This article presents a multispectral image analysis approach for probing the spectral backscattered irradiance from algal cultures. It was demonstrated how this spectral information can be used to measure algal biomass concentration, detect invasive species, and monitor culture health in real time. To accomplish this, a conventional RGB camera was used as a three band photodetector for imaging cultures of the green alga Chlorella sp. and the cyanobacterium Anabaena variabilis. A novel floating reference platform was placed in the culture, which enhanced the sensitivity of image color intensity to biomass concentration. Correlations were generated between the RGB color vector of culture images and the biomass concentrations for monocultures of each strain. These correlations predicted the biomass concentrations of independently prepared cultures with average errors of 22 and 14%, respectively. Moreover, the difference in spectral signatures between the two strains was exploited to detect the invasion of Chlorella sp. cultures by A. variabilis. Invasion was successfully detected for A. variabilis to Chlorella sp. mass ratios as small as 0.08. Finally, a method was presented for using multispectral imaging to detect thermal stress in A. variabilis. New biosorbent material derived from ubiquitous marine algae has been examined in packed-bed flow for Pb2+ removal through sorption columns. Mixed biomass of marine algae has been used, consisting of representative species of the following algae: Ulva lactuca (green algae), Jania rubens (red algae), and Sargassum asperifolium (brown algae). A mixture of these three species showed a promising removal capacity for Pb2+ from aqueous solution. Lead uptake up to 281.8 mg/g dry algal mixture was observed. Equilibrium was achieved after 120 min. No significant effect of changing the flow rate on the removal capacity was noticed. It was found that Langmuir model expresses the system at pH 4. Mineral acids exhibited good elution properties (a mean of 93%) for recovery of sorbed biomass ions as compared with the tested alkalies (about 60%). The influence of far‐red light (FRL) was studied on the chemical composition of Spirulina platensis biomass. The following light compositions were used during the culture white light, blue‐red LED light (BRL) and BRL supplemented with FRL (BRFRL). Chlorophyll and phenol contents were measured by spectrophotometric methods, whereas presence of carotenoids, lipids, and phycobiliproteins were estimated by Fourier‐transform Raman spectrometry. Additionally, phenol content was investigated by fluorescence intensity of algae culture in the range of 430–650 nm. The content of chlorophyll and phenols in algae cells depended on the spectral composition of light and was the highest under BRL (16.7 ± 0.5 and 9.1 ± 0.6, respectively). It was shown that there is a positive linear correlation (R = 0.902 at p < 0.0000001) between the ratio of relative fluorescence intensity of S. platensis suspensions at 450 nm to the suspensions at 540 nm (F450/F540) and the content of phenolic compounds in the biomass. Changes in the F450/F540 ratio can explain approximately 80% changes of phenol contents in algae cells. Spirulina platensis Raman spectra demonstrated that the biomass of algae growing under white light and BRL had a significantly higher intensity of phycobiliprotein bands than the algae growing under BRFRL.  相似文献

A nutritional assay has been conducted with rainbow trout fry (Oncorhynchus mykiss) using a biomass of photosynthetic micro-organisms. The algal biomass was incorporated in the feed at increasing levels (12.5%, 25% and 50%) in order to verify the effects on survival (%), growth (length and mass) and carcass quality of the fish (skin color, water, protein and lipid content). Two commercial feeds (CF, as sold, and 0% A, reprocessed without algae) have been used as controls. After eight weeks at 10 degrees C, the growth has been significantly reduced in fish fed the feeds containing 25% and 50% algae diets (P<0.01). Moreover, an incorporation of algae higher than 12.5% in the feed led to a increase of water content (CF versus other diets; P<0.01) and TL (CF versus 50% A; P<0.01) (wet basis) content of carcasses. Survival was 100% in all treatment groups. Under the experimental conditions used, the results obtained show that a maximum of 12.5% of algal biomass can be incorporated in the feed for rainbow trout fry (O. mykiss) without negative consequences on growth and body content in lipids and energy of fish.  相似文献

Experimental studies of the effect of phosphorus, nitrogen, and silicon on the phytoplankton of the shallow highly eutrophic Lake Nero have been carried out. It is shown that the enrichment of lake water with nitrogen leads to an increased concentration of chlorophyll a and the growth of the biomass of dominant algae species. Phosphorus limits the abundance of a certain species of algae; the limiting role of silicon in the development of spring phytoplankton has not been experimentaly confirmed.  相似文献

Two representative methods for quantitative estimation of soil algae, the culture dilution method and chlorophyll a extraction, were compared using soil samples collected from irrigation land in the flood plain of the River Ili, Kazakhstan, where the distribution of soil algae had been studied in the previous year. The estimate by the culture dilution method was almost the same as in the previous year, except for one site, which was enclosed by shrubs of sakusaul, Haloxylon aphyllum (Minkw.) Iljin. The important role of wind in transport of airborne algal cells was pointed out. There was a significant correlation (p<0.001) between the logarithm of the number of colonies by the culture dilution method andthe logarithm of the concentration of chlorophyll a, when data from all samples were analyzed. However, no significant correlation was observed with the data of cropland sites alone. Furthermore, the seasonal variations of both values at each site did not necessarily agree with each other. One reason for the inconsistency may have been the over estimation of chlorophyll a caused by inclusion of litter from vascular plants. Other reasons may relate to differences between the methodologies. The density of soil algae estimated by culture dilution reflects the algal biomass in a certain previous period of time. Therefore, it is suggested that the method is suitable for spatial, but not for seasonal studies. In this study, the ability of algae to treat a wood-based pulp and paper industry wastewater was investigated. Tests were performed in batch reactors seeded with a mixed culture of algae. Under different lighting and initial wastewater strength conditions, changes in COD, AOX and color contents of reactors were followed with time. Algae were found to remove up to 58% of COD, 84% of color and 80% of AOX from pulp and paper industry wastewaters. No remarkable differences were observed in COD and color when light intensity and wastewater strength were changed, while AOX removals were strongly affected. Algal species identification studies revealed that some green algae (Chlorella) and diatom species were dominant in the treatment. The study also showed that algae grew mixotrophically, while the main mechanism of color and organics removal from pulping effluents was partly metabolism and partly metabolic conversion of colored and chlorinated molecules to non-colored and non-chlorinated molecules. Adsorption onto algal biomass was not so effective.  相似文献

Chemically inert, cylindrical rods positioned in the littoral of two eutrophic Alberta lakes supported higher periphytic algal biomass (measured as total chlorophyll a) than nearby morphologically similar culms of Scirpus validus Vahl. during most of the summer. Upon initiation of macrophyte senescence, biomass on the two substrata became more similar. Experiments were conducted to investigate the basis for these observations. Whole extracts of intact vegetative Scirpus culms had no effect on periphyton photosynthesis, suggesting that the natural substrata do not produce water-soluble allelochemicals. Various modifications of the rod surfaces (roughening, wax coating, wax color) were used to test whether surficial properties of Scirpus culms influenced periphyton accumulation. Roughened rods supported levels of biomass similar to those of smooth rods, and both substrata developed structurally complex periphyton communities. Rods covered with paraffin wax had periphyton communities that were lower in biomass and structurally more simple than those on un-coated rods or on Scirpus culms. Coloring of the wax coating had no consistent effect on periphyton accumulation. We hypothesize that the hydrophobic cuticle on actively growing Scirpus culms retards the development of precursors for attachment by periphytic algae. Upon senescence of the culm and loss of epidermal integrity, colonization of culm surfaces by periphytic algae may occur in a manner similar to that on artificial substrata.  相似文献

Microalgae are a very diverse group of organisms that consist in both prokaryotic and eukaryotic forms. Some species of microalgae can be induced to overproduce particular fatty acids through simple manipulations of the physical and chemical properties of the culture medium. In this paper, the effect of different extraction techniques on the recovery of fatty acids from the freeze-dried biomass from two lipid-producing microalgal strains: Botryococcus braunii LB 572 (green algae) and Synechocystis sp. PCC 6803 (cyanobacteria) was examined. Five procedures were used: after conversion of the lipid material into the corresponding fatty acid methyl esters (FAMEs) via suitable derivatization reactions (extraction-transesterification) and direct transesterification of biomass to produce FAMEs (without the initial extraction step) that used differential types of catalysts and processing conditions. This study has shown that procedure 3, a one step practical procedure for lipid extraction and in situ methyl ester derivation could be used successfully for the determination of the fatty acid compositions of microalgae and cyanobacteria.  相似文献

A summary is presented of estimates of distribution and growth of filamentous algae and its effect on the structure and functioning of epibenthic fauna and fish communities in shallow bays on the Swedish west coast. As a consequence of coastal eutrophication vegetation cover has gradually increased during the last decade, and during 1990's most bays in the Skagerrak-Kattegat area were variously covered with filamentous algae during spring and summer (May–July). In some areas filamentous algae (mainlyCladophora andEnteromorpha) completely covered the bottom. In field studies it was demonstrated that increased cover and dominance of filamentous algae result in structural changes of the epibenthic fauna community. Field studies showed that species richness and biomass of epibenthic fauna increased in a sandy bay with a moderate increase (30 to 50%) of filamentous algae cover. At higher cover (90%), biomass of epibenthic fauna was reduced, however, to the same level as for the sandy habitat, although the dominant epibenthic species were different. Heavy growth of epiphytic filamentous algae on eelgrass resulted in reduced biomass and a shift in the species composition of the epibenthic fauna community. Fish assemblage structure was also related to changes in vegetation. In eelgrass beds, fish species numbers were reduced with increasing cover of epiphytic filamentous algae, and at rocky bottoms with kelp algae (dominated byFucus), fish biomass decreased with increasing cover of attached filamentous algae. Further, foraging efficiency of juvenile cod and settling success of plaice were reduced as a response to increasing dominance of filamentous algae.  相似文献

海洋环境中,细菌和微藻之间的物质交换是生源要素在自然界中迁移转化的重要方式。为进一步了解生源要素的生物地球化学循环,在实验室模拟条件下,研究了共培养体系中营养盐和有机物在细菌和微藻之间的转换。通过纯培养中肋骨条藻(Skeletonema costatum)、东海原甲藻(Prorocentrum donghaiense)、天然海水中的细菌以及藻菌混合培养,分析了营养盐和有机物随藻菌生物量的变化情况,并计算了溶解有机碳(DOC)和溶解有机氮(DON)的浓度比值[(DOC/DON)a]。结果发现,在共培养体系中,细菌对中肋骨条藻的生长有抑制作用,对东海原甲藻影响不明显;中肋骨条藻有利于细菌生长,东海原甲藻抑制细菌生长,这种不同可能与微藻的粒径有关。海洋细菌在2种藻的指数生长均期均会促进微藻吸收氨氮(NH_4-N),但在生长末期NH_4-N以释放为主。硝氮(NO_3-N)的浓度与藻的生长呈负相关,但在衰亡期NO_3-N略有增加,表明NO_3-N再生所需时间较长。细菌对硝氮的吸收量较少,但对其再生有贡献。细菌和中肋骨条藻对磷酸盐(PO_4-P)的吸收存在竞争,但与东海原甲藻的竞争关系不明显。不同培养体系中DOC浓度变化不同,在藻菌共培养体系中增加较快,纯藻培养体系中增加缓慢,在纯菌培养体系中缓慢减少。通过对DOC与DON浓度比值的分析,发现用判断颗粒有机碳(POC)来源的方法可以分析DOC的来源。  相似文献

Mass-cultured algal biomass has been tested as a food source for a number of aquaculture animals because of its low cost and convenience. This paper reviews the results of nutritional studies on processed microalgae with respect to mollusc, crustacean, rotifer and fish culture. Research using species of Spirulina, Chlorella, Scenedesmus and other mass-produced algae indicates that microalgae can be an effective dietary component provided that processing, diet formulation and presentation requirements are met. Processed microalgae can be used to correct specific dietary deficiencies in artificial diets. Our research found that the growth and pigmentation of marron, Cherax tenuimanus (Decapoda, Crustacea), can be significantly enhanced by the incorporation of Dunaliella salina in its artificial diet. Likewise, rainbow trout, Oncorhynchus mykiss, were pigmented by Haematococcus pluvialis.  相似文献

In outdoor thin-layer sloping reactors algae are batch cultured and harvested at biomass concentrations of about 15 g (dw) I-1 whereafter a portion is used as inoculum for the next cycle. Light saturation curves of the oxygen evolution (PII curves) of the algae were measured using diluted aliquots of suspension taken from the reactors. The maximum specific photosynthetic rates (P B max) and the light intensity at the onset of saturated photosynthesis (I k ) decreased whilst the maximum specific photosynthetic efficiency ( B ) increased with an increase in the biomass concentration, during the production cycle. These differences reflect transition from light- to dark-acclimated state of the algae that occurs as a result of an increase of the suspension concentration during the production cycle. During these experiments the thin-layered smooth sloping cultures (TLSS, culture depth 5–7 mm) had higher photosynthetic rates per volume than the thin-layered baffled sloping cultures (TLBS, culture depth 5–15 mm). This was ascribed to the higherP B max values of the algae grown in the TLSS cultures, allowing them to utilise high incident irradiancies more effectively. However, the areal productivity of the TLBS was higher than the TLSS indicating a higher photosynthetic efficiency of the TLBS reactors. The specific productivity decreased rapidly with an increase in the biomass concentration, but the yield remained linear during the batch production cycle, even at high areal densities.  相似文献

【目的】研究自养和兼养两种培养方式对蛋白核小球藻(Chlorella pyrenoidosa)生长、细胞分裂和生化组分积累的影响,探讨人工培养蛋白核小球藻的昼夜节律响应机制和优化技术。【方法】小球藻自养培养采用BG11培养基,兼养培养基在BG11培养基中添加4种不同浓度(1、5、10、20 g/L)的葡萄糖,培养周期为10 d。血球板计数法测定藻细胞浓度,干重法测定藻细胞生物量。显微观察藻细胞大小和分裂情况。脂染色法测定小球藻总脂的含量,藻细胞的叶绿素、蛋白和淀粉分别采用甲醇、氢氧化钠、硝酸钙浸提后通过紫外分光光度法定量测定。【结果】葡萄糖兼养培养对蛋白核小球藻具有显著的促生长效应,最适浓度为10 g/L。10 d收获时,兼养组(10 g/L葡萄糖)藻细胞浓度和干重分别是自养组的2.57倍和6.73倍。分析一昼夜中的藻细胞增殖规律可知,第2天和第5天时自养组中增殖的新生子细胞约有76.00%在黑暗期分裂产生,而兼养组中第2天和第5天光照期的新细胞增殖量占比分别达到40.90%和67.50%。一昼夜内藻细胞大小的迁移动态监测表明,第2天自养组藻细胞的体积变化静息期为8 h,兼养组只有4 h;第5天两组藻细胞大小迁移动态的昼夜节律明显,但兼养组黑暗结束后较大细胞(D6μm)占比显著高于自养组。第8天时,兼养组藻细胞已处于稳定期,总脂和蛋白含量均显著高于自养组,藻细胞总脂和色素含量在一昼夜中相对稳定,但蛋白和淀粉含量分别在光照8 h和12 h左右达到峰值。从第2天开始,对兼养组细胞每天进行2 h光延长,收获时藻细胞浓度和干重分别比对照组提高13%和11%。【结论】葡萄糖兼养培养能大幅提高蛋白核小球藻的生物量。蛋白核小球藻生长增殖与生化组分积累均受昼夜节律调控,自养条件下藻细胞以光照期生长黑暗期增殖为主。兼养培养提高藻细胞生物量的机制在于缩短藻细胞生长静息期,在昼夜节律中加速藻细胞生长并显著提高通过细胞周期检查点的细胞比例,光照期效应尤其明显。藻细胞蛋白和淀粉含量昼夜节律明显,最佳收获时间分别在光照8 h和12 h后。  相似文献

Since the establishment of abalone farming, there has been an increase in the demand for Macrocystis as a food source. Therefore, the pressure on natural stock has also augmented and the sustainability of the actual harvesting practices has been questioned. In this article, an attempt to farm Macrocystis pyrifera by zoospores in northern Chile is described; initially under laboratory conditions and subsequently by cultivation in the sea. The experiments were executed during 1 year and two different cultivation methodologies were used: a direct and an indirect method. A maximum frond length of 175 cm was reached and 22 kg m−1of rope was produced after 120 to 150 days of cultivation in the sea. The algae grew under both methodologies, and no differences in algal length and biomass were detected between the two cultivation systems. However, the direct culture method can be recommended for productive and practical reasons.  相似文献

Development of bioreactor systems utilizing plant suspension cultures has been hindered by the lack of on-line sensors for monitoring important process variables such as biomass concentration and aggregate size. An optical technique, the focused beam reflectance method (FBRM developed by Lasentec Inc., Redmond, WA), was used to characterize several plant suspension cultures: rice (Oryza sativa), tobacco (Nicotiana benthamiana) and wild Chinese cucumber (Trichosanthes kirilowii). These cultures differ in a number of respects such as individual cell size and morphology, aggregate shape and size distribution, initial culture density, and color. For plant suspensions comprised of relatively spherical aggregates (rice and cucumber), the area under the cube-weighted FBRM chord length distribution was linearly correlated to biomass concentration (R 2>0.99) while the mean of the cube-weighted FBRM chord length distribution was nonlinearly related to aggregate size.  相似文献

Studies were carried out in Lake Mutek (Mazurian Lakeland) on the effect of artificial aeration and destratification upon quantitative changes in the phytoplankton. These studies were carried out from 1977 until 1983. Two different methods of aeration were used. Low intensity mixing resulted initially in a two-fold, and later on in a four-fold increase of the phytoplankton biomass. Increase of phytoplankton biomass during lake aeration was due to the development of Ceratium hirudinella. Use of a highly effective air-compressor caused an inhibition of algal development, so that biomass dropped to levels noted in the control year. It was found that the effect of aeration depended on the ratio between lake area and effectiveness of the aerator. Only intensive mixing of the water masses resulted in an inhibition of the development of algae. The effect of artificial destratification was also reflected in changes of the species structure, seasonal succession of the algae, and physiological state of the phytoplankton. Artificial circulation stimulated development of algae characterized by relatively high specific weight, i.e. most of all of Pyrrophyta, Bacillariophyceae and some species of Chlorophyta. Various aspects were discussed of the use of direct aeration as a technical method of lake restoration.  相似文献

In microbial cultures, both cellular growth rate and yield (defined as the degree of substrate conversion into the biomass) are important. Although effect of culture conditions on growth kinetics has been well documented for various microbial strains, there is almost no literature concerning the effect of environmental conditions on growth equilibrium, expressed as biomass yield coefficients from substrate. The present paper discusses the effect of culture conditions: irradiance (physical substrate) and glucose concentration (chemical substrate) on biomass yield coefficients from two chemical substrates: glucose and nitrate-nitrogen in photoautotrophic, heterotrophic and mixotrophic culture of blue-green alga Spirulina (Arthrospira) sp. The efficiency of substrates incorporation into the biomass can be precisely determined only if the elemental composition of the biomass is known. The experimental results showed that culture conditions had a substantial influence on biomass yield coefficients (biomass yield from glucose and nitrate-nitrogen). It was found that, the increase of irradiance favoured increase of biomass yield coefficient from both, glucose and nitrate-nitrogen. However, in the case of yield from nitrogen in mixotrophic culture, the effect was opposite. The effect of glucose concentration was different: the higher the initial glucose concentration, the lower the biomass yield coefficients from chemical substrates.  相似文献

Development of an attached microalgal growth system for biofuel production   总被引:4,自引:0,他引:4  
Algal biofuel production has gained a renewed interest in recent years but is still not economically feasible due to several limitations related to algal culture. The objective of this study is to explore a novel attached culture system for growing the alga Chlorella sp. as biodiesel feedstock, with dairy manure wastewater being used as growth medium. Among supporting materials tested for algal attachment, polystyrene foam led to a firm attachment, high biomass yield (25.65 g/m2, dry basis), and high fatty acid yield (2.31 g/m2). The biomass attached on the supporting material surface was harvested by scraping; the residual colonies left on the surface served as inoculum for regrowth. The algae regrowth on the colony-established surface resulted in a higher biomass yield than that from the initial growth on fresh surface due to the downtime saved for initial algal attachment. The 10-day regrowth culture resulted in a high biodiesel production potential with a fatty acid methyl esters yield of 2.59 g/m2 and a productivity of 0.26 g/m−2 day−1. The attached algal culture also removed 61–79% total nitrogen and 62–93% total phosphorus from dairy manure wastewater, depending on different culture conditions. The biomass harvested from the attached growth system (through scraping) had a water content of 93.75%, similar to that harvested from suspended culture system (through centrifugation). Collectively, the attached algal culture system with polystyrene foam as a supporting material demonstrated a good performance in terms of biomass yield, biodiesel production potential, ease to harvest biomass, and physical robustness for reuse.  相似文献

The principal deviation between parameters of spectral characteristics of surface structures and whole cells of unicellular algae Dunaliella tertiolecta and Tetraselmis viridis before and after influence of electromagnetic field (EMF) has been determined.
