium on Taxol production in LF and SSF.  相似文献

The endophytic fungi Acremonium loliae and a Gliocladium-like sp. were isolated from Lolium perenne; A. coenophialum and a Phialophora-like sp. from Festuca arundinacea; and Epichloe typhina from F. rubra. All five fungi infected endophyte-free seedlings of the host grasses and F. arundinacea after artificial inoculation. All fungi except A. coenophialum were able to infect L. perenne. The inoculation technique involved placing endophyte mycelium into the coleoptile tissue of sterile seedlings growing on water agar in Petri dishes. Infection of mature plants with endophytes was not achieved. The presence of some endophytes in grasses can be beneficial to plant growth and persistence but deleterious to the health of animals which graze them. The desirability of infecting cultivars of grasses with endophytes is discussed.  相似文献

Solid-state fermentation (SSF) with Agaricus brasiliensis and Agaricus bisporus on corn was carried out. The results showed that SSF with the two fungi made up the deficiency of tryptophan in corn and improved the protein nutritional value of corn. The conjugated polyphenols contents in fermented corn decreased and free polyphenols (FPP) contents increased. FPP contents in corn fermented with the two fungi reached respectively 25 and 88 times of control, total polyohenols contents reached respectively 1.4 and 3.3 times of control. The antioxidant properties (i.e. 1,1-diphenyl-2-picrylhydrazyl radical scavenging capacity, reducing power, ferrous ion chelating ability and superoxide anion radical scavenging ability) of fermented corn were improved significantly. SSF with A. bisporus was more favorable to the enhancement in protein nutritional value and antioxidant properties of corn than that of A. brasiliensis. The results indicated that SSF with the two fungi could upgrade the protein nutritional value, FPP content and antioxidant properties of corn.  相似文献

Natural gypsum (NG) and phosphogypsum (PG) were tested for resistance to fungal growth based on standard test ASTM D 3273-86, with the recommended mixture of three fungal species, and using the same test modified by the use of a Cladosporium sp. A, isolated from a gypsum plaster ceiling. In the standard test little growth occurred on any of the test specimens. However, abundant fungal growth was produced by the Cladosporium sp. A on phosphogypsum, which was much more susceptible than natural gypsum. Phosphogypsum heated to 600°C to destroy organic residues was resistant to growth of Cladosporium sp. A, as well as other fungi isolated from phosphogypsum panels stored in the environment for 2 years: Cladosporium sp. B, Aspergillus niger and Trichoderma sp. Phosphogypsum moulded in Petri dishes was susceptible to growth of a wide range of fungi, although Fusarium sp. and Rhizopus sp. caused practically no discoloration of the substrate. Six biocides were separately incorporated into the phosphogypsum at concentrations recommended by the suppliers and test specimens incubated on Sabouraud agar inoculated with various fungal isolates. The biocide 2-N-octyl-4-isothiazolin-3-one was the most efficient compound. It prevented the growth of the fungi most likely to cause health problems in buildings, but not that of Helminthosporium sp., isolated from powdered phosphogypsum in the factory. This was the most resistant fungus showing growth on all biocide-containing specimens.  相似文献

White-rot basidiomycetes are the main decomposers of woody biomass in forest ecosystems. Little is known, however, about the interactions between white-rot fungi and other microorganisms in decayed wood. A wood-rotting fungus, Stereum sp. strain TN4F, was isolated from a fruit body, and its coexisting cultivable bacteria were isolated from its substrate; natural white-rot decayed wood. The effects of bacteria on fungal growth were examined by confrontational assay in vitro. A growth-promoting bacterium for this Stereum strain was identified as Curtobacterium sp. TN4W-19, using 16SrRNA sequencing. A confrontational assay revealed that Curtobacterium sp. TN4W-19 significantly promoted the mycelial growth of Stereum sp. TN4F in the direction of the bacterial colony, without direct contact between the mycelium and bacterial cells. This is the first report of a positive interaction between a white-rot fungus and a coexisting bacterial strain in vitro.  相似文献

Summary In this study flake chitin, crab shell chitin, mushroom stalk, fungal cell wall, wheat bran and rice bran were used as substrate for chitinase production by Enterobacter sp. NRG4 under submerged and solid state fermentation (SSF) conditions. Enterobacter sp. NRG4 produced 72 and 49.7 U/ml of chitinase in presence of cell walls of Candida albicans and Fusarium moniliforme in submerged fermentation. Under SSF, maximum chitinase production was 965 U/g solid substrate with flake chitin and wheat bran (1:3 ratio) at 75% moisture level after 144 h. The purified chitinase inhibited hyphal extension of Fusarium moniliforme, Aspergillus niger, Mucor rouxi and Rhizopus nigricans. The chitinase was effective in release of protoplasts from Trichoderma ressei, Pleurotus florida, Agaricus bisporus and Aspergillus niger. Protoplasts yield was maximum with 60 mg of 24 h old fungal mycelium incubated with 60 U of chitinase and 60 U of cellulase.  相似文献

The aim of this study was to find the antifungal activities of seven essential oils from some Iranian medicinal plants that have maximum (100%) inhibition effect on the mycelium growth of postharvest phytopathogenic fungi. Among 20 examined species belonging to three families, only 7 species could stop the mycelium growth of phytopathogenic fungi. The selected plants include Trachyspermum ammi, Zataria multiflora Boiss., Satureia hortensis, Caryophillum aromaticus, Menthe piperita, Cuminum cyminum L. and Carum carvi, and fungi include Aspergillus flavus, Botrytis cinerea, Penicillium italicum, Penicillium expansum, Penicillium commune, Rhizopus stolonifer and Rhizopus lyococcus. The results showed that the essential oil of these plants could stop the mycelium growth at 500 ppm, but could not completely inhibit the spore germination, however reduced the spore germination to 80–90%. Among the fungi Rhizopus stolonifer and Rhizopus lyococcus are more resistant to the inhibition effects of essential oils. Among the plants, Cuminum cyminum L. and Carum carvi were slightly weaker than other plants. Also except for Cuminum cyminum L. and Carum carvi, the essential oils of other plants had fungicide effect while these two plants in most cases had fugistatic effect. The results showed that these essential oils can be used as an effective alternative control method.  相似文献

This study identified the main pathogens causing fruit rots of mature peaches in northern Greece, the major peach producing area of Greece. The brown rot pathogen Monilinia laxa was responsible for approximately 70% and 78% of rotted peaches in 2005 and 2006 respectively. Serious damage (up to 5%) was also caused with the fungus Phomopsis amygdali. Other pathogens isolated from rotted peaches at a low percentage were Alternaria alternata, Aspergillus niger, Aspergillus flavus, Botrytis cinerea, Sclerotinia sclerotiorum, Fusarium spp., Colletotrichum gloeosporioides, Rhizopus stolonifer and Gilbertella persicaria. Most fungal isolates originated from the rotted peaches were tested for their sensitivity to the fungicides iprodione, carbendazim, thiophanate methyl and tebuconazole at label recommended concentrations. All fungicides inhibited the growth of M. laxa, A. niger, A. flavus, S. sclerotiorum, P. amygdali and B. cinerea on poisoned agar. Apart from iprodione, all other fungicides inhibited the mycelium growth of the pathogen Fusarium sp. The mycelium growth of Fusarium sp. was significantly less with iprodione than control. Only iprodione and tebuconazole were effective against A. alternata and R. stolonifer. Tebuconazole inhibited the mycelium growth of R. stolonifer, while iprodione reduced significantly in comparison to control. The mycelium growth of the fungus C. gloeosporioides was inhibited by tebuconazole and reduced significantly by the fungicides thiophanate methyl, carbendazim and iprodione. Among all the fungi tested, only M. laxa and B. cinerea isolates were found resistant to benzimidazoles [the EC50 (50% effective concentration) value was 100–200 mg/l and 200–300 mg/l for the largest number of thiophanate methyl‐ and carbendazim‐resistant M. laxa isolates respectively, while the biggest number of B. cinerea thiophanate methyl‐ and carbendazim‐resistant isolates showed EC50 value 200–300 mg/l and 300–400 mg/l, respectively]. However, these strains were sensitive to tebuconazole and iprodione. Therefore, these fungicides can be used as an alternative method to control benzimidazole‐resistant Monilinia and Botrytis isolates.  相似文献

Investigation of the crude extracellular chitinase of Bacillussp. 739, an antagonist of phytopathogenic fungi, discerned a relationship between the chitinase and antifungal activities of this bacterium. Purified chitinase lost its ability to inhibit the growth of micromycetes. The antagonistic (antifungal) activity of crude chitinase was found to be located in a low-molecular-weight fraction of the enzyme, which does not possess chitinase activity. Both crude and purified chitinase were able to lyse the cell walls of intact mycelium. Accordingly, it may be inferred that the antagonistic activity of Bacillussp. 739 against micromycetes is largely determined by low-molecular-weight nonenzymatic substances, whereas the role of chitinase is to utilize chitin, which is ubiquitously present in soil.  相似文献

The ectomycorrhizal (ECM) fungi associated with Pinus thunbergii seedlings grown on sand dune were identified by molecular method, and the diversity of bacteria associated with ECM and Extraradical mycelium were examined by Denaturing Gradient Gel Electrophoresis (DGGE) of PCR-amplified 16S rDNA. The mycorrhizal formation rate of 1-year old P. thunbergii seedlings was more than 95%. Cenococcum geophilum was the most dominant ECM fungus, followed by T01, RFLP-8, Russula spp., and Suillus sp. Bacterial community was most diverse with C. geophilum- and RFLP-8-mycorrhiza. Sequencing analysis showed that Burkholderia spp. and Bradyrhizobium spp. were on the surface of ECM short root of seven ECM. The fungi detected as extraradical mycelium using DGGE of 18S rDNA were Suillus bovinus and RFLP-8-mycorrhiza. Bacterial community on the extraradical mycelium was more diverse than those on ECM root tip. Burkholderia spp. and Bradyrhizobium spp. were found also on extraradical mycelium.  相似文献

To examine the relative importance of leaf age and season on the occurrence of phyllosphere fungi, temporal patterns of epiphytic and endophytic phyllosphere fungi of giant dogwood (Swida controversa) were studied with reference to leaf emergence at first occurrence and in the middle of the growing season. A total of 15 and 44 species were isolated from the surface and interior of leaves, respectively. On the leaf surface, detection rate of fungi was consistently 100% and their frequencies increased during the growing season, whereas in the leaf interior, detection rate of fungi and their frequencies were low at leaf emergence and gradually increased during the growing season. Six epiphytic and two endophytic fungi were observed frequently. A white sterile mycelium was frequent only on the surface of newly emerged leaves in the first-order shoot in May. The other 7 species increased during the growing season. The frequencies of Phomopsis sp., Pestalotiopsis sp. 1, and Trichoderma viride were higher on the leaves of first-order shoots than those of higher-order shoots that emerged between July and September, suggesting the possible effects of leaf age on their occurrence. On the other hand, the frequencies of Colletotrichum gloeosporioides, Clonostachys rosea, Cladosporium cladosporioides, and Phoma sp. 1 were not different between the first- and higher-order shoots, suggesting the negligible effect of leaf age. The influence of phenological patterns of leaf emergence of deciduous trees on the diversity and composition of assemblages of phyllosphere fungi is discussed.  相似文献

New fossil remains have been discovered from the well-known Lakhanda Microbiota (1015–1025 Ma, Uchur-Maya Region, Southeastern Siberia). The microfossils have characters observed in microscopic fungi, such as a reticulate mycelium, formed by anastomizing hyphae of non-cellular structure and of variable width. The fungal mycelium shows that hyphae grew in a certain direction, both forwards and backwards. As their length increased, the hyphae became curved and merged with neighboring hyphae to form enclosed spaces. The presence on hyphae of knoblike trapping structures and secretion of adhesive enzymes suggest a similarity with modern nematophagous fungi. Microorganisms associated with the adhesive hyphae consist of colonial green unicellular algae and thin bacterial sheaths, which may indicate mutually beneficial relationships between morphologically and biologically different partners. Microfossils of fungal origin are systematically described. They include Aimonema ramosa gen. et sp. nov. and the green unicellular alga Eoprotoderma neruenica gen. et sp. nov., forming a symbiotic association.  相似文献

Twenty-seven species of freshwater Hyphomycetes from two five-state areas in the northern Appalachian Highland, New England, and three Coastal Plain states were collected, isolated, and identified. The spores of three fungi which are probably aquatic Moniliales were also studied. Sigmoidea, a new Hyphomycete genus, includes submerged aquatic fungi which produce phialides laterally along the mycelium and sigmoid, multicellular phialospores. Varicosporium giganteum sp. n. produces terminal, hyaline, branched, and multiseptate aleuriospores. The main spore axis has one to four lateral branches which arise successively on one side of this axis. Tricladium marylandicum sp. n. is similar to T. splendens in that the lateral branches are constricted where each branch joins the main spore axis. It differs from this species in that the spore has a blunt apical cell on the main axis and on each of the lateral axes. Three possible methods of dispersal are presented to explain the cosmopolitan distribution of freshwater Hyphomycetes.  相似文献

To discover novel and effective antifungal candidates, a series of new curcumol derivatives were designed, synthesized, and evaluated their antifungal activity against five phytopathogenic fungi by the mycelium growth rate method. Derivatives c4 , c22 and c23 exhibited excellent antifungal activity against Phomopsis sp. with EC50 values of 3.06, 3.07, and 3.16 μM, respectively. Specifically, compound c4 exhibited the strongest antifungal activity against Phomopsis sp., which was 44 times that of pyrimethanil (EC50=134.37 μM). The results of scanning electron microscopy (SEM) and transmission electron microscopy (TEM) indicated that compound c4 could cause cell senescence and death of Phomopsis sp. by changing the normal hyphal morphology and disrupting the normal metabolism of hyphal cells. Moreover, compound c4 showed excellent curative effect against Phomopsis sp. on kiwifruit. These findings confirmed that compound c4 has great potential as a potent antifungal agent.  相似文献

Nineteen filamentous fungi, isolated from estuarine sediments in Brazil, were screened for degradation of polycyclic aromatic hydrocarbons (PAH). The fungal isolates were incubated with pyrene. The cultures were extracted and metabolites in the extracts were detected by high performance liquid chromatography (HPLC) and u.v. spectral analyses. Six fungi were selected for further studies using [4,5,9,10-14C]pyrene. Cyclothyrium sp., Penicillium simplicissimum, Psilocybe sp., and a sterile mycelium demonstrated the ability to transform pyrene. Cyclothyrium sp. was the most efficient fungus, transforming 48% of pyrene to pyrene trans-4,5-dihydrodiol, pyrene-1,6-quinone, pyrene-1,8-quinone and 1-hydroxypyrene. This fungus was also evaluated with a synthetic mixture of PAH. After 192 h of incubation, Cyclothyrium sp. was able to degrade simultaneously 70, 74, 59 and 38% of phenanthrene, pyrene, anthracene and benzo[a]pyrene, respectively.  相似文献

We estimated the biomass and growth of arbuscular mycorrhizal (AM) mycelium in sand dunes using signature fatty acids. Mesh bags and tubes, containing initially mycelium-free sand, were buried in the field near the roots of the dune grass Ammophila arenaria L. AM fungal mycelia were detected at a distance of about 8.5 cm from the roots after 68 days of growth by use of neutral lipid fatty acid (NLFA) 16:1ω5. The average rate of mycelium extension during September and October was estimated as 1.2 mm day−1. The lipid and fatty acid compositions of AM fungal mycelia of isolates and from sand dunes were analysed and showed all to be of a similar composition. Phospholipid fatty acids (PLFAs) can be used as indicators of microbial biomass. The mycelium of G. intraradices growing in glass beads contained 8.3 nmol PLFAs per mg dry biomass, and about 15% of the PLFAs in G. intraradices, G. claroideum and AM fungal mycelium extracted from sand dunes, consisted of the signature PLFA 16:1ω5. We thus suggest a conversion factor of 1.2 nmol PLFA 16:1ω5 per mg dry biomass. Calculations using this conversion factor indicated up to 34 μg dry AM fungal biomass per g sand in the sand dunes, which was less than one tenth of that found in an experimental system with Glomus spp. growing with cucumber as plant associate in agricultural soil. The PLFA results from different systems indicated that the biomass of the AM fungi constitutes a considerable part of the total soil microbial biomass. Calculations based on ATP of AM fungi in an experimental growth system indicated that the biomass of the AM fungi constituted approximately 30% of the total microbial biomass. Environmental pollution in addition to direct damage on plant growth, with the destruction of biological control agents, causes indirect damage to plants. The aim of this research was to study the effects of different concentrations (0, 500, 1000, 1500 and 2000 ppm) of heavy metals including Ag, Co, Cu, Fe, Hg, Mn, Pb and Zn on the mycelial growth and to assess the fungicidal or fungistatic effects of these salts on five Nematophagus fungi including Trichoderma harzianum (T8), Trichoderma virens (T21), Trichoderma hamatum (T9), Pochonia chlamydosporia var. chlamydosporia and Arthrobotrys oligospora. The results show that Ag, Co, Cu, Fe and Hg could stop the mycelium growth of all fungi, but Mn, Pb and Zn cannot inhibit the growth of these fungi completely. Among the first group, Hg and Cu stopped the growth of fungi even in 500 ppm. Among these metals that inhibit the growth of fungi, Cu has fungistatic effect and others have fungicide effect. The experiment was conducted in vitro condition, using potato dextrose agar (PDA) under complete randomised design with four replications. The data of mycelium growth were recorded at seven days after inoculation at 25 ± 2°C.  相似文献
