Bacillus thuringiensis (Berliner) bears essential characteristics in the control of insect pests, such as its unique mode of action, which confers specificity and selectivity. This study assessed cry gene contents from Bt strains and their entomotoxicity against Diatraea saccharalis (F.) and Diatraea flavipennella (Box) (Lepidoptera: Crambidae). Bioassays with Bt strains were performed against neonates to evaluate their lethal and sublethal activities and were further analyzed by PCR, using primers to identify toxin genes. For D. saccharalis and D. flavipennella, 16 and 18 strains showed over 30% larval mortality in the 7th day, respectively. The LC50 values of strains for D. saccharalis varied from 0.08 × 105 (LIIT-0105) to 4104 × 105 (LIIT-2707) spores + crystals mL?1. For D. flavipennella, the LC50 values of strains varied from 0.40 × 105 (LIIT-2707) to 542 × 105 (LIIT-2109) spores + crystals mL?1. For the LIIT-0105 strain, which was the most toxic to D. saccharalis, the genes cry1Aa, cry1Ab, cry1Ac, cry1B, cry1C, cry1D, cry1F, cry1I, cry2Aa, cry2Ab, cry8, and cry9C were detected, whereas for the strain LIIT-2707, which was the most toxic to D. flavipennella, detected genes were cry1Aa, cry1Ab, cry1Ac, cry1B, cry1D, cry1F, cry1I, cry2Aa, cry2Ab, and cry9. The toxicity data and toxin gene content in these strains of Bt suggest a great variability of activity with potential to be used in the development of novel biopesticides or as source of resistance genes that can be expressed in plants to control pests.  相似文献

A new anti-Prelog short-chain dehydrogenase/reductase (SDR) encoding gene lcsdr was cloned from Lactobacillus composti DSM 18527, and heterologously expressed in Escherichia coli. LcSDR is nicotinamide adenine dinucleotide phosphate (NADPH)-dependent and has a molecular weight of approximately 30 kDa. The optimal pH and temperature were 6.5 and 30?°C, respectively. The maximal reaction rate Vmax was 133.9 U mg?1; the Michaelis–Menten constant K m of LcSDR were 0.345 mM for acetophenone (1a), and 0.085 mM for NADPH. Through introducing an EsGDH-catalyzed NADPH regeneration system, a biocatalytic process for (R)-1-phenylethanol ((R)-1b) was developed with outstanding time–space yield. Under the optimized conditions, 50 g l?1 1a was converted to (R)-1b in 2 h with a yield of 93.8%, enantiomeric excess of product (e.e.p) above 99% and space–time yield of 562.8 g l?1 d?1.  相似文献

Objectives

To prepare (R)-phenyl-1,2-ethanediol ((R)-PED) with high enantiomeric excess (ee p) and yield from racemic styrene oxide (rac-SO) at high concentration by bi-enzymatic catalysis.

Results

The bi-enzymatic catalysis was designed for enantioconvergent hydrolysis of rac-SO by a pair of novel epoxide hydrolases (EHs), a Vigna radiata EH3 (VrEH3) and a variant (AuEH2A250I) of Aspergillus usamii EH2. The simultaneous addition mode of VrEH3 and AuEH2A250I, exhibiting the highest average turnover frequency (aTOF) of 0.12 g h?1 g?1, was selected, by which rac-SO (10 mM) was converted into (R)-PED with 92.6% ee p and 96.3% yield. Under the optimized reaction conditions: dry weight ratio 14:1 of VrEH3-expressing E. coli/vreh3 to AuEH2A250I-expressing E. coli/Aueh2 A250I and reaction at 20 °C, rac-SO (10 mM) was completely hydrolyzed in 2.3 h, affording (R)-PED with 98% ee p. At the weight ratio 0.8:1 of rac-SO to two mixed dry cells, (R)-PED with 97.4% ee p and 98.7% yield was produced from 200 mM (24 mg/ml) rac-SO in 10.5 h.

Conclusions

Enantioconvergent hydrolysis of rac-SO at high concentration catalyzed by both VrEH3 and AuEH2A250I is an effective method for preparing (R)-PED with high ee p and yield.

Dermacentor nitens tick is commonly found in the equine auditory canal, where it causes economic losses due to its direct damage, causing blood spoliation, stress, transmission of pathogens, and predisposition to myasis and secondary bacterial infection in its hosts. In this study we evaluated the effect of ethanolic extracts of Cerrado plants on biological parameters of engorged females of D. nitens. Ethanolic extracts were prepared from the leaves of Schinopsis brasiliensis, Piptadenia viridiflora, Ximenia americana, and Serjania lethalis at 25–150 mg mL?1. Groups of 10 engorged adult females were treated with these extracts and compared with a control containing distilled water and another control with organophosphate, using five replicates for each group. Compared with the control with water, S. lethalis and X. americana extracts at 100 and 150 mg mL?1 significantly inhibited the posture ability. Differently, extracts of S. brasiliensis and P. viridiflora were the most effective in inhibiting larval hatching. Extracts of X. americana and P. viridiflora showed effective inhibition of reproductive parameters of the tick, presenting dose-dependent effect with IC90 78.86 and 78.94 mg mL?1, respectively. Theses effective extracts contained low condensed tannin levels and their HPLC chromatograms revealed the presence of flavonoids. The efficacies of P. viridiflora and X. americana extracts were higher than 90% indicating that these extracts are promising as alternative agents for D. nitens control.  相似文献

A spirostane with an attached trisaccharide, (25R)-5α-spirostane-2α,3β,5α-triol 3-O-(O-α-l-rhamnopyranosyl-(1 → 2)-O-(β-d-galactopyranosyl-(1 → 3))-β-d-glucopyranoside), was isolated and identified from the aerial parts of Agapanthus africanus by activity-guided fractionation. Fungicidal properties of the crude extract, semi-purified fractions as well as the purified active saponin from A. africanus were screened in vitro against Fusarium oxysporum. At a concentration of 1 mg mL?1, the crude extract and semi-purified ethyl acetate and dichloromethane fractions showed significant antifungal activity. The purified saponin inhibited the in vitro mycelial growth of F. oxysporum completely (100 %) at a concentration of 125 µg mL?1. Furthermore, to verify previously observed induced resistance by crude extracts of A. africanus towards leaf rust, intercellular PR-protein activity was determined in wheat seedlings following foliar application of the purified saponin at 100 µg mL?1. In vitro peroxidase enzyme activity increased significantly (60 %) in wheat seedlings 48 h after treatment with the purified saponin, demonstrating its role as an elicitor to activate a defence reaction in wheat.  相似文献

The aim of this work was to evaluate the effects of co-inoculation with phosphate-solubilizing and nitrogen-fixing rhizobacteria on growth promotion, yield, and nutrient uptake by wheat. Out of twenty-five bacteria isolated from the rhizosphere soils of cereal, vegetable, and agro-forestry plants in eastern Uttar Pradesh, three superior most plant growth-promoting (PGP) isolates were characterized as Serratia marcescens, Microbacterium arborescens, and Enterobacter sp. based on their biochemical and 16S rDNA gene sequencing data and selected them for evaluating their PGP effects on growth and yield of wheat. Among them, Enterobacter sp. and M. arborescens fixed significantly higher amounts (9.32?±?0.57 and 8.89?±?0.58 mg Ng?1 carbon oxidized, respectively) of atmospheric nitrogen and produced higher amounts (27.06?±?1.70 and 26.82?±?1.63 TP 100 µg mL?1, respectively) of IAA in vitro compared to S. marcescens (8.32?±?0.39 mg Ng?1 carbon oxidized and 21.29?±?0.99 TP 100 µg mL?1). Although both M. arborescens and S. marcescens solubilized remarkable amounts of phosphate from tricalcium phosphate likely through production of organic acids, however, Enterobacter sp. was inactive. The effects of these three rhizobacteria were evaluated on wheat in alluvial soils of the Indo-Gangetic Plain by inoculation of plants with bacterial isolates either alone or in combinations in both pot and field conditions for two successive years. Rhizobacterial inoculation either alone or in consortium of varying combinations significantly (P?≤?0.05) increased growth and yield of wheat compared to mock inoculated controls. A consortium of two or three rhizobacterial isolates also significantly increased plant height, straw yield, grain yield, and test weight of wheat in both pot and field trials compared to single application of any of these isolates. Among the rhizobacterial treatment, co-inoculation of three rhizobacteria (Enterobacter, M. arborescens and S. marcescens) performed best in promotion of growth, yield, and nutrient (N, P, Cu, Zn, Mn, and Fe) uptake by wheat. Taken together, our results suggest that co-inoculation of Enterobacter with S. marcescens and M. arborescens could be used for preparation of an effective formulation of PGP consortium for eco-friendly and sustainable production of wheat.  相似文献

A halotolerant bacterial isolate-MHC10 with broad spectrum antibacterial activity against clinical pathogens was isolated from saltpans located in Tuticorin and Chennai (India). 16S rRNA gene analysis of MHC10 revealed close similarity to that of Bacillus methylotrophicus. The culture conditions of B. methylotrophicus MHC10 strain were optimized for antibacterial production using different carbon and nitrogen sources, as well as varying temperature, pH, sodium chloride (NaCl) concentrations and incubation periods. The maximum antibacterial activity of B. methylotrophicus MHC10 was attained when ZMB was optimized with 1 % (w/v) glucose, 0.1 % (w/v) soybean meal which corresponded to a C/N ratio of 38.83, temperature at 37 °C, pH 7.0 and 8 % NaCl. The activity remained stable between 72 and 96 h and then drastically decreased after 96 h. Solvent extraction followed by chromatographic purification steps led to the isolation of hydroquinone (benzene-1,4-diol). The structure of the purified compound was elucidated based on FTIR, 1H NMR, and 13C NMR spectroscopy. The compound exhibited efficient antibacterial activity against both Gram-positive and Gram-negative bacterial pathogens. The minimum inhibitory concentration (MIC) for Gram-positive pathogens ranged from 15.625 to 62.5 µg/mL?1, while it was between 7.81 and 250 µg/mL?1 for Gram-negative bacterial pathogens. This is the first report of hydroquinone produced by halotolerant B. methylotrophicus exhibiting promising antibacterial activity.  相似文献

The prokaryotic communities of four salterns (Bingöl, Fadlum, Kemah, and Tuzlagözü) in Turkey were examined and compared using the cultivation and cultivation-independent methods [fluorescence in situ hybridization (FISH) and 454 pyrosequencing]. FISH analysis with universal probes revealed that feeding waters carried 1.6 × 102–1.7 × 103 cells mL?1, while crystallization ponds carried 3.8 × 106–2.0 × 107 cells mL?1 that were mostly haloarchaea, including square cells (except for Kemah). High-throughput 16S rRNA-based gene sequencing showed that the most frequent archaeal OTUs in Bingöl, Fadlum, Tuzlagözü, and Kemah samples were affiliated with Haloquadratum (76.8 %), Haloarcula (27.8 %), Halorubrum (49.6 %), and Halonotius (59.8 %), respectively. Bacteroidetes was the dominant bacterial phylum in Bingöl and Fadlum, representing 71.5 and 79.5 % of the bacterial OTUs (respectively), while the most abundant bacterial phylum found in the Kemah saltern was Proteobacteria (79.6 %). The majority of the bacterial OTUs recovered from Tuzlagözü belonged to the Cyanobacteria (35.7 %), Bacteroidetes (35.0 %), and Proteobacteria (25.5 %) phyla. Cultivation studies revealed that the archaeal isolates were closely related to the genera Halobacterium, Haloarcula, and Halorubrum. Bacterial isolates were confined to two phyla, Proteobacteria (Alphaproteobacteria and Gammaproteobacteria classes) and Bacteroidetes. Comparative analysis showed that members of the Euryarchaeota, Bacteroidetes, Proteobacteria, and Cyanobacteria phyla were major inhabitants of the solar salterns.  相似文献

The combination of in situ measurements, hydrodynamical modeling, and satellite imagery processing presents a complete tool to improve seaweed culture management. This study measured the evolution of carrageenan content during 1-year period and estimated the biomass of Kappaphycus alvarezii at a bay scale in Malasoro Bay, Indonesia. It allowed the determination of the carrageenan stock at the scale of the bay. The carrageenan content was assessed from different parts, i.e., basal and apical, of the thallus. The biomass from T 0 (beginning of the cultivation) to T 45 (harvesting time) was determined at two seasons. Satellite image processing was performed to estimate the biomass at bay scale using parcels resulting from a semi-automatic delineation process. As no long-term in situ environmental data existed in the study area, a 3D hydrodynamical model (Model for Application at Regional Scale 3D) was implemented to obtain high-frequency salinity, water temperature, and currents. The high carrageenan yield was obtained from April to September 2015 (54–63%) when temperature ranged from 25 to 30 °C, salinity range from 33.8 to 34.8 psu, and precipitation below 0.5 mm. The biomass at bay scale was estimated at 2590 t with the highest carrageenan stock at 1.8 t ha?1 in May 2015. The carrageenan yields and quality obtained in this study fulfilled the specification recommended for industry, and the cultivation of K. alvarezii can be carried out at optimal environmental conditions on April–September. The comprehensive approaches used in this study provide information for carrageenan stock and seaweed culture management as an important economic activity to support Indonesian coastal communities.  相似文献

Xylanases (EC 3.2.1.8) are hydrolytic enzymes that have found widespread application in the food, feed, and paper-pulp industries. Streptomyces sp. FA1 xynA was expressed as a secreted protein in Pichia pastoris, and the xylanase was applied to the production of Chinese steamed bread for the first time. The optimal pH and the optimal temperature of XynA were 5.5 and 60 °C, respectively. Using beechwood as substrate, the K m and V max were 2.408 mg mL?1 and 299.3 µmol min?1 mg?1, respectively. Under optimal conditions, a 3.6-L bioreactor produced 1374 U mL?1 of XynA activity at a protein concentration of 6.3 g L?1 after 132 h of fermentation. Use of recombinant XynA led to a greater increase in the specific volume of the CSB than could be achieved using commercial xylanase under optimal conditions. This study provides the basis for the application of the enzyme in the baking industry.  相似文献

This study examined the co-immobilization of the cyanobacterium Synechococcus elongatus with the plant growth-promoting bacterium Azospirillum brasilense in alginate beads and its potential application for the removal of phosphorus from aquaculture wastewater. Co-immobilization of both microorganisms significantly increased the cell density of S. elongatus (2852.5?×?104 cells mL?1) compared with that of immobilization of cyanobacteria alone (1325.2?×?104 cells mL?1). Chlorophyll a content was similar in co-immobilized (11.1?±?3.5 pg cell?1) and immobilized S. elongatus (14.5?±?4.9 pg cell?1). Azospirillum brasilense showed continuous growth until day 2, after which its cell concentration declined until the end of the assay. Co-immobilized S. elongatus removed more phosphorus (44.8 %) than immobilized cyanobacteria cells alone (32.0 %). In conclusion, phosphate removal was greater with free cells of S. elongatus but overlapped with the values that were obtained with the treatment of co-immobilization of cells. Our results demonstrate that A. brasilense enhances the growth of S. elongatus and improves its removal of phosphorus when they are co-immobilized in alginate beads compared with only immobilization of cyanobacteria cells alone.  相似文献

The citrus fruit borer, Ecdytolopha aurantiana (Lima, 1927) (Lepidoptera: Tortricidae), is responsible for major losses to the citrus industry because it causes rot and drop of fruits. The current study aimed to select and characterize Bacillus thuringiensis (Berliner, 1911) strains toxic to E. aurantiana. For this purpose, 47 B. thuringiensis strains were evaluated in selective bioassays using first instar larvae of E. aurantiana. The lethal concentration (LC50) of the most toxic strains was estimated, and the strains were characterized by morphological, biochemical, and molecular methods. Of the 47 strains tested, 10 caused mortality above 85% and showed mean lethal concentrations between 1.05E+7 and 1.54E+8 spores mL?1. The lowest LC50 values were obtained for the HD-1 standard strain and the BR145, BR83, BR52, and BR09 strains. The protein profile showed the presence of Cry proteins of 60, 65, 70, 80, and 130 kDa. The molecular characterization showed the presence of cry1, cry2, cry3, and cry11 genes. The morphological analysis identified three different crystalline inclusions: bipyramidal, round, and cuboidal. The cry1 and cry2 genes were the most frequent among the B. thuringiensis strains evaluated and encode Cry proteins toxic to insects of the order Lepidoptera, which agree with the toxicity results obtained by the selective bioassays against E. aurantiana. The results showed four different B. thuringiensis strains toxic to E. aurantiana at the same level as the HD-1 standard strain, and these strains have biotechnological potential for E. aurantiana control through the production of transgenic plants or the formulation of biopesticides.  相似文献

A recombinant alcohol dehydrogenase (ADH) from Kangiella koreensis was purified as a 40 kDa dimer with a specific activity of 21.3 nmol min?1 mg?1, a K m of 1.8 μM, and a k cat of 1.7 min?1 for all-trans-retinal using NADH as cofactor. The enzyme showed activity for all-trans-retinol using NAD + as a cofactor. The reaction conditions for all-trans-retinol production were optimal at pH 6.5 and 60 °C, 2 g enzyme l?1, and 2,200 mg all-trans-retinal l?1 in the presence of 5 % (v/v) methanol, 1 % (w/v) hydroquinone, and 10 mM NADH. Under optimized conditions, the ADH produced 600 mg all-trans-retinol l?1 after 3 h, with a conversion yield of 27.3 % (w/w) and a productivity of 200 mg l?1 h?1. This is the first report of the characterization of a bacterial ADH for all-trans-retinal and the biotechnological production of all-trans-retinol using ADH.  相似文献

We report the bifunctional activity of the native ent-kaurene oxidase from Montanoa tomentosa (MtKO) and its N-terminal modified version (LMtKO) for producing both isokaurenoic acid and kaurenoic acid in Saccharomyces cerevisiae. The Km app of MtKO showed more affinity for ent-kaurene (80.5 µM) than for isokaurene (96.4 µM). Interestingly, LMtKO exhibited an increase of the affinity for isokaurene (79.6 µM) but simultaneously showed an enhancement in the Vmax for both substrates (32.6–38.9 μmol?1 mg?1 h?1). Biotransformation assays using isokaurene and yeasts containing LMtKO, resulted in 70% more production of isokaurenoic acid, when compared with the yields from yeasts expressing MtKO. Likewise, biotransformation assays using geranylgeraniol and double transformed cells of S. cerevisiae containing an optimized version the ent-kaurene synthase from Phaeosphaeria sp. L487 (optKS) and the LMtKO, produced ~25% more kaurenoic acid than the yeasts containing optKS and MtKO. The isokaurenoic acid synthesized by transgenic yeasts was tested for its anti-acetylcholinesterase and antimicrobial properties. Isokaurenoic acid generated a non-competitive inhibition on acetylcholinesterase, decreasing the Vmax from 0.0249 to 0.0104 mM min?1 but not affecting the Km (0.714 mM). The same diterpene showed antifungal activity against Fusarium oxysporum, Aspergillus niger and Phytophtora infestans with a minimum inhibitory concentration of 15.3, 18.3 and 19.2 µg mL?1, respectively.  相似文献

The cotton bollworm, Helicoverpa armigera (Hubner) (Lepidoptera: Noctuidae) is a polyphagous pest that exist all over the world. It is also very destructive in Turkey on various agricultural products. In this study, we have detected an alphabaculovirus from Helicoverpa armigera larvae collected from cotton field. The virus isolate was named as Helicoverpa armigera nucleopolyhedrovirus (HearNPV-O1) based on morphological and molecular properties. This is the first record of baculovirus from H. armigera in Eurasia region. Scanning electron microscopy examinations of polyhedral inclusion bodies (PIBs) showed their irregular morphology with average diameter of 0.85 to 1.25 μm. Transmission electron microscopy studies revealed that the nucleocapsids were multiple enveloped and bacilliform shaped. The size of nucleocapsid was found as 279 nm with a width of 56 nm. Digestion of viral genome by PstI generated 8 fragments with total size of 130.9 kbp. According to the phylogenetic analysis of the partial polyhedrin (polh), late expression factor 8 (lef8) and late expression factor 9 (lef9) genes, HearNPV-O1 isolate is very close to H. armigera MNPV Indian isolate-443. Five different concentrations of HearNPV-O1 between 103 and 107 were used in dose-response test on neonate, 3rd and 5th instars larvae of H. armigera. The highest dose (107) showed 92%, 88% and 57% mortality, respectively within 14 days. LC50 values of HearNPV-O1 isolate were calculated as 6?×?104, 7?×?104 and 8?×?106 PIBs/mL?1 against neonate, 3rd and 5th instars larval stages, respectively. These results demonstrate that HearNPV-O1 isolate may be a potential biocontrol agent to be utilized against H. armigera.  相似文献

Sublethal concentrations of chemical insecticides may cause changes in some behavioral characteristics of natural enemies such as functional responses. The residual effect of three synthetic insecticides including deltamethrin, fenvalerate and azadirachtin were studied on functional response of Habrobracon hebetor Say to Ephestia kuehniella Zeller larvae. Seven host densities (2, 4, 8, 16, 32, 64 and 96) were used during a 24 h period. The resulting data were appropriately fit to Type II functional response models in all treatments: (1) control (0.0916 h?1; and T h  = 0.2011 h); (2) deltamethrin (a = 0.0839 h?1; and T h  = 0.3560 h); (3) fenvalerate (a = 0.0808 h?1 and T h  = 0.3623 h); and (4) azadirachtin (a = 0.0900 h?1 and T h  = 0.2042 h). Maximum theoretical parasitism rate (T/T h ) was 119.34 estimated for control wasps. There was no significant difference between the values of attack rates (a and a + D a ) in all treatments while the handling time was statistically affected in female wasps treated with fenvalerate. Our findings will be useful in safe application of these insecticides in pest management programmes.  相似文献

The α-acetolactate decarboxylase (ALDC) can reduce diacetyl fleetly to promote mature beer. A safe strain Bacillus subtilis WB600 for high-yield production of ALDC was constructed with the ALDC gene saald from Staphylococcus aureus L3-15. SDS-PAGE analysis revealed that S. aureus α-acetolactate decarboxylase (SaALDC) was successfully expressed in recombinant B. siutilis strain. The enzyme SaALDC was purified using Ni-affinity chromatography and showed a maximum activity at 45 °C and pH 6.0. The values of K m and V max were 17.7 μM and 2.06 mM min?1, respectively. Due to the unstable property of SaALDC at low pH conditions that needed in brewing process, site-directed mutagenesis was proposed for improving the acidic stability of SaALDC. Homology comparative modeling analysis showed that the mutation (K52D) gave rise to the negative-electrostatic potential on the surface of protein while the numbers of hydrogen bonds between the mutation site (N43D) and the around residues increased. Taken together the effect of mutation N43D-K52D, recombinant SaALDCN43D-K52D showed dramatically improved acidic stability with prolonged half-life of 3.5 h (compared to the WT of 1.5 h) at pH 4.0. In a 5-L fermenter, the recombinant B. subtilis strain that could over-express SaALDCN43D-K52D exhibited a high yield of 135.8 U mL?1 of SaALDC activity, about 320 times higher comparing to 0.42 U mL?1 of S. aureus L3-15. This work proposed a  strategy for improving the acidic stability of SaALDC in the  B. subtilis host.  相似文献

Biocidal natural substances of botanical origin offer a promising ecofriendly option for controlling toxic cyanobacteria. Herein, we study 11 essential oils and some of their major components for their activity on Aphanizomenon gracile. On the basis of our results we support that Origanum vulgare and O. dictamnus, Ocimum basilicum, Eucalyptus meliodora, Melissa officinalis, and Pimpinella anisum exhibited the strongest activities, and the IC50/1d values of the extracts were calculated to be between 168.43 and 241.97 μg mL?1. When the major components of the biocidal essential oils were tested individually, (E)-anethole was found active, exhibiting an IC50/1d value of 71.35 μg mL?1. On the other hand, the half-life (t 1/2 ) of (E)-anethole was calculated at 1 h. A preliminary attempt of (E)-anethole microencapsulation was conducted, in order to slowly release this biocidal agent, increasing the residual life under open air conditions and thus the biological activity. Results were promising since the microencapsulated product exhibited better activity than did the non-formulated (E)-anethole. This is a first report on the biocidal activity of EOs and (E)-anethole on A. gracile and a preliminary indication of the microencapsulated (E)-anethole potential use as a natural biocidal in fresh waters.

The Antarctic Lake Wujka (62°09′28.3″S, 58°27′56.3″W), a shallow water body (Z m  = 1.38 m), situated at c.15 m from the seashore was sampled at two points (Sp 1 and Sp 2) at 3-day intervals from December 2003 to June 2004. The two sampling points differing in location and depth: Sp 1 (Z m  = 0.50 m) was the shallowest site, located near the lake outlet, while Sp 2 (Z m  = 1.38 m) was the deepest spot of the lake. The population density of Notholca squamula salina peaked in June (at 114 ind. l?1) at Sp 1, while at Sp 2 peaked in January (80 ind. l?1) and May (150 ind. l?1). Spearman non-parametric correlations with temperature, salinity, total dissolved solids, conductivity and pH revealed effects that characterize N. squamula salina as a species capable of surviving in a range of aquatic environments, but with a preference for high salinity, food and low temperature. It occurred in highest numbers when the diatom Achnanthes lanceolata var. rostrata (Øestrup) Hust., normally a benthic species, was stirred up into the water during storms that also raised the lake’s salinity to above 20 psu.  相似文献

Impact of different levels of elevated CO 2 on the activity of Frankia (Nitrogen-fixing actinomycete) in Casuarina equisetifolia rooted stem cuttings has been studied to understand the relationship between C. equisetifolia, Frankia and CO2. The stem cuttings of C. equietifolia were collected and treated with 2000 ppm of Indole Butyric Acid (IBA) for rooting. Thus vegetative propagated rooted stem cuttings of C. equisetifolia were inoculated with Frankia and placed in the Open top chambers (OTC) with elevated CO2 facilities. These planting stocks were maintained in the OTC for 12 months under different levels of elevated CO2 (ambient control, 600 ppm, 900 ppm). After 12 months, the nodule numbers, bio mass, growth, and photosynthesis of C. equisetifolia rooted stem cuttings inoculated with Frankia were improved under 600 ppm of CO2. The rooted stem cuttings of C. equisetifolia inoculated with Frankia showed a higher number of nodules under 900 ppm of CO2 and cuttings without Frankia inoculation exhibited poor growth. Tissue Nitrogen (N) content was also higher under 900 ppm of CO2 than ambient control and 600 ppm levels. The photosynthetic rate was higher (17.8 μ mol CO2 m?2 s?1) in 900 ppm of CO2 than in 600 ppm (13.2 μ mol CO2 m?2 s?1) and ambient control (8.3 μ mol CO2 m?2 s?1). This study showed that Frankia can improve growth, N fixation and photosynthesis of C. equietifolia rooted stem cuttings under extreme elevated CO2 level conditions (900 ppm).  相似文献
