Three hundred sixty healthy Ross×Ross 1-day-old broilers were used to study the effects of zinc glycine chelate (Zn-Gly) on growth performance, hematological, and immunological characteristics. All broilers were randomly assigned into six treatments. Diets were as follows: (1) control (containing 29.3 mg Zn kg−1 basic diet [0–3 weeks] and 27.8 mg Zn kg−1 [4–6 weeks]); (2) basic diet plus 30 mg Zn kg−1 from Zn-Gly; (3) basic diet plus 60 mg Zn kg−1 from Zn-Gly; (4) basic diet plus 90 mg Zn kg−1 from Zn-Gly; (5) basic diet plus 120 mg Zn kg−1 from Zn-Gly; (6) positive control, basic diet plus 120 mg Zn kg−1 from zinc sulfate (ZnSO4). After the 21- and 42-day feeding trials, the results showed that both of Zn-Gly and ZnSO4 could improve the growth performance of broilers, with the greatest average daily feed intake observed in the broilers fed 90 mg Zn kg−1 from Zn-Gly, but the greatest average daily gain observed with 120 mg Zn kg−1 from Zn-Gly (0–3 weeks) and 90 mg Zn kg−1 from Zn-Gly (4–6 weeks). Adding additional Zn-Gly improved the levels of immunoglobulins (IgA, IgM, and IgG) and the contents of total protein and Ca in serum and increased the immune organs index especially with 90 mg Zn kg−1 as Zn-Gly. However, there were no significant differences in responses to complements (C3 and C4) and albumin in serum among the treatments.  相似文献

β-Endorphin (BE) infusion at rest can influence insulin and glucagon levels and thus may affect glucose availability during exercise. To clarify the effect of BE on levels of insulin, glucagon and glucose during exercise, 72 untrained male Sprague-Dawley rats were infused i.v. with either: (1) BE (bolus 0.05 mg · kg−1 +0.05 mg · kg−1 · h−1, n = 24); (2) naloxone (N, bolus 0.8 mg · kg−1 + 0.4 mg · kg−1, n = 24); or (3) volume-matched saline (S, n = 24). Six rats from each group were killed after 0, 60, 90 or 120 min of running at 22 m · min−1, at 0% gradient. BE infusion resulted in higher plasma glucose levels at 60 min [5.93 (0.32) mM] and 90 min [4.16 (0.29) mM] of exercise compared to S [4.62 (0.27) and 3.41 (0.26 mM] and N [4.97 (0.38) and 3.44 (0.25) mM]. Insulin levels decreased to a greater extent with BE [21.5 (0.9) and 18.3 (0.6) uIU · ml−1] at 60 and 90 min compared to S [24.5 (0.5) and 20.6 (0.6) uIU · ml−1] and N [24.5 (0.4) and 21.6 (0.7) uIU · ml−1] groups. Plasma C-peptide declined to a greater extent at 60 and 90 min of exercise with BE infusion compared to both S and N. BE infusion increased glucagon at all times during exercise compared to S and N. These data suggest that BE infusion during exercise influences plasma glucose by augmenting glucagon levels and attenuating insulin release. Accepted: 26 February 1997  相似文献

Gross rates of N mineralization and nitrification, and soil–atmosphere fluxes of N2O, NO and NO2 were measured at differently grazed and ungrazed steppe grassland sites in the Xilin river catchment, Inner Mongolia, P. R. China, during the 2004 and 2005 growing season. The experimental sites were a plot ungrazed since 1979 (UG79), a plot ungrazed since 1999 (UG99), a plot moderately grazed in winter (WG), and an overgrazed plot (OG), all in close vicinity to each other. Gross rates of N mineralization and nitrification determined at in situ soil moisture and soil temperature conditions were in a range of 0.5–4.1 mg N kg−1 soil dry weight day−1. In 2005, gross N turnover rates were significantly higher at the UG79 plot than at the UG99 plot, which in turn had significantly higher gross N turnover rates than the WG and OG plots. The WG and the OG plot were not significantly different in gross ammonification and in gross nitrification rates. Site differences in SOC content, bulk density and texture could explain only less than 15% of the observed site differences in gross N turnover rates. N2O and NO x flux rates were very low during both growing seasons. No significant differences in N trace gas fluxes were found between plots. Mean values of N2O fluxes varied between 0.39 and 1.60 μg N2O-N m−2 h−1, equivalent to 0.03–0.14 kg N2O-N ha−1 y−1, and were considerably lower than previously reported for the same region. NO x flux rates ranged between 0.16 and 0.48 μg NO x -N m−2 h−1, equivalent to 0.01–0.04 kg NO x -N ha−1 y−1, respectively. N2O fluxes were significantly correlated with soil temperature and soil moisture. The correlations, however, explained only less than 20% of the flux variance.  相似文献

The objective of this study was to evaluate the effect of N fertilization and the presence of N2 fixing leguminous trees on soil fluxes of greenhouse gases. For a one year period, we measured soil fluxes of nitrous oxide (N2O), carbon dioxide (CO2) and methane (CH4), related soil parameters (temperature, water-filled pore space, mineral nitrogen content, N mineralization potential) and litterfall in two highly fertilized (250 kg N ha−1 year−1) coffee cultivation: a monoculture (CM) and a culture shaded by the N2 fixing legume species Inga densiflora (CIn). Nitrogen fertilizer addition significantly influenced N2O emissions with 84% of the annual N2O emitted during the post fertilization periods, and temporarily increased soil respiration and decreased CH4 uptakes. The higher annual N2O emissions from the shaded plantation (5.8 ± 0.3 kg N ha−1 year−1) when compared to that from the monoculture (4.3 ± 0.1 kg N ha−1 year−1) was related to the higher N input through litterfall (246 ± 16 kg N ha−1 year−1) and higher potential soil N mineralization rate (3.7 ± 0.2 mg N kg−1 d.w. d−1) in the shaded cultivation when compared to the monoculture (153 ± 6.8 kg N ha−1 year−1 and 2.2 ± 0.2 mg N kg−1 d.w. d−1). This confirms that the presence of N2 fixing shade trees can increase N2O emissions. Annual CO2 and CH4 fluxes of both systems were similar (8.4 ± 2.6 and 7.5 ± 2.3 t C-CO2 ha−1 year−1, −1.1 ± 1.5 and 3.3 ± 1.1 kg C-CH4 ha−1 year−1, respectively in the CIn and CM plantations) but, unexpectedly increased during the dry season.  相似文献

Wheat leaf growth is known to be spatially affected by salinity. The altered spatial distribution of leaf growth under saline conditions may be associated with spatial changes in tissue mineral elements. The objective of this study was to evaluate the spatial distributions of mineral elements and their net deposition rates in the elongating and mature zones of leaf 4 of the main stem of spring wheat (Triticum aestivum L. cv. Lona) during its linear growth phase under saline soil conditions. Plants were grown in an illitic-chloritic silty loam with 0 and 120 mM NaCl. Three days after emergence of leaf 4, sampling was begun at 3 and 13 h into the 16-h light period. Spatial distributions of fresh weight (FW), dry weight (DW), and Na+, K+, Cl, NO 3, Ca2+, Mg2+, total P, and total N in the elongating and mature tissues were determined on a millimeter scale. The patterns of spatial distribution of Na+, Cl, K+, NO3 , and Ca2+ in the growing leaves were affected by salinity, while those of Mg2+, total P, and total N were not. Sodium, K+, Cl, Ca2+, Mg2+, and total N concentrations (mmol · kg−1 FW) were consistently higher at 120 mM NaCl than at 0 mM NaCl along the leaf axis from the leaf base, whereas NO3 concentration was lower at 120 mM NaCl. Deposition rates of all nutrients were greatest in the elongation zone. The elongation zone was the strongest sink for mineral elements in the leaf tissues. Local net deposition rates of Na+, Cl, Ca2+, and Mg2+ (mmol · kg−1 FW · h−1) in the most actively elongating zone were enhanced by 120 mM NaCl, whereas for NO3 this was depressed. The lower supply of NO 3 to growing leaves may be responsible for the inhibition of growth under saline conditions. Higher tissue concentrations of Na+ and Cl may cause ion imbalance but probably did not result in ion toxicity in the growing leaves. Potassium, Ca2+, Mg2+, total P, and total N are less plausibly responsible for the reduction in leaf growth in this study. Higher tissue K+ and Ca2+ concentrations at 120 mM NaCl are probably due to the presence of high Ca2+ in the soil of this study. Received: 13 March 1997 / Accepted: 9 June 1997  相似文献

The effect of trace metal ions (Co2+, Cu2+, Fe2+, Mn2+, Mo6+, Ni2+, Zn2+, SeO4 and WO4 ) on growth and ethanol production by an ethanologenic acetogen, Clostridium ragsdalei was investigated in CO:CO2-grown cells. A standard acetogen medium (ATCC medium no. 1754) was manipulated by varying the concentrations of trace metals in the media. Increasing the individual concentrations of Ni2+, Zn2+, SeO4 and WO4 from 0.84, 6.96, 1.06, and 0.68 μM in the standard trace metals solution to 8.4, 34.8, 5.3, and 6.8 μM, respectively, increased ethanol production from 35.73 mM under standard metals concentration to 176.5, 187.8, 54.4, and 72.3 mM, respectively. Nickel was necessary for growth of C. ragsdalei. Growth rate (μ) of C. ragsdalei improved from 0.34 to 0.49 (day−1), and carbon monoxide dehydrogenase (CODH) and hydrogenase (H2ase)-specific activities improved from 38.45 and 0.35 to 48.5 and 1.66 U/mg protein, respectively, at optimum concentration of Ni2+. At optimum concentrations of WO4 and SeO4 , formate dehydrogenase (FDH) activity improved from 32.3 to 42.6 and 45.4 U/mg protein, respectively. Ethanol production and the activity of FDH reduced from 35 mM and 32.3 U/mg protein to 1.14 mM and 8.79 U/mg protein, respectively, upon elimination of WO4 from the medium. Although increased concentration of Zn2+ enhanced growth and ethanol production, the activities of CODH, FDH, H2ase and alcohol dehydrogenase (ADH) were not affected by varying the Zn2+ concentration. Omitting Fe2+ from the medium decreased ethanol production from 35.7 to 6.30 mM and decreased activities of CODH, FDH, H2ase and ADH from 38.5, 32.3, 0.35, and 0.68 U/mg protein to 9.07, 7.01, 0.10, and 0.24 U/mg protein, respectively. Ethanol production improved from 35 to 54 mM when Cu2+ was removed from the medium. The optimization of trace metals concentration in the fermentation medium improved enzyme activities (CODH, FDH, and H2ase), growth and ethanol production by C. ragsdalei.  相似文献

To evaluate the effect of ectomycorrhizal colonization on growth and physiological activity of Larix kaempferi seedlings grown under soil acidification, we grew L. kaempferi seedlings with three types of ectomycorrhizae for 180 days in acidified brown forest soil derived from granite. The soil had been treated with an acid solution (0 (control), 10, 30, 60, and 90 mmol H+ kg−1). The water-soluble concentrations of Ca, Mg, K, Al, and Mn increased with increasing amounts of H+ added to the soil. Ectomycorrhizal development significantly increased in soil treated with 10 and 30 mmol H+ kg−1 but was significantly reduced in soil treated with 60 and 90 mmol H+ kg−1. The concentrations of Al and Mn in needles or roots increased with increasing H+ added to the soil. The total N in seedlings significantly increased with increasing H+ in soil and colonization with ectomycorrhiza. The maximum net photosynthetic rate at light and CO2 saturation (P max) was greater in soil treated with 10 mmol H+ kg−1 than in controls, and was less is soils treated with greater than with 30 mmol H+ kg−1, especially with 60 and 90 mmol H+ kg−1. However, colonization with ectomycorrhiza significantly reduced the concentration of Al and Mn in needles or roots and increased the values of P max and total dry mass (TDM). The relative TDM of L. kaempferi seedlings was approximately 40% at a (BC, base cation)/Al ratio of 1.0. However, ectomycorrhizal seedlings had a 100–120% greater TDM at a BC/Al ratio of 1.0 than non-ectomycorrhizal seedlings, even though the acid treatment reduced their overall growth.  相似文献

Sandy clay loam soil was contaminated with 5000 mg kg−1 diesel, and amended with nitrogen (15.98 atom% 15N) at 0, 250, 500, and 1000 mg kg−1 to determine gross rates of nitrogen transformations during diesel biodegradation at varying soil water potentials. The observed water potential values were −0.20, −0.47, −0.85, and −1.50 MPa in the 0, 250, 500, and 1000 mg kg−1 nitrogen treatments respectively. Highest microbial respiration occurred in the lowest nitrogen treatment suggesting an inhibitory osmotic effect from higher rates of nitrogen application. Microbial respiration rates of 185, 169, 131, and 116 mg O2 kg−1 soil day−1 were observed in the 250, 500, control and 1000 mg kg−1 nitrogen treatments, respectively. Gross nitrification was inversely related to water potential with rates of 0.2, 0.04, and 0.004 mg N kg−1 soil day−1 in the 250, 500, and 1000 mg kg−1 nitrogen treatments, respectively. Reduction in water potential did not inhibit gross nitrogen immobilization or mineralization, with respective immobilization rates of 2.2, 1.8, and 1.8 mg N kg−1 soil day−1, and mineralization rates of 0.5, 0.3, and 0.3 mg N kg−1 soil day−1 in the 1000, 500, and 250 mg kg−1 nitrogen treatments, respectively. Based on nitrogen transformation rates, the duration of fertilizer contribution to the inorganic nitrogen pool was estimated at 0.9, 1.9, and 3.2 years in the 250, 500, and 1000 mg kg−1 nitrogen treatments, respectively. The estimation was conservative as ammonium fixation, gross nitrogen immobilization, and nitrification were considered losses of fertilizer with only gross mineralization of organic nitrogen contributing to the most active portion of the nitrogen pool.  相似文献

Somatic embryogenesis and whole plant regeneration was achieved in callus cultures derived from immature zygotic embryos of Prosopis laevigata (Humb. & Bonpl. ex Willd.) M.C. Johnst., recently identified as chromium (Cr), cadmium (Cd), lead (Pb) and nickel (Ni) accumulator. Embryogenic calli were induced on Murashige and Skoog (MS) medium added with a mixture of organic components plus N-6 benzyladenine (BA) (6.62 μM) and 2,4-dichlorophenoxyacetic acid (2,4-D) (2.26 μM) or thidiazuron (4.54–9.08 μM) and indole-3-acetic acid (1.42 μM). Embryogenic calli transferred onto half-strength MS medium without plant growth regulators developed globular embryos, of which 20% matured when treated with 3.75% (w/v) polyethylene glycol (PEG), and of these 50% fully differentiated into plantlet embryo. Regenerated plants were successfully acclimatized (90%), while in vitro seedlings transferred to MS medium containing 0.5 mM Cd, Cr, Ni or Pb, exhibited high heavy metals accumulation (627 mg Cr kg−1, 5,688 mg Cd kg−1, 1,148 mg Ni kg−1, and 3,037 mg Pb kg−1 dry weight) and efficient roots to shoots translocation (42–73%).  相似文献

Although common bean (Phaseolus vulgaris L.) has good potential for N2 fixation, some additional N provided through fertilizer usually is required for a maximum yield. In this study the suppressive effect of N on nodulation and N2 fixation was evaluated in an unfertile soil under greenhouse conditions with different levels of soil fertility (low=no P, K and S additions; medium = 50, 63 and 10 mg kg–1 soil and high = 200, 256 and 40 mg kg–1 soil, respectively) and combined with 5, 15, 60 and 120 mg N kg–1 soil of 15N-labelled urea. The overall average nodule number and weight increased under high fertility levels. At low N applications, nitrogen had a synergistic effect on N2 fixation, by stimulating nodule formation, nitrogenase activity and plant growth. At high fertility and at the highest N rate (120 mg kg–1 soil), the stimulatory effect of N fertilizer on N2 fixation was still observed, increasing the amounts of N2 fixed from 88 up to 375 mg N plant–1. These results indicate that a suitable balance of soil nutrients is essential to obtain high N2 fixation rates and yield in common beans.  相似文献

Hypocotyls explants of 1-week-old Ruta gravoelens L. seedlings showed high regeneration ability on Murashige and Skoog’s medium supplemented with 5.2 mM H2PO4 , 250 mM adenine sulphate, 3.0% sucrose and 8.87 μM BA. Increasing phosphate and adenine sulphate concentrations had pronounced effects on organogenesis and shoot growth compared to the stander MS medium. Multiple shoots were successfully maintained for 60 days in static liquid medium without any hyperhydricity phenomena. Uncountable shoot buds and shoots were developed on the leaf surface which was in contact with the medium. The growth index increased linearly during the culture course (R 2 = 0.992–0.999) and the highest GI (30.43) was achieved on medium enriched with 90 mM nitrogen and 4.5% sucrose. Medium supplemented with 6.0% sucrose and 60 mM N yielded the highest biomass (4.37 g per flask). However, medium with the stander N content and 6.0% sucrose developed the maximum percentage of biomass (6.67%). The highest percentage of furanocoumarins (11.46 mg g−1 dry weight) calculated as xanthotoxin was produced in medium enriched with 90 mM N and 4.5 or 6.0% sucrose, whereas the maximum rutin (73.5 mg g−1 dry weight) was on medium supplemented with 90 mM N and 4.5% sucrose. Developing system seems to be used for secondary metabolites production because the shoot rooting did not appear. So, all energy is devoted for shoots multiplication, their growth and secondary metabolites production.  相似文献

High-rate biological conversion of sulfide and nitrate in synthetic wastewater to, respectively, elemental sulfur (S0) and nitrogen-containing gas (such as N2) was achieved in an expanded granular sludge bed (EGSB) reactor. A novel strategy was adopted to first cultivate mature granules using anaerobic sludge as seed sludge in sulfate-laden medium. The cultivated granules were then incubated in sulfide-laden medium to acclimate autotrophic denitrifiers. The incubated granules converted sulfide, nitrate, and acetate simultaneously in the same EGSB reactor to S0, N-containing gases and CO2 at loading rates of 3.0 kg S m−3 d−1, 1.45 kg N m−3 d−1, and 2.77 kg Ac m−1 d−1, respectively, and was not inhibited by sulfide concentrations up to 800 mg l−1. Effects of the C/N ratio on granule performance were identified. The granules cultivated in the sulfide-laden medium have Pseudomonas spp. and Azoarcus sp. presenting the heterotrophs and autotrophs that co-work in the high-rate EGSB-SDD (simultaneous desulfurization and denitrification) reactor.  相似文献

An alkaline protease from marine Engyodontium album was characterized for its physicochemical properties towards evaluation of its suitability for potential industrial applications. Molecular mass of the enzyme by matrix-assisted laser desorption ionization-mass spectrometry (MALDI-MS) analysis was calculated as 28.6 kDa. Isoelectric focusing yielded pI of 3–4. Enzyme inhibition by phenylmethylsulfonyl fluoride (PMSF) and aprotinin confirmed the serine protease nature of the enzyme. K m, V max, and K cat of the enzyme were 4.727 × 10−2 mg/ml, 394.68 U, and 4.2175 × 10−2 s−1, respectively. Enzyme was noted to be active over a broad range of pH (6–12) and temperature (15–65°C), with maximum activity at pH 11 and 60°C. CaCl2 (1 mM), starch (1%), and sucrose (1%) imparted thermal stability at 65°C. Hg2+, Cu2+, Fe3+, Zn2+, Cd+, and Al3+ inhibited enzyme activity, while 1 mM Co2+ enhanced enzyme activity. Reducing agents enhanced enzyme activity at lower concentrations. The enzyme showed considerable storage stability, and retained its activity in the presence of hydrocarbons, natural oils, surfactants, and most of the organic solvents tested. Results indicate that the marine protease holds potential for use in the detergent industry and for varied applications.  相似文献

Mature seed-derived embryogenic calli of indica rice (Oryza sativa L. cv. PAU201) were induced on semisolid Murashige and Skoog medium supplemented with 2.5 mg dm−3 2,4-dichlorophenoxyacetic acid + 0.5 mg dm−3 kinetin + 560 mg dm−3 proline + 30 g dm−3 sucrose + 8 g dm−3 agar. Using OsglyII gene, out of 3180 calli bombarded, 32 plants were regenerated on medium containing hygromycin (30 mg dm−3). Histochemical GUS assay of the hygromycin selected calli revealed GUS expression in 50 % calli. Among the regenerants, 46.87 % were GUS positive. PCR analysis confirmed the presence of the transgene of 1 kb in 60 % of independent plants. Further, these plants have been grown to maturity in glasshouse. In vitro screening for salt tolerance showed increase in fresh mass of OsglyII putative transgenic calli (185.4 mg) as compared to control calli (84.2 mg) on 90 mM NaCl after 15 d. When exposed to 150 mM NaCl, OsglyII putative transgenic plantlets showed normal growth while the non-transgenic control plantlets turned yellow and finally did not survive.  相似文献

Summary Nitrogen balance studies were made on rice (Oryza sativa) grown in flooded soil in pots. A low rate of fertilizer (5.64 mg N. kg−1 soil) did not depress the N gain, but a high rate (99.72 mg N. kg−1 soil) elminated the N gain. Soil N loss was negligible since15N applied as ammonium sulfate and thoroughly mixed with the soil was recovered from the soil-plant system after 3 crops. The observed N gain, therefore, was caused by N2-fixation, not by a reduction of soil N loss. Straw enhanced N gain at the rate of 2–4 mg per g straw. However, this gain was not observed when soil N availability was high. Dry fallow between rice crops decreased the N gain.  相似文献

The aim of the present work was to estimate the contribution of different point and diffuse sources to the regional N2O emission strength of steppe in the Xilin river catchment, Inner Mongolia, People’s Republic of China. Transect studies showed that the topographic effect on N2O emissions from upland soils was negligible and that upland steppe is only a very weak net source of N2O during the growing season (0.8 ± 0.4 μg N2O–N m−2 h−1). Slightly higher emissions were found for riparian areas (1.8 ± 0.3 μg N2O–N m−2 h−1), which cover ∼4% of the landscape. Even faeces or urine additions stimulated N2O emissions from steppe soils only weakly (<2.5 μg N2O–N m−2 h−1 for a 5 days period). Due to low moisture contents, N2O emissions from dung heaps were also rather low (6.2 ± 0.8 μg N2O–N kg−1 dry matter h−1). In contrast, three orders of magnitude higher N2O emissions were found at sheepfolds (2.45 mg N2O–N m−2 h−1 on average). By calculating N2O emissions on a landscape scale, we show that point sources, and especially sheepfolds, become the dominating regional N2O source during the growing season if stocking rates are >1 sheep ha−1. Our results indicate that the common grazing management in the Xilin river region leads to a translocation of nitrogen from large source areas towards defined spots. This finding is further supported by measurements of NH3 concentrations at different sites. Since most of the nitrogen accumulated in these hot spots is finally lost through burning of the dried excrements by the farmers for heating and cooking purposes, the ecosystem faces a significant human perturbation of regional N cycling, which may contribute to an accelerated degradation of steppe in the Xilin river region. The response of the aquatic plant Sparganium emersum to different sediment nutrient levels was studied in three mesocosm experiments. The aim was to assess plant growth parameters and nutrient accumulation in the plant tissue under conditions relevant for habitats with sediments affected by anthropogenic nutrient enrichment. The experimental treatments were produced by fertilisation of the rooting medium (washed river sand) with differing doses of either NPK mineral fertiliser or digested sludge from solid pig slurry waste. Growth inhibition by high nutrient levels was not observed in any treatment (highest nutrient concentrations in the sediment with mineral fertiliser: N 250 mg kg−1, P 50 mg kg−1; organic fertiliser: N 6300 mg kg−1, P 1800 mg kg−1), which confirms the tolerance of S. emersum to high nutrient loads. The sediment nutrient concentration was best reflected in shoot dry mass. Nutrient contents in plant tissues were similar for most nutrient concentrations in the rooting media; only N increased significantly with N levels in the sediment in belowground parts. Nutrient standing stocks in plants, however, generally corresponded to the nutrient supply, and reached highest values (max. N 3.7 g m−2, P 1.2 g m−2) in the richest treatments with organic fertiliser. The capability of S. emersum to use nutrients from high sediment concentrations and in organically polluted environments recommends this species for use in water quality management including tertiary wastewater treatment.  相似文献

Four temperature treatments were studied in the climate controlled growth chambers of the Georgia Envirotron: 25/20, 30/25, 35/30, and 40/35 °C during 14/10 h light/dark cycle. For the first growth stage (V3-5), the highest net photosynthetic rate (P N) of sweet corn was found for the lowest temperature of 28–34 μmol m−2 s−1 while the P N for the highest temperature treatment was 50–60 % lower. We detected a gradual decline of about 1 P N unit per 1 °C increase in temperature. Maximum transpiration rate (E) fluctuated between 0.36 and 0.54 mm h−1 (≈5.0–6.5 mm d−1) for the high temperature treatment and the minimum E fluctuated between 0.25 and 0.36 mm h−1 (≈3.5–5.0 mm d−1) for the low temperature treatment. Cumulative CO2 fixation of the 40/35 °C treatment was 33.7 g m−2 d−1 and it increased by about 50 % as temperature declined. The corresponding water use efficiency (WUE) decreased from 14 to 5 g(CO2) kg−1(H2O) for the lowest and highest temperature treatments, respectively. Three main factors affected WUE, P N, and E of Zea: the high temperature which reduced P N, vapor pressure deficit (VPD) that was directly related to E but did not affect P N, and quasi stem conductance (QC) that was directly related to P N but did not affect E. As a result, WUE of the 25/20 °C temperature treatment was almost three times larger than that of 40/35 °C temperature treatment.  相似文献

The behavior of Streptomyces peucetius var. caesius N47 was studied in a glucose limited chemostat with a complex cultivation medium. The steady-state study yielded the characteristic constants μ max over 0.10 h−1, Y XS 0.536 g g−1, and mS 0.54 mg g−1 h−1. The product of secondary metabolism, ɛ-rhodomycinone, was produced with characteristics Y PX 12.99 mg g−1 and m P 1.20 mg g−1 h−1. Significant correlations were found for phosphate and glucose consumption with biomass and ɛ-rhodomycinone production. Metabolic flux analysis was conducted to estimate intracellular fluxes at different dilution rates. TCA, PPP, and shikimate pathway fluxes exhibited bigger values with production than with growth. Environmental perturbation experiments with temperature, airflow, and pH changes on a steady-state chemostat implied that an elevation of pH could be the most effective way to shift the cells from growing to producing, as the pH change induced the biggest transient increase to the calculated ɛ-rhodomycinone flux.  相似文献

Phytate, the major organic phosphorus in soil, is not readily available to plants as a source of phosphorus (P). It is either complexed with cations or adsorbed to various soil components. The present study was carried out to investigate the extracellular phytase activities of tobacco (Nicotiana tabacum variety GeXin No.1) and its ability to assimilate external phytate-P. Whereas phytase activities in roots, shoots and growth media of Pi-fed 14-day-old seedlings were only 1.3–4.9% of total acid phosphatase (APase) activities, P starvation triggered an increase in phytase secretion up to 914.9 mU mg−1 protein, equivalent to 18.2% of total APase activities. Much of the extracellular phytase activities were found to be root-associated than root-released. The plants were not able to utilize phytate adsorbed to sand, except when insoluble phytate salts were preformed with Mg2+ and Ca2+ ions for supplementation. Tobacco grew better in sand supplemented with Mg-phytate salts (31.9 mg dry weight plant−1; 0.68% w/w P concentration) than that with Ca-phytate salts (9.5 mg plant−1; 0.42%), presumably due to its higher solubility. We conclude that insolubility of soil phytate is the major constrain for its assimilation. Improving solubility of soil phytate, for example, by enhancement of citrate secretion, may be a feasible approach to improve soil phytate assimilation.  相似文献
