Transferrin: Evidence for two common subtypes of the TfC allele   总被引:2,自引:0,他引:2  
Summary Evidence is presented for an extended polymorphism of human transferrin (Tf). Three common phenotypes were observed among TfC individuals after isoelectric focusing of sera on polyacrylamide gels. They are explained in terms of two subtypes of the Tfc allele, tentatively designated TfC1 and TfC2. The distribution of the phenotypes Tf C1, C2-1, and C2 provides a good fit to the Hardy-Weinberg equation. In our population sample (n=942) the following frequencies were calculated: TfC1=0.8195, TfC2=0.1720, TfB2=0.0064, TfB1–2=0.0016, and TfD1=0.0005. Family studies (n=112) indicate an autosomal codominant way of inheritance. The observed subheterogeneity is detectable in purified transferrin after isofocusing and subsequent immunofixation. The subtypes are still present after treatment of sera with neuraminidase.  相似文献

Summary Simultaneous subtyping of two genetic markers—group-specific component (Gc) and transferrin (Tf)—by electrofocusing enabled us to compute the following gene frequencies for the Tunisian population: Gc IS .0.525; Gc IF , 0.260; Gc 2, 0.215; Tf CI , 0.770; Tf C2 , 0.215; Tf D1 , 0.015.The frequencies of Tf D , Tf C2 , and Gc 1 are higher than those found in Caucasoid populations and can be explained by Negroid contribution. A selective advantage related to the metabolic role of this vitamin D-binding protein does not seem very likely for any particular Gc type or subtype. It is postulated that the differences in the frequencies of the Gc alleles might be related to selective advantage for genes belonging to other genetic systems originally closely linked to either Gc 1 or to Gc 2 alleles.This work was supported in part by the Faculté de Pharmacie et de Médecine Dentaire of Monastir and by a grant from the Ambassade de France in Tunisia  相似文献

Transferrin types and glutathione (GSH) levels were determined for 38 rhesus monkeys (Macaca mulatto). Eight transferrin alleles were found: their gene frequencies were: TfA 0.04, Tfc 0.50, TfD 0.03, TfD 0.09, TF 0.11, TF 0.05, TfG 0.11, and Tf H' 0.08. The 16 phenotypes found and the number of each was: Tf AC 3, Tf CC 9, Tf CD‘ 2, Tf CD 2, Tf CF 5, Tf CF 3, Tf CH’ 3, Tf CG 2, Tf D'D‘ 1, Tf D'F 1, Tf D'G 1, TfD'H’ l, TfFG2, TfF'G ‘l.TfCG l.and Tf H'H’ 1. Mean GSH level was 310.6 mg/100 ml red cells. The Tf CC group had significantly lower GSH levels than Tfc heterozygotes or non-Tfc homozygotes. The Tfc heterozygotes had significantly lower GSH levels than non-Tfc homozygotes. Tfc, TfD' and TF alleles were associated with lower GSH levels. Glutathione levels were higher in older animals and were higher in September than in December. Sex differences in glutathione levels were not found.  相似文献

Summary TransferrinC (TfC) subtypes were determined by isoelectric focusing (PAGIF) on samples from 90 carriers of the TfB and TfD alleles. In all cases of CB and CD heterozygotes only one of the two common subtypes of the TfC allele, TfC1 or TfC2, was observed. This is considered strong support for the hypothesis of two common alleles at the Tf locus. The different isofocusing patterns of rare B and D variants are compared with the results obtained after agarose gel electrophoresis (AGE).  相似文献

Distribution of transferrin phenotypes in selected troops of Kenya baboons   总被引:1,自引:0,他引:1  
The distribution of transferrin alleles in a group of common baboons, Papio cynocephalus of East Africa, was determined after starch gel electrophoresis of plasma and autoradiography of electrophoretograms. Three alleles, TfA, TfB, and TfC, were found, in agreement with results previously reported. The frequencies of these alleles are TfA = 0.276, TfB = 0.515, Tfc = 0.209. The difference between the frequencies reported here and those reported by Buettner-Janusch (1963 Folia Primat., 1: 73–87) for the same species (TfA = 0.205, TfB = 0.332, Tfc = 0.463) is significant. Transferrin of P. cynocephalus contains four residues of sialic acid per molecule of transferrin, as determined by electrophoresis and autoradiography of plasma treated with neuraminidase.  相似文献

Summary The genetic types of orosomucoid (ORM) were analyzed by isoelectric focusing (IEF) on polyacrylamide gels and subsequent print lectinofixation with a lectin from the beetle, allo A. In this paper, the newly devised print lectinofixation for ORM typing is described. This technique is faster, easier to perform, and has been found to be a useful tool in population genetics and forensic medicine. The results of typing for two alleles, ORM *1 and ORM *2 are described for a population of Northern Japan (n=500). We use the designation “lectinofixation” to denote the method using lectin in place of monospecific antibody in the immunofixation  相似文献

Summary Phenotyping for Gc variants by conventional electrophoresis in 835 Papio hamadryas baboons demonstrated a monomorphic population. Gc subtyping by polyacrylamide IEF gels, pH 4–6, on 394 of these baboons revealed the existence of two common alleles which we named Gc Papio 1 and Gc Papio 2 . Pedigree data confirmed the inheritance of a single locus, two allele system and the observed gene frequencies were 0.593 for Gc Papio 1 and 0.407 for Gc Papio 2 .  相似文献

Summary Malays, Chinese, and Indians from Peninsular Malaysia; Ibans and Bidayuh from Sarawak State; Kadazans from Sabah State, Northern Borneo; and Bataks, Minangkabau, and Javanese from North Sumatra, Indonesia, were subtyped for transferrin C by polyacrylamide gel isoelectric focusing. All nine populations studied are polymorphic for two alleles, TfC1 and TfC2. TfC3 was polymorphic in six populations and present as a rare variant in the other three. The frequency of TfC1 ranged from 0.855 in Bidayuh to 0.711 in Javanese, that of TfC2 from 0.231 in Indians to 0.113 in Bidayuh, and that of TfC3 from 0.030 in Javanese and Chinese to 0.008 in Bidayuh. TfDchi is polymorphic in all the populations that we studied except in Minangkabau, in whom it is present as a rare variant, and in Indians, in whom it is absent.  相似文献

Abstract

Blood plasma from three populations of feral goats (Capra hircus L.) and from five domestic breeds was analysed by polyacrylamide gel electrophoresis. The albumin locus was monomorphic in all samples. The macroglobulin locus was variable but lacked resolution on the gel. The transferrin (Tf) locus was polymorphic. The only two Tf alleles observed (Tf A and Tf B) did not vary from those described by other authors. Gene frequencies of the two Tf alleles showed that one feral population could be distinguished from the other two. Gene frequencies of domestic breeds were variable and showed no definitive pattern. Domestic breed type could not be recognised in the feral populations on the basis of transferrin analysis alone.  相似文献

Summary The role of specific transferrin (Tf) and Tf receptor interaction on brain capillary endothelial cells in iron transport from the plasma to the brain was investigated by using Tf from several species of animals labeled with 59Fe and 125I, and 15-day and adult rats. The rate of iron transfer was much greater in the 15-day rats. It was greatest with Tf from the mammals, rat, rabbit and human, but much lower with chicken ovotransferrin and quokka (a marsupial), toad, lizard, crocodile, and fish Tf. The uptake of Tf by the brain showed a similar pattern, except for a very high uptake of ovotransferrin (ovo Tf). Iron uptake by the femurs (a source of bone marrow) was also high with Tf from the mammalian species and low with the other types of Tf, but showed little change with aging of the animals. It is concluded that iron transport into the brain is dependent on the function of Tf receptors, probably on capillary endothelial cells, and that these receptors show the same type of species specificity as the receptors on immature erythroid cells. Also, the decrease in iron uptake by the brain as rats age from 15 days to adulthood is specific for the brain and is not a general effect of the aging process.Abbreviations Tf transferrin - ovo Tf ovotransferrin  相似文献

The polymorphism of Gc (group-specific components) has been investigated for a series of 3,160 individual samples from 11 Mongoloid populations in Asia and North and South America by isoelectric focusing on polyacrylamide gels. The samples fall into six Gc phenotypes which can be explained by the three common alleles, Gc1F, Gc1S, and Gc2, together with several variant phenotypes explained as the heterozygotes for the three common alleles. The distribution of Gc1F suballele appears to be considerably different from population to population among Mongoloids, ranging from 0.105 (Machiguenga Indans, Peru) to 0.609 (Kadazan, Borneo). A clear geographic cline from Southeast Asia into South America in Gc1F allele was observed in the populations. In general, Gc1F allele frequencies are lower in European populations and higher in African populations. The range of variability in the Gc1F values observed among the Asiatic populations is between the Africans and the Europeans.  相似文献

The pathway and frequency of species' introductions can affect the extent, impact, and management of biological invasions. Here, we examine the pathway of introduction of the aquatic plant Cabomba caroliniana (fanwort) into Canada and the northern United States using plastid DNA sequence (intergenic spacers atpFatpH, trnHpsbA, and trnLtrnF) and DNA content analyses. We test the hypothesis that the spread of fanwort is a result of commercial trade by comparing a Canadian population (Kasshabog Lake, ON) to native populations from southern U.S., introduced populations in northern U.S., and plants from commercial retailers. Thirteen plastid haplotypes were identified throughout North America, including one dominant haplotype, which was present in all C. caroliniana populations. Several rare haplotypes were used to infer shared colonization history. In particular, the Canadian population shared two rare alleles with a population from Massachusetts, suggesting range expansion of C. caroliniana from the northern U.S. However, the possibility of a commercial introduction cannot be excluded, as common alleles were shared between the Canadian population and both commercial and southern U.S. sources. Variation in C. caroliniana genome size was bimodal and populations were classified into “high” and “low” categories. The Canadian population had DNA contents similar to several northern U.S. populations (low DNA content). This may provide additional support for range expansion from these introduced populations rather than from commercial sources or populations in the southern U.S., which had high DNA content.  相似文献

As the serum transferrin polymorphism was observed in several macaque species, we considered it as one of the best markers for the study of population genetics of Japanese macaques,Macaca fuscata. In this work the genetic variants of transferrin (Tf) of 1,451 blood samples from 37 troops of this species were tested. The troops showing the variation of Tf were Fukushima, Yugawara T, Ihama, Ryozenyama, Mikata I and II, Takahama, Takahama (Otomi), Arashiyama A, Minoh A and B, Kohchi, Mihara, Shimane, and Tomogashima. The wild-type allele of this species was Tf F, and the variant alleles detected in these troops were E, G, G, and H′. The alleles E, G and H′ were probably identical with those reported in several macaque species byIshimoto (1972), but the identification of allele G could not be done.  相似文献

A total of 397 persons living in six villages of three Brazilian Indian tribes were studied in relation to the Gc subtypes. The corresponding gene frequencies are more similar between the Gorotire and Caingang than between the Gorotire and the Krahó, despite the considerable geographical distance that separates the villages of these two first tribes and their lignuistic differentiation. An uncommon variant pattern (1C7) was observed in eight Gorotire Indians; it had been described for the first time in a Tibetan sample, furnishing additional evidence on the Asiatic origin of these Indians. The distinct Gc subtype frequencies observed in our main ethnic groups provide an important new tool for anthropological analyses. Tf subtypes were studied among the Caingang only. The frequencies of TfC1 and TfC2 are similar to those obtained by other researchers in Hessen, Germany.  相似文献

Glucoamylase[α-1,4: 1,6-glucan-4: 6-glucohydroease, EC 3.2.1.3] from Rhizopus niveus was entrapped in polyacrylamide gels and adsorbed onto SP-Sephadex C–50 to elucidate the thermostability mechanism of immobilized enzymes. The thermal stability of immobilized glucoamylase entrapped in polyacrylamide gels was enhanced slightly compared with glucoamylase in free solution, and was independent of the acrylamide monomer concentration and N, N′-methylene-bis (acrylamide) content. To explain this phenomenon, the cellular structure of polyacrylamide gel was taken into consideration in addition to interactions between glucoamylase and gel, and a decrease in dielectric constant in the gel [S. Moriyama et al., Agric. Biol. Chem., 41, 1985 (1977)1)]. On the other hand, immobilized glucoamylase bound to SP-Sephadex by ionic interaction showed lower stability than free glucoamylase, and much greater stability than glucoamylase in the presence of dextran sulfate, a constituent of SP-Sephadex. Thermal stabilities for the free and immobilized enzymes were also compared at the pH not in the bulk solution, but in the SP-Sephadex.  相似文献

The main objective of this study was to determine the preliminary Diagnostic Reference Levels (DRLs) in terms of Kerma Area Product (KAP) and fluoroscopy time (Tf) during Endoscopic Retrograde Cholangio-Pancreatography (ERCP) procedures. Additionally, an investigation was conducted to explore the statistical relation between KAP and Tf.Data from a set of 200 randomly selected patients treated in 4 large hospitals in Greece (50 patients per hospital) were analyzed in order to obtain preliminary DRLs for KAP and Tf during therapeutic ERCP procedures. Non-parametric statistic tests were performed in order to determine a statistically significant relation between KAP and Tf.The resulting third quartiles for KAP and Tf for hospitals (A, B, C and D) were found as followed: KAPA = 10.7 Gy cm2, TfA = 4.9 min; KAPB = 7.5 Gy cm2, TfB = 5.0 min; KAPC = 19.0 Gy cm2, TfC = 7.3 min; KAPD = 52.4 Gy cm2, TfD = 15.8 min. The third quartiles, calculated for the total 200 cases sample, are: KAP = 18.8 Gy cm2 and Tf = 8.2 min. For 3 out of 4 hospitals and for the total sample, p-values of statistical indices (correlation of KAP and Tf) are less than 0.001, while for the Hospital A p-values are ranging from 0.07 to 0.08. Using curve fitting, we finally determine that the relation of Tf and KAP is deriving from a power equation (KAP = Tf1.282) with R2 = 0.85.The suggested Preliminary DRLs (deriving from the third quartiles of the total sample) for Greece are: KAP = 19 Gy cm2 and Tf = 8 min, while the relation between KAP and Tf is efficiently described by a power equation.  相似文献

Transferrin phenotypes of 403 Malagasy were determined by autoradiography after electrophoresis of plasma or serum. Three phenotypes, Tf C, Tf CD, and Tf D, were found. The frequency of TfD was 0.051 in the total sample, 0.041 in tribes of the Madagascar plateau, and 0.075 in tribes of the coastal and lowland regions. Haptoglobin phenotypes of 402 Malagasy were determined. The frequency of Hp1 was 0.41 in the total sample, 0.38 in tribes of the plateau, and 0.49 in coastal-lowland tribes. There is a significant difference in the frequency of Hp1 between the tribes of the plateau and those of the coastal and lowland regions. This confirms earlier observations of significant differences in frequencies of haptoglobins and hemoglobin S. Plasma or serum samples from 405 Malagasy were examined by electrophoresis to determine ceruloplasmin phenotypes. Seven distinct ceruloplasmin bands were observed. These were A, B, Galveston, Bridgeport, New Haven, Tananarive, and X, the last two described for the first time. Ten different phenotypes composed of one or two of the bands were observed. The frequency of the allele for ceruloplasmin B (CpB) was 0.78 in the total sample, 0.73 in tribes of the plateau, and 0.92 in coastal and lowland tribes. There is a significant difference in the frequency of this allele in the two major tribal groups, and this provides additional evidence of genetic differences between the peoples of the plateau and those of the coast and lowland regions of Madagascar.  相似文献

Hemoglobin, haptoglobin, and transferrin phenotypes were determined by means of starch-gel electrophoresis for a sample of 226 Kenya hospital patients. Allele frequencies were: HbβS=0.081; Hp1=0.057; TfD=0.038. Hemoglobin S was the only aberrant hemoglobin found in this sample. Transferrins C and D were the only transferrins found. Hemoglobin and transferrin phenotypes were also determined for a sample of 201 newborn Kenya infants. One of these infants had hemoglobins F, S, and C, eight had hemoglobins A, F, and S, and the remainder had hemoglobins A and F. Transferrins C, B, and D were found in this sample. Hypervariability of intronic simple (gt)n(ga)m repeats in HLA-DRB genes   总被引:2,自引:2,他引:0  
We have investigated the extent of DNA variability in intronic simple (gt)n(ga)m repeat sequences and correlated this to sequence polymorphisms in the flanking exon 2 of HLA-DRB genes. The polymerase chain reaction (PCR) was used to amplify a DNA fragment containing exon 2 and the repeat region of intron 2. The PCR products were separated on sequencing gels in order to demonstrate length hypervariability of the (gt)n(ga)m repeats. In a parallel experiment, the PCR products were cloned and sequenced (each exon 2 plus adjacent simple repeats) to characterize the simple repeats in relation to the HLA-DRB sequences. In a panel of 25 DRB1, DRB4, and DRB5 alleles new sequences were not detected. Restriction fragment length polymorphism (RFLP) subtyping of serologically defined haplotypes corresponds to translated DNA sequences in 85% of the cases, the exceptions involving unusual DR/DQ combinations. Many identical DRB1 alleles can be distinguished on the basis of their adjacent simple repeats. We found group-specific organization of the repeats: the DRw52 supergroup repeats differ from those of DRB1*0101, DRB4*0101, and DRB5*0101 alleles and from those of pseudogenes. Finally, we amplified baboon DNA and found a DRB allele with extensive similarity to DRB1 sequences of the DRw52 supergroup. The simple repeat of the baboon gene, however, resembles that of human pseudogenes. In addition to further subtyping, the parallel study of polymorphic protein and hypervariable DNA alleles may allow conclusions to be drawn on the relationships between the DRB genes and perhaps also on the theory of trans-species evolution.The nucleotide sequence data reported in this paper have been submitted to the GenBank nucleotide sequence database and have been assigned the accession number M 34258.  相似文献

Interspecific hybridization has resulted in the recent origin of several hybrid Senecio taxa at diploid, tetraploid and hexaploid levels. As part of research aimed at constructing and comparing genomic maps of each of these taxa and their parents, we have isolated microsatellite loci from genomic DNA libraries of S. vulgaris and S. squalidus. Primers of 35 loci amplified microsatellites resolved in agarose gels from one or more of S. vulgaris, S. squalidus, S. aethnensis and S. chrysanthemifolius. Approximately 71% of primers amplified a product in all four species. A survey of microsatellite variation in S. chrysanthemifolius over a subset of 14 loci resolved 2–11 alleles per locus in polyacrylamide gels with expected heterozygosity (HE) ranging from 0.26 to 0.87.  相似文献
