Summary During the rapid growth phase ofStreptomyces clavuligerus in a 10 litre fermentor, the level of dissolved oxygen (DO) was found to drop to almost zero for a period of approximately 10 h, delaying the appearance of and lowering the production of the antibiotic cephamycin C. Controlling the DO at either 50% or 100% throughout the fermentation did not significantly alter the specific growth rate of the culture, but did elevate final antibiotic levels two- and three-fold respectively. The improved oxygen availability affected antibiotic production both by increasing the rate of specific cephamycin C bisosynthesis and by maintaining this higher rate throughout the production period. These results demonstrate that controlling dissolved oxygen levels close to saturation during periods of rapid growth markedly improves the efficiency and duration of cephamycin C biosynthesis inS. clavuligerus.  相似文献

Streptomyces sp. P6621-RS1726, a high cephamycin C producing mutant, usually produces cephamycin C together with its precursor, penicillin N, in flask fermentation. The fermentation conditions for selective and high yield production of cephamycin C were investigated using 20-l jar fermentors. As a result, cephamycin C was preferentially produced without penicillin N when the DO level was maintained at least higher than at 10% during the production phase. However, when the agitation speed was too high at the initial stage of the fermentation, the cell growth was inhibited, resulting in suppression of both cephamycin C and penicillin N production, even if the DO was maintained at a high level. Based on these experiments, a computer control system for preferential and high-yield production of cephamycin C was constructed. By using this computer control system, scale-up from a 20-l jar fermentor to a 1,500-l pilot fermentor was carried out with control of the DO level at 20% of saturation during the cephamycin C production phase. As a result, the profile of cephamycin C production in the former was favorably reproduced in the latter.  相似文献

Different dissolved oxygen concentrations and aeration rates were imposed on a stable mutant of Streptomyces fradiae during the antibiotic-producing phase. At high aeration rate (1 vvm), the tylosin yield in the fermentor broth with dissolved oxygen (DO) concentrations controlled close to 100% saturation (6-8 ppm) increased 10% as against uncontrolled. The rates of cellular growth, oil consumption, and tylosin production were severely reduced when DO concentration fell below 25% saturation, but all resumed to their initial rates when DO was raised to saturation level again. The DO concentration in combination with air flow rate affected the pattern of the antibiotics produced. At high DO levels, an additional macrolide antibiotic, macrocin, was synthesized to more than one-third the amount of tylosin at high aeration rate (1 vvm). On the other hand, tylosin production rate remained constant and no significant amount of macrocin was produced at low aeration rate (0.2 vvm).  相似文献

The growth kinetics, sporulation, and toxicity of Bacillus thuringiensis var. israelensis were evaluated through the analysis of batch cultures with different dissolved oxygen (DO) profiles. Firstly, DO was maintained constant at 5%, 20%, or 50% throughout fermentation in order to identify the most suitable one to improve the main process parameters. Higher biomass concentration, cell productivity, and cell yield based on glucose were obtained with 50% DO. The higher aeration level also resulted in higher spore counts and markedly improved the toxic activity of the fermentation broth, which was 9-fold greater than that obtained with 5% DO (LC50 of 39 and 329 mg/L, respectively). Subsequently, using a two-stage oxygen supply strategy, DO was kept at 50% during the vegetative and transition phases until the maximum cell concentration was achieved. Then, DO was changed to 0%, 5%, 20%, or 100% throughout sporulation and cell lysis phases. The interruption of oxygen supply strongly reduced the spore production and thoroughly repressed the toxin synthesis. On the contrary, when DO was raised to 100% of saturation, toxic activity increased approximately four times (LC50 of 8.2 mg/L) in comparison with the mean values reached with lower DO levels, even though spore counts were lower than that from the 50% DO assay. When pure oxygen was used instead of normal air, it was possible to obtain 70% of the total biomass concentration achieved in the air assays; however, cultures did not sporulate and the toxin synthesis was consequently suppressed.  相似文献

When dissolved oxygen (DO) was maintained at saturation level during batch fermentations of Streptomyces clavuligerus (NRRL 3585), the accumulation of the intermediate penicillin N was lowered while formation of the end product cephamycin C was increased relative to fermentations without DO control. The specific activity of the penicillin ring-expansion enzyme deacetoxycephalosporin C synthase (DAOCS) was increased 2.3-fold under oxygen saturated conditions, whereas the penicillin ring-cyclizing enzyme isopenicillin N synthase (IPNS) showed only a 1.3-fold increase. Thus oxygen derepression of DAOCS appears to be an important regulatory mechanism in the conversion of penicillin N to cephamycin C in S. clavuligerus. IPNS, an early acting enzyme in cephamycin C biosynthesis, and DAOCS, which acts late in the pathway, both disappeared from cell extracts at 60 h, just prior to cessation of cephamycin production.  相似文献

研究了溶氧浓度对产甘油假丝酵母分批发酵生产甘油过程的影响。实验结果表明:当溶氧浓度控制在30%时,C. glycerinogenes的甘油产量、得率和产率达到最高,分别为120.7 g/L、0.575 g/g和1.69 g/(L•h),而糖酵解代谢副产物形成最少。当溶氧浓度为10%时,发酵过程呈现出“巴斯德效应”的特征,生成的酵解代谢副产物维持在较高水平。在快速生长阶段,随着溶氧从10%增加到60%,细胞呼吸类型表现为从厌氧呼吸向好氧呼吸转变,酵解代谢副产物依次减少。在生长稳定期,控制的溶氧浓度越高,酵解代谢副产物乙醇、乙酸等的生成减少。分别选用Logistic方程、Luedeking-Piret方程和Luedeking-Piret-like方程,能较好地模拟细胞生长、甘油合成和葡萄糖消耗的动力学过程。  相似文献

Summary The multi-subunit enzyme, -(L--aminoadipyl)-L-cysteinyl-D-valine (ACV) synthethase catalyses the first step in the biosynthetic pathway of the -lactam antibiotic, cephamycin C. In batch fermentations of Streptomyces clavuligerus, ACV synthetaase activity appeared during the rapid growth phase. Over the same period the dissolved oxygen (DO) content of the medium was depleted to zero and remained there for nearly 10 h. Maintainance of the DO at saturation throughout the fermentation did not change the maximum ACV synthetaase specific activity, but did reduce the in-vivo stability of the enzyme. Oxygen saturation lowered the maximum intracellular ACV levels to one-sixth of those accumulated in the fermentor with no oxygen control, due principally to an improvement in the conversion of ACV to the penicillin N intermediate. Increased oxygenation also improved ACV conversion to cephamycin C, which demostrated that the activity of both an early and a later enzymatic step in cephamycin biosynthesis was limiting antibiotic production under restricted oxygen conditions. When the level of dissolved oxygen was increased to saturation in defined media fermentations of Streptomyces clavuligerus, the total duration of activity of the penicillin ring cyclization enzyme, isopenicillin N synthase (IPNS), was extended by at least 20 h; however, no increase in the stability of the ring expansion enzyme, desacetoxycephalosporin C synthase (DAOCS), was observed. Consequently, the conversion of the excreted intermediate penicillin N to cephamycin C was 15-20% less efficient at this high oxygen concentration. The increased dissolved oxygen level also led to the complete loss of IPNS and DAOCS activities for 4 h during the period of fastest growth, and the rate of specific cephamycin C production fell to zero. A several hundred fold increase in the level of iron in the defined media resulted in a sixfold improvement in the rate of specific cephamycin C production after 60 h fermentation. This increased rate appeared to be due to an elevation in the in vivo activities of a number of the cephamycin biosynthetic enzymes, particularly those catalysing later pathway steps.  相似文献

Enterotoxin B, nuclease, and total exoprotein production by Staphylococcus aureus strain S-6 was studied in a 0.5-liter fermentor system. While these extracellular products were elaborated over a wide range of aeration rates, maximal production occurred within the very narrow range of 125 to 150 cm(3) of air per min. The levels attained at the optimal aeration rate were not increased by maintaining a constant pH, although yield of enterotoxin:cell mass was highest at a constant pH of 7.0. During the growth cycle of the cultures, when aeration rate alone or aeration rate and pH were held constant, the dissolved oxygen (DO) levels, initially set at 100% of saturation, decreased to 5 to 10% 4 to 5 h after inoculation. The oxygen demand of the culture then maintained this level for an additional 4 to 6 h. This interval of low DO was characterized by maximal growth and exoprotein production. When the DO was controlled at a constant value throughout growth (by increasing or decreasing the airflow rate as appropriate), the culture demonstrated different optima for maximal growth and exoprotein production. A constant DO of 100% stimulated growth to extremely high densities, but the accumulation of toxin and nuclease was not observed. On the other hand, maintaining constant DO levels at 50 or 10% raised exoprotein levels higher than those achieved in a culture grown at the optimal aeration rate. Compared to the optimal aeration rate culture, the 10% DO culture yielded 20% more nuclease, 25% more toxin, and 40 to 50% more total exoprotein. These results indicate that it is the DO and not the aeration rate, per se, that is influential in controlling growth, toxin, nuclease, and total exoprotein production.  相似文献

Azospirillum lipoferum crt1 was grown in batch cultures under standard conditions at 85% saturation of dissolved oxygen (DO) and 30-g/liter glucose concentrations. Kinetic studies revealed nutritional limitations of growth and the presence of an initial lag phase prior to consumption of glucose. The influences of various gaseous environments and shear stress on growth, i.e., various conditions of agitation-aeration, were characterized. Faster growth in the first stages of the culture and shorter duration of the lag phase were observed at DO concentrations of <30% saturation. The possible influences of dissolved CO(2) concentration or shear stress or both were discounted, and we confirmed the detrimental effect of high DO levels (up to 80% saturation) and the favorable influence of low DO concentrations (lower than 30% saturation) on growth. It was concluded that the gaseous environment, i.e., the DO concentration, needs to be considered as an operating parameter and be accurately controlled to ensure optimal growth of Azospirillum cells.  相似文献

The growth of Saccharomyces carlsbergensis in continuous culture has been studied when dissolved oxygen and glucose concentrations were held constant at a series of steady-state levels. Both oxygen and glucose controlled the degree of aerobic metabolism and of ethanolic fermentation. When the glucose uptake rate was low (between 1.2 and 2.8 mmoles per hour per gram of yeast) the relative distribution of glucose between ethanolic and aerobic fermentation was sensitive to oxygen: when dissolved oxygen was near to saturation, glucose metabolism was 0.98 aerobic; when dissolved oxygen was 0.01 saturated, 0.8 of intake glucose metabolism was by ethanolic fermentation. On the other hand when glucose intake was high (between 7.6 and 18.2 mmoles) metabolism was predominately by ethanolic fermentation even when dissolved oxygen concentration was at saturation. The extent, to which catabolism proceeded by an anaerobic or aerobic pathway, as judged by ethanol production, was controlled more by the uptake of glucose than of oxygen.  相似文献

The aim of this study was to examine nitrogen removal from slaughterhouse wastewater in a laboratory-scale sequencing batch reactor (SBR) operated at low dissolved oxygen (DO) levels under two aeration strategies: intermittent aeration (IA) and continuous aeration (CA). Under the IA strategy, during the aeration periods, the maximum DO was controlled at 10% saturation; under the CA strategy, in the first hour of the react phase, the DO was maintained at 10% saturation, and then it was kept at 2–3% saturation in the remaining react phase. Total nitrogen removals of up to 95 and 91% were achieved under the IA and CA aeration strategies, respectively. It is proposed that in situ measurement of oxygen utilization rates can be used to control the operation of SBRs for nitrogen removal.  相似文献

甲醇营养型毕赤酵母表达外源蛋白是在醇氧化酶(alcohol oxidase,AOX)启动子(PAOXI)严格调控下进行的,然而这种启动子在转录水平受到葡萄糖的阻遏。本文研究了毕赤酵母在葡萄糖替代甘油为生长相碳源时表达重组植酸酶蛋白的发酵特征。结果表明:初始葡萄糖浓度为20dL的细胞得率高,为0.39g[DCW]/g。通过基于实时参数(溶氧和呼吸商)调控的葡萄糖补料策略,生长相40h后细胞密度达到100g[DCW]/L,甲醇诱导100h后植酸酶产量达到2200FTUphytase/mL,甲醇得率系数为0.25FTU phytase/gmethnol。因此,在毕赤酵母高表达重组蛋白培养中葡萄糖能够用作生长相基质,并能实现重组蛋白的高效表达。  相似文献

为进行高密度发酵并实现外源基因的高表达,在表型为MutS的重组毕赤酵母(Pichia pastoris)表达人血管生长抑制素的诱导阶段,采用了甘油甲醇混合补料的培养方式。以溶氧水平作为甘油代谢指针来控制甘油限制性流加既可维持一定菌体生长,又不会发生发酵液中残余甘油及有害代谢产物(乙醇)阻遏蛋白表达。当表达阶段的菌体平均比生长速率控制于0.012h-1,菌体浓度达150 g/L,血管生长抑制素浓度最高达到108 mg/L,血管生长抑制素的平均比生产速率为0.02 mg/(g·h),菌体关于甘油的表观得率为0.69 g/g,菌体关于甲醇的表观得率为0.93g/g,较没有采用甘油限制性流加时都有所提高。  相似文献

Experiments confirmed dissolved oxygen (DO) definitely affects cyclic adenosine monophosphate (cAMP) production by Arthrobacter A302. Production of cAMP by batch fermentation was investigated under various DO conditions. A two-stage DO control strategy was proposed to achieve optimal production of cAMP based on the kinetic analysis: the DO level was controlled at 40% during the first 18?h and then switched to 30%. Relatively high cAMP production (9.9?g?L(-1)) was achieved by applying this strategy. The cAMP productivity (0.14?g?L(-1)?h(-1)) was also successfully improved by 85.1, 59.3, 15.1 and 28.0%, compared to cases in which DO was uncontrolled or DO levels were held at 20, 30 and 40%, respectively. This is the first report of the use of a two-stage DO control strategy in cAMP production, and it was verified to be an effective method for enhancing the cAMP yield via this strain.  相似文献

Magnetotactic bacteria are difficult to grow under defined conditions in culture, which has presented a major obstacle to commercial application of magnetosomes. We studied the relationships among the cell growth, magnetosome formation, dissolved oxygen concentration (DO), and the ability to supply oxygen to the cells. Mass culture of Magnetospirillum gryphiswaldense MSR-1 for the production of magnetosomes was established in a 42-L fermentor under the following conditions: (1) sterile air was the sole gas supplied in the fermentor, and DO could be regulated at any level below 10% saturation by cascading the stir rate to DO, (2) to resolve the paradoxical situation that the cell growth requires higher DO whereas magnetosome formation requires low DO below the detectable range of regular oxygen electrode, DO was controlled to optimal level using the change of cell growth rate, rather than reading from the highly sensitive oxygen electrode, as the signal for determining appropriate DO, and (3) timing and rate of supplying the substrates were determined by measuring cell density and Na-lactate concentration. Under these conditions, cell density (OD565) of strain MSR-1 reached 7.24 after 60-h culture in a 42-L fermentor, and cell yield (dry weight) was 2.17 g/L, the highest yield so far being reported. The yield of magnetosomes (dry weight) was 41.7 mg/L and 16.7 mg/L/day, which were 2.8 and 2.7 times higher than the previously reported yields.  相似文献

在7 L发酵罐中研究了溶氧和pH对产朊假丝酵母分批发酵生产谷胱甘肽的影响。结果表明,当葡萄糖浓度为30 g/L且通气量控制在5 L/min时,搅拌转速达到300 r/min即可满足细胞生长和谷胱甘肽合成对溶解氧的需求。不同pH控制方式对谷胱甘肽分批发酵的影响有较大差异。不控制pH时,细胞干重和谷胱甘肽产量比控制pH为55的发酵分别低27%和95%,且有50%的谷胱甘肽向胞外渗漏。研究了将pH控制在4.0、4.5、5.0、5.5、6.0和6.5的谷胱甘肽分批发酵过程,发现在pH 5.5时谷胱甘肽总产量最高。用前期研究建立的动力学模型模拟了不同pH (4.0~6.5)下的分批发酵过程,并从动力学角度解释了pH对细胞生长和谷胱甘肽合成的影响。  相似文献
