Distinguishing kin from non-kin profoundly impacts the evolution of social behaviour. Individuals able to assess the genetic relatedness of conspecifics can preferentially allocate resources towards related individuals and avoid inbreeding. We have addressed the question of how animals acquire the ability to recognize kin by studying the development of olfactory kin preference in zebrafish (Danio rerio). Previously, we showed that zebrafish use an olfactory template to recognize even unfamiliar kin through phenotype matching. Here, we show for the first time that this phenotype matching is based on a learned olfactory imprinting process in which exposure to kin individuals on day 6 post fertilization (pf) is necessary and sufficient for imprinting. Larvae that were exposed to kin before or after but not on day 6 pf did not recognize kin. Larvae isolated from all contact with conspecifics did not imprint on their own chemical cues; therefore, we see no evidence for kin recognition through self-matching in this species. Surprisingly, exposure to non-kin odour during the sensitive phase of development did not result in imprinting on the odour cues of unrelated individuals, suggesting a genetic predisposition to kin odour. Urine-born peptides expressed by genes of the immune system (MHC) are important messengers carrying information about 'self' and 'other'. We suggest that phenotype matching is acquired through a time-sensitive learning process that, in zebrafish, includes a genetic predisposition potentially involving MHC genes expressed in the olfactory receptor neurons.

Genomic imprinting corresponds to the differential expression of a gene according to its paternal or maternal origin. The kinship theory of genomic imprinting proposes that maternally or paternally inherited genes may be in conflict over their effects on kin differently related along the paternal or maternal line. Most examples supporting the kinship theory of imprinting deal with competition between offspring for maternal resources. However, genomic imprinting may also explain differential behavioral expression toward kin whenever sibs are more related to each other via one parental sex than the other. Unfortunately, nothing is currently known about imprinting associated with a behavioral phenotype in insects. Here we report the first evidence of such a maternally imprinted behavior. We show that the solitary parasitoid larvae of Aleochara bilineata Gyll (Coleoptera; Staphylinidae), which avoid superparasitizing their full sibs, also avoid their cousins when they are related to them through their father, but not when they are related to them through their mother. A genetic kin recognition mechanism is proposed to explain this result and we conclude that genomic imprinting could control the avoidance of kin superparasitism in this species and have a profound influence on decision-making processes.

House mice (Mus musculus domesticus) avoid mating with individuals that are genetically similar at the major histocompatibility complex (MHC). Mice are able recognize MHC-similar individuals through specific odour cues. However, to mate disassortatively for MHC genes, individuals must have a referent, either themselves (self-inspection) or close kin (familial imprinting), with which to compare the MHC identity of potential mates. Although studies on MHC-dependent mating preferences often assume that individuals use self-inspection, laboratory experiments with male mice indicate that they use familial imprinting, i.e. males learn the MHC identity of their family and then avoid mating with females carrying ''familial'' MHC alleles. To determine if female mice use familial imprinting, we cross-fostered wild-derived female mouse pups into MHC-dissimilar families, and then tested if this procedure reversed their mating preferences compared with in-fostered controls. Our observations of the female''s mating behaviour in seminatural social conditions and the genetic typing of their progeny both indicated that females avoided mating with males carrying MHC genes of their foster family, supporting the familial imprinting hypothesis. We show that MHC-dependent familial imprinting potentially provides a more effective mechanism for avoiding kin matings and reducing inbreeding than self-inspection.

When facing decisions about where to live, juveniles have a strong tendency to choose habitats similar to where their parents successfully bred. Developing larval fishes can imprint on the chemical cues from their natal habitat. However, to demonstrate that imprinting is ecologically important, it must be shown that settlers respond and distinguish among different imprinted cues, and use imprinting for decisions in natural environments. In addition, the potential role innate preferences play compared to imprinted choices also needs to be examined. As environmental variability increases due to anthropogenic causes these two recognition mechanisms, innate and imprinting, could provide conflicting information. Here we used laboratory rearing and chemical choice experiments to test imprinting in larval anemonefish (Amphiprion percula). Individuals exposed to a variety of benthic habitat or novel olfactory cues as larvae either developed a preference for (spent >50 % of their time in the cue) or increased their attraction to (increased preference but did not spend >50 % of their time in the cue) the cue when re-exposed as settlers. Results indicate not only the capacity for imprinting but also the ability to adjust innate preferences after early exposure to a chemical cue. To test ecological relevance in the natural system, recruits were collected from anemones and related to their parents, using genetic parentage analysis, providing information on the natal anemone species and the species chosen at settlement. Results demonstrated that recruits did not preferentially return to their natal species, conflicting with laboratory results indicating the importance imprinting might have in habitat recognition.

The ability to recognize close relatives in order to cooperate or to avoid inbreeding is widespread across all taxa. One accepted mechanism for kin recognition in birds is associative learning of visual or acoustic cues. However, how could individuals ever learn to recognize unfamiliar kin? Here, we provide the first evidence for a novel mechanism of kin recognition in birds. Zebra finch (Taeniopygia guttata) fledglings are able to distinguish between kin and non-kin based on olfactory cues alone. Since olfactory cues are likely to be genetically based, this finding establishes a neglected mechanism of kin recognition in birds, particularly in songbirds, with potentially far-reaching consequences for both kin selection and inbreeding avoidance.

Kin recognition in Bufo scaber tadpoles: ontogenetic changes and mechanism
Ontogenetic changes in kin-recognition behavior, effect of social environment on kin-recognition ability, and use of visual and chemical cues in kin recognition have been studied in tadpoles of Bufo scaber after rearing them with kin, in mixed groups, or in isolation from Gosner stage 12 (gastrula). By use of a rectangular choice tank the tadpoles were tested for their ability to choose between (a) familiar siblings and unfamiliar non-siblings, (b) unfamiliar siblings and familiar non-siblings, and (c) unfamiliar siblings and unfamiliar non-siblings. When tested without any stimulus groups in the end compartments of the tank, random distribution was observed for the tadpoles and no bias for the apparatus or the procedure. In the presence of kin and non-kin in the end compartments, significantly more tadpoles spent most of their time near kin (familiar or unfamiliar) rather than near non-kin during early larval stages, up to stage 37. After stage 37 (characterized by the differentiation of toes), test tadpoles showed no preference to associate with kin, suggesting an ontogenetic shift in the kin-recognition ability in B. scaber. In experiments involving selective blockade of visual or chemical cues the test tadpoles preferentially associated near their kin on the basis of chemical rather than visual cues. These findings suggest that familiarity with siblings is not necessary for kin recognition and that kin-recognition ability is not modified after exposure to non-kin by mixed rearing. The findings for B. scaber indicate a self referent phenotype matching mechanism of kin recognition which is predominantly aided by chemical rather than visual cues.

Perfluorooctane sulfonate (PFOS) is a widely spread environmental contaminant. It accumulates in the brain and has potential neurotoxic effects. The exposure to PFOS has been associated with higher impulsivity and increased ADHD prevalence. We investigated the effects of developmental exposure to PFOS in zebrafish larvae, focusing on the modulation of activity by the dopaminergic system. We exposed zebrafish embryos to 0.1 or 1 mg/L PFOS (0.186 or 1.858 µM, respectively) and assessed swimming activity at 6 dpf. We analyzed the structure of spontaneous activity, the hyperactivity and the habituation during a brief dark period (visual motor response), and the vibrational startle response. The findings in zebrafish larvae were compared with historical data from 3 months old male mice exposed to 0.3 or 3 mg/kg/day PFOS throughout gestation. Finally, we investigated the effects of dexamfetamine on the alterations in spontaneous activity and startle response in zebrafish larvae. We found that zebrafish larvae exposed to 0.1 mg/L PFOS habituate faster than controls during a dark pulse, while the larvae exposed to 1 mg/L PFOS display a disorganized pattern of spontaneous activity and persistent hyperactivity. Similarly, mice exposed to 0.3 mg/kg/day PFOS habituated faster than controls to a new environment, while mice exposed to 3 mg/kg/day PFOS displayed more intense and disorganized spontaneous activity. Dexamfetamine partly corrected the hyperactive phenotype in zebrafish larvae. In conclusion, developmental exposure to PFOS in zebrafish induces spontaneous hyperactivity mediated by a dopaminergic deficit, which can be partially reversed by dexamfetamine in zebrafish larvae.

The genetic basis of inbreeding avoidance in house mice
Animals might be able to use highly polymorphic genetic markers to recognize very close relatives and avoid inbreeding. The major histocompatibility complex (MHC) is thought to provide such a marker because it influences individual scent in a broad range of vertebrates. However, direct evidence is very limited. In house mice (Mus musculus domesticus), the major urinary protein (MUP) gene cluster provides another highly polymorphic scent signal of genetic identity that could underlie kin recognition. We demonstrate that wild mice breeding freely in seminatural enclosures show no avoidance of mates with the same MHC genotype when genome-wide similarity is controlled. Instead, inbreeding avoidance is fully explained by a strong deficit in successful matings between mice sharing both MUP haplotypes. Single haplotype sharing is not a good guide to the identification of full sibs, and there was no evidence of behavioral imprinting on maternal MHC or MUP haplotypes. This study, the first to examine wild animals with normal variation in MHC, MUP, and genetic background, demonstrates that mice use self-referent matching of a species-specific polymorphic signal to avoid inbreeding. Recognition of close kin as unsuitable mates might be more variable across species than a generic vertebrate-wide ability to avoid inbreeding based on MHC.

Genes of the major histocompatibility complex (MHC) that underlie the adaptive immune system may allow vertebrates to recognize their kin. True kin-recognition genes should produce signalling products to which organisms can respond. Allelic variation in the peptide-binding region (PBR) of MHC molecules determines the pool of peptides that can be presented to trigger an immune response. To examine whether these MHC peptides also might underlie assessments of genetic similarity, we tested whether Xenopus laevis tadpoles socially discriminate between pairs of siblings with which they differed in PBR amino acid sequences. We found that tadpoles (four sibships, n = 854) associated preferentially with siblings with which they were more similar in PBR amino acid sequence. Moreover, the strength of their preference for a conspecific was directly proportional to the sequence similarity between them. Discrimination was graded, and correlated more closely with functional sequence differences encoded by MHC class I and class II alleles than with numbers of shared haplotypes. Our results thus suggest that haplotype analyses may fail to reveal fine-scale behavioural responses to divergence in functionally expressed sequences. We conclude that MHC–PBR gene products mediate quantitative social assessment of immunogenetic similarity that may facilitate kin recognition in vertebrates.

The ability to recognise kin requires the individual to possess a variety of abilities. Individuals must produce a cue which indicates relatedness, they must process this cue to determine relatedness and then must act on this cue. Research has concentrated on the first and final stage of this process, i.e., the cues of kinship and kin correlated behaviour. Little attention has been paid to how individuals process cues to determine relatedness. This paper discusses how individuals ‘recognise’ kin, the exhibition of kin correlated behaviour and considers the role of the MHC in these processes. Two broad theories have emerged to explain how individuals recognise their kin: either a recognition gene(s) or some experiential mechanism. In mammals there is no evidence to suggest that recognition (the processing of the cue) is under genetic control but rather is the result of experience, learning about related individuals during development. Moreover studies on kin recognition in the domestic dog suggest that all kin are not recognised by the same process but different classes of kin, parents, siblings may well be recognised using different means. Studies of kin correlated behaviour suggest that the behaviour exhibited towards kin by Mongolian gerbils is mediated by the environment. Factors of environmental familiarity, sex and developmental age all affect the response of individuals to kin and non‐kin. In these situations the ability to recognise kin does not change but the exhibition of kin correlated behaviour changes according to environmental conditions. These studies indicate that kin recognition may not be the ‘unified’ process previously thought and thus any explanations of the proximate and ultimate causation of kin recognition need to encompass this complexity. The question remains of whether the MHC is complex enough to do so. This revised version was published online in July 2006 with corrections to the Cover Date.

Kin recognition, the ability to detect relatives, is important for cooperation, altruism and also inbreeding avoidance. A large body of research on kin recognition mechanisms exists for vertebrates and insects, while little is known for other arthropod taxa. In spiders, nepotism has been reported in social and solitary species. However, there are very few examples of kin discrimination in a mating context, one coming from the orb-weaver Argiope bruennichi. Owing to effective mating plugs and high rates of sexual cannibalism, both sexes of A. bruennichi are limited to a maximum of two copulations. Males surviving their first copulation can either re-mate with the current female (monopolizing paternity) or leave and search for another. Mating experiments have shown that males readily mate with sisters but are more likely to leave after one short copulation as compared with unrelated females, allowing them to search for another mate. Here, we ask whether the observed behaviour is based on chemical cues. We detected family-specific cuticular profiles that qualify as kin recognition cues. Moreover, correlations in the relative amounts of some of the detected substances between sexes within families indicate that kin recognition is likely based on subsets of cuticular substances, rather than entire profiles.

Locomotor behaviors were examined in two experiments using zebrafish (Danio rerio) larvae at 4, 5, 6 and 7 days post fertilization (dpf). Larvae were observed in individual wells of a 12-well plate for 1 h a day. In Experiment 1, the same larvae were observed for four consecutive days beginning on post-fertilization day 4; in Experiment 2, different groups of larvae from the same egg collection were observed at 4, 5, 6 and 7 dpf. Automated images collected every 6 s were analyzed for information about larval location, orientation and general activity. In both experiments, 4 dpf larvae rested significantly more, used a smaller area of the well more frequently, and were generally less active than older larvae. All larvae exhibited a preference for facing away from the center of the well and for the edge of the well. However, prolonged exposure to the well influenced overall activity, orientation, and preference for the edge region. The implications of these results for understanding the development of larval behavior and for the design of procedures to measure the effects of experience in zebrafish larvae are discussed.

In experiments with specially designed choice tanks, tadpoles of Bufo melanostictus spend significantly greater amounts of time near kin than near non-kin. However, in the absence of kin members, they prefer to spend more time near non-kin rather than stay away in isolation in the opposite blank zone with no company. This implies that association of toad tadpoles with their kin is due to attraction rather than repulsion from non-kin. Experiments designed to elucidate the sensory basis of kin recognition showed that toad tadpoles recognize their kin based on chemical cues rather than visual cues. They can also discriminate between homospecific non-kin and heterospecific (Sphaerotheca breviceps) tadpoles since the tadpoles spent significantly greater amounts of time near the former than near the latter. These findings suggest that where kin members are unavailable, selection may have favoured living with non-kin so as to derive benefits from group living and that a phenotype-matching mechanism may operate for both kin and species discrimination in B. melanostictus.

KIN RECOGNITION: FUNCTIONS AND MECHANISMS A REVIEW
1. The aim of this paper has been to review the theory behind kin recognition to examine the benefits individuals obtain by recognizing their kin and to review the mechanisms used by individuals in their recognition of kin. 2. The ability to discriminate between kin and non-kin, and between different classes of kin gives individuals advantages in fitness greater than individuals unable to recognize their kin. Four specific areas of benefit were considered: altruistic behaviour, co-operative behaviour, parental care and mate choice. Finally the possibility that kin recognition has arisen as a byproduct from some other ability was discussed. 3. Mechanisms of kin recognition were considered with respect to three essential components of kin recognition. The cue used to discriminate kin, how individuals classify conspecifics as kin, etc. and how the ability to recognize kin develops. 4. Individuals can use a number of cues to discriminate kin from non-kin. These were divided into cues presented by conspecifics (conspecific cues), of which three types were considered: individual, genetic and group/colony cues, and non-conspecific cues, environmental, state and no cues. Kin recognition could be achieved by use of all these cues. 5. How individuals classify their conspecifics as kin, etc. can be achieved in a number of ways; dishabituation or self-matching, which require no learning of kinship cues, or by phenotype matching or familiarity, both of which require the learning of kinship information. 6. It may be necessary for individuals to acquire information concerning kinship. This may be learned, and can be achieved in a number of ways; physiological imprinting, exposure learning or associative learning. Acquisition by these means is non-selective, in that the cues which are most salient in the individual's environment will be learned. Selectivity can be introduced into this process to increase the probability of acquiring kinship information by a number of means; learning from parents, sensitive periods for learning and prenatal learning. Finally, kinship information could be supplied by recognition genes. 7. A distinction is drawn between cues which are used by an individual in the discrimination of kin, discriminators, and cues which are used by individuals in the acquisition of information about kinship, acquisitors. 8. Experiments used to support previous categories of mechanisms of kin recognition were examined in the light of this discussion and it was found that the results were open to a number of different interpretations and yielded little specific information about the mechanisms of kin recognition. 9. It was concluded that there was much evidence, both theoretical and experimental to support the proposed benefits individuals gain from recognizing kin, but much more research is required before the mechanisms of kin recognition are fully understood.

Cannibalism is widespread among many different organisms, and can have both negative and positive fitness consequences. Avoiding eating relatives can minimize negative fitness consequences of cannibalism. Such avoidance requires kin discrimination, but evidence for this ability among cannibals is limited with little data that address the cues used in such discrimination. We examined whether larvae of the ladybird beetle Harmonia axyridis avoid eating their relatives. We further manipulated environmental factors to create individuals that had similar or dissimilar rearing environments to begin to test for endogenous versus exogenous recognition cues. In our experiments, third-instar larvae were much less likely to cannibalize if they were interacting with a relative. Larvae that did cannibalize kin required more encounters, and significantly delayed cannibalism, compared to larvae cannibalizing unrelated individuals. Acquired cues were less important. Even though the different rearing environment resulted in significantly different phenotypic effects, similarity or dissimilarity of rearing environment had no effect on cannibalism, and there was no interaction between environments and relatedness in cannibalism. We suggest that H. axyridis has a well-developed kin discrimination system, and that kin recognition in this ladybird beetle is based on endogenous rather than exogenous cues. We also argue that these cues reflect either direct or indirect genetic effects on larval phenotypes.
