Summary Fixation of prothoracic glands of Galleria mellonella with a solution containing saponin permits immunocytochemical staining of the entire gland. Steroid transport through the cell surface of the giant polyploid prothoracic gland cells of Galleria mellonella L. was studied by an ultracytochemical method. The alkaloid digitonin, known to form a complex with all sterols having a free-OH radical in position 3, proved to be suitable for studying the interiorisation of moulting hormone precursors and the release of synthesized hormones. The results suggest that cholesterol uptake in the last larval instar occurs by macropinocytosis during the feeding period, while the release of the steroids produced by the gland occurs by reverse micropinocytosis mostly on days 5-7 of the instar. The two processes are not simultaneous. The intracytoplasmic localisation of the reaction product confirms the steroidogenic role of the prothoracic gland.  相似文献

Galleria mellonella larvae reared in a light:dark (LD) 12:12 regime terminate feeding and pupate 1 day earlier than insects kept in constant darkness. Rearing conditions have no influence on the body weight attained at pupation. In both rearing conditions body weights attained by females were greater on the average by 50% than the body weights of males.In LD 12:12 all distinctive changes in the juvenile hormone titre and juvenile hormone esterase activity occurred 1 day earlier than in constant darkness. The peak value of juvenile hormone esterase activity was by 22% higher in insects reared under light conditions as compared with animals kept in constant darkness.Last-instar G. mellonella larvae reared in constant darkness were more sensitive to cooling and juvenile hormone analogue application. The chilling-induced elevation of juvenile hormone titre was also higher in constant darkness. The chilling-induced elevation of the brain allatotropic activity was not influenced by rearing conditions (brain activity measured as the number of extra-larval moults produced by hosts implanted with chilled brains). However, rearing in an LD 12:12 regime decreases the host sensitivity to implanted brains.  相似文献

Summary The formation and secretion of ecdysteroid by the prothoracic gland cells of Galleria mellonella (Insecta, Lepidoptera) were investigated electron microscopically and immunocytochemically. The moulting hormone ecdysone becomes first evident in vesicles and tubules of the smooth endoplasmic reticulum (SER). The SER forms secretory granules in which ecdysone was shown immunocytochemically. The Golgi apparatus seems not to be directly involved in ecdysone secretion. A 2 × 2 factorial experiment was performed with each of 18 age classes within the last instar of Galleria mellonella. The two experimental factors applied to each age class were chilling and space deprivation; combinations of each factor were tested at two levels (present or absent). The 3-parameter Weibull function was used to calculate the proportion of animals undergoing larval-pupal ecdysis. Significant differences among time-trend statistics were related to interaction between chilling and space deprivation, as well as to age class. In general, chilling without space deprivation produced a small delay relative to the unchilled, not space-deprived condition. With space deprivation, however, chilling accelerated pupation compared to space deprivation alone. The order in which the chilled, space-deprived and space-deprived groups pupated expresses the essence of the interaction. Contrary to an additive effect, delayed development resulting from space deprivation appeared to be mitigated by chilling. Salivary gland stiffening was observed in chilled, space-deprived animals. With space deprivation, stiffening was associated with faster development compared to that of unchilled animals. These observations are discussed in the context of extant hypotheses of endocrine mechanisms responsible for the separate effects of chilling and space deprivation.  相似文献

BACKGROUND: The common marmoset (Callithrix jacchus) is a New World primate that has been used increasingly in toxicological evaluations including testing for testicular toxicity of pharmaceutical and environmental chemicals. Information on structural and functional characteristics of the testis in common marmosets ("marmoset" in this review) is critical for designing experiments, interpreting data collected, and determining relevance to humans in risk assessment. METHODS: This study provides a comprehensive review on testicular development, structure, function, and regulation in common marmosets. RESULTS: There is little information regarding testicular formation and development during gestation. Based on the overall pattern of embryonic development in marmosets, it is postulated that gonadal formation and testicular differentiation most likely takes place during gestational Week 6-12. After birth, the neonatal period of the first 2-3 weeks and the pubertal period from Months 6-12 are critical for establishment of spermatogenesis in the adult. In the adult, a nine-stage model has been used to describe the organization of seminiferous epithelium and multiple stages per tubular cross-section have been observed. Seminiferous epithelium is organized in a wave or partial-wave manner. There are on average two stages per cross-section of seminiferous tubules in adult marmoset testis. Sertoli cells in the marmoset have a uniform morphology. Marmoset spermatogenesis has a high efficiency. The prime determinant of germ cell production is proliferation and survival of spermatogonia. Sertoli cell proliferation during the neonatal period is regulated by follicle-stimulating hormone (FSH), but chorionic gonadotropin (CG), instead of luteinizing hormone (LH), is the only gonadotropin with luteinizing function in marmoset. The receptor gene for CG in marmoset is unique in that it does not have exon 10. Marmosets have a "generalized steroid hormone resistance," i.e., relatively high levels of steroid hormones in circulation and relatively low response to exogenous steroids. Blockage of FSH, CG, and testosterone production during the first 3 months after birth does not cause permanent damage to the male reproductive system. Initiation of spermatogenesis in the marmoset requires unique factors that are probably not present in other mammals. Normal male marmosets respond to estradiol injection positively (increased LH or CG levels), a pattern seen in normal females or castrated males, but not usually in normal males of other mammalian species. CONCLUSIONS: It seems that the endocrine system including the testis in marmosets has some unique features that have not been observed in rodents, Old World primates, and humans, but detailed comparison in these features among these species will be presented in another review. Based on the data available, marmoset seems to be an interesting model for comparative studies. However, interpretation of experimental findings on the testicular effects in marmosets should be made with serious caution. Depending on potential mode of testicular actions of the chemical under investigation, marmoset may have very limited value in predicting potential testicular or steroid hormone-related endocrine effects of test chemicals in humans.  相似文献

The cytopathology of the Seriscethis iridescent virus (SIV) infection of Galleria hemocytes was studied by phase-contrast microscopy, [3H]thymidine autoradiography, and fluorescent antibody and acridine orange staining. Of the five hemocyte classes only the oenocytoid was not infected. Infected plasmatocytes and adipohemocytes undergo nuclear and cytoplasmic hypertrophy accompanied by nuclear DNA synthesis. The timing and identity of nuclear and cytoplasmic DNA synthesis are discussed.  相似文献

The complete amino acid sequence of apolipophorin-III (apoLp-III), a lipid-binding hemolymph protein from the greater wax moth,Galleria mellonella, was determined by protein sequencing. The mature protein consists of 163 amino acid residues forming a protein of 18,075.5 Da. Its sequence is similar to apoLp-III from other Lepidopteran species, but remarkably different from the apoLp-IIIs of insects from other orders. As shown by mass spectrometric analysis, the protein carries no modifications. Thus, all of its known physiological functions, including its recently discovered immune response-stimulating activity, must reside in the protein itself.  相似文献

Under in vitro conditions the prothoracic gland nerve of the last larval instar of Periplaneta americana shows the same efferent nervous activity as under in situ conditions–ie, low activity at the 9th day and high activity at the 20th day of the molting interval. Isolation of the prothoracic ganglion from the subesophageal ganglion provokes an increase in this nerve activity, suggesting an inhibitory effect of the subesophageal ganglion on prothoracic gland nerve activity in vivo. Only in 20-day-old larvae does electrical stimulation of isolated prothoracic glands in vitro via the gland nerve result in a slightly increased release of ecdysteroids from the gland. This effect could not be influenced by different lengths of stimulation periods. Denervation of the prothoracic gland by transection of the gland nerve on the 13th day of the molting interval results in a complete abolition of the first peak of ecdysteroid production in the gland but has no influence on the occurrence and the amount of the main ecdysteroid peak just before the molt. The results suggest the participation of nervous activity in special periods of prothoracic gland regulation in the cockroach.  相似文献

Summary Localization of intracellular calcium was demonstrated by precipitation with potassium hexahydroxoantimonate in the fixation medium containing osmium tetroxide or osmium tetroxide and glutaraldehyde. The presence of calcium in the precipitates was confirmed by X-ray microanalysis. Cells from active prothoracic glands contain more calcium deposits than inactive glands. The calcium precipitates are mainly localized in the nucleus, in the smooth endoplasmic reticulum, in the hyaloplasm and to a lesser degree in the mitochondria. These findings are consistent with the proposed role of calcium in the stimulation of steroidogenesis.  相似文献

为明确不同温度对大蜡螟(Galleria mellonella L.)生长发育和繁殖的影响,在24℃、28℃和31℃三个温度条件下,采用人工饲料饲养观察了大蜡螟的生长发育和繁殖状况。结果表明,温度对大蜡螟发育历期、生长发育速率以及繁殖具有显著影响(P0. 05)。在24~31℃范围内,随着温度升高,各虫态发育历期缩短,幼虫体长增长加快,发育速率加快。在24℃下世代历期最长(47. 96 d),31℃下世代历期最短(33. 68 d)。在同一温度下,雄蛾的寿命明显较雌蛾的寿命长,但成虫寿命、产卵前期和产卵期均随着温度升高而缩短。在24℃下产卵量最高为2 781. 50粒/雌,而在31℃下产卵量最低为1 943. 17粒/雌。  相似文献

Silk gland cells of silkworm larvae undergo multiple cycles of endomitosis for the synthesis of silk proteins during the spinning phase. In this paper, we analyzed the endomitotic DNA synthesis of silk gland cells during larval development, and found that it was a periodic fluctuation, increasing during the vigorous feeding phase and being gradually inhibited in the next molting phase. That means it might be activated by a self‐regulating process after molting. The expression levels of cyclin E, cdt1 and pcna were consistent with these developmental changes. Moreover, we further examined whether these changes in endomitotic DNA synthesis resulted from feeding or hormonal stimulation. The results showed that DNA synthesis could be inhibited by starvation and re‐activated by re‐feeding, and therefore appears to be dependent on nutrition. DNA synthesis was suppressed by in vivo treatment with 20‐hydroxyecdysone (20E). However, there was no effect on DNA synthesis by in vitro 20E treatment or by either in vivo or in vitro juvenile hormone treatment. The levels of Akt and 4E‐BP phosphorylation in the silk glands were also reduced by starvation and in vivo treatment with 20E. These results indicate that the activation of endomitotic DNA synthesis during the intermolt stages is related to feeding and DNA synthesis is inhibited indirectly by 20E.  相似文献

The effect of temperature on the infection of larvae of the greater wax moth, Galleria mellonella, by Heterorhabditis megidis H90 and Steinernema carpocapsae strain All, was determined. For both species, infection, reproduction, and development were fastest at 20 to 24 °C. Infection by both H. megidis and S. carpocapsae occurred between 8 and 16 °C; however, neither species reproduced at 8 °C. Among the nematodes used in experiments at 8 °C, no H. megidis and very few S. carpocapsae developed beyond the infective juvenile stage. Compared with H. megidis, S. carpocapsae invaded and killed G. mellonella larvae faster at 8 to 16 °C. By comparing invasion rates, differences in infectivity between the two nematode species were detected that could not be detected in conventional petri dish bioassays where mortality was measured after a specified period. Invasion of G. mellonella larvae by H. megidis was faster at 24 than at 16 °C.  相似文献
