The mycocinogenous strain Tilletiopsis flava VKM Y-2823 was found to possess fungicidal activity at pH 3.5–4.5, which was retained after curing the strain by eliminating the extrachromosomal genetic elements. The mycocin produced by the strain had a molecular mass of more than 10 kDa and was readily inactivated by heating and treatment with protease K. This mycocin was found to be active against species of the anamorphic genus Tilletiopsis. The overwhelming majority of other representatives of the order Tilletiales, as well as ascomycetous and basidiomycetous (including ballistosporous) yeasts of the orders Sporidiales and Tremellales, were resistant to it.  相似文献

A strain of Kluyveromyces lactis was found to secrete a fungicidal mycocin active in the pH range from 6 to 9 and exhibiting the highest activity at pH of approximately 7. A few yeast species of the families Saccharomycetaceae and Wickerhamomycetaceae were sensitive to the mycocin. Some genera and species were heterogeneous in this respect. UV treatment of the mycocinogenic strain resulted in loss of its antifungal activity. Although prokaryotes were not sensitive to the mycocin, the strain under study inhibited growth of some bacteria.  相似文献

The yeast Williopsis mrakii produces a mycocin or yeast killer toxin designated HMK; this toxin exhibits high thermal stability, high pH stability, and a broad spectrum of activity against other yeasts. We describe construction of a synthetic gene for mycocin HMK and heterologous expression of this toxin in Aspergillus niger. Mycocin HMK was fused to a glucoamylase protein carrier, which resulted in secretion of biologically active mycocin into the culture media. A partial purification protocol was developed, and a comparison with native W. mrakii mycocin showed that the heterologously expressed mycocin had similar physiological properties and an almost identical spectrum of biological activity against a number of yeasts isolated from silage and yoghurt. Two food and feed production systems prone to yeast spoilage were used as models to assess the ability of mycocin HMK to act as a biocontrol agent. The onset of aerobic spoilage in mature maize silage was delayed by application of A. niger mycocin HMK on opening because the toxin inhibited growth of the indigenous spoilage yeasts. This helped maintain both higher lactic acid levels and a lower pH. In yoghurt spiked with dairy spoilage yeasts, A. niger mycocin HMK was active at all of the storage temperatures tested at which yeast growth occurred, and there was no resurgence of resistant yeasts. The higher the yeast growth rate, the more effective the killing action of the mycocin. Thus, mycocin HMK has potential applications in controlling both silage spoilage and yoghurt spoilage caused by yeasts.  相似文献

Wickerhamomyces bovis type strain was found to secrete a mycocin with a fungistatic effect at pH from 3.5 to 6.0. The peak of its activity occurred at pH 5.0 in the presence of 3% NaCl. Yeast species sensitive to this mycocin were located within the family Wickerhamomycetaceae and belonged to phylogenetically related genera Ambrosiozyma, Nakazawaea, Ogataea, and Peterozyma.  相似文献

The viral particles (about 30 nm in diameter) that contain dsRNAs (2.0 and 6.3 kbp) encapsidated by a coat of protein were detected in a mycocin-secreting strain of Cystofilobasidium infirmominiatum isolated from plants in an oak forest (Moscow region). The mycocin with a molecular mass above 15 kDa is fungicidal (maximum activity at pH 4.5) and active mainly against some species of the Cystofilobasidiales and Filobasidiales ('Cryptococcus aerius' clade). Curing by incubation at elevated temperature resulted in the concomitant loss of dsRNAs and mycocinogenic activity, and cured derivatives became sensitive to the mycocin produced by the parent strain.  相似文献

作者多年来从我国禾本科等植物叶表分离了大量掷孢酵母,本文重点报道70株掷孢酵母与类酵母的初步分类,共分为三属,即掷孢酵母属(Sporobolomyces),布勒掷孢酵母属(Bul-lera)与腥掷孢菌属(Tilletiopsis);其中掷孢酵母属占75%。至于53株掷孢酵母,共分为三种,即玫红掷孢酵母(Sporobolomyces roseus)、赭色掷孢酵母(Sporobolomyces salmonicolor)、Sporobolomyces shibatanus);经统计,后者最为常见,占此属的84%。这些种在我国尚未见报道,作为新记录。  相似文献

Isolates of five species of the yeast-like fungus Tilletiopsis Derx (Tilletiopsis albescens Gokhale, Tilletiopsis fulvescens Gokhale, Tilletiopsis minor Nyland, Tilletiopsis pallescens Gokhale, and Tilletiopsis washingtonensis Nyland) were screened for exo- and endo--beta-1,3-glucanase and chitinase production in a liquid broth used to produce inoculum for biological control studies. There were significant differences among the species, and highest overall enzyme activity was present in T albescens and T. pallescens and lowest in T. washingtonensis. A time-course study of beta-1,3-glucanase and chitinase production in T pallescens ATCC 96155 in broth culture with 2.5% glucose as the carbon source showed that enzyme activity gradually increased over a 3- to 21-day period. Maximum enzyme activity was found between pH 4.0 and 5.0. SDS-PAGE of beta-1,3-glucanase isozymes revealed a range of molecular masses from 18 to 29 kDa. Five isozymes were present in both T albescens and T. pallescens and two in T washingtonensis. Antifungal compounds were also detected in ethyl acetate extracts of culture filtrates of T. pallescens ATCC 96155 after 6 days of incubation, while no activity was detected at 14 days. One active fraction was selected following fractionation and preparative chromatography and was bioassayed against Podosphaera (sect. Sphaerotheca) xanthii (Castagne) U. Braun & N. Shishkoff and a number of other fungi. A concentration of 130 microg/mL inhibited germ tube development in P. xanthii, and mildew spores appeared plasmolyzed. Other fungi were inhibited at higher concentrations. Collapse of hyphae and conidiophores was also observed on mildewed leaves treated with the active fraction. Proton NMR analysis indicated that the inhibitory compound was a fatty acid ester. In 3- to 6-day-old cultures of T pallescens ATCC 96155 demonstrating biological control activity, antifungal compound production may have a primary role in restricting growth of mildew fungi and other competitors when applied to leaves.  相似文献

A Kluyveromyces strain secreting a fungicidal proteinaceous toxin has been isolated. Its maximal activity is observed at pH 5.0 and an increased osmotic pressure. Four strains of ballistoconidium-forming yeast-like fungi (K-95, K-125, K-132 and K139), isolated from plants collected in Bangkok, Thailand, were assigned to the genus Tilletiopsis based on morphological and chemotaxonomical characteristics. On the basis of sequence data of 18S rDNA and the D1/D2 region of 26S rDNA, strains K-95, K-125 and K-132 were close to T. flava and T. fulvescens, and strain K-139 each formed related to T. minor. DNA-DNA reassociation experiments with related species revealed that strains K-125, K-132 and K-139 each formed a new and distinct species whereas strain K-95 was identified as T. flava. Tilletiopsis derxii Takashima et Nakase sp. nov. (JCM 10217T; K-125), Tilletiopsis oryzicola Takashima et Nakase sp. nov. (JCM 10218T; K-132), and Tilletiopsis penniseti Takashima et Nakase sp. nov. (JCM 10216T; K-139) are the names proposed for the new taxa.  相似文献

Five yeast strains belonging to the genus Cryptococcus Vuillemin were isolated from steppe plants and turf collected in the Prioksko-terrasny biosphere reserve (Moscow region, Russia). Sequence analyses of the D1/D2 domains of the 26S rDNA and of the internal transcribed spacer region revealed that these yeast strains and strain CBS 8016 have almost identical sequences and belong to the Holtermannia clade of the Tremellomycetidae (Basidiomycota, Hymenomycetes). A novel species named Cryptococcus festucosus (type strain VKM Y-2930) is proposed to accommodate these strains. Physiological characteristics and mycocin sensitivity patterns distinguishing Cryptococcus festucosus from the other species of this clade are presented.  相似文献

Double-stranded RNA viruses of about 35 nm in diameter were isolated from a mycocin-secreting strain of Cryptococcus aquaticus. A derivative of this strain, lacking small dsRNA, was non-mycocinogenic and sensitive to its own toxin. The killing pattern of this mycocin was restricted to some species of the Cystofilobasidiales clade. Despite the differences in genome size of dsRNA viruses in mycocinogenic strains of Cryptococcus aquaticus, Cystofilobasidium sp. CBS 6569, Cystofilobasidium bisporidii, Cystofilobasidium infirmominiatum, Trichosporon pullulans and Xanthophyllomyces dendrorhous and killing patterns of their mycocins, the viral genomes showed homology in hybridisation experiments.  相似文献

The mycocin with a molecular mass of at least 15 kDa secreted by Cryptococcus pinus exhibited fungicidal activity at pH values below 6.5. It was thermolabile and resistant to proteases. Sporidiobolus salmonicolor ATCC 623 and Tilletiopsis ashingtonensis NRRL Y-2555 grew on carbon resources provided as volatiles by ripe Golden Delicious; apples. This ability was not correlated with the reported natural habitats of the 21 species (26 strains) tested. Ethylene, the major volatile produced, was not utilized but butyl acetate, hexyl acetate and hexyl-2-methyl-butanoate (identified by GC-MS) were. These yeasts also assimilated ethanol, butanol, hexanol (Tilletiopsis excepted), acetate, propionate, butyrate and ethyl acetate at appropriately low concentrations. Ethanol and acetate aside, this is the first report of such assimilations by any yeast.  相似文献

Botryodiplodia disease caused by Botryodiplodia theobromae is a recently disease of some Annona cultivars in Egypt, particularly in Behera Governorate, characterized by stem purple lesions, dieback, flowers, and fruits dry and soft rot. Six fungal and bacterial bioagents, i.e., Trichoderma koningii, Trichoderma hamatum, Pseudomonas fluorescens, Pseudomonas putida, Tilletiopsis minor, and Tilletiopsis washingtonensis were tested either solely or at different amalgamations against Botryodiplodia disease, as foliar spraying using three Annona cultivars, i.e., Balady and Abd El-Razik (Annona squamosa) and Hindy (Annona cherimola). In vitro, studies revealed a significant inhibition towards the conidial germination of B. theobromae as well as on the disease incidence on artificially inoculated branches and fruits in the presence of the aforementioned bioagents. An unmistakable reduction in the disease was conspicuous under the action of multi-bioagent conduct. The bioagents were tested during 2003 and 2004 agricultural seasons under the field conditions at Nobaria, (Behera Governorate). A single application and all possible mixture of two or three of the bioagents were applied at 15 days intervals as a foliar spray. Botryodiplodia disease severity and sporulation of the pathogen were always reduced, when the multi-bioagents were applied. When Trichoderma spp. and Pseudomonads spp. were blended together, the disease was greatly abridged in the three tested cultivars compared to any of the sole bioagents. The multi-bioagents were more effective than any sole or even double treatments. The application of multi-bioagents also resulted in a significant increase of fruit yield.  相似文献

Colonization of apples by ballistoconidium-forming fungi causes a new disorder, here named 'white haze'. White haze may occur in mild form in the field, but only becomes problematic after Ultra-Low Oxygen storage, and, therefore, may be considered as a postharvest disorder. All isolates, obtained using the spore-fall method, were morphologically identified as anamorphs of smut fungi belonging to the genus Tilletiopsis. Sequence analysis of the D1/D2 and the ITS domains of the rDNA revealed nine novel taxa scattered among the Exobasidiomycetidae (Ustilaginomycetes). Field experiments confirmed the erratic incidence of white haze over the years, and the development of the disorder seems to be enhanced at lower temperatures and a high relative humidity. Several scab fungicide treatments showed diminishing effects on the incidence of white haze.  相似文献

Aquatic Metschnikowia species (M. australis, M. bicuspidata, M. krissii, and M. zobellii) are sensitive to 10 out of 12 mycocins secreted by Pichia membranifaciens strains. Terrestrial species M. pulcherrima and M. reukaufii are resistant to all these mycocins, while M. gruessii and M. lunata are sensitive to one of them.
