ABSTRACT. To determine whether the production of sexuals in the fire ant, Solenopsis invicta Buren, is related to queen number in the field, the numbers of sexuals in monogyne (M) and polygyne (P) colonies of this species were compared. Large colonies ( n =25 M and 25 P) were collected at random during spring, summer and fall when sexuals are actively produced, and the numbers of sexuals present were counted. The numbers of alates plus sexual pupae and larvae in M colonies far exceeded those in P colonies in each season. Significant differences between M and P colonies were also found when the numbers of alates and sexual immatures (pupae + larvae) were compared separately in each season. In addition, M colonies contained significantly more female sexuals (alates + pupae) than did P colonies in the summer and fall, and significantly more males in all three seasons. The negative relationship between queen number and number of sexuals provides evidence that queen control over the production of sexuals, previously established in laboratory experiments, also occurs under natural conditions.  相似文献

Red imported fire ant, Solenopsis invicta, colonies were infected horizontally by introducing live brood (mainly larvae and pupae) infected with Thelohania solenopsae. Live, infected brood introduced into uninfected colonies were adopted and raised to adulthood instead of being executed by the recipient colony. Introductions of infected larvae with uninfected pupae, which eclose into adult worker caste fire ants, resulted in an 80% infection rate of the inoculated colonies. Infections from introductions of infected pupae with uninfected larvae resulted in a 37.5% infection of inoculated colonies. Infections were also detected in 11.6 and 3.7% of the adult worker caste ants that eclosed from uninfected large larvae and pupae, respectively, that were held with infected adult workers. Microscopic examination of infected brood revealed sporoblasts and large numbers of spores of T. solenopsae in S. invicta pupae.  相似文献

The fire ant Solenopsis invicta and its close relatives display an important social polymorphism involving differences in colony queen number. Colonies are headed by either a single reproductive queen (monogyne form) or multiple queens (polygyne form). This variation in social organization is associated with variation at the gene Gp-9, with monogyne colonies harboring only B-like allelic variants and polygyne colonies always containing b-like variants as well. We describe naturally occurring variation at Gp-9 in fire ants based on 185 full-length sequences, 136 of which were obtained from S. invicta collected over much of its native range. While there is little overall differentiation between most of the numerous alleles observed, a surprising amount is found in the coding regions of the gene, with such substitutions usually causing amino acid replacements. This elevated coding-region variation may result from a lack of negative selection acting to constrain amino acid replacements over much of the protein, different mutation rates or biases in coding and non-coding sequences, negative selection acting with greater strength on non-coding than coding regions, and/or positive selection acting on the protein. Formal selection analyses provide evidence that the latter force played an important role in the basal b-like lineages coincident with the emergence of polygyny. While our data set reveals considerable paraphyly and polyphyly of S. invicta sequences with respect to those of other fire ant species, the b-like alleles of the socially polymorphic species are monophyletic. An expanded analysis of colonies containing alleles of this clade confirmed the invariant link between their presence and expression of polygyny. Finally, our discovery of several unique alleles bearing various combinations of b-like and B-like codons allows us to conclude that no single b-like residue is completely predictive of polygyne behavior and, thus, potentially causally involved in its expression. Summary We examined the relationship between queen number and worker size in colonies of the fire antSolenopsis invicta. Worker size in monogyne colonies was significantly greater than in polygyne colonies; furthermore, polygyne colonies snowed a strong negative linear relationship between queen number and worker size. Higher queen pheromone level and/or decreased food availability accompanying an increase in queen number likely play important roles in producing the observed patterns.  相似文献

Abstract.— Recent studies of the introduced fire ant Solenopsis invicta suggest that introduced polygyne (with multiple queens per nest) populations are strongly influenced by male-mediated gene flow from neighboring monogyne (single queen per nest) populations and selection acting on a single locus, general protein-9 (Gp-9) . This investigation formally tests this hypothesis and determines if these processes can account for the genotypic structure of polygyne S. invicta . To increase the statistical power of this test, we considered the genotypes of polygyne queens and workers at both Gp-9 and the closely linked, selectively neutral locus Pgm-3 . We then constructed and analyzed a novel mathematical model to delimit the effects of monogyne male gene flow and selection on the joint genotypes at the Pgm-3/Gp-9 superlocus. Using this framework, a hierarchical maximum-likelihood method was developed to estimate the best-fitting gene flow and selection parameters based on the fit of our model to data from both the current study and an earlier one of the same population. In each case, selection on polygyne queens and workers alone, with no monogyne male gene flow, provides the most parsimonious explanation for the observed genotype frequencies. The apparent discrepancy between this result and the empirical evidence for monogyne male gene flow indicates that undocumented factors, such as other forms of selection in polygyne males or workers, are operating in introduced polygyne S. invicta .  相似文献

Thelohania solenopsae is a pathogen of the red imported fire ant, Solenopsis invicta, which debilitates queens and eventually causes the demise of colonies. Reductions of infected field populations signify its potential usefulness as a biological control agent. Thelohania solenopsae can be transmitted by introducing infected brood into a colony. The social forms of the fire ant, that is, monogyny (single queen per colony) or polygyny (multiple queens per colony), are associated with different behaviors, such as territoriality, that affect the degree of intercolony brood transfer. T. solenopsae was found exclusively in polygyne colonies in Florida. Non-synchronous infections of queens and transovarial transmission favor the persistence and probability of detecting infections in polygynous colonies. However, queens or alates with the monogyne genotype can be infected, and infections in monogyne field colonies have been reported from Louisiana and Argentina. Limited independent colony-founding capability and shorter dispersal of alate queens with the polygyne genotype relative to monogyne alates may facilitate the maintenance of infections in local polygynous populations. Demise of infected monogyne colonies can be twice as fast as in polygyne colonies and favors the pathogen's persistence in polygyne fire ant populations. The social form of the fire ant reflects different physiological and behavioral aspects of the queen and colony that will impact T. solenopsae spread and ultimate usefulness for biological control.  相似文献

Little is known about the genetic foundations of colony social organization. One rare example in which a single major gene is implicated in the expression of alternative social organizations involves the presumed odorant-binding protein gene Gp-9 in fire ants. Specific amino acid substitutions in this gene invariably are associated with the expression of monogyny (single queen per colony) or polygyny (multiple queens per colony) in fire ant species of the Solenopsis richteri clade. These substitutions are hypothesized to alter the abilities of workers to recognize queens and thereby regulate their numbers in a colony. We examined whether these same substitutions underlie the monogyny/polygyny social polymorphism in the distantly related fire ant S. geminata. We found that Gp-9 coding region sequences are identical in the polygyne and monogyne forms of this species, disproving our hypothesis that one or a few specific amino acid replacements in the protein are necessary to induce transitions in social organization in fire ants. On the other hand, polygyne S. geminata differs genetically from the monogyne form in ways not mirrored in the two forms of S. invicta, a well-studied member of the S. richteri clade, supporting the conclusion that polygyny did not evolve via analogous routes in the two lineages. Specifically, polygyne S. geminata has lower genetic diversity and different gene frequencies than the monogyne form, suggesting that the polygyne form originated via a founder event from a local monogyne population. These comparative data suggest an alternative route to polygyny in S. geminata in which loss of allelic variation at genes encoding recognition cues has led to a breakdown in discrimination abilities and the consequent acceptance of multiple queens in colonies.  相似文献

Two cytochrome P450 genes, CYP4AB1 and CYP4AB2, and the Gp-9 gene were identified as being specifically overexpressed in workers of the red imported fire ant using PCR-selected subtractive hybridization and cDNA array techniques. Full-length CYP4AB1 and CYP4AB2 were cloned and sequenced. The cDNA sequences of CYP4AB1 and CYP4AB2 have open reading frames of 1389 and 1533 nucleotides encoding proteins of 463 and 511 amino acid residues, respectively. Northern blot analysis was performed to compare expression levels of CYP4AB1, CYP4AB2, and Gp-9 for different developmental stages and castes of fire ants. We demonstrate that the expression of these three genes is developmentally and caste specifically regulated in red imported fire ants. Levels of CYP4AB1 mRNA were undetectable in 3rd+4th instars, worker pupae, and alate (mixed sex) pupae; readily detectable in male and female alates; increased (approximately 3-fold) in the queens; and rose to a maximum (13-fold) in workers. Similarly, the expression of CYP4AB2 mRNA was undetectable in 3rd+4th instars, worker pupae, and alate pupae; low in male and female alates and queens; and increased (approximately 7-fold) in workers. Levels of Gp-9 mRNA were readily detectable in male alates; increased (approximately 3-fold) in female alates; and reached a maximum (approximately 12-fold) in workers. Their caste-specific overexpression suggests the functional importance of CYP4AB1, CYP4AB2, and Gp-9 in workers of the red imported fire ant.  相似文献

The fire ant Solenopsis invicta exists in two social forms, one with colonies headed by a single reproductive queen (monogyne form) and the other with colonies containing multiple queens (polygyne form). This variation in social organization is associated with variation at the gene Gp-9, with monogyne colonies harboring only the B allelic variant and polygyne colonies containing b-like variants as well. We generated new Gp-9 sequences from 15 Solenopsis species and combined these with previously published sequences to conduct a comprehensive, phylogenetically based study of the molecular evolution of this important gene. The exon/intron structure and the respective lengths of the five exons of Gp-9 are identical across all species examined, and we detected no evidence for intragenic recombination. These data conform to a previous suggestion that Gp-9 lies in a genomic region with low recombination, and they indicate that evolution of the coding region in Solenopsis has involved point substitutions only. Our results confirm a link between the presence of b-like alleles and the expression of polygyny in all South American fire ant species known to possess colonies of both social forms. Moreover, phylogenetic analyses show that b-like alleles comprise a derived clade of Gp-9 sequences within the socially polymorphic species, lending further support to the hypothesis that monogyny preceded polygyny in this group of fire ants. Site-specific maximum likelihood tests identified several amino acids that have experienced positive selection, two of which are adjacent to the inferred binding-pocket residues in the GP-9 protein. Four other binding-pocket residues are variable among fire ant species, although selection is not implicated in this variation. Branch-specific tests revealed strong positive selection on the stem lineage of the b-like allele clade, as expected if selection drove the amino acid replacements crucial to the expression of polygyne social organization. Such selection may have operated via the ligand-binding properties of GP-9, as one of the two amino acids uniquely shared by all b-like alleles is predicted to be a binding-pocket residue.  相似文献

We describe genetic structure at various scales in native populations of the fire ant Solenopsis invicta using two classes of nuclear markers, allozymes and microsatellites, and markers of the mitochondrial genome. Strong structure was found at the nest level in both the monogyne (single queen) and polygyne (multiple queen) social forms using allozymes. Weak but significant microgeographic structure was detected above the nest level in polygyne populations but not in monogyne populations using both classes of nuclear markers. Pronounced mitochondrial DNA (mtDNA) differentiation was evident also at this level in the polygyne form only. These microgeographic patterns are expected because polygyny in ants is associated with restricted local gene flow due mainly to limited vagility of queens. Weak but significant nuclear differentiation was detected between sympatric social forms, and strong mtDNA differentiation also was found at this level. Thus, queens of each form seem unable to establish themselves in nests of the alternate type, and some degree of assortative mating by form may exist as well. Strong differentiation was found between the two study regions using all three sets of markers. Phylogeographic analyses of the mtDNA suggest that recent limitations on gene flow rather than longstanding barriers to dispersal are responsible for this large-scale structure.  相似文献

Eight laboratory-reared ant species were fed baits of house fly, Musca domestica L., pupae treated with hydramethylnon. Two fire ant species, Solenopsis invicta Buren and Solenopsis geminata (F.), and Pheidole morrissi (Forel) were killed; average percentage of mortality of the five other species was less than 20%. In contrast, all species that were fed the commercial fire ant bait containing hydramethylnon (Amdro) died or were adversely affected. In the field, applications of house fly pupae and eye gnat, Hippelates pusio Loew, pupae dipped in acetone solutions of fenoxycarb significantly reduced population indices of the red imported fire ant, S. invicta, compared with commercial formulations of fenoxycarb (Logic) and hydramethylnon (Amdro). Field observations showed that the pupae of either species can be carried or moved by one or two worker ants. The smooth, hard cuticle of the pupae make them easy to handle and apply with application equipment. The current cost of house fly pupae is considerably more than the cost of a granular carrier, pregel defatted corn grits. However, if mass-production methods reduce this price differential, fly pupae could become an effective and more species-specific fire ant bait carrier.  相似文献

A series of oligonucleotide primer pairs covering the entire genome of Solenopsis invicta virus 1 (SINV-1) were used to probe the genome of its host, S. invicta, for integrated fragments of the viral genome. All of the oligonucleotide primer sets yielded amplicons of anticipated size from cDNA created from an RNA template from SINV-1. However, no corresponding amplification was observed when genomic DNA (from 32 colonies of S. invicta) was used as template for the PCR amplifications. Host DNA integrity was verified by amplification of an ant-specific gene, SiGSTS1. The representation of fire ant colonies included both social forms, monogyne and polygyne, and those infected and uninfected with SINV-1. Furthermore, no amplification was observed from genomic DNA from ant samples collected from Argentina or the US. Thus, it appears that SINV-1 genome integration, or a portion therein, has not likely occurred within the S. invicta host genome.  相似文献

了解我国红火蚁Solenopsis invicta Buren的耐低温能力并为其风险分析提供科学依据,本文对采自我国4个地点红火蚁蚁群中不同等级和虫态的过冷却点,以及不同季节和室内不同饥饿程度的工蚁过冷却点进行了测定。结果表明:蚁群中不同等级和虫态的过冷却点差异显著。工蚁成虫的过冷却点(-11.8±0.4℃)显著低于有翅雌蚁(-8.1±0.6℃)及蚁后(-7.9±0.5℃),工蚁蛹和幼虫的过冷却点分别为-13.6±0.3℃和-12.1±0.4℃,蚁后蛹和幼虫的分别为-13.3±0.3℃和-12.2±0.3℃。工蚁蛹的过冷却点均显著低于其成虫和幼虫。蚁后蛹与其幼虫的过冷却点没有显著差异,但与其成虫的差异显著。同一未成熟期不同等级之间的也没有显著差异;不同季节的工蚁过冷却点有显著差异,其中以春季的最高(-4.3±0.2℃),夏季(-6.6±0.2℃)、秋季(-9.4±0.1℃)的次之,冬季的(-12.0±0.3℃)最低;深圳、珠海、广州和陆川4个地理种群的工蚁过冷却点在春、秋和冬季无显著差异,但夏季陆川种群的过冷却点显著低于其他3个种群的(P<0.05)。饥饿可使工蚁的过冷却点明显升高,但大工蚁的过冷却点较小工蚁升高的幅度更大,饥饿15 d后的大工蚁比饥饿前的过冷却点升高了7.0℃,而小工蚁仅升高了4.9℃。这些结果为评价红火蚁在我国潜在的传播风险提供了理论依据。  相似文献

A recent study is the first to sequence a gene known to underlie a complex social phenotype. In the fire ant, Solenopsis invicta, a single allelic difference at the Gp-9 locus specifies the number of queens a colony has, and hence the social structure of the colony. Gp-9 appears to encode a protein implicated in chemical recognition of nestmates, consistent with workers determining queen number by selectively executing queens as a function of workers' and queens' Gp-9 genotypes. Other Solenopsis species exhibit the same social and genetic polymorphism. This study pioneers the integrated understanding of the evolution of social behaviour at molecular, individual and social levels.  相似文献

【目的】本研究旨在分析红火蚁Solenopsis invicta毒腺细菌群落多样性,并与热带火蚁Solenopsis geminata和聚纹双刺猛蚁Diacamma rugosum比较毒腺细菌群落差异。【方法】采用Illumine Hiseq 2500测序平台对红火蚁(工蚁、有翅蚁和蚁后)、热带火蚁(工蚁)及聚纹双刺猛蚁(工蚁)毒腺细菌群落16S rRNA基因V3-V4区测序,基于测序数据进行生物信息学分析。【结果】变形菌门(Proteobacteria)在红火蚁工蚁、有翅蚁和蚁后以及热带火蚁工蚁毒腺中占优势,而厚壁菌门(Firmicutes)在聚纹双刺猛蚁工蚁毒腺中占优势。与红火蚁工蚁和有翅蚁相比,柔壁菌门(Tenericutes)在红火蚁蚁后毒腺中更丰富。广州红火蚁蚁后毒腺中假单胞杆菌Pseudomonas相对丰度显著高于其在有翅蚁及工蚁中的。螺原体Spiroplasma相对丰度在热带火蚁毒腺中显著高于在聚纹双刺猛蚁工蚁毒腺中的。毒腺中细菌多样性分析发现,芽孢杆菌属Bacillus和乳酸杆菌属Lactobacillus在广西红火蚁工蚁中的相对丰度显著高于在广州红火蚁工蚁中的。然而,乳酸杆菌属细菌在聚纹双刺猛蚁工蚁毒腺中的相对丰度显著高于广西的热带火蚁工蚁毒腺中的。【结论】毒腺细菌组成和多样性在3种不同种类蚂蚁工蚁和红火蚁不同品级中存在差异。  相似文献

Red imported fire ant, Solenopsis invicta, colonies were successfully infected with the microsporidium Vairimorpha invictae by introducing live larvae, pupae, or dead adults from V. invictae-infected field colonies collected in Argentina. Introductions with 4th instar larvae or non-melanized pupae obtained from infected field colonies, resulted in infection of 40% of the inoculated colonies. Introductions of 4th instars or melanized pupae produced from colonies that were initially infected in the laboratory, resulted in infections of 83% of the colonies, thus perpetuating the infection in other colonies. Infection was detected in 2 of 6 colonies after introducing adult worker caste ants that had died with V. invictae. The average number of adults and the volume of immature ants per colony were significantly lower in the infected than in the control colonies. Infected colonies had 86% fewer adults per colony and 82% less immature ants than the controls. A portion of the 16S rRNA gene of the V. invictae identified from these studies was amplified, cloned, and sequenced; the 1251 nucleotide amplicon was 100% identical to the 16S rRNA gene sequence recorded previously in the GenBank database, thus verifying the species as V. invictae. This is the first report of the artificial transmission of this pathogen to uninfected ant colonies, and demonstration of its ability to hinder growth in individual colonies.  相似文献

Understanding how exotic invasive species are spread is fundamental for ecology and conservation biology. Human transport has become one of the primary modes of dispersal for exotic species. We examined how the multiple queen, or polygyne, social form of the fire ant Solenopsis invicta is spread along roadsides in Tallahassee, Florida, USA. We then determined the likely source of this expanding population, which was a central soil depot. A survey of road maintenance practices in counties of several southeastern states and Texas revealed that the use of a central soil depot is a common practice. Road maintenance therefore may be the primary source for the establishment of new polygyne fire ant populations in this region and elsewhere. Control efforts focused on the soil depots will help to limit further spread of polygyne fire ants and perhaps other invasive organisms, particularly invasive weeds.  相似文献

Oligonucleotide primer pairs were designed to unique areas of the small subunit (16S) rRNA gene of Thelohania solenopsae and a region of the Gp-9 gene of Solenopsis invicta. Multiplex PCR resulted in sensitive and specific detection of T. solenopsae infection of S. invicta. The T. solenopsae-specific primer pair only amplified DNA from T. solenopsae and T. solenopsae-infected S. invicta. This primer pair did not produce any amplification products from DNA preparations from uninfected S. invicta, seven additional species of microsporidia (including Vairimorpha invictae), or Mattesia spp. The Gp-9-specific primers recognized and amplified DNA from Solenopsis xyloni, Solenopsis richteri, Solenopsis geminata, the invicta/richteri hybrid, and monogyne and polygyne S. invicta, but not from T. solenopsae, and, as such, served as a positive control verifying successful DNA preparation. Multiplex PCR detected T. solenopsae in worker fire ants infected with as few as 5000 spores. Furthermore, multiplex PCR detected T. solenopsae in all developmental stages of S. invicta. However, detection could be made more sensitive by using only the T. solenopsae-specific primer pair; ants infected with as few as 10 spores were able to be discerned. Multiplex PCR detection of T. solenopsae offers the advantages of a positive control, a single PCR amplification, detection of all developmental stages, and increased sensitivity and specificity compared with microscopy.  相似文献
