A naïve (unimmunized) human semisynthetic phage display library was employed to isolate recombinant antibody fragments against the cyanobacterial hepatotoxin microcystin-LR. Selected antibody scFv genes were cloned into a soluble expression vector and expressed in Escherichia coli for characterization against purified microcystin-LR by competition enzyme-linked immunosorbent assay (ELISA). The most sensitive single-chain antibody (scAb) isolated was capable of detecting microcystin-LR at levels below the World Health Organization limit in drinking water (1 μg liter−1) and cross-reacted with three other purified microcystin variants (microcystin-RR, -LW, and -LF) and the related cyanotoxin nodularin. Extracts of the cyanobacterium Microcystis aeruginosa were assayed by ELISA, and quantifications of microcystins in toxic samples showed good correlation with analysis by high-performance liquid chromatography. Immobilized scAb was also used to prepare immunoaffinity columns, which were assessed for the ability to concentrate microcystin-LR from water for subsequent analysis by high-performance liquid chromatography. Anti-microcystin-LR scAb was immobilized on columns via a hexahistidine tag, ensuring maximum exposure of antigen binding sites, and the performance of the columns was evaluated by directly applying 150 ml of distilled water spiked with 4 μg of purified microcystin-LR. The procedure was simple, and a recovery rate of 94% was achieved following elution in 1 ml of 100% methanol. Large-scale, low-cost production of anti-microcystin-LR scAb in E. coli is an exciting prospect for the development of biosensors and on-line monitoring systems for microcystins and will also facilitate a range of immunoaffinity applications for the cleanup and concentration of these toxins from environmental samples.  相似文献

The toxic dinoflagellate Alexandrium ostenfeldii is the only bioluminescent bloom-forming phytoplankton in coastal waters of the Baltic Sea. We analysed partial luciferase gene (lcf) sequences and bioluminescence production in Baltic A. ostenfeldii bloom populations to assess the distribution and consistency of the trait in the Baltic Sea, and to evaluate applications for early detection of toxic blooms. Lcf was consistently present in 61 Baltic Sea A. ostenfeldii strains isolated from six separate bloom sites. All Baltic Sea strains except one produced bioluminescence. In contrast, the presence of lcf and the ability to produce bioluminescence did vary among strains from other parts of Europe. In phylogenetic analyses, lcf sequences of Baltic Sea strains clustered separately from North Sea strains, but variation between Baltic Sea strains was not sufficient to distinguish between bloom populations. Clustering of the lcf marker was similar to internal transcribed spacer (ITS) sequences with differences being minor and limited to the lowest hierarchical clusters, indicating a similar rate of evolution of the two genes. In relation to monitoring, the consistent presence of lcf and close coupling of lcf with bioluminescence suggests that bioluminescence can be used to reliably monitor toxic bloom-forming A. ostenfeldii in the Baltic Sea.  相似文献

We studied if biomass, cell dimensions and microcystin concentration of the cyanobacterium Anabaena sp. (strain BIR 257; recently also referred to as Dolichospermum) change in the presence of turbidity caused by clay and a eukaryotic green algal species. Anabaena sp. was incubated in experimental lab bioassays, with added clay and the chlorophyte Brachiomonas submarina. A control was established containing a single species of Anabaena sp. Biomass of Anabaena, given as carbon concentration increased in response to clay addition, suggesting that Anabaena was stimulated by lower light conditions whereas B. submarina was not. Cell lengths increased most likely as a response to lower light conditions. Dissolved toxin concentration decreased in treatments containing clay. Increased clay turbidity may favour the cyanobacterium Anabaena sp. over its eukaryotic competitors in the future Baltic Sea, especially in low-salinity estuaries.  相似文献

Abstract

Few major groups of cyanophytes contribute significantly to the productivity of oceanic oligotrophic ecosystem: unicellular Synechococcus spp., and filamentous nitrogen-fixing Trichodesmium and Richelia species occur as abundant populations in open seas and oceans. In addition, oceanic eutrophic waters support the development of massive summer-blooms of Nodularia, Aphanizomenon and Anabaena in the Baltic Sea. The importance of the planktic cyanophytes as primary producers and their contribution to the input of new fixed nitrogen in the euphotic zone is underlined.  相似文献

This is the first report on microcystins, cyclic heptapeptide hepatotoxins, from Brazilian water supplies. A colony isolate (NPJB-1) of the colonial cyanobacteriumMicrocystis aeruginosa from Lagoa das Garças, São Paulo, was cultured under non-axenic conditions. Exponential phase cells were harvested, concentrated and lyophilized for mouse bioassays and toxin extraction. The LD100 of lyophilized cell suspensions was approximately 31 mg kg–1 (dry cell weight/animal weight). Isolation, purification and characterization of the toxins were carried out by reversed phase HPLC, HPLC amino acid analysis and fast atom bombardment mass spectrometry. Strain NPJB-1 produces two different hepatotoxic heptapeptide microcystins. The main one was microcystin-LR, the most commonly reported microcystin from cyanobacteria. The other was microcystin-LF, the phenylalanine variant of microcystin-LR. This is the first published report for microcystin-LF.  相似文献

We studied the vertical structure of the phytoplankton community in two toxic cyanobacterial blooms in the offshore Baltic Sea. In 1994, vertically separated potentially toxic, diazotrophic and mixotrophic species (belonging to Cyanophyceae, Dinophyceae and Prymnesiophyceae) dominated. In 1997, picocyanobacteria, mainly in colonies, made up 40–50% of the total phytoplankton carbon biomass in the top 20 m both day and night. Colony-forming species of picocyanobacteria seem to be occasionally important and hitherto underestimated in the Baltic Sea.We found species-specific depth distribution patterns. Nodularia spumigena and Anabaena spp. were observed mainly above 10 m depth, while Aphanizomenon sp. was mostly found deeper, especially at night. Dinophysis norvegica was only abundant near the seasonal pycnocline and showed very limited diurnal migration. Other flagellates, including small Cryptophyceae and 10 identified Chrysochromulina species, occurred down to 40 m depth. Their vertical migration may help to retrieve nutrients from below the summer pycnocline.We conclude that considerable differences in dominating functional groups may occur between years/bloom stages, and that the vertical distribution pattern of many species is recurring at similar environmental conditions, suggesting species-specific niche-separation.  相似文献

Abstract Toxic cyanobacterial mass occurrences have caused animal poisonings worldwide and may pose a health hazard for humans. Strains of the genus Anabaena are either non-toxic or produce hepatotoxins, microcystins (MCYST), or neurotoxins (such as anatoxin-a). In order to study which growth conditions favor hepatotoxic vs neurotoxic strains and how production of toxins varies, we compared the responses of two microcystin- and two anatoxin-a-producing Anabaena strains in continuous turbidostat cultures, at different temperatures, under growth-limiting light levels. Growth rates consistently remained <0.8 divisions per 24 h. Differences were strain-specific and not associated with hepatotoxicity or neurotoxicity. Thus, differential adaptation of strains to temperature and to growth-limiting light levels cannot explain why, in some cyanobacterial water blooms, hepatotoxic strains, and in others, neurotoxic ones become dominant. A statistical analysis of field data showed that the most significant discriminating factors between different types of blooms were the concentrations of dissolved PO4-phosphorus and NO3-nitrogen. Anabaena blooms with unknown neurotoxicity associated with low PO4-phosphorus and high NO3-nitrogen concentrations. Among other Anabaena blooms, the hepatotoxic ones associated with the lowest, and most of the non-toxic ones with higher concentrations of PO4-phosphorus. Anabaena blooms that contained anatoxin-a and hepatotoxic Microcystis blooms showed tendencies towards the highest concentrations of PO4-phosphorus. Non-toxic blooms dominated by genera other than Anabaena occurred over a wide range of growth conditions. In turbidostat cultures, maximal production of microcystins correlated with maximal growth rates. Light regulated the production of MCYST-LR variants, and temperature affected the production of MCYST-RR variants. Anatoxin-a seemed to be produced most under temperatures and light levels slightly suboptimal for growth. Two species of the calanoid copepod Pseudocalanus, P. acuspes and P. elongatus, have been reported from the Baltic and North Seas. Morphometric analysis based on Frost (Can J Zool 67:525–551, 1989) did not identify the species in samples collected during the German GLOBEC program; the majority of measurements fell outside reported ranges for Baltic and North Sea specimens. A multiplexed species-specific PCR (SS-PCR) protocol was designed from a ~700 bp sequence of mitochondrial cytochrome oxidase I (mtCOI), with primers designed to amplify regions of different sizes for the two species to allow detection by gel electrophoresis. The accuracy and reliability of the multiplexed SS-PCR reaction was confirmed by sequencing the mtCOI region. Preliminary SS-PCR analysis of geographic distributions of the two species indicated that P. acuspes occurred exclusively at sampling sites in the Baltic Sea, whereas P. elongatus was found in the southern North Sea and at one site in the Baltic. The SS-PCR protocol can be used for routine identification of the two species for studies of population, community and ecosystem dynamics.  相似文献

This study assessed the major histocompatibility complex (MHC) and neutral genetic variation and structure in two percid species, perch Perca fluviatilis and zander Sander lucioperca, in a unique brackish ecosystem, the Baltic Sea. In addition, to assess the importance of MHC diversity to disease susceptibility in these populations, comparisons were made to an introduced, disease susceptible, P. fluviatilis population in Australia. Eighty‐three MHC class II B exon 2 variants were amplified: 71 variants from 92 P. fluviatilis samples, and 12 variants from 82 S. lucioperca samples. Microsatellite and MHC data revealed strong spatial genetic structure in S. lucioperca, but not P. fluviatilis, across the Baltic Sea. Both microsatellite and MHC data showed higher levels of genetic diversity in P. fluviatilis from the Baltic Sea compared to Australia, which may have facilitated the spread of an endemic virus, EHNV in the Australian population. The relatively high levels of genetic variation in the Baltic Sea populations, together with spatial genetic structure, however, suggest that there currently seems to be little risk of disease epidemics in this system. To ensure this remains the case in the face of ongoing environmental changes, fisheries and habitat disturbance, the conservation of local‐scale genetic variation is recommended.  相似文献

The presence of toxin-producing cyanobacteria is of concern for the management of recreational waters. The abundance of toxic cyanobacteria and environmental concentrations of their toxins may fluctuate substantially within a short time emphasising the need for rapid methods to estimate toxin concentrations in the water. During late June-early September 2002 the abundance, biomass and characteristic pigments of Nodularia spumigena Mertens and the hepatotoxin nodularin produced by N. spumigena were analysed in water samples collected from the Baltic entrance area. Significant relationships were found between cell-bound concentrations of nodularin and the abundance and biomass of N. spumigena with a relationship of approximately 1 pg nodularin per Nodularia-cell. It is suggested that simple counts of Nodularia under the microscope may be used as a rapid on-site technique to estimate potential nodularin concentrations in recreational waters. Comparison with data from Australia shows that cell-bound concentration of nodularin per Nodularia-cell differs between geographically distant areas and therefore such relationships should be established for individual areas. The carotenoids echinenone, canthaxanthin and a cis-canthaxanthin-like caroteniod, identified from a laboratory culture of N. spumigena isolated from the Baltic Sea, were also significantly correlated with concentrations of cell-bound nodularin. However, Aphanizomenon and Anabaena, two genera commonly co-occurring with Nodularia, also contain these pigments and thus the significant correlations obtained presumably originate from Nodularia being the dominant cyanobacterium in all samples collected in 2002.  相似文献

Numbers of luminous bacteria were counted at three stations of the brackish water ecosystem of the western Baltic Sea from July 1985 to July 1986. Additional samples were taken during three cruises from stations at the North Atlantic Ocean, the Norwegian Sea and adjacent marine areas. — In Kiel Bight (western Baltic) values varied between 0 and 68,000 luminous cfu 1−1. With exception of the coastal station a distinct seasonal distribution pattern was shown in a water depth of 20 m: high numbers found in summer were opposed to low numbers in winter, the peaks being rather high in comparison to those of other areas. Statistical analysis showed that the results of 20 m were significantly different from those of 0 and 10 m depth; however, there was no correlation with temperature and salinity. Taxonomic studies revealed that the population consisted primarily of the genus Photobacterium. — The optimum of salinity was not a brackish but a marine one and was about 30% for the majority of the strains tested. A smaller number of strains grew best at a salinity between 10 and 15%. Optima of temperature ranged from 15 to 20 °C for most of the test strains. — Taxonomic analysis was also performed with luminous strains from marine areas adjacent to the western Baltic Sea, Photobacterium being the dominant genus here, too. Luminous bacteria were also enriched from the external surface and the gut contents of whitings (Merlangius merlangus) and cods (Gadus morhua). A model is proposed which explains the distribution pattern found. According to this, the gut-dwelling luminous bacteria are transported by their hosts from the North Sea into the western Baltic Sea. Here they are released into the environment, thus inhabiting another niche.  相似文献

The potential fecundity of cod in the Baltic Sea from 1993 to 1999   总被引:2,自引:1,他引:1  
Investigations on the reproduction biology of cod in the Baltic Sea (Gadus morhua L. and G. morhua callarias L.) were carried out beginning in 1993, with yearly sampling of gonads to estimate the potential absolute and relative individual fecundity in the Belt Sea (subdivision, SD: 22) and in the Arkona Sea (SD: 24). Additional samples were taken in the Bornholm Sea (SD: 25). The potential absolute fecundity of the total area of investigation is described by the regression models: Fa = 6.89 × TL3.09 and Fa = ?40 586 + 977.7 × W. As a further result the mean relative fecundity was 978 oocytes per gram body weight. Furthermore, it was shown that there was a difference in development of the potential individual fecundity between the Belt Sea cod and the original eastern Baltic cod stock. In contrast to the stable fecundity of cod in the Belt Sea, the fecundity of the large females in the Bornholm Sea significantly increased from 1993 to 1999. This change is related to a decrease in the dry weight of oocytes in the ovaries. In the same period this cod stock dramatically decreased.  相似文献

The first distribution, biomass and toxicity study of a newly established bloom of the colonial cyanobacteria Microcystis aeruginosa was conducted on October 15, 2003 in the upper San Francisco Bay Estuary. Microcystis aeruginosa was widely distributed throughout 180 km of waterways in the upper San Francisco Bay Estuary from freshwater to brackish water environments and contained hepatotoxic microcystins at all stations. Other cyanobacteria toxins were absent or only present in trace amounts. The composition of the microcystins among stations was similar and dominated by demethyl microcystin-LR followed by microcystin-LR. In situ toxicity computed for the >75 m cell diameter size fraction was well below the 1 g l–1 advisory level set by the World Health Organization for water quality, but the toxicity of the full population is unknown. The toxicity may have been greater earlier in the year when biomass was visibly higher. Toxicity was highest at low water temperature, water transparency and salinity. Microcystins from the bloom entered the food web and were present in both total zooplankton and clam tissue. Initial laboratory feeding tests suggested the cyanobacteria was not consumed by the adult copepod Eurytemora affinis, an important fishery food source in the estuary.  相似文献

Three water bloom samples were collected in August 1986 from the southern Baltic Sea. Acute toxicity of the samples was determined by mouse bioassay and the toxins were further studied by HPLC. The bloom samples contained equal amounts of cyanobacteria Nodularia spumigena and Aphanizomenon flos-aquae and were hepatotoxic. Two hepatotoxic Nodularia spumigena strains were isolated from the samples. The isolates produce a toxic peak indistinguishable from the bloom material in the HPLC analysis. The toxicity of the fractions was verified by mouse bioassay. Thus the toxicity of the bloom samples was in all likelihood caused by Nodularia spumigena.  相似文献

The community structure of the bacteria associated with Nodularia spumigena (Mertens) cyanobacterial aggregates in the Baltic Sea was studied with temperature gradient gel electrophoresis (TGGE), using a 16S rRNA gene fragment as a target. Various developmental stages of the aggregates and free-floating cyanobacterial filaments were sampled to reveal possible changes in associated microbial community structure during development and senescence of the aggregates. The microbial community structures of all samples differed, and the communities of young and decaying aggregates were separated by cluster analysis of the TGGE fingerprint data. Sequencing of the TGGE fragments indicated the presence of bacteria from the α-, β-, and γ-proteobacterial groups, as well as members of Cytophaga–Flexibacter–Bacteroides lineages and gram-positive Actinobacteria spp. The majority of the Nodularia-associated sequences were not closely related to previously reported 16S rDNA sequences from the Baltic Sea or any other environment. The structure of the bacterial assemblage reflects the environmental changes associated with the succession and decay of the cyanobacterial aggregates. In addition, the sequence data suggest that the N. spumigena (Mertens) blooms in the Baltic Sea may host thus far uncharacterized bacterial species.  相似文献

In a macrogeographic survey, the population genetic structure of mussels from various regions of the Baltic Sea, a large semi-enclosed brackish-water basin, was examined with reference toMytilus edulis andM. galloprovincialis samples from the North Sea, Irish coast and southern Portugal. Electrophoretically detectable variation was analysed at 6 polymorphic enzyme loci (Ap, Est-D, Lap-2, Odh, Pgi andPgm). Evidence was provided of a remarkably large amount of biochemical genetic differentiation among ecologically and morphologically divergent mussel populations in the Baltic. Patterns of allele frequencies in low-salinity populations from the area of the Baltic Proper were demonstrated to be widely homogeneous but contrast strongly with those of the western Baltic, the latter resembling populations from marine habitats of the North Sea. Associated with a pronounced salinity gradient, the spatial heterogeneity in gene-pool structure is indicated by steep clines of allele frequency changes in the area of the eastern Danish isles. The adaptive significance of the observed allozymic variation is suggested. From genetic distance estimates, the subdivision of population structure is discussed in relation to the significant amount of differentiation detected withinMytilus populations to date and to the evolutionary time required for the divergence of Baltic mussel populations. The allozymic data provide evidence for the genetic distinctiveness of mussels from the low-salinity areas of the Baltic. Their position at the specific or subspecific level of classification requires further consideration.  相似文献

Recent studies showing consequences of species’ genetic diversity on ecosystem performance raise the concern of how key ecosystem species are genetically structured. The bladder wrack Fucus vesiculosus L. is a dominant species of macroalga in the northern Atlantic, and it is particularly important as a habitat‐forming species in the Baltic Sea. We examined the genetic structure of populations of F. vesiculosus with a hierarchical approach from a within‐shore scale (10 m) to a between‐seas scale (Baltic Sea–Skagerrak, 800 km). Analysis of five microsatellite loci showed that population differentiation was generally strong (average FST = 12%), being significant at all spatial scales investigated (101, 103, 104–5, 106 m). Genetic differentiation between seas (Baltic Sea and Skagerrak) was substantial. Nevertheless, the effects of isolation by distance were stronger within seas than between seas. Notably, Baltic summer‐reproducing populations showed a strong within‐sea, between‐area (70 km) genetic structure, while Baltic autumn‐reproducing populations and Skagerrak summer‐reproducing populations revealed most genetic diversity between samples within areas (<1 km). Despite such differences in overall structure, Baltic populations of summer‐ and autumn‐reproducing morphs did not separate in a cluster analysis, indicating minor, if any, barriers to gene flow between them. Our results have important implications for management and conservation of F. vesiculosus, and we raise a number of concerns about how genetic variability should be preserved within this species.  相似文献

Since 1985, the nonindigenous polychaete species Marenzelleria neglecta has been found in the Baltic Sea. The species, which was introduced by ship ballast water, spreads rapidly and dominates in many habitats today. Using three gene segments of the mitochondrial DNA (16S rDNA, Cytochrom oxidase I, Cytochrom b), we investigated four populations of the western and northern Baltic Sea in a preliminary survey and compared them with four other populations from the North Sea, the Baltic Sea and from the Arctic. First, we could demonstrate the applicability of the markers to discriminate the species with certainty. Second, with M. viridis and M. arctia, we could detect two more species of the same genus, which have recently been introduced into the Baltic Sea. One of these, M. arctia, was hitherto known as an exclusive arctic member of the genus. The impact of these two recently invaded Marenzelleria species onto the autochthonous fauna needs to be evaluated in the future. The Baltic Sea as a ‘natural aquarium’ now offers the possibility to investigate sibling species simultaneously. However, correct identification and denomination of Marenzelleria species are indispensable prerequisites for all future studies. Molecular markers allow the exact identification of all Marenzelleria species and must be used whenever a classical taxonomic identification is uncertain.  相似文献

Ceramium tenuicorne (Kützing) Wærn is a red alga that is widely distributed in the brackish Baltic Sea. We studied the growth response of Ceramium to low salinity and nutrient enrichment, using isolades from two regions of the Baltic Sea where the alga approaches its inner distribution limit. Ecotypic differentiation was observed in that differences in growth response among isolates corresponded to salinity conditions in their regions of origin. Isolates from the Gulf of Bothnia (4 psu) had inherently lower growth rates that were not increased when transferred to higher salinity, but were better adapted to very low salinity levels than isolates from the Baltic Proper (7 psu). Further, the results indicate that Ceramium from different regions of the Baltic Sea vary in their responses to nutrient enrichment. The observed differences may be best described as a quantitative difference in the proportion of isolates with hyposaline adaptation. The results indicate that the wide distribution of Ceramium in the Baltic Sea is better explained by the occurrence of locally adapted genotypes than by a generalist life strategy, and provide example of adaptive differentiation in a marine edge environment.  相似文献

Stable isotope analysis of Baltic Sea cod from the Bornholm Sea (ICES‐SD 25) and western Baltic (Belt Sea, ICES‐SD 22) revealed significant differences in the δ15N and δ13C values of the dorsal muscle, as well as in the δ18O values of otoliths. The method pledges to become especially appropriate to the Baltic due to the high variability in oxygen isotope ratios associated with its estuarine nature.  相似文献
