This study was designed to measure ovarian hormones prior to and during the first estrous cycle in guinea pigs. Blood was obtained from 12 animals throughout the first estrous cycle. Ovaries and peripheral serum were obtained from 25 additional animals at various stages of development prior to and after first ovulation. Estradiol, progesterone, androstenedione, and testosterone were measured in all sera and half of the ovaries. The remaining ovaries were fixed for histology. Serum estradiol was nondetectable until a few days before first ovulation, but was present in the ovary throughout development. Serum progesterone was nondetectable until the day of ovulation, but the luteal phase pattern was similar to that observed in adults. Serum androgens were detectable throughout development, with androstenedione higher than testosterone. The immature ovary contained more testosterone than androstenedione, but this pattern was reversed after ovulation. These results indicate that the immature ovary in the guinea pig contains minimal amounts of estradiol and progesterone, the first estrous cycle is similar to that in adults, and that the pattern of ovarian androgen content changes during the peripubertal period. 相似文献

The program of acquisition of adult metabolic phenotypes was studied in three jaw muscles in order to determine the link between muscle metabolism and functional development. During early postnatal stages, there were similar transitions in the masseter, anterior digastric, and internal pterygoid muscles with respect to fiber growth, fiber type composition, and whole muscle energy metabolism. Oxidative capacity, as judged by the activities of the enzymes succinate dehydrogenase (SDH), malate dehydrogenase (MDH), and beta-hydroxyacyl CoA dehydrogenase (beta OAC), rose sharply after birth to reach near maximal levels by 3 weeks. The capacities for glycolytic metabolism represented by lactate dehydrogenase (LDH), and for high-energy phosphate metabolism represented by adenylokinase (AK) and creatine kinase (CK) activities, rose gradually, not reaching peak values until 6 weeks or later. Thus, the maturation of oxidative metabolism preceded that of glycolytic metabolism in the developing jaw muscles. This was documented for individual fibers in the masseter muscle. Differential metabolic maturation among the jaw muscles was evident beyond 3 weeks. All three jaw muscles attained their specific adult fiber-type profile by about 6 weeks. This maturation program differed from that of hindlimb muscles [Nemeth et al., J Neurosci 9:2336-2343, 1989] and diaphragm muscle [Kelly et al., J Neurosci 11:1231-1242, 1991], reflecting their differential energy demands for contractile performance. 相似文献

Expression, activity, and subcellular localization of testicular hormone-sensitive lipase during postnatal development in the guinea pig 总被引:3,自引:0,他引:3
The present work reports on testicular hormone-sensitive lipase (HSL), the biological significance of which has been documented in male fertility. The HSL protein levels and enzymatic activity were measured, respectively, by densitometry of immunoreactive bands in Western blots, performed with antibodies against recombinant rat HSL, and by spectrophotometry in seminiferous tubules (STf) and interstitial tissue (ITf) enriched fractions generated from neonatal, pubertal, and adult guinea pig testes. In addition, HSL was studied in subcellular fractions obtained from STf isolated from adult testes and in epididymal spermatozoa (Spz). A 104-kDa HSL protein was detected in STf and ITf, the expression and activity of which increased with testicular development. Three immunoreactive bands of 104, 110, and 120 kDa were detected in the lysosomal subfraction, and two bands of 104 and 120 kDa were detected in Spz. The HSL activity was positively correlated with free (FC) and esterified (EC) cholesterol ratios in STf and ITf, but not with triglyceride (TG) levels, during testicular development. Immunolabeling localized HSL to elongated spermatids and Sertoli cells, where its distribution was stage-dependent, and within the cells lining the excurrent ducts of the testis. The findings of the 104- and 120-kDa HSL immunoreactive bands and of HSL activity in Spz as well, as the detection of the 104-, 110-, and 120-kDa immunoreactive bands in lysosomes, suggest that part of HSL may originate from germ cells and be imported in Sertoli cells. The HSL protein levels and enzymatic activity in ITf and STf were positively correlated with serum testosterone levels during development. To the best of our knowledge, this study is the first to contribute insights regarding the impact of HSL on FC:EC cholesterol ratios and TG levels in the interstitial tissue and tubules in relation to serum testosterone levels during postnatal development, and regarding the immunolocalization of the enzyme in regions of the male gamete consistent with spermatozoa-oocyte interaction. 相似文献

The effects of anoxia on cardiac action potentials were studied at different stages of development of embryonic chick heart. The plateau phase of the action potential was markedly depressed by anoxia in old (15–16 days old) embryonic hearts without any significant change in other configurations of the action potential. Raising the concentration of glucose in the external fluid prevented the shortening of the action potential plateau by anoxia, and, conversely, a further reduction was observed in glucose-free media. In young (3–4 days old) embryonic hearts, the shortening of the action potential plateau was not produced by anoxia, but was produced by a combination of anoxia and glucose deprivation. When the action potential was shortened by anoxia in old hearts and by anoxia plus glucose deprivation in young hearts, isoproterenol (10?5M), dibutyryl cyclic 3′,5′-adenosine monophosphate (dBcAMP: 1 mM) plus aminophylline (1 mM), and calcium ion (3–6 mM), partially reversed the shortened action potential in old hearts, but did not produce any prolongation in the young hearts. Therefore, the cation channels responsible for the action potential plateau in young hearts may be pharmacologically different from those in old hearts. Permeability of fetal skin to tritiated water was measured in vitro using samples taken from the back and flanks of 21 guinea pig fetuses whose gestational age ranged from 30 to 67 days (term = 68 days). From 30 to 45 days, fetal skin was relatively permeable to water, with a permeability coefficient for unidirectional, diffusional transfer of labelled water that averaged 0.372 +/- 0.041 (SEM) X 10(-4) cm/s. Then during a 5-10 day interval, the measured permeability coefficient decreased abruptly to very low and barely detectable levels. These changes took place at the time during gestation when others have shown the skin becomes keratinized and growth of new hair follicles is completed. Thus these findings are consistent with a relatively free exchange of water between amniotic fluid and fetal interstitium across the skin during the first two-thirds of gestation and then with further maturation an abrupt functional separation between these fluid compartments during the last third of gestation. 相似文献

The pattern of adrenergic nerves was visualized and norepinephrine (NE) quantified in guinea pig ovaries from fetal day 50 to term and on days 1, 5, 10, 20 and 28 postnatally. Before birth, there were a few perivascular adrenergic nerves and correspondingly low ovarian NE levels. After birth and through day 20, nerves gradually grew into adjacent stroma. By day 28, there was a proliferation of beaded adrenergic nerves around blood vessels and into adjacent stroma, which was reflected by significantly increased ovarian levels of NE. These findings are discussed in relation to antral follicle development, atresia, and interstitial gland formation in the ovaries of prepubertal guinea pigs. 相似文献

During development fetal arterial oxygen tension falls, whereas cerebral oxygen consumption rises due to an increase in cerebral metabolism. To compensate for this increase in oxygen consumption, blood flow and therefore oxygen delivery to the cerebrum rises. To determine whether during development oxygen delivery to the cerebrum meets cerebral oxygen consumption, we measured the concentrations of high-energy phosphates and glycolytic intermediates in the cerebral cortex of fetal guinea pigs at different gestational ages. During development there was no change in the concentrations of adenosine triphosphate, creatine phosphate, adenosine monophosphate, and lactate. However, cerebral concentrations of adenosine diphosphate increased and those of glucose decreased. Our results suggest that the increase in fetal cerebral oxygen delivery during development meets cerebral oxygen consumption with increasing gestational age. We speculate that the measured rise in the concentrations of adenosine diphosphate may accelerate glycolysis during development and therefore may cause a rise in both cerebral blood flow to maintain oxygen delivery. 相似文献

Summary The villi of the caecal mucosa in postnatal rats were studied using both scanning electron and light microscopy.On the day of birth, numerous villi of various sizes and shapes were present on the caecal mucosa. After the 5th day, the villi decreased very rapidly in length and in number. A strong constriction was observed at the basal region of the caecal villi. During postnatal days 5 9 the villi probably separated and disappeared from the caecal mucosa. No villi were observed in rats that were over 10 days of age. 相似文献

Brief ischemia or hypoxia has been found to protect the heart against susbsequent long-lasting ischemia and to improve contractile dysfunction as well to reduce cell necrosis and the incidence of lethal arrhythmias. This phenomenon, termed preconditioning (PC) has been demonstrated in different species. However, little is known about PC in guinea pigs. Moreover, electrophysiological changes underlying protection have not been studied so far in conjuntion with force recovery in a setting of PC. The aim of the study was to study PC in a guinea pig papillary muscle, using recovery of contractility after long hypoxic challenge as the main end-point of protection, and to investigate concominant electrophysiological alterations. In guinea pig papillary muscle preparations contracting isometrically (paced at 2 Hz), transmembrane action potentials (AP) and developed force (DF) were recorded by conventional microelectrode technique and a force tranducer. In addition, effective refractory periods (ERP) were determined. Hypoxia was induced by superfusion with 100% N2 (pO2 < 5 kPa) and pacing at 3,3 Hz. In the control group, long hypoxia lasted for 45 min and was followed by 30 min reoxygenation. In the PC group, muscles were subjected to 5 min hypoxia followed by 10 min recovery prior to sustained hypoxia/reoxygenation. Results: Long hypoxia induced a similar depression of DF in both, PC and control groups. However, a loss of contractile activity occured earlier in the PC group. AP duration and ERP decreased faster and were significantly shorter after PC. Upon reoxygenation, preconditioned muscles showed significantly better recovery of function (DF 86% of prehypoxic value vs. 36% in controls; p < 0,05). AP and ERP were completely restored in both, PC and control groups. Guinea pig papillary muscle can be preconditioned with a brief hypoxic challenge against contractile dysfunction upon long-lasting hypoxia/reoxygenation. Shortening of AP and loss of contractility occured more quickly during hypoxia and may participate in the protective effect of preconditioning. Possible mechanisms might involve facilitated opening of KATP-dependent channels. 相似文献

本研究旨在探讨组胺在哮喘豚鼠气道重塑中的作用。将50只健康雄性豚鼠随机分为5组。正常对照组:雾化吸入蒸馏水8周;哮喘模型组:致敏后雾化吸入卯白蛋白(ovalbumin,OVA)8周;哮喘模型延续组:致敏后雾化吸入OVA14周;组胺组:致敏后雾化吸入OVA14周,最后6周同时加用组胺:组胺受体拈抗剂组:致敏后雾化吸入OVA14周,最后6周同时加用组胺受体拈抗剂。检测血清组胺、Na^+、Cl^-浓度、动脉血氧分压(PaO2)、动脉血二氧化碳分压(PaCO2)、pH、实际碳酸氢盐(actual bicarbonate,AB)、标准碳酸氧盐(standard bicarbonate,SB)以及气道壁粘膜层、基底膜层和平滑肌层厚度。所得结果如下:(1)哮喘模型组血清组胺浓度、气道擘厚度明显高于正常对照组(P〈0.01);哮喘模型延续组明显高于哮喘模型组(P〈0.01);组胺组明显高于哮喘模型延续组(P〈0.01):组胺受体拮抗剂组低于哮喘模型延续组(P〈0.05,0.01)。(2)哮喘模型组PaO.小于正常对照组(P〈0.01);哮喘模型延续组PaO2、pH、AB、SB小于哮喘模型组(P〈0.01),而PaCO2大于哮喘模型组(P〈0.01);组胺组PaO2、pH、AB、SB小于哮喘模型延续组(P〈0.01),而PaCO2大于哮喘模型延续组(P〈0.01);组胺受体拮抗剂组PaO2、pH、AB、SB高于哮喘模型延续组(P〈0.01),而PaCO2低于哮喘模型延续组(P〈0.01);血清Na^+、Cl^-浓度各组间差异不明显。以上结果提示:(1)组胺在哮喘气道重塑中起介导作用。(2)应用组胺受体拈抗剂对防治哮喘气道重塑有一定作用。(3)PaO2、pH与气管、肺门支气管、肺外周小气道气道擘厚度呈负相关(P〈0.01),而PaCO2、阴离子间隙(anion gap,AG)值与上述气道壁厚度呈止相关(P〈0.01)。 相似文献
