In order to determine ortho-acetyl-L-carnitine, two biosensors were proposed. The biosensors were designed using physical immobilization of L-amino acid oxidase (L-AAOD) and horseradish peroxidase (HRP). Electrode characteristics were obtained and compared for the two carbon paste (graphite powder and paraffin oil) biosensors. The linear concentration ranges for the proposed biosensors were in the ranges of fmol/L to nmol/L, magnitude order with low limits of detection. Due to their reliability, the biosensors were used as detectors in a sequential injection analysis system, and gave reliable results for on-line assay of ortho-acetyl-L-carnitine in synthesis process control with a frequency of 75 samples per hour.  相似文献

Abstract

Carbon paste based biosensors for the determination of creatine and creatinine have been integrated into a sequential injection system. Applying the multi‐enzyme sequence of creatininase (CA), and/or creatinase (CI) and sarcosine oxidase (SO), hydrogen peroxide has been detected amperometrically. The linear concentration ranges are of pmol/L to nmol/L magnitude, with very low limits of detection. The proposed SIA system can be utilized reliably for the on‐line simultaneous detection of creatine and creatinine in pharmaceutical products, as well as in serum samples, with a rate of 34 samples per hour and RSD values better than 0.16% (n=10).  相似文献

Glycolysis is the primary metabolic pathway in all living organisms. Maintaining the balance of glycolysis flux and biosynthetic pathways is the crucial matter involved in the microbial cell factory. Few regulation systems can address the issue of metabolic flux imbalance in glycolysis. Here, we designed and constructed a bifunctional glycolysis flux biosensor that can dynamically regulate glycolysis flux for overproduction of desired biochemicals. A series of positive-and negative-response biosensors were created and modified for varied thresholds and dynamic ranges. These engineered glycolysis flux biosensors were verified to be able to characterize in vivo fructose-1,6-diphosphate concentration. Subsequently, the biosensors were applied for fine-tuning glycolysis flux to effectively balance the biosynthesis of two chemicals: mevalonate and N-acetylglucosamine. A glycolysis flux-dynamically controlled Escherichia coli strain achieved a 111.3 g/L mevalonate titer in a 1L fermenter.  相似文献

A sequential injection analysis (SIA) is proposed for the simultaneous determination of L- and D-methotrexate (Mtx) using amperometric biosensors as detectors. A SIA system is proposed due to the highest precision and accuracy and lower consumption of sample and buffer. The amperometric biosensors used as detectors in SIA system were based on L-amino acid oxidase (L-AAOD) or/and L-glutamate oxidase (L-Glox) and horseradish peroxidase (HRP) for the assay of L-Mtx and D-amino acid oxidase (D-AAOD) and HRP for the assay of D-Mtx were selected. The linear concentration ranges are of pmol/l to nmol/l magnitude order, with very low limits of detection. The SIA/biosensors system can be used reliably on-line in synthesis process control, for the simultaneous assay of L- and D-Mtx with a frequency of 34 samples per hour.  相似文献

MethodsThis study enrolled 207 patients with chronic cholestasis who were ordered to test for ATP8B1 and/or ABCB11 from January 2012 to December 2015. Additional 17 patients with ATPB81 or ABCB11 deficiency diagnosed between January 2004 and December 2011 were also enrolled in this study. 600 population-matched children served as controls. Clinical data were obtained by retrospectively reviewing medical records.ResultsA total of 26 patients were diagnosed with ATP8B1 deficiency and 30 patients were diagnosed with ABCB11 deficiency. GGT levels were similar between the two disorders at any observed month of age, but varied with age. The peak GGT value was <70U/L in the 2nd~6th month of life, <60U/L in the 7th~12th month and <50U/L beyond one year. GGT levels in patients with a genetic diagnosis were different from that in patients without a genetic diagnosis and controls. Larger ranges for GGT were found in patients without a genetic diagnosis. Some controls had GGT≥70U/L in the 2nd~6th month. Of the 207 patients, 39 (18.8%) obtained a genetic diagnosis. 111 patients met the ranges described above, including all the 39 patients with ATP8B1 or ABCB11 deficiency. The sensitivity was 100.0%. The rate of a positive molecular diagnosis increased to 35.1% (39/111 vs. 39/207, X2 = 10.363, P = 0.001). The remaining 96 patients exceeded the ranges described above and failed to receive a genetic diagnosis. These patients accounted for 43.8% of sequencing cost.ConclusionsGGT levels in patients with ATP8B1 or ABCB11 deficiency varied with age. The peak GGT value was <70U/L in the 2nd~6th month of life, <60U/L in the 7th~12th month and <50U/L beyond one year.  相似文献

BackgroundHuman biomonitoring studies of trace elements in biological fluids are mostly limited to a certain number of elements or biological materials. In this study, we describe the significant extension of a biomonitoring to 73 elements being present in concentration ranges from ng/L to g/L in clinically relevant specimens such as blood, serum, erythrocytes and urine.MethodsThe samples were collected from 102 occupationally non-exposed inhabitants of northern Germany. The elements were determined either by inductively coupled plasma tandem mass spectrometry (ICP-MS/MS) in the low concentration range or by inductively coupled plasma optical emission spectrometry (ICP-OES) for essential trace elements and electrolytes.ResultsMean values and selected percentiles of element concentrations are presented for all sample materials. From the results, we calculated the distribution of elements between plasma and blood cells. Application of ICP-MS/MS improves selectivity and accuracy in the determination of elements that are strongly spectrally interfered, such as Cr, Ge, Pd or Ti in blood samples.ConclusionsThis publication provides very valuable information for occupational or environmental hygienists, toxicologists and clinical chemists due to the particularly high number of determined elements and presented concentration ranges.  相似文献

Abstract

At Trounson Kauri Park, we monitored the movements of 21 feral cats (Felis catus), 11 stoats (Mustela erminea) and one male ferret (Mustelafuro). In feral cats, the average minimum home range was 446 ha (±82 SE) for 14 males, significantly larger than the average minimum of 117 ha (±40 SE) for seven females. In stoats the average minimum home range was 107 ha (±20 SE), for nine males compared with 81 ha (±31 SE) for two females. The single male ferret had a minimum home range of 197 ha. Adult male feral cats lived on apparently separate, non‐overlapping home ranges; females occupied exclusive home ranges which were overlapped by adult males; home ranges of sub‐adult male feral cats overlapped those of other sub‐adult male, adult male and female feral cats. The home ranges of two neighbouring male stoats overlapped, although their core ranges did not; both these and the ferret's home ranges overlapped those of the neighbouring feral cats. The feral cats were often located in cover in pastureland or near the edge of larger tracts of forest within their home ranges; stoats near waterways in the larger tracts of forest; and the ferret near the forest/pasture margins of Trounson Kauri Park. Our results suggest that control devices targeting all these species should be set at a minimum spacing of 800 m in order to put the majority of the resident and immigrant predators at risk.  相似文献

BackgroundDYMIND DH76 (DYMIND BIOTECH, China) is a new automated hematology system designed to provide CBC count, including a 5-part WBC differential count, and its analytical performance should be assessed before adoption for clinical use.MethodsThe analyzer was evaluated according to the International Council for Standardization in Haematology guideline. The purposes of this study were to assess its analytical performance in comparison to SYSMEX XN 1000 hematology analyzer currently used in our laboratory, as well as to compare the automated and manual WBC differential.ResultsWithin-run precision in all concentration ranges was very good with coefficients of variation (CVs) between 0.02% and 2.5% except for platelets over 500×109/L (CV 9.5%). Within-batch imprecision showed CVs lower the declared deviation ranges. Accuracy (defined as trueness) was excellent for all CBC and white cell differential parameters, compared with the state of the art%. Linearity was confirmed with excellent regression coefficients (0.999-1.000), even in the lowest values, and carryover was ≤ 1%. Comparison between DYMIND DH76 and SYSMEX XN 1000 was also very good with correlation coefficients (R2) for WBC (1.000), RBC (0.999), hemoglobin (0.999) and PLT over 50×109/L (0.994) and R2 was lower but still acceptable (0.910) for PLT<50×109/L. R2 for neutrophils, lymphocytes, eosinophils, basophils, and monocytes were 0.974, 0.982, 0.957, 0.625, and 0.836, respectively, in the comparison between the manual and DYMIND DH76 automated differential WBC counts.ConclusionsWith excellent analytical performance and acceptable comparative analysis, DYMIND DH76 hematology analyser covered the predefined international standards and requirements and is fully appropriate for clinical application.  相似文献

Capsule Since pesticide controls, it has expanded from a pre-pesticide baseline of 41 pairs to over 100 pairs.

Aims To describe the population's pattern of recovery and identify constraints on its annual productivity.

Methods Occupancy and productivity data were collected from all known and new ranges in Cumbria, UK. Statistical methods were employed to describe temporal and spatial patterns. Possible effects of weather were investigated using meteorological data.

Results The population recovered from eight pairs, with birds preferentially recolonizing 41 traditional ranges. The highest occupancy levels were 100 ranges in 1997 and 1998. Many new ranges were in peripheral and atypical locations, although some resulted from the splitting of traditional ones. The central core now has the highest recorded density (> 9 pairs per 100 km2). As occupied ranges increased, the mean nearest neighbour distance declined to an asymptote (approximately 3.5 km). Simultaneously, maximum values were observed for the percentages of known and occupied ranges in which eggs were laid. The mean number fledged by occupied ranges peaked in the early 1990s. New ranges seem to be more susceptible to the effects of May rainfall but overall there was little evidence that new and traditional ranges differ in productivity. Persecution has continued, with over 10% of examined ranges experiencing intentional disturbance in the worst years.

Conclusion The recovery of the Cumbrian population seems to be related to two processes: (1) greater productivity and survival forcing birds to occupy more peripheral and atypical sites; (2) a very high density in the central core possibly due to increased food supply resulting from the channelling of prey through its valley systems.  相似文献

Abstract

Non-covalent complexes of urease/polyethylene glycol (PEG)-aldehyde were synthesized using regular molar ratios of urease and PEG-aldehyde at room temperature. The physical properties of the non-covalent complexes were analyzed in order to investigate the impact of coupling ratio, temperature, pH, storage stability, and thermal stability. Urease activity was analyzed by UV–Vis spectrophotometer at 630?nm. The results showed that the strongest thermal resistance was obtained using nU/nPEG:1/1 (mg/mL) complex within all molar ratios tested. The enzymatic activity of nU/nPEG:1/1 complex doubled the activity of the free enzyme. Therefore, this complex was chosen to be used in the analyses. When coupled with PEG-aldehyde, urease exhibited improved activity between pH 4.0–9.0 and the optimum pH was found to be 7.0. The thermal inactivation results of the complex demonstrated that higher activity remained (40%) when compared with the free enzyme (10%) at 60?°C. The storage stability of the non-covalent complex was 4 weeks which was greater than the storage stability of the free enzyme. A kinetic model was suggested in order to reveal the mechanism of enzymatic conversion. Potentiometric urea biosensor was prepared using two different membranes: carboxylated poly vinyl chloride (PVC) and palmitic acid containing PVC. The potentiometric responses of both sensors were tested against pH and temperature and the best results were obtained at pH 7.0 and 20–30?°C. Also, selectivity of the suggested biosensors toward Na+, Li+ Ca2+, and K+ ions was evaluated and the reproducibility responses of the urea biosensors were measured with acceptable results.  相似文献

目的 多层生物介质的生物传感器被广泛应用于各大领域,其检测特性对于传感器优劣的评估尤为重要。本文目的在于量化表征多层生物介质的电学特征。方法 基于生物电阻抗谱技术来探究多层生物介质的电化学阻抗谱特性,并结合保角映射的方法来量化表征多层生物介质,阐明其对阻抗的影响规律,继而为生物传感器的研制与开发提供理论基础。有效提取各生物介质层修饰后电阻抗参数(Z*),从而量化表征多层生物介质层的电阻抗谱特性。结果 对多层模型进行了理论计算并构建了相关试验测试系统,研究结果表明,随着生物介质层的逐步修饰,检测区域电阻抗参数(Z*)在f=0.1~50 MHz下持续上升,理论计算结果趋势与试验结果趋势较好吻合,论证了此理论计算方法的正确性。结论 本文证实了可根据生物电阻抗谱和保角映射方法量化表征多层生物介质的电阻抗谱特性,对生物传感器的研制与开发有一定的实用价值。  相似文献

BackgroundLow serum Vitamin D is considered to be associated with tuberculosis while the “dangerous” level was not clear. The aim of this study was to identify the association between tuberculosis and serum Vitamin D levels via synthesis of available evidence.MethodsA search of EMBASE, Medline, ISI Web of knowledge, and Pubmed was conducted. The number of subjects of tuberculosis and no-tuberculosis groups in four Vitamin D range. Meta-analyses were performed and presented by odds ratios (ORs) and corresponding 95% confidence intervals (CIs).ResultsA total of 15 studies involving 1440 cases and 2558 controls were included. A significantly increased risk of tuberculosis was found in two ranges: ≤ 12.5 nmol/L: pooled OR = 4.556, 95% CI = 2.200-9.435; 13-25 nmol/L: pooled OR = 3.797, 95% CI = 1.935-7.405. No statistically significant risk of tuberculosis was found in the range of 26–50 nmol/L (pooled OR = 1.561, 95% CI =0.997-2.442). In range 51–75 nmol/L, no positive association was found (pooled OR =1.160, 95% CI = 0.708-1.900).ConclusionsThis study found that a serum Vitamin D level ≤ 25 nmol/L was significantly associated with an increased risk of tuberculosis while the range of 51–75 nmol/L was not. The range 26-50nmol/L posed potential high tuberculosis risk. Future large-scale, well-designed studies are needed to verify these results.  相似文献

目的:探讨血清IgE水平与冠心病的相关性。方法:将我院收治的135例患者根据冠脉造影结果分为冠心病组(CHD)和非冠心病组(non-CHD),冠心病组根据临床症状分为稳定型心绞痛组(SAP)、不稳定型心绞痛组(UAP)和急性心肌梗死组(AMI),动脉堵塞程度用Gensini评分量化,采用SIMENS BNII全自动免疫散射比浊仪测定总IgE水平。比较CHD和non-CHD组患者及SAP、UAP和AMI组患者血清IgE水平的差异,并进一步分析血清IgE等级与冠心病Gensini积分的相关性。结果:non-CHD患者血清IgE水平(32.3(13.5,61)KU/L)明显低于CHD患者(69(26.4,169)KU/L)(P=0.001),UAP和AMI患者血清IgE水平(78.6(37.0,191.0)KU/L、118.5(75.3,148.1)KU/L)均显著高于SAP组(36.7(20.7,96.7)KU/L)(P=0.034和P=0.001),且多支血病变组患者血清IgE水平(67.2(30.9,249.0)KU/L)显著高于单支血管病变患者(34.6(18.1,59.0)KU/L)(P=0.039)。IgE水平根据四分位间距分为四个等级,随着IgE分级水平增加冠心病Gensini积分增加。结论:冠心病患者血清IgE水平升高,且与冠心病类型和血管堵塞程度都存在显著相关性,可能辅助冠心病的诊断及病情监测。  相似文献

In this study, recombinant bacterial biosensors were immobilized in an agarose matrix and used for the simple and disposable field monitoring of phenolic compounds. In brief, Escherichia coli cells harboring the pLZCapR plasmid, which was previously designed to express the β-galactosidase reporter gene in the presence of phenolic compounds, were immobilized in agarose gel with or without a substrate [chlorophenol red β-galactopyranoside (CPRG)] and dispensed to the wells of a 96-well plate. Analytes were added to the wells, and color development was monitored either directly from wells containing intact cells co-immobilized with CPRG (SYS I), or using cells that were lysed prior to the addition of CPRG (SYS L). SYS L showed relatively higher intensities and faster color development than SYS I. However, both systems developed a red color (representing hydrolysis of CPRG) in the presence of 10 μM to 10~100 mM phenol, with maximum responses seen at 1~5 and 50 mM phenol for SYS I and SYS L, respectively. Other phenolic compounds (2-chlorophenol, 2-methylphenol, 3-methylphenol, 4-chlorophenol, 2-nitrophenol, resorcinol, catechol, and 2,5-dimethylphenol) were also detected by the systems, with varied detection ranges and responses. The agarose-immobilized biosensors were stable for 28 days, retaining 39~69% of their activities when stored at 4°C without nutrients or additives. The immobilized biosensors described herein do not require the on-site addition of a substrate (in the case of SYS I), the pretreatment of samples, or the use of unwieldy instruments for the on-site monitoring of phenolic compounds from environmental samples.  相似文献

Abstract

Early detection of tuberculosis (TB) reduces the interval between infection and the beginning of treatment. However, commercially available tests cannot discriminate between BCG-vaccinated healthy persons and patients. Also, they are not suitable to be used for immunocompromised persons. In recent years, biosensors have attracted great attention due to their simple utility, accessibility, and real-time outputs. These sensors are increasingly being considered as pioneering tools for point-of-care diagnostics in communities with a high burden of TB and limited accessibility to reference laboratories. Among other types of biosensors, the electrochemical sensors have the advantages of low-cost operation, fast processing, simultaneous multi-analyte analyzing, operating with turbid samples, comparable sensitivity and readily available miniaturization. Electrochemical biosensors are sub-divided into several categories including: amperometric, impedimetric, potentiometric, and conductometric biosensors. The biorecognition element in electrochemical biosensors is usually based on antibodies (immunosensors), DNAs or PNAs (genosensors), and aptamers (aptasensors). In either case, whether an interaction of the antigen–antibody/aptamer or the hybridization of probe with target mycobacterial DNA is detected, a change in the electrical current occurs that is recorded and displayed as a plot. Therefore, impedimetric-based methods evaluate resistance to electron transfer toward an electrode by a Nyquist plot and amperometric/voltammetric-based methods weigh the electrical current by means of cyclic voltammetry, square wave voltammetry, and differential pulse voltammetry. Electrochemical biosensors provide a promising scope for the new era of diagnostics. As a consequence, they can improve detection of Mycobacterium tuberculosis traces even in attomolar scales.  相似文献

ObjectiveTo establish pediatric reference ranges for plasma fractionated free metanephrines by enzyme immunoassay (EIA) and to evaluate its performance in the diagnosis of catecholamine-secreting tumors in the pediatric population.MethodsNormotensive children and children with suspected catecholamine-secreting tumors underwent measurement of plasma fractionated metanephrines by EIA to establish pediatric reference ranges. Children with suspected pheochromocytoma or paraganglioma also underwent magnetic resonance imaging or computed tomography from the neck to the pelvis and were followed up for a minimum of 1 year. Diagnosis of pheochromocytoma/ paraganglioma was confirmed by histologic examination. Pheochromocytoma/paraganglioma was excluded in children who had a histologic diagnosis other than pheochromocytoma/paraganglioma and in those who had no imaging evidence of tumor and no progression on follow-up.ResultsPlasma fractionated metanephrines were measured in 78 normotensive children (age range, 1.5-17 years) and in 38 children with suspected catecholamine-secreting tumors. Of the 38 children (age range, 6-17 years) with suspected pheochromocytoma/paraganglioma, 17 had a histopathologically proven catecholamine-secreting tumor. The newly derived pediatric upper reference limit for metanephrine (128 pg/mL) was higher than in adults (90 pg/mL), whereas the pediatric upper reference limit for normetanephrine (149 pg/mL) was lower than in adults (180 pg/mL). The manufacturer’s reference range for plasma fractionated metanephrines yielded a sensitivity of 100% and a specificity of 85.7%. Use of newly established pediatric reference ranges increased the specificity to 95.2% without altering the sensitivity (100%).ConclusionsPlasma fractionated metanephrines by EIA provide an accurate test with good sensitivity and specificity for the diagnosis of pheochromocytoma and paraganglioma in children. Capsule Singing territories were well separated.

Aims To examine the spatial distribution of Sardinian Warbler males during the breeding period in Mediterranean shrubland and, specifically, their territories, home ranges and spatial overlaps.

Methods We studied a 12-ha plot of Mediterranean shrubland in the 1997 to 1999 breeding seasons. Sardinian Warblers were captured using mist-nets, colour-ringed and their territories mapped. In 1999, seven breeding males were radiotracked in order to map home ranges.

Results The Sardinian Warbler had an average of 7.3 breeding pairs/10 ha in the study plot. The mean territory size was 8779 m2 and the mean home range was 22 321 m2. A positive relationship was found between the area of the home range and singing territory. Home ranges of males born in 1998 were about half the size of those of the oldest males. Paired males who moved with a female had smaller home ranges than those that were either unpaired or whose mate was incubating. The degree of home range overlap was high with some overlap between neighbouring territories. The estimate of home range area increased by 10% when the information generated by a mapping method was added and the estimated territory area increased by 31% when data generated by radiotracking were added. Transmitters remained attached to birds for an average of 9.63 ± 3.46 days (mean ± se).

Conclusions Singing territories were segregated to a considerable degree. In contrast, the wide overlaps among home ranges was best explained by the presence of food resources that the males exploit at the same time and also by the search for extra-pair copulation in nearby territories. We consider radiotracking in this species to be feasible and valid, with no evidence of negative effects on activity levels, weight or mortality.  相似文献
