We discovered that extracts of the female sex pheromone gland of the carpenterworm moth Isoceras sibirica Alpheraky, a pest of Asparagus officinalis Linn., contained (Z)-7-tetradecen-1-ol (Z7-14:OH), (Z)-9-tetradecen-1-ol (Z9-14:OH), (Z)-7-tetradecenyl acetate (Z7-14:Ac), (Z)-9-tetradecenyl acetate (Z9-14:Ac), and (Z)-9-hexadecadecenyl acetate (Z9-16:Ac). The average levels of the chemicals in a single sex pheromone gland of a calling moth were (0.71 +/- 0.24) ng, (1.42 +/- 0.44) ng, (4.36 +/- 0.32) ng, (8.71 +/- 0.26) ng, and (0.82 +/- 0.38) ng, respectively. The electroantennography (EAG) analysis of these chemicals and their analogues demonstrated that Z9-14:Ac triggered significantly the male EAG response. Traps with rubber septa lure impregnated with Z9-14:Ac (500 microg/septum), Z7-14:Ac (250 microg/septum), and Z9-16:Ac (50 microg/septum) were more effective in catching male moths than traps with other baits or virgin females. Addition of Z7-14:OH and Z9-14:OH to rubber septa did not enhance the efficiency of the trap.  相似文献

New geometric isomers, methyl (9Z,11Z)-13-hydroperoxy-9,11-octadecadienoate and methyl (10Z,12Z)-9-hydroperoxy-10,12-octadecadienoate, were proved to be present in methyl linoleate hydroperoxide produced by autoxidation. They were identified from their UV, MS, and 1H-NMR spectra after conversion to the corresponding oxo derivatives: methyl (9Z,11Z)-13-oxo-9,11-octadecadienoate and methyl (10Z,12Z)-9-oxo-10,12-octadecadienoate. The behavioural significance of (Z)-9-tetradecenal to male H. assulta was tested by comparing the number of moths attracted to lures containing a standard synthetic female sex pheromone with lures in which (Z)-9-tetradecenal was also added. The standard pheromone mixture used contained 1000 g (Z)-9-hexadecenal, 50 g (Z)-11-hexadecenal, 300 g (Z)-9-hexadecenyl acetate and 15 g (Z)-11-hexadecenyl acetate impregnated on rubber septa. Addition of (Z)-9-tetradecenal to the standard pheromone was shown to significantly reduce the caught of male H. assulta when added in amounts greater than 10 g or 1% of the major pheromone component in both field and net-house experiments. The reduction in catch was found to be dependent on the quantity of (Z)-9-tetradecenal added to the standard pheromone. Chemical and electrophysiological analyses and field trials were used to identify the female sex pheromone of Copitarsia decolora (Guenée) (Lepidoptera: Noctuidae). Coupled gas chromatographic-electroantennographic detection (GC-EAD) analysis of the female gland extract showed the presence of two EAD-active peaks, which were identified by GC-mass spectrometric (MS) analysis as (Z)-9-tetradecenyl acetate (Z9-14:Ac) and (Z)-9-tetradecenol (Z9-14:OH). The field evaluation of the EAD-active compounds indicated that traps baited with either Z9-14:Ac or Z9-14:OH caught few males. In contrast, traps baited with the binary blend of both components caught significantly more males than traps baited with the single compounds. Captures in traps baited with a mixture of Z9-14:Ac and Z9 -14:OH at 4:1, 10:1, and 100:1 ratios were not significantly different from the catches in traps baited with virgin females. Few males were captured in traps baited with a blend of Z9-14:Ac and Z9-14:OH at 1:4, 1:10, and 1:100 ratios.  相似文献

The effects on olfaction of N-ethylmaleimide (NEM), a specificreagent of free sulfhydryl groups, were studied in the mothMamestra brassicae. The antennae of male M.brassicae bear twotypes of specialist receptor neurons involved in pheromone communication.One type is tuned to (Z)-11-hexadecenyl acetate (Z11-16:Ac),the main pheromone component; the second type is tuned to (Z)-9-tetradecenylacetate (Z9-14:Ac), an interspecific inhibitor not producedby the females of this species. Vapours of NEM irreversiblyinhibited the electro-antennographic (EAG) responses to Z11-16:Acand Z9-14:Ac. When Zll-16:Ac was applied before and during NEMtreatment, the responses to Z9-14:Ac were preserved and someprotection was observed in the responses to Zll-16:Ac. In return,Z9-14:Ac partially prevented the disappearance of responsesto Zll-16:Ac but not to Z9-14:Ac. A third compound, hexadecylacetate (16:Ac), found in the pheromone gland, but not detectedby the antennal receptors, did not prevent the inhibition causedby NEM.  相似文献

The delta 5,9 fatty acids (5Z,9Z)-5,9-hexadecadienoic acid, (5Z,9Z)-5,9-nonadecadienoic acid, and (5Z,9Z)-5,9-eicosadienoic acid were synthesized for the first time in four steps (9-12% overall yield) starting from commercially available 2-(2-bromoethyl)-1,3-dioxolane. The synthetic approach provided enough material to corroborate the structure and stereochemistry of (5Z,9Z)-5,9-nonadecadienoic acid which was recently identified in the flowers of Malvaviscus arboreus (Malvaceae). The novel phospholipids 1-hexadecanoyl-2-[(5Z,9Z)-5,9-eicosadienoyl]-sn-glycer o-3-phosphocholine and 1-octadecanoyl-2-[(5Z,9Z)-5,9-eicosadienoyl]-sn- glycero-3-phosphocholine were also synthesized from commercially available L-alpha-phosphatidylcholine (egg yolk) and characterized by positive ion electrospray mass spectrometry. These are the first examples of unsymmetrical phospholipids with saturated fatty acids at the sn-1 position and delta 5,9 fatty acids at the sn-2 position.  相似文献

Two Helicoverpa species, H. armigera and H. assulta use (Z)-11-hexadecenal and (Z)-9-hexadecenal as their sex attractant pheromone components but in opposite ratios. Since both female and male interspecific hybrids produced by female H. assulta and male H. armigera have been obtained in our laboratory, we can make a comparative study of sex pheromone composition and biosynthesis in the two species and their hybrid. With GC and GC-MS analyses using single gland extracts, the ratio of (Z)-9-hexadecenal to (Z)-11-hexadecenal was determined as 2.1:100 in H. armigera, and 1739:100 in H. assulta. The hybrid has a ratio of 4.0: 100, which is closer to that of H. armigera, but significantly different from H. armigera. We investigated pheromone biosynthesis with labeling experiments, using various fatty acid precursors in H. armigera, H. assulta and the hybrid. In H. armigera, (Z)-11-hexadecenal is produced by delta11 desaturation of palmitic acid, followed by reduction and terminal oxidation; (Z)-9-hexadecenal results from delta11 desaturation of stearic acid, followed by one cycle of chain shortening, reduction and terminal oxidation. delta11 desaturase is the unique desaturase for the production of the two pheromone components. In our Chinese strain of H. assulta, palmitic acid is used as the substrate to form both the major pheromone component, (Z)-9-hexadecenal and the minor one, (Z)-11-hexadecenal. Our data suggest that delta9 desaturase is the major desaturase, and delta11 desaturase is responsible for the minor component in H. assulta, which is consistent with previous work. However, the weak chain shortening acting on (Z)-9 and (Z)-11-octadecenoic acid, which is present in the pheromone glands, does occur in this species to produce (Z)-7 and (Z)-9-hexadecenoic acid. In the hybrid, the major pheromone component, (Z)-11-hexadecenal is produced by delta11 desaturation of palmitic acid, followed by reduction and terminal oxidation. The direct fatty acid precursor of the minor component, (Z)-9-hexadecenoic acid is mainly produced by delta9 desaturation of palmitic acid, but also by delta11 desaturation of stearic acid and one cycle of chain shortening. The greater relative amounts of (Z)-9-hexadecenal in the hybrid are due to the fact that both palmitic and stearic acids are used as substrates, whereas only stearic acid is used as substrate in H. armigera. The evolutionary relationships between the desaturases in several Helicoverpa species are also discussed in this paper.  相似文献

Analysis of sex pheromone glands of the apple leafroller Bonagota cranaodes Meyrick by gas chromatography coupled with mass spectrometry or electroantennographic detection showed the presence of 14 structurally related acetates and alcohols of the chain length 10-18, including the main pheromone component (E,Z)-3,5-dodecadienyl acetate (E3,Z5-12Ac). Male antennae responded to the main compound, its Z,Z isomer, (E,Z) -3,5-tetradecadienyl acetate (E3,Z5-14Ac), and the monoenes (Z)-5-dodecenyl acetate (Z5-12Ac) and (Z)-9-hexadecenyl acetate (Z9-16Ac). Traps baited with a four-component blend of E3,Z5-12Ac, Z5-12Ac, E3,Z5-14Ac, and Z9-16Ac in a 100:5:5:100 ratio were significantly more attractive than the main compound alone. This improved trap lure is more suitable for monitoring population densities of B. cranaodes, and for detection of the onset of the seasonal flight period. A more complete pheromone blend is of importance also with respect to current attempts to develop mating disruption for control of this major pest of apple in Brazil.  相似文献

Using gas chromatography the relative amounts of (Z)-9-tricosene (muscalure) and some other hydrocarbons on the cuticle of 1- to 20-day-old houseflies (Musca domestica L.) from different strains were determined. Flies from a WHO strain, in culture since 1961, and first-generation laboratory-cultured flies from two wild-type strains from a poultry breeding and a cow-house with pigsty, respectively, were compared. On WHO females hydrocarbons with 23–25 C atoms constituted about 65% of the total hydrocarbons, whereas on wild-type females less than 2% of these compounds was present. (Z)-9-tricosene comprised up to 20–30% of the total hydrocarbons on 5- to 20-day-old WHO females, whereas less than 0.5% (Z)-9-tricosene was present on the wild-type females. We also compared the amounts of (Z)-9-tricosene and some other hydrocarbons on female houseflies, kept in culture in the laboratory for several generations. It appeared that whereas on first-generation wild-type females hardly or no (Z)-9-tricosene could be detected, the amounts of this substance had increased considerably after some tens of generations in the laboratory. It is suggested that this was due to selection in subsequent generations of high-density populations. Production of (Z)-9-tricosene and of tricosane was shown to be closely linked. Selection did not affect the production of other cuticular hydrocarbons by the females. It is suggested that in mixed populations (both sexes together in a cage) in the course of time (Z)-9-tricosene is transferred from females to males and (Z)-9-heptacosene from males to females. It is concluded that reproductive ability of houseflies does not primarily depend on the amounts of (Z)-9-tricosene on females, although higher amounts of this substance may increase contacts between males and females.  相似文献

Pheromone gland extracts from calling female Plodia interpunctella contained at least seven compounds that consistently elicited electroantennographic responses from male antennae upon gas chromatographic analysis. Three of these compounds were found to be the previously identified gland constituents, i.e., (Z,E)-9,12-tetradecadienyl acetate (Z9,E12-14:OAc), (Z,E)-9,12-tetradecadienal (Z9,E12-14:Ald) and (Z,E)-9,12-tetradecadienol (Z9,E12-14:OH). A fourth EAD-active compound was identified as (Z)-9-tetradecenyl acetate (Z9-14:OAc). The homologue (Z)-11-hexadecenyl acetate (Z11-16:OAc) was also identified in the extracts, but showed no EAD activity. The identity of all five compounds was confirmed by comparison of GC retention times and mass spectra with those of synthetic standards. In flight tunnel tests there were no significant differences in response of male P. interpunctella to the bait containing all four EAD-active compounds and the responses to female gland extacts. A behavioural assay of different two-compound blends in the flight tunnel showed that only addition of the corresponding aldehyde to the major pheromone component Z9,E12-14:OAc raised the male response. A subtractive assay, however, revealed that the exclusion of any of the compounds from the complete four-compound blend reduced its activity significantly. We thus conclude that the female-produced sex pheromone of P. interpunctella consists of at least four components, i.e., Z9,E12-14:OAc, Z9,E12-14:Ald, Z9,E12-14:OH and Z9-14:OAc.In a field trapping test performed in a storage facility, the four-component blend attracted significantly more males of P. interpunctella than traps baited with Z9,E12-14:OAc alone. In contrast, the highest number of Ephestia kuehniella males was found in the traps baited with this major component, suggesting that the secondary pheromone components contribute to the species specificity of the blend.  相似文献

Guava (Psidium guajava) hydroperoxide lyase (HPL) preparations were incubated with [1-(14)C](9Z,11E,13S,15Z)-13-hydroperoxy-9,11,15-octadecatrienoic acid for 1 min at 0 degrees C, followed by rapid extraction/trimethylsilylation. Analysis of the trimethylsilylated products by gas chromatography-mass spectrometry and radio-high-performance liquid chromatography revealed a single predominant (14)C-labelled compound, identified by its (1)H-nuclear magnetic resonance, ultraviolet and mass spectra as the trimethylsilyl ether/ester of (9Z,11E)-12-hydroxy-9,11-dodecadienoic acid. Longer time incubations afford smaller yield of this enol due to its partial tautomerization into (9Z)-12-oxo-9-dodecenoic acid. The data obtained demonstrate that formation of (9Z)-12-oxo-9-dodecenoic acid in the HPL reaction is preceded by unstable enol oxylipin, and further suggest that hemiacetals are the true products of HPL catalysis.  相似文献

Four EAG-active components were detected in GC-EAG analyses of hexane extracts from virgin Etiella behrii (Zeller) (Lepidoptera: Pyralidae) females. These components were identified as dodecyl acetate (12:Ac), (E)-9-dodecenyl acetate (E9-12:Ac), either (Z)-9- or (E)-11-tetradecenyl acetate (Z9- or E11-14:Ac), and (Z)-11-tetradecenyl acetate (Z11-14:Ac) by comparison of retention indices on both nonpolar and polar GC columns. The available extract was insufficient for further GC-MS or other chemical analyses (<0.2 ng/female). In field tests carried out in East Java, a 10:90 mixture of E9-12:Ac and Z11-14:Ac showed attractiveness to male moths and addition of 12:Ac and/or E11-14:Ac significantly increased the trap catches while addition of Z9-14:Ac showed no significant effect. Maximum attraction was obtained with 5.35 or 10.7 g/rubber septum of a mixture of E9-12:Ac, Z11-14:Ac, 12:Ac and E11-14:Ac at the ratio of 10:90:0.7:6.3, respectively. The role of pheromone blends in species discrimination between E. behrii and the related E. zinckenella (Treitschke) is discussed.  相似文献

槐庶尺蛾Semiothisa cinerearia Bremer et Grey (鳞翅目: 尺蛾科)是我国北方国槐Sophora japonica L.上的重要食叶害虫。本研究的主要目的是阐明槐庶尺蛾性信息素成分化学结构的绝对构型, 为在城市地区环境友好地防控槐庶尺蛾的为害提供一种新方法。经与标准品比较气相色谱保留时间和质谱特征离子, 从槐庶尺蛾处女雌蛾(2-3日龄)性信息素腺体溶剂提取物中检测到顺6, 顺9-顺-3, 4-环氧-十七碳二烯烃和顺3, 顺6, 顺9-3, 6, 9-十七碳三烯烃2种成分, 在腺体中以100∶4.8±1.3 (N=12)的比例存在。槐庶尺蛾性信息素腺体提取物进一步经手性毛细管色谱柱(CycloSil-B, 30 m×0.25 mm×0.25 μm 液膜厚)分离, 在优化的程序升温条件下发现腺体成分顺6, 顺9-顺-3, 4-环氧-十七碳二烯烃具有3R, 4S的绝对构型。两种合成的对映异构体混合物顺6, 顺9-3R, 4S-环氧-十七碳二烯烃和顺6, 顺9-3S, 4R-环氧-十七碳二烯烃以1.28∶1的比例加到腺体提取物中, 比例变为1.55∶1。根据这一分析, 腺体成分顺6, 顺9-顺-3, 4-环氧-十七碳二烯烃进一步确认具有3R, 4S的绝对构型。该研究结论将为生产上研发高效的槐庶尺蛾性信息素诱芯奠定坚实的基础。  相似文献

Sex pheromones of many Lepidopteran species have relatively simple structures consisting of a hydrocarbon chain with a functional group and usually one to several double bonds. The sex pheromones are usually derived from fatty acids through a specific biosynthetic pathway. We investigated the incorporation of deuterium-labeled palmitic and stearic acid precursors into pheromone components of Helicoverpa zea and Helicoverpa assulta. The major pheromone component for H. zea is (Z)11-hexadecenal (Z11-16:Ald) while H. assulta utilizes (Z)9-hexadecenal (Z9-16:Ald). We found that H. zea uses palmitic acid to form Z11-16:Ald via delta 11 desaturation and reduction, but also requires stearic acid to biosynthesize the minor pheromone components Z9-16:Ald and Z7-16:Ald. The Z9-16:Ald is produced by delta 11 desaturation of stearic acid followed by one round of chain-shortening and reduction to the aldehyde. The Z7-16:Ald is produced by delta 9 desaturation of stearic acid followed by one round of chain-shortening and reduction to the aldehyde. H. assulta uses palmitic acid as a substrate to form Z9-16:Ald, Z11-16:Ald and 16:Ald. The amount of labeling indicated that the delta 9 desaturase is the major desaturase present in the pheromone gland cells of H. assulta; whereas, the delta 11 desaturase is the major desaturase in pheromone glands of H. zea. It also appears that H. assulta lacks chain-shortening enzymes since stearic acid did not label any of the 16-carbon aldehydes.  相似文献

The cossid moth (Coryphodema tristis) has a broad range of native tree hosts in South Africa. The moth recently moved into non-native Eucalyptus plantations in South Africa, on which it now causes significant damage. Here we investigate the chemicals involved in pheromone communication between the sexes of this moth in order to better understand its ecology, and with a view to potentially develop management tools for it. In particular, we characterize female gland extracts and headspace samples through coupled gas chromatography electro-antennographic detection (GC-EAD) and two dimensional gas chromatography mass spectrometry (GCxGC-MS). Tentative identities of the potential pheromone compounds were confirmed by comparing both retention time and mass spectra with authentic standards. Two electrophysiologically active pheromone compounds, tetradecyl acetate (14:OAc) and Z9-tetradecenyl acetate (Z9-14:OAc) were identified from pheromone gland extracts, and an additional compound (Z9-14:OH) from headspace samples. We further determined dose response curves for the identified compounds and six other structurally similar compounds that are common to the order Cossidae. Male antennae showed superior sensitivity toward Z9-14:OAc, Z7-tetradecenyl acetate (Z7-14:OAc), E9-tetradecenyl acetate (E9-14:OAc), Z9-tetradecenol (Z9-14:OH) and Z9-tetradecenal (Z9-14:Ald) when compared to female antennae. While we could show electrophysiological responses to single pheromone compounds, behavioral attraction of males was dependent on the synergistic effect of at least two of these compounds. Signal specificity is shown to be gained through pheromone blends. A field trial showed that a significant number of males were caught only in traps baited with a combination of Z9-14:OAc (circa 95% of the ratio) and Z9-14:OH. Addition of 14:OAc to this mixture also improved the number of males caught, although not significantly. This study represents a major step towards developing a useful attractant to be used in management tools for C. tristis and contributes to the understanding of chemical communication and biology of this group of insects.  相似文献

Two cDNAs encoding acyl-CoA Z9-desaturase from the fat body and Z10-desaturase from the pheromone gland of the greenhead leafroller moth, Planotortrix octo, were obtained by RACE PCR. The Z9-desaturase (Pocto-Z9) cDNA spans 2291 nt with an ORF encoding a 352 amino-acid protein, which has 65% identity to Trichoplusia ni Delta 9 desaturase (Tni-Z9). The Z10-desaturase (Pocto-Z10) cDNA spans 2777 nt with an ORF encoding a protein with 356 amino acids. Pocto-Z10 shows lower identity to Pocto-Z9 and Tni-Z9 (48 and 46%, respectively) and relatively higher identity to the Delta 11 desaturases of T. ni and Helicoverpa zea (57 and 56%, respectively). The ORFs of these two P. octo cDNAs were constructed into an expression vector, YEpOLEX, that complemented the unsaturated fatty acid (UFA) auxotrophy of a desaturase-deficient ole1 strain of Saccharomyces cerevisiae. Expression of Pocto-Z9 produced a 5:2 ratio of Z9-16 and Z9-18 acids, with minor amounts (<4%) of Z9-14, Z9-15, and Z9-17 acids. Pocto-Z10 was successfully expressed in the YEpOLEX system when complemented with Z11-18:Me, and the major desaturase product proved to be Z10-16:Acid. The results confirm the regio- and stereo-selectivity of this unusual Delta 10 desaturase.  相似文献

电生理试验显示出:大螟Sesamia inferens(Walker)雄蛾触角对Z11-16:Ac(A)oZ11-16:OH(B)激起强烈的触角电位(EAG)反应,Z11-16:ALD?,E11-16:Ac(D),Z9-14:Ac(E)和Z7-12:Ac(F)四种化合物也能激起较强的EAG反应;A化合物的刺激阈比B高,此两种化合物5:5比例时,激起的反应最强烈。电生理的试验推测,雄蛾触角对A和B两种化合物的感受,是通过触角内两种不同类型的嗅觉感受器。A和B不同比例混合后,田间诱蛾试验证明:在9:1,8:2,5:5的比例时,其诱蛾量远远超过单独的A化合物;E加入A与B的混合物中,起抑制诱蛾的效果,上述试验表明,Z11-16:OH是大螟的性诱剂。  相似文献

The epicuticular and internal waxes of male and female houseflies were examined by capillary gas chromatography-mass spectrometry at closely timed intervals from emergence until day-6 of adulthood. New components identified included tricosan-10-one, 9,10-epoxyheptacosane, heptacosen-12-one, a series of odd-carbon numbered dienes from C31 to C39, several positional isomers of monoenes including (Z)-9- and 7-pentacosene and a number of methyl- and dimethylalkanes. (Z)-9-tricosene appears in internal lipids prior to appearing on the surface of the insect, suggesting that it is transported in the hemolymph to its site of deposition on the epicuticle. The large increases in the amount of (Z)-9-tricosene in females from day-2 until day-6 is compensated for by a concomitant decrease in (Z)-9-heptacosene. The C23 epoxide and ketone only appear in females after the production of (Z)-9-tricosene is induced, and are only abundant in epicuticular waxes, suggesting they are formed after (Z)-9-tricosene is transported to the cells which are involved in taking them to the surface of the insect. Mathematical analysis indicated that the time shift between internal production and external accumulation in females is more than 24 h. The divergence between male and female lipid production occurs at an early stage, when insects are less than one day old.  相似文献
