Severing the dorsal vessel (DV) behind the corpus allatum (CA), or in the anterior part of the abdomen of Rhodnius prolixus, greatly reduces egg production, an effect which is abolished by the topical application of juvenile hormone l (JH l). Severing the DV in the posterior abdomen does not result in a marked reduction of egg production, although severing the alary muscles in segments V and VI has a similar effect to severing the DV in the anterior abdomen. Reduced egg production caused by severing the DV on day 8 postemergence does not occur if the nerves connecting the CA to the brain are severed on day 1 post emergence. However, egg production is reduced if the DV is severed on day 1 post emergence and the connections between the brain and the CA severed on day 8, suggesting that inhibition of the CA caused by severance of the DV requires innervation from the brain. An isolated CA implanted into an animal decapitated immediately after feeding escapes from the inhibition imposed by severance of the DV. Conversely, the CA in an insect, the head of which has been decapitated just anterior to the CA, remains inhibited. This result suggests that the head posterior to the brain must be present to maintain inhibition. It is concluded that DV severance acts on the brain via some humoral influence to impose inhibition on the CA, and that an endocrine center in the head is required in order to maintain the inhibition.  相似文献

At eclosion, the ovaries of female Corn earworm Heliothis zea do not contain mature eggs. Virgin-unfed females produced approximately 400 mature eggs in 8 days; mating or feeding doubled this number, and mating plus feeding more than tripled it. Females allatectomized or decapitated at day O matured few eggs. Egg production was restored by implantation of active corpora allata (CA) or by treatment with the juvenile hormone (JH) analogue methoprene at day 0. 20-Hydroxyecdysone, on the other hand, had no effect. Females in which the CA had been denervated or in which the median neurosecretory cells of the brain had been ablated at day O produced fewer eggs than sham-operated animals. These results indicate that egg maturation is controlled by JH and that continuous input from the brain is required for sustained CA activity for maintaining a high rates of egg maturation.The rate of JH biosynthesis by CA in vitro was determined with a radiochemical assay. The major hormones produced were JH-II and JH-III with small quantities of JH-I. The rates of JH synthesis were similar in all experimental groups which may indicate that the in vitro rate of JH synthesis does not reflect the actual state of CA activity in the female.  相似文献

We assessed the effects of age and mating status on in vitro juvenile hormone (JH) biosynthesis, oocyte growth, egg production and vitellogenin (Vg) accumulation in the tortricid moths, Choristoneura fumiferana and C. rosaceana. To determine whether vitellogenesis is dependent on the presence of JH, we also examined the effects of decapitation and JH analog treatments on egg production. In both species, the corpora allata (CA) of adult females released fmol quantities of JH, with JH II being the major homolog produced. The CA began producing detectable quantities of JH around the time of emergence. Full activation of the CA was observed a few hours sooner in C. fumiferana than in C. rosaceana. In pharate adults and young virgin females of both species, growth of the basal oocyte reflected changes in CA activity. Decapitation of newly emerged females significantly reduced egg production, but treatment of decapitated females with the JH analog methoprene resulted in egg production that was similar to (C. fumiferana) or greater than (C. rosaceana) that of controls, indicating that JH is required for oocyte maturation. Vg was first observed in the hemolymph before the presumptive time of CA activation, suggesting that the synthesis of this protein is not dependent on JH. The presence of normal quantities of Vg in the hemolymph of pupae decapitated before CA activation confirmed this hypothesis. The Vg titer underwent a transient decline following CA activation and was significantly lower in mated than in virgin females of both species 3 and 5 days after copulation. Since CA activation at emergence and mating are both expected to cause a rise in the JH titer, we suggest that the declines in the levels of Vg result from JH-enhanced Vg uptake by the developing oocytes. Mating induced a significant increase in egg production but had no measurable impact on rates of JH biosynthesis in vitro.  相似文献

The hemolymph juvenile hormone (JH) titer in sexually immature female adults of Locusta migratoria (Ibaraki strain, Japan) was lower than in sexually mature females; nevertheless, JH synthetic activity by the corpora allata (CA) in vitro was considerably higher in immature females than in sexually mature females ([Okuda et al., 1996]). We carried out experiments to explain this contradiction. The CA activity of sexually immature female adults was very low when the CA were incubated as a complex together with the corpora cardiaca (CC) and brain. When the same complex was assayed after cutting the nerve cord connecting the CC and CA (NCA1), JH synthesis by the CA was enhanced tenfold. When this pair of CA was incubated in fresh medium without the CC and brain, JH synthesis was further increased. Therefore, the higher in-vitro JH production by CA from immature female adults was the result of isolation of the CA from the brain and CC. A methanolic extract of brain-CC complexes contained a factor that inhibited JH synthetic activity by CA in vitro in both immature and mature insects, and this inhibition was reversible. The factor was heat-resistant but lost allatostatic activity after pronase digestion. These results indicate that the allatostatic factor is probably a heat-stable peptide.  相似文献

Abstract. Hormonal factors influencing reproductive development were examined in adult boll weevils, Anthonomus grandis (Boheman) (Coleoptera: Curculionidae). Long-day, high-temperature rearing conditions promote reproduction whereas short-day, low-temperature conditions do not. Implants of corpora allata (CA), brains, or brains plus retrocerebral complexes taken from long-day donors, or hormone analogue treatments were used to examine onset of vitellogenin synthesis and uptake in decapitated bodies of adult weevils reared in short-day, low-temperature conditions. Weevils decapitated within 2 days after eclosion and reared in short-day, low-temperature conditions never initiated vitellogenin production or ovarian development. Females and males decapitated on day 2 showed haemolymph vitellogenin within 5 days following treatment with Juvenile Hormone (JH) analogue or implantation of CA, but not after implantation of brain alone or implantation of muscle (sham). Uptake of vitellogenin into the oocytes did not occur unless both JH analogue and brain were given as replacement therapy. These experiments indicated that JH is necessary and sufficient to stimulate vitellogenin synthesis in this species but that a brain factor must be present for vitellogenin uptake.  相似文献

Allatotropic activity was found in the methanolic extract of the suboesophageal ganglia (SOG) and the corpora cardiaca (CC) of the Mythimna loreyi virgin males. No allatotropic activity was observed in the extract of brain or corpora allata (CA). Although CA can be activated by the SOG and CC extract, respectively, CC extract inhibited the response to the SOG extract. A significant in vitro allatotropic effect was exerted by the SOG and CC extract within 10 and 15 min, respectively, and this effect can be sustained for several hours even after transferring to fresh medium without extracts. The time course pattern of the CA activation ratio in both the SOG and CC extract-treated group is very similar to, but with significantly higher level than, that in the control group, suggesting the existence of an intrinsic pacemaker or an in vitro effect that controls the fluctuation of the CA biosynthetic activity. Synthetic Manduca sexta allatotropin had no significant effect on the M. loreyi CA. The results of treatment with the adenylate cyclase activator forskolin, the phosphodiesterase inhibitor IBMX, and the cAMP analogue dibutyryl-cAMP did not indicate that cAMP might be involved in the allatotropic control of CA. The characteristics of vitellogenin (Vg) and the relationship between Vg production and egg production in the tobacco budworm, Heliothis virescens, were studied. The relationship between Vg production and juvenile hormone (JH) and the impact of mating on Vg and egg production were also investigated. Vg appears in the hemolymph of H. virescens about 6 h after moth eclosion. Vg may be separated into two apoproteins (ApoVg-I and ApoVg-II) by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The molecular weights were calculated to be 156,065 ± 800 for ApoVg-I and 39,887 ± 323 for ApoVg-II. SDS-PAGE analysis revealed that the female hemolymph Vg polypeptides appear to be identical to those from eggs but are absent in male hemolymph. Vg concentration was significantly higher in mated females than in virgin females of the same age at 48 h after emergence. Rates of egg production increased as Vg production increased; rates of egg production in mated females were significantly higher than those of virgin females at 48, 72, 96, and 120 h postemergence. Vg production is dependent on JH, because hemolymph from decapitated females lacked Vg while that of decapitated females treated with synthetic JH had Vg at levels comparable to similarly aged, normal H. virescens females. Hemolymph JH titers in mated females were significantly higher compared with those in virgin females at all sampling periods. Allatotropic activity is found in methanolic extracts of the brain–suboesophageal ganglion (SOG)–corpora cardiaca (CC) complex from virgin males of Mythimna loreyi Duponchel (Lepidoptera, Noctuidae). Corpora allata (CA) from 6‐day‐old virgin males exhibit low rates of release of Juvenile Hormone (JH) acid (JHA) in vitro. Release of JHA can be activated by the addition of an extract of brain–SOG–CC complex in a dose‐dependent manner, and this allatotropic activation can be sustained consistently in the continuous presence of such extracts. Based on its trypsin sensitivity and heat stability, the allatotropic factor is most likely a peptide. The allatotropic activity is dependent on the concentration of calcium ions in the medium, with the highest activation achieved beyond 2 m m . The results of nerve transection experiments suggest that both nervi corporis allati I (NCA I) and NCA II are involved in mediating the allatotropic control of CA in vitro. Isolated CA alone show significantly higher rates of release of JHA than the intact brain–SOG–CC–CA complex during the first 3 h of incubation, but the release of JHA reaches almost the same range in both groups by the end of the fourth hour of incubation.  相似文献

Studies were undertaken to investigate vitellogenesis and its regulation in female adults of the fall armyworm, Spodoptera frugiperda. A single female-specific protein, likely to be the S. frugiperda vitellogenin (Vg), appeared approximately 5 h after adult eclosion in the hemolymph of virgin females. The concentration of the protein increased with age as sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) revealed. A protein with the same relative molecular mass was also present in egg extracts, but absent from hemolymph samples from male moths. The relative molecular mass of the designated S. frugiperda Vg was determined as 164.5+/-2.5 kDa. Vitellogenic oocytes became visible 36-48 h after emergence and egg deposition began on day 3 of adult life. Vg could not be detected in the hemolymph of females decapitated directly after eclosion. When decapitated virgin females were injected with the JH-mimic methoprene (MP), the level of Vg was comparable to that in non-decapitated moths, indicating that vitellogenesis in S. frugiperda depends on juvenile hormone (JH). However, the number of vitellogenic oocytes was somewhat lower than in non-decapitated virgin females. Injection of 20-hydroxyecdysone (20E) promoted Vg production to a similar extent in decapitated female moths, but in contrast to methoprene injection, treatment with 20E never resulted in the production of vitellogenic oocytes. In vitro cultivated ovaries of adult females dissected directly after eclosion produced lower amounts of ecdysteroids than those isolated on day 1 after emergence. Our results suggest a crucial role for 20E in the induction of vitellogenesis in the noctuid S. frugiperda, while JH seems to be essential for the continued uptake of Vg by developing oocytes and may trigger 20E biosynthesis in the ovary.  相似文献

A previous study by Davey [Can J Zool 4:243–249 (1987)] showed that egg production in the blood-feeding insect Rhodnius prolixus is greatly enhanced when the corpus allatum (CA) is denervated. This result supported findings of others that the brain of Rhodnius imposes an inhibition on the CA via its connections to the CA. The present study identifies the nervus corporis cardiacum II (NCCII) as the nerve responsible for this inhibitory influence. Transecting the NCCII before feeding causes a dramatic increase in the number of eggs made. Since the NCCII is a relatively small nerve that may contain only axons of lateral and posterior neurosecretory cells in the protocerebrum, this result suggests that one or both of these cell types produce the brain factor inhibiting egg production in this insect. The corpora allata (CA) of various insects express enzymes with fixation resistant NADPHdiaphorase activity. In female grasshoppers, juvenile hormone (JH) released from the CA is necessary to establish reproductive readiness, including sound production. Previous studies demonstrated that female sound production is also promoted by systemic inhibition of nitric oxide (NO) formation. In addition, allatotropin and allatostatin expressing central brain neurons were located in close vicinity of NO generating cells. It was therefore speculated that NO signaling may contribute to the control of juvenile hormone release from the CA.This study demonstrates the presence of NO/cGMP signaling in the CA of female Chorthippus biguttulus. CA parenchymal cells exhibit NADPHdiaphorase activity, express anti NOS immunoreactivity and accumulate citrulline, which is generated as a byproduct of NO generation. Varicose terminals from brain neurons in the dorsal pars intercerebralis and pars lateralis that accumulate cGMP upon stimulation with NO donors serve as intrinsic targets of NO in the CA. Both accumulation of citrulline and cyclic GMP were inhibited by the NOS inhibitor aminoguanidine, suggesting that NO in CA is produced by NOS. These results suggest that NO is a retrograde transmitter that provides feedback to projection neurons controlling JH production. Combined immunostainings and backfill experiments detected CA cells with processes extending into the CC and the protocerebrum that expressed immunoreactivity against the pan-neural marker anti-HRP. Allatostatin and allatotropin immunopositive brain neurons do not express NOS but subpopulations accumulate cGMP upon NO-formation. Direct innervation of CA by these peptidergic neurons was not observed.  相似文献

1. A study of the activity of cysteamine in relation to juvenile hormone (JH) production in adult females of Blattella germanica was carried out. 2. In vivo assays showed that cysteamine stimulates protein synthesis in the left colleterial gland and, in some instances, enhances oocyte growth. 3. In vitro assays demonstrated that cysteamine enhances JH release by incubated corpora allata (CA), and that this effect is more pronounced when using CA from 10-day-old females (period of ootheca transport), either connected to the corpora cardiaca (CC) or to the CC and to the brain. 4. In the present study the melatonin receptor 1A gene (MTNR1A) was proposed to be a candidate gene for egg production in Yangzhou geese. A total of 210 goose blood samples were collected to investigate the association of the MTNR1A gene with the number of eggs produced. Using a direct sequencing method, a single nucleotide polymorphism (SNP; g.177G>C) was detected in the 5? regulatory region of the MTNR1A gene (Genbank ss1985399687). Two alleles (G and C) and three genotypes were identified. Association analysis results showed that the g.177G>C SNP significantly affected the level of egg production within a 34‐week egg‐laying period (< 0.05). Furthermore, the geese with the GG genotype produced significantly more eggs compared to the geese with the CC genotype. Quantitative real‐time PCR analysis showed that the MTNR1A gene was highly expressed in small intestine, granulosa cell and ovary compared to other examined tissues. In addition, the mRNA expression level of MTNR1A in ovary indicated that significantly higher expression levels were recorded for geese with the GG genotype compared to those with the CC genotype. Moreover, a luciferase reporter assay showed that the CC genotype had significantly lower promoter activity than did GG. These results suggest that the identified SNP in the MTNR1A gene may influence the number of eggs produced and mRNA expression levels in Yangzhou geese and could be considered as a useful molecular marker in goose selection and improvement, especially for egg production.  相似文献

(Department of Entomology, North Dakota State University of South Carolina, School of Medicine, Columbia, U.S.A.) — Acta zool. (Stockh.) 61(2): 65–77. The corpora cardiaca (CC) and corpora allata (CA) of the pink bollworm, Pectinophora gossypiella, were studied at the ultrastructural level during induction maintenance, and termination of larval diapause. The CC is composed of 5–6 intrinsic secretory (IS) cells, glial cells, and axons that carry electron dense (ED) and electron lucent (EL) granules from the brain. The IS cells produce ED granules with an average diameter of 110 ± 26 nm. During diapause, the axons with ED granules showed large accumulations of neurosecretory granules, but the axons with the EL granules contained lesser amounts. The CA is made up of approximately 25 large cells and axons that pass through the CC from the brain. Most of the axons take up a peripheral position. A characteristic feature of CA cells during diapause was the presence of stacks of convoluted tubules of the smooth endoplasmic reticulum (SER) that may be associated with juvenile hormone synthesis. Previous studies demonstrate that virgin female adult Helicoverpa armigera (Lepidoptera: Noctuidae) moths exhibit calling behaviour and produce sex pheromone in scotophase from the day after emergence, and that mating turns off both of these pre-mating activities. In the fruit fly Drosophila melanogaster, a product of the male accessory glands, termed sex peptide (SP), has been identified as being responsible for suppressing female receptivity after transfer to the female genital tract during mating. Juvenile hormone (JH) production is activated in the D. melanogaster corpus allatum (CA) by SP in vitro. We herein demonstrate cross-reactivity of D. melanogaster SP in the H. armigera moth: JH production in photophase virgin female moth CA in vitro is directly activated in a dose-dependent manner by synthetic D. melanogaster SP, and concurrently inhibits pheromone biosynthesis activating neuropeptide (PBAN)-activated pheromone production by isolated pheromone glands of virgin females. Control peptides (locust adipokinetic hormone, AKH-I, and human corticotropin, ACTH) do not inhibit in vitro pheromone biosynthesis. Moreover, SP injected into virgin H. armigera females, decapitated 24 h after eclosion, or into scotophase virgin females, suppresses pheromone production. In the light of these results, we hypothesize the presumptive existence of a SP-like factor among the peptides transmitted to female H. armigera during copulation, inducing an increased level of JH production and depressing the levels of pheromone produced thereafter.  相似文献

An active corpus allatum (CA) in the blood feeding insect, Rhodnius prolixus, releases a substance that inhibits metamorphosis in larvae, and activates egg production in adults. In adults, transecting the nervus corporis cardiacum II's (NCCII), which are attached posteriorly to each protocerebral lobe, greatly increases egg production indicating that the adult CA is activated and receives neural inhibition from cells associated with the NCCII [Chiang, Arch. Insect. Biochem. Physiol. 39:126-131 (1998)]. In the present study, the NCCII's in fifth instar larvae were transected immediately before or after feeding to determine if these nerves normally inhibit CA activity in the last larval instar. Approximately 20 to 25 days following ingestion of a blood meal, L5's with transected NCCII's emerged as fully-formed adults with no larval characteristics. Examination of the brain in these recently emerged adults revealed that the NCCII's were absent. Since fifth instar larvae with transected NCCII's emerged with no juvenile characteristics, cutting the NCCII's did not activate the CA, indicating that the mechanism for inhibition of the CA differs in the last larval instar and adult animals.  相似文献

Ovariectomized Aedes aegypti do not synthesize vitellogenin after a blood meal, unless an ovary from a blood-fed donor is implanted. Decapitation, however, prior to implantation inhibits vitellogenin synthesis. A female ovariectomized and decapitated 6 hr after a blood meal, synthesizes vitellogenin if an ovary from a blood-fed donor is implanted. On the other hand, females that are fed on blood and immediately decapitated can not be stimulated to synthesize vitellogenin with implanted ovaries removed from blood-fed donors. These experiments led to the hypothesis that the blood meal stimulates the ovary to secrete a corpus cardiacum stimulating factor, that in turn promotes release of egg development neurosecretory hormone stored in the corpus cardiacum.Injection of 20-hydroxy-ecdysone or ovarian extract prepared from ovaries removed from unfed females does not release egg development neurosecretory hormone. Thus corpus cardiacum stimulating factor is not 20-hydroxy-ecdysone, and ovaries removed from unfed females do not store it.The rate of inactivation of egg development neurosecretory hormone released from the corpus cardiacum after a blood meal was investigated by implanting an ovary into females that were blood fed for various intervals than decapitated and ovariectomized. Seventy per cent of implants grow when the operation is done 18 hr after feeding, and 30% when the operation is done between 18 and 24 hr after feeding, indicating that egg development neurosecretory hormone is stable for the first 18 hr after a blood meal.Aedes taeniorhynchus females ovariectomized 24 hr after adult emergence do not synthesize vitellogenin. When such a female is implanted with an ovary removed from a sugar-fed or blood-fed Aedes aegypti donor vitellogenin synthesis is initiated, and the implant grows. Decapitation prior to implantation inhibit vitellogenin synthesis and implants do not grow. These results indicate that corpus cardiacum stimulating factor is not species specific.  相似文献

This paper deals with apical dominance using a dicotylar model obtained after decapitation of pea seedlings with two shoots — one dominant and the other inhibited. When the dominant shoot was decapitated the inhibited one is released from inhibition and after 24 to 72 h begins to grow. However, the levels of trans-zeatin and production of ethylene increase within 4 and 6 hours respectively after release from inhibition, and within an interval of 72 h the levels of both phytohormones begin gradually to decrease. This indicates that also in this model, the release from apical dominance is associated with an increase in the level of cytokinin zeatin and, thereafter, also with an increased production of ethylene. If indolyl-3-acetic acid (IAA) is applied on the decapitated main stem after decapitation of the dominant shoot, the growth of the initially inhibited one is very strongly retarded; if, however, IAA is applied on the decapitated dominant shoot, this inhibition is significantly weaker. This means that the inhibiting effect of IAA on the inhibited shoot originates to a greater degree from the main stem rather than from the dominant shoot. The effect of benzyladenine (BA) is transferred equally from the decapitated main stem and from the decapitated dominant shoot because the initially inhibited shoot begins to grow as well as also other shoots from serial cotyledonary buds.  相似文献

Mating and availability of substrate were found to have a dramatic effect on the level of egg production and juvenile hormone III (JH III) titer in Teleogryllus commodus. Mated females with egg laying opportunity produced twice as many eggs, and exhibited an 8-fold increase in JH titer, as compared with virgin females or mated females forced to retain most of their eggs due to the absence of substrate for egg deposition. Denervation of the ovaries, or nerve cord transection, also caused accumulation of eggs and lowered the JH level, suggesting a feedback mechanism in which the state of egg depletion of the ovaries determines the level of JH secretion by the corpora allata (CA). Results from ovariectomy of mated or virgin animals suggest that the feedback mechanism includes inhibitory humoral factors secreted by the ovary.  相似文献

Molting and metamorphosis are essential events for arthropod development, and juvenile hormone (JH) and its precursors play critical roles for these events. We examined the regulation of JH biosynthesis by the corpora allata (CA) in Bombyx mori, and found that intact brain-corpora cardiaca (CC)–CA complexes produced a smaller amount of JH than that in CC–CA complexes and CA alone throughout the 4th and 5th (last) instar stadium. The smaller amount of synthesis was due to allatostatin-C (AST-C) produced by the brain. The CC synthesized short neuropeptide F (sNPF) that also suppressed the JH synthesis, but only in day 3 4th stadium and after the last larval ecdysis. For the suppression, both peptides prevented the expression of some of the distinct JH biosynthetic enzymes in the mevalonate pathway. Allatotropin (AT) stimulated sNPF expression in the CC of day 1 5th instar stadium, not of day 3 4th; therefore the stage-specific inhibition of JH synthesis by sNPF was partly due to the stimulative action of AT on the sNPF expression besides the stage-specific expression of the sNPF receptors in the CA, the level of which was high in day 2 4th and day 0 5th instar larvae. The cessation of JH biosynthesis in the last instar larvae is a key event to initiate pupal metamorphosis, and both sNPF and AST-C are key factors in shutting down JH synthesis, along with the decline of ecdysone titer and dopamine.  相似文献
