The metamorphic response of pediveliger larvae of Mytilus galloprovincialis to the neurotransmitter blockers chlorpromazine, amitriptyline, rauwolscine, idazoxan, atenolol and butoxamine, and to tetraethylammonium chloride (TEA) was investigated through a series of bioassays. Chlorpromazine, amitriptyline and idazoxin inhibited larval metamorphosis induced by 10?4 M epinephrine. The concentration that inhibited metamorphosis by 50% (IC50) for chlorpromazine and amitriptyline was 1.6 × 10?6 M and 6.6 × 10?5 M, respectively. Idazoxan was less effective with an IC50 of 4.4 × 1013 M. Moreover, these three inhibitors showed no toxicity at any of the concentrations tested. The larval metamorphic response to K+ was not inhibited by 10?3 M tetraethylammonium chloride after 96 h. Thus, the neurotransmitter blockers chlorpromazine and amitriptyline are inhibitors of larval metamorphosis, and will be useful tools for antifouling studies.  相似文献

Mussels have a seasonal reproduction and cadmium is a common stressor in estuarine and coastal environments. In previous studies, we have shown that exposure to subtoxic doses of cadmium produced alterations in the properties of winter Mytilus galloprovincialis sperm protamine‐like (PL) proteins. In this study, it was analyzed the possibility that these cadmium effects may be seasonal. Winter and summer mussels were exposed to CdCl2, and it was tested the PL‐proteins for cadmium bioaccumulation, electrophoretic pattern, DNA binding, and potentiality to induce DNA oxidative damage. It was found that cadmium exposure did not produce the same effects on PL‐proteins of summer mussels that were produced on PL‐proteins of winter mussels, that is: cadmium bioaccumulation, alterations in the acetic acid‐urea polyacrylamide gels (AU‐PAGE) and sodium dodecyl sulfate‐PAGE pattern, a reduced DNA binding affinity and the ability to induce DNA oxidative damage. PL‐proteins from summer mussels, apart from not being affected by all the abovementioned effects of cadmium, also showed a very low DNA binding affinity, independent of cadmium exposure. This study reveals clock‐associated seasonal responses to cadmium in M. galloprovincialis. Understanding the mechanisms through which environmental signals guide biological rhythms is fundamental to understanding the seasonal sensitivity of this bioindicator, to use M. galloprovincialis in appropriate seasonal periods.  相似文献

Pediveliger larvae of Mytilus galloprovincialis were subjected to a series of bioassays to investigate the induction of metamorphosis using neuroactive compounds, K(+), NH(4)(+) and organic solvents. Growth and survival of post-larvae obtained using ethanol and methanol were also observed. Epinephrine, phenylephrine, clonidine and metanephrine induced larval metamorphosis at 10(-6) to 10(-4) M in both 24-h and continuous exposure assays. In 24-h exposure assays, alpha-methyldopa at 5 x 10(-5) M and methoxyphenamine at 5 x 10(-5)-10(-4) M induced 55-94% metamorphosis. Similarly, excess K(+) at 3 x 10(-2) M induced 39% metamorphosis and NH(4)(+) at 1-5 x 10(-2) M induced 63-78% metamorphosis. The EC50s of seven organic solvents ranged from 0.04 to 0.82 M. Post-larvae that metamorphosed using ethanol and methanol survived as juveniles and grew at the same rate as those from microbial biofilm. Thus, the above compounds can be useful inducers of metamorphosis for antifouling studies using larvae and juveniles of M. galloprovincialis.  相似文献

The adverse effect of seafood consumption on human health is related to the bioaccessibility in contrast with the total heavy metal level in the tissues. In this study, bioaccessibility of Cd, Cr, Cu, Mn, Ni, Pb, and Zn in Mediterranean mussel (Mytilus galloprovincialis Lamarck, 1819) edible soft tissues collected along the southern Black Sea coast were investigated using in vitro digestion model. Total Pb concentration in the tissues was found to be higher than the maximum permissible limits set by European Commission. The bioaccessibility of metals in the tissues was found to be decreased in the order: Ni (83.11%) > Cu (80.47%) > Zn (76.86%) > Cd (73.32%) > Mn (69.11%) > Pb (61.07%) > Cr (58.44%). While significant positive linear regression was observed between total and bioaccessible concentrations for Mn, Pb, and Ni, significant negative linear regression was found for Cd (p < .05). The hazard quotients (HQ) calculated using bioaccessible amount were detected lower than the limit (HQ <1), which may not pose a potential hazard to humans reported by US Environmental Protection Agency. In sum, mussel consumption from sampling sites may cause a potential risk concerning human health especially for Cd and Pb in case of increased portion sizes.  相似文献

Global aquaculture production of blue mussel has increased over last years. This work reaffirms the great potential of cryopreservation technique on mussel industry and overcome economic barriers a cause of a traditional and rudimentary management and continue growing. The aim of this work is to set some preliminary basis attending to toxicity of cryoprotecting agents (CPAs) on different development stages of Mytilus galloprovincialis as a start point to develop a stable cryopreservation protocol. Toxicity tests were carried out by using common CPAs (dimethyl-sulfoxide (Me2SO), glycerol, (GLY), propylene glycol (PG) and ethylene glycol (EG)) in a range from 0.5 to 3 M on fertilized egg, trochophore larva, and D-larva of Mytilus galloprovincialis. Results evidenced more resistance of older development stages to toxicity. Of all CPAs tested, toxicity testing highlights PG or EG as suitable CPAs for cryopreservation of early development stages; whereas D-larva was unaffected by any of the CPAs tested. Preliminary cryopreservation trials were developed to obtain information into cell cryoprotection. Further research should be focused on membrane permeability and other parameters, such as the balance between toxicity and cryoprotective effect of CPAs.  相似文献

A Southern hemisphere lineage of the blue mussel Mytilus galloprovincialis has been diverging in allopatry from Northern hemisphere conspecifics for 0.84–1.2 million years. Secondary contact between Southern and Northern hemisphere mussels in Chile, New Zealand and Australia provides an opportunity to better understand the extent and consequences of extensive range expansion. Non-native M. galloprovincialis and hybrids, as detected from RFLP assays of nuclear and mitochondrial DNA, are present in all three countries and significant cytonuclear disequilibria exist for native homozygotes in Chile and New Zealand, non-native homozygotes in Chile and non-native heterozygotes in New Zealand. Introductions into Australia are rare events given that no pure non-native mussels were detected. Immigration from one or both taxa into the hybrid zone may underlie disequilibria in New Zealand, whilst gender-directional crossing with limited ongoing hybridization contributes to disequilibria in Chile. Hybridization dynamics do not pose a threat to the Southern lineage in Chile and Australia, but in New Zealand, introgression, continued immigration and slight hybridization gender bias towards non-native maternal parents could lead to the regional extirpation of the native lineage.  相似文献

This paper examines the genetic variability of the Pacific mussel Mytilus trossulus and an introduced Atlantic species, M. galloprovincialis, in the northwestern Sea of Japan (Peter the Great Bay and Kievka Bay). The genotyping of individuals from eight populations was carried out using eight polymorphic enzyme loci and two nuclear DNA markers (Me-5 and ITS-1,2); the occurrence frequency of parent species and their hybrids was determined. The enzyme and nuclear markers demonstrated concordant genetic variation. The genotypes of the native species M. trossulus were predominant in the samples studied. The frequency of the introduced species M. galloprovincialis in the total material was relatively low; however, it reached 42 ± 2% in samples that were collected in Possjet Bay near the town of Zarubino in a zone of active international navigation. In this area the greatest number of hybrids was found as well. It is concluded that the invasion of M. galloprovincialis in the northwestern Sea of Japan is continuing; permanent populations of this mussel appeared in Possjet Bay that were not recorded here previously.  相似文献

Interactions between anthropogenic disturbances and introduced and native species can shift ecological communities, potentially leading to the successful establishment of additional invaders. Since its discovery in New Jersey in 1988, the Asian shore crab (Hemigrapsus sanguineus) has continued to expand its range, invading estuarine and coastal habitats in eastern North America. In estuarine environments, H. sanguineus occupies similar habitats to native, panopeid mud crabs. These crabs, and a variety of fouling organisms (both NIS and native), often inhabit man-made substrates (like piers and riprap) and anthropogenic debris. In a series of in situ experiments at a closed dock in southwestern Long Island (New York, USA), we documented the impacts of these native and introduced crabs on hard-substrate fouling communities. We found that while the presence of native mud crabs did not significantly influence the succession of fouling communities compared to caged and uncaged controls, the presence of introduced H. sanguineus reduced the biomass of native tunicates (particularly Molgula manhattensis), relative to caged controls. Moreover, the presence of H. sanguineus favored fouling communities dominated by introduced tunicates (especially Botrylloides violaceous and Diplosoma listerianum). Altogether, our results suggest that H. sanguineus could help facilitate introduced fouling tunicates in the region, particularly in locations where additional solid substrates have created novel habitats.  相似文献

A focused ultrastructural study of Marteilia spp. found in cultured Ostrea edulis, Mytilus edulis and Mytilus galloprovincialis from France and Spain was conducted with emphasis placed on haplosporosomes, striated plate-like inclusions and spore wall morphology. Two types of haplosporosome were identified, sphaeroid and oblate, which were common to the parasite in all 3 host species. A total of 492 haplosporosomes were measured; those from the Marteilia sp. in Mytilus spp. were marginally smaller than those in Ostrea edulis. Spore wall morphology was found to vary depending on the state of maturity of the parasite--the more mature the parasite, the thicker the wall surrounding it. It is suggested that the current criteria used to distinguish M. maurini from M. refringens are invalid and that M. maurini was relegated to a junior synonym of M. refringens.  相似文献

1. An alkaline elution procedure for the detection of DNA damage in marine invertebrate haemolymph has been developed. 2. Provided that three criteria are optimized, such as buffer composition, small filter pores (0.22 microns GVWP 025 00, Millipore), and optimal amounts of haemolymph applied, flow rates may be changed within the range of 0.2 ml/min to 0.05 ml/min without adverse back-pressure on the filter and without blocking filter pores. 3. Under optimal conditions, 70% of mussel haemolymph DNA, and 80% of crab haemolymph DNA will be retained on the filter after 6 hr of elution, indicating shorter DNA in mussel haemolymph. 4. The technique is applicable for testing the in vivo effects of different compounds on DNA in marine invertebrates, and to measurements of DNA damage in naturally exposed mussels. 5. This argues an important case for the use of alkaline elution technique for assessment of environmental genotoxicity, and especially for investigation of DNA damage in different marine organisms which cover a broad range in their DNA molecular weights.  相似文献

In Hawaii, invasive plants have the ability to alter litter-based food chains because they often have litter traits that differ from native species. Additionally, abundant invasive predators, especially those representing new trophic levels, can reduce prey. The relative importance of these two processes on the litter invertebrate community in Hawaii is important, because they could affect the large number of endemic and endangered invertebrates. We determined the relative importance of litter resources, represented by leaf litter of two trees, an invasive nitrogen-fixer, Falcataria moluccana, and a native tree, Metrosideros polymorpha, and predation of an invasive terrestrial frog, Eleutherodactylus coqui, on leaf litter invertebrate abundance and composition. Principle component analysis revealed that F. moluccana litter creates an invertebrate community that greatly differs from that found in M. polymorpha litter. We found that F. moluccana increased the abundance of non-native fragmenters (Amphipoda and Isopoda) by 400% and non-native predaceous ants (Hymenoptera: Formicidae) by 200%. E. coqui had less effect on the litter invertebrate community; it reduced microbivores by 40% in F. moluccana and non-native ants by 30% across litter types. E. coqui stomach contents were similar in abundance and composition in both litter treatments, despite dramatic differences in the invertebrate community. Additionally, our results suggest that invertebrate community differences between litter types did not cascade to influence E. coqui growth or survivorship. In conclusion, it appears that an invasive nitrogen-fixing tree species has a greater influence on litter invertebrate community abundance and composition than the invasive predator, E. coqui.  相似文献

The intertidal mussel Mytilus galloprovincialis is a successful invader worldwide. Since its accidental introduction onto the South African west coast in the late 1970s, it has become the most successful marine invasive species in South Africa. One possible explanation for this phenomenon is that M. galloprovincialis suffers less from phototrophic shell-degrading endoliths in its invasive than in its native range. We assessed photoautotrophic endolithic pressure on M. galloprovincialis in native (Portugal) and invasive (South Africa) ranges. Invasive populations were more heavily infested than native populations. In Portugal, only the biggest/oldest mussels displayed endolithic erosion of the shell and the incidence of infestation was greater at higher shore levels where more prolonged exposure to light enhances endolith photosynthesis. In South Africa, even the smallest size classes of mussels were heavily infested throughout the shore. In Portugal, endolithic-induced mortality was observed at only one location, while in South Africa it occurred at all locations and at significantly higher rates than in Portugal. Important sub-lethal effects were detected in infested native mussels, confirming previous studies of invasive populations and suggesting an energy trade-off between shell repair and other physiological constraints. We observed a positive relationship between infestation rates and barnacle colonization on mussel shells, suggesting possible facilitation of barnacle settlement/survival by shell-boring pathogens. Identification of endoliths revealed common species between regions. However, two species were unique in the invasive range while another was unique in the native region. Different levels of endolithic infestation in the invasive and the native range were not explained by the effect of major environmental determinants (Photosynthetically Available Radiation and wave height). The results reject our initial hypothesis, indicating that invasion success of M. galloprovincialis is not simply explained by escape from its natural enemies but results from complex interactions between characteristics of the invaded community and properties of the invader.  相似文献

Sharp environmental gradients encountered within the intertidal zone have driven the evolution of physiological adaptations that allow its inhabitants to maintain cellular function in the presence of fluctuating abiotic factors. These adaptations are mediated by gene-regulatory networks that, despite their inherent complexity, must remain evolvable and capable of responding to different selection pressures associated with specific ecological niches. Phosphorylation events catalyzed by cell-signaling enzymes represent a parsimonious mechanism to integrate new functional or regulatory properties into these gene-regulatory networks. In this study, proteins phosphorylated on consensus sequences for protein kinases A, B, and C; cyclin-dependent kinases; and mitogen-activated protein kinases, as well as the abundance of phosphorylated stress-activated protein kinase (phospho-SAPK/JNK), were quantified in order to ascertain whether phosphorylation events are divergent among native (Mytilus californianus and Mytilus trossulus) and invasive (Mytilus galloprovincialis) species of mussels that differ in their tolerance toward environmental stress. Abundances of phosphorylated substrate proteins for each of the major signaling proteins that were investigated, as well as the abundance of phospho-SAPK/JNK, differed both within and between species during thermal and osmotic stress. These data suggest that modulating protein function via phosphorylation may be an important mechanism to integrate novel properties into stress-regulatory networks. In turn, differential phosphorylation during environmental stress may contribute to species-specific tolerances toward abiotic stress, interspecies dynamics, and biogeographic patterns in Mytilus congeners.  相似文献

1. The accumulation and subcellular distribution of cadmium and resulting induction of metallothionein has been studied in mussels, Mytilus galloprovincialis, exposed to 400 μg Cd 1−1.2. Cadmium accumulated by mussels was primarily associated with metallothionein: a maximum four-fold increase in concentrations of the metal-binding protein (3 mg g−1 to 12 mg g−1) was observed after 30 days.3. Results for M. galloprovincialis confirm the importance of metallothionein in the detoxification of intracellular cadmium and indicate that induction rates are identical to those observed in the closely-related M. edulis.4. Intercomposition of data between these species may therefore be valid when employing metallothionein assays to determine sub-lethal response to cadmium contamination.  相似文献
