Several studies of intraspecific and interspecific DNA sequence variation from Drosophila loci have revealed a pattern of low intraspecific variation from genomic regions of low recombination. The mechanisms consistently invoked to explain these patterns are the selective sweep of advantageous mutations together with genetic hitchhiking of linked loci. To examine the effect of selective sweeps on genetic divergence during speciation, we studied two loci in different genomic regions thought to be subject to selective sweeps. We obtained DNA sequences from 1.1kb pair portions of the fourth chromosome locus cubitus interruptus Dominant (ciD) and from the asense locus near the telomere of the X chromosome. At ciD, we found very low variation among multiple lines of Drosophila mauritiana and D. sechellia. This finding is consistent with an earlier report of very low variation in D. melanogaster and D. simulans at ciD and supports the conclusion of selective sweeps and genetic hitchhiking on the nonrecombining fourth chromosome. The pattern of variation found at asense suggests that a selective sweep has occurred recently at the tip of the X chromosome in D. simulans, but not in D. melanogaster or D. mauritiana. The data from ciD and asense are compared with data from three X chromosome loci (period, zeste, and yolk protein 2) that experience normal levels of recombination. By examining estimated genealogies and the rates at which different classes of mutations have accumulated, we conclude that selective sweeps are common occurrences on the fourth chromosome but less common near the tip of the X chromosome. An interesting pattern of low variation at ciD among D. simulans, D. mauritiana, and D. sechellia suggests that a selective sweep may have occurred among these forms even after divergence into separate species had begun.  相似文献

The temperature sensitive period for mutant expression was determined incubitus interruptus ofDrosophila melanogaster. Two stocks carrying the dominant alleleci D were compared, in which under the influence of temperature, mutant expression of the most conspicuous character (gap in fourth vein) changes in opposite directions.The effective period (T.E.P.) ofci D was found to be very similar in both stocks. It is relatively short and begins after puparium formation. The T.E.P. is also the same for mutant expression on the fourth and the fifth vein. But in contrast to the 4th vein character, the 5th vein reacted in the same direction in both stocks.The T.E.P. was again determined in the same two stocks after the recessive alleleci had been introduced instead ofci D . The T.E.P. ofci in both stocks occupies a large part of the larval period including the first instar. The genetic background difference that causes opposite temperature reactions ofci D , does not show up in the temperature reactions ofci.The significance of these facts and of T.E.P.'s in general are discussed.  相似文献

Several alleles were found to determine the colour of the dorsal pronotum in Chorthippus brunneus; there was evidence for at least two loci (C and V). Brown (CB)was the universal recessive and green (CC) was dominant to all other colours. The white allele (CW)was codominant with green(CG)and purple (CP). Wing-patterns were determined by a separate, probably linked locus (W). A dominant plain wing-pattern (WP) was associated with colours other than brown. Striped(WS)and mottled(Wmo) were codominant and a plain recessive allele (WP) was also found. All three alleles were associated with the brown phenotype. A purple-sided allele (SPu) was sometimes obmd with Cpu.. SPu was dominant to brown sides (SB), A series of markings on the dorsal and lateral pronotum (linea intermedia, fascia postocularis, linea media, carina media and zona lateralis) were investigated and found to be controlled at separate loci which may be linked to W. These characters were expressed by dominant alleles. Epistatic effects by modifier loci were shown to have an important effect on the determination of wing phenotype. Allele Wo+, for example, suppressed the stripe-wing pattern, linea media, carina media and zona lateralis. It was concluded that colour patterns appear to be under genetic control and that dominant alleles were rare in the wild. Changes in shades of colours were shown to be age-dependent and minor.  相似文献

Somatolactin (SL) plays an essential role in body-color regulation in medaka and is encoded by the color interfere (ci) locus. The ci mutant fish possess constitutively increased numbers of leucophores and a concomitant decrease in visible xanthophores. However, the mechanism of action of SL on these cell types, and the role of SL in body-color regulation in other species, is unknown. In this study, we verified an SL–xanthophore relationship in ci mutant fish using the leucophore free (lf) gene. Histological observation of lf larvae indicated that these mutants do not possess differentiated leucophores. The cilf double mutant, whose genotype was confirmed using DNA markers, lacked leucophores; however, the number of xanthophores remained low, demonstrating that leucophores are not necessary for mediating SL signaling to xanthophores. This finding suggests a conserved function for SL in xanthophore regulation across species, rather than the evolution of a medaka-specific and leucophore-dependent role of SL in body-color regulation. Our results also demonstrate that the lf gene has an indispensable role in leucophore development epistatic to SL signaling. The lf gene has not been cloned. The high-resolution recombination map surrounding the lf locus constructed in this study, together with medaka whole genome sequences that will be released soon, will allow the rapid cloning of the lf gene by forward genetic approaches.  相似文献

This paper theoretically analyses the relationship between surplus energy, which is available for either somatic growth or reproduction, and body weight. From the data of metabolism and growth of the biwamasu, Oncorhynchus rhodurus, obtained by Miura et al., a Bernoulli's differential equation is induced to represent the relationship between body weight and the sum of surplus energy and active metabolic rate. Solving this equation gives the amount of surplus energy, f(Wx), as follows:f(Wx) = (αWx1−γ1−γ)1/(1−γ)−Wx, in which α, β and γ are constants and Wx is body weight at age x. The function is applied to ten fish populations and consequently it is found to be useful for a wider age range and a wider variety of fishes than the conventional function.  相似文献

Decreased neutrophils and megakaryocytes in anemic mice of genotype W/W   总被引:12,自引:0,他引:12  
The concentration of neutrophils and megakaryocytes was determined in the marrow of anemic mice of genotype W/Wv and their normal (+/+) litter mates. In all groups studied, the humerus of W/Wv mice contained significantly less neutrophils and megakaryocytes than did normal animals. Blood neutrophil concentration was less in all groups of W/Wv mice but in only one group which was the youngest group studied, did this value differ significantly from normal. The blood and marrow neutrophil response to endotoxin was similar in W/Wv and “+/+” animals. This suggests that the neutrophilic system of W/Wv mice responds to this stimulus in a relatively normal manner, much as their erythroid system responds to hypoxia, and androgens.  相似文献

The concept of critical nitrogen concentration(%N c) has been proposed as the minimum%N in shoots required to produce the maximum aerial biomassat a given time. Several authors have shown that%N c declines as a function of aerial biomassaccumulation (W) and the %N cW relationship has been proposed as a diagnostic tool of N statusin different crops, excluding maize. From data obtained in five nitrogenfertilisation experiments in irrigated maize crops, 26 critical data-pointswere selected with a precise statistical procedure. An allometric relationwas fitted and a critical %NW relationshipmodel is proposed in maize as: If W < 1 t ha-1%N c = 3.40 If 1 t ha-1W ≤ 22 t ha-1%N c = 3.40(W)−0.37 The model is applicable to maize crop development between emergenceand silking + 25 days. The model was tested and validated with dataobtained in a network of 17 N fertilisation experiments conducted inFrance under contrasting pedoclimatic conditions. In only nineout of 280 data-points (3.2%), the plant N status was mispredictedwhen ±5% error around %N c wasallowed. A critical N uptake model (Nuc, kg Nha-1) is proposed as Nuc = 34 (W)0.63 A comparison between Nuc and N uptake observedin N treatments giving the maximal grain yields has shown that maizecrops assimilate at least 30 kg N ha-1 in a storage N poolat the silking stage. The significance of the critical%NW and Nu−W relationships is discussed in relation to theoretical models proposed inwhole plant ecophysiology. Different relationships calculated betweenleaf area index and aerial biomass accumulation, and between N uptakeand leaf area were consistent with previous results for other crops.This strengthens the interest of the critical%NW relationship for use as diagnostictool of nitrogen status in maize crops. Abstract

The autoregulation of testicular LH receptors was studied in Wx/Wv mice with germ cell aplasia and in normal (±/±) mice. To assess the effects of each individual allele, Wx/± and Wv/± mice were also examined. Basal testicular LH receptor concentration was higher in Wx/Wv mice than in all other genotypes, and higher in Wx/± than in ±/± mice. Twenty-four h after injection of 0.3 IU hCG/g bw, LH receptor concentration was decreased in ±/± and Wv/± mice, but not in Wx/Wv or Wx/± animals. Administration of hCG caused a significant increase in plasma testosterone levels in all genotypes. However, injection of the highest dose of hCG used (0.9 IU/g bw) caused a significantly greater elevation in plasma testosterone in Wx/Wv than in ±/± mice. Plasma gonadotropin levels were significantly higher in Wx/Wv mice than in all other genotypes. The present results indicate that the Wx allele is responsible for the changes in testicular function observed in Wx/Wv mice, and suggest that this allele may be involved in the genetic regulation of testicular LH receptors in the mouse.  相似文献

Walking is the most common type of physical activity prescribed for the treatment of obesity. The net metabolic rate during level walking (W/kg) is ~10% greater in obese vs. normal weight adults. External mechanical work (Wext) is one of the primary determinants of the metabolic cost of walking, but the effects of obesity on Wext have not been clearly established. The purpose of this study was to compare Wext between obese and normal weight adults across a range of walking speeds. We hypothesized that Wext (J/step) would be greater in obese adults but Wext normalized to body mass would be similar in obese and normal weight adults. We collected right leg three-dimensional ground reaction forces (GRF) while twenty adults (10 obese, BMI=35.6 kg/m2 and 10 normal weight, BMI=22.1 kg/m2) walked on a level, dual-belt force measuring treadmill at six speeds (0.50–1.75 m/s). We used the individual limb method (ILM) to calculate external work done on the center of mass. Absolute Wext (J/step) was greater in obese vs. normal weight adults at each walking speed, but relative Wext (J/step/kg) was similar between the groups. Step frequencies were not different. These results suggest that Wext is not responsible for the greater metabolic cost of walking (W/kg) in moderately obese adults.  相似文献

How does a Drosophila wing grow to the appropriate size and shape? Although a collaboration of cell division with the patterning of cell fates seems obvious, almost nothing is known about how these two processes are coordinated during development. A recent paper1 finds that blocking cell division uncouples cell growth from the cell division cycle, displaying remarkable flexibility in the ability of the wing primordia to achieve the right proportions with fewer than normal cells. Cell death and its effect on wing size have been described in some wing mutants of Drosophila hydei. Dead cells in the imaginal discs were localized by Nile-bule and acridine-orange staining. Various Notch (N) alleles, the mutation Costal-nick (Cnk) and the compound N/Cnk show characteristic patterns of cell death in the imaginal wing disc. Some but not all of the structural features of the adult wing can be related to the site of cell death during larval stages. In NAx types, extensive cell death is followed by regenerative growth, invalidating a simple relation between size of the disk and size of the wing. In Nts/Cnk cell death and wing morphology depend on the breeding temperature. From temperature experiments we conclude that cell death starts between day 4 and 5 after egg laying and can be induced by a shift to the restrictive temperature during the critical phase. Patterns of wing incisions and cell death in Nts/Cnk genotypes seem not to be delimited by any of the known compartment boundaries.  相似文献

The proliferation and differentiation of hemopoietic cells from genetically anemic Wv/Wx,W/Wv, and Wv/Wv mice, and from nonanemic carrier W/+, Wb/+, and Wv/+ mice have been evaluated in vivo by transplantation techniques and in vitro by the agar gel culture method. Marrow from anemic and carrier mice contained progenitor cells which were decreased in number and formed small, often rudimentary, colonies in the spleens of irradiated recipient mice. Proliferation and differentiation of both erythropoietic and leukopoietic progenitor cells were delayed and reduced, but erythropoiesis was more severely affected than leukopoiesis. The severity of the hemopoietic impairment was gene-dose dependent. The W gene effect on leukopoietic progenitor cells was not secondary to anemia or to abnormal erythropoiesis. The marrow cells of anemic and carrier mice which form colonies of granulocytic and mononuclear cells in vitro were neither decreased in number nor impaired in proliferation and differentiation. Hypertransfusion of red blood cells increased the frequency of in vitro colony-forming cells, but not that of in vivo progenitor cells. The data demonstrate that colony-forming cells which proliferate in the agar gel cultures in vitro are distinct from the in vivo colony-forming cells and suggest that the former are primitive members of the granulocytic cell line. Perhaps in vitro CFU are in an intermediate stage of differentiation between in vivo CFU and myeloblasts, analogous to that which has been suggested for the erythropoietin-sensitive cell in the red cell series. W mutant alleles appear to act, therefore, at or very near the beginning of hemopoietic differentiation.  相似文献

Zusammenfassung In einem Windkanal wurde für eine Myotis lucifugus die Abhängigkeit der Fluggeschwindigkeit (vF) und der Geschwindigkeit über Grund (vG) von der Windgeschwindigkeit (vW) bestimmt. Die Fledermaus flog bei Windstille mit einer mittleren vF von etwa 4,5 m/sec. Bei zunehmenden Gegenwinden erhöhte sie vF und verringerte vG, um bei vW=7,7 m/sec für kurze Zeit stationären Flug (vG=0) zu erreichen. Die Flügelschlagfrequenz lag bei Gegenwinden von 0–7,7 m/sec zwischen 10–11/sec. Bei zunehmenden Rückenwinden wurde der Flug immer mehr dem Rüttelflug ähnlich und die Flügelschlagfrequenz stieg bis 16/sec an. Die vG blieb nahezu konstant in einem Bereich zwischen 4,5–5 m/sec. Bei Myotis lucifugus, Chilonycteris rubiginosa, Carollia perspillicata und Rhinolophus ferrum-equinum wurde die Flügelstellung während der Lautaussendung ermittelt. Alle Arten erzeugten entweder einen Einzellaut oder eine Gruppe von Lauten pro Flügelschlag.
Bats in the wind tunnel
Summary In an experimental wind tunnel air speed (vF) and ground speed (vG) of a Myotis lucifugus were measured as a function of wind speed (vW). The bat had a vF of about 4,5 m/sec in still air. With head winds it increased vF and lowered vG to reach stationary flight (vG=0) at a vW of 7,7 m/sec. The rate of wing motion remained at about 10–11/sec at head winds from 0–7,7 m/sec. With tail winds the bat changed to a semi-hovering flight with wing beat frequencies rising to about 16/sec and vF dropping to almost zero at 4–5 m/sec tailwinds. The vG remained nearly constant between about 4,5–5 m/sec. (Figs. 2 and 3). The wing positions during which orientation sounds were emitted were determined for Myotis lucifugus (Fig. 4), Chilonycteris rubiginosa, Carollia perspillicata and Rhinolophus ferrum-equinum (Fig. 5). All bats emitted either one single sound or a group of sounds per wing beat.

The kinetics of growth of bone marrow cells from normal or genetically anemic mice (Sl/Sld and W/Wv) were studied in irradiated normal and genetically anemic hosts. The parameters followed included total cellularity, the number of peroxidase positive cells, and the number of cells capable of forming colonies in vivo (CFU-S) or in culture (CFU-C). The results of these experiments demonstrate that W and Sl defects alter the growth of CFU-C and peroxidase-positive cells to a modest degree; that the defects are more obvious when studied in spleen rather than in bone marrow; and that there is no additivity of W and Sl defects. Nineteen irradiated recipients of marrow from W/Wv mice were studied after three to six months. Of these, 18 showed host-type erythrocytes, while in one mouse the erythrocytes had the size distribution of W/Wv cells. This finding indicated that occasionally genetically defective stem cells may repopulate irradiated hosts.  相似文献

In shake-flask culture, Methylobacterium extorquens accumulated poly(3-hydroxybutyrate) (PHB) possessing a substantially higher weight-average molecular mass (MW) than previously reported for this organism. The MW of PHB produced by M. extorquens was dependent on the initial concentration of methanol or sodium succinate, used as sole carbon sources. The highest MW values (0.6 and 1.7 × 106) were obtained with low initial concentrations of methanol or sodium succinate (4.0 and 3.0 g l–1, respectively) and the latter substrate always yielded PHB of higher MW than that produced from methanol. Thus PHB with an MW in the range 0.2–1.7 × 106 could be produced by selection of the carbon source and its concentration. In contrast to the findings with M. extorquens, the MW of PHB produced by Alcaligenes eutrophus was high (1.1–1.6 × 106) and generally unaffected by the choice or concentration of the carbon source. The use of glycerol as sole carbon source did, however, result in the accumulation of PHB with a markedly lower MW (5.5–8.5 × 105) than that produced from other sole carbon sources by this organism under similar conditions. We hypothesized that inhibition and activation of basolateral to luminal chloride transport mechanisms were associated with respective decreases and increases in basolateral to luminal water fluxes. The luminal to basolateral (J W L→B ) and basolateral to luminal (J W B→L ) water fluxes across ovine tracheal epithelia were measured simultaneously. The mean J W L→B (6.5 μl/min/cm2) was larger than J W B→L (6.1 μl/min/cm2). Furosemide reduced J W B→L from 6.0 to 5.6 μl/min/cm2. Diphenylamine-2-carboxylate (DPC) reduced J W B→L from 7.9 to 7.3 μl/min/cm2 and reduced the membrane potential difference by 38%. Furosemide together with DPC decreased J W L→B by 30% and J W B→L by 15%. Norepinephrine increased J W B→L from 4.9 to 6.0 μl/min/cm2. Neuropeptide Y in the presence of norepinephrine decreased J W L→B (6.4 to 5.2 μl/min/cm2) and returned J W B→L to its baseline value. Vasopressin increased J W B→L from 4.1 to 5.1 μl/min/cm2. Endothelin-1 induced a simultaneous increase in J W B→L (7.0 to 7.7 μl/min/cm2) and decrease in J W L→B (7.4 to 6.4 μl/min/cm2); and decreased the membrane resistance. These data indicate that in tracheal epithelia under homeostatic conditions J W B→L has a ∼15% actively coupled component. Consistent with our hypothesis, inhibition and receptor-induced stimulation of chloride effluxes were associated with decreases and increases in J W B→L , respectively. However, as inhibition of transcellular chloride transport always decreased J W L→B more than J W B→L , reducing transepithelial chloride transport did not result in less water being transported into the airway lumen. We recently reported the establishment of transgenic mouse lines carrying the mouse metallothionein/ret fusion gene in which severe melanosis and melanocytic tumors developed. In the present study, we demonstrate that a significant number of pigmented hairs developed in Wv/Wv mice crossed to one of the transgenic mouse lines. The pigmented hair of Wv/Wv mice carrying the ret oncogene did not lose color during aging and reappeared after shaving, indicating that the melanocytes in the hair follicle function. The melanocytic tumors also developed in these mice, although the incidence was lower than that in the wild transgenic mice. Furthermore, the neural tube culture of mouse embryos indicated that neural crest cells of the transgenic mice gave rise to a cell population that autonomously produced melanin even in the absence of melanocyte stimulating hormone. These results strongly suggested that the introduced ret oncogene could compensate for the defect of c-kit in Wv mice during both embryogenesis and postnatal life and induce a high level of melanin synthesis in the process of melanocyte development.  相似文献

In deletion-mapping of W-specific RAPD (W-RAPD) markers and putative female determinant gene (Fem), we used X-ray irradiation to break the translocation-carrying W chromosome (W Ze ). We succeeded in obtaining a fragment of the W Ze chromosome designated as Ze W, having 3 of 12 W-RAPD markers (W-Bonsai, W-Yukemuri-S, W-Yukemuri-L). Inheritance of the Ze W fragment by males indicates that it does not include the Fem gene. On the basis of these results, we determined the relative positions of W-Yukemuri-S and W-Yukemuri-L, and we narrowed down the region where Fem gene is located. In addition to the Ze W fragment, the Z chromosome was also broken into a large fragment (Z1) having the + sch (1-21.5) and a small fragment (Z2) having the + od (1-49.6). Moreover, a new chromosomal fragment (Ze WZ2) was generated by a fusion event between the Ze W and the Z2 fragments. We analyzed the genetic behavior of the Z1 fragment and the Ze WZ2 fragment during male (Z/Z1 Ze WZ2) and female (Z1 Ze WZ2/W) meiosis using phenotypic markers. It was observed that the Z1 fragment and the Z or the W chromosomes separate without fail. On the other hand, non-disjunction between the Ze WZ2 fragment and the Z chromosome and also between the Ze WZ2 fragment and the W chromosome occurred. Furthermore, the females (2A: Z/Ze WZ2/W) and males (2A: Z/Z1) resulting from non-disjunction between the Ze WZ2 fragment and the W chromosome had phenotypic defects: namely, females exhibited abnormal oogenesis and males were flapless due to abnormal indirect flight muscle structure. These results suggest that Z2 region of the Z chromosome contains dose-sensitive gene(s), which are involved in oogenesis and indirect flight muscle development.  相似文献
