To obtain a homothallic mutant in Lentinula edodes, basidiospores derived from the common Bmut dikaryon (A1B1mut × A2B1mut) were treated with UV irradiation. Of a total of approximately 5000 monosporous cultures recovered, a single basidiospore isolate was found to produce the hyphae bearing clamp connections without mating. This mutant strain could form fruit bodies, and all its single basidiospore isolates developed into colonies with clamp connections. Such homothallic behaviors were transmitted from the mutant strain to the next generation. During the germination and following hyphal elongation in a single basidiospore of mutant strain, clamp connections were clearly detected in multicellular hyphae, which contained two nuclei in each cell. Their clamp connections were morphologically variable, viz., pseudo, abnormal, and true clamps. Amplified fragment length polymorphism (AFLP) profiles among the basidiospore isolates of mutant strain were identical, indicating that the mutant strain produced isogenic basidiospore progeny. Agaricus brasiliensis is a medicinal mushroom native to Brazil. It was first identified as Agaricus blazei and its scientific name continues to be debated. We examined the cytology of different Brazilian commercial strains of A. brasiliensis and the nuclear behavior of strain CS1 during basidiospore development using fluorescent microscopy. All strains have multinucleate hyphae and no significant differences in nuclei numbers were observed between them. Basidia from A. brasiliensis strain CS1 are typically tetrasporics and produce binucleate basidiospores, demonstrating that a postmeiotic mitosis occurs during basidiospore development. This result suggests that A. brasiliensis is primarily a heterothallic species.  相似文献

Exobasidium symploci-japonicae var. carpogenum, causing Exobasidium fruit deformation on Symplocos lucida collected in Fukuoka Prefecture, Japan, is newly described based on morphological observations of hymenial structure and mode of basidiospore germination. This new variety differs morphologically from the type variety, particularly in the septal number of basidiospores and in the shapes and sizes of conidia formed on the medium. Spongipellis litschaueri, described in 1931 from Austria, is often synonymised with S. delectans in current literature. Morphological and molecular approaches were applied to revise the taxonomic position of this species. The basidiospore dimensions and DNA sequences confirmed differences between S. delectans and S. litschaueri, thus the latter should be kept as a distinct species. However, the relationship between S. litschaueri and its North American kin S. unicolor is complex. Although most of the examined specimens of S. unicolor differ significantly from S. litschaueri in morphological and molecular characters, one American isolate showed markedly high DNA sequence similarity to the European species. Moreover, specimens of S. unicolor from the more or less western USA have larger basidiospores than the others and the respective values are similar to S. litschaueri. Thereby, S. unicolor may split in two taxa where one is closely related to S. litschaueri. Additionally, DNA sequences of a large subunit of the nuclear ribosomal RNA gene were analysed to elucidate the position of Spongipellis within the Polyporales. The results revealed unexpected polyphyly when the generic type S. spumeus fell into a separate lineage from S. delectans, S. litschaueri, S. pachyodon, and S. unicolor. In addition, Tyromyces fissilis occasionally kept in Spongipellis is not related to any species of the genus nor to Tyromyces chioneus, the generic type of Tyromyces. Coprinopsis novorugosobispora (Basidiomycota, Psathyrellaceae), a new ammonia fungus, fructificated from the soil sample collected in Beijing, China. In macro- and micro-morphology, this species is similar to C. rugosobispora reported from Belgium, but differs in having smaller basidiospore and rougher basidiospore ornamentation. The Phellinus igniarius group comprises several closely related wood-decaying basidiomycetes with poroid hymenophores that are sometimes difficult to identify on a morphological basis. The delimitation of pileate species belonging to the group was the subject of ITS (internal transcribed spacer of ribosomal DNA) and tefa (translation elongation factor 1 alpha) DNA sequence analyses applied to specimens from central and northern Europe. The results confirmed the distinctiveness of P. alni, P. igniarius, P. lundellii, P. nigricans, P. populicola, P. tremulae, and P. tuberculosus in Europe. The specimens of the previously distinguished species P. cinereus were found to be identical with either P. nigricans or P. alni. Thus, Phellinus cinereus does not follow the species criteria of phylogenetic species recognition. In addition, a recently described species, P. neolundellii, was grouped within the P. alni clade. The ITS and tefa analyses produced a different topology for P. populicola and P. igniarius. P. alni had the largest spectrum of hosts, including woody plants from nine families, and records on Aesculus hippocastanum and Ulmus glabra are reported for the first time. P. igniarius s.s. has been collected not only on Salix spp. as expected but also, though rarely, on Populus nigra, and it has been observed once on Malus domestica. The host specificity of the remaining species resembles previous data. An additional analysis of basidiospore dimensions did not reveal any differences between P. alni and P. igniarius, but both are distinguishable from those of P. nigricans.  相似文献

Effects of pH, NH4-N, and temperature on basidiospore germination in Coprinopsis austrophlyctidospora from New Zealand, C. phlyctidospora from Japan, C. aff. rugosobispora from Canada, and C. echinospora from Canada were investigated. The Coprinopsis spp. required the presence of ammonium-nitrogen under weak alkaline to neutral conditions for germination, regardless of their different areas of occurrence. The former two species had a wider concentration of NH4Cl solution and pH range for germination in comparison to the latter two species. The optimum concentration of NH4Cl solution for the germination was 0.01 M in C. austrophlyctidospora and 0.1 M in the other three species. The pH optimum for germination in the former two species was 8.0 whereas that for germination in the latter two species was 8.0–8.5. The temperature range (5.0–40.0°C) for the former two species was wider than that (5–30°C) for the latter two species. Temperature optima for the germination in the former two species, C. aff. rugosobispora and C. echinospora, were 30, 20–25 and 15°C, respectively. The germination abilities of these Coprinopsis species in a wide range of temperatures are relevant to their natural temperature regime, showing their potential ability to propagate in tropical to subarctic regions.  相似文献

Structure and ornamentation of basidiospore walls in Xerula (inclusive of Dactylosporina) are illustrated using scanning electron microscope (SEM) images. Oudemansiella spores are not depicted, but all sections of Xerula are represented by 17 taxa. Spore wall sculpturing cannot be used as an infrageneric diagnostic character, nor to separate Oudemansiella and Rhizomarasmius from Xerula. Xerula caulovillosa is proposed as a new species, and Xerula radicata f. marginata as a new combination.  相似文献

Mentzelia hualapaiensis, a new species of Mentzelia sect. Bartonia, is described from the Grand Canyon region of Arizona. The new species is closely related to M. puberula, which is found west of M. hualapaiensis along the Colorado River, and to M. oreophila, M. polita, and M. tiehmii. It shares with these species a suffrutescent shoot system characterized by a subterranean, branching caudex, multiple annual branches, and similar leaves that have shallowly lobed or toothed to entire laminas. The flowers of M. hualapaiensis differ from those of its closest relatives in having cream-white, linear to narrowly spatulate petals and staminodes, characteristics that are convergent with those of the flowers of the Chihuahuan Desert species M. humilis.  相似文献

A new yeast species, Kazachstania wufongensis, is proposed in this paper based on six strains isolated from soil in Taiwan. The species may produce one to four ellipsoidal ascospores in each ascus, directly transformed from diploid cells. Genus assignment and distinction of the species from other recognized species of Kazachstania is based on morphological and physiological characteristics, and on phylogenetic analysis of nucleotide sequences of the D1/D2 domains of the large subunit (LSU) rRNA gene. Sequence analysis of the D1/D2 domains of the LSU rRNA gene reveals that K. wufongensis is a member of the Kazachstania exigua complex, and its phylogenetically closest relatives are K. exigua, K. barnettii, K. bulderi, and K. turicensis. The species can be further differentiated from the other phylogenetically related species based on internal transcribed spacer sequence and electrophoretic karyotype. Therefore, the new species Kazachstania wufongensis sp. nov. is proposed. The type strain of this new species, which was isolated from forest soil in Wufong, Hsinchu, Taiwan, is FN21S03T (=CBS 10886T = BCRC 23138T).  相似文献

Agave manantlanicola, a new endemic species from the high altitude mountains of the Sierra de Manantlán in western Mexico, is described and illustrated. The species belongs to the subgenus Littaea and Gentry’s Amolae group. The new species is compared to other morphologically similar species from western Mexico, including Agave attenuata subsp. attenuata, A. attenuata subsp. dentata, A. bakeri, A. chazaroi, and A. vazquezgarciae. The aim of the present work was to determine phylogenetic relationships among Astraeus species and to support macroscopic and microscopic characters of Astraeus with analysis of the ITS rDNA region. Collections of Astraeus basidiomes were made from different geographical areas in Thailand and compared with existing collections made worldwide. The marriage of observations on morphological features, including basidiospore ornamentation and molecular data demonstrated the presence of several Astraeus species. Sequences for 41 Astraeus collections were compared and the phylogenetic analyses grouped Thai Astraeus collections into two distinct groups. One contained A. odoratus and an Asian species described herein as A. asiaticus. There are at least two additional species: A. pteridis, and one so far un-named from North America. Our results show that molecular data can be used in combination with traditional morphological characteristics to resolve taxonomic uncertainties in the genus Astraeus.  相似文献

Kuehneola japonica has a microcyclic life cycle with a regular alternation of generations. Single basidiospore inoculations onto Rosa wichuraiana resulted in teliospore production, indicating its homothallic nature. Dikaryotization in a vegetative mycelium in the host seemed to occur through nuclear division that was not followed by septum formation. Karyogamy and meiosis took place through teliospore and metabasidium development; this fungus was considered to reproduce genetically homogeneous progenies. Puccinia lantanae and P. patriniae were also microcyclic in their life cycle; however, these fungi differed from K. japonica in the mode of nuclear behavior. In the former two fungi, both vegetative and reproductive cells were uninucleate. No karyogamy was observed, and nuclear division in the metabasidium development was thought to be mitotic. In P. lantanae, a basidiospore was formed on a sterigma, whereas a whiplike hypha emerged from each metabasidium cell in P. patriniae. Inoculations of Justicia procumbens with a single basidiospore of P. lantanae resulted in teliospore production. The fungus seemed to remain uninucleate, either haploid or diploid, throughout the life cycle. Thus, reproduction was considered to be apomictic. Streptomyces albovinaceus, Streptomyces caviscabies, Streptomyces griseus, Streptomyces setonii, and Streptomyces virginiae selected as antagonists of Moniliophthora (ex Crinipellis) perniciosa, the causal agent of cacao Witches’ broom, were examined in vitro to detect production of chitinases, β-1,3-glucanases, and cellulases. All the species produced chitinases, but not β-1,3-glucanases or cellulases, when grown on a liquid mineral medium containing glucose, colloidal chitin, or cell walls of M. perniciosa as a carbon source. There were no quantitative differences among species in the production of chitinase, however, the germination inhibition of basidiospores of M. perniciosa was higher when they were cultivated using glucose as a carbon source, followed by colloidal chitin and cell walls. All the species also produced hydroxymate type siderophores in similar quantities, and the quantity of siderophores did not correlate with the inhibition of basidiospore germination. The germination inhibition was more pronounced when S. albovinaceus, S. griseus, and S. virginiae were cultivated on iron-deficient medium, suggesting involvement of siderophores in the antagonism by these species of actinomycetes.  相似文献

Basidiomycetes produce large quantities of spores that are dispersed by the wind over long distances. Some species can release billions of spores in a day, many of which are important aeroallergens associated with asthma and allergic rhinitis. The results of a survey carried out in the city of Seville during two consecutive years, using a Hirst-type spore trap, showed that airborne basidiospores are present throughout the year, although there is a certain seasonal distribution, with the highest average concentration occurring in November (23,586 basidiospores). Eighteen different types of basidiospores were identified, with Coprinus and Ustilago being the most frequent, followed in descending order of abundance by Agaricus, Phylacteria, Boletaceae, Ganoderma, Cortinarius, Calvatia, Agrocybe, Bovista and Puccinia; the remaining spore types comprised less than 1% of the total basidiospore count.  相似文献

Seven species of the genus Cosmospora collected in Japan are reported in this article. Among them, Cosmospora japonica is described as a new species. Cosmospora henanensis, C. rishbethii, and C. triqua, all of which are known only from their type localities, are added to the Japanese mycobiota. The other species, C. chaetopsinaecatenulatae, C. diminuta, and C. peponum, are new records for Japan. Additional distribution records are given for Cosmospora species hitherto known in Japan.  相似文献

A new corticolous species of Byssoloma, B. rubromarginatum Messuti and de la Rosa from Nothofagus dombeyi forests is described and illustrated. The new species is characterized by the morphology of the thallus and apothecia, the 3–5 (7)-septate ascospores, the presence of an anthraquinone in the exciple, and its ecology. It is the first species within the genus known to have gyrophoric acid in the apothecial margin. A key to Byssoloma species known from Nothofagus temperate forests in South America is provided.  相似文献

The new species is similar to S. grandifolius, S. scorpioides, and S. umbellatus, and morphological differences among the taxa are discussed.
