Summary The ratio of deuterium to hydrogen (expressed as D) in hydrogen released as water during the combustion of dried plant material was examined. The D value (metabolic hydrogen) determined on plant materials grown under controlled conditions is correlated with pathways of photosynthetic carbon metabolism. C3 plants show mean D values of-132 for shoots and -117 for roots; C4 plants show mean D values of -91 for shoots and-77 for roots and CAM plants a D value of-75 for roots and shoots. The difference between the D value of shoot material from C3 and C4 plants was confirmed in species growing under a range of glasshouse conditions. This difference in D value between C3 and C4 species does not appear to be due to differences in the D value (tissue water) in the plants as a result of physical fractionation of hydrogen isotopes during transpiration. In C3 and C4 plants the hydrogen isotope discrimination is in the same direction as the carbon isotope discrimination and factors contributing to the difference in D values are discussed. In CAM plants grown in the laboratory or collected from the field D values range from-75 to +50 and are correlated with 13C values. When deprived of water, the D value (metabolic hydrogen) in both soluble and insoluble material in leaves of Kalanchoe daigremontiana Hamet et Perr., becomes less negative. These changes may reflect the deuterium enrichment of tissue water during transpiration, or in field conditions, may reflect the different D value of available water in areas of increasing aridity. Whatever the origin of the variable D value in CAM plants, this parameter may be a useful index of the water relations of these plants under natural conditions.  相似文献

Methods are described to correlate aromatic 1H 2/13C 2 or 1H 1/15N 1 with aliphatic 13C chemical shifts of histidine and tryptophan residues, respectively. The pulse sequences exclusively rely on magnetization transfers via one-bond scalar couplings and employ [15N, 1H]- and/or [13C, 1H]-TROSY schemes to enhance sensitivity. In the case of histidine imidazole rings exhibiting slow HN-exchange with the solvent, connectivities of these proton resonances with -carbons can be established as well. In addition, their correlations to ring carbons can be detected in a simple [15N, 1H]-TROSY-H(N)Car experiment, revealing the tautomeric state of the neutral ring system. The novel methods are demonstrated with the 23-kDa protein xylanase and the 35-kDa protein diisopropylfluorophosphatase, providing nearly complete sequence-specific resonance assignments of their histidine -CH and tryptophan -NH groups.  相似文献

Zusammenfassung Serumproben von 1322 Blutspendern aus Hessen, 40 Familien mit 89 Kindern, 20 Mutter-Kind-Kombinationen und 268 Sera einer Bantupopulation aus Portugiesisch Angola wurden mit einer modifizierten Technik der Hochspannungs-Dünnschichtelektrophorese auf Agarosegel hinsichtlich des C3-Polymorphismus untersucht. Die Genfrequenzen für Weiße (C3S=0.779, C3F=0.215) und Neger (C3S=0.95, C3F=0,048) stimmen gut mit den Werten anderer Autoren überein. Insgesamt ließen sich bei Weißen 9 Phänotypen sicher abgrenzen, bei Negern 3. Familienstudien bestätigten den für die Allele C3S und C3F angenommenen Vererbungsmodus (autosomal codominant) ausnahmslos. Die Frage der Lagerungsstabilität des C3 wurde abschließend untersucht.
Investigations on C3-polymorphism ( 1c-Globulin)Gene frequencies and family studies in blood donors from Hessen and a Bantu population
Summary Serum samples of 1322 unrelated individuals from Hessen (Germany), 40 families with 89 children, 20 mother-child-combinations and 268 sera of a Bantu population from Angola were examined for C3 polymorphism using a modified technique of high voltage agarose gel electrophoresis. The gene frequencies for Caucasians (C3S=0.779, C3F=0.215) and negroes (C3S=0.95, CF=0.048) are in good accordance with those obtained by other authors. In total 9 different phenotypes were observed in Caucasians, 3 phenotypes in negroes. Family studies verify the supposed way of inheritance (autosomal codominant for C3S and C3F) without exception. Finally the problem of C3-inactivation by storage was investigated.

Charophyte oosporangia and water samples from a highly calcareous lake were measured for stable carbon and oxygen isotopic composition. The time period over which the oosporangia calcify is short, thus any biochemical relationship between the water and oosporangia"s calcite represents only one time window (late Summer in Malham Tam). This important temporal restraint must also apply to interpretations of all fossil material measured. The 18Oc of the charophyte oosporangia is deduced to be in equilibrium with the 18O of the water for a given temperature. The 13 Cc of the charophyte oosporangia was approximately 2.5 per mil lower than the 13CDIC in the water we measured. With the release Of CO2 with phosphoric acid from the charophyte oosporangia, there was no significant difference in the 18Oc values obtained, regardless of whether or not the carbonate was separated from the organic center, however 13Cc values were marginally lower for carbonate plus organic center measurements. Our results indicate that fossil charophyte gyrogonites can be used to elucidate the geochemistry of the ancient water body in which they lived.  相似文献

Summary To determine whether sweet potato (Ipomoea batatas (L.) Poir.) takes up organic matter through the roots from the medium, the concentrations of natural14C (14C) in plant organic matter, atmospheric CO2 and compost applied to media were examined under soil and sand culture conditions. In these experiments, three kinds of composts of different 14C were used. CO2 derived from the mineralization of compost was continuously pumped out from the pots and its direct uptake by the leaves was prevented.14C of plant parts harvested after the 43 days experimental period were affected by the 14C of the compost in the treatments where the compost of rice straw was applied, and which suggested that a significant amount of plant carbon was derived from the compost.  相似文献

Successful application of stable-hydrogen isotope measurements (Df) of feathers to track origins of migratory birds and other wildlife requires a fundamental understanding of the correlation between Df and deuterium patterns in rainfall (Dp) over continental scales. A strong correlation between Dp and Df has been confirmed for birds and insects in North America, but not yet for other continents. Here, we compare Df data from resident European birds to new Dp basemaps for Europe. Three maps, representing growing-season and mean annual Dp estimates from an elevation-explicit, detrended interpolation model and growing-season Dp estimates from simple Kriging, all indicate that strong isotope gradients occur across Europe with a general depletion occurring in a northeast direction. The feather data, representing 141 individuals of 25 avian species from 38 sites, ranged from –131 to –38. Regression analysis showed that strong correlations existed between both mean annual and growing-season Dp estimated by detrended interpolation and Df of non-aquatic and non-corvid birds (r2=0.66 and 0.65, respectively). We also examined mean annual and growing-season 18Op vs. 18Of for our samples. Both oxygen regressions were similar (r2=0.56 and 0.57, respectively) but poorer than for deuterium. Our study reveals that D measurements of feathers from migratory birds in Europe may be used to track their origin and movements, and so provide a powerful investigative tool for avian migration research in Europe.  相似文献

Rotational diffusion properties have been derived for the DNA dodecamer d(CGCGAATTCGCG)2 from 13C R1 and R1 measurements on the C1, C3, and C4 carbons in samples uniformly enriched in 13C. The narrow range of C-H bond vector orientations relative to the DNA axis make the analysis particularly sensitive to small structural deviations. As a result, the R1/R1 ratios are found to fit poorly to the crystal structures of this dodecamer, but well to a recent solution NMR structure, determined in liquid crystalline media, even though globally the structures are quite similar. A fit of the R1/R1 ratios to the solution structure is optimal for an axially symmetric rotational diffusion model, with a diffusion anisotropy, D||/D, of 2.1±0.4, and an overall rotational correlation time, (2D||+4D)–1, of 3.35 ns at 35 °C in D2O, in excellent agreement with values obtained from hydrodynamic modeling.  相似文献

Summary Stable carbon isotope ratios of roots and soil organic matter were measured in Curlew Valley, Utah to determine if changes in the relative dominance of two shrub species had occurred in this salt-desert community. Measurements were made on soil cores along transects stretching from monospecific stands of Ceratoides lanata, a C3 shrub, to monospecific stands of Atriplex confertifolia, a C4 shrub. 13C values of roots and soil organic matter under Ceratoides cover appeared to be in equilibrium with the current plant community. By contrast, 13C values of roots and soils under Atriplex portions of the transects were more negative than would be expected for a C4-dominated community. These results indicate that a change in relative C3/C4 dominance has occurred, and suggest that the C4 shrub Atriplex confertifolia is increasing in importance in this salt-desert community.  相似文献

Summary In a lactic acid fermentation by Streptococcus faecalis, the specific consumption rate of glucose (v) and the specific production rate of lactic acid () were represented by the following simple equations as functions of the specific growth rate (): 1/=(1/) + 1/ = (1/) + By use of data from a batch culture, these two equations were derived from enzyme kinetics of the product inhibition. These equations were successfully applied to the results of batch culture and chemostat culture. In addition, calculation of ATP yield by these equations agreed with the experimental results better than the conventional Leudeking-Piret type equation, which includes two terms associated with growth and not with growth. Summary Natural carbon and nitrogen isotope ratios were measured in different compartments (needles and twigs of different ages and crown positions, litter, understorey vegetation, roots and soils of different horizons) on 5 plots of a healthy and on 8 plots of a declining Norway spruce (Picea abies (L.) Karst.) forest in the Fichtelgebirge (NE Bavaria, Germany), which has recently been described in detail (Oren et al. 1988a; Schulze et al. 1989). The 13C values of needles did not differ between sites or change consistently with needle age, but did decrease from the sun-to the shade-crown. This result confirms earlier conclusions from gas exchange measurements that gaseous air pollutants did no long-lasting damage in an area where such damage was expected. Twigs (13C between-25.3 and-27.8) were significantly less depleted in 13C than needles (13C between-27.3 and-29.1), and 13C in twigs increased consistently with age. The 15N values of needles ranged between-2.5 and-4.1 and varied according to stand and age. In young needles 15N decreased with needle age, but remained constant or increased in needles that were 2 or 3 years old. Needles from the healthy site were more depleted in 15N than those from the declining site. The difference between sites was greater in old needles than in young ones. This differentiation presumably reflects an earlier onset of nitrogen reallocation in needles of the declining stand. 15N values in twigs were more negative than in needles (-3.5 to-5.2) and showed age- and stand-dependent trends that were similar to the needles. 15N values of roots and soil samples increased at both stands with soil depth from-3.5 in the organic layer to +4 in the mineral soil. The 15N values of roots from the mineral soil were different from those of twigs and needles. Roots from the shallower organic layer had values similar to twigs and needles. Thus, the bulk of the assimilated nitrogen was presumably taken up by the roots from the organic layer. The problem of separation of ammonium or nitrate use by roots from different soil horizons is discussed.  相似文献

To determine if changes in microbial community composition and metabolic capacity alter decomposition patterns of young and old soil carbon pools, we incubated soils under conditions of varying temperature, N-availability, and water content. We used a soil from a pineapple plantation (CAM; 13C litter = –14.1) that had previously been under tropical forest (C3; 13C soil carbon = –26.5). Forest derived carbon represented 'old' carbon and plantation inputs represented 'new' carbon. In order to differentiate utilization of young (< 14 years) and old (> 14 years) soil carbon, we measured the 13C of respired CO2 and microbial phospholipid fatty acids (PLFAs) during a 103 day laboratory incubation. We determined community composition (PLFA and bacterial intergenic transcribed spacer (ITS) analysis) in addition to carbon degrading and nutrient releasing enzyme activities. We observed that greater quantities of older carbon were respired at higher temperatures (20 and 35°C) compared to the lower temperature (5°C). This effect could be explained by changes in microbial community composition and accompanying changes in enzyme activities that affect C degradation. Nitrogen addition stimulated the utilization of older soil carbon, possibly due to greater peroxidase activity, but microbial community composition was unaffected by this treatment. Increasing soil moisture had no effect on the utilization of older SOM, but enzyme activity typically declined. Increased oxidative enzyme activities in response to elevated temperature and nitrogen additions point to a plausible mechanism for alterations in C resource utilization patterns.  相似文献

Summary A new 1H-detected 3D NMR experiment is described that permits quantitative measurement of two- and three-bond 13C–1H couplings in proteins with selectively 13C-enriched methyl sites. The method is demonstrated for staphylococcal nuclease selectively [5,5 13C]-labeled in all 11 leucine positions and ligated with thymidine 3,5-biphosphate and Ca2+. Two- and three-bond 13C methyl-proton couplings are reported and, together with the measured three-bond JCC in uniformly 13C-enriched staphylococcal nuclease, the 2- and the stereospecific assignments of the C methyl group with respect to the prochiral -protons were determined. The same residues that were previously found to have high degrees of internal mobility on the basis of 13C relaxation times have measured coupling constants that are indicative of motional averaging.  相似文献

Summary Five subunits (-, -, -, - and -subunits) of the six -and -subunits) in the F1 portion (F1ATPase) of sweet potato (Ipomoea batatas) mitochondrial adenosine triphosphatase were isolated by an electrophoretic method. The - and -subunits were not distinguishable immunologically but showed completely different tryptic peptide maps, indicating that they were different molecular species. In vitro protein synthesis with isolated sweet potato root mitochondria produced only the -subunit when analyzed with anti-sweet potato F1ATPase antibody reacting with all the subunits except the -subunit. Sweet potato root poly(A)+RNA directed the synthesis of six polypeptides which were immunoprecipitated by the antibody: two of them immunologically related to the -subunit and the others to the - and -subunits. We conclude that the -subunit of the F1ATPase is synthesized only in the mitochondria and the -, - and -subunits are in the cytoplasm.  相似文献

The study deals with a comparative analysis of the relative abundances of the carbon isotopes 12C and 13C in the metabolites and biomass of the Burkholderia sp. BS3702 and Pseudomonas putida BS202-p strains capable of utilizing aliphatic (n-hexadecane) and aromatic (naphthalene) hydrocarbons as sources of carbon and energy. The isotope compositions of the carbon dioxide, biomass, and exometabolites produced during the growth of Burkholderia sp. BS3702 on n-hexadecane (13C = –44.6 ± 0.2) were characterized by the values of 13CCO 2 = –50.2 ± 0.4, 13Cbiom = –46.6 ± 0.4, and 13Cexo = –41.5 ± 0.4, respectively. The isotope compositions of the carbon dioxide, biomass, and exometabolites produced during the growth of the same bacterial strain on naphthalene (13C = –21 ± 0.4) were characterized by the isotope effects 13CCO 2 = –24.1 ± 0.4, 13Cbiom = –19.2 ± 0.4, and 13Cexo = –19.1 ± 0.4, respectively. The possibility of using the isotope composition of metabolic carbon dioxide for the rapid monitoring of the microbial degradation of petroleum hydrocarbons in the environment is discussed.  相似文献

The present paper reports our attempts to determine whether the inclusion of 0.0014 mM Zn++ within a hydroponic culture medium affects the ability of 12-day-old Zea mays, cv. SS-522 to take-up [3H]-aflatoxin B1. Data from the corollary experiment, i.e., whether inclusion of aflatoxin affects the ability of Zea mays, cvs. Truckers White, X-Sweet and Merit to take-up 65ZnCl2 are presented also. This report is a preliminary to one regarding an in-progress analysis of whether pollutant levels of Zn++ affect aflatoxin uptake and distribution. In the absence of irrigating seedlings, which were grown in Perlite containing 65ZnCl2, with a solution containing mixed aflatoxins, the stem contained the greatest amount of label with root plus seed the next highest and the leaf the least for each of the cvs. In contrast, when the seedlings were irrigated with a solution containing mixed aflatoxins, the root plus seed contained either an amount nearly identical to (cv. Truckers White) or in excess of that within the stem (cvs. X-Sweet and Merit). Calculation of the percentages of aflatoxin-induced diminutions in leaf, stem and root label suggested that the aflatoxins interfered with the translocation of 65ZnCl2 from the root to the stem and leaf, at least for cvs X-Sweet and Merit. When 0.0014mM Zn++ as ZnSO4 was added to an incubation medium in which 12-day-old seedlings were suspended and plant growth assessed over 72 hours, a 15% increase (significant at 0.05 level) in seedling height over that of Zn++-deficient plants was observed. No differences in [3H]-aflatoxin B1 uptake were noted between those seedlings which were grown in either Zn++-containing or lacking media. Less than one % of the[3H]-aflatoxin B1 which was taken-up was recovered within chloroform extracts of the seedlings. The distributions of radioactivity from [3H]-aflatoxin B1 for leaf, stem, seed and root were 0, 57, 26 and 19% and 0, 26, 58 and 18% for Zn++-containing and -lacking media, respectively.  相似文献

Carbon-isotope ratios were examined as 13C values in several C3, C4, and C3–C4 Flaveria species, and compared to predicted 13C, values generated from theoretical models. The measured 13C values were within 4 of those predicted from the models. The models were used to identify factors that contribute to C3-like 13C values in C3–C4 species that exhibit considerable C4-cycle activity. Two of the factors contributing to C3-like 13C values are high CO2 leakiness from the C4 pathway and pi/pa values that were higher than C4 congeners. A marked break occurred in the relationship between the percentage of atmospheric CO2 assimilated through the C4 cycle and the 13C value. Below 50% C4-cycle assimialtion there was no significant relationship between the variables, but above 50% the 13C values became less negative. These results demonstrate that the level of C4-cycle expression can increase from, 0 to 50% with little integration of carbon transfer from the C4 to the C3 cycle. The inheritance of host plant resistance and its effect on the relative infection efficiency for leaf blast was studied in the crosses IR36/CO39 (partially resistant × highly susceptible) and IR36/IR64 (both partially resistant). On the natural scale, gene action appeared multiplicative. After log transformation, additive effects described most of the genetic variation in the cross IR36/CO39, while additive and dominance effects were about equal in magnitude in the cross IR36/IR64. Dominance was towards increased resistance. No transgressive segregation occurred in the cross IR36/CO39. The number of genes that reduce lesion number was estimated to be zero in CO39 and five or more in IR36. The cross IR36/IR64 showed transgressive segregation in both directions, and IR36 and IR64 each contain at least one gene that is not present in the other cultivar. The heritabilities (narrow sense) in the F2 were low (range 0.06–0.16), while narrow sense heritabilities based on F3 lines were much higher (range 0.41–0.68). Lesion numbers in F3 lines were reasonably correlated with those in F5 progenies derived from the same F2 plant (r was±0.6 in both crosses). Partial resistance can be effectively improved by selecting the most resistant plants from the most resistant F3 lines.  相似文献

As preparation for a below ground food web study, the spatial variability of three soil properties (total N, total C and pH) and two stable isotopes (13C and 15N of whole soil) were quantified using geostatistical approaches in upland pastures under contrasting management regimes (grazed, fertilised and ungrazed, unfertilised) in Scotland. This is the first such study of upland, north maritime grasslands. The resulting patterns of variability suggest that to obtain statistically independent samples in this system, a sampling distance of 13.5 m is required. Additionally, temporal change (a decline of 1) was observed in whole soil 15N for the grazed, fertilised plot. This may have been caused by new inputs of symbiotically-fixed atmospheric N2.  相似文献

We present the results of a 5-year examination of variation in the stable carbon isotope composition () of three C3 graminoid species from a Sandhills prairie: Agropyron smithii, Carex heliophila and Stipa comata. Although consistent species-specific patterns for mean were seen, there was also significant and substantial among-year and within-season variation in . A smaller contribution to variation in came from topographic variation among sampling sites. Effects of species, year, season and topography contribute to variation in in an additive manner. An association between climate and exists that is consistent with previous work suggesting that reflects the interplay between photosynthetic gas exchange and plant water relations. Within the growing season, the time over which integrates plant response to the environment ranges from days to months.  相似文献

Cyanobacterial mats (CBM) are important components of wetland ecosystems in limestone-based regions of the Caribbean. During two sampling periods (July 1999 and January 2000) we measured N2-fixation in samples from 23 different marshes simultaneously with measurements of relevant environmental factors. Samples were evaluated for abundance of five groups of cyanobacteria: (1) Leptolyngbya, (2) Oscillatoria, (3) Chroococcales, (4) Nostoc-& Stigonematales, and (5) dead sheaths. Differences in nitrogen fixation, expressed as nitrogenase activity in nmol C2H4 cm–2 h–1, were best explained by the proportion of heterocyst-forming cyanobacteria. The samples were analyzed for the natural abundance of 15N. 15N values ranged from –1.99 to 11.44 and were strongly negatively correlated with N2-fixation. With all data included, 15N was also strongly correlated with nitrates in water. With the samples from Little Belize (high nitrate content marshes) excluded, the effect of nitrate became insignificant. N2-fixation predicted from 15N measured on an independent data set from September 2000 was moderately accurate (r2 = 0.68, 0.52 and 0.54 for predictions based on July 1999, January 2000 and combined data sets, respectively). When individual sample sets were divided into two groups with 15N < 2 and 15N > 2, the two groups were always highly significantly different in terms of their N2-fixation. The presented evidence suggests that 15N can be used as a reliable indicator of N2-fixation by CBM.  相似文献
