B. licheniformis MML2501 which was isolated from groundnut rhizosphere soil showed increased populations on spermozphere colonisation and significantly increased the seed germination and other growth parameters in groundnut under in vitro conditions. B. licheniformis MML2501 did not solubilise phosphate but produced indole acetic acid (IAA), with a maximum of 23 μg/ml under optimised conditions such as pH 7.0, temperature 35°C, tryphtophan at a concentration of 16 mM and at 200 rpm shaken conditions. The production of IAA by B. licheniformis MML2501 was further confirmed by TLC and HPLC analyses, in which the Rf value and retention time of 0.66 and 4.1 min respectively, match with that of the authentic IAA. Seed treatment of B. licheniformis MML2501 in groundnut showed a significant increase in seed germination, other growth parameters and yield parameters under potted plant experiments.  相似文献

Root rot caused by Rhizoctonia bataticola is a serious threat in cotton. Field experiments were conducted to study the influences of intercropping system in cotton with inorganic fertilizer and two bioinoculants (Azospirillum and Pseudomonas) on root rot incidence and yield of cotton. The results revealed that among the intercropping systems, cotton intercropping with Sesbania aculeata (1 : 1 ratio) recorded the highest rhizosphere colonization of Pseudomonas fluorescens in the year 2007 and 2008 and the lowest root rot incidence of 1.40, 2.49 and 3.90; 1.02, 2.22 and 5.98% at the vegetative, flowering and maturity stages in the year 2007 and 2008, respectively. From nutrient management practices, integration of Azospirillum and Pseudomonas with 50% recommended dose of NPK recorded the highest rhizosphere colonization of P. fluorescens in both years and the lowest root rot incidence of 1.40, 2.32 and 3.36; 1.07, 2.01 and 5.25% at vegetative, flowering and maturity stages in 2007 and 2008, respectively. Cotton + S. aculeata recorded the maximum number of sympodial branches (23.5 and 20.62/plant in 2007 and 2008, respectively) and the highest seed cotton yield of 2010 and 1894 kg/ha. The highest cotton equivalent yield (CEY) of 2052 and 1895 kg/ha was recorded in cotton + onion system, which was closely followed by cotton + S. aculeata system that had the CEY of 2010 and 1894 kg/ha in 2007 and 2008, respectively. The increased CEY is due to increased cost of onion compared with S. aculeata. Combined application of 100% recommended dose of NPK and bioinoculants recorded the seed cotton yield of 2227 and 1983 kg/ha and CEY of 2460 and 2190 kg/ha in 2007 and 2008, respectively. The lowest root rot incidence and increased yield in cotton + S. aculeata combined with 50% NPK and bioinoculants could be due to synergistic effect among the bioinoculants and S. aculeata.  相似文献

We are developing a collection of Bacillus strains, isolated from different environments, for use in controlling Sclerotinia sclerotiorum on oilseed rape in China and elsewhere. Strain BY-2, isolated from internal tissues of an oilseed rape root, was demonstrated to be Bacillus subtilis based on biochemical and morphological characteristics and on 16S RNA gene sequence. Photographic evidence from gnotobiotic studies using the lacZ-tagged strain BY-2(pUC18) confirmed that this strain was capable of colonizing internal root tissues. Strain BY-2 did not effectively colonize the ectorhizosphere or the surface of the stems or leaves of oilseed rape when applied in pellet or wrap seed treatment formulations. Populations of BY-2 dropped from 108 CFU seed−1 to 104 CFU g root−1 and ⩽102 CFU g stem−1 or leaf−1 after 60 days. Strain BY-2 was applied as a pellet seed treatment formulation alone, as a spray at flowering alone, and as the pellet seed treatment formulation combined with the spray application in two field trials at the Wuxue location conducted in two consecutive years. These three treatments containing BY-2 provided disease control (disease incidence) and mean seed yield that was similar to the chemical control treatment and significantly greater than the pellet without bacteria and non-treated control treatments. All three of these BY-2 treatments were similar to each other with regard to these two metrics and to treatments containing B. subtilis Tu-100, a genetically distinct strain previously shown to be effective against this disease. In two additional field trials, conducted in consecutive years at the Wuhan location, strain BY-2 applied as a wrap seed treatment formulation alone, as a spray at flowering alone, and as the wrap seed treatment combined with the spray application provided disease control (disease incidence) and mean seed yield that was similar to the chemical control treatment. These three BY-2 treatments also were significantly greater than the non-treated control treatments and compared favorably to treatments containing Tu-100. There was no evidence of BY-2 promoting growth of oilseed rape when applied in the pellet or wrap seed treatment formulations in field trials conducted at the Wuxue or at the Wuhan locations. We now have three Bacillus strains (B. subtilis strains BY-2 and Tu-100, B. megaterium A6) that control S. sclerotiorum on oilseed rape in the field that can be tested in strain combinations for enhanced disease control. We also have multiple methods for application of Bacillus strains as both seed treatment and foliar applications were effective.  相似文献

Balloon flower (Platycodon grandiflorum) is widely cultivated vegetable and used as a remedy for asthma in East Asia. Experiments were conducted to isolate endophytic bacteria from 1-, 3-, and 6-year-old balloon flower roots and to analyze the enzymatic, antifungal, and anti-human pathogenic activities of the potential endophytic biocontrol agents obtained. Total 120 bacterial colonies were isolated from the interior of all balloon flower roots samples. Phylogenetic analysis based on 16S rRNA gene sequences showed that the population of ‘low G + C gram-positive bacteria’ (LGCGPB) gradually increased 60.0–80.0% from 1 to 6 years balloon flower sample. On the other hand, maximum hydrolytic enzyme activity showing endophytic bacteria was under LGCGPB, among the bacterial strains, Bacillus sp. (BF1-1 and BF3-8), Bacillus sp. (BF1-2 and BF3-5), and Bacillus sp. (BF1-3, BF3-6, and BF6-4) showed maximum enzyme activities. Besides, Bacillus licheniformis (BF3-5 and BF6-6) and Bacillus pumilus (BF6-1) showed maximum antifungal activity against Phytophthora capsici, Fusarium oxysporum, Rhizoctonia solani, and Pythium ultimum. Moreover, Bacillus licheniformis was found in 3 and 6 years balloon flower roots, but Bacillus pumilus was found only in 6 years sample. It is presumed that older balloon flower plants invite more potential antifungal endophytes for there protection from plant diseases. In addition, Bacillus sp. (BF1-2 and BF3-5) showed maximum anti-human pathogenic activity. So, plant age is presumed to influence diversity of balloon flower endophytic bacteria.  相似文献

In order to obtain high productivity for a cotton crop, one of the major requirements is to establish an adequate plant population. The use of good-quality seed may ultimately be the best approach to attain this goal problem. The objective of this research was to study the effect of N-fertilization (at rates of 95.2 and 142.8 kg of N ha−1), foliar application of K (at rates of 0, 0.38, 0.77, 1.15 kg of K2O ha−1, applied twice during square initiation and boll development stages) and the plant growth retardant (PGR), mepiquat chloride (applied twice, 75 days after planting at 0.0 [control] and 0.048 kg a.i. ha−1, and 90 days after planting at 0.0 [control] and 0.024 kg a.i. ha−1), on seed yield, viability, and seedling vigor of Egyptian cotton (Gossypium barbadense cv. Giza 86). A field experiment was conducted at the Agricultural Research Center, Giza, Egypt in two growing seasons. Growth, mineral uptake, seed yield per plant and per ha, seed weight, seed viability, seedling vigor and cool germination test performance were all found to increase significantly due to the addition of the high N-rate, the foliar application of three potassium concentrations, and the PGR mepiquat chloride. The N and K rates as well as application of mepiquat chloride had no significant effect on the germination rate index in both seasons. Under the conditions of this study, applying N at a rate of 142.8 kg ha−1 combined with spraying cotton plants with K2O at 1.15 kg ha−1 and with mepiquat chloride at 0.048 + 0.024 kg ha−1 were found to improve seed yield as well as seed viability and seedling vigor in the next season.  相似文献

This study is concerned with the co-production of alkaline proteases and thermostable α-amylase by some feather-degrading Bacillus strains: B. mojavensis A21, B. licheniformis NH1, B. subtilis A26, B. amyloliquefaciens An6 and B. pumilus A1. All strains produced both enzymes, except B. pumilus A1, which did not exhibit amylolytic activity. The best enzyme co-production was obtained by the NH1 strain when chicken feathers were used as nitrogen and carbon sources in the fermentation medium. The higher co-production of both enzymes by B. licheniformis NH1 strain was achieved in the presence of 7.5 g/l chicken feathers and 1 g/l yeast extract. Strong catabolic repression on protease and α-amylase production was observed with glucose. Addition of 0.5% glucose to the feather medium suppressed enzyme production by B. licheniformis NH1. The growth of B. licheniformis NH1 using chicken feathers as nitrogen and carbon sources resulted in its complete degradation after 24 h of incubation at 37°C. However, maximum protease and amylase activities were attained after 30 h and 48 h, respectively. Proteolytic activity profiles of NH1 enzymatic preparation grown on chicken feather or casein-based medium are different. As far as we know, this is the first contribution towards the co-production of α-amylase and proteases using keratinous waste. Strain NH1 shows potential use for biotechnological processes involving keratin hydrolysis and industrial α-amylase and proteases co-production. Thus, the utilization of chicken feathers may result in a cost-effective process suitable for large-scale production.  相似文献

Bacillus strains are broadly studied for their beneficial role in plant growth and biological control of plant disease and pest; however, little is known about their underlying mechanisms. In this study, we assessed the controlling and defence‐related mechanisms of three Bacillus strains including rice seed‐associated strain B. subtilis A15, rhizobacterial strains B. amyloliquefaciens D29 and B. methylotrophicus H8, all of which are against bacterial leaf blight (BLB) caused by Xanthomonas oryzae pv. oryzae. Results indicated that all three strains showed strong biofilm formation ability. The culture filtrates of each strain significantly suppressed the growth and biofilm formation of X. oryzae, while changes in bacterial cell morphology such as cell swell and severe cell wall alterations were observed through the transmission electron microscopy images. PCR analysis revealed that all three strains harbour the antimicrobial‐associated genes that are responsible for biosynthesis of bacillomycin, fengycin, iturin and surfactin. Subsequent real‐time qPCR analysis revealed the upregulated expression of fenD and srfAA genes in D29 and H8, and fenD and ituC genes in A15 during their in vitro interaction with X. oryzae. It suggests that the antibacterial mechanisms of the three strains may be at least partially associated with their ability to secrete corresponding lipopeptides. Interestingly, the applications of the three strains in greenhouse conditions were found to be effective in controlling the BLB disease, which was achieved through the activation of inducing systemic resistance resulted from the enhanced activities of defence‐related enzymes. This is the first report of demonstration of the mode of antibacterial effect of Bacillus strains against X. oryzae. Overall, data from the current study provide valuable information for biological control of BLB disease in rice.  相似文献

Multispecies cropping systems contribute to sustainable agriculture with multiple ecosystem services. Effects of intercropping of various crops with faba beans on growth and yield parameters and disease severity of root rot, damping off and broomrape were investigated. This study was implemented in the laboratory, greenhouse and field to investigate the effect of the intercropping systems (fenugreek + faba bean, lupine + faba bean, garlic + faba bean and sole faba bean). The intercropping systems were combined with the application of arbuscular mycorrhiza fungi (AMF) and yeast as bio-control agents, compared to chemical application of herbicides (Glyphosate) and fungicides (Rizolex-T50), to control rot root diseases and broomrape weeds, Orobanche spp., of faba bean plants in vivo and under the naturally infested field. In vitro, yeast and Rizolex-T50 significantly inhibited mycelial growth of root pathogenic fungi. Intercropping with garlic and/or application of Rizolex-T, significantly decreased the incidence and disease index of root rot and damping-off diseases, meanwhile increased percentage of survival plants. In vivo, intercropping with fenugreek and/or application of Glyphosate, significantly reduced the number/weight of spikes/plot of broomrapes. Intercropping with fenugreek combined with AMF application promoted crop growth and significantly increased yield components. The AMF enhanced seed yield/ha when applied to the intercropping of faba bean + fenugreek and faba bean + garlic, showing the highest seed yield/ha with 3.722 and 3.568 ton/ha, respectively. Intercropping of faba bean with garlic integrated with AMF revealed the highest values of LER, 2.45, and net return, 2341 US$/ha. Our results suggested that using faba bean–garlic intercrop along with AMF inoculation can reduce root rot disease, damping off and broomrapes, as well as enhance the profitability of Egyptian farmer and sustainable production.  相似文献

Various seed dressing and soil application formulations were developed from Trichoderma viride, T. virens and T. harzianum to increase the shelf life of bio-formulations used to manage dry root rot (Rhizoctonia bataticola) of mungbean (Vigna radiata), a major yield limiting factor in mungbean production. The shelf life of the formulations developed in the present study was monitored by counting colony forming units (cfu) up to 25 months of storage at room temperature (26 ± 8 °C). A newly developed seed dressing formulation, Pusa 5SD based on peat powder (47.5%), Sabudana powder (Manihot esculenta) (47.5%) and carboxymethyl cellulose (5%) and a newly developed soil application formulation, Pusa Biopellet (PBP) based on sodium alginate, aluminium silicate, Sabudana powder and tap water (1:5:5:100 w/w/w/v) exhibited longer shelf life. Another formulation Pusa Biogranule (PBG) based on wheat and pulse brans varied in cfu counts during different periods of storage. Pusa 5SD could be used up to 25 months of storage while PBP 10G and PBG 5 could be used up to 15 months of storage (>105 cfu). The efficacy of the formulations was evaluated in pot experiments against the disease. In these experiments, T. harzianum based PBP 10G and PBG 5 for soil application, and Pusa 5SD for seed treatment were found to be superior to others in reducing the dry root rot incidence, and increasing the seed germination and shoot and root lengths. However, a combination of soil application of PBP 10G (T. harzianum) and seed treatment with T. harzianum based Pusa 5SD + carboxin was found superior to the use of any of these formulations alone in reducing the dry root rot incidence (87.2%) and increasing the seed germination (43.0%), shoot length (40.3%), root length (37.0%) and grain yield (54.6%) of mungbean crop over those of untreated control under sick field conditions.  相似文献

We examined if germination and seedling emergence of species from the soil seed bank of mesic grassland in South Africa differed in their response to smoke or heat treatments alone or combined. Soil seed bank samples taken from 0 to 5 cm depth of the topsoil were treated with smoke-water solution, heat and heat + smoke-water combined and subsequent emergence of seedlings monitored over 90 days. In total, 790 individuals from 11 different plant families representing 37 graminoid (Poaceae) and non-graminoid species of flowering plants were identified. The most abundant plant families that emerged were Poaceae followed by Asteraceae. Grasses contributed 18% of the germinable seed bank, while non-grass (forbs and trees) species contributed >80%. The most abundant grass species that emerged was Themeda triandra and the most common forb species was Centella asiatica. Compared to the control, smoke-water (SW) and/or smoke-water + heat (H + SW) treatment resulted in a 1.6-fold increase in mean number of seedling emergence. SW and H + SW treatment of soil seed bank also resulted in fourfold to fivefold greater biomass production. These results suggest that emergence of some smoke-responsive perennial grasses (e.g. Themeda triandra) from the soil seed bank can be enhanced using smoke techniques. Moreover, plant-derived smoke could potentially have a significant role in the restoration of degraded grasslands in South Africa and elsewhere.  相似文献

To evolve eco-friendly management of fenugreek root rot caused by Rhizoctonia solani, a field trial was conducted during Kharif 2002 and Rabi seasons of 2002–2003 and 2003–2004. Experiments were conducted with eight treatments and three replications in RBD using the variety CO-2. The pooled analysis of the three season data showed that seed treatment with Trichoderma viride at 4g/kg of seed + soil application of Trichoderma viride at 5 kg/ha + soil application of neem cake at 150 kg/ha (T3) recorded a percent disease index (PDI) of 23.1 versus 65.5 PDI in the control which accounted for a disease reduction of 64.7%. It was on par with seed treatment with Trichoderma viride at 4g/kg of seed + soil application of T. viride at 5 kg/ha (T2) which reduced the disease incidence by 62.3% (24.7 PDI). The chemical treatment used for comparison, i.e. seed treatment with carbendazim + soil drenching at 0.1% + soil application of neem cake at 150 kg/ha recorded the lowest PDI of 16.8 with 74.4% disease reduction. Among the various treatments T3 gave a seed yield of 572.7 kg/ha followed by T2 (555.7 kg/ha). Treatment T7 recorded the highest yield of 578.7 kg/ha. In the control plot the recorded yield was only 359.3 kg/ha. Though T3 was more effective at reducing the disease incidence than T2, the C:B ratio was higher (1:9.1) in respect of T2 than T3, which gave a C:B ratio of only 1:3.9. Hence, seed treatment with T. viride at 4g/kg + soil application of T. viride at 5kg/ha is a cost effective, eco-friendly management strategy for fenugreek root rot.  相似文献

A Gram-positive, rod-shaped, motile and spore-forming bacterium, designated ZLD-8T, was isolated from a desert soil sample collected from Xinjiang Province in north-west China, and subjected to a polyphasic taxonomic analysis. This isolate grew optimally at 30°C and pH 7.0. It grew with 0–4% NaCl (optimum, 0–1%). Comparative 16S rRNA gene sequence analysis showed that strain ZLD-8T was closely related to members of the genus Bacillus, exhibiting the highest 16S rRNA gene sequence similarity to Bacillus kribbensis DSM 17871T (98.0%). The levels of 16S rRNA gene sequence similarity with respect to other Bacillus species with validly published names were less than 96.3%. The DNA G + C content of strain ZLD-8T was 40.1 mol%. The strain contained MK-7 as the predominant menaquinone. The diagnostic diamino acid in the cell-wall peptidoglycan was meso-diaminopimelic acid. The major polar lipids were diphosphatidylglycerol, phosphatidylglycerol and phosphatidylethanolamine. The major fatty acids (>5% of total fatty acids) were anteiso-C15:0 (39.56%), iso-C14:0 (25.69%), C16:1 ω7c alcohol (10.13%) and iso-C15:0 (5.27%). These chemotaxonomic results supported the affiliation of strain ZLD-8T to the genus Bacillus. However, low DNA–DNA relatedness values and distinguishing phenotypic characteristics allowed genotypic and phenotypic differentiation of strain ZLD-8T from recognized Bacillus species. On the basis of the polyphasic evidence presented, strain ZLD-8T is considered to represent a novel species of the genus Bacillus, for which the name Bacillus deserti sp. nov. is proposed. Mechanisms for the undesired persistence of Bacillus species in paper machine slimes were investigated. Biofilm formation was measured for industrial Bacillus isolates under paper machine wet-end-simulating conditions (white water, pH 7, agitated at 45°C for 1–2 days). None of the 40 tested strains of seven Bacillus species formed biofilm on polished stainless steel or on polystyrene surfaces as a monoculture. Under the same conditions, Deinococcus geothermalis E50051 covered all test surfaces as a patchy thick biofilm. The paper machine bacilli, however, formed mixed biofilms with D. geothermalis E50051 as revealed by confocal microscopy. Biofilm interactions between the bacilli and the deinococci varied from synergism to antagonism. Synergism in biofilm formation of D. geothermalis E50051 was strongest with Bacillus coagulans D50192, and with the type strains of B. coagulans, B. amyloliquefaciens or B. pumilus. Two B. licheniformis, one B. amyloliquefaciens, one B. pumilus and four B. cereus strains antagonized biofilm production by D. geothermalis. B. licheniformis D50141 and the type strain of B. licheniformis were the strongest antagonists. These bacteria inhibited deinococcal growth by emitting heat-stable, methanol-soluble metabolite(s). We conclude that the persistence of Bacillus species in paper machine slimes relates to their ability to conquer biofilms formed by primary colonizers, such as D. geothermalis. Ravenna grass, Tripidium ravennae (L.) H. Scholz, is known to produce an abundance of biomass, but how plant density affects its biomass potential remains unknown. The objectives were to determine the effects of plant density on biomass yield; plant growth traits; biomass?carbon, nitrogen, and ash concentrations; heating value; nitrogen removal; and sucrose concentration in leaves and culms. The treatments consisted of five plant densities (1,250; 2,500; 5,000; 10,000; and 20,000 plants per hectare) in a randomized complete block design with four blocks. Plots were nonirrigated, unfertilized, and harvested once during the dormant season each year. Data were collected from 2015?2019. Dependent variables that varied with plant population density (p < .05) were biomass yield, number of reproductive culms per plant, reproductive culm diameter, reproductive culm sucrose concentration, and nitrogen removal with biomass. Biomass yield ranged from 5.6 to 16.3 Mg/ha for plant densities of 1,250–20,000 plants per hectare, respectively. Combined over years, nonlinear regression of the data showed the equation for biomass yield to plateau at 16.2 Mg/ha at a plant density of 10,640 plants per hectare. As plant density increased, the number of reproductive culms per plant, culm diameter, and culm sucrose concentration significantly decreased. At 1,250 plants per hectare, the number of reproductive culms per plant, culm diameter, and culm sucrose averaged 70, 10.2 mm, and 63.2 g/kg, respectively. Nitrogen removed with biomass significantly increased as biomass yield increased with plant density. At a density of 10,000 and 20,000 plants per hectare, the amount of nitrogen removed annually in the harvested biomass averaged 88 kg/ha. The data suggest that 10,000 plants per hectare would produce the greatest annual biomass yields; however, research is needed to determine the nutrient requirement for Ravenna grass to sustain biomass production at that density.  相似文献

The effect of fluctuations of salinity in three different seasons on diazotrophic populations and N2 fixation in six mono cropped rice field soils of the coastal region of the Gangetic delta of West Bengal, India, was studied. The average pH, ECe, organic carbon and total nitrogen of the soils ranged from 4.99–7.08, 2.02–19.58 dSm−1, 4.68–12.03 g kg−1 and 0.44–1.70 g kg −1, respectively. The average log colony forming units of the bacterial populations and N2-fixation in the soils varied from 4.61 to 5.86 and 2.74 to 4.52 mg N2 fixed 50 ml −1 culture media respectively, with the lowest value recorded in summer. Recovery of microorganisms and N2- fixation gradually decreased with extraneous addition of NaCl in the culture media. All the eight isolates were Gram positive, spore and capsule formers. They could utilize glucose, sucrose, mannitol, starch, citrate and nitrate, and were catalase and gelatinase positive, but indole, methyl red and Vogues Proskauer reaction negative. The organisms produced alkaline reaction on TSI agar slant. The acetylene reduction assay of the isolates at 0 and 1% NaCl in the culture media were 4.51–164.52 and 1.72–100.6 nmole C2H4 ml−1 culture media in 72 h, respectively. The isolates could fix 2.42–4.45 and 2.04–4.08 mg N2 fixed 50 ml−1 culture media at 0 and 1% NaCl in the culture media respectively. 16S rDNA sequences of the isolates were similar to the species: Bacillus sp. isolate 28A, Bacillus sp. MOLA 87, Bacillus sp. By113 (B)Ydz-dh, Bacillus sp. PN13, Bacillus licheniformis strain RH101, Bacterium Antarctica 14, Bacillus sp. PN13 and Bacillus megaterium.  相似文献

The spermosphere, an important habitat to the plant micro-ecosystem, has a unique significance to seed microbial ecology, but has been poorly researched. In this study, the mature seeds of reciprocal cross maize (Zea mays L., Nongda108) were collected to investigate the diversity and population succession dynamics of indigenous spermosphere bacteria at 12, 24 and 36 h into seed germination using 16S rDNA library construction. In the spermosphere of Nongda108A (Huang C × 178), the dominant bacteria genera identified were Pseudomonas and Burkholderia. The proportion of Pseudomonas increased from 59.60 to 75.00% then 82.61%; while Burkholderia decreased from 39.39 to 25.00% then 15.22% at 12, 24 and 36 h, respectively. Bacillus, Paenibacillus and Stenotrophomonas were the dominant genera in Nongda108B. The proportion of Paenibacillus after 12, 24 and 36 h into germination decreased from 68.00 to 46.15 to 13.27%, respectively. The proportion of non-Paenibacillus genera increased from 32.00 (Stenotrophomonas) to 53.85 (Bacillus) to 77.55% (Burkholderia) from 12 h to 24 h to 36 h, respectively. Sixteen Bacillus strains isolated from rhizosphere, histoplane and phyllosphere of different plant species were identified by 16S rDNA gene sequencing and evaluated for in vitro auxin production as well as growth stimulation of Vigna radiata (L.) Wilczek. Auxin production by Bacillus spp. in L-broth medium supplemented with 1,000 μg ml−1 L-tryptophan ranges from 0.60 to 3.0 μg IAA ml−1 as revealed by gas chromatography and mass spectrometric (GC–MS) analysis. Rhizospheric isolates exhibit relatively more IAA synthesis than histoplane and phyllosphere isolates. Plant microbe interaction experiments conducted under gnotobiotic conditions recorded 55.55, 46.46 and 46.20% increase in shoot length with Bacillus megaterium MiR-4, B. pumilus NpR-1 and B. subtilis TpP-1, respectively, over control. Bacillus inoculations also increased shoot fresh weight with B. megaterium MiR-4 (60.94%) and B. pumilus NpR-1 (37.76%). Highly significant positive correlation between auxin production analyzed by GC–MS and shoot length (r = 0.687**, P = 0.01) and shoot fresh weight (r = 0.703**, P = 0.01) was noted under gnotobiotic conditions. Similarly, significant correlation was also found between auxin production by Bacillus spp. (GC–MS analysis) and different growth parameters such as shoot length (r = 0.495*, P = 0.05), number of pods (r = 0.498*, P = 0.05) and grain weight (r = 0.537*, P = 0.05) at full maturity under natural wire house conditions. Results showed that auxin production potential of plant associated Bacillus spp. can be effectively exploited to enhance the growth and yield of V. radiata.  相似文献

A strain of Bacillus produced an amylase with properties characteristically different from known bacterial amylases. The purified 80 kDa protein of pI 5.1 dextrinized starch, glycogen and pullulan. The temperature and pH optima of the enzyme were 60 °C and 6.6 respectively. In the presence of 0.05 M CaCl2, the enzyme retained stability for 15 min at 80 °C. Antibodies raised to the amylase protein showed no reaction with -amylases of Bacillus sp. and B. licheniformis. In culture, proteolytic degradation of the enzyme was observed.  相似文献

Numerous studies have shown that early-fruit removal enhances vegetative growth and development of cotton (Gossypium hirsutum L.). However, few studies have examined changes in leaf senescence and endogenous hormones due to fruit removal. The objective of this study was to determine the correlation between some endogenous phytohormones, particularly the cytokinins and abscisic acid (ABA), and leaf senescence following fruit removal. Cotton was grown in pots and in the field during 2005 and 2006. Two early-fruiting branches were excised from plants at squaring to form the fruit removal treatment while the non-excised plants served as control. Plant biomass, seed cotton yield, cytokinins and ABA levels in main-stem leaves and xylem sap as well as main-stem leaf photosynthetic rate (Pn) and chlorophyll (Chl) concentration were determined after removal or at harvest. Fruit removals increased the leaf area, root and shoot dry weight and plant biomass at 35 days after removal (DAR), whether in potted or field-grown cotton; under field conditions, it also improved plant biomass and seed cotton yield at harvest. The Pn and Chl concentration in excised plants were significantly higher than in control plants from 5 to 35 DAR, suggesting that fruit removal considerably delayed leaf senescence. Fruit-excised plants contained more trans-zeatin and its riboside (t-Z + t-ZR), dihydrozeatin and its riboside (DHZ + DHZR), and isopentenyladenine and its riboside (iP + iPA) but less ABA in both main-stem leaves and xylem sap than control plants from 5 to 35 DAR. These results suggest that removal of early fruiting branches delays main-stem leaf senescence, which can be attributed to increased cytokinin and/or reduced ABA. Cytokinin and ABA are involved in leaf senescence following early fruit removal.  相似文献

This study presents the improved biodegradation of Congo red, a toxic azo dye, using mutant Bacillus sp. obtained by random mutagenesis of wild Bacillus sp. using UV and ethidium bromide. The mutants obtained were screened based on their decolorization performance and best mutants were selected for further studies. Better decolorization was observed in the initial Congo red concentration range 100–1000 mg/l for wild species whereas mutant strain was found to offer better decolorization up to 3000 mg/l. Mutant strain offered 12–30% reduction in time required for the complete decolorization by wild strain. The optimum pH and temperature were found to be 7.0 and 37 °C, respectively. Two efficient strains such as Bacillus sp. ACT 1 and Bacillus sp. ACT 2 were isolated from the various mutants obtained. Bacillus sp. ACT 2 showed improved enzymatic production and Bacillus sp. ACT 1 showed improved growth compared to wild strain. The enzyme responsible for the degradation was found to be azoreductase by SDS–PAGE and about 53% increased production of enzyme was achieved with mutant species. The experimental data were modeled using growth and substrate inhibition models.  相似文献
