Insect α-amylase inhibiting and/or growth inhibiting activities of proteinaceous inhibitors from red kidney bean (Phaseolus vulgaris) and hard red winter wheat (Triticum aestivum) were examined. The bean inhibitor was most effectivein vitro against α-amylases from the red flour beetle (Tribolium castaneum) and the confused flour beetle (T. confusum), followed by those from the rice weevil (Sitophilus oryzae) and yellow mealworm (Tenebrio molitor). The insect enzymes were from two- to 50-fold more susceptible than human salivary α-amylase. When the inhibitors were added at a 1% level to a wheat flour plus germ diet, the growth of red flour beetle larvae was slowed relative to that of the control group of larvae, with the bean inhibitor being more effective than the wheat inhibitor. Development of both the red flour beetle and flat grain beetle (Cryptolestes pusillus) was delayed by 1% bean inhibitor, but development of the sawtoothed grain beetle (Oryzaephilus surinamensis) and lesser grain borer (Rhyzopertha dominica) was not affected by either the bean or wheat inhibitor at the 1% level. Rice weevil adults fed a diet containing 1% bean or wheat inhibitor exhibited more mortality than weevils fed the control diet. When the wheat amylase inhibitor was combined with a cysteine protease inhibitor, E-64, and fed to red flour beetle larvae, a reduction in the growth rate and an increase in the time required for adult eclosion occurred relative to larvae fed either of the inhibitors separately. The bean inhibitor was just as effective alone as when it was combined with the protease inhibitor. These results demonstrate that plant inhibitors of insect digestive enzymes act as growth inhibitors of insects and possibly as plant defense proteins, and open the way to the use of the genes of these inhibitors for genetically improving the resistance of cereals to storage pests. Bruchid beetle larvae cause major losses in grain legume crops throughout the world. Some bruchid species, such as the cowpea weevil (Callosobruchus maculatus) and the Mexican bean weevil (Zabrotes subfasciatus), are pests that damage stored seeds. The Mediterranean flour moth (Anagasta kuehniella) is of major economic importance as a flour and grain feeder; it is often a severe pest in flour mills. Plant lectins have been implicated as antibiosis factors against insects. Bauhinia monandra leaf lectin (BmoLL) was tested for anti-insect activity against C. maculatus, Z. subfasciatus and A. kuehniella larvae. BmoLL produced ca. 50% mortality to Z. subfaciatus and C. maculatus when incorporated into an artificial diet at a level of 0.5% and 0.3% (w/w), respectively. BmoLL up to 1% did not significantly decrease the survival of A. kuehniella larvae, but produced a decrease of 40% in weight. Affinity chromatography showed that BmoLL bound to midgut proteins of the insect C. maculatus. 33 kDa subunit BmoLL was not digested by midgut preparations of these bruchids. BmoLL-fed C. maculatus larvae increased the digestion of potato starch by 25% compared with the control. The transformation of the genes coding for this lectin could be useful in the development of insect resistance in important agricultural crops.  相似文献

Insects feeding on stored grains cause considerable damage to harvested cereals and legumes every year. The use of α-amylase inhibitors to interfere with the pest’s digestion process has become an interesting alternative biocontrolling agent. In this study, we have studied the interactions of α-amylase inhibitors from Albizia lebbeck seeds with the amylases of coleopteran and lepidopteran insect pests. We isolated and purified the α-amylase inhibitor using acetone precipitation and gel filtration chromatography. Two prominent activity bands of α-amylase inhibitors were detected in electrophoretic analysis using 8% starch PAGE. We found that the α-amylase inhibitor, isolated as a monomer, had a molecular weight of 14.4 kDa. The α-amylase inhibitor was purified 36.15-fold with gel filtration chromatography. Its specific activity was determined at 14.4 U/mg/min. Feeding analysis of Tribolium confusum larvae on a diet containing purified α-amylase inhibitor from Albizia lebbeck revealed that survival of the larvae was severely affected, with the highest mortality rate occurring on the fifth day of feeding. We found that the isolated α-amylase inhibitor inhibits T. confusum and Helicoverpa armigera α-amylases in electrophoretic analysis as well as in solution assays. The isolated α-amylase inhibitor was found to be resistant to commercial protease as well as T. confusum and H. armigera digestive proteinases. The isolated α-amylase inhibitor was degraded by heating above 60°C. Our results suggest that A. lebbeck α-amylase inhibitor could be a useful future biocontrolling agent.  相似文献

The effects of eight diets (atta flour, wheat flour, self-rising flour, rice flour, custard powder, corn flour, tapioca starch, and potato starch) on the development of the red flour beetle, Tribolium castaneum (Herbst), reared at 29-31 degrees C and 66-70% RH were assessed. Five pairs of male and female T. castaneum were reared on the respective diets for 28 d before the experimental setup was dismantled and adult counts were recorded. In another experiment, the insects were allowed to mate and oviposit in each flour or starch type over a period of 7 d before being removed. The counting of pupae and adult emergence began on the day of emergence and was continued on a daily basis until day 140. Proximate analysis was performed for chemical composition of each diet, and the numbers of new adults that developed were found to be positively correlated (r2 = 0.97; P < 0.05) with the protein content and negatively correlated (r2 = 0.93; P < 0.05) with the carbohydrate content. For T. castaneum, the suitable diets were ranked as follows: atta flour > wheat flour > self-rising flour > rice flour > custard powder > corn flour > tapioca starch > potato starch. T. castaneum larval development to the pupal and adult stages developed significantly faster in atta flour (P < 0.05) than in the other diets, and the greatest number of progeny was produced from beetles reared on atta flour. Fewer adults emerged from wheat flour, self-rising flour, and rice flour, and no new emergences were recorded for the remaining diets. Developmental rate was much slower in beetles reared on diets in which a low number in progeny was produced. These data illustrate that different diets can influence the sustainability of these insects and affect their development and growth.  相似文献

Tribolium confusum is a major pest of grains and grain products. Some legumes hitherto not used as major food are gaining interest due to increase in human population and the need of food and feeds. Susceptibility of fresh and processed Glycine max and Cajanus cajan flours, and white wheat flour (control) to T. confusum was studied towards effective storage techniques and cheap insect diet. Ten copulating pairs of the beetle were introduced into twenty grams of each flour. Number of eggs laid, emerged larvae, pupae, eclosed adults, and duration of development of each stage was used as indices of survival of T. confusum. Proximate composition, saponin; and α-amylase and protease inhibitors determined suitability of flours as insect diet. This work discusses the significance of proximate composition and anti-nutrient properties in fresh and roasted processed flours on the development and survival of T. confusum.  相似文献

Amylase inhibitors (AIs) are suitable candidates for protecting plants and their products from attacks by herbivorous and granivorous insects. However, detailed studies of the suppressive effects of AIs on target and non‐target insects are necessary before their application in post‐harvest protection. To address this issue, laboratory bioassays were used to test the effect of the non‐proteinaceous inhibitor acarbose on a stored product pest, the flour moth Ephestia kuehniella (Zeller) (Lepidoptera: Pyralidae), and its parasitoid Venturia canescens (Gravenhorst) (Hymenoptera: Ichneumonidae). Two sublethal concentrations (0.001 and 0.0001%, wt/wt) of acarbose were incorporated into the diet of parasitized and unparasitized larvae of E. kuehniella. Development time and fresh body weight of the larvae, together with the size of the wasps, were compared for insects reared on acarbose‐treated and control diets. On the diet containing 0.001% acarbose, the developmental time was longer and relative weight gains of the E. kuehniella larvae were lower, but the weight of the larvae prior to pupation was similar to that of the control. The acarbose did not have a suppressive effect on the parasitoid V. canescens; in fact the wasps that emerged from the hosts reared on a diet containing 0.0001% acarbose were on average larger and heavier than the controls. These results demonstrate that it might be possible to enhance the control of stored product pests by using both biological control and AIs.  相似文献

Larvae of the Angoumois grain moth readily established in refined diets moulded into pellets. A bioassay based on larval establishment, weight, and numbers of adults was used to determine the insect's major nutritional requirements. The results showed that test diets containing corn starch (49–64%), glycerol (15–19%), casein (9%), yeast (7·5%), and wheat germ (2·6%) permitted good establishment and normal development. No further improvements were obtained when diets were supplemented with a mineral or vitamin mixture, cholesterol, or ascorbic acid. It was concluded that yeast and wheat germ supplied the insect with adequate amounts of lipids, vitamins, and minerals. However, even the best diets lacked some undetermined growth promoting factor because larvae grew better in a natural product control diet.  相似文献

Females ofSpodoptera littoralis Boisd. (Lepidoptera: Noctuidae) with different feeding experiences during their larval development were tested for their ovipositional response to methanol extracts of larval frass and semisynthetic diets. The effect of the following frass, diet and diet component extracts was tested: (a) frass fromS. littoralis orAgrotis segetum larvae fed on a potato-based diet; (b) frass fromS. littoralis larvae fed on a wheat germ-based diet; (c) potato and wheat germ-based diets; and (d) potatoes and wheat germ. Ovipositing females without prior experience of the potato diet were deterred by extracts of: (1) larval frass from either species fed on potato diet; (2) the potato-based diet; (3) potato. Also females with experience of the potato diet during only a part of their larval development were deterred from oviposition by frass of larvae reared on the potato diet and by the diet itself. However, for females reared on the potato diet for their entire larval development, oviposition was no longer deterred by either of the three extracts listed above. Extracts of: (1) frass from larvae of either species reared on wheat germ diet: (2) the wheat germ diet; or (3) wheat germ did not significantly affect oviposition. Females with ablated antennae were still deterred by frass extracts from larvae fed on potato diet, when they had been reared on the wheat germ diet. In feeding experiments, larvae of larval stage one and of larval stage three-four reared on either of the two diets preferred to feed on the wheat germ diet. However, the preference was significantly stronger for larvae with no prior contact with the potato diet. The effect of larval experience on the loss of oviposition-deterring activity by extracts of larval frass, diets and diet components is discussed in view of induction and selection.  相似文献

Biological and biochemical parameters of a flightless strain of Harmonia axyridis, fed on a pork liver-based artificial diet and on Ephestia kuehniella eggs as controls, were compared. The diet-grown larvae showed a significantly longer developmental time and a lower adult emergence rate compared to control larvae. The weights of the newly emerged adults were significantly higher for adults fed E. kuehniella eggs during their larval stages than fed the artificial diet. In contrast, larval food source had no effect on the duration of the pre-oviposition period or adult longevity. For adults fed on E. kuehniella eggs as larvae, a significantly longer pre-oviposition period, lower daily weight gain and fecundity were found for the diet-fed females compared to those fed on E. kuehniella eggs throughout the life span. The adult food source had no significant effect on longevity and fertility. Lower amino acid and fatty acid contents (in particular C16:1 and C18:3n-3) were found for the prepupae and newly emerged females obtained from diet-reared larvae compared to controls. Deficiencies in fatty acids C16:1 and C18:3n-3 were also observed in females obtained from E. kuehniella egg-reared larvae and fed on diet from adult emergence. The analyses of the foods showed deficiencies in artificial diet, especially for some amino and fatty acids. The results suggest a non-optimal composition of the artificial diet and some possibilities for its improvement. However, this polyphagous predator could be reared from first instar larvae to fully reproductive adults on a pork liver-based artificial diet.  相似文献

Ladybird beetles have successfully been used to control different pests. Cheilomenes sexmaculata (Fabricius) and Oenopia conglobata contaminata)Menetries((Coleoptera: Coccinellidae) are two dominant and efficient predators of the common pistachio psylla, Agonoscena pistaciae (Hem.: Psyllidae) in Iran. In the current study, the impact of two diets, i.e., nymphs of A. pistaciae and eggs of Ephestia kuehniella Zeller (Lep.: Pyralidae), were investigated on the cold hardiness, supercooling point (SCP), and lethal temperature of different life stages of the coccinellids. The results suggested that the eggs of E. kuehniella are a suitable diet for both predators. In general, beetles of O. conglobata contaminata were more cold tolerant than those of C. sexmaculata. The SCP of the adults of C. sexmaculata, feeding on psyllid was about −15 °C, whereas the SCP of the beetles, feeding on the eggs of flour moth, was about −19 °C. However, the diets had no significant effects on the SCP of O. conglobata contaminata. For both ladybird beetles, eggs were the most sensitive stage, and adulthood was the most tolerant developmental stage. No eggs survived at zero and subzero temperatures. The survival of C. sexmaculata increased from 6.25% for adults fed on psyllids to 13.75% for those fed on flour moth eggs after 24 h exposure to cold at −4 °C. The survival of O. conglobata contaminata adults after exposure to cold at −4 °C for 24 h raised from 28.75% for adults fed on psyllids to 42.50% for those fed on flour moth eggs. Regardless of the diet, both beetles were considered to be chill-intolerant insects as the most mortality occurred above the SCP.  相似文献

The larval midgut amylase of Tenebrio molitor L. may be classified as a typical -amylase on the basis of its action pattern on amylopectin, glycogen, and β-amylase limit dextrin. When the development is on the preferred diet of wheat-bran, the amylatic activity of the larvae is impaired by an -amylase inhibitor present in wheat. The inhibitor has been found to be stable to digestion by the larvae. Wheat β-amylase plays the major role in the digestion of starch by larvae. The carrion beetle subfamily Silphinae (Coleoptera: Silphidae) contains dominant macroinvertebrates of soil ecosystems in temperate zones. However, their feeding habits, which determine the role of each species in the ecosystem, have not been sufficiently studied. Moreover, although a diet shift from necrophagy on vertebrate carcasses to predatory feeding on invertebrates is known to occur in this subfamily, the processes and mechanisms of this shift have also been inadequately addressed. We examined female fecundity and larval development on various diets in a Silphinae species, Necrophila (Eusilpha) japonica (Motschulsky). The experimental diets included a meat diet and various invertebrate diets, which reflect the ‘ancestral’ feeding habit in Silphinae, necrophagy, and the ‘derivative’ feeding habit, predatory feeding. Female fecundity was significantly higher on the meat diet (minced beef) than on an insect larvae diet (mealworms and dipteran larvae) but did not significantly differ from that on an earthworm diet. Larval developmental performance was significantly higher on the earthworm diet than on the meat and insect larvae diets. Our results for larval development were consistent with those of previous stable isotope analyses of the same species, in which isotopic values of larval samples agree with those of hypothetical consumers that utilize earthworms. The consistency of results among different methods indicates that N. japonica larvae are most likely earthworm feeders. In contrast, our results for the female fecundity experiment differed from those of previous stable isotope analyses, in which vertebrate carcasses unlikely serve as the staple diet of adults in the field; thus, the feeding habits of N. japonica adults remain unresolved. Our observations that females and larvae performed best on the meat and earthworm diets, respectively, may indicate that, in Silphinae, the diet shift from necrophagy to predatory habits occurs earlier in larvae than in adults.  相似文献

Developing grains of pearl millet ( Pennisetum typhoides Burm. S & H cv. PIB 155) were sampled and analyzed for starch and its free-sugar precursors. The activities of invertase, sucrose-ADP (UDP) glucosyl transferase and of α-amylase and β-amylase in relation to the rate of starch accumulation in the developing grain were assayed. By culturing detached ears, the incorporation of 14C from free sugar precursors to starch was studied. The starch content gradually increased until grain maturity. The rate of starch accumulation was maximum around 12 days after anthesis. Around this period, the activities of sucrose-ADP(UDP) glucosyl transferase and α-amylase, β-amylase were also at a peak. Invertase activity was high during the early period of grain development but gradually declined as the grains matured. In the most actively metabolising milky grains, incorporation of 14C from [14C]-sugars to starch was maximum in the mid mid-milky grains. Addition of 20 m M K+ to the culture solution did not affect the incorporation of 14C from supplied sucrose to the free sugar pool and to the starch of the grain, but Mg2+ supply at 20 m M concentration lowered 14C incorporation from exogenous sucrose to grain free sugars, although the utilization of the latter for starch synthesis was enhanced.  相似文献

Winter wheat was compared with a barley which has a normal amylopectin/amylose starch ratio, and waxy barleys which have essentially 100% amylopectin starch. In one trial, broiler chicks were fed on one of seven diets which had grain portions of all wheat (100W), 50% wheat–50% Compana (normal) barley (50W–50C), 25% wheat–75% Compana (25W–75C), 100% Compana (0W–100C), 50% wheat–50% Wapana (waxy) barley (50W–50Wa), 25% wheat–75% Wapana (25W–75Wa), or 100% Wapana (0W–100Wa). Body weights (g) at 7 weeks were (100W) 1659, (50W–50C) 1523, (25W–75C) 1401, (0W–100C) 1396, (50W–50Wa) 1503, (25W–75Wa) 1403 and (0W–100Wa) 1293. Dressing percentage was not affected by diet but mortality was greater on 0W–100C. Metabolizable energy (kJ/g dry matter) values determined on non-laying Leghorn hens were 12.93 ± 0.38 and 12.76 ± 0.46 for the Compana and Wapana barley, respectively. In a second trial, addition of a food-grade diastase at 2 g per kg of feed did not affect body weight on wheat-based diets, increased weights over corresponding control diets for Wapana-based diets but gave inconsistent results for Washunopana, a hull-less waxy barley with an active α-amylase-like activity on the starch granules. Group feed values indicated that enzyme supplements improved feed efficiency for barley diets but metabolizable energy values were similar for diets with or without diastase additions.  相似文献

Aims:  To clarify the taxonomic status of strain ZG0656 and analyse the effects of its acarviostatin products on blood glucose levels in mammalian systems.
Methods and Results:  Our program to screen for new α-amylase inhibitors led to the isolation of strain ZG0656. The polyphasic taxonomic study revealed that strain ZG0656 represents a novel variation of Streptomyces coelicoflavus , for which we propose the name S. coelicoflavus var. nankaiensis . Four chemically distinct α-amylase inhibitors, acarviostatins I03, II03, III03 and IV03, were isolated from strain ZG0656. Acarviostatins III03 and IV03 are both novel oligomers. All four acarviostatins are mixed noncompetitive porcine pancreas α-amylase inhibitors. Acarviostatin III03 is the most potent α-amylase inhibitor known to date. Moreover, in the in vitro and in vivo experiments, acarviostatins III03 showed significant inhibition of starch hydrolysis and glucose transfer to blood.
Conclusions:  Strain ZG0656 is a novel variation of S. coelicoflavus, whose products are novel effective α-amylase inhibitors. Among the products, acarviostatins III03 could significantly depress blood glucose levels in mammalian systems and be developed towards a possible therapeutic agent for diabetes.
Significance and Impact of the Study:  Acarviostatin III03 is the most potent α-amylase inhibitor known to date. The oligomer will benefit the research on the relationship between α-amylase and various inhibitors and will offer more choices in diabetes treatments.  相似文献

The objective of this research was, based on biological studies, to determine and adequate diet for rearing Corcyra cephalonica (Stainton, 1865) in the laboratory so as to permit the rearing of this factitious host for Trichogramma mass production. The research was conducted at 25 +/- 1 degrees C, RH 60 +/- 10% and photophase of 14 hours. Six artificial diets were compared: a) whole wheat flour (48.5%), ground rice (48.5%) and sugar (3%); b) ground rice (97%) and sugar (3%); c) whole wheat flour (48.5%), rice flour (48.5%) an sugar (3%); d) whole wheat flour (97%) and yeast (3%); e) wheat germ (97%) and yeast (3%); f) rice bran (94%), sugar (3%) and yeast (3%); f) rice bran (94%), sugar (3%) and yeast (3%). All of the diets studied permitted the development of C. cephalonica although the diets with wheat germ and yeast and that consisting of rice bran, sugar and yeast proved to be the most adequate for rearing the moth. These diets reduced the total (egg-adult) cycle, shortened the egg laying period, and produced heavier adults. Studies on the fertility life tables showed that higher net reproduction rates (Ro) and finite ratio of increase (lambda) were obtained from adults reared on these diets.  相似文献

Rice (Oryza sativa var. Nipponbare) was transformed with an artificial avidin gene. The features of this construct are as follows: (1) a signal peptide sequence derived from barley alpha amylase was added at the N-terminal region, (2) codon usage of the gene was optimized for rice, and (3) the gene was driven by rice glutelin GluB-1, an endosperm-specific promoter. Avidin was produced in the grain of the transgenic rice but not in the leaves. The concentration of avidin in the kernels was about 1,800 ppm. All larvae of the confused flour beetle (Tribolium confusum) and Angoumois grain moth (Sitotroga cerealella) died when fed transgenic avidin rice powder or kernels, respectively, whereas most of the test insects developed into adults when they were fed a nontransgenic rice control diet. Avidin extracted from the transgenic rice kernel lost most biotin-binding activity after 5 min heating at 95 degrees C.  相似文献

1. Combinations of a cysteine proteinase inhibitor (CPI) and serine proteinase inhibitors (SPI) in wheat germ diets were toxic to larvae of the red flour beetle, Tribolium castaneum, when tested at levels where individual inhibitors were nontoxic.2. Mixtures of 0.1% (w/w) CPI (E-64) plus 1% of either of three plant SPIs (soybean Kunitz trypsin inhibitor, soybean Bowman-Birk trypsin-chymotrypsin inhibitor, or lima bean trypsin inhibitor) inhibited T. castaneum growth, resulting in 82–97% reduction in larval weight gain 17 days after hatching and 40–60% mortality.3. Supplemention of diet containing 0.1% E-64 plus 1% soybean Kunitz trypsin inhibitor (STI) with a mixture of amino acids at 7% caused a partial reversal of the growth inhibition, with 91% of the larvae surviving.4. Diet containing 0.1% E-64 plus either 5 or 10% STI resulted in 100% mortality of the larvae during the first or second instar.5. Addition of a mixture ofamino acids at 20% to the 0.1% E-64 plus 10% STI diet allowed 89% of the larvae to develop into adults.6. The synergism between different classes of proteinase inhibitors in the insect's diet that enhances growth inhibition and toxicity demonstrates the potential for an insect pest management strategy involving the coordinated manipulation of two or more types of digestive enzyme inhibitor genes in plants.  相似文献

Abstract: Water-soluble carbohydrates from (a) living sapwood of Scots pine ( Pinus sylvestris L); (b) dry sapwood blocks from Scots pine (EUROPEAN STANDARD EN 46, 1988); and (c) faeces from Hylotrupes bajulus (L) larvae feeding from these blocks were analysed. Living sapwood contains high amounts of sucrose, glucose, fructose and starch. These carbohydrates seem to be hydrolysed and metabolized during the production of the sapwood blocks. The faeces contain carbohydrates which are not present either in living sapwood or in the sapwood blocks: xylose and an oligosaccharide of uncertain composition. This oligosaccharide is the dominant water-soluble carbohydrate in the faeces. In contrast to living sapwood and sapwood blocks, the faeces contain considerable amounts of soluble β-glucans. The results show that the composition and the amounts of carbohydrates differ significantly from that in the sapwood blocks (fodder for the larvae). The α-glucans (starch) present in the wood are not significantly utilized by the larvae. Regarding soluble carbohydrates, diet wood has only little in common with the living wood of the standing tree.  相似文献

We determined that the number of insect fragments, quantified using the standard flotation method, in flour milled from wheat infested with larvae, pupae, or preemergent adults of the lesser grain borer, Rhyzopertha dominica (F.), was proportional to infestation level. Wheat infested with a single preemergent adult contributed 28 and 10x as many fragments as wheat infested with a single larva or pupa, respectively. Using regression models that were developed from these data, we predicted that the maximum infestation level that would result in flour with fragment counts below the Food and Drug Administration defect action level (75 fragments/50 g of flour) was 0.95 and 1.5% (380-640 infested kernels/kg of wheat) for pupae and larvae, but it decreased to 0.05% (20 infested kernels/kg) when the grain was infested with preemergent adults. We also reexamined the accuracy and sensitivity of near-infrared spectroscopy (NIRS) for detecting insect fragments in flour by testing three different NIR spectrometers. NIRS-predicted numbers of insect fragments were correlated with the actual number of fragments. NIRS is less precise than the standard flotation method, but it is rapid, nondestructive, does not require extensive sample preparation, and could easily be automated for a more sophisticated sampling protocol for flour based on prescreening samples with NIRS followed up by use of the standard flotation method when necessary.  相似文献
