Factor analysis was used to identify the processes responsible for the changes in water quality in a stratified wastewater reservoir. These reservoirs are well suited to study hypertrophic conditions. In the epilimnion, the main source of variablility was the external loading due to variations in wastewater inputs and water level (volume) of the reservoir. The second source was the seasonal climatic changes affecting photosynthesis. The third source was differences between night and day processes affecting REDOX potential. In the hypolimnion, the main source of variability was organic loading but due to both external loading (wastewater inputs) and internal loading (compounds released from the sediments). The second source was related to the input of chlorophyll into the hypolimnion, which were external (entry of chlorophyll rich inflow from a waste stabilization pond) and/or internal (input of chlorophyll from upper layers due to water column mixing).  相似文献

The ultrastructural ontogeny of Commelina benghalensis minor-vein elements was followed. The mature minor vein has a restricted number of elements: a sheath of six to eight mestome cells encloses one xylem vessel, three to five vascular parenchyma cells, a companion cell, a thin-walled protophloem sieve-tube member and a thick-walled metaphloem sieve-tube member. The protophloem sieve-tube member (diameter 4–5 m; wall thickness 0.12 m) and the companion cell originated from a common mother cell. The metaphloem sieve-tube member (diameter 3 m; wall thickness 0.2 m) developed from the same precursor cell as the phloem parenchyma cells. Counting the plasmodesmatal frequencies demonstrated a symplastic continuum from mesophyll to the minor-vein phloem. The metaphloem sievetube member and the phloem parenchyma cells are the termini of this symplast. The protophloem sieve-tube member and companion cell constitute an insulated symplastic domain. The symplastic route, mesophyll to metaphloem sieve tube, appears to offer a path for symplastic loading; the protophloem sieve tube may be capable of accumulation from the apoplast. A similar two-way system of loading may exist in a number of plant families. Plasmodesmograms (a novel way to depict cell elements, plasmodesmatal frequencies and vein architecture) of some other species also displayed the anatomical requirements for two routes from mesophyll to sieve tube and indicate the potential coexistence of symplastic and apoplastic loading.  相似文献

The nature of phloem loading of photosynthesis products – either symplastic or apoplastic – has been a matter of debate over the last two decades. This controversy was reconciled by proposing a multiprogrammed loading mechanism. Different modes of phloem loading were distinguished on the basis of the variety of plasmodesmatal connectivity between the minor vein elements. Physiological evidence for at least two phloem loading mechanisms as well as recent support for coincidence between plasmodesmatal connectivity and the loading mechanism is shortly reviewed. The present paper attempts to correlate the plasmodesmatal connectivity between sieve element/companion cell complex and the adjacent cells (the minor vein configuration) – and implicitly the associate phloem loading mechanisms – with different types of climate. The minor vein configuration is a family characteristic. This enables one to relate vein configuration with ecosystem using the family distribution over the globe. The uneven distribution of vein types between terrestrial ecosystems indicates that apoplastic phloem loading predominates in cold and dry climate zones. Projection of the minor vein configuration on the Takhtajan system of flowering plants suggests evolution from apoplastic to symplastic phloem loading. Accordingly, the distribution of minor vein configurations suggests that drought and temperature stress have led to the transformation of the ancient symplastic mode into the more advanced apoplastic mode of loading.  相似文献

The cotyledons of castor bean (Ricinus communis L.) act as absorption organs for amino acids, which are supplied to the medium. The analysis of the sieve-tube sap, which exudes from the cut hypocotyl, demonstrated the ability of the cotyledons to load particular amino acids into the phloem and to reject the loading of others. The sieve-tube sap of cotyledons, which were embedded in the endosperm, contained 150 mM amino acids, with 50 mM glutamine as the major amino acid, and 10–15 mM each of valine, isoleucine, lysine and arginine. Removal of the endosperm led to a drastic decline in the amino-acid content of sieve-tube sap down to 16 mM. Addition of single amino acid species to the medium increased the amino acid concentration in the sieve-tube sap in specific manner: glutamine caused the largest increase (up to 140 mM in exudate), glutamate and alanine smaller increases (up to 60 mM), and arginine the smallest. In addition, the amino acid composition of the sieve-tube sap changed, for instance, glutamine or alanine readily appeared in the sieve-tube sap upon incubation in glutamine or alanine, respectively, whereas glutamate was hardly discernible even in the case of incubation with glutamate; arginine was loaded into the sieve tubes only reluctantly. In general, glutamine and alanine accumulated four- to tenfold in the sieve tubes. The uptake of amino acids and of sucrose into the sieve tubes was interdependent: the loading of sucrose strongly reduced the amino acid concentration in the sieve-tube exudate and loading of amino acids decreased the sucrose concentration. Comparison of the concentrations of various amino acids on their way from the endosperm via the cotyledon-endosperm interface, through the cotyledons and into the sieve tubes showed that glutamine, valine, isoleucine and lysine are accumulated on this pathway, whereas glutamate and arginine are more concentrated in the cotyledons than in the sieve tubes. Obviously the phloem-loading system has a transport specificity different from that of the amino acid uptake system of the cotyledon in general and it strongly discriminates between amino acids within the cotyledons.  相似文献

Eau Galle Reservoir, Wisconsin, was treated with a hypolimnetic dose of aluminum sulfate (alum) in 1986 to diminish excessive phytoplankton production associated with high phosphorus loading from anoxic, profundal sediments. Prior to treatment, internal total phosphorus (TP) loading was 3 to 6 times greater than external TP loading during summer stratification. Periodic increases in epilimnetic TP mass and chlorophylla concentrations closely corresponded with elevated internal TP loading. For one year following treatment, internal TP loading and concentrations of soluble reactive phosphorus (SRP) in the hypolimnion were substantially reduced. However, abnormally high external TP loading during the stratified period of 1986 resulted in high TP mass and chlorophylla in the epilimnion. During the summers of 1987 and 1988, effects of alum treatment on internal TP loading were essentially negated, and epilimnetic TP mass and chlorophylla remained unchanged from pretreatment years. Multiple potential sources of P input to this reservoir make it difficult to reduce epilimnetic P and phytoplankton growth.  相似文献

A model predicting phosphorus and nitrogen concentrations from loading rates was tested using data collected from a shallow, nutrient-rich lake where both internal and external loading occurred. Predicted nutrient concentrations agreed closely with obaerved values and it is suggested that the model could be used to predict the reduction in loading rate required to effect lake restoration.  相似文献

蛋白质组技术难点之一是如何获取尽可能多的细胞或组织的蛋白信息.双向电泳蛋白斑点数目直接反映了实验蛋白质组信息的完整性.除样品制备外,蛋白上样方式对双向电泳图谱的质量和完整性有直接的影响.实验论文从以下3个方面考察不同的蛋白上样方式对双向电泳图谱的影响;即:水化上样与杯上样;一次上样与重复上样;以及酸性端加样与碱性端上样.实验结果发现;蛋白上样量较大时,杯上样方式的图谱斑点数目较水化上样方式明显增多;样品蛋白浓度较高时,稀释多次上样明显优于一次性浓缩蛋白上样;蛋白裂解液(MCF7乳腺癌细胞)在酸性端加样对偏碱性蛋白的分离未发现明显优势.相反,在等电聚焦伏小时(Vh) 足够的前提下,碱性端加样对偏碱性端蛋白反而有利,表现为斑点数目较多,而且等电点方向拖尾减轻.实验结果对提高双向电泳的质量以及相关蛋白质组信息的完整性提供了有益的技术参考.  相似文献

The extent to which sediments of aquatic systems immobilize or release phosphorus can affect dramatically the P content of overlying waters. Data from 48 different aquatic systems suggests that there may be a major difference between fresh- and salt-water systems in this immobilization. Under oxic conditions (water overlying sediments had dissolved oxygen > 0.5 mg/L) P is strongly immobilized in sediments of most fresh-water systems. In sediments of most salt-water systems P is released from sediments and behaves, essentially, as a conservative tracer of benthic decomposition. This difference in P cycling is large enough to have an influence on the often cited difference in phytoplankton nutrient limitation between fresh- and salt-water systems.  相似文献

People following arthroscopic partial medial meniscectomy (APM) are at increased risk of developing knee osteoarthritis. High impact loading and peak loading early in the stance phase of gait may play a role in the pathogenesis of knee osteoarthritis. This was a secondary analysis of longitudinal data to investigate loading-related indices at baseline in an APM group (3 months post-surgery) and a healthy control group, and again 2 years later (follow-up). At baseline, 82 participants with medial APM and 38 healthy controls were assessed, with 66 and 23 re-assessed at follow-up, respectively. Outcome measures included: (i) heel strike transient (HST) presence and magnitude, (ii) maximum loading rate, (iii) peak vertical force (Fz) during early stance. At baseline, maximum loading rate was lower in the operated leg (APM) and non-operated leg (non-APM leg) compared to controls (p≤0.03) and peak Fz was lower in the APM leg compared to non-APM leg (p≤0.01). Over 2 years, peak Fz increased in the APM leg compared to the non-APM leg and controls (p≤0.01). Following recent APM, people may adapt their gait to protect the operated knee from excessive loads, as evidenced by a lower maximum loading rate in the APM leg compared to controls, and a reduced peak Fz in the APM leg compared to the non-APM leg. No differences at follow-up may suggest an eventual return to more typical gait. However, the increase in peak Fz in the APM leg may be of concern for long-term joint health given the compromised function of the meniscus.  相似文献

Muscle activity reduces soft-tissue resonance at heel-strike during walking   总被引:1,自引:0,他引:1  
Muscle activity has previously been suggested to minimize soft-tissue resonance which occurs at heel-strike during walking and running. If this concept were true then the greatest vibration damping would occur when the input force was closest to the resonant frequency of the soft-tissues at heel-strike. However, this idea has not been tested. The purpose of this study was to test whether muscle activity in the lower extremity is used to damp soft-tissue resonance which occurs at heel-strike during walking. Hard and soft shoe conditions were tested in a randomized block design. Ground reaction forces, soft-tissue accelerations and myoelectric activity were measured during walking for 40 subjects. Soft-tissue mass was estimated from anthropologic measurements, allowing inertial forces in the soft-tissues to be calculated. The force transfer from the ground to the tissues was compared with changes in the muscle activity. The soft condition resulted in relative frequencies (input/tissue) to be closer to resonance for the main soft-tissue groups. However, no increase in force transmission was observed. Therefore, the vibration damping in the tissues must have increased. This increase concurred with increases in the muscle activity for the biceps femoris and lateral gastrocnemius. The evidence supports the proposal that muscle activity damps soft-tissue resonance at heel-strike. Muscles generate forces which act across the joints and, therefore, shoe design may be used to modify muscle activity and thus joint loading during walking and running.  相似文献

The purpose of this study was to quantify the load on the lumbar spine of subjects when they are asked to adjust from a slouched sitting posture into an upright posture with one of three different strategies: “free” (no instruction) and two coached patterns: “lumbopelvic” dominant and “thoracic” dominant. The activity of selected muscles and kinematic data was recorded from 20 volunteers while performing the three movement patterns to adjust sitting posture. Moments and forces at the lumbar spine were computed from an anatomically detailed model that uses kinematics and muscle activation as input variables.The lumbopelvic pattern produces less joint moment on the lumbar spine (on average 31.2 ± 3.9 N m) when compared to the thoracic pattern (43.8 ± 5.8 N m). However, the joint compression force was similar for these two patterns, but it was smaller in the free pattern, when no coaching was given (lumbopelvic: 1279 ± 112 N, thoracic: 1367 ± (125 N, free: 1181 ± 118 N). Lower thoracic erector muscle activity and higher lumbar erector activity were measured in the lumbopelvic pattern in comparison with the other two. In summary the lumbopelvic pattern strategy using predominantly the movement of anterior pelvic tilt results in smaller joint moments on the lumbar spine and also positions the lumbar spine closest to the neutral posture minimizing passive tissue stress. This may be the strategy of choice for people with low back flexion intolerance.  相似文献

Nutrient and chlorophyll concentrations in Lake Newnan (27 km2, mean depth 1.5 m), Florida showed dramatic increases from 1991 to 1998. Historical data showed Lake Newnan never had sufficient aquatic macrophyte abundance for a shift in alternate stable states to account for increases in trophic state characteristics. External phosphorus and nitrogen loads from incoming streams were monitored from August 1997 to July 1998 to determine if external supplies of nutrients were responsible for increases in lake nutrient and chlorophyll concentrations. During the study period, external nutrient loading rates were not correlated to lake nutrient concentrations. Phosphorus and nitrogen models based on the external loading estimates predicted the lake total phosphorus and total nitrogen concentrations to be 370% and 680% less, respectively, than the observed lake total phosphorus and total nitrogen mean concentrations. Consequently, phosphorus and nitrogen exports were 280% and 540% greater, respectively, than stream input loading. Data during the study period revealed strong inverse relations between lake stage and total phosphorus (r=–0.78), total nitrogen (r=–0.71), and chlorophyll (r=–0.90) concentrations. Long-term data (1965–1998) also revealed inverse correlations (r=–0.48 to –0.52) between lake stage and total phosphorus, total nitrogen, and chlorophyll concentrations. Applying fundamental wave theory and using a bathymetric map, it is probable that as much as 70% of the lake bottom sediment could be subjected to resuspension 50% of the time when the lake stage falls below 19.9 m mean sea level (msl). Above a lake stage of 19.90 m msl, less than 20% of the lake bottom sediment can potentially be resuspended 50% of the time. A percent frequency distribution from 1991 to 1998 showed that over 30% of the lake stages fell below 19.9 m msl. However, from 1967 to 1990, only 8% of the lake stage values fell below 19.9 m msl. Increases in total phosphorus, total nitrogen and chlorophyll concentrations in Lake Newnan were likely caused by an increased probability of internal loading due to decreased lake levels, and not to external loading of phosphorus and nitrogen.  相似文献

Abstract

The interventricular septum is the structure that separates the left and right ventricles of the heart. Under normal loading conditions, it is concave to the left ventricle, but under abnormal loading the septum flattens and occasionally inverts. In the past, the septum has frequently been modelled as integral to the left ventricle with the effects of pressure from the right ventricle being ignored. Under abnormal loading, the septum has been described as behaving equivalent to a “flapping sail”. There has been no consideration of structural behaviour under these conditions. A 2-D plane stress FE model of the septum was used to investigate the difference in structural behaviour of the septum during diastole between normal and abnormal loading. The biaxial stress patterns that develop are distinctively disparate. Under normal loading, the septum behaves much like a thick-walled cylinder subject to internal and external pressure, with the resulting stresses being circumferential tension and radial compression, both varying with radius. These stresses are very low throughout most of diastole. However, under abnormal loading, the septum behaves in an arch-like fashion, with high compressive stresses almost circumferential in direction, combined with radial compression. We conclude that right ventricular pressures cause bending effects in the wall of the heart, and that under abnormal loading, the compressive stresses that develop in the septum may lead to an understanding of certain, previously unexplained, pathological conditions.  相似文献

Biomechanical aspects of running injuries are often inferred from external loading measurements. However, previous research has suggested that relationships between external loading and potential injury-inducing internal loads can be complex and nonintuitive. Further, the loading response to training interventions can vary widely between subjects. In this study, we use a subject-specific computer simulation approach to estimate internal and external loading of the distal tibia during the impact phase for two runners when running in shoes with different midsole cushioning parameters. The results suggest that: (1) changes in tibial loading induced by footwear are not reflected by changes in ground reaction force (GRF) magnitudes; (2) the GRF loading rate is a better surrogate measure of tibial loading and stress fracture risk than the GRF magnitude; and (3) averaging results across groups may potentially mask differential responses to training interventions between individuals.  相似文献

A Comparison of the Sucrose Transporter Systems of Different Plant Species   总被引:5,自引:0,他引:5  
Abstract: The sucrose uptake behaviour of many different plant species is characterised by the presence of at least two components with distinct kinetic properties. These include at least one high-affinity and one low-affinity transport system. All known sucrose transporters from higher plants fall into one of three large subfamilies, according to phylogenetic analysis. Apparently, the largest subfamily, the SUT1 subfamily, exclusively consists of high-affinity sucrose transporters from dicotyledons, whereas none of the transporters from monocotyledonous plants groups within this subfamily. The other two subfamilies of sucrose transporter-like proteins are either low-affinity transporter or putative sucrose-sensing proteins. Most of the known sucrose transporters from monocotyledons are closely related to the SUT2 subfamily and include high-affinity transporters, suggesting a different evolutionary origin of dicotyledonous and monocotyledonous sucrose transporter gene families.  相似文献

To study the export of sugars from leaves and their long-distance transport, sucrose-proton/co-transporter activity of potato was inhibited by antisense repression of StSUT1 under control of either a ubiquitously active (CaMV 35S ) or a companion-cell-specific (rolC) promotor in transgenic plants. Transformants exhibiting reduced levels of the sucrose-transporter mRNA and showing a dramatic reduction in root and tuber growth, were chosen to investigate the ultrastructure of their source leaves. The transformants had a regular leaf anatomy with a single-layered palisade parenchyma, and bicollateral minor veins within the spongy parenchyma. Regardless of the promoter used, source leaves from transformants showed an altered leaf phenotype and a permanent accumulation of assimilates as indicated by the number and size of starch grains, and by the occurrence of lipid-storing oleosomes. Starch accumulated throughout the leaf: in epidermis, mesophyll and, to a smaller degree, in phloem parenchyma cells of minor veins. Oleosomes were observed equally in mesophyll and phloem parenchyma cells. Companion cells were not involved in lipid accmulation and their chloroplasts developed only small starch grains. The similarity of ultrastructural symptoms under both promotors corresponds to, rather than contradicts, the hypothesis that assimilates can move symplasmically from mesophyll, via the bundle sheath, up to the phloem. The microscopical symptoms of a constitutively high sugar level in the transformant leaves were compared with those in wild-type plants after cold-girdling of the petiole. Inhibition of sugar export, both by a reduction of sucrose carriers in the sieve element/companion cell complex (se/cc complex), or further downstream by cold-girdling, equally evokes the accumulation of assimilates in all leaf tissues up to the se/cc complex border. However, microscopy revealed that antisense inhibition of loading produces a persistently high sugar level throughout the leaf, while cold-girdling leads only to local patches containing high levels of sugar. Partial-thickness tears of the supraspinatus tendon frequently occur at its insertion on the greater tubercule of the humerus, causing pain and reduced strength and range of motion. The goal of this work was to quantify the loss of loading capacity due to tendon tears at the insertion area. A finite element model of the supraspinatus tendon was developed using in vivo magnetic resonance images data. The tendon was represented by an anisotropic hyperelastic constitutive law identified with experimental measurements. A failure criterion was proposed and calibrated with experimental data. A partial-thickness tear was gradually increased, starting from the deep articular-sided fibres. For different values of tendon tear thickness, the tendon was mechanically loaded up to failure. The numerical model predicted a loss in loading capacity of the tendon as the tear thickness progressed. Tendon failure was more likely when the tendon tear exceeded 20%. The predictions of the model were consistent with experimental studies. Partial-thickness tears below 40% tear are sufficiently stable to persist physiotherapeutic exercises. Above 60% tear surgery should be considered to restore shoulder strength.  相似文献

A large data set (n = 154) of phytoplankton production and biomass in relation to physico-chemical environmental factors was collected from 1979 to 1986 in a recently created sand-pit lake (Paris suburbs). These data are well suited to interpret the oligotrophication observed along the 8 years period, characterized by a regular decrease in chlorophyll (from 16 to 4 µg l-1 as annual averages).A model describing the ecological functioning of the lake has been established. Biological processes related to phyto-, bacterio- and zooplankton as well as sediment-water interactions, are described within several submodels. Most of the parameters involved were determined by in situ measurements in this or similar environments The model provides a good simulation of observed data and confirms that the reduction of nutrient loading, resulting from the diversion — in 1981 — of a sewer previously discharging into the lake, was responsible for the oligotrophication of the system. The model allows to explore the response of planktonic compartments accross a gradient of nutrient loading. The role of hydrology is also tested. The systematic run of the model with and without zooplankton leads to a better understanding of top-down control.  相似文献

Electromyographic (EMG) amplitude and mechanical tension are directly related during isometric contraction. Maximal voluntary isometric contractions are typically elicited through two different procedures; resisting a load, which is eccentric in nature, and contracting against an immovable object, which is concentric in nature. A wealth of literature exists indicating that EMG amplitude during concentric contractions is greater than that of eccentric contractions of the same magnitude. However, the effects of different methods to elicit isometric contraction on EMG amplitude have yet to be investigated. The purpose of this study was to compare EMG amplitudes under different loading configurations designed to elicit isometric muscle contraction. Twenty healthy volunteers (10 males and 10 females, age = 23 ± 2 yrs, height = 1.7 ± 0.09 m, mass = 69.9 + 16.8 kg) performed a maximal voluntary plantarflexion effort for which the vertical ground reaction force (GRFv) sampled from a force plate and surface EMG of the soleus were recorded. Participants then performed isometric plantarflexion at 20%, 30%, 40%, and 50% GRFvmax in a seated position, from a neutral ankle position, under two different counterbalanced isometric loading conditions (concentric and eccentric). For concentric loading conditions, the subject contracted against an immovable resistance to the specified %GRFv identified via visual and auditory feedback. For eccentric loading conditions, subjects contracted against an applied load placed on the distal anterior thigh that produced the specified %GRFv. This applied load had the tendency to force the ankle into dorsiflexion. Therefore, plantarflexion force, in an attempt to maintain the ankle in a neutral position, resisted lengthening of the plantarflexor musculature, thus representing eccentric loading during an isometric contraction. Mean EMG amplitude was compared across loading levels and types using a 2 (loading type: concentric, eccentric) × 4 (loading level: 20%, 30%, 40%, 50% GRFv) repeated-measures ANOVA. The main effect for loading level was significant (p = 0.007). However, the main effect for loading type, and the loading type × loading level interaction were non-significant (p > 0.05). The present findings provide evidence that isometric muscle contractions loaded in either concentric or eccentric manners elicit similar EMG amplitudes, and are therefore comparable in research settings.  相似文献

Populations of beef cattle represent a potential non-point source of environmental contamination for Cryptosporidium parvum if on-farm management practices fail to minimize transport from bovine manure to adjacent water sources. Characterizing this risk of contamination requires several parameters to be estimated, the most important being a valid and precise estimate of the oocyst loading rate per animal unit. The oocyst loading rate is defined in this study as the total number of oocysts excreted by a cohort of adult beef cows during a 24[emsp4 ]h period. We propose a methodology for estimating this parameter for low prevalent populations whereby the majority of individuals are test negative. Under specific degrees of confidence and at the population scale, this methodology generates estimates for maximal oocyst loading based on the sensitivity of the diagnostic test and the point prevalence and intensity of fecal shedding from a cross-sectional survey of the target population.Our cross-sectional survey on California beef cows generated a prevalence of infection of 1.1 % (6/557) and an intensity of oocyst shedding ranging from 219 to 5,491 oocysts/g, with a geometric mean of 835 oocysts/g from six positive cows. Negative binomial estimate of the percent recovery of the diagnostic assay was 0.235. Based on this percent recovery and using approximately 19.4[emsp4 ]mg of feces per assay, the DT90 of our assay, defined as the concentration of oocysts at which our diagnostic assay had a 90 % probability of detecting one or more oocysts in a sample, was 755 oocyst/g feces. At a 95 % confidence level, the estimated maximum number of oocysts being excreted in the feces of California beef cows ranged from 4.8 to 14.4 oocysts/g feces/cow, or 7.7×104 to 2.3×105 oocysts/beef cow/day.  相似文献
