On many Australian commercial pig farms, groups of growing pigs are mass-medicated through their drinking water with selected antimicrobials for short periods to manage herd health. However, delivery of medication in drinking water cannot be assumed to deliver an equal dose to all animals in a group. There is substantial between-animal variability in systemic exposure to an antimicrobial (i.e. the antimicrobial concentration in plasma), resulting in under-dosing or over-dosing of many pigs. Three sources of this between-animal variability during a water medication dosing event are differences in: (1) concentration of the active constituent of the antimicrobial product in water available to pigs at drinking appliances in each pen over time, (2) medicated water consumption patterns of pigs in each pen over time, and (3) pharmacokinetics (i.e. oral bioavailability, volume of distribution and clearance between pigs and within pigs over time). It is essential that factors operating on each farm that influence the range of systemic exposures of pigs to an antimicrobial are factored into antimicrobial administration regimens to reduce under-dosing and over-dosing.  相似文献

The emergence of pathogens resistant to antimicrobials has prompted political initiatives targeting a reduction in the use of veterinary antimicrobials in Denmark, especially for pigs. This study elucidates the tendency of pig farms with a significantly higher antimicrobial use to remain in clusters in certain geographical regions of Denmark. Animal Daily Doses/100 pigs/day were calculated for all three age groups of pigs (weaners, finishers and sows) for each quarter during 2012–13 in 6,143 commercial indoor pig producing farms. The data were split into four time periods of six months. Repeated spatial cluster analyses were performed to identify persistent clusters, i.e. areas included in a significant cluster throughout all four time periods. Antimicrobials prescribed for weaners did not result in any persistent clusters. In contrast, antimicrobial use in finishers clustered persistently in two areas (157 farms), while those issued for sows clustered in one area (51 farms). A multivariate analysis including data on antimicrobial use for weaners, finishers and sows as three separate outcomes resulted in three persistent clusters (551 farms). Compared to farms outside the clusters during this period, weaners, finishers and sows on farms within these clusters had 19%, 104% and 4% higher use of antimicrobials, respectively. Production type, farm type and farm size seemed to have some bearing on the clustering effect. Adding these factors as categorical covariates one at a time in the multivariate analysis reduced the persistent clusters by 24.3%, 30.5% and 34.1%, respectively.  相似文献

Because antimicrobial resistance in food-producing animals is a major public health concern, many countries have implemented antimicrobial monitoring systems at a national level. When designing a sampling scheme for antimicrobial resistance monitoring, it is necessary to consider both cost effectiveness and statistical plausibility. In this study, we examined how sampling scheme precision and sensitivity can vary with the number of animals sampled from each farm, while keeping the overall sample size constant to avoid additional sampling costs. Five sampling strategies were investigated. These employed 1, 2, 3, 4 or 6 animal samples per farm, with a total of 12 animals sampled in each strategy. A total of 1,500 Escherichia coli isolates from 300 fattening pigs on 30 farms were tested for resistance against 12 antimicrobials. The performance of each sampling strategy was evaluated by bootstrap resampling from the observational data. In the bootstrapping procedure, farms, animals, and isolates were selected randomly with replacement, and a total of 10,000 replications were conducted. For each antimicrobial, we observed that the standard deviation and 2.5–97.5 percentile interval of resistance prevalence were smallest in the sampling strategy that employed 1 animal per farm. The proportion of bootstrap samples that included at least 1 isolate with resistance was also evaluated as an indicator of the sensitivity of the sampling strategy to previously unidentified antimicrobial resistance. The proportion was greatest with 1 sample per farm and decreased with larger samples per farm. We concluded that when the total number of samples is pre-specified, the most precise and sensitive sampling strategy involves collecting 1 sample per farm.  相似文献

The behaviour of growing pigs housed on two different commercial farms was studied to analyse daily rhythms related to behaviour. During the hot summer in Castilla La Mancha (Spain), 25 pigs were used, 10 from the first farm and 15 from the second one, so as to be able to work with the same stocking density (0.75 m2/pig). Their behaviours were recorded for 16 days in two separate periods with a video camera and subsequently observed for studying one photogram every 10 minutes (2035 observations for each farm), in which six predominant behaviours were assessed. The existence of a periodic pattern in most of the behaviours was verified with a mathematical model, demonstrating substantial differences between the models obtained for each farm.  相似文献

The between- and within-herd variability of porcine reproductive and respiratory syndrome virus (PRRSV) antibodies were investigated in a cross-sectional study of 103 British pig herds conducted 2003–2004. Fifty pigs from each farm were tested for anti-PRRSV antibodies using ELISA. A binomial logistic model was used to investigate management risks for farms with and without pigs with PRRSV antibodies and multilevel statistical models were used to investigate variability in pigs' log ELISA IRPC (relative index × 100) in positive herds.  相似文献

The objectives of this study were to compare and characterize the prevalence of antimicrobial-resistant (AR) Campylobacter in conventional and antimicrobial-free (ABF) production systems on farms, at slaughter, and in the environment. Fecal and environmental samples were collected from ABF farms (pigs, 1,239; environment, 797) and conventional farms (pigs, 1,650; environment, 1,325). At slaughter, we collected samples from carcasses, including postevisceration swabs, postchill swabs, and mesenteric lymph nodes from ABF systems (postevisceration swabs, 182; postchill swabs, 199; mesenteric lymph nodes, 184) and conventional systems (postevisceration swabs, 272; postchill swabs, 271; mesenteric lymph nodes, 255) at separate processing facilities. We also sampled the processing plant environment, including truck and lairage floor swab samples (ABF, 115; conventional, 90). Overall, a total of 2,908 Campylobacter isolates, including Campylobacter coli (farm, 2,557, 99.8%; slaughter, 341, 98.3%) and Campylobacter jejuni (farm, 4, 0.2%; slaughter, 6, 1.7%), were isolated in the study. There was no significant difference in the prevalence of Campylobacter between ABF and conventionally raised pigs (farrowing, P = 0.20; nursery, P = 0.06; finishing, P = 0.24) and the environment (P = 0.37). At slaughter, Campylobacter was isolated from all of the stages, including postchill. The highest frequencies of resistance were exhibited against tetracycline (ABF, 48.2%; conventional, 88.3%). Ciprofloxacin-resistant C. coli isolates were observed in conventionally raised (17.1%) and ABF (1.2%) pigs (P = 0.11). Antimicrobial use data from conventional farms indicated significant associations between oxytetracycline use and tetracycline resistance in the nursery pigs (P = 0.01), between tiamulin exposure and azithromycin and erythromycin resistance in nursery (P < 0.01) and finishing (P < 0.01) pigs, and between enrofloxacin exposure and ciprofloxacin and nalidixic acid resistance in farrowing (P < 0.01) and nursery (P < 0.01) pigs. Identical antimicrobial resistance profiles were observed in the pigs and their environments on farms and at slaughter. In summary, our results highlight the persistence and dissemination of AR Campylobacter from farm to slaughter in ABF and conventionally raised pigs and their environments.  相似文献

Monitoring animal welfare (AW) in pig farms requires both proper indicators and a feasible approach. Animal-based measures (ABMs) are well-established AW indicators. Furthermore, AW screening at the slaughterhouses could be useful for identifying problems on farm. The aim of this study was to evaluate ABMs at the slaughterhouse and, when possible, to compare these ABMs with those collected on the farm. The study was carried out in northern Italy in a commercial abattoir and in a sample of farms. Animal-based measures were recorded on pigs from 62 batches of 54 farms, during ante-mortem (n=10 085 pigs) and post-mortem (n=7952 pigs) inspections. Sixteen of 54 farms were selected to compare ABMs collected at the slaughterhouse with ABMs collected on the farm. Overall, 2295 pigs (mean pigs examined per farm 119±45) were inspected at the slaughterhouse (group S) and 420 pigs (mean pigs per farm 26±5) on the farm (group F). Non-animal-based measures were also collected at the 16 farms. Differences between groups S and F, at the animal level, were assessed by a two-tailed paired Wilcoxon–Mann–Whitney test. Differences at the site of observation level (farm and slaughterhouse) were assessed by Fisher’s exact test using a hierarchical log-linear modelling for contingency tables. The most frequent ABMs at the slaughterhouse were manure on the body (47.7%), followed by dermatitis (28.0%), white spot (25.4%) and bursitis (24.7%). Recording ABMs at the slaughterhouse and on the farm usually yielded similar results; however, there were some exceptions. In particular, significant differences were found for non-uniformity of size (P<0.05) and dermatitis (P<0.001), which were higher at the slaughterhouse than on the farm. Results of log-linear modelling underlined the effect of the farm of origin on the percentage of pigs with bursitis, manure on the body and ear injuries that were observed at the slaughterhouse. In group S, significant associations between manure on the body and insufficient presence of clean and dry areas in the corresponding farm were found (P<0.05). Although these results should be interpreted with care due to the limited sample of farms, the slaughterhouse could be a feasible site of observation of ABMs, which could then be integrated in monitoring of AW on farm. Considering the number of slaughtered batches per farm, it would be possible to repeat assessments several times throughout the year for each farm, which could help provide an index for the continuous monitoring of AW.  相似文献

Recent developments in cattle tick control have incorporated the use of recombinant Bm86 vaccines against this ectoparasite. The vaccine developed by our group (Gavac) contains an antigen expressed in Pichia pastoris, and has been successfully employed for the control of tick infestations and transmission of tick-borne diseases. Here, we examined the safety and effect of the Gavac vaccine on reproductive parameters in cattle. Toxicity tests in mice and guinea pigs demonstrated the safety of Gavac. To study the adverse effects of vaccination on reproduction, a field trial involving 9,500 animals in Cuba was conducted. The cattle at 3 farms were vaccinated while those on a fourth farm were left unvaccinated and served as the control. Following vaccination, the control of tick infestation and the transmission of babesiosis were used to demonstrate the efficacy of the vaccine. No adverse effects were observed in any of the reproductive parameters studied when comparing the data before and after vaccination with Gavac and between the vaccinated farms and the control farm. These results demonstrate that under the conditions of our study vaccination with Gavac is safe for use on cattle.  相似文献

Aims: Yersinia enterocolitica 4/O:3 isolates of slaughter pigs originating from different farms were characterized to study the distribution of different genotypes at farm. A correlation between the genotypes and the resistance patterns was also examined. Methods and Results: Hundred and eighty‐seven ail‐positive Y. enterocolitica 4/O:3 isolates recovered from pigs originating from 31 Bavarian farms in 2000, 2003 and 2004 were characterized. PFGE using NotI, ApaI and XhoI enzymes revealed 31 genotypes. The most common genotype was found in 13% of the pigs. From most farms (71%), only one genotype was found. Some genotypes were found during different years. Low resistance was noted to streptomycin (9%), sulphamethoxazole (9%), amoxicillin/clavulanic acid (5%) and tetracycline (1%) by agar disc diffusion method. Conclusions: Several genotypes were found. Some genotypes were widely distributed and persisted for years. Farm‐specific genotypes may exist. No clear relation between the genotypes and antimicrobial patterns was found. Significance and Impact of the Study: This study provides data on the genetic diversity of Bavarian pig strains and antimicrobial resistance. It may be of interest for other countries where Y. enterocolitica strains are genotyped to get more information about the strain distribution of this pathogen.  相似文献

A repeated cross-sectional study was conducted to determine the patterns of antimicrobial resistance in 1,286 Escherichia coli strains isolated from human septage, wildlife, domestic animals, farm environments, and surface water in the Red Cedar watershed in Michigan. Isolation and identification of E. coli were done by using enrichment media, selective media, and biochemical tests. Antimicrobial susceptibility testing by the disk diffusion method was conducted for neomycin, gentamicin, streptomycin, chloramphenicol, ofloxacin, trimethoprim-sulfamethoxazole, tetracycline, ampicillin, nalidixic acid, nitrofurantoin, cephalothin, and sulfisoxazole. Resistance to at least one antimicrobial agent was demonstrated in isolates from livestock, companion animals, human septage, wildlife, and surface water. In general, E. coli isolates from domestic species showed resistance to the largest number of antimicrobial agents compared to isolates from human septage, wildlife, and surface water. The agents to which resistance was demonstrated most frequently were tetracycline, cephalothin, sulfisoxazole, and streptomycin. There were similarities in the patterns of resistance in fecal samples and farm environment samples by animal, and the levels of cephalothin-resistant isolates were higher in farm environment samples than in fecal samples. Multidrug resistance was seen in a variety of sources, and the highest levels of multidrug-resistant E. coli were observed for swine fecal samples. The fact that water sample isolates were resistant only to cephalothin may suggest that the resistance patterns for farm environment samples may be more representative of the risk of contamination of surface waters with antimicrobial agent-resistant bacteria.  相似文献

The features and the constitution of the microbial population of fresh feces were compared between pigs fed a diet supplemented with dietary microbes and pigs given nonsupplemented diets. The former were reared on farm C and the latter on farms A and B. The concentrations of ammonia-N, indole, and skatole of fresh feces were not significantly different between pigs reared on farm C and those raised on farms A and B, but the concentrations of ammonia-N and the skatole of fresh feces were significantly different between pigs reared on farms A and B. The total VFA (volatile fatty acids) concentration of fresh feces in pigs on farm C was slightly lower than in those on farms A and B. Moreover, the molar proportion of the acetic acid in feces in pigs on farm C was lower; inversely, that of propionic and butyric acids was higher in comparison with those on farms A and B. No differences were evident in the total viable counts of feces among pigs reared on the three different farms. Clostridium perfringens was abundant in feces of pigs raised on farms A and B, but it was not detected in pigs reared on farm C. Megasphaerae, bifidobacteria, and clostridia except for C. perfringens were more abundant in the feces of pigs fed a diet supplemented with dietary microbes on farm C, compared with pigs given the nonsupplemented diets on farms A and B.  相似文献

The aims of this study were to determine the ability of amplified fragment length polymorphism (AFLP) to differentiate Salmonella isolates from different units of swine production and to demonstrate the relatedness of Salmonella between farms and abattoirs by AFLP. Twenty-four farms in the midwestern United States were visited four times from 2006 to 2009. At each farm or abattoir visit, 30 fecal samples or 30 mesenteric lymph nodes were collected, respectively. A total of 220 Salmonella isolates were obtained, serotyped, and genotyped by multilocus sequence typing (MLST) and AFLP. These 220 isolates clustered into 21 serotypes, 18 MLST types, and 14 predominant AFLP clusters based on a genetic similarity threshold level of 60%. To assess genetic differentiation between farms, harvest cohorts, and pigs, analysis of molecular variance was conducted using AFLP data. The results showed 65.62% of overall genetic variation was attributed to variance among pigs, 27.21% to farms, and 7.17% to harvest cohorts. Variance components at the farm (P = 0.003) and pig (P = 0.001) levels were significant, but not at the harvest cohort level (P = 0.079). A second analysis, a permutation test using AFLP data, indicated that on-farm and at-abattoir Salmonella from pigs of the same farms were more related than from different farms. Therefore, among the three subtyping methods, serotyping, MLST, and AFLP, AFLP was the method that was able to differentiate among Salmonella isolates from different farms and link contamination at the abattoir to the farm of origin.  相似文献

Japan depends heavily on imports for its food supply. Since 2000, the food self‐sufficiency ratio has remained approximately 40% on a caloric basis. Japanese food wastage (i.e., food losses and food waste) is estimated to have been 6.42 million tonnes (50 kg per capita of wastage) in 2012. These values indicate that food wastage leads to wasted natural resources and excessive greenhouse gas (GHG) emissions both in Japan and in countries that export to Japan. This study estimates Japanese food wastage by food item to evaluate impacts on land and water resources and global GHG emissions during the processing, distribution, and consumption phases of the food supply chain while also considering the feed crops needed for livestock production. Despite uncertainties due to data limitations, in 2012, 1.23 million hectares of harvested land were used to produce food that was eventually wasted, and 413 million m3 of water resources were wasted due to Japanese food wastage in agricultural production. Furthermore, unnecessary GHG emissions were 3.51 million tonnes of CO2 eq. in agricultural production and 0.49 million tonnes of CO2 eq. in international transportation. The outcomes of the present study can be used to develop countermeasures to food wastage in industrializing Asian countries where food imports are projected to increase and food wastage issues in the consumption stage are expected to become as serious as they currently are in Japan.  相似文献

This study was conducted in a rural region where there are conventional and organic farms, the agricultural production includes more than 20 million people, and the effect on environmental quality is still poorly known in terms of indicators. Our objectives were: (1) compare soils attributes to reference areas, (2) verifying if cultivated areas under different farm systems presented differences in the soils attributes, (3) evaluate the attributes of quality water of watersheds and comparing the results with limiting values established by environmental legislation, and (4) analyze the values considering three criterion: watersheds, climatic season, and region of the landscapes. The study was conducted in two rural watersheds that have similar biophysical features and located in the Ibiúna municipality, São Paulo State, Brazil. However, one watershed encompasses farms where landowners largely use conventional agricultural systems. In the other watershed approximately 25% of the farms there are using an organic farm system. In the two watersheds soil samples were collected in sites covered with natural forest and in sites with agriculture (one watershed being organic and other being conventional). The attributes analyzed were soil bulk density (BD), concentrations of Carbon (C) and Nitrogen (N), C:N ratio, C Management Index, and the abundance of 13C and 15N in the soil organic matter. Water attributes were analyzed onsite or in laboratory after analysis of samples. Analyses included: air and water temperature, pH, dissolved oxygen, salinity, total of dissolved solids, total solids, electric conductivity, turbidity, total chloride, nitrate, total phosphorus and potassium. Regarding the soil attributes our database revealed that (1) the soils from cultivated sites of both watersheds presented significant differences from their respective forested areas, (2) Soil attributes are of equal quality in both farm systems. Concerning water attributes: (1) almost all attributes presented values better than the limiting values stipulated by Brazilian legislation; (2) the watersheds did not present significant differences of most of the attributes; (3) in the criteria climatic season data showed some significant differences. The data showed that the soils from the areas used for agricultural ends present belief that significantly worse soil quality in comparison to soils from sites still covered with natural forest. Neither the land cover nor farming system are altering the superficial water quality of the studied watershed and this appears to be related to the extensive percentage of natural remaining vegetation that still exists in both watersheds. The seasonality is an important force that drives the quality characteristics of the water. We highlight that the principles of organic agriculture should be practiced more efficiently and influences such as deforestation should be rigorously avoided.  相似文献

This study's objective was to determine the prevalence of Salmonella spp. in pigs and their farm environments in Korea, and to investigate the relationship between the strains based on their phenotypic and genotypic characteristics. A total of 36 Salmonella spp. were isolated in this study: 18 isolates from 492 pigs (3.7%) and 18 isolates from 418 (4.3%) farmhouse environmental samples from 16 different pig farms. Of the Salmonella strains isolated from the numerous environmental samples, the highest prevalence was observed in slurry or manure, followed by partitions, farmer's hands, floors, water/ nipples, ventilation sources, and feed, respectively. All the Salmonella isolates originating from different farms were genetically distinct. In three farms, however, identical phage types and pulse-field gel electrophoresis patterns were observed among Salmonella isolates from pig feces and environmental samples. This study suggests that environments contaminated with Salmonella could pose an infection risk to pigs on pig farms.  相似文献

To be able to analyze the relationship between the level of resistance and the use of antimicrobials, it is necessary to collect detailed data on antimicrobial usage. For this reason, data on antimicrobial use on 495 pig farms from entire Germany were collected and analyzed. In Germany, each application and dispensing of medicines to food-producing animals is documented in detail obligatorily by the veterinarian. This information was collected retrospectively for the year 2011. The analyses undertook separate examinations on the age groups sow, piglet, weaner and fattening pig; both the route of administration and indication per active ingredient, and active ingredient class, were evaluated. In total, 20,374 kg of antimicrobial substances were used in the study population. Tetracyclines were used in highest amounts, followed by beta-lactams, trimethoprim-sulfonamides and macrolides. Concerning the frequency of using an active substance per animal, polypeptides were most commonly administered. In all age groups, respiratory infections were the main indication for using antimicrobials, followed by intestinal diseases in piglets, weaners and fattening pigs and diseases of reproductive organs in sows. Over a period of 100 days, the median number of treatment days with one antimicrobial substance for piglets was 15 days, for weaners about 6 days, for fattening pigs about 4 days and for sows about 1 day. A multifactorial ANOVA was conducted to investigate which factors are associated with the treatment frequency. The factors “veterinarian” and “age group” were related to the treatment frequency, just as the interaction between “veterinarian” and “farm size” as well as the interaction between “veterinarian” and “age group”.  相似文献
