A total of 102 strains of Clostridium botulinum (56 strains of type A, 43 type B, and 3 nontoxigenic strains which could not be typed) was examined for resistance to gamma rays. When these organisms were suspended in neutral phosphate buffer in concentrations of 104 spores per tube, the threshold sterilizing dose appeared to be 1.4 Mrad. Partial survival to 1.4 Mrad was shown by 10.7% of the type A strains, 18.6% of the type B strains, and one of three nontoxigenic strains. Over-all, type A strains indicated higher radioresistance than type B strains, although there was overlapping. Representatives of the most resistant strains had D values of 0.317 to 0.336 Mrad; the D values of an intermediate group were 0.224 to 0.253 Mrad, and the most sensitive strain studied, 51B, had a D value of 0.129 Mrad. The radioresistance of Putrefactive Anaerobe 3679, strain S-2, was comparable to the intermediate C. botulinum group (D = 0.209).  相似文献

S ummary . Spores of Bacillus cereus were treated with thioglycollic acid which ruptures at least 10–30% of the spore disulphide bonds by reducing them to thiol groups. The treated spores were still viable and were sensitive to lysozyme but remained as resistant to γ-irradiation and to heat as untreated spores. Neither treated nor untreated spores were sensitized to irradiation by reagents which block thiol groups. The results did not indicate that the high content of disulphide bonds in spore coat protein protects spores against inactivation by irradiation or heat.  相似文献

Dried preparations with Streptococcus faecium, strain A(2)1, and spores of Bacillus sphaericus, strain C(I)A, normally used for control of the microbiological efficiency of radiation sterilization plants and preparations with spores of Bacillus subtilis, normally used for control of sterilization by dry heat, formalin, and ethylene oxide, as well as similar preparations with Micrococcus radiodurans, strain R(1), and spores of Bacillus globigii (B. subtilis, var. niger) were gamma irradiated with dose rates from 16 to 70 krad/h at temperatures from 60 to 100 C. At 80 C the radiation response of the spore preparations was the same as at room temperature, whereas the radiation resistance of the preparations with the two vegetative strains was reduced. At 100 C the radiation response of preparations with spores of B. subtilis was unaffected by the high temperature, whereas at 16 and and 25 krad/h the radiation resistance of the radiation-resistant sporeformer B. sphaericus, strain C(I)A, was reduced to the level of radiation resistance of preparations with spores of B. subtilis. It is concluded that combinations of heat and gamma irradiation at the temperatures and dose rates tested may have very few practical applications in sterilization of medical equipment.  相似文献

SYNOPSIS. Stentor polymorphus was irradiated with 60Co gamma rays at 525 rads/minute to examine the effect on survival, cell division, oral membranellar frequency and oxygen uptake. Both survival and cell division were studied on single cells. The LD50 is 285 kilorads but cell division is inhibited below this dose; 110 kilorads doubles the 1st post-irradiation division interval, and a delay of 240 hours occurs at the maximum tolerated dose of 310 kilorads. Conjugating cells are more sensitive, with an LD50 of approximately 40 kilorads.
The frequency of cilia in the membranellar band, measured stroboscopically, is reduced by 30% after 14.5 kilorads, and stopped by higher doses. Recovery has a similar time scale to recovery of fission. A Clarke electrode was used to measure changes in oxygen uptake after irradiation. Depressions of up to 50% were found. Recovery followed a similar pattern to that of ciliary activity and cell division.  相似文献

Radiation exposure is a threat to public health because it causes many diseases, such as cancers and birth defects, due to genetic modification of cells. Compared with the past, a greater number of people are more frequently exposed to higher levels of radioactivity today, not least due to the increased use of diagnostic and therapeutic radiation-emitting devices. In this study, ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOF-MS)-based metabolic profiling was used to investigate radiation- induced metabolic changes in human fibroblasts. After exposure to 1 and 5 Gy of γ-radiation, the irradiated fibroblasts were harvested at 24, 48, and 72 h and subjected to global metabolite profiling analysis. Mass spectral peaks of cell extracts were analyzed by pattern recognition using principal component analysis (PCA) and partial least squares-discriminant analysis (PLS-DA). The results showed that the cells irradiated with 1 Gy returned to control levels at 72 h post radiation, whereas cells irradiated with 5 Gy were quite unlike the controls; therefore, cells irradiated with 1 Gy had recovered, whereas those irradiated with 5 Gy had not. Lipid and amino acid levels increased after the higher-level radiation, indicating degradation of membranes and proteins. These results suggest that MS-based metabolite profiling of γ-radiation-exposed human cells provides insight into the global metabolic alterations in these cells.  相似文献

The effects of γ-irradiation on wheat gluten were studied by means of gel filtration on Sephadex G-100, starch gel electrophoresis and analysis of amino acid composition. Analyses of gluten at a moisture content of 2% revealed no significant change in amino acid composition except for cystine which was decreased by about 8% with irradiation at 10 Mrad. Changes in the chromatogram from gel filtration were interpreted in terms of random depolymerization resulting from irradiation. The results of starch gel electrophoresis suggested that irradiation levels greater than 3 Mrad resulted in characteristic changes in the molecular configuration of gliadin components.  相似文献

Although safe doses of radiation have been determined, concerns about the harmful effects of low-dose radiation persist. In particular, to date, few studies have investigated the correlation between low-dose radiation and disease development. Asthma is a common chronic inflammatory airway disease that is recognized as a major public health problem. In this study, we evaluated the effects of low-dose-rate chronic irradiation on allergic asthma in a murine model. Mice were sensitized and airway-challenged with ovalbumin (OVA) and were exposed to continuous low-dose-rate irradiation (0.554 or 1.818 mGy/h) for 24 days after initial sensitization. The effects of chronic radiation on proinflammatory cytokines and the activity of matrix metalloproteinase-9 (MMP-9) were investigated. Exposure to low-dose-rate chronic irradiation significantly decreased the number of inflammatory cells, methylcholine responsiveness (PenH value), and the levels of OVA-specific immunoglobulin E, interleukin (IL)-4, and IL-5. Furthermore, airway inflammation and the mucus production in lung tissue were attenuated and elevated MMP-9 expression and activity induced by OVA challenge were significantly suppressed. These results indicate that low-dose-rate chronic irradiation suppresses allergic asthma induced by OVA challenge and does not exert any adverse effects on asthma development. Our findings can potentially provide toxicological guidance for the safe use of radiation and relieve the general anxiety about exposure to low-dose radiation.  相似文献

S ummary : Strains of Salmonella senftenberg isolated from Norwegian herring meal and strain 775W were exposed to gamma radiation from a 60Co source. When they were irradiated in phosphate buffered saline solution, the average D10 values for all the strains was 19·3 krad. On irradiating strains 56 and 775W in herring meal the D10 values were 192·1 and 188·5 krad, respectively, thus indicating that the suspending medium had a great effect on the radiation resistance of the organisms. Radiation doses of the order of 0·8–1·3 Mrad are recommended for the decontamination of herring meal.  相似文献

喹诺酮类是目前临床上应用较多的抗菌药。然而,喹诺酮类耐药菌时有出现。就细菌对喹诺酮类抗菌药主要耐药机制从分子水平作一综述。(1)一般认为,喹诺酮类抗菌药通过结合细菌Ⅱ类拓扑异构酶,干扰细菌复制,而发挥抑菌作用。Ⅱ类拓扑异构酶变异时,细菌可逃脱喹诺酮类的抑菌作用。高水平的耐药由DNA回旋酶和拓扑异构酶Ⅳ同时发生变异造成。(2)细菌细胞壁是抗菌药进入的屏障。细胞壁组分脂多糖和孔蛋白的改变,可减少喹诺酮类的通透。(3)有些细菌可利用“外排泵”主动将喹诺酮类排出,降低喹诺酮类在菌体内的积累浓度。(4)细菌的其他一些代谢因素也可影响喹诺酮类的抑菌作用。  相似文献

A considerable browning was observed especially in cortex tissue and along xylem of potato tubers harvested at Sakai in Osaka Prefecture, after irradiation with 10, 20 and 50 krad doses of cobalt-60 gamma rays. This phenomenon was accompanied by the marked increase in polyphenol content and peroxidase activity, and the transient increase in o-diphenol oxidase activity. Total reducing compounds in the tissue were also increased by gamma irradiation.

The browning phenomenon depended on the storage period from the harvest to gamma irradiation treatment. The browning and the transient increase in o-diphenol oxidase activity were completely suppressed in the case of tubers irradiated 3 months after harvest.

There was no significant change in α-amylase activity in all tubers tested.  相似文献

When the urediniospores of three races of Melampsora medusae, the causal agent of leaf rust of poplars, were exposed to Co60 gamma irradiation (range 100–1000 Gy), there were significant differences in the radiosensitivity of the races as assessed, in vitro, by incubation period to flecking and the uredinial survivability percentage. In race 5A the frequency of mutation to virulence on Populus deltoides cv. T-173, normally a resistant cultivar, was maximum (1.688 %) at an irradiation dose of 400 Gy. Importance of mutations as source of variability in pathosystems and usefullness of artificially induced mutations in the context of host-pathogen interactions are discussed.  相似文献

The free amino acids of potatoes irradiated with the doses of 7,000, 15,000 and 30,000 rad were determined by ion-exchange chromatography.

After 15 days storage following irradiation, it was shown that the concentration of asparatic acid, proline and aliphatic amino acids increased with increasing irradiation doses, while that of basic amino acids and glutamic acid especially decreased. However, after 105 days of storage, the similarity of the free amino acid content of irradiated potatoes to that of non-irradiated and non-stored potatoes was observed.

On the concentration of protein-bound amino acids, there were no significant differences between non-irradiated and 15,000 rad irradiated potatoes.  相似文献

An inoculated, irradiated beef pack (1,240 cans) study was conducted for the determination of microbiological safety for unrestricted human consumption. Each can contained a mixture of 106 spores of each of 10 strains of Clostridium botulinum (5 type A and 5 type B), or a total of 107 spores/can. The cans were irradiated to various doses (100 cans/dose) with 60Co gamma rays at -30 ± 10 C, incubated at 30 ± 2 C for 6 months, and examined for swelling, toxicity, and recoverable botulinal cells. The minimal experimental sterilizing dose based on nonswollen, nontoxic sterile cans was 2.2 < experimental sterilizing dose ≤ 2.6 Mrad. Using recoverable cells as the most stringent criterion of spoilage, and assuming the conventional simple exponential (without an initial shoulder) rate of spore kill, the “12D” dose was 3.7 Mrad when estimated on the basis of a mixture of 10 strains totaling 107 spores/can, and 4.3 Mrad if it is assumed that each can of beef contained 106 spores of a single most resistant strain and all of these spores were of identical resistances. However, an analysis of the data by extreme value statistics indicated with 90% confidence that the spore death rate was not a simple exponential but might be a shifted exponential (with an initial shoulder), Weibull, lognormal, or normal, with a “12D” equivalent of about 3.0 Mrad regardless of the initial spore density per can. There was an apparent antagonism between the irradiated type A and B strains in the cans. Some of the cans contained type B toxin but did not include type B viable cells. Other cans had a mixture of type A and B toxins, but a large number of these cans did not yield recoverable type B cells. However, type A viable cells could always be demonstrated in those cans containing type A toxin.  相似文献

The application of the radiopasteurization method to fermentation media consisting mainly of molasses was investigated. γ-Irradiation was found to have an excellent pasteurization effect on the fermentation media and at the same time to bring about an increase in the fermentation rate and yield of ethanol. Percent survivals in molasses decreased to ca. 70% by heating at 80°C for 30 min, to ca. 10% by irradiation with 3.0 × 105 rad and to ca. 1% by 6.0 × 105 rad. Irradiated mash was suitable for the medium of the “starter”, since the rate and the degree of the growth of Saccharomyces cerevisiae in irradiated mash did not differ from those of the growth in heat-pasteurized mash.

In the case of the molasses mash supplemented with nitrogen sources, the fermentation rate and yield of ethanol in irradiated mash were larger than those in heated mash. Besides, in the absence of nitrogen sources a 14% difference in fermentation yield was seen between the mash irradiated with 3.0 × 105 rad and the mash heated at 80°C. With the doses ranging from 1.0 × 105 to 9.5 × 105 rad, concentrations of total sugar and direct reducing sugar, pH, and optical density of molasses were little affected by irradiation.  相似文献
