In normal pentobarbital-anaesthetized rats (20 mg/kg, i.p.), Ketanserine (2.5 to 10 mg/kg, i.v.) increases the temperature of the paw skin and decreases the central one. A strong thermogenetic shivering appears when central temperature falls below 35 degrees C. This shivering is a specific characteristics of Ketanserine activity, probably related to its fixation on 5-HT2 central receptors.  相似文献

The heat production and oxygen consumption of intact virgin, pregnant, lactating and postlactating rats has been investigated both in groups fed a "cafeteria" diet as well as in controls. A third group of rats fed the cafeteria diet after parturition has been investigated. Pregnant rats fed a "cafeteria" diet increased their weight faster than controls. During lactation no increases in weight were observed, and in postlactation both "cafeteria" groups attained the same values higher than controls. The ingestion of a "cafeteria" diet resulted in higher heat production in all groups except lactating rats, which--in addition--showed higher heat outputs than all the other groups when the actual data were corrected by metabolic weight according to the surface law. The high lactation heat production (and oxygen consumption) can be a consequence of increased metabolic activity in the rat organism, devoted to milk production. It can be concluded that during lactation the dam energy output through the milk must absorb any increases in energy input due to the more densely-packed energy in the "cafeteria" diet, and this did not result in increased heat production.  相似文献

In the calidrine sandpiper red knot (Calidris canutus), the weeks preceding takeoff for long-distance migration are characterized by a rapid increase in body mass, largely made up of fat but also including a significant proportion of lean tissue. Before takeoff, the pectoral muscles are known to hypertrophy in preparation for endurance flight without any specific training. Because birds facing cold environments counterbalance heat loss through shivering thermogenesis, and since pectoral muscles represent a large proportion of avian body mass, we asked the question whether muscle hypertrophy in preparation for long-distance endurance flight would induce improvements in thermogenic capacity. We acclimated red knots to different controlled thermal environments: 26 degrees C, 5 degrees C, and variable conditions tracking outdoor temperatures. We then studied within-individual variations in body mass, pectoral muscle size (measured by ultrasound), and metabolic parameters [basal metabolic rate (BMR) and summit metabolic rate (M(sum))] throughout a 3-mo period enclosing the migratory gain and loss of mass. The gain in body mass during the fattening period was associated with increases in pectoral muscle thickness and thermogenic capacity independent of thermal acclimation. Regardless of their thermal treatment, birds showing the largest increases in body mass also exhibited the largest increases in M(sum). We conclude that migratory fattening is accompanied by thermoregulatory side effects. The gain of body mass and muscle hypertrophy improve thermogenic capacity independent of thermal acclimation in this species. Whether this represents an ecological advantage depends on the ambient temperature at the time of fattening.  相似文献

Triiodothyroacetic acid (TRIAC) is a physiological product of triiodothyronine (T(3)) metabolism, with high affinity for T(3) nuclear receptors. Its interest stems from its potential thermogenic effects. Thus this work aimed 1) to clarify these thermogenic effects mediated by TRIAC vs. T(3) in vivo and 2) to determine whether they occurred predominantly in adipose tissues. To examine this, control rats were infused with equimolar T(3) or TRIAC doses (0.8 or 4 nmolx100 g body wt(-1) x day(-1)) or exposed for 48 h to cold. Both T(3) doses and only the highest TRIAC dose inhibited plasma and pituitary thyroid-stimulating hormone (TSH) and thyroxine (T(4)) in plasma and tissues. Interestingly, the lower TRIAC dose marginally inhibited plasma T(4). T(3) infusion increased plasma and tissue T(3) in a tissue-specific manner. The highest TRIAC dose increased TRIAC concentrations in plasma and tissues, decreasing plasma T(3). TRIAC concentrations in tissues were <10% those of T(3). Under cold exposure or high T(3) doses, TRIAC increased only in white adipose tissue (WAT). Remarkably, only the lower TRIAC dose activated thermogenesis, inducing ectopic uncoupling protein (UCP)-1 expression in WAT and maximal increases in UCP-1, UCP-2, and lipoprotein lipase (LPL) expression in brown adipose tissue (BAT), inhibiting UCP-2 in muscle and LPL in WAT. TRIAC, T(3), and cold exposure inhibited leptin secretion and mRNA in WAT. In summary, TRIAC, at low doses, induces thermogenic effects in adipose tissues without concomitant inhibition of TSH or hypothyroxinemia, suggesting a specific role regulating energy balance. This selective effect of TRIAC in adipose tissues might be considered a potential tool to increase energy metabolism.  相似文献

(?)-Ephedrine has been shown to increase energy expenditure and cause the loss of body fat in rats and mice. The present study compares the effects of (?)- and (+)-ephedrine, (?)- and (+)- pseudoephedrine, (±)- and (+)-norephedrine and (?)- and (+)-norpseudoephedrine on food intake, energy expenditure and body composition in ob/ob and normal mice and food intake in rats. The most potent anorectic and thermogenic compounds had the S configuration at the C-2 position but the orders of potencies for the compounds for anorexia and thermogenesis were not identical. The potencies of the compounds in reducing body lipid content correlated better with their thermogenic than their anorectic potencies.  相似文献

A chronological study was performed to investigate the postnatal development of the thermogenic capacity of the brown adipose tissue (BAT) comparing rats born and reared at 16 degrees C (cold) or 28 degrees C (control). Mitochondrial mass, cytochrome-c-oxidase activity (index of oxidative capacity) and GDP binding to mitochondria (uncoupling test) were investigated in rats from 1 to 33 days of age. Specific cytochrome-c-oxidase activity was the same in both groups during the first week, then increased in the cold group and decreased in controls; from the 9th day it was always twice as high in the former as in the latter. Specific binding of GDP to mitochondrial proteins remained almost constant in control rats during the first week contrasting with a rapid increase in that for cold rats. Afterwards it decreased in both groups until weaning but remained five times as high in cold rats as in control rats. As growth of BAT is faster and mitochondrial content greater in cold reared rats, the capacity of the tissue for thermogenesis appeared to be greatly temperature dependent soon after birth and during the entire suckling period. However the mechanisms of this stimulation remain to be elucidated.  相似文献

Different commercially available plant preparations have been claimed to have anti-obesity action. We investigated the acute effects of oral administration of 12 of these preparations in non-obese women and men. No significant increase in energy expenditure (EE) has been noted after treatment with any of these preparations. No change in respiratory quotient (RQ) was shown, except after treatment with maté (Ilex paraguariensis) extract, where a drop in RQ was observed, indicating a rise in the proportion of fat oxidized. The results suggested the poor potential of these plant preparations in the treatment of obesity, except possibly for the maté extract. Further studies are required to explore the influence of higher dosages of these preparations as well as chronic administration in man.  相似文献

Mammalian adipose tissues are broadly divided into white adipose tissue (WAT) and thermogenic fat tissue (brown adipose tissue and beige adipose tissue). Uncoupling protein 1 (UCP1) is the central protein in thermogenesis, and cells that exhibit induced UCP1 expression and appear scattered throughout WAT are called beige adipocytes, and their induction in WAT is referred to as “beiging”. Beige adipocytes can differentiate from preadipocytes or convert from mature adipocytes. UCP1 was thought to contribute to non-shivering thermogenesis; however, recent studies demonstrated the presence of UCP1-independent thermogenic mechanisms. There is evidence that thermogenic fat tissue contributes to systemic energy expenditure even in human beings. This review discusses the roles that thermogenic fat tissue plays in energy consumption and offers insight into the possibility and challenges associated with its application in the treatment of obesity and type 2 diabetes.  相似文献

Basal metabolic rate (BMR) and the thermogenic effect of food (TEF) after a liquid mixed meal of 2092 kJ (500 kcal) were examined in physically well-trained, elderly men in comparison with sedentary weight- and age-matched controls. BMR tended to be higher and TEF was significantly higher in the physically well-trained men than in the controls. No certain differences were found in plasma thyroid hormones or catecholamines. BMR correlated with whole body potassium while TEF did not. The tendency to elevated BMR in the well-trained men might therefore be due to their greater muscle mass. The elevated TEF, however, probably has other causes and might be associated with the elevated catecholamine sensitivity associated with the physically trained condition.  相似文献

The flowers or inflorescences of certain primitive seed plants are able to regulate their temperature during blooming by modulating the rate of heat production to remain much warmer than the surroundings. A large drop in ambient temperature causes a smaller drop in flower temperature which causes an increase in the rate of heat production by futile involvement of the cytochrome and alternative oxidase respiratory pathways. The result is that the rate of heat production is inversely related to ambient temperature and flower temperature remains high and relatively independent of ambient temperature. While the biophysics of thermal balance in the whole flowers is better understood, the regulation of the biochemical heat-generating pathways is not known.  相似文献

The natural trigger for heat production in the thermogenic inflorescencesof Sauromatum guttatum Schott (voodoo lily) was recently identifiedas salicylic acid (SA), which induced heat production at levelsas low as 13 ng g f. wt–1. Since then the levels of SAwere determined in other thermogenic and non-thermogenic plantspecies. In thermogenic inflorescences of five aroid species,and in male cones of at least four thermogenic cycads SA levelsduring heat production exceeded 1 µg g f. wt–1.SA was not detected in the thermogenic flowers of a water lily,Victoria regia Lindl. (Nymphaeaceae), and Bactris major Jacq.(Palmae). Levels of salicylic acid varied substantially in thefloral parts of seven non-thermogenic species and in the leavesof 27 non-thermogenic species. Resting oxygen consumption was measured in anaesthetized male and female common marmosets (Callithrix jacchus) at 29 degrees C. Injection of noradrenaline caused a marked increase in oxygen consumption (63%), but this response was lower in males. Large deposits (750 mg) of brown adipose tissue (BAT) were dissected from the cervical, subscapular, axillary and perirenal areas. High levels of guanosine diphosphate binding to isolated brown fat mitochondria were observed, indicating that the thermogenic proton conductance pathway is very active in brown fat from the marmoset. High levels of brown adipose tissue thermogenesis in these animals may be related to a requirement for diet-induced thermogenesis.  相似文献

The thermogenic carpellary appendages of the flowers of Victoriacruziana d'Orb. and the thermogenic staminal appendages of Nelumbolutea (Willd.) Pers. possess the cyanide-insensitive, ‘alternative’respiratory pathway. The presence of this pathway was demonstratedin tissue slices as well as in mitochondria. The thermogenicactivity was accompanied by ultrastructural changes in the Victoriamitochondria. Before anthesis, mitochondria with well-developedcristae were present in appendage tissue in large numbers. Duringanthesis, lamelliform cristae appeared in a different orientation. The enzyme content of the mitochondrial respiratory chain was investigated in the heat-producing plant Arum orientale. It is shown that mitochondria isolated from thermogenic tissues of this plant (with respect to non-thermogenic tissues of A. orientale or to Zea mays) demonstrate significantly elevated levels of activities of two non-coupled NADH dehydrogenases oxidizing intramitochondrial and cytoplasmic NADH pools. It is postulated that operation of a completely non-coupled respiratory chain consisting of non-coupled NADH:quinone oxidoreductases and cyanide-resistant alternative quinol-oxidase is the main mechanism of heat production in thermogenic plants.  相似文献
