Endophytic fungi were isolated from healthy, living, and symptomless tissues of inner bark, leaf, and roots of Aegle marmelos, a well-known medicinal plant, growing in different parts of India including Varanasi. A total of 79 isolates of endophytic fungi were isolated, representing 21 genera, adopting a standard isolation protocol. Members of the deuteromycotina were more prevalent than ascomycotina and others. The result was quite encouraging in terms of maximum isolates recovery from hyphomycetes (78.5%) followed by ascomycetes (8.9%) and coelomycetes (7.6%) respectively, which corroborates previous studies in same area. However, 5.1% isolates remained unidentified and were classified under Mycelia Sterilia. No isolate was obtained from either basidiomycotina or from zygomycotina. Fusarium spp. had maximum colonization frequency (8.00%) in this plant. The other dominant endophytic genera were Aspergillus spp., Alternaria sp., Drechslera sp., Rhizoctonia sp., Curvularia sp., Nigrospora sp., and Stenella sp. Only two ascomycetous members Chaetomium globosum and Emericella sp. (perfect state of Aspergillus sp.) were obtained from the bark sample. These results indicated that distribution of endophytic fungi within the A. marmelos is not even. Bark harbors more endophytic fungi than leaf and root.  相似文献

Discs of tissue excised from ripe nonflue-cured and flue-cured tobacco leaves, harvested on six dates and cultured on three selective media, yielded 21 and 24 genera of fungi, respectively. Of 5,094 fungi isolated from 3,240 pieces of nonfluecured leaf tissue, 89.5% comprised five genera, Alternaria, Cladosporium, Epicoccum, Trichoderma, and Nigrospora. Of 2,494 fungi isolated from 3,240 pieces of flue-cured leaf tissue, 70.9% were Alternaria, Cladosporium Epicoccum, Aspergillus, and Nigrospora. Flue-cured and nonflue-cured brown-spot lesions harvested at two locations yielded 12 and 14 genera of fungi, respectively. Alternaria, Penicillium, Phoma, and Stemphyllium comprised 91.5% of the 2,245 fungi isolated from noncured and 87.1% of the 1,118 fungi isolated from the cured lesions. The number and kinds of fungi obtained from diseased and healthy tissue were reduced but not eliminated by flue-curing.  相似文献

Summary In order to study the foliar endophytes from teak (Tectona grandis L.) and rain tree (Samanea saman Merr.) growing in the campus of Chulalongkorn University, healthy leaves were collected at two-monthly intervals during January to December. The number of genera and species, together with their colonization frequency (CF%) in mature teak and rain tree leaves were greater than those in the young leaves. More endophytic isolates in the leaves of both trees were recovered during the rainy season. The fungal genera found in both young and mature teak leaves were Alternaria, Colletotrichum, Nigrospora, Phomopsis and mycelia sterilia. Phomopsis was the dominant genus in both young (newly emerged) and mature leaves. Fusarium, Penicillium, Schizophyllum commune and members of the Xylariaceae were found only in mature leaves. For the rain tree leaves, species of Phomopsis and mycelia sterilia were found in both young newly emerged and mature leaves. Colletotrichum and Penicillium were found only in mature leaves, whereas Nigrospora was found only in young newly emerged leaves. In this study, Phomopsis was the dominant genus in the leaves of both tree species. A total of 37 isolates of endophytic fungi isolated from teak and rain tree leaves were tested for the production of antimicrobial activities. Out of these, 18 isolates could produce inhibitory substances effective against Bacillus subtilis, Staphylococcus aureus and Escherichia coli and 3 isolates inhibited growth of Candida albicans in vitro.  相似文献

In order to study the species composition of endophytes from wheat healthy plants in Buenos Aires Province (Argentina) and to determine their infection frequencies from leaves, stems, glumes and grains, wheat plants were collected from five cultivars at five growth stages from crop emergence to harvest. A total of 1,750 plant segments (leaves, stems, glumes and grains) were processed from the five wheat cultivars at five growth stages, and 722 isolates of endophytic fungi recovered were identified as 30 fungal genera. Alternaria alternata, Cladosporium herbarum, Epicoccum nigrum, Cryptococcus sp., Rhodotorula rubra, Penicillium sp. and Fusarium graminearum were the fungi that showed the highest colonization frequency (CF%) in all the tissues and organs analysed. The number of taxa isolated was greater in the leaves than those in the other organs analysed.  相似文献

Summary By utilizing energy-dispersive X-ray microanalysis the heavy metals manganese, iron, zinc and lead were determined in different tissues of beech (Fagus sylvatica L.) autumn leaves. It was observed that all four metals were accumulated in sieve element-companion cell complexes of the leaf veins in the period from the green (2 October) to the brown (23 October) leaf coloration. Concomitantly, the concentration of these metals decreased in certain tissues of the stems subtending the leaves. The major reservoirs of heavy metals in the stems were periderm, cortex, pith and xylem rays. Since sieve element-companion cell complexes of the stems showed increasing metal concentrations during the transition from the green to the brown leaf stage, and since this temporary increase also occurred in the petiole phloem, it is inferred that symplastic transport can be used in addition to apoplastic transport for the displacement of heavy metals. A collection of endophytic fungi was obtained from living apparently symptomless roots, stems, leaves and fruits of Melia azedarach. This is an exotic tree introduced into Brazil from Asia and is a producer of insecticidal compounds. It grows very well and is apparently resistant to many pathogens. Each segment from the plant was sampled during two periods. A total of 55 fungal isolates were recovered. Hyphomycetes were more prevalent than Ascomycetes, Coelomycetes and Basidiomycetes. The genera Aspergillus and Penicillium were the most common in the plant.  相似文献

Fragments of the desert moss Syntrichia caninervis Mitt. were grown on the surface of moistened sand to assess their regeneration capacity. The plant material was collected in two different years (2014 and 2015) and divided into five fragment classifications (stem apices, green leaves, yellow-green leaves, brown leaves and stems). All fragments of the stem apices, green leaves and stems regenerated within 10 days of culture while some fragments of yellow-green leaves (two 2014 fragments and one 2015 fragment) and brown leaves (three 2014 fragments and three 2015 fragments) died. Fragments of stem, stem apices and green leaves regenerated more quickly, produced longer protonemata and more shoots as compared to fragments of yellow-green and brown leaves. These differences were statistically significant but there was no difference in regeneration between the fragments from 2014 and 2015. Differential regeneration and proliferation of different plant fragments has important implications for the clonal propagation of S. caninervis in the Gurbantunggut Desert.  相似文献

Although the terrestrial and temperate orchids–fungal biology have been largely explored, knowledge of tropical epiphytic orchids–fungus relationships, especially on the ecological roles imparted by non-mycorrhizal fungal endophytes, is less known. Exploitation of the endophytic fungal mycobiota residing in epiphytic orchid plants may be of great importance to further elucidate the fungal ecology in this special habitat as well as developing new approaches for orchid conversations. The composition of fungal endophytes associated with leaves, stems and roots of an epiphytic orchid (Dendrobium nobile), a famous Chinese traditional medicinal plant, was investigated. Microscopic imaging, culture-dependant method and molecular phylogeny were used to estimate their entity and diversity. Totally, there were 172 isolates, at least 14 fungal genera and 33 different morphospecies recovered from 288 samples. Ascomycetes, coelomycetes and hyphomycetes were three major fungal groups. There were higher overall colonization and isolation rates of endophytic fungi from leaves than from other tissues. Guignardia mangiferae was the dominant fungal species within leaves; while the endophytic Xylariaceae were frequently observed in all plant tissues; Colletotrichum, Phomopsis and Fusarium were also frequently observed. Phylogenetic analysis based on ITS gene revealed the high diversity of Xylariacea fungi and relatively diverse of non-Xylariacea fungi. Some potentially promising beneficial fungi such as Clonostachys rosea and Trichoderma chlorosporum were found in roots. This is the first report concerning above-ground and below-ground endophytic fungi community of an epiphytic medicinal orchid, suggesting the ubiquitous distribution of non-mycorrhizal fungal endophytes in orchid plants together with heterogeneity and tissue specificity of the endophyte assemblage. Possible physiological functions played by these fungal endophytes and their potential applications are also discussed briefly. The new species Ligularia zhengyiana from the Hubei Province, China, is described and illustrated. It is characterized by dense blackish purple pilose hairs on the stems and by coarsely triangular‐dentate leaves. Ligularia zhengyiana is similar to L. fischeri (Ledeb.) Turcz., but L. zhengyiana has dense long blackish purple hairs on stems, while L. fischeri is glabrous or has short yellowish‐brown hairs on stems. In addition, the pappus of L. zhengyiana is white, while that of L. fischeri is yellowish‐brown.  相似文献

The environmental release of genetically engineered (transgenic) plants may be accompanied by ecological effects including changes in the plant-associated microflora. A field release of transgnic potato plants that produce the insecticidal endotoxin ofBacillus thuringiensis var.tenebrionis (Btt) was monitored for changes in total bacterial and fungal populations, fungal species diversity and abundance, and plant pathogen levels. The microflora on three phenological stages of leaves (green, yellow and brown) were compared over the growing season (sample days 0, 21, 42, 63 and 98) for transgenic potato plants, commercial Russet Burbank potato plants treated with systemic insecticide (Di-Syston) and commercial Russet Burbank potato plants treated with microbialBtt (M-Trak). In addition, plant and soil assays were performed to assess disease incidence ofFusarium spp.,Pythium spp.,Verticillium dahliae, potato leaf roll virus (PLRV) and potato virus Y (PVY). Few significant differences in phylloplane microflora among the plant types were observed and none of the differences were persisent. Total bacterial populations on brown leaves on sample day 21 and on green leaves on sample day 42 were significantly higher on the transgenic potato plants. Total fungal populations on gree leaves on sample day 63 were significantly different among the three plant types; lowest levels were on the commerical potato plants treated with systemic insecticide and highest levels were on the commercial potato plants treated with microbialBtt. Differences in fungal species assemblages and diversity were correlated with sampling dates, but relatively consistent among treatments.Alternaria alternata, a common saprophyte on leaves and in soil and leaf litter, was the most commonly isolated fungus species for all the plant treatments. Rhizosphere populations of the soilborne pathogensPythium spp.,Fusarium spp. andV. dahliae did not differ between the transgenic potato plants and the commercial potato plants treated with systemic insecticide. The incidence of tuber infection at the end of the growing season by the plant pathogenV. dahliae was highest for the transgenic potato plants but this difference was related to longer viability of the transgenic potato plants. This difference in longevity between the transgenic potato plants and the commercial + systemic insecticide potato plants also made comparison of the incidence of PVY and PLRV problematic. Our results indicate that under field conditions the microflora of transgenicBtt-producing potato plants differed minimally from that of chemically and microbially treated commerical potato plants.  相似文献

该文采用传统形态学方法结合r DNA-ITS序列分析,对我国重要药用植物罗汉果中的内生真菌进行了鉴定并研究其多样性。结果表明:采用组织培养法从罗汉果健康植株中共分离得到150株内生真菌,包括罗汉果中雌株的内生真菌96株、雄株的内生真菌54株。122株内生真菌经形态学结合r DNA-ITS序列分析鉴定为9个属,均归属为子囊菌门,包含座囊菌纲(Dothideomycetes)和子囊菌纲(Sordariomycetes)。其中,座囊菌纲(Dothideomycetes)真菌包含3科、3属;子囊菌纲(Sordariomycetes)真菌包含6科、6属。优势属为刺盘孢属(Colletotrichum)和镰刀菌属(Fusarium)。罗汉果雌、雄植株不同组织中内生真菌的定殖率及分离率的变化规律均不相同,雌株中以根中内生真菌的定殖率和分离率最高,叶片中的最低;在雄株中以叶片中的定殖率和分离率最高,根中的最低。不同菌株在雌、雄两种植株的不同组织中的分布均不同,结合内生真菌群落组成的相似性比较结果,表明部分内生真菌对罗汉果雌株和雄株,以及同一植株中的不同组织均具有偏好性。不同组织中内生真菌的多样性指数在0.11~0.69的范围,其中雌株根部的内生真菌多样性指数最高。以上研究结果为后期探究内生真菌与罗汉果互作关系奠定了基础。  相似文献

为探讨黑老虎(Kadsura coccinea)根际土壤和组织内生真菌菌群的组成及其生态功能,该研究采用ITS高通量测序技术对成熟黑老虎(根、茎、叶)内生真菌及根际土壤真菌群落结构、多样性和生态功能进行了分析。结果表明:(1)从12个样品中共获得2 241个可操作分类单元(OTU),涉及10门、41纲、95目、212科、367属,内生真菌(根、茎、叶)和根际土壤真菌OTU数分别为386、536、258、1 435个,其中共有的OTU为18个。在门水平上,黑老虎内生真菌及根际土壤真菌优势群落均为子囊菌门和担子菌门,其中子囊菌门在叶和茎中占比分别高达96.99%和95.37%;在属水平上,黑老虎根际土壤真菌中腐生真菌被孢霉属占比较高(为13.5%),叶和茎等生长旺盛的组织中子囊菌门未分类属和痂囊腔菌属占比较高。(2)α多样性分析结果显示,黑老虎根际土壤真菌群落的丰度和多样性明显高于内生真菌,茎中内生真菌丰度显著高于根和叶,而根、茎和叶组织间内生真菌多样性差异不显著;PCoA分析结果显示,叶和茎的真菌群落结构相似性更高。(3)利用FUNGuild数据库进行的功能预测分析结果显示,黑老虎根际土...  相似文献

通过用两种传统培养基(PDA、SDA)分离方法对5个自然野生分布的桃儿七群落(分布于青海省、甘肃省和四川省)茎叶组织内的内生真菌多样性进行研究,并用形态学与分子生物学的方法鉴定菌株。实验结果显示,720个茎叶组织块中共分离到141株内生真菌。依据真菌在培养基上的形态初步划分为52个分类单元,经鉴定归属于19属,其中茎组织中16属叶组织中6属,桃儿七茎叶组织中的真菌优势属为拟青霉属,相对分离频率为26.34%。5个采样点间内生真菌的香浓维纳多样性指数为0.71—1.41,Sorenson相似性系数为0.13—0.50,定殖率为14.58%—28.47%。通过对PDA、SDA两种培养基以及茎、叶组织的定殖率进行统计,结果显示:PDA(17.50%)SDA(21.67%),茎(29.72%)叶(9.44%)。研究结果表明,桃儿七茎叶组织中内生真菌的多样性较低,不同采样点间宿主植物内内生真菌群落的相似性较低,真菌群落结构存在差异。研究为进一步扩大从桃儿七中筛选产鬼臼毒素等活性物质内生真菌提供了一种新的思路。  相似文献

Fungal endophytes of 17 genera were found in stems of four native Gossypium species (G. australe, G. bickii, G. nelsonii, G. sturtianum) collected from inland areas in Queensland, the Northern Territory, and South Australia in 2001. Phoma, Alternaria, Fusarium, Botryosphaeria, Dichomera, and Phomopsis were common, accounting for 58, 18, 11, 3, 1, and 1 % of the 281 recovered isolates, respectively, and occurring in 47, 29, 19, 5, 5, and 4 % of the 79 sampled populations. Among the four Gossypium species in Queensland and the Northern Territory, Alternaria spp. and Fusarium spp. had the greatest recovery frequency in G. bickii stems. The recovery frequencies of Phoma spp. and Alternaria spp. were significantly greater in the G. sturtianum stems collected from South Australia than in those from Queensland and the Northern Territory. Pathogenicity of 42 representative isolates was tested on cultivated cotton (G. hirsutum). All isolates caused some localized discoloration in stem tissue when inoculation was conducted with the stem puncturing method, but none of the isolates could induce any foliar symptoms during the five-week experimental period by either inoculation method (root dipping or stem puncturing), suggesting that the endophytic fungi of native Gossypium species are unlikely sources of cotton pathogens.  相似文献

This study builds upon past work investigating seedling leaf physiology and structure among tropical trees. We seek to explain how related and unrelated species and genera co‐occur in relation to varying amounts of shade. Seedlings of eight Sri Lankan rain forest tree species in three genera (Dipterocarpus, Mesua, Shorea section Doona) were grown for 2 years in four treatments that simulated a variety of shade environments across the understorey of a rain forest. All three genera comprise major canopy tree species of mixed dipterocarp forest, a widespread and important Asian tropical forest type. Compared with the other genera, Dipterocarpus spp. had the largest leaves, the thinnest leaf blades and relatively high rates of stomatal conductivity across all shade treatments, making them water‐loving species sensitive to droughty soils. Mesua spp. had intermediate sized leaves, with the thickest leaf blades and palisade mesophyll layers, the highest stomatal densities, the smallest aperture sizes and the lowest rates of stomatal conductance, making them the most water conservative. Shorea spp. were generally intermediate in blade and palisade mesophyll dimensions between Dipterocarpus spp. and Mesua spp., but they had the smallest leaves. Greater differences among genera than among species within genera were apparent, but species differences within genera were also apparent. 对我国古老特有植物青檀叶片进行内生和附生真菌的研究,以了解青檀叶片内生和附生真菌的组成特点和探讨内生和附生真菌菌群之间的可能联系,为研究真菌资源多样性、植物附生和内生真菌的相互演化关系及真菌与宿主植物协同进化等提供有益参考资料。研究结果表明,从健康的青檀叶片获得可培养内生真菌839株,附生真菌1857株,共计2696株,鉴定其分属于4目,5科,43属。在目的分类水平上,内生和附生真菌均以丛梗孢目Moniliales为优势菌群,分别占90.23%和92.51%;在科的水平上,内生真菌以暗梗孢科Dematiaceae和丛梗孢科Moniliaceae为优势菌群,分别占47.56%和42.67%,附生真菌以丛梗孢科Moniliaceae和暗梗孢科Dematiaceae为优势菌群,分别占67.04%和25.47%;在属的水平上,内生真菌以黑团孢属Periconia和青霉属Penicillium为优势菌群,分别占31.47%和10.73%,附生真菌以小球霉属Glomerularia、膝葡孢属Gonatobotrys和青霉属Penicillium为优势菌群,分别占20.03%、13.95%和12.22%。青檀叶片内生真菌和附生真菌均存在的菌群数量达到23个属,占53.49%。内生真菌特有的属有6个,共分离19株,占0.70%,附生真菌特有的属有14个,共分离120株,占4.45%。内生真菌的Shannon-Wiener index(H')多样性指数(2.44)和Margalef index(R)丰富度指数(2.88)分别小于附生真菌ShannonWiener index(H')多样性指数(2.57)和Margalef index(R)丰富度指数(3.32),但两者的Evenness index(E)均匀度指数几乎相等。青檀叶片内生和附生真菌菌群组成具有较高的相似性,相似性系数达0.70。通过Fisher's exact test分析表明青檀叶片内生和附生真菌菌群组成无明显差异(P=0.072)。  相似文献

The diet of a group of six brown howlers was studied weekly during 12 months in a reserve of 250 ha of secondary, mesophytic, semi-deciduous forest. The phenology of 186 trees of 72 species and 29 families was monitored simultaneously. Scan sampling was used to record the diet from dawn until dusk on a total of 60 days of observation, yielding 718 hr of animal-observer contact and 2,943 feeding scans. The diet was composed of leaves (73%), flowers (12%), and fruits (5%), from 68 identified plant species.Celtis iguanae, Cassia ferruginea, andInga spp. were the main food sources, accounting for approximately 50% of the diet. Young leaves (59%) were preferred to mature leaves (31%), trees contributing 56% and lianas 41% of the leaf diet. The ingestion of young leaves was correlated to the availability of these items, however, the correlations were not significant for flowers and fruits. The diet was poorer in fruits and richer in young leaves of lianas in comparison to other howler monkey species studies, probably as a consequence of the liana abundance in this forest fragment.  相似文献

Ascomycota are among the fungi that cause serious willow diseases in all natural habitats worldwide. This study was conducted to determine if basket willow used in green wall structures (GWS) built of willow stems were infected by potentially important fungal diseases or their antagonists in urban areas of eastern Canada. In total, 13 different phenotypic genera belonging to eight families of ascomycetous fungi were isolated and identified according to their sexual and/or asexual forms. Venturia pathogenic species complex were represented by three different anamorphs: Fusicladium, Fusicladium-Cladosporium, and Pollaccia as anamorph. They were responsible for the highest incidence value on leaves (IF > 15%). Cryptodiaporthe, Drepanopeziza, and Glomerella dominated on bark (IF > 5%). A significantly higher incidence value of fungal communities was found on first year than on second year GWS. The correspondence analysis using χ2 distance showed that communities of potentially pathogenic species are closely related to diseased plants, while healthy plants often contain biocontrol species such as Cladobotryum mycoparasite on healthy bark and Alternaria sp. antagonist on healthy leaves. The phylogenetic positions of the different fungal taxa and their relationship have been revealed by use of PCR amplified internal transcriber spacer (ITS) region of rDNA.  相似文献

Summary Mosses account for 75% of the annual phosphorus accumulation in aboveground parts of an Alaskan black spruce forest, although they comprise only 17% of the phosphorus pool in aboveground vegetation. Sphagnum subsecundum and feathermosses (Hylocomium splendens and Pleurozium schreberi) have a higher capacity to absorb phosphate than do the fine roots of black spruce (Picea mariana) that are situated beneath the moss layer. In three of the four moss species studied, phosphate absorption capacity increases with increasing age of green tissue and decreases with increasing age of brown tissue. In the two feathermosses, which acquire moisture primarily from the air, and in Sphagnum, phosphate absorption is more rapid in green than in brown tissue. In contrast, the endohydric moss Polytrichum commune, which transports water through stem tissue from soil, absorbs phosphate most rapidly from stems in mineral soil. Two treatments designed to reduce activity of mycorrhizae (cutting of roots extending beneath the moss carpet or application to the moss surface of a fungicide that kills mycorrhizal hyphae) tended to increase phosphate retention by mosses and reduce phosphate transfer out of the experimental plots. This suggests that mycorrhizae are an important avenue of phosphorus movement out of the moss carpet and a means by which the black spruce competes with the overlying mosses for nutrients.  相似文献

链霉菌是自然界中大量存在的一类微生物,具有多种多样的功能,其基内菌丝多核有间隔,气生菌丝上附着孢子链,从孢子萌发到孢子释放完成整个生命周期。链霉菌以菌丝体的形式定殖于多种植物体的根、茎、叶等部位而发挥功能,能分泌多种具有促进植物生长与生物防治功能的代谢物。对近几年链霉菌在提升植物营养吸收、促进植物生长、增强植物应对逆境能力、改善土壤结构、恢复污染水体等方面的研究进行了综述,并对今后链霉菌的研究方向和应用前景进行了展望。  相似文献
