Cauliflower (Brassica oleracea L. botrytis) and broccoli (Brassicaoleracea L. italica) plants were grown in large pots in growthchambers for a range of temperatures (mean air temperaturesfrom 7.0-25.3 C) and irradi-ances (from 9.3-50.8 mol m–2d–1 or 4.7-25.4 MJ m–2 d–1). The extinctioncoefficient for PAR decreased with plant size reaching a valueof 0.55 in cauliflower and 0.45 in broccoli at plant leaf areasof 0.235 m2 and 0.227 m2, respectively. The leaf area expansionrate was unaffected by irradiance when compared at identicalleaf surface temperatures. The response of expansion rate tosurface temperature was fitted to a broken stick model witha base temperature of –0.7C and an optimum temperatureof 21.0C. The radiation conversion coefficient increased withair temperature below 13.8C and remained constant above this.The estimated radiation conversion coefficient above 13.8Cand for a PPFD of 20 mol m–2 d–1 was 0.77 g mol–1in cauliflower and 0.87 g mol–1 in broccoli. The radiationconversion coefficient declined with increasing irradiance levelfrom a maximum of 1.89 g mol–1 at near nil irradiancein cauliflower. Row crops are often inefficient in utilizing soil resources. One reason for this appears to be inefficient rooting of the available soil volume. Five experiments were performed to study the temporal and spatial root development of cauliflower (cv. Plana). The crop was grown with 60 cm between rows, and root development was followed in minirhizotrons placed under the crop rows, 15 cm, and 30 cm from the crop rows. Soil was sampled and analyzed for nitrate content at the final harvest and once during growth. In two of the experiments N fertilizer rate was varied and in two of the other experiments two cultivars were compared (cv. Plana and Siria).The rooting depth of cauliflower was found to be linearly related to temperature sum, with a growth rate of 1.02 mm day-1 °C-1. Depending on duration of growth this leads to rooting depths at harvest of 85–115 cm. Soil analysis showed that the cauliflower was able to utilize soil nitrogen down to at least 100 cm.With Plana differences in root growth between row and interrow soil were only observed during early growth, but with Siria this difference was maintained until harvest. However, at harvest both cultivars had depleted row and interrow soil nitrate equally efficient. Nitrogen fertilizer did not affect overall root development significantly.The branching frequency of actively branching roots was increased in all soil layers from about 6 to 10 branches cm-1 by increasing N fertilizer additions from 130 to 290 kg N ha-1. Increasing N supply increased the number of actively branching roots in the topsoil and reduced it in the subsoil.The average growth rate of the roots was always highest in the newly rooted soil layers, but fell during time. At 74 days after planting very few roots were growing in the upper 60 cm of the soil whereas 70% of the root tips observed in the 80–100 cm soil layer were actively growing. Within each soil layer there was a large variation in growth rate of individual root tips.  相似文献

Jasmonates are a new group of plant hormones; their roles on plant development are still little known. The aim of this work is to determine the action of jasmonates on cabbage, Brassica oleracea L. var Capitata, development both in in vitro cultured explants and in whole plants. Jasmonic acid (JA) enhanced nodal explant development when applied at 2–50 nM and inhibited it when supplied at 1250 and 6000 nM JA. Overall plant development was enhanced most under the 10 nM JA treatment; which significantly increased the explant shoot, leaf, and root dry weight. The root system of the explants cultured under the lower JA concentrations appeared more vigorous. Jasmonic acid also promoted the development of isolated in vitro cultured roots when applied at 2 and 10 nM. Root length and weight significantly increased, while concentrations 250 nM JA and over were detrimental. Isolated roots were progressively thicker as the JA concentration increased. Methyl jasmonate promoted both the below- and above-ground cabbage plant development when applied in a confined atmosphere at a concentration of only 1.225 nl.l–1 MJ: plants were higher and heavier, and showed an improved root system development. On the other hand, the 2.43 nl.l–1 MJ treatment decreased plant growth. The present work reveals a role for jasmonates as enhancers of in vitro and in vivo cabbage plant development. To our knowledge, no corresponding studies on the effects of jasmonates on whole plants have been previously published.  相似文献

A model of nitrogen uptake and distribution is presented whichdescribes these processes in relation to the amount of availablesoil nitrate and the rate of plant growth. Nitrogen uptake iseither sink or source limited. Sink limitation is based on maximumN-concentrations of plant compartments. The N-uptake model iscombined with a photosynthesis model based on the productivity-nitrogenrelationship at the single-leaf level. The model is parameterizedusing cauliflower as an example crop. Applied to an independentdata set, the combined model was able to predict leaf, stemand inflorescence nitrogen concentrations with correlation coefficientsbetween predicted and simulated values of 0.89, 0.66 and 0.86,respectively. The influence of nitrogen supply and light intensityon leaf nitrate-N could also be predicted with good accuracy(r2 = 0.87). Dry matter production based on the productivity-Nrelationship and the partitioning into leaf, stem and inflorescencewas also reproduced satisfactorily (r2 = 0.91, 0.93 and 0.92,respectively). By manipulation of various growth regulators and physical conditions,plants have been regenerated from excised roots, stem segments,cotyledons, leaves, and callus cultures of red cabbage (Brassicaoleracea var. capitata) grown under in vitro conditions. Shootbuds were induced on isolated root segments (1 cm long) culturedon Murashige and Skoog's medium and the frequency of bud formationwas greatly enhanced by the addition of kinetin (0.5 part 10–6).Callus obtained from the seeds, cotyledons, and hypocotyl segmentscultured on a medium fortified with 2,4-D (1 part 10–6),kinetin (0.1 part 10–6), and coconut milk (10%, v/v) hasbeen repeatedly subcultured. The callus is slow growing, andon transference to a kinetin (2 parts 10–6) and IAA (2parts 10–6) medium underwent morphogenesis to give riseto plants. The objective of this study was to determine if plant roots have to take up nitrate at their maximum rate for achieving maximum yield. This was investigated in a flowing-solution system which kept nutrient concentrations at constant levels. Nitrate concentrations were maintained in the range 20 to 1000 μM. Maximum uptake rate for both species was obtained at 100 μM. Concentrations below 100 μM resulted in decreases in uptake rate per cm root (inflow) for both spinach and kohlrabi by 1/3 and 2/3, respectively. However, only with kohlrabi this caused a reduction in N uptake and yield. Thus indicating that this crop has to take up nitrate at the maximum inflow. Spinach, however, compensated for lower inflows by enhancing its root absorbing surface with more and longer roots hairs. Both species increased their root length by 1/3 at low nitrate concentrations.  相似文献

Comparative analysis of the influence of the salts of Cd, Pb, Ni, Al, Cu and Zn at concentrations 10(-6)-10(-3) M on cytogenetic parameters of Allium cepa apical meristem cells was carried out. It was shown that these metal compounds could induce both the clastogenic and the aneugenic effects (including mitosis and cytokinesis disturbances). Based on ability to cause the chromosome aberrations the metal salts were arranged in such row of genotoxicity: ZnSO4 > Pb(CH3COO)2 > Al(NO3)3 approximately NiSO4 > CdCl2 > CuSO4. The investigated compounds of cadmium, lead, nickel and aluminum were found to be more effective aneugens than clastogens. According to our data NiSO4 had the strongest antimitotic activity (it increased the percentage of chromosome lagging of, multipolar anaphases, C-mitoses in 69.6 times, compared to the control). The ability of other metal salts to induce such abnormalities decreased in the range: CdCl2 > Al(NO3)3 > Pb(CH3COO)2 > CuSO4 > ZnSO4. Increasing of binuclear cells on the slides was revealed after treatment by all tested substances, however Ni sulfate was found as the strongest inducer of cytokinesis abnormalities. Presented results allowed to suggest that cytoskeleton could be a cell target for salts of some metals (especially for NiSO4).  相似文献

 Self-incompatibility (SI) in Brassica is a sporophytic system, genetically determined by alleles at the S-locus, which prevents self-fertilization and encourages outbreeding. This system occurs naturally in diploid Brassica species but is introduced into amphidiploid Brassica species by interspecific breeding, so that in both cases there is a potential for yield increase due to heterosis and the combination of desirable characteristics from both parental lines. Using a polymerase chain reaction (PCR) based analysis specific for the alleles of the SLG (S-locus glycoprotein gene) located on the S-locus, we genetically mapped the S-locus of B. oleracea for SI using a F2 population from a cross between a rapid-cycling B. oleracea line (CrGC-85) and a cabbage line (86-16-5). The linkage map contained both RFLP (restriction fragment length polymorphism) and RAPD (random amplified polymorphic DNA) markers. Similarly, the S-loci were mapped in B. napus using two different crosses (91-SN-5263×87-DHS-002; 90-DHW-1855-4×87-DHS-002) where the common male parent was self-compatible, while the S-alleles introgressed in the two different SI female parents had not been characterized. The linkage group with the S-locus in B. oleracea showed remarkable homology to the corresponding linkage group in B. napus except that in the latter there was an additional locus present, which might have been introgressed from B. rapa. The S-allele in the rapid-cycling Brassica was identified as the S29 allele, the S-allele of the cabbage was the S 5 allele. These same alleles were present in our two B. napus SI lines, but there was evidence that it might not be the active or major SI allele that caused self-incompatibility in these two B. napus crosses. Peroxidases (EC 1.11.1.7; donor: hydrogen peroxide oxidoreductase) are part of a large group of enzymes. In this study, peroxidase, a primer antioxidant enzyme, was purified with 19.3 fold and 0.2% efficiency from cauliflower (Brassica oleracea L.) by ammonium sulphate precipitation, dialysis, CM-Sephadex ion-exchange chromatography and Sephadex G-25 purification steps. The substrate specificity of peroxidase was investigated using 2,2'-azino-bis(3-ethylbenz-thiazoline-6-sulphonic acid) (ABTS), 2-methoxyphenol (guaiacol), 1,2-dihydroxybenzene (catechol), 1,2,3-trihyidroxybenzene (pyrogallol) and 4-methylcatechol. Also, optimum pH, optimum temperature, optimum ionic strength, stable pH, stable temperature, thermal inactivation conditions were determined for guaiacol/H(2)O(2), pyrogallol/H(2)O(2), ABTS/H(2)O(2), catechol/H(2)O(2) and 4-methyl catechol/H(2)O(2) substrate patterns. The molecular weight (M(w)) of this enzyme was found to be 44 kDa by gel filtration chromatography method. Native polyacrylamide gel electrophoresis (PAGE) was performed for isoenzyme determination and a single band was observed. K(m) and V(max) values were calculated from Lineweaver-Burk graph for each substrate patterns.  相似文献

A new series of dinuclear squarato-bridged nickel(II) and copper(II) complexes [Ni2(2,3,2-tet)21,3-C4O4)(H2O)2](ClO4)2 (1), [Ni2(aepn)21,3-C4O4)(H2O)2](ClO4)2 (2), [Cu2(pmedien)21,3-C4O4)(H2O)2](ClO4)2.4H2O (3) and [Cu2(DPA)21,2-C4O4)(H2O)2](ClO4)2 (4) where is the dianion of 3,4-dihydroxycyclobut-3-en-1,2-dione (squaric acid), 2,3,2-tet = 1,4,8,11-tetraazaundecane, aepn = N-(2-aminoethyl)-1,3-propanediamine, pmedien = N,N,N′,N″,N″-pentamethyldiethylenetriamine and DPA = di(2-pyridylmethyl)amine were synthesized and structurally characterized by X-ray crystallography. The spectral and structural characterization as well as the magnetic behaviour of these complexes is reported. In this series, structures consist of the groups as counter ions and the bridging the two M(II) centers in a μ-1,3- (1-3) and in a μ-1,2-bis(monodentate) (4) bonding fashions. The coordination geometry around the Ni(II) ions in 1 and 2 is six-coordinate with distorted octahedral environment achieved by N atoms of the amines and by one or two oxygen atoms from coordinated water molecules, respectively. In the Cu(II) complexes 3 and 4, a distorted square pyramidal geometry is achieved by the three N-atoms of the aepn or DPA and by an oxygen atom from a coordinated water molecule. The electronic spectra of the complexes in aqueous solutions are in complete agreement with the assigned X-ray geometry around the M(II) centers. The complexes show weak antiferromagnetic coupling with ∣J∣ = 1.8-4.2 cm−1 in the μ-1,3- bridged squarato compounds 1-3, and J = −16.1 cm−1 in the corresponding μ-1,2- bridged squarato complex 4. The magnetic properties are discussed in relation to the structural data.  相似文献

Summary. The Or mutation in cauliflower (Brassica oleracea L. var. botrytis) leads to abnormal accumulations of -carotene in orange chromoplasts, in tissues in which leucoplasts are characteristic of wild-type plants. Or chromoplasts were investigated by light microscopy of fresh materials and electron microscopy of glutaraldehyde- and potassium permanganate-fixed materials. Carotenoid inclusions in Or chromoplasts resemble those found in carrot root chromoplasts in their optical activity and angular shape. Electron microscopy revealed that the inclusions are made up of parallel, membrane-bound compartments. These stacks of membranes are variously rolled and folded into three-dimensional objects. We classify Or chromoplasts as membranous chromoplasts. The Or mutation also limits plastid replication so that a single chromoplast constitutes the plastidome in most of the affected cells. There are one to two chromoplasts in each cell of a shoot apex. The ability of differentiated chromoplasts to divide in the apical meristems of Or mutant plants resembles the ability of proplastids to maintain plastid continuity from cell to cell in meristems of Arabidopsis thaliana mutants in which plastid replication is drastically limited. The findings are used to discuss the number of levels of regulation involved in plastid replication.  相似文献

结球甘蓝(Brassica oleracea var. capitata)和青花菜(Brassica oleracea var. italica)小孢子胚再生植株频率低是目前影响游离小孢子培养技术有效应用的关键问题之一, 研究其小孢子胚植株再生频率的影响因素, 提高胚再生植株频率, 对促进游离小孢子培养技术在甘蓝类蔬菜育种中更好地应用具有重要意义。该文以结球甘蓝中甘11和青花菜TI-111等基因型为试材, 对影响游离小孢子胚再生成植株的固体培养基类型、琼脂浓度、胚的类型及胚在液体培养基中的滞留时间等因素进行了研究。结果表明: 游离小孢子培养25天的子叶胚在琼脂浓度为1%–1.25%的B5培养基上植株再生频率最高。进一步通过8个不同基因型对上述实验结果进行了验证, 结果显示, 游离小孢子培养25天的子叶胚在1%琼脂浓度的B5培养基上植株再生频率达77.8%–97.2%。  相似文献

Metallo-proteinase from 8-d-old seedlings of kale was isolated. The enzyme was extracted with 1% NaCl, concentrated by ammonium sulfate and finally purified by high-performance liquid chromatography. The isolated enzyme had a molecular weight of 22.4 kDa and showed a maximum activity at pH 9.0 using casein as a substrate. Proteolytic activity of proteinase was inhibited by chelators. The inhibition by ethylenediaminetetraacetate (EDTA) was abolished by some divalent metals ions, especially by Zn2+. The paper compares different approaches for the genetic transformation of cauliflower (Agrobacterium-mediated, PEG-mediated and/or electroporation). Transient expression of the neomycin phosphotransferase II (NPTII) gene could be detected after direct gene transfer. Stable transformation was achieved using both Agrobacterium-mediated and direct gene transfer. Expression as well as incorporation of the NPTII sequence could be demonstrated.  相似文献

Protoplasts of savoy cabbage (Brassica olleracea L. var. subauda), "SA61" (SV), were isolated from leaves and hypocotyls of seedlings grown in vitro, in enzyme mixture containing 2% cellulase (Onozuka R-10) and 0.8% macerozyme RI0. Good results of protoplast collection were obtained by using 18% and 17% sucrose solution floating leaf protoplasts and hypocotyl protoplasts respectively, and centrifugalizing with the rate of 500 r/min. All the collected protoplasts were cultured in 5 different liquid media from which the best results were observed on DPD1 medium for leaf protoplasts and on MS1 medium for hypocotyl protoplasts, with the highest cell division rate and planting efficiency. About 2 weeks of cultures, many cell clusters and a few embryo-like structures were visualized. The cell clusters developed into visible microcalli in 20-30 days and grew up to 1 mm or so in dimeter about 40 days of culture. For growth, the calli were transferred to 7 different agar media and from which two suitable media, MB2 and MB3, were selected. Cultured for 40-50 days, the calli grew up, and were transferred to 4 solid media for organ differentiation. Ideal results of shoot regeneration were obtained on MS, medium. About 2 weeks after rooted on the MS medium without any auxin, intact plants were regenerated.  相似文献
