For a period of 7 months water samples were analyzed for the presence ofKlebsiella pneumoniae and fecal coliforms at 11 sites in a cloud rain forest watershed in Puerto Rico. Diffusion chamber studies were conducted at two sites which were found to contain low numbers of naturally occurringK. pneumoniae and fecal coliforms. These studies indicated that bothK. pneumoniae andEscherichia coli may be capable of surviving environmental conditions for extended periods of time. The presence of both bacteria in pristine natural waters is indicative of their being autochtonous to tropical environments. Thus, as a result, the use of coliform and even fecal coliform bacteria as indicators of fecal pollution may be misleading when applied to countries with a tropical climate.  相似文献

The survival ofStreptococcus faecalis andEscherichia coli was studied in situ in a tropical rain forest watershed using membrane diffusion chambers. Densities were determined by acridine orange direct count and Coulter Counter. Population activity was determined by microautoradiography, cell respiration, and by nucleic acid composition. Densities ofS. faecalis andE. coli decreased less than 1 log unit after 105 hours as measured by direct count methods. Activity as measured by respiration, acridine orange activity, and microautoradiography indicated that both bacteria remained moderately active during the entire study. After 12 hours,E. coli was more active thanS. faecalis as measured by nucleic acid composition. In this tropical rain forest watershed,E. coli andS. faecalis survived and remained active for more than 5 days; consequently, both would seem to be unsuitable as indicators of recent fecal contamination in tropical waters.  相似文献

Vibrio cholerae and Escherichia coli were inoculated into membrane diffusion chambers and placed around two small coral reef islands in Puerto Rico and monitored for 5 days. Several chambers were also buried in the sands of one of the reefs. Both E. coli and V. cholerae densities declined by 2 orders of magnitude, as measured by direct particle counts with a Coulter Counter (Coulter Electronics, Inc., Hialeah, Fla.). However, the density of neither bacteria changed dramatically when the same samples were analyzed by epifluorescent direct counts. Differences in the two direct count methods were accounted for by changes in cell morphology that occurred in both bacteria after exposure to seawater. Morphological changes occurred more rapidly in E. coli compared with those in V. cholerae. Bacteria in chambers exposed to sediment did not show significant changes in morphology and had only a slight decline in density. Physiological activity declined by more than 40% for both bacteria within 24 h. The decline in activity was less severe in the sediments. Tropical coral reef sands and turtle grass beds were shown to be less stressful environments for V. cholerae and E. coli than would have been predicted from temperature and microcosm studies. V. cholerae can survive the in situ conditions of a tropical coral reef and could become a source of bacterial contamination for fish and shellfish in this environment. The simultaneous monitoring of E. coli levels established that this bacteria can not be used as an indicator of V. cholerae or other fecal-borne pathogens in coral reef environments because of the greater stress these environments put on E. coli. Both bacteria could be of greater public health importance in tropical marine areas than previously imagined.  相似文献

Densities of V. cholerae and fecal coliforms were not significantly correlated, even though the highest densities of both bacteria were found at a sewage outfall. High densities of V. cholerae were also found at pristine sites at the highest point in the watershed. The density of Escherichia coli and V. cholerae in membrane diffusion chambers did not change significantly during the course of two such studies. Physiological activity, as measured by electron transport system activity and relative nucleic acid composition, indicated that both E. coli and V. cholerae remained active. This study suggests that V. cholerae is indigenous to tropical fresh waters and that assays other than those that detect fecal coliforms or E. coli must be used for assessing public health risk in tropical waters.  相似文献

Repeated lyophilization of Deinococcus proteolyticus, Micrococcus luteus and Escherichia coli cells results in a successive decrease of their survival. The survival curve is exponential with E. coli and M. luteus, and sigmoidal with a broad shoulder with D. proteolyticus both after repeated lyophilization and after UV- or gamma-irradiation. When cells were subjected to gamma-irradiation after a 20-fold freeze-drying, the corresponding survival curve became exponential without the shoulder. Hence we assume that irradiation and repeated lyophilization afflict the same cellular structures and/or functions.  相似文献

Flow cytometry was used to characterize bacterioplankton from two tropical environments in Brazil: the eutrophic Guanabara Bay and the oligotrophic southwest Atlantic Ocean. Bacterial abundance was evaluated by flow cytometry, and cells were stained with SYTO 13, allowing demonstration of differences in nucleic acid content. Bacterial production was also evaluated by means of 3H-leucine incorporation. Bacterial numbers were different for both sites. In Atlantic Ocean samples, we found a maximum of 5.50 x 10(5) cells ml(-1), and low nucleic acid content organisms predominated. In Guanabara Bay, bacterial numbers were one order of magnitude higher than in the ocean, and they varied from outer bay (1.01 x 10(6) cells ml(-1)) to inner bay (6.90 x 10(6) cells ml(-1)). Bacterial activity in ocean samples varied from 4.6 to 126 ng C l(-1) h(-1), while in the bay, mean values ranged from 1.95 microg C l(-1) h(-1) (outer bay) to 7.35 microg C l(-1) h(-1) (inner bay). Values found for both parameters are characteristic of different trophic situations. These results illustrate the utility of cytometric analyses of bacterioplankton populations in characterizing their large spatial and temporal scales of distribution in aquatic ecosystems.  相似文献

Two methods for determining relative microbial activity in the marine environment were compared. In one method, a single concentration of a labeled substrate was used to calculate rates of substrate utilization; in the other, multiple concentrations of the same substrate (heterotrophic activity method) were used to calculate maximum potential substrate utilization rates. These studies were made on 232 seawater and 79 sediment samples taken from a variety of marine environments. The highest correlations between these two methods were seen in the sediment samples tested. The lowest correlation coerfficient seen in the sediment samples was 0.90, and the highest was 0.98. In seawater samples (six studies), the lowest correlation coefficient was 0.77 and the highest was 0.95. The correlation between these two methods was also substrate concentration dependent. Higher correlation coefficients were observed when higher substrate concentrations were used. Under certain conditions, these two methods appear to be comparable for estimating relative levels of microbial activity in the marine environment.  相似文献

Summary Thein vitro activity of a combination of colimycin and tetracycline againstPseudomonas aeruginosa andEscherichia coli was studied with the aid of tube dilution tests,Elek andHilson's modification of the replica technique, and growth inhibition curves. The interaction between the two antibiotics was as a rule of an additive nature, but a slight synergism was observed in some instances. The values of the minimum inhibitory concentrations did not seem to influence the nature of the interaction. A considerable synergism observed in all tube dilution tests withE. coli strains is attributed partly to addition and partly to suppression of the growth of more colimycin-resistant bacteria by the tetracycline. Possiblein vivo applications of this antibiotic combination are briefly discussed.  相似文献

A silver-resistant mutant ofKlebsiella pneumoniae B-5 was produced by passaging in nutrient broth containing graded concentrations of silver nitrate up to 150 ppm. The development of silver resistance in the strain resulted in rough colonies, decrease in cell size, carbohydrate content and change in klebocin pattern. The virulence of the AgR strain as checked by the burn wound model decreased as the mutant could not establish itself in the skin and spleen of the animals and the organism was cleared more efficiently by human lymphocytes than the parent AgS strain.  相似文献

Sediments in the marine environment are generally subject to higher energy levels than those of lake systems, and lakes are virtually unaffected by tidal range which modifies beach structures formed in response to wave effects. However, despite different energy levels, the textural characteristics of both marine and lacustrine sediments are very similar. The main difference between marine and lacustrine facies is the depth range over which these characteristics remain consistent. In lakes, depth limitation may influence the development of surface waves and restrict textural distributions.Simple textural relationships can be used to describe comparable marine and lacustrine sedimentary conditions. Sediments which have been altered by post-depositional erosion, such as lag deposits, or by ice-drop or wind blown settlement, show comparable textural modifications.The settlement of silt and clay size particulates, in the marine environment and lakes, may differ slightly because of the chemical differences between salt and fresh water.  相似文献

The enterotoxigenicity, klebocinogeny and susceptibility to antibiotics for 100 strains of Klebsiella pneumoniae isolated from various food samples is reported. 49% of the samples supported growth of K. pneumoniae: a high proportion of these strains were isolated from sweets and snacks (51%). Enterotoxigenicity as determined by a coagglutination test with anti-LT4 coated staphylococci was observed in 61 of 100 Klebsiella isolates. 21% of the strains were found to be klebocin producers against the K. pneumoniae WC indicator strain. A significant correlation was observed between klebocinogeny and enterotoxigenicity (p less than 0.05). Most strains (91 and 74% respectively) were inhibited by gentamicin and nalidixic acid. Most of the strains were multiply drug-resistant.  相似文献

The presence and persistence of enterotoxigenic Escherichia coli (ETEC) is poorly investigated in marine habitats. Here we compared ETEC with the more studied fecal contaminant, Salmonella enterica serotype Typhimurium ( S. enterica ) and the marine bacteria Vibrio parahaemolyticus . All three species of bacteria were culturable on agar plates during 8 weeks of incubation in seawater. However, the culturability of ETEC was positively affected by low temperature whereas V. parahaemolyticus was negatively affected. High-nutrient conditions favored the growth of ETEC but not the other bacteria. When the bacteria were fed to blue mussels, V. parahaemolyticus inhibited the filtration activity and the ingestion was lower than that of the enterobacteria. On the other hand, the mussels were less efficient in eliminating V. parahaemolyticus and an in vitro study showed that the hemocytes of three different species of bivalves were not able to kill this strain of V. parahaemolyticus . The bactericidal capacity of bivalves was seemingly an efficient elimination pathway for S. enterica and ETEC. This study showed that ETEC in endemic areas should, to the same degree as S. enterica and V. parahaemolyticus , be taken in consideration when assessing the role of marine environments as a source of enteric infection.  相似文献

AIM: Leucine aminopeptidase (LAP), an enzyme involved in the decomposition of natural peptides, was measured in different marine and brackish ecosystems, together with some environmental and microbiological parameters. METHODS AND RESULTS: The fluorogenic compound L-leucine-7-amido-4-methyl coumarin was specifically used for the determination of this in situ activity. The enzyme data obtained from this comparative study highlighted the strong spatial and temporal variability of the distribution of LAP in aquatic ecosystems, which was sometimes related to the course of environmental variables such as salinity and organic carbon content. CONCLUSIONS: LAP assay has proved to be a rapid method providing useful information on the microbial metabolic processes involved in the mineralization of organic matter. SIGNIFICANCE AND IMPACT OF THE STUDY: The determination of the potential rates of extracellular enzyme activity is of great ecological importance to extend knowledge on the role played by bacteria in aquatic biogeochemical cycles.  相似文献

Abstract The survival pattern and plasmid maintenance of Escherichia coli was examined in an artificial seawater microcosm. It was found that the three strains of E. coli (EK3C, H10407 and 34309) included in the study were able to maintain a portion of cells in the culturable phase for at least 3 years in artificial seawater. Along with retaining culturability, that portion of the cell population also maintained their indigenous plasmids over the 3-year period. It is concluded that cells of E. coli maintaining culturability in seawater are selectively adapted to the salinity of seawater, remaining in a culturable state. The results of the study are significant in that it has been assumed by many public health authorities that E. coli cannot survive, without nutrient addition, in seawater for long periods of time, i.e., years of exposure to seawater.  相似文献

Two rare wild-type strains of Pseudomonas aeruginosa were mixed in membrane diffusion chambers and then introduced into a natural freshwater environment for 72 h. The plasmid-containing strain (R serotype 15) and the plasmidless strain (H serotype 5) had initial bacterial densities of 2 x 10(5) cells per ml. Samples collected from the chambers were analyzed for viable and direct counts and for acquired-resistance frequencies. Suspected transconjugant-to-donor ratios ranged from 0.5 to 1.3; transfer percentages ranged from 13 to 70%. [3H]thymidine uptake indicated DNA synthesis in both strains as well as in transconjugants. These studies indicate that rare wild-type bacterial strains with large plasmid loads can survive as well as can bacteria with low plasmid loads when exposed to the in situ conditions of a tropical freshwater habitat. These results also suggest that genetic modification of indigenous microbiota through conjugation or transformation is feasible when rare wild-type strains or genetically engineered microorganisms are released in large numbers in tropical aquatic ecosystems.  相似文献

Abstract The possible fate in sea water of the two fish pathogenic bacteria Vibrio anguillarum and Vibrio salmonicida is discussed on the basis of microcosm experiments with these and other copiotrophic bacteria. Recent articles dealing with the survival of fish pathogens are reviewed, and the reported survival capacities are discussed in relation to ecological mechanisms, such as death, and predation, leading to the removal of bacteria from the water column.  相似文献
