Fixed-time AI (TAI) after GnRH-PGF(2alpha)-GnRH treatment is a method to achieve pregnancies in dairy herds without estrous detection. However, cows that fail to respond to the initial GnRH may have compromised TAI conception rates due to asynchronous ovarian response. This study documented the percentage of GnRH-treated Holstein cows (n=345) in two herds that displayed estrus at an inopportune time for optimum TAI conception rate (< or =48h post-PGF(2alpha); premature estrus (PE)) and compared conception rates of two TAI protocols in cows that did not display PE. At biweekly herd health exams, cows diagnosed as not pregnant to a previous AI and cows >80 days postpartum with no AI were treated with 100 microg GnRH (day -7) and 25mg PGF(2alpha) (day 0). Cows detected in PE by twice-daily visual observation from day -7 to day 2 were bred by AI 8-12h later. Cows not detected in PE were randomly assigned by parity, body condition score, and postpartum interval to receive either: (1) 100microg GnRH at 48h after PGF(2alpha) and TAI 16 to 18h later (Ovsynch); or (2) TAI at 72h post-PGF(2alpha) and a concurrent 100 microg GnRH injection to those cows not detected in estrus between 48 and 72h post-PGF(2alpha) (modified Ovsynch (MOV)). All hormone injections were im. Twenty percent (68/345) of the cows were detected in estrus before 48 after PGF(2alpha), of which 5% (17/345) were detected in estrus before PGF(2alpha) (< or =day 0). Herd influenced the percentage of cows in the PE group (herd A versus herd B; 25% versus 14%; P<0.05). Conception rates were not affected by treatment (PE versus Ovsynch versus MOV; 32% (21/65) versus 30% (37/125) versus 32% (47/145); P>0.10). However, within MOV-treated cows, conception rates were greater (P<0.05) in cows detected in estrus (46% (23/50)) compared with cows not detected in estrus (25% (24/95)). In conclusion, 20% of GnRH-treated cows displayed PE and necessitates estrous detection during this period if maximal pregnancy rates are to be achieved. Although additional estrous detection is required compared to Ovsynch, reduced cow handling and hormone usage, efficient use of expensive semen through greater conception rates in cows detected in estrus, and comparable TAI conception rates, suggests the MOV protocol may be a cost effective alternative to Ovsynch in many dairy herd reproductive management programs.  相似文献

Objectives were to evaluate progesterone concentrations after cows had initiated estrous cycles following calving and induction of estrous cycles in postpartum anovular high-producing Holstein dairy cows treated with controlled internal drug releasing (CIDR). In experiment 1 (EXP1), 62 cows that had initiated estrous cycles received a new CIDR (NCIDR) containing 1.38 g of progesterone or a 7-d used autoclaved CIDR (UCIDR) 48h after luteolysis for 7 d. Ovaries were examined by ultrasonography, and plasma analyzed for concentrations of progesterone. In experiment 2 (EXP2), 515 cows diagnosed as anestrus were randomly assigned to untreated control, NCIDR or UCIDR for 6d. Plasma was analyzed for concentration of progesterone 12 d after CIDR removal to determine ovulation. In EXP1, milk yield and body condition did not influence progesterone concentrations. Concentration of progesterone tended to increase faster (P=0.10) in cows receiving UCIDR than NCIDR, but both treatments reached a plateau at 90min. Cows receiving the NCIDR had greater (P=0.04) concentrations of progesterone during the 7-d treatment, but they were mostly subluteal (<1.0 ng/mL) after d 2. After removal, concentrations of progesterone were greater for NCIDR than UCIDR for the first 45 min, and were similar thereafter. Multiparous cows had lesser (P=0.004) concentrations than primiparous cows throughout the study. The pattern of ovarian follicular development was not affected by treatment. In EXP2, induction of onset of estrous cycles increased (P<0.01) with progesterone treatments, but was similar between NCIDR and UCIDR. Proportion of cows experiencing shorter than typical length estrous cycles after first AI tended to be greater (P=0.09) for control cows than those receiving the CIDR, and for cows remaining anestrous than those in which onset of estrous cycles was induced. Pregnancy per AI and pregnancy loss were similar among treatments. Cows that resumed estrous cyclicity prior to first AI had greater (P=0.01) pregnancy per AI. Treatment of high-producing Holstein cows that had previously initiated onset of estrous cycles with CIDR resulted in subluteal concentrations of progesterone, but in anestrous high-producing cows increased induction of estrous cycles with no effect on fertility at first insemination.  相似文献

The objective of this study was to evaluate the reproductive performance of lactating cows in seasonal dairy herds after estrus synchronization with PGF2alpha (PG) with or without supplementation with progesterone (P4). In Trial 1, synchronized cows (S1; n = 521) were compared with untreated control cows (C; n = 518) in 5 herds. Estrus of cows in the S1 group was synchronized with 2 treatments of PG (Lutalyse) 13 d apart. The breeding season started 2 d after the second PG. Cows were first bred by AI for 7 wk and then herd sires were used. Compared with C cows, estrus synchronization in the treated cows reduced the conception rate to first AI (61.1 vs 70.5%; P < 0.01) and the intervals from start of the breeding season to conception for cows conceiving to AI (11.0 vs 14.6 d; P < 0.05) or to both AI and natural mating (16.5 vs 18.4 d; P < 0.05). There was no effect on conception rate to second AI (68.8%), on pregnancy rate by Day 24 (72.3%) or Day 49 (86.3%) of the breeding season, or on the percentage of cows not pregnant at end of the breeding season (5.0%). In Trial 2, effects of P4 supplementation before the second PG on reproductive performance were evaluated in 4 herds. Estrus of each cyclic cow was synchronized with PG as in Trial 1. Half of the cows in each herd were treated with an intravaginal P4 device (CIDR) for 5 d before the second PG (S2+P4, n = 608), whereas the remaining half received no CIDR treatment (S2, n = 593). Compared with S2 cows, P4 treatment increased the estrous response rate to the second PG (89.6 vs 82.9%; P < 0.01), the conception rate to first AI (65.1 vs 59.7%; P = 0.07), the pregnancy rate by Day 6 of the breeding season (59.3 vs 49.0%; P < 0.001), and reduced the intervals from start of the breeding season to conception for cows conceiving to AI (8.6 vs 10.4 d; P < 0.10) or to both AI and natural mating (12.7 vs 16.4 d; P < 0.01). Treatment with a used CIDR from Days 16 to 21 after start of breeding to re-synchronize returns to service had no effect on conception rate to first or second AI but may decrease the conception rate to second AI in cows previously treated with CIDR. In conclusion, estrus synchronization with the double PG system can reduce fertility, while P4 supplementation for 5 d before the second PG can improve estrous response and overall reproductive performance. Stage of the estrous cycle at the time of the second PG can affect fertility following synchronization.  相似文献

The objective of the present study was to determine whether treatment of postpartum multiparous and primiparous anestrous beef cows with an intravaginal progesterone-releasing insert (CIDR) and PGF(2alpha), with and without the addition of GnRH or estradiol cypionate (ECP) at the time of CIDR insertion, is effective in stimulating onset of estrous cycles. Postpartum lactating Angus primiparous (n=47, 2 years of age, 495+/-6 kg) and multiparous (n=76, >or=3 years of age, 553+/-9 kg) cows were assigned by calving date to four blocks spaced 21-day apart. Cows were assigned sequentially by calving date to four treatment groups: (1) PGF(2alpha) (n=30), (2) CIDR-PGF(2alpha) (n=30), (3) GnRH-CIDR-PGF(2alpha) (n=33), and (4) ECP-CIDR-PGF(2alpha) (n=27). Intravaginal CIDR inserts were in place from days -7 to 0. A single 100 microg injection of GnRH or 2 mg ECP were administered on day -7, and 25mg PGF(2alpha) was administered on day 0. Day 0 averaged 38+/-1 day postpartum. Blood samples were collected on days -19, -9, 0, 5, 9, 12, 16, 19, 23, 26, and 30 for determination of plasma progesterone concentrations. Pre-treatment luteal activity (progesterone>or=1 ng/ml) was detected in 19% of primiparous and 8% of multiparous cows. Progesterone concentrations on day 0 were greater (P<0.001) in primiparous (3.2+/-0.3 ng/ml) than multiparous (2.0+/-0.2 ng/ml) cows. Following CIDR withdrawal, progesterone concentrations from days 5 to 30 were used to categorize response profiles as either: (1) treatment-induced onset of estrous cycles, (2) continued anestrus, or (3) spontaneous ovulation and subsequent formation of a CL. Incidence of treatment-induced onset of estrous cycles, which was defined as progesterone concentrations >or=1 ng/ml in three or more consecutive samples from days 9 to 19, was influenced by treatment and parity. Percentages of cows initiating estrous cycles were greater (P<0.001) in the three CIDR-treated groups than in the PGF(2alpha) group (55 and 8%, respectively). Percentages of cows initiating estrous cycles in the CIDR-PGF(2alpha), GnRH-CIDR-PGF(2alpha), and ECP-CIDR-PGF(2alpha) groups were 55, 58, and 52%, respectively. Incidence of treatment-induced estrous cycles in the three CIDR-treated groups of cows was greater (P=0.008) in primiparous (76%) than multiparous (43%) cows. Treatment of postpartum anestrous primiparous and multiparous beef cows with CIDR-PGF(2alpha) approximately 40-day postpartum provides an approach to increase the percentage of cows that have reinitiated estrous cycles by the start of the breeding season.  相似文献

Twenty pluriparous, spring-calving Brahman cows were used to determine luteal competency, as measured by serum progesterone concentrations, during the first and the second postpartum estrous cycles. Prior to and after calving, all cows were maintained in good body condition on Coastal bermudagrass pasture (IFN 1-00-703). The calves were allowed to suckle ad libitum, and sterile marker bulls were maintained with the cow herd as an aid in estrus detection throughout the trial. Cow weight and body condition score were recorded within 24 hours after calving and again at the first behavioral estrus observed. From day 1 through day 14 (day 0 = estrus) of both the first and the second postpartum estrous cycles, blood samples were collected from each cow, processed to yield serum and analyzed by radioimmunoassay for progesterone concentrations. There was a higher incidence of abnormal estrous cycles following the first postpartum estrus (35%) than following the second (5%) postpartum estrus (P<0.05). The abnormal first estrous cycles were characterized by either a short luteal phase (four cows) or by standing estrus behavior without luteal tissue formation (three cows). When serum progesterone concentrations were compared for all cows during the first estrous cycle with those during the second estrous cycle, there was less progesterone released during the cycle (P<0.05) and lower peak progesterone concentrations (P<0.10) during the first estrous cycle. However, if the abnormal cows were excluded from the analyses, there was no difference (P>0.10) in either progesterone concentrations through the 14 days measured or in peak progesterone concentrations between the first and the second postpartum estrous cycles. It can be concluded from this study that the higher incidence of abnormal luteal function following the first postpartum estrus may contribute to the decreased conception rates observed when cows are bred at their first postpartum estrus.  相似文献

Two experiments were conducted to determine if progesterone secretion and fertility would be affected by administration of human chorionic gonadotropin (hCG) before or after the first insemination. In Experiment 1, 48 Holstein heifers received 1000 IU of hCG or 1 ml of saline on Days 2, 3, and 4 of an estrous cycle. They were inseminated at the subsequent estrus. In Experiment 2, 110 Jersey and 105 Holstein cows received a single injection of 5000 IU of hCG or 5 ml of saline on Day 3 after estrus. These cows were first inseminated either at the estrus immediately preceding treatment or at the subsequent estrus. In both experiments, blood samples for determination of progesterone were collected thrice weekly for 3 to 4 wk following treatment. In Experiment 1, progesterone concentrations during mid-cycle were higher in hCG-treated heifers than in saline-treated controls. Treatment with hCG resulted in an 11% increase in the first service conception rate (P < 0.48). In Experiment 2, hCG-treated cows displayed higher progesterone secretion during mid-cycle than saline-treated herdmates. The conception rate of cows inseminated prior to hCG-treatment was not affected by treatment, but cows inseminated after treatment had a marginally lower fertility rate. The conception rate of cows receiving a repeat insemination following hCG treatment was higher than for the controls. We conclude that treatment with hCG did not improve the conception rate at the first insemination, but it may be beneficial for cows that require a repeat service.  相似文献

Poor conception rates in highly productive lactating cattle is especially prevalent in large, intensively-managed commercial herds. One of the causative factors is sub-optimal pre-implantation embryonic development which appears to result from inadequate circulating concentrations of progesterone. In the present study, the efficacy of very modest progesterone supplementation, between Days 3.5 and 10 post-AI, on pregnancy rates was determined in a commercial herd where bovine somatotropin (bST) was used as a management tool. All lactating cattle that were deemed to be in estrus and inseminated over a 4-week period were randomly assigned to either a control group (no treatment) or CIDR-1.9g (previously used for estrous synchronization) treatment from Day 3.5 to Day 10 post-AI. Milk samples were collected four times: on the day of AI, at Day 2 or 3, at Day 4 and at Day 22 post-AI and were analyzed for progesterone content. Data from a total of 130 breedings were used in the final analysis. The CIDR treatment increased circulating concentrations of progesterone in treated animals over those of control animals on Day 4 by 0.7ng/ml (P<0.05) and increased pregnancy rate from 35% (22/63) to 48% (32/67) (P=0.068). The effect of treatment was greater in first and second lactation cows, where pregnancy rates were 33% (18/55) in controls and 51% (31/61) in treated animals (P=0.03). The results of this study indicate that the timing of onset of the progesterone influence is important for successful pregnancy outcome, particularly in first and second lactation cows.  相似文献

A new analogue of PGFα, alfaprostol, was used to induce estrus in normally cycling and anestrous cows.Five groups of six animals were treated with single injections of different doses of the drug (4, 5, 6, 7 or 8 mg/animal) on day 10–11 of the estrous cycle. Six cows in anestrus, with palpable corpora lutea, were treated i.m. with 8 mg/animal. The luteolytic properties of alfaprostol were assessed by measuring milk progesterone by RIA for 96 h after treatment and by checking for the subsequent signs of estrus. The dose of 4 mg caused a slight fall in progesterone but did not induce heat; the 5 and 6 mg doses induced estrus in animals and the 7 mg dose was effective in animals. The 8 mg dose produced a sharp reduction in milk progesterone within 24 h, followed by estrus in both the normally cycling and the anestrous groups in animals. No clinical signs of drug intolerance were seen. The present results show that alfaprostol might be of great use for both cycle synchronization in normally cycling cows and for treatment of anestrus caused by a persistent corpus luteum.  相似文献

A controlled trial investigating the effect on conception of administration of 250 μg of gonadotropin-releasing hormone (GnRH) at artificial insemination (AI) in dairy cows in seasonal or split calving herds was conducted. Time of detection of estrus, body condition, extent of estrous expression, treatment, breed, age and milk production from the most recent herd test of the current lactation was recorded. Cows were tested for pregnancy with fetal aging between 35 and 135 days after AI. Sixteen herds provided 2344 spring-calved cows and 3007 inseminations. Logistic regression adjusting for clustering at herd level was used to examine the effect of treatment for first (2344) and second (579) inseminations separately. For first AI, treatment significantly improved conception rate in cows with milk protein concentrations of 3.75% or greater and for cows with milk protein concentrations between 3.00% and 3.50% and less than 40 days calved; increased conception rate from 41.2% to 53.4%. Treatment reduced conception rates in cows with milk protein concentrations of 2.75% or less. Treating only cows identified as responding positively to treatment (11% of all study cows) was estimated to increase first service conception rate in herds from 48.1% to 49.4%. There was no significant effect of treatment on conception to second AI, nor any significant interactions. These findings indicate that GnRH at AI should be limited to the sub-group cows most likely to respond. The positive effect of GnRH at AI may be mediated through improved oocyte maturation and/or improved luteal function, rather than by reducing AI-to-ovulation intervals.  相似文献

The objectives of this study were to evaluate factors associated with resumption of postpartum estrous cycles and embryonic survival in lactating dairy cows. Holstein cows, 6396 from four dairy farms were evaluated to determine the relationships among parity, body condition score (BCS) at calving and at AI, season of year when cows calved, and milk yield on resumption of postpartum estrous cycles by 65 days postpartum, and all the previous variables, estrual or anestrus and AI protocol on conception rates and embryonic survival at the first postpartum insemination. Cows had their estrous cycle pre-synchronized with two PGF injections given 14 days apart and were inseminated between 69 and 82 days postpartum following either an estrous or ovulation synchronization protocol initiated 12–14 days after the presynchronization. Blood was sampled and analyzed for progesterone twice, 12–14 days apart, to determine whether cows had initiated onset of estrous cycles after calving. Cows were scored for body condition in the week after calving, and again at AI, between 69 and 82 days postpartum. Pregnancy was diagnosed at 30 ± 3 and 58 ± 3 days after AI. Farm influenced all reproductive outcomes evaluated. More (P < 0.0001) multiparous than primiparous cows had initiated estrous cycles. Onset of estrous cycles was also influenced (P < 0.01) by BCS at calving and at AI, BCS change, season, and milk yield. More (P < 0.001) cows that had initiated estrous cycles than anestrous cows were pregnant at 30 and 58 days after AI, but anestrus did not affect pregnancy loss. Conception rates were also influenced (P < 0.01) by parity, BCS at calving and AI, BCS change, and season; however, milk yield and insemination protocol were not associated with conception rates at 30 and 58 days after AI. Factors that reduced conception rate on day 30 after AI also increased pregnancy loss between 30 and 58 days of gestation. Improving BCS at calving and AI, minimizing losses of BCS after calving, and hastening onset of estrous cycles early postpartum are all expected to increase conception because of enhanced embryonic survival.  相似文献

Several recent experiments have reported that chronic treatment with bovine somatotropin (bST) increased the number of days open without affecting the services per conception. The physiological basis for these effects was examined. Eleven lactating Holstein cows received daily injections of bST (40 mg) and 10 received daily injections of vehicle. Treatment was initiated between 32 and 85 d post partum and continued for up to 180 d. Eight of 11 bST-treated cows experienced at least one period of extended ovarian acyclicity during treatment. Only 3 of 10 control cows did so (P = 0.05). Concentrations of progesterone during luteal phases were lower in bST-treated cows than in controls (P = 0.06). Baseline concentrations of LH were suppressed in bST-treated cows compared with those of controls (P < 0.04). Neither the pulse frequency of LH nor the expression of estrous behavior was affected by bST (P > 0.30). These results indicate that chronic administration of a high dose of bST can reduce reproduction performance by promoting ovarian acyclicity.  相似文献

To assess endocrine and morphological responses of ovaries to total weaning at parturition, 6 Zebu (Bos indicus) cows 5 years or older were investigated. Following parturition, blood samples were collected daily during the first month and twice weekly thereafter until day 60 to determine concentrations of progesterone (P4) and prostaglandin F2α metabolite. It took between 25 to 32 days to complete uterine involution.The prostaglandin metabolite remained elevated for a mean period of 14.2 days (range, 4-21) postpartum. Five of the animals resumed cyclicity with a short estrous cycle starting between days 7 to 34 and lasting between 7 and 14 days. No estrous behavior was recorded prior to the short estrous cycles, but subsequent normal-length estrous cycles were all preceded by signs of estrus. In the 1 animal that resumed cyclicity with an estrous cycle of normal length on day 37 (length 20 days), the cycle was preceded by estrous behavior. Progesterone concentrations reached a mean maximum of 4.8 nmol liter−1 during the short estrous cycles, and prostaglandin metabolite concentrations peaked while P4 concentrations were decreasing. P4 concentrations reached a mean maximum of 12.2 nmol liter−1 during the estrous cycles of normal length. The interval from parturition to the first estrous cycle of normal length varied between 16 and 48 days, and the length of the cycle was 18 to 22 days. Starting 2 days postpartum, ovaries from 5 of the cows were scanned by ultrasonography every second day until day 30 postpartum. Medium-sized follicles were detected between days 4 to 7 postpartum in 4 of the scanned cows that later had short estrous cycles. The time between parturition and the appearance of the first dominant follicle was 7.6 days (range 6-10 days). The interval between parturition and the appearance of the first ovulatory-sized follicle was 10.2 days (range 8-13 days). In 3 of the scanned cows this ovulatory-sized follicle ovulated. We conclude that cyclic ovarian activity in Zebu cows can start early in the postpartum period in the absence of offspring, and that short luteal phases, not preceded by estrous behavior, may play an important role in establishing normal postpartum ovarian activity.  相似文献

A study was conducted to determine reproductive response of primiparous beef heifers to an ovulation induction regimen during restricted dietary energy intake. Thirty-seven Barzona x Hereford heifers, maintained under drylot conditions, were utilized. Heifers were restricted in TDN on a pen basis to approximately 50% of N.R.C. recommendations for the first 90 days postpartum, then received 120% for 80 days thereafter. All animals received control injections (C) or 30 mg progesterone on day 15 postpartum followed in 48 hours by 2 mg estradiol-17beta (PE). Treated heifers not ovulating at first treatment and/or not cycling, were re-treated at 60 day postpartum, with non-cycling C heifers receiving control injections. Intact fertile bulls were maintained with the heifers from day 1 to 170 days postpartum, with visual observations for signs of estrus and breeding activity conducted twice daily during this period. At first treatment, seven of 18 heifers ovulated, one conceived and four continued to cycle. At second treatment, three of 13 conceived and seven returned to a synchronized estrus 15 to 21 days later. Although intervals to first estrous behavior and estrus favored PE heifers (P< .05), by 90 and 170 days postpartum no advantage in interval to conception or number conceiving was observed.  相似文献

Lactating cows (n = 64) were assigned randomly to shade or no shade treatments for a continuous trial (20 wk) during summer of 1976. Respirations/min and rectal temperatures were higher for no shade cows. Dry matter forage intake was 9.7% higher for shade cows, whereas water intake was 19% greater for no shade cows. Milk yield and conception rates were higher for shade managed cows. Mean plasma corticoid concentrations were high throughout the entire estrous cycle in no shade compared to shade cows (13.04 > 8.72 ng/ml). No shade cows had higher progesterone and LH concentrations, and a lower estradiol to progesterone ratio during the cycle. Results indicated that endocrine changes were indicative of recurring estrous cycles in thermal stressed lactating dairy cows. However, alterations in water intake and thermoregulation of stressed cows were associated with decreased fertility and milk production, and changes in steroid concentrations that may reduce uterine blood flow.  相似文献

Two trials involving 85 heifers and 67 cows were conducted to determine the effect of estrous cycle stage at the time of Syncro-Mate-B((R)) (SMB) treatment on interval to estrus following implant removal and on conception rate at the synchronized estrus. In Trial 1, 57 beef and 28 dairy heifers were treated with SMB on each representative day of a 22-d estrous cycle (estrus = Day 0). Beef heifers were artificially inseminated approximately 48 h after implant removal, whereas dairy heifers were inseminated 0 to 12 h after detection of estrus. Inseminations were scored by the inseminator according to their difficulty. Interval to the onset of estrus was not different between heifers treated early ( Day 11) in the cycle (35.2 +/- 7.2 h). Conception rate at the synchronized estrus was slightly higher in early-cycle heifers (22 47 = 47% ) compared to late-cycle heifers (14 38 = 37% , P = 0.2). Heifers that were difficult to inseminate had lower (P < 0.01) conception rates (2 11 = 18% ) at the synchronized estrus than heifers considered normal (21 51 = 41% ) or easier than normal to inseminate (13 23 = 57% ). In Trial 2, of the 131 beef cows synchronized, 67 that were estimated to be either early or late in the estrous cycle by progesterone analysis were utilized. Cows were treated with SMB and inseminated without regard to estrus 48-h after implant removal. Inseminations were scored as in Trial 1. Calves were separated from cows from the time of implant removal to insemination. Conception rate was higher (P < 0.05) in cows treated with SMB early ( Day 11, 16 35 = 46% ). Cows that were difficult to inseminate had a lower (P < 0.01) conception rate (0 8 = 0% ) than cows that were normal (43 94 = 46% ) or easier than normal to inseminate (13 29 = 45% ).  相似文献

Thirty-two lactating Holstein cows were assigned to 1 of 4 groups in a randomized block design using a 2 X 2 factorial arrangement of treatments. Recombinant bovine growth hormone (rbSt; 25 mg/day) or placebo was administered beginning at Day 35 or 70 postpartum. All cows began treatment approximately 3 days post-estrus. Blood samples were collected at least once daily for a 70-day period to determine the concentration of progesterone and the duration of the luteal and follicular phases. During estrous cycles 1 and 3, frequent blood samples were taken (every 10 min for 8 h) 24 and 60 h after the onset of luteal regression. These samples were assayed for luteinizing hormone (LH), and samples coincident with the second LH pulse detected were assayed for estradiol. Ultrasonography was used to determine the size of the largest ovarian follicle from Day 17 until ovulation in estrous cycles 1 and 3. Luteal life span, length of the follicular phase, and diameter of the largest follicle were not affected by treatment with rbSt. Administration of rbSt increased the concentration of progesterone in plasma during the first two luteal phases (p less than 0.01). Progesterone was elevated during the mid-luteal phase of cycle 3 in rbSt-treated cows that began treatment about Day 35 postpartum but not in cows that began treatment on Day 70 postpartum (Treatment X Stage X Day, p less than 0.01). In a previous study we showed that estrus synchronization with 2 treatments of PGF2 alpha 13 d apart reduced conception rate at the synchronized estrus and that this reduction occurred mainly in cows in the early luteal phase at the second PGF2 alpha treatment. The objective of the present study was to determine the efficacy of a synchronization regimen in which PGF2 alpha was administered during the mid- to late-luteal phase to cows that had previously been synchronized with progesterone. Spring-calving cows from 6 dairy herds were used in this study. On Day -32 (Day 1 = the start of the breeding season), cows that had calved 2 or more weeks ago were randomly assigned to a synchronization (S, n = 732) or control (C, n = 731) group. Cows in Group S were treated with an intravaginal progesterone device (CIDR) for 12 d from Day -32 to Day -20, while those in Group C were left untreated. Similar percentages of cows in Group S (80.6%) and C (82.9%) had cycled by Day -7. The CIDR treatment synchronized the onset of estrus, resulting in 92.9% of cows in estrus being detected within 7 d after CIDR removal. Cows in Group S that had cycled by Day -7 were treated with PGF2 alpha (25 mg, i.m., Lutalyse) on Day -2. Cows in both groups that were anestrous on Day -7 were treated with a combination of progesterone and estradiol benzoate (EB) to induce estrus and ovulation (CIDR and a 10 mg EB capsule on Day -7, CIDR removal on Day -2, and injection of 1 mg EB 48 h after CIDR removal). The PGF2 alpha treatment synchronized the onset of estrus in 87.5% of the cows. Group S and C cows had similar conception rates to first (61.0 vs 58.3%) and second (58.4 vs 60.9%) AI; similar pregnancy rates over the AI period (82.8 vs 79.2%) and over the whole breeding season (91.9 vs 90.6%); and required a similar number of services per pregnancy to AI (1.7 vs 1.8). The interval from the start of the breeding season to conception for cows conceiving to AI or to combined AI and natural mating was shorter (P < 0.001) by 5.7 and 6.2 d, respectively, for the Group S cows. It is concluded that the treatment regimen tested in the present study achieved satisfactory estrus synchronization, had no detrimental effect on fertility at the synchronized estrus, and shortened the interval from start of the breeding season to conception.  相似文献

Our working hypothesis was that the low concentrations of progesterone (P4) and synthetic progestins administered in hormonal regimens to control estrous cycles of cows would have similar effects on secretion of LH and 17 beta-estradiol (E2). In addition, we hypothesized that concentrations of exogenous P4 typical of the midluteal phase of the estrous cycle and the corpus luteum (CL) would have similar effects on LH and E2, and the effects would be different from those of synthetic progestins and low concentrations of P4. Cows (n = 29) were randomly assigned to one of five treatment groups: 1) one Progesterone Releasing Intravaginal Device (1PRID; n = 6); 2) two PRIDs (2PRID; n = 6); 3) norgestomet, as in Syncro-Mate-B regimen (SMB; n = 6); 4) melengestrol acetate (MGA; 0.5 mg/day; n = 5); and 5) control (CONT; n = 6). Treatments were administered for 9 days (Day 0 = initiation of treatment). All cows from 1PRID, 2PRID, SMB, and MGA groups were injected with prostaglandin F2 alpha (PGF2 alpha) on Days 2 and 5 of the treatment period to regress CL. Cows in the 1PRID and SMB groups were also administered exogenous estrogen according to the respective estrous synchronization protocol for these products. Daily blood samples were collected from Day 0 to 35 to determine concentrations of P4. On Day 8, blood samples were collected at 15-min intervals for 24 h to determine pattern of LH secretion. To evaluate by means of blood progesterone levels and estrous expression the effect of the injection of 25 mg of PGF2alpha in Zebu cows, 17 Indobrazil cows with a palpable corpus luteum (CL) were injected with PGF2alpha and blood samples taken every 6 h in Experiment I. In Experiment II, 15 cows from the previous experiment at the same stage of diestrus were injected with PGF2alpha and bled every 4 h for 5 d. Progesterone levels had declined by 50% 6 h after injection in all cows and dropped to below 1 ng/ml by 30 h. Estrus was observed in 47% of the animals in Experiment I and 60% in Experiment II. Ovulation was detected accurately in 29% of the animals at 33.6 +/- 11.6 h after the onset of estrus. The correlation between finding a CL by rectal palpation and levels of progesterone higher than 1 ng/ml of blood was 86 and 93% in Experiments I and II, respectively. These studies indicate that the injection of 25 mg of PGF2alpha is sufficient to produce luteolysis in Zebu cattle, although estrous expression is poor.  相似文献

This study was undertaken to elucidate the possible role of endotcxin in mediating premature luteolysis in the well- documented phenomenon of short estrous cycles in postpartum dairy cows. Four groups of Holstein heifers (n = 4 to 6 each) received either intrauterine infusion of sterile culture medium (Group I); intrauterine infusion of Escherichia coli (E. coli ) endotoxin (5 mug/kg) in sterile culture medium (Group II); intrauterine administration of 10 ml of a 24-h culture of a strain of E. coli isolated from the uterus of a cow with metritis (approximately 10(9) colony forming units/ml; Group III); or intravenous administration of E. coli endotoxin (5 mug/kg; Group IV) on Day 7-9 of the estrous cycle. Blood samples were collected every 48 h during the pretreatment estrous cycle and up to the administration of the experimental treatment, thereafter 4-h samples were collected for 5 d. Sample collection was then performed every 48 h for the remainder of the treatment cycle and the post treatment cycle. Serum concentrations of progesterone and plasma concentrations of 15-keto-13, 14-dihydroprostaglandin F(2alpha) (PGFM) were determined by radionmmunoassay. Intrauterine infusion of endotoxin had no effect on the cycle length or on hormone concentrations, while infusion of viable E. coli organisms tended to shorten the estrous cycle. Intravenous administration of endotoxin produced a sharp increase in both progesterone and PGFM concentrations, followed by a transient decrease in progesterone concentrations. Cycle length remained unchanged. It was concluded that the intact endometrium prevents the uptake of endotoxin although pathogenic E. coli organisms may disrupt the endometrial integrity sufficiently to shorten the estrous cycle by premature luteolysis. It is postulated that intravenous administration of endotoxin influences luteal function by the activation of the arachidonic acid cascade, by a direct effect on the corpus luteum, or via other mediators.  相似文献
