Park wheat (Triticum aestivum cv. Park) is susceptible to stem melanosis when grown on Cu-deficient soils. Three field experiments were conducted to evaluate the effectiveness of various Cu sources, using different methods and rates of application for reducing the incidence of stem melanosis and increasing the grain yield of Park wheat on a Cu-deficient Black Chernozemic soil in central Alberta. A fourth experiment determined the residual effect of Cu over 4 years. In the first experiment, Park wheat had less disease and higher grain yield when Cu-chelate solution at 2 and 4 kg Cu ha−1 or Cu-sulfate solution at 10 and 20 kg Cu ha−1 was applied to the soil surface and incorporated to a depth of 8 cm or was applied at these rates as a foliar spray. The foliar application, however, was phytotoxic and delayed maturity by approximately two weeks. Sidebanding Cu (4 cm to the side and 4 cm below the seed row) was least effective. In the second experiment, Cu-sulfate solution incorporated into the soil was more effective than the soil incorporated granular Cu-sulfate in reducing disease incidence and increasing grain yield in the year of application. In the following year the granular Cu was as effective as solution Cu. The third experiment showed that Cu seed dressings did not have any effect on disease incidence or grain yield of Park wheat. In the fourth experiment, the residual effect of Cu-chelate was evident four years after application. The grain yield in the fourth year was about four times that of the control. In the Mediterranean basin, Verticillium Wilt of Olive (VWO) is diffused throughout its range of cultivation, causing severe yield losses and tree mortality. The disease was reported in almost all the Mediterranean and Middle East countries, and in Lebanon it is of increasing significance also on many valuable crops. The disease has already been reported on potato, peach and almond in the Bekaa valley; however, to date no information is available about the incidence of VWO and the inoculum density of Verticillium dahliae microsclerotia in soil of the main agricultural areas of Lebanon. Results from the present investigations demonstrate a high V. dahliae frequency in soils (75.3%), coupled with a mean soil inoculum density of 17.0 MS g?1, clearly indicating a great impact on the production of susceptible hosts in Lebanon, mainly in Bekaa region. Molecular method to assess the microsclerotia inoculum density in soil allowed the detection of a higher frequency of infested soils, as compared with the traditional plating, thus confirming its higher sensitivity. The overall Verticillium wilt prevalence in the inspected olive orchards was 46.2%, and the frequency of V. dahliae‐infected trees was 25.7%. The widespread presence of V. dahliae in all olive growing areas of Lebanon enforces the adoption of measures aimed at reducing the soil inoculum density before any new olive plantation, and the use of strong phytosanitary regulations to improve the certification schemes of propagating material.  相似文献

Experiments on soil drenching with fungicides against take-all in wheat   总被引:3,自引:0,他引:3  
In short term pot experiments benomyl, iprodione and KWG 0599 applied as soil drenches in several types of soil significantly suppressed take-all symptoms from inoculum placed just below wheat seeds planted 1×5 cm deep, and in sand but not other soils when seeds were 5 cm deep. Benomyl was, however, effective against inoculum below seed planted 5 cm deep in a loam-sand mixture when the drench contained an alcohol ethoxylate surfactant. Computer simulations of fungicide distributions in the soils correlated well with disease control observations. In long term outdoor pot experiments two drenches with benomyl (without surfactant) controlled disease significantly for at least 3 months against inoculum placed 15 cm deep. The significance of these results for the practical control of take-all by fungicides is discussed.  相似文献

The symptoms of a growth disorder of cotton and associated properties of the soil were quantified. Data were collected from 35 sites across two irrigated fields that showed gradients in the severity of early season stunting of cotton. Ordination analysis of soil characteristics distinguished three groups of sites (A, B, and C) which corresponded to patterns of yield and early season growth. Group A and B soils had lower pH, finer texture and higher P, Zn, Mn and exchangeable Mg, K and Na than group C soils. Early season growth and arbuscular mycorrhizal colonisation of cotton at group A and B sites was much slower than at group C sites. Group B sites showed a recovery of yield later in the season while group A sites did not. Nutrient deficiencies, waterlogging, soil compaction, soil sodicity and Mn toxicity were unlikely causes of early season stunting, although soil Mn was a good predictor for the disorder. Thielaviopsis basicola, Verticillium dahliae and unidentified Chytridiomycetes were not associated with stunting and reduced yield, although other fungal pathogens may have been present. Root browning was a symptom of the disorder and suggests that pathogens, perhaps bacteria, play a causal role. This study showed that the growth disorder involved an interaction between cotton and the soil flora which was associated with heavy soil texture.  相似文献

Poor field control of lettuce collar rot by iprodione was observed in southern France and was attributed to enhanced biodegradation of the fungicide. Enhanced biodegradation was obtained in vitro after repeated applications of iprodione to non-degrading soils. Normal soils became biodegrading after mixing with degrading soils (3 vol./1 vol.). Activity of the responsible microflora seemed dependent on soil physico-chemical characteristics.  相似文献

Several bacterial and fungal strains have been evaluated as biocontrol agents (BCAs) against Verticillium dahliae. In these studies, the BCAs were applied as a root drenching inoculum; however, this application method may have an adverse effect on the native, beneficial for the plants, microbial community. In the present study, it was evaluated whether endophytic application by stem injecting a conidial suspension of the non pathogenic Fusarium oxysporum F2 strain, isolated from a V. dahliae suppressive compost amendment, could reduce significantly Verticillium wilt symptom development in eggplants. It was revealed that stem injection of F2 seven days before transplanting the seedlings to soil infested by V. dahliae microsclerotia resulted in reduced disease severity compared to the control treatment. To visualise F2 ramification into the plant vascular system eggplant stems were injected with an EGFP transformed F2 strain. It was shown that F2 colonises effectively the plant vascular tissues over a long period of time as it was assessed by F2 DNA levels. In parallel, qPCR analysis showed that the application of F2 reduced significantly the amount of V. dahliae DNA in the stem tissues compared to the control treatment.  相似文献

Summary Shoot cultures of three Hibiscus moscheutos (L.) cultivars were infested with micro-arthropods (mites). Nodal segments (1 cm long) were excised from these cultures and encapsulated in a sodium alginate gelled Driver and Kuniyuki Walnut DKW medium containing 10, 50, or 100 mg l−1 acephate (insecticide) or 10 mg l−1 acephate plus 0, 50, or 100 mg l−1 benomyl (fungicide), then placed in refrigerated (5°C) darkness for 4 wk. Acephate was tested alone if visible fungus was not touching the shoot masses and benomyl was tested if fungus was in contact with the proliferating shoots. Cold-stored encapsulated nodes were then placed on DKW medium with 0.1 μM thidiazuron for 6 wk for subsequent shoot development. The presence of acephate in the encapsulation medium completely eradicated or killed the mites, with 38–69% of cultures fungus-free; 12% of the fungal-contaminated nodes encapsulated with 100 mg l−1 benomyl were fungus-free.  相似文献

[目的] 研究黄萎病抗性棉(海 7124)根际土壤中酵母菌株对棉花黄萎病病原真菌大丽轮枝菌和全齿复活线虫的拮抗效果,为生物防治棉花黄萎病和全齿复活线虫提供理论依据。[方法] 通过镜检、糖发酵实验、碳源同化实验、26S rRNA测序对菌株的形态、生理生化特征及其系统发育关系进行鉴定,并利用七叶苷筛选、刚果红染色、平皿对峙实验、盆栽实验、平板生测实验测试其产酶活性以及抑制大丽轮枝菌和杀线虫活性。[结果] 从大批黄萎病抗性棉(海 7124)根际土壤中筛选出编号为YIN9的酵母菌菌株,分类鉴定结果表明:YIN9菌株属于假丝酵母属Candida。平皿对峙实验结果表明:菌株YIN9对大丽轮枝菌的抑菌率达59%;将菌株YIN9的无菌发酵滤液与大丽轮枝菌孢子共培养12 h后镜检发现,用菌株YIN9处理的实验组,大部分棉花黄萎病病菌孢子不能正常萌发。盆栽实验结果表明:菌株YIN9对棉花黄萎病的平均防治效果为60.02%,可以显著降低感病棉棉花黄萎病的发病率和病情指数。此外,与从黄萎病抗性棉根际土壤中筛选获得的其他酵母菌株相比,菌株YIN9具备较高的杀线虫活性:菌株作用全齿复活线虫48 和60 h后,线虫死亡率分别为90%和100%。将菌株YIN9发酵液煮沸后,其抑制大丽轮枝菌和杀线虫活性均急剧下降,进一步测试发现,该菌株拥有较高的蛋白酶和纤维素酶活性。[结论] YIN9中的生防因子可能是热不稳定性物质,具备较高的杀线虫活性,可以显著提高感病棉对黄萎病的抗性。  相似文献

The development of vesicular-arbuscular mycorrhizal fungi (VAMF): Glomus mosseae (Nicol and Gerd.) Gerdemann and Trappe, Glomus versiforme (Karsten) Berch, Sclerocystis sinuosa Gerdemann and Bakhi and Verticillium dahliae and the effects of the VAMF on the verticillium wilt of cotton (Gossypium hirsutum L. and Gossypium barbadense L.) were studied with paper pots, black plastic tubes and clay pots under natural growth conditions. All of the tested VAMF were able to infect all the cotton varieties used in the present experiment and typical vesicles and arbuscules were formed in the cortical cells of the cotton roots after inoculation. The cap cells, meristem, differentiating and elongating zones of the root tip were found to be colonized by the VAMF. In the case of most V. dahliae infection, the colonization occurred mostly from the root tip up to 2 cm. VAMF and V. dahliae mutually reduced their percentage of infection when inoculated simultaneously. VAMF inoculation reduced the numbers of germinable microsclerotia in the soil of the mycorrhizosphere, while the quantity of VAM fungal spores in the soil was not influenced by infection of with V. dahliae. The % of arbuscule colonization in roots was negatively correlated with the disease grades, while the numbers of vesicles in roots were not. These results suggest that certain vital competition and antagonistic reactions exist between VAMF and V. dahliae. VAMF reduced the incidence and disease indices of verticillium wilt of cotton during the whole growth phase. It is evident that cotton seedling growth was promoted, flowering was advanced, the numbers of flowers and bolls were increased, and this resulted in an increase in the yield of seed cotton. Among the VAMF species, Glomus versiforme was the most effective, and Sclerocystis sinuosa was inferior. So far as the author is aware, such an effect of VAMF on the increase of cotton wilt tolerance/resistance is reported here far the first time.  相似文献

Soil texture, chemistry and moisture have a profound effect upon the activity and persistence of entomopathogenic nematodes (EPNs). Whereas nematodes’ natural habitat is within the soil, ticks and other arthropod pests prefer to stay on the soil surface and under stones or leaf litter; they spend much of their life cycle in the humid environment of the soil upper layer, therefore consideration of the effect of the soil environment on nematode activity is a pre-requisite for the sucessful use of EPNs against arthropod pests. In the present study we investigated the effects of soil type, and humidity on various nematode strains and on their effectiveness against ticks. Many infective juveniles (IJs) of Steinernema carpocapsae and S. riobrave were found in the uppermost soil layer whereas the heterorhabditid strains were almost absent from the upper 6 cm of the soil profile. The IJs of S. feltiae, and the S. carpocapsae strain S-20, exhibited an intermediate behavior. It was found that the activity of IJs of S. carpocapsae in the soil upper layer (1 cm depth) was strongly affected by soil type: the greatest number of IJs were recorded from sandy loam soil; less were found in the lighter soils – ‘Marine sand’ and ‘Calcareous sandstone’ – and only very few were recovered from heavy soils. Strikingly, even when the soil moisture was low and the number of nematodes found in the upper layer correspondingly low, tick mortality remained high. The results demonstrate: (a) the possible use of the nematodes as an anti-tick agent; (b) the importance of knowing the exact interaction of nematodes with the immediate environment of the pest, in order to optimize the pest-control activity of the nematode.  相似文献

Abstract

Little is known about the effect of modified carbon black nanoparticles (MNCB) on the availability of heavy metals and petroleum degradation in petroleum and heavy metals co-contaminated soils. The overall objective of this study was to investigate the simultaneous effect of MNCB on heavy metal immobilization and petroleum biodegradation in co-contaminated soil in plant and plant-microbe combined remediation. The results showed that the petroleum degradation increased by 50% in petroleum-Cd co-contaminated soil and 65% in petroleum-Ni co-contaminated soil in plant-microbe combined remediation, comparing with the plant remediation and the application of MNCB did not show significant improvement on petroleum degradation in both plant and plant-microbe combined remediation. MNCB could significantly reduce the availability of heavy metals in soil and the uptakes of Cd and Ni by Suaeda salsa by roughly 18 and 10% and improve the growth of plant by alleviating the growth inhibition caused by heavy metals. The application of Bacillus subtilis and Sphingobacterium multivorum (heavy metal tolerant bacteria) inhibited the biomass of Suaeda salsa by enhancing the petroleum degradation. It could be concluded that MNCB played a major role in immobilizing the heavy metals and bacteria dominated the petroleum degradation in petroleum-metal co-contaminated soil.  相似文献

In Spain, Verticillium wilt, caused by Verticillium dahliae, is the most important disease of cotton and olive. Isolates of V. dahliae infecting these crops can be classified into highly virulent, defoliating (D), and mildly virulent, nondefoliating (ND), pathotypes. Infested soil is the primary source of inoculum for Verticillium wilt epidemics in cotton and olive, and severity of disease relates to the prevailing V.dahliae pathotype. In this work we have adapted the use of previously developed primer pairs specific for D and ND V. dahliae for the detection of these pathotypes by nested PCR in artificial and natural soils. Success in the detection procedure depends upon efficiency in extracting PCR-quality DNA from soil samples. We developed an efficient DNA extraction method from microsclerotia infesting the soil that includes the use of acid washed sand during the grinding process and skimmed milk to avoid co-purification of Taq-polymerase inhibitors with DNA. The specific nested-PCR procedure effectively detected 10 or more microsclerotia per gram of soil. The detection procedure has proven efficient when used with a naturally infested soil, thus demonstrating usefullness of the diagnostic method for rapid and accurate assessment of soil contamination by V. dahliae pathotypes.  相似文献

Experimental sites were established at two locations in north-eastern Victoria to define factors limiting the establishment and growth of Trifolium subterranean L. (subterranean clover). Liming the soil, seed inoculation and fungicide application were used in renovating subterranean clover pasture on two acidic soils (Longwood: brown/grey sandy loam DY 3.14 and Seymour: grey brown light clay DY 3.22, Northcote classification) with mean annual rainfall of 650 mm and 600 mm respectively. Soil acidity, low available soil phosphorus and plant disease were identified as factors limiting clover yield on these soils. Significant yield responses to lime (35–140%) were obtained with subterranean clover at both sites, with corresponding decreases in Al in the 0–10 cm soil horizon. Liming the soil, when combined with seed inoculation, increased the number and effectiveness of root nodules at both sites. Soil P available for plant growth was low at both sites (6.1 and 8.4 μg g−1) resulting in sub-optimal P concentrations in the clover herbage (45 mmol kg−1 at Longwood). Levels of root disease were low but Aphanomyces euteiches and Phytophthora clandestina (causal agents of lateral and tap root rot) were detected frequently on roots. Application of fungicide resulted in higher dry matter yields (p=0.05) at both sites. An assessment of the relative contributions of these limiting factors and the benefits to be obtained from better management would provide a clearer picture of the profitability and sustainability of this farming system.  相似文献

Metam sodium (MS) is often applied to potato fields via sprinkler irrigation systems (water-run, WR) to reduce propagules of soil-borne pathogenic fungi, particularly Verticillium dahliae, to prevent yield loss from potato early dying disease. However, this procedure has not been effective for controlling quality defects in tubers caused by Columbia root-knot nematode (Meloidogyne chitwoodi). In five trials from 1996 to 2001, application of MS by soil shank injection (SH) provided better control and tuber quality than that generally obtained by WR MS, in three of five trials. Results were similar when SH MS was injected at one (41–45 cm), two (15 and 30 cm) or three (15, 30 and 45 cm) depths. In the two trials where SH metam potassium was tested, culls were reduced to 3% and 0% and were equivalent to those resulting from a similar rate in kg a.i./ha of SH MS. A shank-injected tank mix of MS plus ethoprop EC and SH MS plus in-season chemigation applications of oxamyl provided acceptable control in trials where SH MS alone was inadequate. In-furrow application of aldicarb at planting following SH MS did not appear to increase performance. Most consistent control (0–2% culled tubers in five trials) occurred when SH MS at 280 liters/ha was used together with 1,3- dichloropropene (140 liters/ha), applied simultaneously or sequentially. This was similar to combinations of 1,3-D and WR MS, but SH MS may be preferred under certain conditions.  相似文献

Soil fungi accumulate radiocaesium from contaminated soil and it has been hypothesised that this may alter the plant availability and movement of the radionuclide in soil. The effect of twice-monthly addition of an aqueous suspension of the fungicide ‘Captan’ on the changes in a peaty podzol soil at 2 sites, contaminated 2 or 3 years earlier by the injection of 134Cs, has been quantified. The sites had different soil acidity and vegetation cover. The less acid soil (pHwater 5.0) had been improved by the addition of lime and fertilizer and was reseeded with grass and clover. The more acid soil (pHwater 3.8) was under hill grasses, herbs and heather. On both sites the addition of fungicide did not alter the amount or concentration of radiocaesium in plant material sampled monthly or the depth distribution of radiocaesium in the soil profile. The concentration of the fungal constituent, ergosterol, in the soil, measured monthly, was unaffected by the fungicide treatment but evidence was obtained from a pot experiment to show that ergosterol decomposes slowly in cold, wet soils. On the more acid soil, two weeks after the last application of fungicide, there was a decline in active fungi as measured by fluorescein diacetate staining. Chloroform fumigation of the more acid soil resulted in a small increase in the amount of 134Cs exchangeable with 1 M ammonium acetate. Radiocaesium in seven different fungi grown in pure culture was found to be almost entirely extractable (> 95%) with 1 M ammonium acetate. Another, Amanita rubescens, showed some retention and 88% was extractable. These findings do not preclude the fungal biomass as an important soil component controlling plant availability of radiocaesium from acid, organic soils by maintaining radiocaesium in a biological cycle, but make it unlikely that any fixation by fungi in a chemical sense is involved.  相似文献

Apple orchard soil treated with Aaterra (etridiazole), A5430 (furalaxyl), copper sulphate, LS 74–783 (aluminium tris(ethyl phosphonate)) or water in mid-August 1977 was assayed for Phytophthora syringae by an apple fruit baiting method immediately after fungicide application and at weekly intervals for 8 wk. There was no infection of baits placed in the orchard and this was attributed to low rainfall. When wetted and baited, samples of surface soil caused from 0 to 85% fruit infection depending on the time of sampling and soil treatment. In control soil, fungus activity was low in the first 4 wk, high in the fifth wk and fluctuated widely thereafter. In samples from soils treated with Aaterra, copper sulphate or LS 74–783 activity was first apparent in the fifth wk and then fluctuated roughly in parallel with that of control soil although at generally lower levels. Changing levels of activity were apparently related to air temperature during baiting. No infection of fruits occurred in any samples from soil treated with furalaxyl. Further assays of this soil indicated a complete suppression of P. syringae to at least 10 cm soil depth. Furalaxyl is sufficiently active to be considered as a soil treatment for controlling Phytophthora fruit rot of apple and pear.  相似文献

Captan was applied to laboratory-incubated agricultural soil and to bacterial cultures to determine its effects on total counts of soil microorganisms, nitrification, ammonification of urea and asymbiotic dinitrogen fixation. In Captan-treated soils, total count of fungi, bacteria and actinomycetes decreased significantly only at a relatively high fungicide concentration (1000 μg.g−1). Fungi and actinomycetes were more affected than bacteria. While oxidation of ammonia in an enriched, actively nitrifying culture was almost totally inhibited by Captan, ammonification of urea in incubated soil was only partly depressed. The depressing effect of Captan was more pronounced in cultures of Micrococcus than in those of Proteus. Asymbiotic dinitrogen fixation in nutrient-ammended soil was promoted during the first week and depressed on prolonged exposure to the fungicide depending on its first concentration. In autoclaved Azotobacter-inoculated soil a similar but less pronounced effect was noticed. Fixation by Azotobacter caltures was insensitive to Captan. In contrast, growth ofRhizobium phaseoli, R. leguminosarum andR. japonicum in yeast-extract-mannitol medium was adversly affected by Captan, particularly at 200 μg.ml−1. Nodulation of pea and mung bean (1 month old potted plants) grown from surface-sterilized inoculated seeds in aptan-treated soil was also significantly depressed. Both total number of nodules decreased with increasing concentration of the fungicide, but the inhibitory effect was more pronounced in the number of effective nodules.  相似文献

Summary Organic (e.g. chitin, green manure, cellulose) amendments to soil induced quantitative and qualitative changes in the rhizosphere microflora of antirrhinum plants infected withVerticillium dahliae Kleb. Whereas reduction in disease severity occurred with chitin and green manure amendments, an increase in disease severity was observed with the application of cellulose. The reduction of the disease severity with chitin and green manure may be correlated with the increased population of actinomycetes in the antirrhinum rhizosphere.  相似文献

We examined whether pinyon mice (Peromyscus truei) and brush mice (P. boylii) could act as directed dispersal agents of pinyon pine (Pinus edulis) through differential responses to soil particle size and rock cover. In field experiments, we allowed mice to either cache pinyon seeds or recover artificially cached seeds (pilfer) from quadrats containing large- or small-particle soils. Both species placed most (70%) seed caches in small-particle soil. Pilfering was the same from both particle sizes in the first year, while more seeds were pilfered from large-particle soils in the second year. In separate experiments, rock cover interacted with soil particle size, with both species placing over 50% of their caches in small-particle soil with rock cover. Overall, we found greater seed-caching in small-particle soils near rocks, with equal or lower pilfering from small-particle soils, suggesting more seeds would survive in small-particle soils near rock cover. Three lines of evidence supported the hypothesis that mice could act as directed dispersers by moving pinyon seeds to beneficial microsites for germination and establishment. First, in greenhouse experiments, pinyon seed germination was 4 times greater in small-particle soil cores than in large-particle soil cores. Second, soils near rocks had significantly higher water content than areas of open soil at the driest time of the year, a critical factor for seedling survival in the arid southwestern USA. Third, 75% of juvenile pinyon trees were growing in small-particle soils, and 45% were growing near rock nurses.  相似文献

Application of a peat-based powder inoculant of Azospirillum brasilense, as well as a granular inoculant (each containing 0.5–1.0×107 Azospirillum/g moist peat), in the seed furrows of Zea mays resulted in significantly increased yields (11 to 14%) in light soils at low rates of N fertilization. In general, there was no effect of inoculation on plant yields in heavier soils nor when N fertilization was high. Pre-emergence application of granular inoculant and inoculation associated with irrigation were more efficient in increasing yield than inoculation post-emergence or seed coating.E.
