Algal pigments were measured, by reverse-phase h.p.l.c., duringgrazing experiments with the protozoan Oxyrrhis marina Dujardinon Rhodomonas sp. Chlorophylls and carotenoids were degradedto colourless residues; the rate of degradation of pigmentswas highest in the light. Very little chlorophyll a (5%) wasdegraded to phaeophytin and none to phaeophorbide during grazing.This suggests that phaeopigment concentrations cannot be usedas a measure of algal mortality due to grazing when heterotrophicProtozoa are a component of the grazer community.  相似文献

Can phaeopigments be used as markers for Daphnia grazing in Lake Constance?   总被引:1,自引:0,他引:1  
The formation of chlorophyll a degradation products was measuredwith natural phytoplankton from Lake Constance and Daphnia magnaand native Daphnia as grazers in grazing experiments duringspring bloom conditions using high-pressure liquid chromatography(HPLC). Chlorophyll a start concentrations were between 1.2and 16.3 µg l–1; phaeopigment weights constituted5% of chlorophyll a weight. Only phaeophorbide a was a markerfor Daphnia grazing; concentrations of other phaeopigments (phaeophytina, chlorophyllide a and two unidentified phaeopigments) didnot increase during Daphnia grazing. Conversion efficiencies(chlorophyll a to phaeophorbide a) were between 0 and 43% ona weight basis, and between 0 and 65% on a molar basis. Conversionefficiencies were highest at high grazer density (40 Daphnial–1) and after a 24 h exposure time. Grazing by microzooplanktonprobably led to the formation of the two unidentified phaeopigments.In Lake Constance, Daphnia density was significantly positivelycorrelated with the phaeophorbide a/chlorophyll a ratio whenit was <5000 Daphnia m–3. However, when higher Daphniadensities were included in calculations, then Daphnia densitywas positively, but insignificantly, correlated with the phaeophorbidea/chlorophyll a ratio. This suggests that when the level offood per Daphnia is low, then grazing is more efficient withless production of phaeophorbide a and a higher production ofcolourless products.  相似文献

The activities of chlorophyllase, contents of pigments including chlorophyll a and b, chlorophyllide a and b, and phaeophorbide a during leaf senescence under low oxygen (0.5% O2) and control (air) were investigated in a non-yellowing mutant and wild-type leaves of snap beans (Phaseolus vulgaris L.). Chlorophyllase from leaf tissues had maximum activity when incubated at 40C in a mixture containing 50% acetone. In both mutant and wild type, chlorophyllase activity was the highest in freshly harvested non-senescent leaves and decreased sharply in the course of senescence, indicating that the loss of chlorophylls in senescing leaves is not directly related to the activity of chlorophyllase and that chlorophyllase activity is not altered in the mutant. The wild type had higher ratios of chlorophyll a to chlorophyll b than the mutant and chlorophyll a : b ratios increased during senescence in both types. In the senescent mutant leaves, accumulations of chlorophyllide a and chlorophyllide b were detected, but no phaeophorbide a was found. Chlorophyllide b had a greater accumulation than chlorophyllide a in the early stage of senescence. Low oxygen treatment not only delayed chlorophyll degradation but also enhanced the accumulations of chlorophyllide a and b and lowered the ratios of chlorophyll a to chlorophyll b.  相似文献

Data from four reservoirs representative of different trophic states and with different apparent optical properties were analyzed to determine the relationship of Secchi depth to algal biomass as measured by chlorophyll a. In the eutrophic reservoir Secchi depth was determined partially by the chlorophyll a content (r2 = 0.31) but only when chlorophyll a data from bloom conditions are included. In the two mesotrophic reservoirs, Secchi depth was entirely determined by non-algal turbidity. In the oligotrophic reservoir, Secchi depth was determined neither by chlorophyll a nor non-algal turbidity and was probably determined by dissolved color. When data from the four reservoirs were pooled (N = 205), 53% of the variation in Secchi depth was explained by: SD = 2.55–0.52 ln (Turbidity) + 0.005 (Chlorophyll a). It is apparent that attempts to estimate algal biomass for trophic state classification or other management practices from Secchi depth data are inappropriate even where moderate amounts of non-algal turbidity are present.  相似文献

To investigate the effect of the length of incubation on grazing parameters, four experiments were run for between 33 and 64 h, using unsorted communities of zooplankton (> 800 μm) in natural sea water. Samples were taken at timed intervals throughout the incubations and the concentrations of particulate chlorophyll, total pigment, and carbohydrate were measured. Chlorophyll and carbohydrate ingestion rates were directly proportional to substrate concentration and the level of each decreased exponentially with time throughout each experiment. A significant proportion of ingested chlorophyll was not egested as phaeopigment but was digested or destroyed. In all four experiments mixed stages of Calanus hyperboreus (Krøyer) and C. glacialis (Jaschnov) accounted for >90% of the zooplankton. Individual filtration rates calculated for chlorophyll and carbohydrate were constant over all four experiments, and were not significantly different from each other so that carbohydrate and chlorophyll were apparently consumed equally readily.  相似文献

We examined photopigment degradation and transformation in sediment microcosms that received different detrital source materials (planktonic, littoral, terrestrial) in the presence or absence of amphidpods (Gammarus sp.). Additions of realistic quantities of particulate organic matter resulted in detectable changes in pigment concentration and composition despite insignificant changes in total organic matter. The transformation of chlorophyll a to total phaeophorbide was significantly higher in all high quality (high nitrogen) detritus treatments containing amphipods. The highest production of phaeophorbide was in the higher quality detritus (blue-green algae, Anabaena cylindrica, and macrophyte, Vallisneria americana) when compared to red maple (Acer rubrum). Phaeophytin formation was not related to amphipod grazing and thus may be determined more by microbial heterotrophic processes. The degradation product of the carotenoid lutein, lutein 5,6 expoxide, was formed in all treatments. Phaeopigment composition can be used to infer differences in heterotrophic activity and will help in the interpretation of photopigment distribution in field samples.  相似文献

Phytoplankton production is determined by growth, senescence, sinking and zooplankton grazing. In an attempt to follow algal senescence and grazing, some authors have used HPLC fluorescence detection of chlorophyll a breakdown products. Laboratory grazing experiments have shown that copepods reduce chlorophyll a from diatoms leading to an increase in pheophytin a rather than pheophorbide a. However, field measurements only indicated a slight increase of pheopigment concentrations in summer. During this period, high heterotrophic activities (zooplankton and bacteria) seemed to be responsible for rapid pheopigment disappearance. On the other hand, highest chlorophyllide a levels appeared to be related to spring accumulation of nutrient-limited senescent algae. While increases in pheophytin a accounted for chlorophyll a consumption, changes in pheophorbide a concentrations could be linked to chlorophyllide a abundance. These results suggest that laboratory studies cannot be uncritically extrapolated to the field.  相似文献

The gf tomato mutant, which retains chlorophyll during ripening, has been found to be affected in leaf senescence. The leaves of the gfmutant show an absolute stay-green phenotype. As leaf senescence and fruit ripening proceed, there is a marked difference in chlorophyll content between wild-type and gf. In both attached and detached leaf studies, or after treatment with ethylene, the leaves withered and abscised in gf with only slight loss of chlorophyll and carotenoids. Total protein content declined and free amino acids increased during leaf senescence in wild-type and gf, but Western analysis showed that LHCII polypeptides were retained at higher levels in gf. Expression of senescence-related mRNAs increased normally in gf whereas those for cab, rbcS and rbcL declined in both mutant and wild-type. The mutant possesses enzyme activity for chlorophyllase, the formation of phaeophorbide a by the action of Mg-dechelatase and the oxygenolytic opening of the porphyrin macrocycle. Analysis of chlorophyll breakdown products in fruit indicated that gf, like other stay-green mutants, accumulates chlorophyllides a and b, but phaeophorbide a does not accumulate in vivo. This may indicate that, in the mutant, in vivo the action of phaeophorbide a-oxygenase is somehow presented, either by altered accessibility or transport of components required for thylakoid disassembly or the absence of another factor.  相似文献

During grazing of the copepodTemora longicornis onThalassiosira eccentrica phaeophorbide-a and several phaeophytins-a appeared in the culture vessels. However, a variable portion of the diatom's chlorophyll-a was degraded to substances that could not be detected either spectrophotometrically or fluorometrically. Chlorophyll-c and fucoxanthin were also lost during diatom cell transit throughTemora's gut. The variability between experiments in the destruction of chl.a to colourless residues is ascribed to differences in coprophagy and in transit time through the copepod gut, processes related to filtering rate and the ambient food concentration. Because the pigment loss is variable, grazing pressure cannot be estimated by simply recording bulk phaeopigment concentrations. The net result of the appearance of a fucoxanthinol-like pigment while all other pigments disappeared is a gradual carotenoid enrichment of the samples during grazing.  相似文献

The amount of particulate matter for different size classes was determined in the North Frisian Wadden Sea over a period of 10 months from November 1973 to August 1974. Particle volume, particulate organic carbon and phaeopigment concentrations decrease with advancing season, reaching minima in June. It is concluded that concentrations of particulate matter including chlorophyll were sufficiently high throughout the study period to support the growth and reproduction of appendicularia which abundantly occur in the Wadden Sea.  相似文献

Samples of the phytoplankton in a freshwater lake, Lake Liddell, New South Wales (Lat: 32° 22 S, Long. 150° 1 E) were collected every 4 weeks between October 1987 and November 1988. Chlorophyll a concentrations ranged from 1.8 g 1–1 to 9.1 g 1–1 and were positively correlated with the following nutrient parameters: total and nett mass additions of nitrate/nitrite-N and total-N, total additions of Kjeldahl-N, and nett mass addition N-P ratios. There was no correlation between lake nutrient concentrations and chlorophyll a. Factors other than nutrient concentrations appeared to be effecting chlorophyll a concentrations as summer levels were low despite nutrient concentrations being at a maximum. In spring and summer the phytoplankton was dominated by chlorophytes, with dinoflagellates and diatoms most abundant in autumn. During winter cyanobacteria were the most abundant. The relative abundance of chlorophytes was positively correlated with in lake nitrate/nitrite-N concentrations whereas the relative abundance of cyanobacteria was negatively correlated with this parameter. Based on chlorophyll a concentrations and the phytoplankton flora Lake Liddell can be classified as mesotrophic.  相似文献

We examined, under laboratory conditions, the influence of temperature (2 °C vs. 10 °C) on the physiological responses of two aquatic bryophytes from a mountain stream to artificially enhanced UV-B radiation for 82 d. These organisms may be exposed naturally to relatively low temperatures and high levels of UV-B radiation, and this combination is believed to increase the adverse effects of UV-B radiation. In the moss Fontinalis antipyretica, UV-B-treated samples showed severe physiological damages, including significant decreases in chlorophyll (Chl) and carotenoid (Car) contents, Chl a/b and Chl/phaeopigment ratios, Chl a fluorescence parameters Fv/Fm and PS2, electron transport rate (ETRmax), and growth. In the liverwort Jungermannia cordifolia, UV-B radiation hardly caused any physiological change except for growth reduction. Thus, this liverwort seemed to be more tolerant to UV-B radiation than the moss under the specific experimental conditions used, maybe partly due to the accumulation of UV-B absorbing compounds. The influence of temperature on the effects of UV-B radiation depended on the species: the higher the UV-B tolerance, the lower the influence of temperature. Also, different physiological variables showed varied responses to this influence. Particularly, the lower temperature used in our study enhanced the adverse effects of UV-B radiation on important physiological variables such as Fv/Fm, growth, and Chl/phaeopigment ratios in the UV-B-sensitive F. antipyretica, but not in the more UV-B-tolerant J. cordifolia. Thus, the adverse effects of cold and UV-B radiation were apparently additive in the moss, but this additiveness was lacking in the liverwort. The Principal Components Analyses (PCA) conducted for both species with the physiological data obtained after 36 and 82 d of culture confirmed the above results. Under natural conditions, the relatively high water temperatures in summer might facilitate the acclimation of aquatic bryophytes from mountain streams to high levels of UV-B radiation. This may be relevant to predict the consequences of concomitant global warming and increasing UV-B radiation.  相似文献

In order to evaluate the validity of the gut pigment methodto assess grazing and diet in two freshwater zooplankters, experimentswere carried out to check chlorophyll a and xanthophyll conservationduring feeding. For both animals, two sets of experiments wereconducted by incubating animals in the laboratory, either isolatedfrom a reservoir (the calanoid copepod, Eudiaptomus gracilis)or cultured under high-food conditions (the cladoceran, Daphniagaleata). For both animals, gut pigments and clearance rateson different types of algae were determined from the same incubations.Chlorophyll a and derivatives, as well as major algal carotenoids,were analysed by High Performance Liquid Chromatography (HPLC).In copepods, the pigment profiles from the gut extracts reflectedthe diet of the animals poorly. The animal extracts containedalmost exclusively alloxanthin (or an alloxanthin-like pigment)in large amounts, whereas the other pigments were lost in highproportions (>70% for lutein and fucoxanthin; 57 and 78%for a-phorbins). The cladocerans fed on the main types of algaeabundant in the suspensions, with a preference, however, forsmall cells. Although the main xanthophylls from these algaewere detected in the Daphnia extracts, some destruction of luteinand fucoxanthin may have occurred (18.7 and 30%). The loss ratefor alloxanthin seemed more variable (0 and 68%), possibly dependingon food concentration. As for the transformation of a-phorbins,E.gracilis and D.galeata behaved quite differently. The HPLCprofiles of copepod extracts always showed a very small chlorophylla peak, along with phaeophytin a and pyrophaeophytin a. Thosefrom the cladoceran exhibited a large phaeophorbide a peak,along with some chlorophyll a and phaeophytin a. In fact, D.galeatadid not destroy a-phorbins under our experimental conditionsbut converted chlorophyll a mainly into phaeophorbide. Froma comparison of our results with data from other studies, itseems that in these two zooplankters, use of gut pigment datafor quantitative grazing assessment should be considered withcaution.  相似文献

Effects of grazing on the quantity and quality of freshwater Aufwuchs   总被引:6,自引:5,他引:1  
Qualitative and quantitative measures of the Aufwuchs (scum flora) on artificial substrates in situ were used to evaluate the effects of grazing by freshwater pulmonate snails in a shallow pond in southeastern Michigan. Grazer densities of 216 snails/m2 marked reduced standing crop so that after 45 days grazed substrata had 6.46 mg dry weight, 604 µg C and 4.18 µg chlorophyll a as compared to controls with 30.62 mg dry weight, 3699 µg C and 6.29 µg chlorophyll a, all on a per dm2 basis.Grazing did not change carbon per mg dry matter but caused significant increases in both µg chlorophyll a (control, 0.206; and grazed, 0.649 µg chlorophyll a/mg dry weight, P < 0.01) and nitrogen (control, 8.3; and grazed, 24.2 µg N/mg dry weight, P < 0.001) after 45 days. Both abundance and diversity of the attached community was reduced by grazing from 24 taxa and 80,889 individuals/cm2 on control to 8 taxa and 501 individuals/cm2 on grazed substrates. Mean productivity of the Aufwuchs was significantly (P < 0.001) reduced by grazing from 76.3 µg C/(dm2·day) on control to 17.7 µg C/(dm2·day) on grazed substrata.Snails were very efficient at clearing smooth surfaces of living cells, detritus, and particulate inorganic matter. There was little evidence of selectivity except for an apparent inability to remove some of the smallest cells (e.g. Cocconeis sp.) probably for mechanical reasons.  相似文献

Summary The effect of the cyanogenic glucosides linamarin and lotaustralin and their hydrolyzing enzyme linamarase was studied in a B2 generation segregating for the genes Ac and Li. Plants containing the glucosides are protected against grazing by snails both in the seedling stage and as adult plants. In seedlings, however, there is a direct effect on survival, whereas in adult plants the leaf area of plants containing linamarin/lotaustralin is less reduced under intense grazing. Linamarase has no effect on grazing by snails, possibly as a result of the presence of -glucosidase activity in the gut of these animals. The genes Ac and Li, or genes tightly linked to them, have other effects as well: plants possessing one dominant Ac allele produce fewer flowers than homozygous ac plants. I compared this difference in flower production to the metabolic cost of producing the cyanogenic glucosides. The energy content of the difference in flower head production far exceeded the metabolic cost of cyanoglucoside production in Acac plants. It is possible that the cost of maintaining a certain level of cyanoglucosides is much more important for the plant than the initial cost of biosynthesis. The importance of the effects of Ac and Li in the maintenance of cyanogenic polymorphism in white clover is discussed.  相似文献

Summary High Performance Liquid Chromatography analysis of algal pigments from inter- and subtidal (deep and shallow) sediments from the Kerguelen Islands showed clear differences in the pigment composition at the different stations. High concentrations of chlorophyll c and fucoxanthin were present at all locations, indicating significant diatom densities, chlorophyll b was detected at all sites. At one station the other green algal pigments were also present; here green algae contributed more to chlorophyll a concentrations than diatoms, as estimated by using pigment ratios and microscopic observations. At another location chlorophyll b was associated with a high concentration of diadinoxanthin, indicating an abundance of euglenoids. This indicates that chemotaxonomy can be powerful tool in microphytobenthos studies since enumeration of living cells are difficult as many algae are attached to sediment particles (epipsammic algae). Ways of diagenesis, carotenoid degradation and the role of grazing are briefly mentioned. Phaeophorbide a-like pigments were the most significant chlorophyll a degradation products, with concentrations up to 110 g · g–1 dry weight sediment, i.e. 10 times the chlorophyll a concentration. Some taxonomic estimations, based on pigments ratios, and their limits, are discussed.  相似文献

Sedimentation of chlorophylls was studied during summer 1997 in Adventfjorden (Spitsbergen, Arctic). During the period of study, the water column was found to be well stratified by a freshened surface layer (salinity <31 PSS). A high load of suspended particulate matter from riverine discharge reduced the euphotic zone to an interval of 0.4–1.1m. Total particulate matter sedimentation rates were about twice as high in June as in July. The following chlorophylls were distinguished in the sedimented particles: chl a and its degradation products (allomer chl a, phaeophytin a, phaeophorbide a, chlorophyllide a), chl b and chl c 1+c 2. The quantitatively most important derivative of chl a was phaeophorbide a (31--41% of porphyrin a). Generally, the sedimentation rate of chlorophylls increased with depth. Linear relationships between concentrations of chl a and phaeophorbide a (r 2=0.92), as well as between concentrations of chl a and phaeophytin a (r 2=0.90) indicated a strong connection between phytoplankton abundance and zooplankton grazing. The significant correlation between chl a and chlorophyllide a concentrations (r 2=0.82) showed that most of the sinking chl a belonged primarily to diatoms, and low chlorophyllide a:chl a ratio (0.03) indicated that cellular senescence was not an important reason for the sinking of chl a. Moreover, very low chl b:chl a ratios (about 0.05 calculated for samples where chl b was detectable) suggest that contributions of green algae and/or higher plant detritus were negligible in sinking particles. The ratio of chl c 1+c 2:chl a was 0.85 indicating that chl c-containing algae were dominating.  相似文献

The release of dissolved organic carbon (DOC) byMysis relicta that occurs while feeding may be an important pathway in the recycling of nutrients in Lake Michigan. Laboratory experiments were conducted to determine the levels of DOC released byM. relicta from Lake Michigan while grazing on laboratory-cultured diatoms. Increasing filtering rates were observed at progressively lower chlorophyll concentrations. Higher filtering rates were accompanied by higher fragmentation of diatoms and a higher concomitant release of DOC from broken cells. While the amount of DOC release is small (0.02–3.30 g C · 1–1· h–1), it nonetheless is of importance to bacteria and possibly some algae. The chlorophyll a content and two operational fractions of carbohydrate (water extractable and EDTA extractable) were measured every three months during one year along transects on a tidal flat in the Ems-Dollard estuary (The Netherlands). Chlorophyll a was used as an indicator of microphytobenthos biomass, which was composed predominantly of epipelic diatoms. Both carbohydrate fractions correlated significantly with chlorophyll a. EDTA extractable carbohydrates were more resistant towards degradation than the water extractable fraction. During most of the year, concentrations of chlorophyll a and carbohydrates were low, but in June, high concentrations of up to 90 g chlorophyll a/g sediment were found in a narrow zone running parallel to the channel. Maximum concentrations of water extractable carbohydrates and EDTA extractable carbohydrates ranged between 800–1200 and 600–800 g/g sediment, respectively. The mud content was high (± 90%) at the margin of the tidal flat. This was not limited to the growth season of the diatoms, but was observed throughout the year. This indicated that the high mud content at the mudflat margin was mainly caused by hydrodynamic factors, and not by biostabilization. In June, exceptionally high diatom densities were found in sediment with a high mud content. There was only minor evidence that biostabilization by epipelic diatoms lead to a further increase in the mud content of the sediment.  相似文献

The development of strategies for selecting and characterizing aluminum-resistant variants from Nicotiana plumbaginifolia Viv. cell cultures is described. Plated cells, smeared callus, in-vitro-grown shoots, and seedlings of wild-type N. plumbaginifolia all showed similar responses to Al, with total growth inhibition at or above 600 M Al. The strict control of both cell density and aggregate size is important in selection experiments for total inhibition of the growth of wild-type cells. Two approaches for the selection of Al-resistant variants were used. In a direct method, cells were plated onto medium containing 600 M Al which inhibited growth and chlorophyll synthesis in wildtype cells. A double selection strategy based on both cell growth and greening was used to isolate 29 Al-resistant variants. In the other approach, a rescue method, suspensions were cultured for 10 d in medium containing 600 M Al, then plated onto standard medium for recovery of survivors. Using this strategy, 217 Al-resistant variants were selected. After six to twelve weeks of growth in the absence of Al, each variant was cloned and reselected from single cells. Al resistance was retained in 31% and 51% of the variants selected by the direct and rescue strategies, respectively. Seedling segregation data are presented for the progeny (selfed and backcrossed) of plants regenerated from one of the variants and are consistent with those expected for a single dominant mutation.  相似文献
