The controls of late dicot embryogenesis and early germination   总被引:14,自引:0,他引:14  
During seed formation, the embryo appears to be germinable as soon as cell division is completed; however, it continues development on the plant. This review describes the stages of development after cell division and provides a summary of important observations and recent use of molecular markers as they apply to the regulation of dicot seed formation. Genetic evidence suggests that abscisic acid may help initiate late embryogenesis, although no evidence firmly establishes that abscisic acid controls any other aspect of late dicot development. Previous studies utilizing cultured embryos have implicated abscisic acid and water potential as endogenous promoters of late embryogenesis and inhibitors of germination. However, these embryo culture experiments have been misinterpreted. The experiments show that both immature and mature embryos respond to environmental water stress by expressing a developmental program that is normally induced in late embryogenesis by abscission of the vascular connection. This postabscission program probably prepares the embryo for its forthcoming desiccation during normal development and is predicted to be important in protecting the embryo from water stress during germination.  相似文献

Cloned mRNAs identify three programs of gene expression in cotton (Gossypium hirsutum L.) embryos that are associated with the maturation (reserve accumulation) stage, the postabscission stage, which is marked by expression of Late-embryogenesis-abundant (Lea) mRNAs, and germination (broadly defined as including all events through early postgerminative growth). In order to test if the regulation of these programs is the same in other dicotyledonous species, their expression was studied in normal and cultured maturation-stage, postabscission-stage, and mature embryo-stage embryos or seed of oilseed rape (Brassica napus L.), soybean (Glycine max [L.] Merr.), and tobacco (Nicotiana tabacum L.) using cotton and other cDNA probes. During postabscission, Lea mRNAs accumulated in all test species and were induced in earlier maturation-stage embryos by excision and culture on basal medium. Abscisic acid often enhanced this induction in the test species. Germinationspecific mRNAs were induced in cultured maturationstage and postabscission-stage embryos of all test species. These results indicate that the regulation of embryonic and germination programs is similar in all dicotyledons tested. Radioimmunoassay has been used to measure levels of wheat-germ agglutinin and barley-germ agglutinin during embryogenesis and germination. The two lectins exhibited similar patterns of accumulation during grain maturation in vivo and both decreased to low levels after imbibition of harvest-ripe grains for 3 d. Precocious germination of immature wheat and barley embryos excised and cultured in vitro could be prevented either by inclusion of abscisic acid or mannitol in the culture medium. Changes in the level of wheat-germ agglutinin induced by in vitro culture depended on the maturation stage of the embryo. No direct correlation was found between application of exogenous abscisic acid and accumulation of the lectin.  相似文献

Osmotic stress and abscisic acid induce expression of the wheat Em genes   总被引:8,自引:0,他引:8  
The early-methionine-labelled (Em) polypeptide is the single most abundant cytosolic protein of dry wheat embryos. It is encoded by messenger RNA which accumulates during the later (maturation) stages of embryogenesis. The accumulation of Em mRNA can be induced in isolated developing embryos, in culture, by the application of the plant growth regulator, abscisic acid, which prevents precocious germination. Precocious germination is also inhibited by the culture of embryos under conditions of osmotic stress when accumulation of Em mRNA is induced. This induction occurs in the absence of any significant increase in the endogenous levels of embryonic abscisic acid although there is a requirement for the continued presence of the growth regulator. Additionally, expression of Em genes can be repeated during early germination, if imbibing embryos are subjected to osmotic stress. Induction of Em-gene expression by osmotic stress is consistent with the proposed role of the Em polypeptide in mediating the remarkable tolerance of cereal embryos to the programmed desiccation undergone during their maturation.  相似文献

We have investigated events which take place in the developing barley (Hordeum vulgare L.) embryo during its acquisition of desiccation tolerance. Excised embryos are capable of precocious germination as early as 8 d after pollination (DAP). At this age, however, they are not capable of resisting a desiccation treatment which induces a loss of 96–98% of their initial water content. At 16 DAP the embryos germinate despite the drastic drying treatment. The pattern of in-vivo and in-vitro proteins synthesized by the developing embryos from 12 DAP (desiccation-intolerant) and 16 DAP (desiccation-tolerant) were compared. A set of 25–30 proteins was identified which is denovo synthesized or enhanced during the developmental period leading to desiccation tolerance. Abscisic acid (ABA; 100 M) applied in vitro for 5 d to 12-DAP embryos induces desiccation tolerance and represses a subset of polypeptides preferentially associated with 16-DAP embryos. During in vitro culture of barley embryos ABA stimulates the appearance of a set of proteins and prevents the precocious germination allowing embryogenesis to continue in vitro. It also suppresses a set of germination-related proteins which appear 4 h after the incubation of the dissected embryo on a germination medium without ABA. 通过对受精后烟草(Nicotiana tabacumL.)胚珠的体外培养,建立了一个简便、实用、高频率的胚胎发生研究系统。采用MS培养基附加6%蔗糖,然后换以2%蔗糖对烟草胚珠进行培养,结果受精后胚珠在体外可以完成正常的胚胎发生并可直接萌发成幼苗。追踪观察表明,合子体外胚胎发生过程中关键发育事件,如合子休眠期间的定向生长、一次不对称分裂的完成、胚柄的形成和解体、胚的分化等过程均可在体外重现。体外培养形成的胚珠及胚胎在形态上与自然状态下形成的胚珠及胚胎几乎没有差异。此体系操作简单,稳定性好,且关键发育事件的时空调控与自然胚胎发育进程相吻合。  相似文献

Somatic embryogenesis of coniferous species was first reported more than 20 years ago. Since then, there has been an explosion of research aimed at developing and optimizing protocols for efficient regeneration of plantlets. Although routinely used both as a means of propagation, as well as a valuable model system for investigating the structural, physiological, and molecular events occurring during embryo development, in vitro embryogenesis is still problematic for some coniferous species. Major problems include: low number of embryos generated; and low frequency of mature embryos able to convert into viable plantlets. Until recent years, despite the fact that embryogenesis is comprised of a sequence of defined steps which include proliferation of embryogenic tissue, embryo maturation, and germination, attempts at improving the whole procedure have been made almost exclusively during the maturation stage. This strategy was based on the assumption that successful regeneration is related to treatments provided during the development of the embryos. Major optimizations of the maturation medium have involved judicious selections of type and concentration of growth regulators, namely abscisic acid, and adjustments of the osmoticum of the culture medium. Extensive work has been conducted in defining the effects of plasmolysing and non-plasmolysing osmoticum agents during maturation, as well as in improving desiccation techniques required for the completion of the maturation program. In the last 2 years, however, work on spruce has clearly demonstrated that the early events in embryogenesis are crucial for the successful completion of the overall embryogenic program. The use of cell tracking techniques, implemented by physiological and molecular studies, has revealed that manipulations of the culture conditions early in the process can increase both number and quality of embryos produced in culture. Additional manipulations of the germination medium can also enhance germination and conversion frequency of somatic embryos matured in a sub-optimal environment. These new findings, together with the unraveling of molecular mechanisms involved in the control/regulation of embryo development hold considerable promise for clonal propagation in conifers.  相似文献

Synthetic seed technology requires the inexpensive production of large numbers of high-quality somatic embryos. Proliferating embryogenic cultures from conifers consist of immature embryos, which undergo synchronous maturation in the presence of abscisic acid and elevated osmoticum. Improvements in conifer somatic embryo quality have been achieved by identifying the conditions in vitro that resemble the conditions during in ovulo development of zygotic embryos. One normal aspect of zygotic embryo development for conifers is maturation drying, which allows seeds to be stored and promotes normal germination. Conditions of culture are described that yield mature conifer somatic embryos that possess normal storage proteins and fatty acids and which survive either partial drying, or full drying to moisture contents similar to those achieved by mature dehydrated zygotic embryos. Large numbers of quiescent somatic embryos can be produced throughout the year and stored for germination in the spring, which simplifies production and provides plants of uniform size. This review focuses on recent advances in conifer somatic embryogenesis and synthetic seed technology, particularly in areas of embryo development, maturation drying, encapsulation and germination. Summary We describe a tissue culture procedure for somatic embryogenesis and plantlet regeneration in cotton (Gossypium hirsutum L. cv. Coker 312). Callused explants or individual globular embryos were transferred to basal media to induce somatic embryogenesis. To determine characteristic early indicators of successful germination and conversion, we identified six types of embryos that developed on basal media. Two of the six embryo types, designated as tulip-shaped and trumpet-shaped, could undergo conversion in preliminary tests, whereas the others had little or no developmental potential. Several media treatments designed to enhance the maturation of globular somatic embryos failed to increase the fraction of embryos which matured to form recoverable types. In efforts to improve plantlet recovery, tulip-shaped embryos were used in limited trials to contrast the effects of chemical and physical desiccation treatments on germination and conversion. The selective use of tulip-shaped somatic embryos, coupled with partial desiccation, seems to have augmented plant recovery. Growth habit, flowering, seed set, and lint production of most of the regenerated plants were comparable to seed-derived plants grown under the same conditions. Low efficiency of embryo maturation, germination and conversion to plantlets is a major problem in many species including Persian walnut. We studied the effects of abscisic acid (ABA) and sucrose, on the maturation and germination of Persian walnut (Juglans regia) somatic embryos. Individual globular somatic embryos were grown on a maturation medium supplemented with different combinations of ABA and sucrose for ca. 1 month, until shoot meristems and radicles had developed. White and opaque embryos in late cotyledonary stage were subjected to desiccation after the culture period on maturation media. The number of germinated somatic embryos was influenced by the concentrations of ABA in the maturation medium. The best treatment for germination, in which both shoot and root were developed contained 2 mg l−1 ABA and resulted in 41% conversion of embryos into plantlets. Regeneration was reduced at higher levels of ABA. While ABA always reduced the rate of secondary embryogenesis, treatments containing 4.0% sucrose significantly increased the number of secondary embryos. On the other hand, sucrose had little influence on maturation. Normal and abnormal embryos were verified anatomically.  相似文献

Abscisic acid (ABA) is required for seed maturation in maize (Zea mays L.) and other plants. Gibberellins (GAs) are also present in developing maize embryos, and mutual antagonism of GAs and ABA appears to govern the choice between precocious germination or quiescence and maturation. Exogenous ABA can also induce quiescence and maturation in immature maize embryos in culture. To examine the role of GAs versus ABA in regulating maize embryo maturation, the effects of modulating GA levels were compared with those of ABA in embryos cultured at successive stages of development. The effects of GA synthesis inhibition or exogenous GA application differed markedly in embryos at different stages of development, indicating changes in both endogenous GA levels and in the capacity for GA synthesis as embryogenesis and maturation progress. In immature embryos, the inhibition of GA synthesis mimicked the effects of exogenous ABA, as shown by the suppression of germination, the acquisition of anthocyanin pigments, and the accumulation of a variety of maturation-phase mRNAs. We suggest that GA antagonizes ABA signaling in developing maize embryos, and that the changing hormone balance provides temporal control over the maturation phase.  相似文献

During precocious germination, Brassica napus L. embryos exhibitfeatures of embryo maturation and features of post-germinativegrowth at the same time, which raises important questions aboutthe nature of these developmental programmes and the transitionbetween them. To investigate the possiblity that the two programmesare segregated spatially rather than temporallhy in these embryos,embryos of different ages (excised at 38, 47, and 60 d post-anthesis)were placed in culture to induce precocious germination, andtotoal RNA was isolated from the cotyledons and axes or hypocotyls.cDNA sequences (two marking embryo maturation and four markingthe onset of post-germinative growth) were used as probes ondot blots to compare mRNA population dynamics in these organs.We find that individual organs accumulate mRNAs characteristicof only one developmental programme at any given time and concludethat the programmes are both distinct and potentially mutuallyexclusive. Embryos are able to ‘switch’ from accumulatingembryo maturation mRNAs to acumulating post-germinative mRNAsin culture, but the transition occurs at different times indifferent organs. Previous studies have identified a set of highly phosphorylated proteins of 23–25 kDa accumulated during normal embryogenesis of Zea mays L. and which disappear in early germination. They can be induced precociously in embryos by abscisic acid (ABA) treatment. Here the synthesis and accumulation of this group of proteins and their corresponding mRNAs were examined in ABA-deficient viviparous embryos at different developmental stages whether treated or not with ABA, and in water-stressed leaves of both wild-type and viviparous mutants.During embryogenesis and precocious germination of viviparous embryos the pattern of expression of the 23–25 kDa proteins and mRNAs closely resembles that found in non-mutant embryo development. They are also induced in young viviparous embryos by ABA treatment. In contrast, leaves of ABA-deficient mutants fail to accumulate mRNA in water stress, yet do respond to applied ABA. In water-stressed leaves of wild type plants the mRNAs are induced and translated into 4 proteins with a molecular weight and isoelectric point identical to those found in embryos.These results indicate that the 23–25 kDa protein set is a new member of the recently described class or proteins involved in generalized plant ABA responses.The different pattern of expression for the ABA-regulated 23–25 kDa proteins and mRNAs found in embryo and in vegetative tissues of viviparous mutants is discussed.  相似文献

Freshly harvested, developing kernels of maize (Zea mays L.)do not germinate up to 77 d after pollination, but can be inducedto do so by fluridone, premature desiccation, and isolationof the developing embryo. The pattern of protein synthesis indeveloping maize embryos is distinct from that during germinationand subsequent seedling growth. Premature desiccation at 35DAP elicits a pattern of protein synthesis upon rehydrationwhich is similar to that in germinated embryos from mature drykernels. Fluridone-induced viviparous germination is accompaniedby changes in the synthesis of some proteins to a post-germinativepattern, but some developmental proteins continue to be synthesized.Embryos isolated from developing kernels at 35 DAP germinatewhen incubated on water; they also produce some developmentalproteins during germination. Kernels from developing cobs at35 DAP which are detached from the mother plant and maintainedin an atmosphere of high relative humidity (moist controls)do not germinate, but neither do they continue a clearly definedpattern of either developmental or germinative protein synthesis.Drying is thus critical to effect a clear transition of proteinsynthesis from a developmental to a germinative mode in maizeembryos. Abscisic acid within the developing embryos is reduced by fluridone,but to a lesser extent by premature drying or maturation drying.Changes in sensitivity to abscisic acid by the developing embryomay be as, or more, important in permitting germination, andthe attendant synthesis of proteins, than changes in abscisicacid content. Embryo development and germination of Cyclamen persicum have been comparatively characterized for zygotic and somatic embryos with regard to mitotic activity and morphology in order to identify developmental abnormalities in somatic embryogenesis. Zygotic embryo development proved to be highly synchronous with distinct periods of cell division, cell elongation and embryo maturation within a total period of 17 weeks of seed development. Somatic embryo development was accomplished within only 3 weeks, resulting in a mixture of morphologically highly variable embryos. No distinct developmental periods could be identified and no reduction of the mitotic activity was discovered for non-desiccated somatic embryos. Controlled desiccation of somatic embryos severely reduced their germination rate, demonstrating resemblance of somatic embryos to recalcitrant seeds, whereas zygotic Cyclamen seeds could be characterized as typically orthodox.  相似文献

Compared to seeds, somatic embryos accumulated relatively low levels and different types of storage carbohydrates. The regulation of starch accumulation was studied to determine its effects on desiccation tolerance and vigor of dry somatic embryos. Somatic embryos of Medicago sativa are routinely matured through three phases: 7 days of development; 10 days of phase I maturation, a rapid growth phase; and 10 days of phase II maturation, a phase leading to the acquisition of desiccation tolerance. The control of starch deposition was investigated in alfalfa somatic embryos by manipulating the composition of the phase I maturation medium with different levels of sucrose, abscisic acid, glutamine and different types of carbohydrates and amino acids. After phase II maturation, mature somatic embryos were collected for desiccation and subsequent conversion, or for biochemical analyses. Starch deposition occurred primarily during phase I maturation, and variations in the composition of this medium influenced embryo quality, storage protein and starch accumulation. A factorial experiment with two levels of glutamine × three levels of sucrose showed that increasing the sucrose concentration from 30 to 80 g/l increased embryo size and starch content, but had minimal effect on accumulation of storage proteins; glutamine also increased embryo size, but decreased starch content and increased accumulation of the high salt soluble S-2 (medicagin) storage proteins. ABA did not influence any of the parameters tested when included in phase I maturation at concentration up to 10 μM. Replicating sucrose with maltose, glucose, or glucose and fructose did not alter embryo size or starch accumulation (mg/g fresh weight), but replacement with fructose alone reduced embryo size, and replacement with glucose alone reduced germination. Suplementation with the amino acids, asparagine, aspartic acid and glutamine increased seedling vigor, but decreased the starch content of embryos. The data indicate that starch accumulation in somatic embryos is regulated by the relative availability of carbon versus nitrogen nutrients in the maturation medium. The quality of mature somatic embryos, determined by the rate of seedling development (conversion and vigor), correlated with embryo size, storage protein and free amino acid but not with starch. Therefore, further improvements in the quality of somatic embryo may be achieved through manipulation of the maturation medium in order to increase storage protein, but not starch deposition.  相似文献

Summary The effect of abscisic acid (ABA) was evaluated during the maturation and germination of holm oak (Quercus ilex L.) somatic embryos. The addition of ABA to the culture medium significantly reduced unwanted recurrent embryogenesis in mature somatic embryos without affecting the germination of embryos subjected to stratification at 4°C. Stratification at 4°C for 2 mo. was the most efficient for stimulating somatic embryo germination of holm oak. The addition of 90 and 450 mM sucrose also improved germination, while higher sucrose concentrations were inhibitory.  相似文献

. In most plants, somatic embryos tend to germinate prematurely, a process that is detrimental to controlled plant production and the conservation of artificial seeds. We investigated the dormancy characteristics of Siberian ginseng somatic embryos induced simply by a high sucrose treatment, a treatment that enables the long-term conservation of artificial seeds following encapsulation and provides embryos with an enhanced resistance to dehydration. Early-cotyledonary stage somatic embryos were mass-produced by means of bioreactor culture. These embryos were then plated on medium supplemented with various levels of sucrose (1%, 3%, 6% or 9%) and allowed to mature. Subsequent germination of these embryos following the maturation period depended significantly on the sucrose level. At concentrations of 9% sucrose, none of the somatic embryos germinated after maturation, and none were recovered after being transferred to half-strength MS medium containing 2% sucrose. Gibberellic acid treatment was necessary to induce germination; other growth regulators such as auxins and cytokinins did not induce a response. Endogenous abscisic acid content in somatic embryos matured at 9% sucrose (487.8 ng/g FW) was approximately double that found in those matured at 3% sucrose (258.4 ng/g FW). This indicates induced dormancy in embryos under high osmotic stress. Alginate encapsulation of embryos facilitated the artificial induction of dormancy to extend the conservation period without germination. The induction of dormancy strengthened resistance to dehydration after the embryos were desiccated to 15% of their normal water content. Reduced chances of embryo survival during long-term desiccation were distinctly delayed in dormant embryos. These results indicate that the induction of dormancy in embryos is a promising application for synthetic seed production.  相似文献
